ENGLISH: This study was undertaken to determine whether meristic characters indicate that more than one major population of anchovetas occurs in the range of the species from Mexico to Peru. Interest in this species lies in the fact that it is the principal bait fish used to catch yellowfin and skipjack tunas in the Eastern Pacific. Specimens examined were from collections made by California tuna fishing vessels at six major baiting localities covering nearly the entire range of the species, namely, Almejas Bay on the outer coast of Baja California, Guaymas and Ahome Point in the Gulf of California, Gulf of Fonseca, Gulf of Panama, and Gulf of Guayaquil. Four meristic characters were selected for study: vertebrae, dorsal fin rays, anal fin rays, and gill rakers on the first gill arch. Vertebral counts, using X-ray film, were taken from a total of 1,500 fish, 250 each from each of the six localities. For the other characters, 125 anchovetas were examined from each locality for a total of 750, the counts being made with the aid of a binocular microscope. Specimens were between 80 and 165 mm. standard length. SPANISH: Este estudio ha sido hecho con el propósito de determinar si los caracteres numéricos de las anchovetas indican que existe más de una población de este pez en la zona en que se encuentra la especie, comprendida entre México y Perú. El interés en dicha especie radica en el hecho de que éste es el pez de carnada usado principalmente para la pesca de los atunes "aleta amarilla" y "barrilete" en el Pacífico Oriental. Los especímenes que han sido examinados, se tomaron de las muestras recogidas por los barcos atuneros de California en seis de las mejores localidades en que se pesca la anchoveta, las cuales comprenden casi toda la zona en donde se encuentra la especie, a saber, Bahía de Almejas en la costa exterior de Baja California, Guaymas y Punta Ahorne en el Golfo de California, el Golfo de Fonseca, el Golfo de Panamá y el Golfo de Guayaquil. Cuatro caracteres numéricos fueron escogidos para su estudio: los que presentan 1) las vértebras, 2) los radios de la aleta dorsal, 3) los radios de la aleta anal y 4) las branquispinas del primer arco branquial. Mediante el uso de películas con rayos X, se contaron las vértebras en un total de 1,500 peces, es decir, 250 de cada una de las seis mencionadas localidades. En relación con los otros caracteres, se examinaron 125 anchovetas de cada área, o sea, un total de 750 ejemplares, habiendo sido hecho el conteo por medio de un microscopio binocular. The meristic and morphometric characteristics of Gymnarchus niloticus are described and linear equations relating various parts of the body to the head length or total length are given. The age of G. niloticus in Lake Chad (Nigeria) was determined from growth marks on the opercular bones. The mean lengths for age, and mean weights for age obtained for the first five years of life are given. ENGLISH: Samples of yellowfin tuna, Thunnus albacares, collected from five areas of the Pacific Ocean (Mexico, Ecuador, Australia, Japan, and Hawaii) between January and May of 1988 and 1990 were examined for spatiotemporal variation in morphometric characters and gill-raker counts. 'Iwo-factor analysis of variance, with area and year treated as grouping factors, indicated a significant difference in the means of the total gill-raker counts among fish from different areas, but no significant difference between fish caught in different years. The morphometric data were adjusted by allometric formulae to remove size effects. The correct classification rates for the five groups, using discriminant function analysis, based on adjusted morphometric characters, were 77.60/0 for the samples from 1988 and 74.40/0 for those from 1990. These are 72.00/0 and 68.00/0 (Cohen's kappa statistic) better than would have occurred chance. The pattern of geographic variability, however, is unstable for these two years, thus requiring separate discriminant functions for each year. Although there is annual variability in the morphometric characters, these results demonstrate that the stocks examined are morphometrically distinguishable and that their phenetic relationships reflect their geographic origin. SPANISH: Se examinaron muestras de atún aleta amarilla, Thunnus albacares, tomadas de cinco áreas del Océano Pacífico (México, Ecuador, Australia, Japón, y Hawaii) entre enero y mayo de 1988 y 1990, para descubrir variaciones espaciotemporales en las características morfométricas y los conteos de branquiespinas. Un análisis de varianza de dos factores, con área y año como factores de agrupación, indicó una diferencia significativa en los promedios de los conteos de branquiespinas totales entre peces de distintas áreas, pero ninguna entre peces capturados en distintos años. Se ajustaron los datos morfométricos con fórmulas alométricas para eliminar los efectos de la talla del pez. En un análisis de función discriminante, las tasas de clasificación correcta de los cinco grupos, basadas en características morfométricas ajustadas, fueron 77.60/0 para las muestras de 1988 y 74.40/0 para aquellas de 1990. Estas cifras son 72.00/0 y 68.00/0 (estadístico de kappa de Cohen) mejores de lo que se hubiera obtenido al azar. Sin embargo, la variabilidad geográfica es inestable en estos dos años, requiriendo por lo tanto funciones discriminantes separadas para cada año. Aunque existe variabilidad anual en las características morfométricas, estos resultados demuestran que los stocks examinados son morfamétricamente distinguibles, y que su relación fenética refleja su origen geográfico. Interleukin-2 (IL-2) is an important mediator in the vertebrate immune system. IL-2 is a potent growth factor that mature T lymphocytes use as a proliferation signal and the production of IL-2 is crucial for the clonal expansion of antigen-specific T cells in the primary immune response. IL-2 driven proliferation is dependent on the interaction of the lymphokine with its cognate multichain receptor. IL-2 expression is induced only upon stimulation and transcriptional activation of the IL-2 gene relies extensively on the coordinate interaction of numerous inducible and constitutive trans-acting factors. Over the past several years, thousands of papers have been published regarding molecular and cellular aspects of IL-2 gene expression and IL-2 function. The vast majority of these reports describe work that has been carried out in vitro. However, considerably less is known about control of IL-2 gene expression and IL-2 function in vivo.

