The objective of this study was to analyze the production of dry mass, forage accumulation rate and the structural composition of Brachiaria brizantha cv. Xaraes pastures, managed under different grazing heights in continuous stocking. The experimental area was 12 hectares, divided in paddocks of one hectare each. The treatments were 15, 30, 45 and 60 cm of defoliation heights. Nellore steers were used to reach the desired heights. Every 28 days four cut samples and eight visual samples were collected for comparative performance in the experimental plots. Two grazing exclusion cages were used per treatment to estimate accumulation rates kg.ha(-1) DM. The cut material was taken to the laboratory for separation of the botanical components (green leaf, stem and senescent material), weighing and determination of dry matter. The experimental design was the completely randomized with three replicates. There were treatments effects at all seasons for the production of total mass, except in the spring. In summer and spring seasons it was observed the highest values for leaf blades (1.100 kg DM.ha(-1)). In the winter, the highest values of senescent material was observed, as expected. The average accumulation showed no significant difference among the treatments, except for stem and total mass in the summer and stem in the fall. The tillers were heavier and higher values for number of green leaves per tiller occurred in the fall, but for senescent leaves per tiller ocurred in the winter. Swards grazed at heights between 45 and 60 cm of defoliation, had good production of forage mass and leaf constituent.

The objective of this study was to determine the best height of the Tanzania grass under grazing, assessing morphological and productive characteristics of the sward. This experiment was conducted in Cidade Gaucha, Parana, where he was rated the accumulation rate and morphological characteristics of Tanzania grass pastures grazed at 20, 40, 60 and 80 cm grazing under continuous stocking with variable stocking rate for beef cattle. The experimental area was 12 hectare divided in paddocks of one hectare each. For each paddock were allocated two grazing exclusion cage for estimating herbage accumulation, and eight samples were taken every 28 days throughout the year 2010. The completely randomized design. The herbage mass of the morphological structures showed a linear positive function of sward heights. The height of defoliation did not influence the rate of herbage accumulation for any of the morphological structures: green leaf, stem + sheaths and senescent material. There were also no differences in leaf: stem ratio. All heights within the range studied provided adequate number of green leaves per tiller. Tanzania grass pastures grazed under continuous stocking between 40 and 60 cm, for providing a good association between accumulation and mass of green leaf blades, and acceptable values of the mass of stem + sheaths and senescent material. Pasture of Tanzania grass under 60 and 80 cm allow of leaves satisfactory accumulation in the autumn.

Purpose: In order to assist in the selection of artificial teeth for complete dentures, this study aimed to assess the relationship between horizontal and vertical measurements of the face and the morphology of the maxillary central incisor. Materials and Methods: This was a study of 50 plaster casts and 100 teleradiographs - 50 in lateral norm and 50 in frontal norm, belonging to 50 individuals, Caucasian, with a naturally optimal occlusion, matching at least four of the six keys of Andrews. Images of the upper central incisors were obtained by scanning the plaster casts (three-dimensional) and subjectively classified by three examiners as oval, triangular or quadrangular. Facial measures (vertical and horizontal) were defined by means of teleradiographs. In order to check inter-examiner agreement on the classification of central incisor, the Kappa test was used. To verify whether data had normal distribution, the Kolmogorov-Smirnov test was used ( P > 0.2) was used. One-way analysis of variance was employed to assess the association between variables (P > 0.05). Results: When vertical measurements were compared with the three incisor shapes, there was no statistically significant difference (P > 0.05): Triangular (0.54), oval (0.63) and quadrangular (0.51). Similarly, no difference (P > 0.05) was found for facial width (139.08, 143.37, 141.65), maxillary width (76.68, 78.99, 76.91) and mandibular width (103.47, 105.50, 103.11). Conclusions: The majority of cases showed that horizontal and vertical measurements of the face cannot be used as a reference for determining the morphology of the maxillary central incisor crown. It is relevant to analyze and compare other morphological structures to improve the oral health-related quality of life for the conventional denture wearer.

