Penaeidins, members of a new family of antimicrobial peptides constitutively produced and stored in the haemocytes of penaeid shrimp, display antimicrobial activity against bacteria, and fungi. Here, a DNA sequence encoding the mature Ch-penaeidin peptide was cloned into the pPIC9K vector and transformed into Pichia pastoris. The transformed cells were screened for multi-copy plasmids using increasing concentrations of G418. Positive colonies carrying chromosomal integrations of the Chp gene were identified by phenotype and PCR. When transformed cells were induced with methanol, SDS-PAGE and Western blotting revealed the production of a similar to6100 Da recombinant CHP (rCHP) expression product. Large scale expression revealed that rCHP was produced at 108 mg/L under optimal conditions in the highest Chp-producing P. pastoris clone. The antimicrobial activities of rCHP were studied by liquid phase analysis, which revealed that rCHP exhibited activities against some Gram-negative and Gram-positive bacteria, but had a relatively low activity against some fungi. Purification of rCHP by cation exchange chromatography and subsequent automated amino acid sequencing revealed the presence of four additional amino acids (YVEF) at the N-terminus that belonged to the cleaved fusion signal peptide; these residues may account for the observed decrease in antifungal activity. Together, these observations indicate that rCHP is an effective antimicrobial peptide that can be successfully produced at high levels in the yeast, and therefore may be a potential antimicrobial candidate for practical use. A new family of neural network architectures is presented. This family of architectures solves the problem of constructing and training minimal neural network classification expert systems by using switching theory. The primary insight that leads to the use of switching theory is that the problem of minimizing the number of rules and the number of IF statements (antecedents) per rule in a neural network expert system can be recast into the problem of minimizing the number of digital gates and the number of connections between digital gates in a Very Large Scale Integrated (VLSI) circuit. The rules that the neural network generates to perform a task are readily extractable from the network's weights and topology. Analysis and simulations on the Mushroom database illustrate the system's performance.

The Haemophilus influenzae HMW1 adhesin is a high-molecular weight protein that is secreted by the bacterial two-partner secretion pathway and mediates adherence to respiratory epithelium, an essential early step in the pathogenesis of H. influenzae disease. In recent work, we discovered that HMW1 is a glycoprotein and undergoes N-linked glycosylation at multiple asparagine residues with simple hexose units rather than N-acetylated hexose units, revealing an unusual N-glycosidic linkage and suggesting a new glycosyltransferase activity. Glycosylation protects HMW1 against premature degradation during the process of secretion and facilitates HMW1 tethering to the bacterial surface, a prerequisite for HMW1-mediated adherence. In the current study, we establish that the enzyme responsible for glycosylation of HMW1 is a protein called HMW1C, which is encoded by the hmw1 gene cluster and shares homology with a group of bacterial proteins that are generally associated with two-partner secretion systems. In addition, we demonstrate that HMW1C is capable of transferring glucose and galactose to HMW1 and is also able to generate hexose-hexose bonds. Our results define a new family of bacterial glycosyltransferases.

A family of stochastic gradient algorithms and their behaviour in the data echo cancellation work platform are presented. The cost function adaptation algorithms use an error exponent update strategy based on an absolute error mapping, which is updated at every iteration. The quadratic and nonquadratic cost functions are special cases of the new family. Several possible realisations are introduced using these approaches. The noisy error problem is discussed and the digital recursive filter estimator is proposed. The simulation outcomes confirm the effectiveness of the proposed family of algorithms.

The microfilamentous green alga Uronema curvatum is widely distributed along the western and eastern coasts of the north Atlantic Ocean where it typically grows on crustose red algae and on haptera of kelps in subtidal habitats. The placement of this marine species in a genus of freshwater Chlorophyceae had been questioned. Molecular phylogenetic analysis of nuclear-encoded small and large subunit rDNA sequences reveal that U. curvatum is closely related to the ulvophycean order Cladophorales, with which it shares a number of morphological features, including a siphonocladous level of organization and zoidangial development. The divergent phylogenetic position of U. curvatum, sister to the rest of the Cladophorales, along with a combination of distinctive morphological features, such as the absence of pyrenoids, the diminutive size of the unbranched filaments and the discoid holdfast, warrants the recognition of a separate genus, Okellya, within a new family of Cladophorales, Okellyaceae. The epiphytic Urospora microscopica from Norway, which has been allied with U. curvatum, is revealed as a member of the cladophoralean genus Chaetomorpha and is herein transferred to that genus as C. norvegica nom. nov.