To gain new insight into the regulation of IL-2 gene expression in vivo, anatomical and developmental patterns of IL-2 gene expression in the mouse were established by employing in situ hybridization and immunohistochemical staining methodologies to tissue sections generated from normal mice and mutant animals in which T -cell development was perturbed. Results from these studies revealed several interesting aspects of IL-2 gene expression, such as (1) induction of IL-2 gene expression and protein synthesis in the thymus, the primary site of T-cell development in the body, (2) cell-type specificity of IL-2 gene expression in vivo, (3) participation of IL-2 in the extrathymic expansion of mature T cells in particular tissues, independent of an acute immune response to foreign antigen, (4) involvement of IL-2 in maintaining immunologic balance in the mucosal immune system, and (5) potential function of IL-2 in early events associated with hematopoiesis.

Extensive analysis of IL-2 mRNA accumulation and protein production in the murine thymus at various stages of development established the existence of two classes of intrathymic IL-2 producing cells. One class of intrathymic IL-2 producers was found exclusively in the fetal thymus. Cells belonging to this subset were restricted to the outermost region of the thymus. IL-2 expression in the fetal thymus was highly transient; a dramatic peak ofiL-2 mRNA accumulation was identified at day 14.5 of gestation and maximal IL-2 protein production was observed 12 hours later, after which both IL-2 mRNA and protein levels rapidly decreased. Significantly, the presence of IL-2 expressing cells in the day 14-15 fetal thymus was not contingent on the generation of T-cell receptor (TcR) positive cells. The second class of IL-2 producing cells was also detectable in the fetal thymus (cells found in this class represented a minority subset of IL-2 producers in the fetal thymus) but persist in the thymus during later stages of development and after birth. Intrathymic IL-2 producers in postnatal animals were located in the subcapsular region and cortex, indicating that these cells reside in the same areas where immature T cells are consigned. The frequency of IL-2 expressing cells in the postnatal thymus was extremely low, indicating that induction of IL-2 expression and protein synthesis are indicative of a rare activation event. Unlike the fetal class of intrathymic IL-2 producers, the presence of IL-2 producing cells in the postnatal thymus was dependent on to the generation of TcR+ cells. Subsequent examination of intrathymic IL-2 production in mutant postnatal mice unable to produce either αβ or γδ T cells showed that postnatal IL-2 producers in the thymus belong to both αβ and γδ lineages. Additionally, further studies indicated that IL-2 synthesis by immature αβ -T cells depends on the expression of bonafide TcR αβ-heterodimers. Taken altogether, IL-2 production in the postnatal thymus relies on the generation of αβ or γδ-TcR^+ cells and induction of IL-2 protein synthesis can be linked to an activation event mediated via the TcR.

