The alpha subunits of the heterotrimeric guanine nucleotide-binding proteins (G proteins) hydrolyze GTP at a rate significantly higher than do most members of the Ras family of approximatelly 20-kDa GTP-binding proteins, which depend on a GTPase-activating protein (GAP) for acceleration of GTP hydrolysis. It has been demonstrated that an inserted domain in the G-protein alpha subunit, not present in the much smaller Ras-like proteins, is responsible for this difference [Markby, D. W., Onrust, R. & Bourne, H. R. (1993) Science 262, 1895-1900]. We report here that ARD1, a 64-kDa protein with an 18-kDa carboxyl-terminal ADP-ribosylation factor (ARF) domain, exhibited significant GTPase activity, whereas the ARF domain, expressed as a recombinant protein in Escherichia coli, did not. Addition of the 46-kDa amino-terminal extension (similarly synthesized in E. coli) to the GTP-binding ARF-domain of ARD1 enhanced GTPase activity and inhibited GDP dissociation. The kinetic properties of mixtures of the ARF and non-ARF domains were similar to those of an intact recombinant ARD1. Physical association of the two proteins was demonstrated directly by gel filtration and by using the immobilized non-ARF domain. Thus, like the alpha subunits of heterotrimeric G proteins, ARD1 appears to consist of two domains that interact to regulate the biological activity of the protein.

Walker & Co. in Feb. 1895. Scale [ca. 1:2,400]. The image inside the map neatline is georeferenced to the surface of the earth and fit to the Massachusetts State Plane Coordinate System, Mainland Zone (in Feet) (Fipszone 2001). Através de uma exaustiva pesquisa em fontes jornalísticas e documentais, o autor introduz o leitor no universo das ideias e práticas políticas que marcaram a tensa implantação do regime republicano no Brasil e especialmente no Rio Grande do Norte.

La ciudad ofrecía en la Baja Edad Media claras oportunidades para aquellos nobles que supieran situarse en el complejo juego de la política urbana y sus diversas instancias de poder. Para ello siempre se hace necesario contar con los apoyos sociales y físicos adecuados. La dinámica de enfrentamientos entre el Arzobispo de Santiago y el concello compostelano presenta un ejemplo inmejorable para valorar en su justa medida la inserción de la nobleza territorial en las relaciones de poder. Para abordar este caso se analizará la participación en la vida pública –también las revueltas urbanas– de la Casa de Moscoso tanto a través de su larga trayectoria de enfrentamientos con la Iglesia como mediante el estudio de su patrimonio urbano, incluyendo aquellos elementos relacionados con la creación de una memoria específica del linaje dentro de la ciudad.

En 1526, Hassan El Wazzan / Jean-Léon l’Africain, achève à Rome la rédaction en italien du manuscrit du Libro della Cosmographia Dell’Africa, œuvre majeure considérée à la Renaissance comme l’une des principales sources de connaissance du continent africain en Europe. En 1550, un savant vénitien du nom de Jean-Baptiste Ramusio publie le texte italien de Jean-Léon dans un recueil de récits de voyages. L’édition, intitulée Descrizione dell’Africa (Description de l’Afrique), diffère significativement du manuscrit original. Elle subit maintes modifications par Ramusio dont l’objectif est de livrer un ouvrage qui répond aux attentes des Européens et qui correspond à l’image que l’Occident chrétien se faisait du monde musulman. Cette version a servi de texte de départ aux nombreuses traductions qui ont suivi. La première traduction française, datant de 1556, est réalisée par Jean Temporal, éditeur et imprimeur lyonnais. La deuxième, parue en 1956 et rééditée en 1980, est l’œuvre d’Alexis Épaulard; elle s’appuie partiellement sur le manuscrit original, mais aussi sur la version imprimée de Ramusio. Notre travail consiste à confronter les deux traductions françaises à l’édition de Ramusio. Nous tenterons de démontrer que les deux traducteurs français sont lourdement intervenus dans le texte traduit, et ce afin de servir des desseins expansionnistes et colonialistes. Notre recherche met en évidence la prise de position des traducteurs et les idéologies qui affectent l’appréciation du livre. Pour ce faire, nous procédons à l’analyse des traductions au niveau textuel et au niveau paratextuel tout en mettant en évidence le contexte historique et politico-idéologique entourant la parution de ces deux traductions françaises. Nous consacrons une attention toute particulière au choix des mots, aux allusions et aux stratégies utilisées par les traducteurs et les éditeurs. Les travaux de Maria Tymoczko sur la traduction et l’engagement politique fournissent le cadre de référence théorique de cette recherche, tout autant que les textes d’Edward Said sur l’orientalisme et le postcolonialisme. Il ressort de cette recherche que ces traductions françaises sont empreintes d’une idéologie eurocentrée visant à conforter les ambitions hégémoniques en terre africaine.

