Tutkielmassa tarkastellaan Pohjois-Suomen savottakämpillä vuosien 1945 1975 välillä työskennelleitä kämppäemäntiä. Kämppäemännät toimivat metsätyöntekijöiden yhteisasunnoissa ruuanlaittajina ja siivoojina. Savottakämpille alettiin palkata kokkeja 1900-luvun alussa ja 1930-luvulta eteenpäin puutavarayhtiöt alkoivat huolehtia heidän palkkaamisestaan. Yhtiöiden palkkaamien ruuanlaittajien ammattinimikkeesi vakiintui kämppäemäntä. Kämppämajoitus väheni 1970-luvun myötä kun metsätyössä siirryttiin työntekijöiden kotikuljetuksiin. Pohjois-Suomessa kämppätyömaita ja kämppäemäntiä oli kuitenkin 1980-luvun lopulle asti. Kämppäemännät työskentelivät maskuliinisella metsäalalla kämppäyhteisöjen ainoina naisina. Tutkielmassa kysytäänkin, minkälaisia käsityksiä ja määritelmiä kämppäemännyyteen yhdistettiin ja miten kämppäemännän sukupuoli näkyy näissä määritelmissä. Lisäksi kysytään, minkälaisina kämpän sisäiset sukupuolten väliset suhteet näyttäytyivät. Tarkastelussa hyödynnetään Yvonne Hirdmanin sukupuolijärjestelmän käsitettä. Tutkimuskysymyksiä lähestytään kolmesta näkökulmasta: Ensin tarkastellaan, miten kämppäemännyyttä määritellään aikalaiskirjallisuudessa. Tässä tarkastelussa tärkeimpänä lähdeaineistona toimivat kämppäemännille suunnatut oppaat. Toiseksi tarkastellaan, miten kämppäemäntinä toimineet naiset vastasivat näihin määritelmiin ja minkälaiseksi he kokivat kämpillä vallinneet sukupuolten väliset suhteet. Kolmanneksi kuvataan, mitä savottakämpillä majoittuneet metsäalalla toimineet miehet näkivät hyvän kämppäemännän ominaisuuksiksi ja minkälaisiksi he kokivat emännän aseman kämppäyhteisössä. Kahden viimeisen näkökulman lähdeaineistona toimii muistitietoaineisto. Kämppäemännät toimivat savottakämpillä erilaisten odotusten ristipaineessa. Kämppäemännän oppaat määrittelevät heidän roolinsa feminiiniseksi ja äidilliseksi. Ne luovat kämppäemännän työstä naisten yhteiskunnallisen roolin mukaista määrittelemällä kämpän kodiksi ja emännän sen hengettäreksi, joka huolehtii miesten hyvinvoinnista. Kämpillä majoittuneet miehet sen sijaan arvostavat kämppäemäntää, joka on rempseä ja huumorintajuinen. Kämppäemännän kuului sopeutua kämpän maskuliiniseen kulttuuriin, mikä onnistui parhaiten osallistumalla sen huumoriin. Kämpän sukupuolijärjestelmä perustui sukupuolitettuun työnjakoon ja kämppätilan sukupuolenmukaiseen jakamiseen. Kämpän keittiö ja emännän huone olivat naisille kuuluvaa yksityisaluetta, josta oltiin yhteydessä miesten puolelle vain tarjoiluluukun välityksellä. Sukupuolten erillään pitämistä perusteltiin kämppäemännän suojelemisella, mutta sen tavoitteena oli myös estää sukupuolisuhteiden syntyminen kämpän miesten ja kämppäemännän välille. Kämppäemäntä olikin virallisesti rauhoitettu ja emännän koskemattomuudesta huolehtiminen oli kämppäyhteisön vastuulla. Kämppäelämässä syntyi kuitenkin seurustelusuhteita ja mahdollisesti myös sukupuolisuhteita. Näistä ei kuitenkaan mielellään kerrota haastatteluissa. Myös seksuaalista häirintää esiintyi. Kämppäemännät kuitenkin korostavat miesten kunnioittavaa suhtautumista heihin. He korostavat, etteivät sukupuolten väliset suhteet olleet ongelmallisia ja painottavat omaa sukupuolimoraaliaan. Kämppäemännät näkevät itsensä kämppäyhteisön jäsenenä, eivätkä halua puhua pahaa muusta yhteisöstä. Vaikeita tilanteita kuvatessaan he korostavat omaa aktiivisuuttaan ja selviytymistään. Kämppäemännät luovat itsestään kuvaa selviytyjinä ja vahvoina naisina.