With regard to tissue specificity of IL-2 gene expression in vivo, analysis of whole body sections obtained from normal neonatal mouse pups by in situ hybridization demonstrated that IL-2 mRNA^+ cells were found in both lymphoid and nonlymphoid tissues with which T cells are associated, such as the thymus (as described above), dermis and gut. Tissues devoid of IL-2 mRNA^+ cells included brain, heart, lung, liver, stomach, spine, spinal cord, kidney, and bladder. Additional analysis of isolated tissues taken from older animals revealed that IL-2 expression was undetectable in bone marrow and in nonactivated spleen and lymph nodes. Thus, it appears that extrathymic IL-2 expressing cells in nonimmunologically challenged animals are relegated to particular epidermal and epithelial tissues in which characterized subsets of T cells reside and thatinduction of IL-2 gene expression associated with these tissues may be a result of T-cell activation therein.

Based on the neonatal in situ hybridization results, a detailed investigation into possible induction of IL-2 expression resulting in IL-2 protein synthesis in the skin and gut revealed that IL-2 expression is induced in the epidermis and intestine and IL-2 protein is available to drive cell proliferation of resident cells and/or participate in immune function in these tissues. Pertaining to IL-2 expression in the skin, maximal IL-2 mRNA accumulation and protein production were observed when resident Vγ_3^+ T-cell populations were expanding. At this age, both IL-2 mRNA^+ cells and IL-2 protein production were intimately associated with hair follicles. Likewise, at this age a significant number of CD3ε^+ cells were also found in association with follicles. The colocalization of IL-2 expression and CD3ε^+ cells suggests that IL-2 expression is induced when T cells are in contact with hair follicles. In contrast, neither IL-2 mRNA nor IL-2 protein were readily detected once T-cell density in the skin reached steady-state proportions. At this point, T cells were no longer found associated with hair follicles but were evenly distributed throughout the epidermis. In addition, IL-2 expression in the skin was contingent upon the presence of mature T cells therein and induction of IL-2 protein synthesis in the skin did not depend on the expression of a specific TcR on resident T cells. These newly disclosed properties of IL-2 expression in the skin indicate that IL-2 may play an additional role in controlling mature T-cell proliferation by participating in the extrathymic expansion of T cells, particularly those associated with the epidermis.

Finally, regarding IL-2 expression and protein synthesis in the gut, IL-2 producing cells were found associated with the lamina propria of neonatal animals and gut-associated IL-2 production persisted throughout life. In older animals, the frequency of IL-2 producing cells in the small intestine was not identical to that in the large intestine and this difference may reflect regional specialization of the mucosal immune system in response to enteric antigen. Similar to other instances of IL-2 gene expression in vivo, a failure to generate mature T cells also led to an abrogation of IL-2 protein production in the gut. The presence of IL-2 producing cells in the neonatal gut suggested that these cells may be generated during fetal development. Examination of the fetal gut to determine the distribution of IL-2 producing cells therein indicated that there was a tenfold increase in the number of gut-associated IL-2 producers at day 20 of gestation compared to that observed four days earlier and there was little difference between the frequency of IL-2 producing cells in prenatal versus neonatal gut. The origin of these fetally-derived IL-2 producing cells is unclear. Prior to the immigration of IL-2 inducible cells to the fetal gut and/or induction of IL-2 expression therein, IL-2 protein was observed in the fetal liver and fetal omentum, as well as the fetal thymus. Considering that induction of IL-2 protein synthesis may be an indication of future functional capability, detection of IL-2 producing cells in the fetal liver and fetal omentum raises the possibility that IL-2 producing cells in the fetal gut may be extrathymic in origin and IL-2 producing cells in these fetal tissues may not belong solely to the T lineage. Overall, these results provide increased understanding of the nature of IL-2 producing cells in the gut and how the absence of IL-2 production therein and in fetal hematopoietic tissues can result in the acute pathology observed in IL-2 deficient animals.