Interleukin(IL)-18 is a pleiotrophic cytokine with functions in immune modulation, angiogenesis and bone metabolism. In this study, the potential of IL-18 as an immunotherapy for prostate cancer (PCa) was examined using the murine model of prostate carcinoma, RM1 and a bone metastatic variant RM1(BM)/B4H7-luc. RM1 and RM1(BM)/B4H7-luc cells were stably transfected to express bioactive IL-18. These cells were implanted into syngeneic immunocompetent mice, with or without an IL-18-neutralising antibody (αIL-18, SK113AE4). IL-18 significantly inhibited the growth of both subcutaneous and orthotopic RM1 tumors and the IL-18 neutralizing antibody abrogated the tumor growth-inhibition. In vivo neutralization of interferon-gamma (IFN-γ) completely eliminated the anti-tumor effects of IL-18 confirming an essential role of IFN-γ as a down-stream mediator of the anti-tumor activity of IL-18. Tumors from mice in which IL-18 and/or IFN-γ was neutralized contained significantly fewer CD4+ and CD8+ T cells than those with functional IL-18. The essential role of adaptive immunity was demonstrated as tumors grew more rapidly in RAG1−/− mice or in mice depleted of CD4+ and/or CD8+ cells than in normal mice. The tumors in RAG1−/− mice were also significantly smaller when IL-18 was present, indicating that innate immune mechanisms are involved. IL-18 also induced an increase in tumor infiltration of macrophages and neutrophils but not NK cells. In other experiments, direct injection of recombinant IL-18 into established tumors also inhibited tumor growth, which was associated with an increase in intratumoral macrophages, but not T cells. These results suggest that local IL-18 in the tumor environment can significantly potentiate anti-tumor immunity in the prostate and clearly demonstrate that this effect is mediated by innate and adaptive immune mechanisms.

We examined the effects of progressive resistance training (PRT) and supplementation with calcium-vitamin D(3) fortified milk on markers of systemic inflammation, and the relationship between inflammation and changes in muscle mass, size and strength. Healthy men aged 50-79 years (n = 180) participated in this 18-month randomized controlled trial that comprised a factorial 2 x 2 design. Participants were randomized to (1) PRT + fortified milk supplement, (2) PRT, (3) fortified milk supplement, or (4) a control group. Participants assigned to PRT trained 3 days per week, while those in the supplement groups consumed 400 ml day(-1) of milk containing 1,000 mg calcium plus 800 IU vitamin D(3). We collected venous blood samples at baseline, 12 and 18 months to measure the serum concentrations of IL-6, TNF-alpha and hs-CRP. There were no exercise x supplement interactions, but serum IL-6 was 29% lower (95% CI, -62, 0) in the PRT group compared with the control group after 12 months. Conversely, IL-6 was 31% higher (95% CI, -2, 65) in the supplement group compared with the non-supplemented groups after 12 and 18 months. These between-group differences did not persist after adjusting for changes in fat mass. In the PRT group, mid-tibia muscle cross-sectional area increased less in men with higher pre-training inflammation compared with those men with lower inflammation (net difference similar to 2.5%, p < 0.05). In conclusion, serum IL-6 concentration decreased following PRT, whereas it increased after supplementation with fortified milk concomitant with changes in fat mass. Furthermore, low-grade inflammation at baseline restricted muscle hypertrophy following PRT.
