Il termine biochar definisce il prodotto solido derivante dalla pirolisi di un qualsiasi materiale organico, con lo specifico scopo di essere applicato nei suoli sia per fini agronomici che di gestione ambientale. Un suo utilizzo in maniera "responsabile" richiede però una piena comprensione delle sue proprietà e dei meccanismi che controllano la sua attività nel terreno, che dipendono dalla biomassa di partenza e dalle condizioni di sintesi tramite pirolisi. Infatti le condizioni di pirolisi, in particolare la temperatura di processo e il tempo di residenza, determinano biochar con caratteristiche differenti. In questo lavoro di tesi sono stati prodotti biochar da due diverse tipologie di biomassa residuale ampiamente disponibili (stocchi di mais e pollina). Per ciascuna biomassa sono state scelte tre condizioni di pirolisi (400°C x 20 minuti, 500°C x 10 minuti e 600°C x 5 minuti). Sui biochar ottenuti sono state effettuate le seguenti determinazioni: analisi elementare, Pirolisi‐GC‐MS, idrocarburi policiclici aromatici (IPA), acidi grassi volatili (VFA), azoto ammoniacale (N‐NH4 +), pH, conduttività elettrica e ritenzione idrica. Infine i biochar sintetizzati sono stati utilizzati per fare due test di germinazione per valutare l'effetto sulla formazione delle prime strutture di crescita delle plantule, tramite test di tossicità brevi con piastre Petri. Il primo test è stato condotto a concentrazione crescente di miscele acqua/biochar (2, 5, 40 e 100 g/L sulla base delle quantità di biochar utilizzate come ammendante nel suolo), sulla germinazione seguendo la metodologia normata dalla ISO 11269:2012. I semi utilizzati nel primo test sono stati quelli del crescione (Lepidium sativum L.) come specie dicotiledone, e del sorgo (Sorghum saccharatum M.) come monocotiledone. Il secondo saggio di tossicità eseguito è stato quello descritto dalla normativa in materia UNI 11357, valutando l'eventuale effetto di tossicità alla massima concentrazione delle varie tipologie di biochar, utilizzando come specie dicotiledoni il cetriolo (Cucumis sativus L.) ed il crescione (Lepidium sativum L.), come monocotiledone il sorgo (Sorghum saccharatum M.). Per i biochar da stocchi di mais, rappresentativi di biomasse erbacee e con diverso grado di carbonizzazione, non si osservano effetti apprezzabili alle condizioni di uso agricolo. Nel caso dei biochar da pollina si osservano invece inibizioni alla germinazione sin dalle concentrazioni più basse. In particolare, quello pirolizzato a 400°C mostra un potenziale effetto tossico più marcato, probabilmente associato ad un contenuto di IPA e VFA superiore a quello degli altri biochar.

In this thesis the potential risks associated to the application of biochar in soil as well the stability of biochar were investigated. The study was focused on the potential risks arising from the occurrence of polycyclic aromatic hydrocarbons (PAHs) in biochar. An analytical method was developed for the determination of the 16 USEPA-PAHs in the original biochar and soil containing biochar. The method was successfully validated with a certified reference material for the soil matrix and compared with methods in use in other laboratories during a laboratory exercise within the EU-COST TD1107. The concentration of 16 USEPA-PAHs along with the 15 EU-PAHs, priority hazardous substances in food, was determined in a suite of currently available biochars for agricultural field applications derived from a variety of parent materials and pyrolysis conditions. Biochars analyzed contained the USEPA and some of the EU-PAHs at detectable levels ranging from 1.2 to 19 µg g-1. This method allowed investigating changes in PAH content and distribution in a four years study following biochar addition in soils in a vineyard (CNR-IBIMET). The results showed that biochar addition determined an increase of the amount of PAHs. However, the levels of PAHs in the soil remained within the maximum acceptable concentration for European countries. The vineyard soil performed by CNR-IBIMET was exploited to study the environmental stability of biochar and its impact on soil organic carbon. The stability of biochar was investigated by analytical pyrolysis (Py-GC-MS) and pyrolysis in the presence of hydrogen (HyPy). The findings showed that biochar amendment significantly influence soil stable carbon fraction concentration during the incubation period. Moreover, HyPy and Py-GC-MS were applied to biochars deriving from three different feedstock at two different pyrolysis temperatures. The results evidenced the influence of feedstock type and pyrolysis conditions on the degree of carbonisation.