Tutkimukseni tarkoitus on paikallistaa polkuja hyvän ja pahan tuolle puolen. Tutkimukseni termit hyvä ja paha määrittyvät moraalisesti. Täten tutkimusaiheeni on ihmisen ja moraalin välinen suhde. Tutkin ihmisen moraalista olotilaa, siinä tapahtuvaa muutosta ja tämän muutoksen ehtoja. Tutkimuksellani on kaksi pääkysymystä: kuinka on mahdollista päästä hyvän ja pahan tuolle puolen ja miksi meidän tulisi siirtyä hyvän ja pahan tuolle puolen. Polut hyvän ja pahan tuolle puolen vievät myös etiikan tuolle puolen. Tutkimukseni pyrkii selvittämään mitä vikaa etiikassa on ja kuinka on mahdollista perustella toimintaa ilman etiikkaa. Tutkimukseni metodi on systemaattinen analyysi. Johdanto-osassa käsittelen yleisesti moraalia, sen vaikutuspiiriä ja niitä syitä, joiden vuoksi se täytyy hylätä. Tarkastelen myös etiikkaa ja selvitän sitä laajempaa kontekstia mihin se liittyy. Ensimmäisessä luvussa käsittelen Martti Lutherin teologiaa. Lutherilla moraalia edustaa Jumalan laki, jonka alaisuudesta kristitty pääsee vapaaksi kun Jeesus ottaa ristinkuolemassaan ihmisten synnin itselleen ja kärsii Jumalan lain mukaisen kuolemanrangaistuksen. Uskon yhteydessä Kristukseen kristitty kohoaa lain ulkopuolelle tekemään rakkauden tekoja. Toinen luku on omistettu Søren Kierkegaardin kristilliselle eksistentialismille. Kierkegaardin eksistenssitasojen filosofiassa uskonnollinen olemisen taso paljastuu korkeammaksi kuin eettinen. Tässä nousee tärkeäksi Kierkegaardin kirja Pelko ja vavistus. Kierkegaard tutkii Raamatun kertomusta Aabrahamista, jolle Jumala antaa käskyn uhrata oma poikansa Iisak. Kierkegaardille tarina on esimerkki eettistä korkeammasta ja sitä tilapäisesti vastustavasta olemisen tavasta. Kolmannessa luvussa käsittelen Dietrich Bonhoefferin teologiaa. Bonhoefferin mukaan Jeesus kutsuu ihmisiä seuraamaan häntä. Tämä on kutsu hyvän ja pahan tuolle puolen paratiisin viattomuuteen, joka rikkoo eettisyyden ihmisen ja Jeesuksen välillä. Neljännessä luvussa tarkastelen Friedrich Nietzschen filosofiaa ja pyrin ymmärtämään mitä hänen ohjelmansa hyvän ja pahan tuolle puolen tarkoittaa. Nietzschen vallantahdon filosofiassa moraali on heikkojen keino alistaa vahvat. Tarkastelen Nietzschen käsitystä moraalin synnystä ja sen haitallisuudesta ja pyrin selvittämään kuinka herrat elävät hyvän ja pahan tuolla puolen. Viidennessä luvussa tutkin Jaques Derridan grammatologiaa. Derridan mukaan teksti on suljettu tila, jonka ulkopuolelle on mahdoton päästä. Derrida sanoittaa kuitenkin sanoittamatonta tilattomuutta tekstin tuolla puolen. Tästä ei-paikasta käsin hypereettinen dekonstruktio purkaa perinteistä etiikkaa. Kuudennessa luvussa tarkastelen Mark C. Taylorin epä/teologiaa. Taylorille olotilan muutosta kuvaa termi harhautuminen , joka vie ihmisen lainsuojattomuuteen hyvän ja pahan tuolle puolen. Selvitän mitä harhautuminen tarkoittaa ja miten se liittyy Taylorin Jumalan kuoleman epä/teologiaan. Johtopäätösluvussa tuon esiin tutkimustuloksia, nostan esille joitain pääluvuissa esiintyviä teemoja ja vertaan tutkimuskohteitteni näkemyksiä toisiinsa. Lutherin ja Nietzschen perinteissä on paljon yhteistä, mutta pohjimmiltaan ne sotivat toisiaan vastaan. Hyvän ja pahan tuolle puolen johtaa kaksi polkua. Toinen niistä vie paratiisiin, toinen tuhoon.

