Grâce aux nombreuses études sur le sujet, nous savons qu’une stimulation inflammatoire vasculaire excessive entraîne un débalancement des fonctions homéostatiques de l’endothélium. Ce débalancement est à l’origine d’une dysfonction endothéliale définie comme étant l’étape clé contribuant au développement de l’athérosclérose. Le Toll-like receptor-2 (TLR2) est impliqué dans l’activation cellulaire via la transcription des gènes liés à l’inflammation. Il reconnaît des molécules microbiennes mais également des facteurs endogènes non-infectieux tels que sécrétés par les tissus endommagés provenant de la dysfonction endothéliale. Ainsi, l’activation et la signalisation du TLR2 sont en étroite relation avec le développement de l’athérosclérose. Les études épidémiologiques ont confirmé le rôle athéroprotecteur de l’œstrogène via de nombreux mécanismes d’action. Ainsi, nous avons cherché à identifier de nouvelles cibles moléculaires permettant de mieux interpréter les bénéfices potentiels de l’œstrogène sur le système cardiaque. Pour la première fois chez les cellules endothéliales (CE) vasculaires de souris, nos travaux ont confirmé l’effet anti-inflammatoire de l’œstrogène via la diminution de l’expression et de l’activité du TLR2. Nous avons également déterminé l’influence de l’œstrogène sur le profil de la réponse inflammatoire de ce récepteur en mesurant les potentiels endothéliaux de migration et d’adhésion. De plus, nous avons caractérisé les voies de signalisation impliquées en démontrant l’influence négative de l’œstrogène sur la phosphorylation des kinases activées par le TLR2; illustrant l’interaction entre l’œstrogène et la signalisation de ce récepteur. Nos travaux amènent ainsi de nouvelles connaissances sur la régulation endothéliale du TLR2 et mettent en lumière les effets anti-inflammatoires et vasculaires rapides de l’œstrogène.