O tipo de alimentação depende do ambiente em que o animal se encontra, sendo este um fator responsável pela alteração da morfologia, como a estratificação e o nível de queratinização da língua, e a funcionalidade da mesma. Dentre as estruturas morfológicas funcionais da língua, as papilas vêm merecendo destaque devido a sua estreita relação com a dieta. Foram utilizadas duas espécies de cervídeos: cinco Mazama gouazoubira e duas Mazama americana, dividindo-se a língua em três partes: ápice, corpo e raiz. Analisou comparativamente a língua de duas espécies por meio de microscopia de luz e eletrônica de varredura. As papilas filiforme, fungiforme e valada apresentaram na língua das duas espécies estudadas, e com a mesma distribuição, mudando apenas a quantidade e formato de papilas filiformes no ápice lingual e a quantidade e disposição das papilas valadas na raiz da língua, fator este que pode ser ligado à dieta dos animais. . Além disto, sua distribuição é semelhante à de outras espécies de herbívoros.

This thesis was driven by the ambition to create suitable model systems that mimic complex processes in nature, like intramolecular transitions, such as unfolding and refolding of proteins, or intermolecular interactions between different cell compo-nents. Novel biophysical approaches were adopted by employing atomic force mi-croscopy (AFM) as the main measurement technique due to its broad diversity. Thus, high-resolution imaging, adhesion measurements, and single-molecule force distance experiments were performed on the verge of the instrumental capabilities. As first objective, the interaction between plasma membrane and cytoskeleton, me-diated by the linker protein ezrin, was pursued. Therefore, the adsorption process and the lateral organization of ezrin on PIP2 containing solid-supported membranes were characterized and quantified as a fundament for the establishment of a biomimetic model system. As second component of the model system, actin filaments were coated on functionalized colloidal probes attached on cantilevers, serving as sensor elements. The zealous endeavor of creating this complex biomimetic system was rewarded by successful investigation of the activation process of ezrin. As a result, it can be stated that ezrin is activated by solely binding to PIP2 without any further stimulating agents. Additional cofactors may stabilize and prolong the active conformation but are not essentially required for triggering ezrin’s transformation into an active conformation. In the second project, single-molecule force distance experiments were performed on bis-loop tetra-urea calix[4]arene-catenanes with different loading rates (increase in force per second). These macromolecules were specifically designed to investigate the rupture and rejoining mechanism of hydrogen bonds under external load. The entangled loops of capsule-like molecules locked the unbound state of intramolecular hydrogen bonds mechanically, rendering a rebinding observable on the experimental time scale. In conjunction with Molecular Dynamics simulations, a three-well potential of the bond rupture process was established and all kinetically relevant parameters of the experiments were determined by means of Monte Carlo simulations and stochastic modeling. In summary, it can be stated that atomic force microscopy is an invaluable tool to scrutinize relevant processes in nature, such as investigating activation mechanisms in proteins, as shown by analysis of the interaction between F-actin and ezrin, as well as exploring fundamental properties of single hydrogen bonds that are of paramount interest for the complete understanding of complex supramolecular structures.

In this work, metal nanoparticles produced by nanosphere lithography were studied in terms of their optical properties (in connection to their plasmon resonances), their potential application in sensing platforms - for thin layer sensing and bio-recognition events -, and for a particular case (the nanocrescents), for enhanced spectroscopy studies. The general preparation procedures introduced early in 2005 by Shumaker-Parry et al. to produce metallic nanocrescents were extended to give rise to more complex (isolated) structures, and also, by combining colloidal monolayer fabrication and plasma etching techniques, to arrays of them. The fabrication methods presented in this work were extended not only to new shapes or arrangements of particles, but included also a targeted surface tailoring of the substrates and the structures, using different thiol and silane compounds as linkers for further attachment of, i.e. polyelectrolyte layers, which allow for a controlled tailoring of their nanoenvironment. The optical properties of the nanocrescents were studied with conventional transmission spectroscopy; a simple multipole model was adapted to explain their behaviour qualitatively. In terms of applications, the results on thin film sensing using these particles show that the crescents present an interesting mode-dependent sensitivity and spatial extension. Parallel to this, the penetrations depths were modeled with two simplified schemes, obtaining good agreement with theory. The multiple modes of the particles with their characteristic decay lengths and sensitivities represent a major improvement for particle-sensing platforms compared to previous single resonance systems. The nanocrescents were also used to alter the emission properties of fluorophores placed close to them. In this work, green emitting dyes were placed at controlled distances from the structures and excited using a pulsed laser emitting in the near infrared. The fluorescence signal obtained in this manner should be connected to a two-photon processes triggered by these structures; obtaining first insight into plasmon-mediated enhancement phenomena. An even simpler and faster approach to produce plasmonic structures than that for the crescents was tested. Metallic nanodiscs and nanoellipses were produced by means of nanosphere lithography, extending a procedure reported in the literature to new shapes and optical properties. The optical properties of these particles were characterized by extinction spectroscopy and compared to results from the literature. Their major advantage is that they present a polarization-dependent response, like the nanocrescents, but are much simpler to fabricate, and the resonances can be tailored in the visible with relative ease. The sensing capabilities of the metallic nanodiscs were explored in the same manner as for the nanocrescents, meaning their response to thin layers and to bio-recognition events on their surface. The sensitivity of these nanostructures to thin films proved to be lower than that of the crescents, though in the same order of magnitude. Experimental information about the near field extension for the Au nanodiscs of different sizes was also extracted from these measurements. Further resonance-tailoring approaches based on electrochemical deposition of metals on the nanodiscs were explored, as a means of modifying plasmon resonances by changing surface properties of the nanoparticles. First results on these experiments would indicate that the deposition of Ag on Au on a submonolayer coverage level can lead to important blue-shifts in the resonances, which would open a simple way to tailor resonances by changing material properties in a local manner.

