Giardia duodenalis isolates from asymptomatic or symptomatic patients and from animals present similarities and differences in the protein composition, antigenic profile, pattern of proteases and isoenzymes, as well as in nucleic acids analysis. In the present overview, these differences and similarities are reviewed with emphasis in the host-parasite interplay and possible mechanisms of virulence of the protozoon.

On solid substrates, yeast colonies pass through distinct developmental phases characterized by the changes in pH of their surroundings from acidic to nearly alkaline and vice versa. At the beginning of the alkali phase colonies start to produce ammonia, which functions as a quorum-sensing molecule inducing the reprogramming of cell metabolism. Such reprogramming includes, among others, the activation of several plasma membrane transporters and is connected with colony differentiation. In the present study, we show that colony cells can use two transport mechanisms to import lactic acid: a ‘saturable’ component of the transport, which requires the presence of a functional Jen1p transporter, and a ‘non-saturable’ component (diffusion) that is independent of Jen1p. During colony development, the efficiency of both transport components changes similarly in central and outer colonial cells. Although the lactate uptake capacity of central cells gradually decreases during colony development, the lactate uptake capacity of outer cells peaks during the alkali phase and is also kept relatively high in the second acidic phase. This lactate uptake profile correlates with the localization of the Jen1p transporter to the plasma membrane of colony cells. Both lactic acid uptake mechanisms are diminished in sok2 colonies where JEN1 expression is decreased. The Sok2p transcription factor may therefore be involved in the regulation of non-saturable lactic acid uptake in yeast colonies.

O Biodiesel é uma fonte de energia renovável que actualmente se encontra em expansão. O Biodiesel é constituído por uma mistura de ésteres alquílicos de ácidos gordos. A existência de ácidos gordos insaturados torna o Biodiesel quimicamente menos estável, podendo ocorrer oxidação, degradação e polimerização do combustível, se este for inadequadamente armazenado ou transportado. O objectivo deste trabalho consistiu em avaliar a eficiência da utilização de antioxidantes fenólicos (ácido protocatecuico, ácido gálico, ácido 3,4 di-hidroxifenilacético, ácido cafeico, ácido hidrocafeico, ácido 3,4,5-tri-hidroxicinâmico, ácido m-coumárico e ácido p-coumárico), na estabilização do Biodiesel. O estudo envolveu a análise da influência do uso de cada um dos antioxidantes na inibição da peroxidação lipídica do ácido linoleico um dos principais ácidos gordos insaturados presentes na matéria-prima utilizada na produção de Biodiesel. A avaliação do efeito de inibição dos antioxidantes na peroxidação do ácido linoleico foi efetuada usando o método do tiocianato de ferro (III). Os resultados obtidos demonstraram, que todos os ácidos fenólicos estudados, apresentam uma elevada capacidade para inibir a peroxidação lipídica do ácido linoleico. As percentagens de inibição da peroxidação do ácido linoleico variaram entre os 72%, observada para o ácido p-coumárico, e os 82 %, verificada para o ácido protocatecuico. A eficiência de inibição da peroxidação por parte dos antioxidantes fenólicos em estudo foi comparada com a obtida utilizando um antioxidante de referência, o trolox. A eficiência de inibição obtida para todos os antioxidantes fenólicos estudados foi muito superior à observada para o trolox. Os resultados obtidos nesta dissertação permitem concluir que a utilização de ácidos fenólicos constitui uma boa alternativa para a estabilização de matrizes lipídicas, particularmente em combustíveis como o Biodiesel.

Previous experiments revealed that DHH1, a RNA helicase involved in the regulation of mRNA stability and translation, complemented the phenotype of a Saccharomyces cerevisiae mutant affected in the expression of genes coding for monocarboxylic-acids transporters, JEN1 and ADY2 (Paiva S, Althoff S, Casal M, Leao C. FEMS Microbiol Lett, 1999, 170∶301–306). In wild type cells, JEN1 expression had been shown to be undetectable in the presence of glucose or formic acid, and induced in the presence of lactate. In this work, we show that JEN1 mRNA accumulates in a dhh1 mutant, when formic acid was used as sole carbon source. Dhh1 interacts with the decapping activator Dcp1 and with the deadenylase complex. This led to the hypothesis that JEN1 expression is post-transcriptionally regulated by Dhh1 in formic acid. Analyses of JEN1 mRNAs decay in wild-type and dhh1 mutant strains confirmed this hypothesis. In these conditions, the stabilized JEN1 mRNA was associated to polysomes but no Jen1 protein could be detected, either by measurable lactate carrier activity, Jen1-GFP fluorescence detection or western blots. These results revealed the complexity of the expression regulation of JEN1 in S. cerevisiae and evidenced the importance of DHH1 in this process. Additionally, microarray analyses of dhh1 mutant indicated that Dhh1 plays a large role in metabolic adaptation, suggesting that carbon source changes triggers a complex interplay between transcriptional and post-transcriptional effects.