Il processo di compostaggio è il metodo più antico ed al tempo stesso innovativo di trasformazione dei rifiuti. La trasformazione in impianti di compostaggio industriale ricrea lo stesso processo semplice ma in scala maggiore processando molte tonnellate/anno di rifiuto con particolare riguardo a quello proveniente da raccolta differenziata. Il presente elaborato, oltre ad illustrare le nuove tecnologie di produzione nelle realtà locali della zona Romagnola, ha inteso indagare un aspetto alternativo ed innovativo di valutazione del compost. A supporto di quanto già previsto alla specifica normativa di settore e nell'ottica dell'ottimizzazione del processo, è stata valutata la possibilità di uso della tecnica analitica di microestrazione in fase solida (SPME) e della tecnica strumentale con naso elettronico. Si è inteso verificare anche come l'utilizzo di carbone vegetale, aggiunto alla fase di maturazione, possa apportare un contributo significativo nel controllo delle emissioni odorigene, problematica diffusa nelle aree limitrofe ad impianti di compostaggio. L'utilizzo delle tecniche di estrazione in fase solida (SPME) ha dimostrato che durante il processo le componenti odorigene tendono a ridursi notevolmente modificandosi in modo apprezzabile e valutabile mediante analisi GC-MS. L'aggiunta di carbone vegetale alla fase di maturazione contribuisce alla riduzione delle emissioni odorigene. I risultati ottenuti evidenziano come l'utilizzo dei sistemi proposti possa portare ad un'economicità qualora si riscontrassero tempi di maturazione variabili per le tipologie di materiale trattato, riducendo così i tempi di permanenza nei tunnel di maturazione e come le tecniche SPME e naso elettronico possano essere accoppiate per ottenere risultati complementari così da costruire un'informazione complessiva completa. Possibili sviluppi dello studio potrebbero essere improntati alla verifica puntuale del grado di maturazione in relazione alla tipologia del materiale in ingresso ed alla stagionalità nonché all'utilizzo di tali tecniche ad altre matrici e nell'ambito dell'individuazione della presenza di composti indesiderati in matrici ambientali.

Le tecniche di imaging al seno per la rilevazione di tumori stanno interessando un notevole numero di gruppi di ricerca in campo biomedico soprattutto negli ultimi anni. In particolare si cercano metodologie innovative in quanto le attuali tecniche già sviluppate e utilizzate in campo clinico, mammografia a raggi X e risonanza magnetica (MRI), presentano alcuni limiti. Questa tesi si focalizzerà in particolare su una di queste tecniche emergenti: l’imaging a microonde (MWI). Questo metodo infatti, limita notevolmente i costi, evita disagi per la paziente come la compressione del seno, penetra in modo ottimale nei tessuti e non li ionizza. La MWI si basa sulle diverse proprietà dielettriche, permittività e conduttività, dei vari tessuti che costituiscono la mammella, in particolare tra tessuto sano e maligno. Lo scopo di questa tesi è quello di analizzare tali proprietà dielettriche, le diversità che i vari tessuti presentano e come tutto ciò venga sfruttato per ottenere l’imaging della mammella. In particolare questo lavoro si propone di riportare e analizzare i principali studi sui tessuti biologici della mammella compiuti nel corso degli anni riguardo queste proprietà dielettriche e i rispettivi risultati ottenuti. Si tratteranno inoltre i mezzi di accoppiamento: soluzioni in cui è immerso l’assetto antenne-oggetto che minimizzano la riflessione del segnale sulla pelle e assicurano una migliore qualità di immagine.