Host defense peptides (HDPs) are an evolutionarily conserved component of the innate immune response found in all living species. They possess antimicrobial activities against a broad range of organisms including bacteria, fungi, eukaryotic parasites, and viruses. HDPs also have the ability to enhance immune responses by acting as immunomodulators. We discovered a new family of HDPs derived from pathogenic helminth (worms) that cause enormous disease in animals and humans worldwide. The discovery of these peptides was based on their similar biochemical and functional characteristics to the human defense peptide LL-37. We propose that these new peptides modulate the immune response via molecular mimicry of mammalian HDPs thus providing a mechanism behind the anti-inflammatory properties of helminth infections.

The real time implementation of an efficient signal compression technique, Vector Quantization (VQ), is of great importance to many digital signal coding applications. In this paper, we describe a new family of bit level systolic VLSI architectures which offer an attractive solution to this problem. These architectures are based on a bit serial, word parallel approach and high performance and efficiency can be achieved for VQ applications of a wide range of bandwidths. Compared with their bit parallel counterparts, these bit serial circuits provide better alternatives for VQ implementations in terms of performance and cost. © 1995 Kluwer Academic Publishers.

For the first time, the hydrogenation/hydrogenolysis of a range of disulfides has been achieved over a supported palladium catalyst using hydrogen under relatively benign conditions. These unexpected results demonstrate that it is possible to avoid the poisoning of the catalyst by either the nitrogen-containing groups or the sulfur species, allowing both efficient reaction and recycling of the catalyst under the proper conditions (e.g., at low temperatures). A slight loss in activity was found on recycling; however, the catalyst activity can be recovered using hydrogen pretreatment. The reaction mechanism for the hydrogenolysis and hydrogenation of ortho-, meta-, and para-dinitrodiphenyldisulfide to the corresponding aminothiophenol has been elucidated. Density functional theory calculations were used to investigate the adsorption mode of the dinitrodiphenyldisulfides; a clear dependence on adsorption geometry was found regarding whether the molecule is cleaved at the S-S bond before the reduction of the nitro group or vice versa. This study demonstrates the versatility of these catalysts for the hydrogenation/hydrogenolysis of sulfur-containing molecules, which normally are considered poisons, and will extend their use to a new family of substrates. Sponge classification has long been based mainly on morphocladistic analyses but is now being greatly challenged by more than 12 years of accumulated analyses of molecular data analyses. The current study used phylogenetic hypotheses based on sequence data from 18S rRNA, 28S rRNA, and the CO1 barcoding fragment, combined with morphology to justify the resurrection of the order Axinellida Lévi, 1953. Axinellida occupies a key position in different morphologically derived topologies. The abandonment of Axinellida and the establishment of Halichondrida Vosmaer, 1887 sensu lato to contain Halichondriidae Gray, 1867, Axinellidae Carter, 1875, Bubaridae Topsent, 1894, Heteroxyidae Dendy, 1905, and a new family Dictyonellidae van Soest et al., 1990 was based on the conclusion that an axially condensed skeleton evolved independently in separate lineages in preference to the less parsimonious assumption that asters (star-shaped spicules), acanthostyles (club-shaped spicules with spines), and sigmata (C-shaped spicules) each evolved more than once. Our new molecular trees are congruent and contrast with the earlier, morphologically based, trees. The results show that axially condensed skeletons, asters, acanthostyles, and sigmata are all homoplasious characters. The unrecognized homoplasious nature of these characters explains much of the incongruence between molecular-based and morphology-based phylogenies. We use the molecular trees presented here as a basis for re-interpreting the morphological characters within Heteroscleromorpha. The implications for the classification of Heteroscleromorpha are discussed and a new order Biemnida ord. nov. is erected.

Friedel–Crafts alkylation of benzene with 1-decene was catalysed by a new family of liquid Lewis acids: liquid coordination complexes (LCCs). LCCs are prepared by mixing a metal halide (e.g. GaCl3) and a donor molecule (e.g. N,N-dimethylacetamide, urea, or trioctylphosphine oxide), with the metal halide typically used in excess. This leads to the formation of a eutectic mixture comprised of charged and neutral species in a dynamic equilibrium. GaCl3-based LCCs were used in catalytic amounts, giving high reaction rates under ambient conditions, with selectivities to 2-phenyldecane superior to those previously reported in the literature. The influence of reaction conditions and catalyst composition on the reaction rate and selectivity was investigated. Optimised reaction conditions were suggested. This exploratory study offers promise with regard to the development of safer, LCC-based alternatives to HF in industrial alkylations.