Os radicais livres formam-se naturalmente nos organismos vivos, pois a sua produção/geração está interligada com o processo de produção de energia (respiração), processos inflamatórios (fagocitose), regulação do crescimento celular, sinalização intercelular e síntese de substâncias biológicas relevantes. Estes também podem ser introduzidos por vias exógenas (poluição, radiação, tabaco, alimentação, etc). Os radicais livres têm capacidade de reagir com o material nucleico (ADN e ARN), proteínas e substâncias oxidáveis, causando danos oxidativos responsáveis pelo envelhecimento e originar doenças degenerativas, tais como, o cancro, arteriosclerose, artrite reumatoide, entre outras. De forma a combater os efeitos pejorativos provocados pelos radicais, os organismos vivos desenvolveram complexos sistemas de defesa antioxidante. Estes sistemas são constituídos por antioxidantes endógenos, produzidos pelos seres vivos, tais como enzimas ou por antioxidantes exógenos obtidos por via da alimentação (por exemplo o ácido ascórbico). Neste sentido, um antioxidante tem capacidade de eliminar ou reduzir a propagação da cadeia de geração de radicais livres. Neste trabalho foi desenvolvido um biossensor enzimático para a quantificação da capacidade antioxidante total de matrizes alimentares. A construção deste biossensor consistiu na eletroimobilização da adenina no elétrodo de pasta de carbono (EPC) ou na adsorção física da dA20 na superfície do EPC. O dano oxidativo foi induzido pelo radical hidroxilo gerado pela reação de Fenton. Nesta dissertação, foi estudada a capacidade de alguns antioxidantes em eliminar o efeito pejorativo dos radicais livres e combater a integridade das bases de adenina ou do dA20.Os antioxidantes estudados foram o ácido ascórbico e alguns ácidos fenólicos como o ácido hidroxibenzoico (ácido gálico) e ácidos hidroxicinâmicos (ácido cafeico e ácido cumárico). Estes antioxidantes têm a capacidade de neutralizar o radical hidroxilo e proteger a adenina/dA20 imobilizado na superfície do EPC. O comportamento da Lacase foi estudado na presença do ácido gálico e do ácido ascórbico. Os estudos eletroquímicos foram realizados através da voltametria de onda quadrada (VOQ), sendo que a interação entre a adenina/ou o dA20 imobilizada na superfície do EPC e os radicais livres na ausência e presença de antioxidantes foi avaliada por meio de mudanças no pico anódico produzido pela oxidação da adenina /dA20. Os resultados demonstraram que estes biossensores permitem a avaliação da capacidade antioxidante total em águas aromatizadas.

New lipophilic hydroxycinnamic acid based derivatives were designed and synthesized and their antioxidant and neuroprotective activities evaluated. The chemical modification introduced in the cinnamic acid scaffold leads to compounds with amplified lipophilicity and in general with increased antioxidant activity when compared to natural models (caffeic and ferulic acids). The compounds did not display cytotoxicity and present a significant neuroprotective effect against 6-OH-DA induced damage to SH-SY5Y cells. Compound 6 stands out as an efficient radical scavenger and iron(II) chelator that ensures drug-like properties. Moreover, neuroprotection against oxidative damage was observed even at low concentration (1 μM). Therefore, compound 6 developed by a biology-oriented approach displays a combination of important features for a further optimization process that will generate a new effective antioxidant with therapeutic application for oxidative-stress-related events, namely neurodegenerative diseases.

OBJECTIVE: The adjusted effect of long-chain polyunsaturated fatty acid (LCPUFA) intake during pregnancy on adiposity at birth of healthy full-term appropriate-for-gestational age neonates was evaluated. STUDY DESIGN: In a cross-sectional convenience sample of 100 mother and infant dyads, LCPUFA intake during pregnancy was assessed by food frequency questionnaire with nutrient intake calculated using Food Processor Plus. Linear regression models for neonatal body composition measurements, assessed by air displacement plethysmography and anthropometry, were adjusted for maternal LCPUFA intakes, energy and macronutrient intakes, prepregnancy body mass index and gestational weight gain. RESULT: Positive associations between maternal docosahexaenoic acid intake and ponderal index in male offspring (β=0.165; 95% confidence interval (CI): 0.031-0.299; P=0.017), and between n-6:n-3 LCPUFA ratio intake and fat mass (β=0.021; 95% CI: 0.002-0.041; P=0.034) and percentage of fat mass (β=0.636; 95% CI: 0.125-1.147; P=0.016) in female offspring were found. CONCLUSION: Using a reliable validated method to assess body composition, adjusted positive associations between maternal docosahexaenoic acid intake and birth size in male offspring and between n-6:n-3 LCPUFA ratio intake and adiposity in female offspring were found, suggesting that maternal LCPUFA intake strongly influences fetal body composition.