A universal and robust analytical method for the determination of Δ9-tetrahydrocannabinol (THC) and two of its metabolites Δ9-(11-OH)-tetrahydrocannabinol (11-OH-THC) and 11-nor-Δ9-carboxy-tetrahydrocannabinol (THC-COOH) in human whole blood was developed and validated for use in forensic toxicology. Protein precipitation, integrated solid phase extraction and on-line enrichment followed by high-performance liquid chromatography separation and detection with a triple quadrupole mass spectrometer were combined. The linear ranges used for the three cannabinoids were from 0.5 to 20 ng/mL for THC and 11-OH-THC and from 2.5 to 100 ng/mL for THC-COOH, therefore covering the requirements for forensic use. Correlation coefficients of 0.9980 or better were achieved for all three analytes. No relevant hydrolysis was observed for THC-COOH glucuronide with this procedure--in contrast to our previous GC-MS procedure, which obviously lead to an artificial increase of the THC-COOH concentration due to the hydrolysis of the glucuronide-conjugate occurring at high pH during the phase-transfer catalyzed methylation step.

Synthetic polymers constitute a wide class of materials which has enhanced the quality of human life, providing comforts and innovations. Anyway, the increasing production and the incorrect waste management, are leading to the occurrence of polymers in the environment, generating concern. To understand the extent of this issue, analytical investigation holds an essential position. Standardised methods have not established yet, and additional studies are required to improve the present knowledge. The main aim of this thesis was to provide comprehensive information about the potential of pyrolysis coupled with gas-chromatography and mass spectrometry (Py-GC-MS) for polymers investigation, from their characterisation to their identification and quantification in complex matrices. Water-soluble (poly(dimethylsiloxanes), PDMS bearing poly(ethylene glycol), PEG, side chains) and water-insoluble polymers (microplastics, MPs, and bioplastics) were studied. The different studies revealed the possibility to identify heterogeneous classes of polymers, fingerprinting the presence of PDMS copolymers and distinguishing chemically different polyurethanes (PURs). The occurrence of secondary reactions in pyrolysis of polymer mixtures was observed as possible drawback. Pyrolysis products indicative of secondary reactions and their reaction mechanisms were identified. Py-GC-MS also revealed its fundamental role for the identification of polymers composing commercial bioplastics items based. The results aided to identify chemicals that have the potential to migrate in sea waters. Investigations of environmental samples demonstrated the capability of Py-GC-MS to provide reliable, reproducible and comparable results about polymers in complex matrices (PEG-PDMS in sewage sludges and PURs and other MPs in road dusts and spider webs). Criticisms were especially found in quantitation, such as the retrieval reference materials, the construction of reliable calibration protocols and the occurrence of bias due to interferences between pyrolysis products. This thesis pursues the greater purpose to develop harmonised and standardised methods for environmental investigations of polymers, that is fundamental to assess the real state of the environment.

Essential oils (EO) obtained from twenty medicinal and aromatic plants were evaluated for their antimicrobial activity against the oral pathogens Candida albicans, Fusobacterium nucleatum, Porphyromonas gingivalis, Streptococcus sanguis and Streptococcus mitis. The antimicrobial activity of the EO was evaluates by microdilution method determining Minimal Inhibitory Concentration. Chemical analysis of the oils compounds was performed by Gas chromatography-mass spectrometry (CG-MS). The most active EO were also investigated as to their actions on the biolfilm formation. The most of the essential oils (EO) presented moderate to strong antimicrobial activity against the oral pathogens (MIC--Minimal Inhibitory Concentrations values between 0.007 and 1.00 mg/mL). The essential oil from Coriandrum sativum inhibited all oral species with MIC values from 0.007 to 0.250 mg/mL, and MBC/MFC (Minimal Bactericidal/Fungicidal Concentrations) from 0.015 to 0.500 mg/mL. On the other hand the essential oil of C. articulatus inhibited 63.96% of S. sanguis biofilm formation. Through Scanning Eletronic Microscopy (SEM) images no changes were observed in cell morphology, despite a decrease in biofilm formation and changes on biofilm structure. Chemical analysis by Gas Chromatography-Mass Spectrometry (GC-MS) of the C. sativum essential oil revealed major compounds derivatives from alcohols and aldehydes, while Cyperus articulatus and Aloysia gratissima (EOs) presented mono and sesquiterpenes. In conclusion, the crude oil from C. articulatus exhibited the best results of antimicrobial activity e ability to control biofilm formation. The chemical analysis showed the presence of terpenes and monoterpenes such as a-pinene, a-bulnesene and copaene. The reduction of biofilms formation was confirmed from SEM images. The results of this research shows a great potential from the plants studied as new antimicrobial sources.