O conhecimento sobre famílias envelhecidas é ainda escasso. Neste âmbito, a pesquisa tem incidido nos cuidados familiares a idosos dependentes, focando os problemas de saúde, dependência funcional e declínio cognitivo. Esta investigação pretende contribuir para aprofundar o conhecimento sobre as famílias envelhecidas, assumindo uma perspetiva normativa e desenvolvimental, e contemplando a diversidade de contextos de vida e envelhecimento. O capítulo 1 centra casais compostos por pessoas idosas, e tem por objetivos: caracterizar a estrutura, dinâmica e valores do agregado familiar dos casais idosos; evidenciar valores e dinâmica relacional dos casais idosos. A amostra compreende 136 participantes, a quem foi administrado um questionário sobre a fase última do ciclo de vida familiar (Cerveny,1997). A análise de dados efetuou-se com recurso ao programa de análise de dados estatística SPSS 17.1. Os resultados indicam que os casais vivem predominantemente em casal, com uma dinâmica relacional do agregado caracterizada pelo respeito, diálogo e carinho; dinâmica relacional do casal caracterizada por clima afetuoso, amizade e diálogo, e valores assentes no amor, diálogo e convívio familiar. A dinâmica relacional do casal é pautada por atividades de lazer realizadas em conjunto e vida sexual tão boa como antes; os valores dão ao casamento significados de realização pessoal e perpetuação através dos filhos na juventude, e adaptação e descoberta na velhice. O capítulo 2 foca a construção da integridade familiar considerando a diversidade de contextos socioeconómicos (pessoas idosas que viveram em contexto de pobreza ao longo da vida), socioculturais (ex-emigrantes portugueses) e novas formas de famílias (homens homossexuais). Foi aplicada uma entrevista semiestruturada (King & Wynne, 2004) a uma amostra de 12, 20 e 10 pessoas, respetivamente. A análise de dados foi efetuada com base na análise de conteúdo com recurso a juízes independentes baseada na grounded theory, contudo no caso do contexto socioeconómico recorreu-se ao programa de análise de dados qualitativa N-Vivo 7. Os resultados sugerem que a diversidade de contextos analisada coloca desafios à rutura familiar o que pode potenciar o caminho da desconexão e alienação. Contudo, o contexto das significações exerce um papel fundamental na construção da integridade familiar. A redefinição da identidade associada a uma filosofia de vida que enfatize as forças em vez dos fracassos parece determinar a construção da integridade familiar, contudo existem especificidades. Relativamente ao contexto socioeconómico: as pessoas idosas no caminho da integridade revelam um sentido de autovalorização (ter vivido uma vida significativa) apesar da pobreza; as pessoas idosas no caminho da desconexão/alienação alimentam sentimentos de insignificância devido à escassez de recursos económicos. Ainda neste contexto, os valores (princípios de conduta) reinterpretam a identidade ao longo da vida e permitem compreender que a integridade familiar ocorre quando ser pobre é encarado pelas conquistas; a desconexão/alienação emerge quando ser pobre incorpora sentimentos de desvalorização e inferioridade. No contexto sociocultural, as pessoas idosas ex-emigrantes cujo processo de emigração se desenvolveu em família (a família está envolvida no processo de emigração e funciona como um pilar desde a fase de decisão até ao regresso) desenvolveram uma filosofia de vida assente numa atitude ativa e solidária e estão em integridade familiar; as pessoas em desconexão relatam episódios de conflito familiar que marcam a trajetória de emigração, e uma atitude passiva na resolução desses conflitos até à atualidade; as pessoas em alienação familiar, cujo processo de emigração se desenrolou de forma solitária, desenvolvem uma filosofia de vida assente na luta solitária: a sua força e identidade estão em enfrentar tudo sem precisar de ninguém. Relativamente às novas formas de família, a integridade familiar evolui desde a revelação da homossexualidade (em idade jovem) e conclui-se na velhice quando a homossexualidade se torna um legado. A desconexão parece evoluir da luta constante da falha da aceitação da homossexualidade pela família e outras pessoas significativas. O capítulo 3 analisa as trajetórias de vida de homens homossexuais atualmente idosos, para compreender melhor a influência da homossexualidade e os principais eventos. Adotou-se a técnica da linha de acontecimentos de vida (Acquaviva et al., 2007), aplicada a 10 participantes com 60 anos ou mais. Os resultados sugerem que vários eventos de vida influenciam o curso de vida: i) o autoconhecimento da homossexualidade; ii) tentar passar por heterossexual; iii) assumir a homossexualidade (explicita ou implicitamente); iv) sentir limitações e desafios relacionados com o ser idoso e homossexual. O capítulo 4 procurou alargar a perspetiva do envelhecimento considerando uma abordagem transcultural. Assim, realizou-se um estudo numa comunidade indígena (Guarani Mbya, Brasil). Neste estudo analisase o modo de viver e ser idoso nessa comunidade. A amostra compreende 6 participantes a quem foi administrada uma entrevista aberta. Este estudo contemplou ainda a observação com registo etnográfico e realização de um diário de bordo. A análise de conteúdo efetuou-se com apoio do software de dados qualitativa WebQDA 1.4.3. Os resultados sugerem o papel das pessoas idosas na preservação de uma cultura ágrafa, garantindo que as tradições estejam presentes nas gerações atuais através da oralidade. A adoção de lentes normativas no estudo e compreensão das famílias envelhecidas permite compreender as tarefas desenvolvimentais e normativas no fim da vida.