ABSTRACTINTRODUCTION:The larvicidal activity of oils, fatty acids, and methyl esters of Solanum lycocarpum fruit against Culex quinquefasciatus is unknown.METHODS:The larvicidal activity of samples of ripe and unripe fruit from S. lycocarpum was evaluated against third and fourth instar larvae of C. quinquefasciatus .RESULTS:The oils, fatty acids, and methyl esters of S. lycocarpum showed the greatest larvicidal effect (57.1-95.0%) at a concentration of 100mg/L (LC 50values between 0.70 and 27.54mg/L).CONCLUSIONS:Solanum lycocarpum fruit may be a good source of new natural products with larvicidal activity.

The cell wall of Staphylococcus aureus is a highly complex network mainly composed of highly cross-linked peptidoglycan (PG) and teichoic acids (TAs), both important for the maintenance of the integrity and viability of bacteria. The penicillin binding proteins (PBPs), which catalyse the final stage of PG biosynthesis, are targets of β-lactam antibiotics and have been a key focus of antibacterial research. S. aureus has four native PBPs, PBP1-4 carried by both methicillin-sensitive (MSSA) and –resistant (MRSA) strains. PBP4 is required for the synthesis of the highly cross-linked PG and, as shown in recent studies, is essential for the expression of β-lactam resistance in community-acquired strains (CA-MRSA). This protein has a septal localization that seems to be spatially and temporally regulated by an unknown intermediate of the wall teichoic acids (WTA) biosynthesis pathway. Therefore, if WTA synthesis is compromised, PBP4 becomes dispersed throughout the entire cell membrane. The aim of this project was to identify the WTA precursor responsible for the septal recruitment of PBP4. In order to do so, inducible mutants of tarB and tarL genes in the background of NCTCPBP4-YFP were constructed allowing for the study of PBP4 localization in the presence and absence of these specific tar genes.With this work we were able to show that the absence of TarB or TarL leads to the delocalization of PBP4, indicating that TarL or a protein/WTA precursor whose localization/synthesis is dependent on TarL is responsible for the recruitment of PBP4.

Context: Caffeic acid is described as antibacterial, but this bioactive molecule has some issues regarding solubility and stability to environmental stress. Thus, encapsulation devices are required. Objective: The aim of this work was to study the effect of the caffeic acid encapsulation by cyclodextrins on its antibacterial activity. Materials and methods: The interactions between the caffeic acid and three cyclodextrins (-cyclodextrin (CD), 2-hydroxypropyl--cyclodextrin (HPCD) and methyl--cyclodextrin were study. Results and discussion: The formation of an aqueous soluble inclusion complex was confirmed for CD and HPCD with a 1:1 stoichiometry. The CD/caffeic acid complex showed higher stability than HPCD/caffeic acid. Caffeic acid antibacterial activity was similar at pH 3 and pH 5 against the three bacteria (K. pneumoniae, S. epidermidis and S. aureus). Conclusions: The antibacterial activity of the inclusion complexes was described here for the first time and it was shown that the caffeic acid activity was remarkably enhanced by the cyclodextrins encapsulation.

The synthesis of a novel fused nitrogen heterocycle, benzoquinolone, for evaluation as a photocleavable protecting group is described for the first time, by coupling to model amino acids (alanine, phenylalanine and glutamic acid). Conversion of the phenylalanine ester conjugate to the thionated derivative was accomplished by reaction with Lawesson’s reagent. Photocleavage studies of the carbonyl and thiocarbonyl benzoquinolone conjugates in various solvents and at different wavelengths (300, 350 and 419 nm) showed that the most interesting result was obtained at 419 nm for the thioconjugate, revealing that the presence of the thiocarbonyl group clearly improved the photolysis rates, giving practicable irradiations times for the release of the amino acids (less than 1 minute).

An ion chromatography procedure, employing an IonPac AC15 concentrator column was used to investigate on line preconcentration for the simultaneous determination of inorganic anions and organic acids in river water. Twelve organic acids and nine inorganic anions were separated without any interference from other compounds and carry-over problems between samples. The injection loop was replaced by a Dionex AC15 concentrator column. The proposed procedure employed an auto-sampler that injected 1.5 ml of sample into a KOH mobile phase, generated by an Eluent Generator, at 1.5 mL min-1, which carried the sample to the chromatographic columns (one guard column, model AG-15, and one analytical column, model AS15, with 250 x 4mm i.d.). The gradient elution concentrations consisted of a 10.0 mmol l-1 KOH solution from 0 to 6.5 min, gradually increased to 45.0 mmol l-1 KOH at 21 min., and immediatelly returned and maintained at the initial concentrations until 24 min. of total run. The compounds were eluted and transported to an electro-conductivity detection cell that was attached to an electrochemical detector. The advantage of using concentrator column was the capability of performing routine simultaneous determinations for ions from 0.01 to 1.0 mg l-1 organic acids (acetate, propionic acid, formic acid, butyric acid, glycolic acid, pyruvate, tartaric acid, phthalic acid, methanesulfonic acid, valeric acid, maleic acid, oxalic acid, chlorate and citric acid) and 0.01 to 5.0 mg l-1 inorganic anions (fluoride, chloride, nitrite, nitrate, bromide, sulfate and phosphate), without extensive sample pretreatment and with an analysis time of only 24 minutes.