Os gêneros de peixes fósseis Oshunia e Placidichthys são holósteos pertencentes à Ordem Ionoscopiformes e provenientes do Cretáceo Inferior do Brasil, das bacias do Araripe e de Tucano. No clado Ionoscopiformes sensu Grande & Bemis (1998) estão incluídas as famílias Ionoscopidae e Ophiopsidae, todavia as relações internas deste grupo ainda são duvidosas. Oshunia e Placidichthys fazem parte das famílias Ionoscopidae e Ophiopsidae, respectivamente, sendo o gênero Oshunia considerado como mono-específico (cf., O. brevis), enquanto que Placidichthys apresenta duas espécies nominais (cf., P. bidorsalis e P. tucanensis). Em função destas espécies terem sido descritas a partir de poucos espécimes, ainda existiam várias lacunas no conhecimento em relação as mesmas, como, por exemplo, a possibilidade da existência de outras espécies no gênero Oshunia e a falta de informações anatômicas, especialmente do crânio, da região occipital, dos ossos da face e da nadadeira caudal das espécies de Placidichthys. Outro ponto em aberto na literatura era a posição filogenética dos dois gêneros. Frente a estas questões, o objetivo da presente dissertação foi realizar uma revisão anatômica dos gêneros Oshunia e Placidichthys, a fim de ampliar o conhecimento anatômico e taxonômico acerca dos mesmos, além realizar uma análise filogenética da Ordem Ionoscopiformes, baseada em matrizes de caracteres existentes na literatura, para se obter um melhor posicionamento dessas espécies brasileiras. Em função da facilidade de acesso a material mexicano, também foram incluídos nesta revisão os gêneros Teoichthys e Tuetzalichthys provenientes do Cretáceo da Formação Tlayúa, estes também peixes fósseis holósteos pertencentes à Ordem Ionoscopiformes. Do ponto de vista taxonômico, não foi possível confirmar a existência de novas espécies para o gênero Oshunia, entretanto ficou clara a presença de uma nova espécie pertencente ao gênero mexicano Teoichthys. A presente revisão proporcionou uma série de novas informações sobre a anatomia destas espécies de Ionoscopiformes, tais como a descrição dos ossos circumorbitais e do teto craniano e uma reinterpretação acerca da nadadeira dorsal de Placidichthys bidorsalis, ou ainda sobre a forma do rostral de Teoichthys kallistos. Da mesma maneira, esta revisão também ofereceu novos dados para a construção de uma nova hipótese filogenética para Ionoscopiformes, a qual se mostrou consideravelmente distinta das hipóteses filogenéticas anteriores (cf., relações internas de Ionoscopidae e o posicionamento do gênero Teoichthys). O baixo suporte para grande parte dos clados torna evidente a fragilidade das hipóteses de relacionamento interno do clado Ionoscopiformes, bem como a necessidade de uma revisão mais aprofundada das outras espécies deste grupo e dos caracteres a serem utilizados em futuras análises filogenéticas.

Os teleósteos são o grupo mais diversificado entre os vertebrados e seu registro mais antigo data do Jurássico. Sua atual classificação inclui quatro clados, dentre os quais Euteleostei é o mais avançado e variado. Apesar de todos os trabalhos a respeito do grupo, ele ainda não possui diagnose, definição e composição precisas. A discordância entre autores é ilustrada pelas nove diferentes propostas filogenéticas elaboradas nos últimos 30 anos. Muitos fósseis do Cretáceo são classificados como euteleósteos basais por falta de conhecimento morfológico, enquanto outros fósseis possuem classificação sistemática controversa ou compartilham aspectos estruturais com euteleósteos basais. Nesse contexto, os objetivos da presente dissertação são avaliar o monofiletismo de euteleósteos basais e recuperar relações filogenéticas de táxons do Nordeste do Brasil, África, Europa, Ásia e América do Norte atribuídos aos euteleósteos basais. Sete táxons brasileiros (i.e., Beurlenichthys ouricuriensis, Britoichthys marizalensis, Clupavus brasiliensis, Santanasalmo elegans, Santanichthys diasii, Scombroclupeoides scutata e novo euteleósteo da Bacia de Pelotas) e 14 táxons de localidades estrangeiras (i.e., Avitosmerus canadensis, Barcarenichthys joneti, Chanoides macropoma, Clupavus maroccanus, Gaudryella gaudryi, Humbertia operta, Kermichthys daguini, Leptolepides spratiiformis, Lusitanichthys characiformis, Nybelinoides brevis, Orthogonikleithrus leichi, Pattersonella formosa, Wenzichthys congolensis e Tchernovichthys exspectatum) foram analisados através de observação direta, fotografias, desenhos e descrições e submetidos a uma análise de Sistemática Filogenética utilizando o princípio da parcimônia. Três espécies recentes (i.e., Elops saurus, Hoplias malabaricus e Salmo trutta) foram usadas como grupo externo. Sessenta e dois caracteres foram selecionados e, como resultado, seis árvores igualmente parcimoniosas foram obtidas com 325 passos, índice de consistência (CI) de 0,2523 e índice de retenção (RI) de 0,4309. O consenso estrito é representado pela seguinte topologia: ((C. marocanus), (C. brasiliensis, (H. malabaricus + S. diasii))) ((G. gaudryi, (C. macropoma + L. characiformis)), (K. daguini), ((A. canadensis, (novo euteleósteo, (S. elegans + W. congolensis), (B. ouricuriensis + B. marizalensis)), (L. spratiiformis, (S. scutata, (N. brevis + P. formosa))), (B. joneti), (O. leichi), (H. operta + T. exspectatum), indicando que euteleósteos basais não formam um grupo monofilético e que as atuais sinapomorfias propostas são insuficientes para suportar o grupo.