Es wurden Glycopeptide mit einer Partialsequenz aus der N-terminalen Domäne des natürlichen Selektinliganden PSGL-1 synthetisiert, welche prinzipiell als kompetitive Inhibitoren unerwünschter selektinvermittelter Zelladhäsionsphänomene fungieren könnten. Grundsätzlich könnte es möglich sein, auf diesem Wege entsprechende chronisch entzündliche Krankheiten wie rheumatoide Arthritis zu behandeln und bestimmte akut eintretende schwere Schädigungen von gesundem Gewebe sowie die Metastasenbildung maligner Tumore zu unterdrücken. Das tatsächliche Potential der hergestellten Glycopeptide als Liganden der Selektine kann nun in biologischen Tests geprüft werden. Der gewählte Ausschnitt aus dem P-Selektin-Glycoprotein-Liganden-1 (PSGL-1) reicht von Tyr48 bis Pro59 und umfasst so sämtliche Aminosäurereste der Sequenz, die für das Auftreten einer hochaffinen Rezeptorbindung erforderlich sind. Dabei ist die Seitenkette von Thr57 mit einem O-Glycan modifiziert, welches das in natürlichen Selektinliganden häufig vorkommende Tetrasaccharid Sialyl-Lewisx bzw. ein Mimetikum desselben enthält und die für Mucine typische Form der Anbindung an das peptidische Rückgrat über eine N-Acetyl-α-D-galactosamineinheit aufweist. Zum Aufbau der komplexen Glycopeptidstrukturen wurde zunächst eine Strategie für die Synthese des an die Hydroxylaminosäure gebundenen Oligosaccharids im Gramm-Maßstab ausgearbeitet. Dabei kam der Wahl eines geeigneten Schutzgruppenmusters besondere Bedeutung zu. Das entwickelte Konzept basiert allein auf chemischen Methoden und ermöglicht die parallele Herstellung potentieller Mimetika. So wurde in dieser Arbeit L-Fucose durch D-Arabinose und N-Acetyl-D-neuraminsäure durch (S)-Cyclohexylmilchsäure ersetzt. Die erhaltenen Glycosylaminosäure-Bausteine wurden schließlich in die Glycopeptid-synthesen an der festen Phase eingebracht, welche nach vollständiger Deblockierung die gewünschten Zielverbindungen lieferten.rn

All’ascolto dell’incompiuto Matrimonio di Musorgskij, Borodin aveva sentenziato lapidariamente: «une chose manquée». Uguale commento viene riservato tuttora alla produzione operistica di Sergej Rachmaninov. In questo caso però non ci troviamo di fronte a un teatro rivoluzionario che si trova costretto a fare un passo indietro rispetto ai principi programmatici di cui si vuol far portatore, bensì a un potenziale inespresso, a un profilo drammaturgico mai giunto a piena maturazione. L’evidente eterogeneità della produzione operistica lasciataci in eredità dal compositore non permette infatti di determinarne con chiarezza le ‘linee guida’ e ne rende problematica la collocazione nel contesto musicale a lui coevo. Scopo della presente dissertazione è indagare l’apprendistato e il debutto operistico del compositore russo attraverso l’analisi di materiale documentario, testi letterari, partiture nonché delle fonti critiche in lingua russa, difficilmente accessibili per lo studioso italiano. Il cuore dell’elaborato è un’analisi dettagliata di Aleko, un atto unico presentato nel 1892 dal compositore come prova finale al corso di ‘Libera composizione’ del Conservatorio di Mosca. L’opera viene considerata nel suo complesso come organismo drammatico-musicale autonomo (rapporto fonte/libretto, articolazione interna, sistema delle forme, costellazione dei personaggi, distribuzione delle voci, ecc.), ma inquadrata al contempo nel più ampio contesto del teatro musicale coevo (recezione critica sia da parte della pubblicistica coeva sia da parte della storiografia musicale). Viene fornita in appendice un’edizione del libretto con traduzione a fronte. Incorniciano l’analisi dell’opera un capitolo in cui si offre una rilettura estetica del ‘caso Rachmaninov’ e un aperçu sulla produzione operistica matura del compositore, alla luce delle riflessioni proposte nel corso della dissertazione.