La mémoire et l’apprentissage sont des phénomènes complexes qui demeurent encore incertains quant aux origines cellulaire et moléculaire. Il est maintenant connu que des changements au niveau des synapses, comme la plasticité synaptique, pourraient déterminer la base cellulaire de la formation de la mémoire. Alors que la potentialisation à long-terme (LTP) représente un renforcement de l’efficacité de transmission synaptique, la dépression à long-terme (LTD) constitue une diminution de l’efficacité des connexions synaptiques. Des études ont mis à jour certains mécanismes qui participent à ce phénomène de plasticité synaptique, notamment, les mécanismes d’induction et d’expression, ainsi que les changements morphologiques des épines dendritiques. La grande majorité des synapses excitatrices glutamatergiques se situe au niveau des épines dendritiques et la présence de la machinerie traductionnelle près de ces protubérances suggère fortement l’existence d’une traduction locale d’ARNm. Ces ARNm seraient d’ailleurs acheminés dans les dendrites par des protéines pouvant lier les ARNm et assurer leur transport jusqu’aux synapses activées. Le rôle des protéines Staufen (Stau1 et Stau2) dans le transport, la localisation et dans la régulation de la traduction de certains ARNm est bien établi. Toutefois, leur rôle précis dans la plasticité synaptique demeure encore inconnu. Ainsi, cette thèse de doctorat évalue l’importance des protéines Staufen pour le transport et la régulation d’ARNm dans la plasticité synaptique. Nous avons identifié des fonctions spécifiques à chaque isoforme; Stau1 et Stau2 étant respectivement impliquées dans la late-LTP et la LTD dépendante des récepteurs mGluR. Cette spécificité s’applique également au rôle que chaque isoforme joue dans la morphogenèse des épines dendritiques, puisque Stau1 semble nécessaire au maintien des épines dendritiques matures, alors que Stau2 serait davantage impliquée dans le développement des épines. D’autre part, nos travaux ont permis de déterminer que la morphogenèse des épines dendritiques dépendante de Stau1 était régulée par une plasticité synaptique endogène dépendante des récepteurs NMDA. Finalement, nous avons précisé les mécanismes de régulation de l’ARNm de la Map1b par Stau2 et démontré l’importance de Stau2 pour la production et l’assemblage des granules contenant les transcrits de la Map1b nécessaires pour la LTD dépendante des mGluR. Les travaux de cette thèse démontrent les rôles spécifiques des protéines Stau1 et Stau2 dans la régulation de la plasticité synaptique par les protéines Stau1 et Stau2. Nos travaux ont permis d’approfondir les connaissances actuelles sur les mécanismes de régulation des ARNm par les protéines Staufen dans la plasticité synaptique. Antecedentes: En la actualidad no es clara la relación de la DM 2 con respecto al deterioro de la función pulmonar y menos aún si el tipo de tratamiento modifica parámetros espirométricos e inflamatorios. Objetivo: Comparar la función pulmonar de pacientes con DM 2 tratados con metformina vs secretagogos y la combinación con insulinas. Establecer el nivel de biomarcadores inflamatorios entre los grupos de tratamiento. Metodología: Estudio observacional analítico de corte transversal 495 pacientes diabéticos, entre julio 2005 y septiembre de 2007. Se obtuvieron variables espirométricas, niveles de biomarcadores inflamatorios como ferritina, fibrinógeno, PCR, Iinterleukina 6, TNF-α. Se realizó análisis de residuales de función pulmonar (valores esperados-observados) entre tipo de tratamiento con respecto al deterioro en la función pulmonar (variables espirométricas) y los niveles plasmáticos de biomarcadores. Resultados: Sin embargo un resultado valioso que se muestra en nuestro estudio, es que Después de ajustar por determinantes conocidos de la función pulmonar; los pacientes tratados con metformina tenían una tendencia no significativa de menor residual del VEF1 siendo de -133.2 vs -174.8 ml en el grupo de secretagogos. Al igual que un residual de CVF menor en los pacientes tratados con metformina en comparación con secretagogos, siendo de -212.1 ml vs -270.2 ml respectivamente con una p de 0.039. En el grupo de pacientes tratados con Metformina, los niveles de biomarcadores inflamatorios fueron menores. Conclusiones: Este estudio sustenta que la metformina parece evitar el deterioro de la función pulmonar de los pacientes diabéticos, al igual que parece tener un efecto antiinflamatorio.

Se realizó un estudio genético – poblacional en dos grupos etarios de población colombiana con la finalidad de evaluar las diferencias genéticas relacionadas con el polimorfismo MTHFR 677CT en busca de eventos genéticos que soporten la persistencia de este polimorfismo en la especie humana debido que este ha sido asociado con múltiples enfermedades. De esta manera se genotipificaron los individuos, se analizaron los genotipos, frecuencias alélicas y se realizaron diferentes pruebas genéticas-poblacionales. Contrario a lo observado en poblaciones Colombianas revisadas se identificó la ausencia del Equilibrio Hardy-Weinberg en el grupo de los niños y estructuras poblacionales entre los adultos lo que sugiere diferentes historias demográficas y culturales entre estos dos grupos poblacionales al tiempo, lo que soporta la hipótesis de un evento de selección sobre el polimorfismo en nuestra población. De igual manera nuestros datos fueron analizados junto con estudios previos a nivel nacional y mundial lo cual sustenta que el posible evento selectivo es debido a que el aporte de ácido fólico se ha incrementado durante las últimas dos décadas como consecuencia de las campañas de fortificación de las harinas y suplementación a las embarazadas con ácido fólico, por lo tanto aquí se propone un modelo de selección que se ajusta a los datos encontrados en este trabajo se establece una relación entre los patrones nutricionales de la especie humana a través de la historia que explica las diferencias en frecuencias de este polimorfismo a nivel espacial y temporal.