HfO2 is one of the most important high refractive index materials for depositing high power optical mirrors. In this research, HfO2 thin films were prepared by dual-ion beam reactive sputtering method, and the laser-induced damage thresholds (LIDT) of the sample were measured in 1-on-1 mode for laser with 1064 nm wavelength. The results indicate that the LIDT of the as-grown sample is only 3.96 J/cm(2), but it is increased to 8.98 J/cm(2) after annealing under temperature of 200 degrees C in atmosphere. By measuring the laser weak absorption and SIMS of the samples, we deduced that substoichiometer is the main reason for the low LIDT of the as-grown sample, and the experiment results were well explained with the theory of electronic-avalanche ionization. The evolutionary associations between closely related fish species, both contemporary and historical, are frequently assessed by using molecular markers, such as microsatellites. Here, the presence and variability of microsatellite loci in two closely related species of marine fishes, sand seatrout (Cynoscion arenarius) and silver seatrout (C. nothus), are explored by using heterologous primers from red drum (Sciaenops ocellatus). Data from these loci are used in conjunction with morphological characters and mitochondrial DNA haplotypes to explore the extent of genetic exchange between species offshore of Galveston Bay, TX. Despite seasonal overlap in distribution, low genetic divergence at microsatellite loci, and similar life history parameters of C. arenarius and C. nothus, all three data sets indicated that hybridization between these species does not occur or occurs only rarely and that historical admixture in Galveston Bay after divergence between these species was unlikely. These results shed light upon the evolutionary history of these fishes and highlight the genetic properties of each species that are influenced by their life history and ecology.

A família Zingiberaceae é a mais representativa da ordem Zingiberales, contendo mais de 1.000 espécies divididas em quatro subfamílias e seis tribos, amplamente distribuídas por todos os continentes. Nas últimas décadas houve um incremento nos estudos relativos à distribuição, o grau de especialização e a funcionalidade dos elementos de vaso em diversas famílias de monocotiledôneas. Entretanto, ainda existem poucos estudos que contribuam para o delineamento dos aspectos evolutivos e ecológicos das tribos de Zingiberaceae. Os objetivos desse trabalho são os de comparar a anatomia dos órgãos subterrâneos e aéreos de oito espécies de Zingiberaceae e estabelecer a distribuição dos elementos traqueais, bem como, o de determinar a especialização dos elementos de vaso de vinte e oito espécies pertencentes a três tribos Alpineae, Zingibereae e Globbeae. As espécies foram coletadas em áreas naturais protegidas e em áreas de cultivos particulares no estado do Rio de Janeiro. Os órgãos subterrâneos e aéreos foram processados de acordo com as técnicas usuais de microscopia óptica e eletrônica de varredura. A análise estrutural do eixo vegetativo das oito espécies pertencentes aos gêneros Alpinia, Renealmia, Curcuma, Hedychium e Zingiber indicam uma similaridade e mostram que os elementos de vaso estão restritos às raízes. Alguns caracteres estruturais dos elementos de vaso, como o tipo da placa de perfuração, o número de barras e o tipo de espessamento parietal se mostraram importantes para o estabelecimento da relação entre as subfamílias e tribos. Zingibereae e Globbeae reúnem estados de caracteres mais basais, como placa de perfuração escalariforme e espessamento parietal espiralado, e os mais derivados são encontrados na tribo Alpinieae, incluindo placa de perfuração simples e espessamento parietal parcialmente pontoado