Niobium oxides have been pointed as an alternative to tantalum in the production of solid electrolytic capacitors, with advantages regarding the dielectric constant, density and price. In this work, it is intended to create a new family of niobium oxides based capacitors, adapting the technology and production line currently used with tantalum. Despite the known potentialities of niobium oxides, and many types of niobates, in several technological applications, the understanding of these oxide systems is still noticeably insufficient. Hence, a careful bibliographic review is shown, which evidences the complexity of these materials, the difficulty in identifying of their different phases and polymorphs, as well as in the interpretation of their properties. In this context, several fundamental studies on niobium oxides are presented, namely structural, microstructural, optical and electrical characterizations, which allow not only to contribute in an important way for the general knowledge of the physical properties of these materials, but also to advance to a sustained development of the niobium oxides based solid electrolytic capacitors. Several processing parameters were studied, clearing the way towards the creation of a prototype. It was also decided to perform a preliminary study on the synthesis and characterization of other oxide systems based in niobium, namely rare-earth orthoniobates (RENbO4), which interest has been related to their optical properties and protonic conductivity. Hence, single and polycrystalline samples of RENbO4 were synthesized and characterized structural, optical and electrically, leaving open an interesting future work.

Work in the area of molecule-based magnetic and/or conducting materials is presented in two projects. The first project describes the use of 4,4’-bipyridine as a scaffold for the preparation of a new family of tetracarboxamide ligands. Four new ligands I-III have been prepared and characterized and the coordination chemistry of these ligands is presented. This project was then extended to exploit 4,4’-bipyridine as a covalent linker between two N3O2 macrocyles. In this respect, three dimeric macrocycles have been prepared IV-VI. Substitution of the labile axial ligands of the Co(II) complex IV by [Fe(CN)6]4- afforded the self-assembly of the 1-D polymeric chain {[Co(N3O2)H2O]2Fe(CN)6}n•3H2O that has been structurally and magnetically characterized. Magnetic studies on the Fe(II) complexes V and VI indicate that they undergo incomplete spin crossover transitions in the solid state. Strategies for the preparation of chiral spin crossover N3O2 macrocycles are discussed and the synthesis of the novel chiral Fe(II) macrocyclic complex VII is reported. Magnetic susceptibility and Mössbauer studies reveal that this complex undergoes a gradual spin crossover in the solid state with no thermal hysteresis. Variable temperature X-ray diffraction studies on single crystals of VII reveal interesting structural changes in the coordination geometry of the macrocycle accompanying its SCO transition. The second project reports the synthesis and characterization of a new family of tetrathiafulvalene derivatives VIII – XII, where a heterocyclic chelating ligand is appended to a TTF donor via an imine linker. The coordination chemistries of these ligands with M(hfac)2.H2O (M( = Co, Ni, Mn, Cu) have been explored and the structural and magnetic properties of these complexes are described.