X-ray photoemission spectroscopy (XPS) is one of the most universal and powerful tools for investigation of chemical states and electronic structures of materials. The application of hard x-rays increases the inelastic mean free path of the emitted electrons within the solid and thus makes hard x-ray photoelectron spectroscopy (HAXPES) a bulk sensitive probe for solid state research and especially a very effective nondestructive technique to study buried layers.rnThis thesis focuses on the investigation of multilayer structures, used in magnetic tunnel junctions (MTJs), by a number of techniques applying HAXPES. MTJs are the most important components of novel nanoscale devices employed in spintronics. rnThe investigation and deep understanding of the mechanisms responsible for the high performance of such devices and properties of employed magnetic materials that are, in turn, defined by their electronic structure becomes feasible applying HAXPES. Thus the process of B diffusion in CoFeB-based MTJs was investigated with respect to the annealing temperature and its influence on the changes in the electronic structure of CoFeB electrodes that clarify the behaviour and huge TMR ratio values obtained in such devices. These results are presented in chapter 6. The results of investigation of the changes in the valence states of buried off-stoichiometric Co2MnSi electrodes were investigated with respect to the Mn content α and its influence on the observed TMR ratio are described in chapter 7.rnrnMagnetoelectronic properties such as exchange splitting in ferromagnetic materials as well as the macroscopic magnetic ordering can be studied by magnetic circular dichroism in photoemission (MCDAD). It is characterized by the appearance of an asymmetry in the photoemission spectra taken either from the magnetized sample with the reversal of the photon helicity or by reversal of magnetization direction of the sample when the photon helicity direction is fixed. Though recently it has been widely applied for the characterization of surfaces using low energy photons, the bulk properties have stayed inaccessible. Therefore in this work this method was integrated to HAXPES to provide an access to exploration of magnetic phenomena in the buried layers of the complex multilayer structures. Chapter 8 contains the results of the MCDAD measurements employing hard x-rays for exploration of magnetic properties of the common CoFe-based band-ferromagnets as well as half-metallic ferromagnet Co2FeAl-based MTJs.rnrnInasmuch as the magnetoresistive characteristics in spintronic devices are fully defined by the electron spins of ferromagnetic materials their direct measurements always attracted much attention but up to date have been limited by the surface sensitivity of the developed techniques. Chapter 9 presents the results on the successfully performed spin-resolved HAXPES experiment using a spin polarimeter of the SPLEED-type on a buried Co2FeAl0.5Si0.5 magnetic layer. The measurements prove that a spin polarization of about 50 % is retained during the transmission of the photoelectrons emitted from the Fe 2p3/2 state through a 3-nm-thick oxide capping layer.rn

Over the past ten years, the cross-correlation of long-time series of ambient seismic noise (ASN) has been widely adopted to extract the surface-wave part of the Green’s Functions (GF). This stochastic procedure relies on the assumption that ASN wave-field is diffuse and stationary. At frequencies <1Hz, the ASN is mainly composed by surface-waves, whose origin is attributed to the sea-wave climate. Consequently, marked directional properties may be observed, which call for accurate investigation about location and temporal evolution of the ASN-sources before attempting any GF retrieval. Within this general context, this thesis is aimed at a thorough investigation about feasibility and robustness of the noise-based methods toward the imaging of complex geological structures at the local (∼10-50km) scale. The study focused on the analysis of an extended (11 months) seismological data set collected at the Larderello-Travale geothermal field (Italy), an area for which the underground geological structures are well-constrained thanks to decades of geothermal exploration. Focusing on the secondary microseism band (SM;f>0.1Hz), I first investigate the spectral features and the kinematic properties of the noise wavefield using beamforming analysis, highlighting a marked variability with time and frequency. For the 0.1-0.3Hz frequency band and during Spring- Summer-time, the SMs waves propagate with high apparent velocities and from well-defined directions, likely associated with ocean-storms in the south- ern hemisphere. Conversely, at frequencies >0.3Hz the distribution of back- azimuths is more scattered, thus indicating that this frequency-band is the most appropriate for the application of stochastic techniques. For this latter frequency interval, I tested two correlation-based methods, acting in the time (NCF) and frequency (modified-SPAC) domains, respectively yielding esti- mates of the group- and phase-velocity dispersions. Velocity data provided by the two methods are markedly discordant; comparison with independent geological and geophysical constraints suggests that NCF results are more robust and reliable.