Phthalates are industrial additives widely used as plasticizers. In addition to deleterious effects on male genital development, population studies have documented correlations between phthalates exposure and impacts on reproductive tract development and on the metabolic syndrome in male adults. In this work we investigated potential mechanisms underlying the impact of DEHP on adult mouse liver in vivo. A parallel analysis of hepatic transcript and metabolic profiles from adult mice exposed to varying DEHP doses was performed. Hepatic genes modulated by DEHP are predominantly PPARalpha targets. However, the induction of prototypic cytochrome P450 genes strongly supports the activation of additional NR pathways, including Constitutive Androstane Receptor (CAR). Integration of transcriptomic and metabonomic profiles revealed a correlation between the impacts of DEHP on genes and metabolites related to heme synthesis and to the Rev-erbalpha pathway that senses endogenous heme level. We further confirmed the combined impact of DEHP on the hepatic expression of Alas1, a critical enzyme in heme synthesis and on the expression of Rev-erbalpha target genes involved in the cellular clock and in energy metabolism. This work shows that DEHP interferes with hepatic CAR and Rev-erbalpha pathways which are both involved in the control of metabolism. The identification of these new hepatic pathways targeted by DEHP could contribute to metabolic and endocrine disruption associated with phthalate exposure. Gene expression profiles performed on microdissected testis territories displayed a differential responsiveness to DEHP. Altogether, this suggests that impacts of DEHP on adult organs, including testis, could be documented and deserve further investigations.

The aim of the present study was to investigate the effect of probiotic immobilization onto wheat grains, both wet and freeze dried, on the adhesion properties of the probiotic cells and make comparisons with wet and freeze dried free cells. Lactobacillus casei ATCC 393 and Lactobacillus plantarum NCIMB 8826 were used as model probiotic strains. The results showed satisfactory adhesion ability of free cells to a monolayer of Caco-2 cells (> 1000 CFU/100 Caco-2 cells for wet cells). Cell immobilization resulted in a significant decrease in adhesion, for both wet and freeze dried formulations, most likely because immobilized cells did not have direct access to the Caco-2 cells, but it still remained in adequate levels (> 100 CFU/100 Caco-2 cells for wet cells). No clear correlation could be observed between cell adhesion and the hydrophobicity of the bacterial cells, measured by the hexadecane adhesion assay. Most notably, immobilization enhanced the monolayer integrity of Caco-2 cells, demonstrated by a more than 2-fold increase in transepithelial electrical resistance (TEER) compared to free cells. SEM micrographs ascertained the adhesion of both immobilized and free cells to the brush border microvilli. Finally, the impact of the food matrix on the adhesion properties of probiotic bacteria and on the design of novel functional products is discussed.

Platelet aggregation and phosphorylation of phospholipase Cγ2 induced by collagen were attenuated in ADAP(-/-) platelets. However, aggregation and signaling induced by collagen-related peptide (CRP), a GPVI-selective agonist, were largely unaffected. Platelet adhesion to CRP was also unaffected by ADAP deficiency. Adhesion to the α(2) β(1) -selective ligand GFOGER and to a peptide (III-04), which supports adhesion that is dependent on both GPVI and α(2) β(1), was reduced in ADAP(-/-) platelets. An impedance-based label-free detection technique, which measures adhesion and spreading of platelets, indicated that, in the absence of ADAP, spreading on GFOGER was also reduced. This was confirmed with non-fluorescent differential-interference contrast microscopy, which revealed reduced filpodia formation in ADAP(-/-) platelets adherent to GFOGER. This indicates that ADAP plays a role in mediating platelet activation via the collagen-binding integrin α(2) β(1). In addition, we found that ADAP(-/-) mice, which are mildly thrombocytopenic, have enlarged spleens as compared with wild-type animals. This may reflect increased removal of platelets from the circulation.