Bacterial strains and metagenomic clones, both obtained from petroleum reservoirs, were evaluated for petroleum degradation abilities either individually or in pools using seawater microcosms for 21 days. Gas Chromatography-Flame Ionization Detector (GC-FID) and Gas Chromatography-Mass Spectrometry (GC-MS) analyses were carried out to evaluate crude oil degradation. The results showed that metagenomic clones 1A and 2B were able to biodegrade n-alkanes (C14 to C33) and isoprenoids (phytane and pristane), with rates ranging from 31% to 47%, respectively. The bacteria Dietzia maris CBMAI 705 and Micrococcus sp. CBMAI 636 showed higher rates reaching 99% after 21 days. The metagenomic clone pool biodegraded these compounds at rates ranging from 11% to 45%. Regarding aromatic compound biodegradation, metagenomic clones 2B and 10A were able to biodegrade up to 94% of phenanthrene and methylphenanthrenes (3-MP, 2-MP, 9-MP and 1-MP) with rates ranging from 55% to 70% after 21 days, while the bacteria Dietzia maris CBMAI 705 and Micrococcus sp. CBMAI 636 were able to biodegrade 63% and up to 99% of phenanthrene, respectively, and methylphenanthrenes (3-MP, 2-MP, 9-MP and 1-MP) with rates ranging from 23% to 99% after 21 days. In this work, isolated strains as well as metagenomic clones were capable of degrading several petroleum compounds, revealing an innovative strategy and a great potential for further biotechnological and bioremediation applications.

Essential oil from the leaves of Guatteria australis was obtained by hydrodistillation, analyzed by Gas Chromatography coupled to Mass Spectromery (GC-MS) and their antiproliferative, antileishmanial, antibacterial, antifungal and antioxidant activities were also evaluated. Twenty-three compounds were identified among which germacrene B (50.66%), germacrene D (22.22%) and (E)-caryophyllene (8.99%) were the main compounds. The highest antiproliferative activity was observed against NCI-ADR/RES (TGI = 31.08 μg/ml) and HT-29 (TGI = 32.81 μg/ml) cell lines. It also showed good antileishmanial activity against Leishmania infantum (IC50 = 30.71 μg/ml). On the other hand, the oil exhibited a small effect against Staphylococcus aureus ATCC 6538, S. aureus ATCC 14458 and Escherichia coli ATCC 10799 (MIC = 250 μg/ml), as well as small antioxidant activity (457 μmol TE/g) assessed through ORACFL assay. These results represent the first report regarding chemical composition and bioactivity of G. australis essential oil.

The composition and biological activities of propolis, a resinous hive product collected by honeybees from various plant sources, depends on various factors such as season and vegetation of the area. The aim of this study was to evaluate the influence of the seasonal effect on the ethanolic extracts of Brazilian propolis (EEP) type 6 and type 12, collected during 6 months in terms of antibacterial activity and phenolic composition. The antimicrobial properties were evaluated by MIC and MBC on S. mutans Ingbritt 1600 and the profile of chemical composition by UV-visible spectrophotometry, HPLC-RF and GC-MS. The results demonstrated that the season in which propolis is collected influences its chemical composition, resulting in modifications in its antibacterial activity.