Zentrales Thema der Arbeit war die Aufklärung von Verwandtschaftsverhältnissen im „Tree of Life“ der vielzelligen Tiere (Metazoa) unter Einsatz großer DNA-Sequenzdatensätze und phylogenomischer Methoden. Zur Untersuchung der internen Phylogenie der Syndermata (= meist freilebende Rädertiere („Rotifera“) + endoparasitische Kratzwürmer (Acanthocephala)) sowie ihrer Position im Metazoen-Stammbaum wurden insgesamt sieben neue mitochondriale (mt) Genome sowie neue Transkriptom-Sequenzdaten von sieben verschiedenen Syndermata-Spezies generiert und/oder analysiert. Die Stammbaumrekonstruktionen auf Grundlage dieser sowie orthologer Sequenzen anderer Spezies in Form von phylogenomischen Datensätzen mit bis zu 82.000 Aminosäurepositionen ergaben folgende Aussagen zur Evolution: (i) Innerhalb der Acanthocephala bilden monophyletische Palaeacanthocephala das Schwestertaxon zu den Eoacanthocephala. Die Archiacanthocephala sind Schwestertaxon zu allen vorgenannten. (ii) Innerhalb der Syndermata bilden die epizoisch lebenden Seisonidea das Schwestertaxon zu den endoparasitischen Acanthocephala (= Pararotatoria), die Bdelloidea sind das Schwestertaxon zu den Pararotatoria (= Hemirotifera) und die Monogononta das Schwestertaxon zu den Hemirotifera. Die klassischen Eurotatoria (= Bdelloidea + Monogononta) sind demnach paraphyletisch. (iii) Innerhalb der Metazoa bilden die Syndermata gemeinsam mit den Gnathostomulida die Gnathifera. Diese sind die Schwestergruppe zu allen anderen Spiralia-Taxa, welche sich in Rouphozoa (= Platyhelminthes + Gastrotricha) sowie die Lophotrochozoa aufspalten. Die Platyzoa (= Gnathifera + Platyhelminthes + Gastrotricha) sind demnach paraphyletisch. Diese phylogenetischen Hypothesen wurden im Hinblick auf ihre Implikationen für die Evolution morphologischer und ökologischer Merkmale interpretiert. Demnach sind während der Evolution dieser Tiergruppen mehrfach sekundäre Verlustereignisse von komplexen morphologischen Merkmalen aufgetreten (laterale sensorische Organe innerhalb der Acanthocephala und das Räderorgan (Corona) innerhalb der Syndermata), was die Verwendung dieser Merkmale im Sinne einer klassisch-morphologischen Phylogenetik kritisch erscheinen lässt. Der Endoparasitismus der Acanthocephala hat sich wahrscheinlich über ein epizoisches Zwischenstadium, wie man es heute noch bei den Seisonidea findet, entwickelt. Der letzte gemeinsame Vorfahre der Spiralia war vermutlich klein und unsegmentiert und besaß keine echte Leibeshöhle (Coelom). Demnach hätten sich Segmentierung und Coelome innerhalb der Metazoa mehrfach unabhängig voneinander (konvergent) entwickelt. Die Arbeit beinhaltete folgende weitere, zum Teil methodische Aspekte: (i) die Analyse der Architektur der mt Genome der Monogononta bestätigte die aberrante Organisation in zwei Subgenomen für die Brachionidae. (ii) Eine Prüfung der Tauglichkeit ribosomaler Proteine für molekular-phylogenetische Arbeiten ergab das Vorhandensein widersprüchlicher phylogenetischer Signale in diesen speziellen Proteinsequenzen. (iii) Es konnte nachgewiesen werden, dass systematische Fehler wie „long-branch attraction“ bei der Positionierung der Syndermata im Stammbaum der Metazoa eine große Rolle spielen und adressiert werden müssen.