Platelet-specific deletion of CLEC-2, which signals through Src and Syk kinases, or global deletion of its ligand podoplanin results in blood-filled lymphatics during mouse development. Platelet-specific Syk deficiency phenocopies this defect, indicating that platelet activation is required for lymphatic development. In the present study, we investigated whether CLEC-2-podoplanin interactions could support platelet arrest from blood flow and whether platelet signalling is required for stable platelet adhesion to lymphatic endothelial cells (LECs) and recombinant podoplanin under flow. Perfusion of human or mouse blood over human LEC monolayers led to platelet adhesion and aggregation. Following αIIbβ3 blockade, individual platelets still adhered. Platelet binding occurred at venous but not arterial shear rates. There was no adhesion using CLEC-2-deficient blood or to vascular endothelial cells (which lack podoplanin). Perfusion of human blood over human Fc-podoplanin (hFcPDPN) in the presence of monoclonal antibody IV.3 to block FcγRIIA receptors led to platelet arrest at similar shear rates to those used on LECs. Src and Syk inhibitors significantly reduced global adhesion of human or mouse platelets to LECs and hFcPDPN. A similar result was seen using Syk-deficient mouse platelets. Reduced platelet adhesion was due to a decrease in the stability of binding. In conclusion, our data reveal that CLEC-2 is an adhesive receptor that supports platelet arrest to podoplanin under venous shear. Src/Syk-dependent signalling stabilises platelet adhesion to podoplanin, providing a possible molecular mechanism contributing to the lymphatic defects of Syk-deficient mice.

From consideration of children's rights in general and equal opportunities for disabled children in particular, it is important to consult children about barriers and supports to learning and participation. Finding appropriate and feasible ways, however, to incorporate this into educational programmes for younger children can present challenges. Here we report on what happened when teachers from reception classes in England for children aged 4–5 years implemented activities designed to access pupils' views about what helps or hinders at school. Teachers evaluated the feasibility and usefulness of the activities and, together with a small sample of children's responses, this showed that young children could indeed identify aspects of school life they like or dislike, laying the foundations for identifying barriers and supports to learning. Teachers' responses highlighted the importance of careful choice of activity to meet the needs of young children, particularly those with communication difficulties and/or low self-confidence, with staff in some cases adapting and merging activities to suit pupils' needs. Sensitive issues emerged concerning the introduction of consultation activities early in children's school careers. The implications of a compliant rather than collaborative approach by teachers are discussed in the context of children's right to have their views heard, and their developing understanding of difference.

Mutations in Na+-glucose transporters (SGLT)-2 and hepatocyte nuclear factor (HNF)-1 alpha genes have been related to renal glycosuria and maturity-onset diabetes of the young 3, respectively. However, the expression of these genes have not been investigated in type 1 and type 2 diabetes. Here in kidney of diabetic rats, we tested the hypotheses that SGLT2 mRNA expression is altered; HNF-1 alpha is involved in this regulation; and glycemic homeostasis is a related mechanism. The in vivo binding of HNF-1 alpha into the SGLT2 promoter region in renal cortex was confirmed by chromatin immunoprecipitation assay. SGLT2 and HNF-1 alpha mRNA expression (by Northern and RT-PCR analysis) and HNF-1 binding activity of nuclear proteins (by EMSA) were investigated in diabetic rats and treated or not with insulin or phlorizin (an inhibitor of SGLT2). Results showed that diabetes increases SGLT2 and HNF-1 alpha mRNA expression (similar to 50%) and binding of nuclear proteins to a HNF-1 consensus motif (similar to 100%). Six days of insulin or phlorizin treatment restores these parameters to nondiabetic-rat levels. Moreover, both treatments similarly reduced glycemia, despite the differences in plasma insulin and urinary glucose concentrations, highlighting the plasma glucose levels as involved in the observed modulations. This study shows that SGLT2 mRNA expression and HNF-1 alpha expression and activity correlate positively in kidney of diabetic rats. It also shows that diabetes-induced changes are reversed by lowering glycemia, independently of insulinemia. Our demonstration that HNF-1 alpha binds DNA that encodes SGLT2 supports the hypothesis that HNF-1 alpha, as a modulator of SGLT2 expression, may be involved in diabetic kidney disease.