Phytochemical investigation of the hexane extract from the stem of Xylopia laevigata led to the isolation of the ent-kaurane diterpenoids, ent-kaur-16-en-19-oic acid, 4-epi-kaurenic acid, ent-16β-hydroxy-17-acetoxy-kauran-19-al, ent-3β-hydroxy-kaur-16-en-19-oic acid, and ent-16β,17-dihydroxy-kauran-19-oic acid, as well as spathulenol and a mixture of β-sitosterol, stigmasterol and campesterol. The identification of the compounds was performed on the basis of spectrometric methods including GC-MS, IR, and 1D and 2D NMR. Potent larvicidal activity against Aedes aegypti larvae with LC50 of 62.7 µg mL-1 was found for ent-3β-hydroxy-kaur-16-en-19-oic acid. This compound also showed significant antifungal activity against Candida glabrata and Candida dubliniensis with MIC values of 62.5 µg mL-1.

Croton macrobothrys Baill, Euphorbiaceae, is a tree from the Atlantic Forest in Southern Brazil, used in traditional medicine and popularly known as "dragon's blood" and "pau-sangue". Leaf n-hexane, dichloromethane and methanol extracts were analyzed by GC/MS and evaluated for their in vitro antiproliferative activity on cell lines 786-0 (kidney), HT-29 (colon), K562 (leukemia), NCI-ADR/RES (drug resistant ovary), NCI-H460 (lung), MCF-7 (mammary), PC-3 (prostate), OVCAR-3 (ovary), U251 (glioma) and UACC-62 (melanoma). The dicloromethane extract exhibited activity against all cell lines at the concentration 25 µg/mL, in particular on cell lines NCI-H460 (GI50 0.33 μg/mL) and K5662 (GI50 0.91 μg/mL). Relevant constituents in dichloromethane extract are the alkaloids corydine and salutaridine, as well as the diterpenes geranylgeraniol and crotonin-derived clerodanes.

A series of nine new [3-(disubstituted-phosphate)-4,4,4-trifluoro-butyl]-carbamic acid ethyl esters (phosphate-carbamate compounds) was obtained through the reaction of (4,4,4-trifluoro-3-hydroxybut-1-yl)-carbamic acid ethyl esters with phosphorus oxychloride followed by the addition of alcohols. The products were characterized by ¹H, 13C, 31P, and 19F NMR spectroscopy, GC-MS, and elemental analysis. All the synthesized compounds were screened for acetylcholinesterase (AChE) inhibitory activity using the Ellman method. All compounds containing phosphate and carbamate pharmacophores in their structures showed enzyme inhibition, being the compound bearing the diethoxy phosphate group (2b) the most active compound. Molecular modeling studies were performed to investigate the detailed interactions between AChE active site and small-molecule inhibitor candidates, providing valuable structural insights into AChE inhibition.

An analytical comparison of three different techniques for quantitative profile of esters in cachaça is reported. The Ministério da Agricultura Pecuária e Abastecimento (MAPA) recommends the use of GC/FID or volumetry. Despite being laborious and lacking in chemical speciation, the volumetric technique for total ester content shows to be appropriate, reproducible, and accurate for the analysis of cachaça. However, the GC/FID suggested by MAPA, considering only ethyl acetate, shows inaccuracy, underestimating the total ester content by a median factor of 72%, mainly due to the absence of ethyl lactate analysis. On the other hand, the GC/MS technique that comprises the analysis and speciation of nine esters, including ethyl lactate, proved to be reproducible, simple, fast and accurate for the analysis of total ester content in cachaça. Thus, the total ester content results obtained using GC/FID must be considered with precaution.

Herein, we report the concentration of ethyl carbamate (EC) and copper in 380 samples of sugar-cane spirit and 45 samples of manioc spirit as determined by GC-MS and FAAS respectively. The cyanide content determined spectrophotometrically is reported for the manioc spirit. Sugar cane spirit produced by alembic distillation (70,0 µg L-1) shown a lower content of EC than samples produced by column distillation (270 µg L-1). No simple correlation between the content of EC and copper for sugar cane spirit as well among the concentration of EC, copper, and cyanide for manioc spirit could be observed.