Leaf and needle biomasses are key factors in forest health. Insects that feed on needles cause growth losses and tree mortality. Insect outbreaks in Finnish forests have increased rapidly during the last decade and due to climate change the damages are expected to become more serious. There is a need for cost-efficient methods for inventorying these outbreaks. Remote sensing is a promising means for estimating forests and damages. The purpose of this study is to investigate the usability of airborne laser scanning in estimating Scots pine defoliation caused by the common pine sawfly (Diprion pini L.). The study area is situated in Ilomantsi district, eastern Finland. Study materials included high-pulse airborne laser scannings from July and October 2008. Reference data consisted of 90 circular field plots measured in May-June 2009. Defoliation percentage on these field plots was estimated visually. The study was made on plot-level and methods used were linear regression, unsupervised classification, Maximum likelihood method, and stepwise linear regression. Field plots were divided in defoliation classes in two different ways: When divided in two classes the defoliation percentages used were 0–20 % and 20–100 % and when divided in four classes 0–10 %, 10–20 %, 20–30 % and 30–100 %. The results varied depending on method and laser scanning. In the first laser scanning the best results were obtained with stepwise linear regression. The kappa value was 0,47 when using two classes and 0,37 when divided in four classes. In the second laser scanning the best results were obtained with Maximum likelihood. The kappa values were 0,42 and 0,37, correspondingly. The feature that explained defoliation best was vegetation index (pulses reflected from height > 2m / all pulses). There was no significant difference in the results between the two laser scannings so the seasonal change in defoliation could not be detected in this study.

The location and location guidance of shopping centers has been under much public discussion in Finland in the recent years. The Ministry of the Environment has expressed concern over the sustainability of ‘out-of-town’ shopping centers. Shopping centers outside the urban form are seen to cause more traffic, thus contributing to climate change by increasing carbon dioxide emissions. The sustainability of urban form has been researched in several studies and factors like urban density, public transport and a comfortable living environment were found to be the most important. This study presents the views of Finnish shopping center stakeholders on the sustainability of shopping center locations. These views were gathered using focus groups. Stakeholders included managers, consultants, investors, developers, architects and tenants of shopping centers and public sector actors dealing with shopping industry. As one theme in the discussions, participants were asked to present their views on the sustainability of shopping centers’ current locations. The study is part of the Aalto University of Technology KOKKKA project, which has its main focus upon shopping centers and sustainability. Shopping centers were seen to affect sustainability mainly through their location. A sustainable location was thought of as one that involved locating in an economically successful place, inside the urban form. A sustainable location was also easily accessible, with good access via public transport and the shopping center also had to create comfortable living environment in its surroundings. The views of the focus groups participants are similar to the views in sustainable urban structure theories and, inter alia, Finland’s national sustainable development strategy.

Tutkielman aiheena on kirjoitetun digitaalisessa muodossa olevan tekstin kielen automaattinen tunnistaminen. Tekstin kieli on usein tarpeellista identifioida, jotta tekstin jatkokäsittelyssä osataan toimia käyttäen oikeita menetelmiä. Tekstin kielen automaattinen tunnistaminen on internetissä olevan tekstimäärän ja kielivalikoiman kasvaessa tullut tärkeäksi näiden dokumenttien automaattisen käsittelyn esivaiheeksi. Kielen tunnistaminen on kieleltään tuntemattoman tekstin vertaamista joukkoon annettuja kieliä. Samoja tai hyvin läheisiä menetelmiä voidaan käyttää myös tekstin lajitteluun esimerkiksi aihealueiden suhteen. Tässä tutkielmassa esitellään tutkielmaa varten rakennetun kielentunnistimen kehitysvaiheet sekä sen suorituskyvyn evaluointia. Aluksi tutkielmassa paneudutaan muutamaan eri kielentunnistamisen menetelmään, sekä esitellään myös eräiden toteutettujen kielentunnistinten rakennetta niistä kirjoitettujen artikkelien perusteella. Tämän jälkeen tutkielmassa kerrotaan kuinka kielentunnistimen rakentamiseen luotiin harjoituskorpuksia. Harjoituskorpukset luotiin Wikipedian artikkeleista yhteensä 103 kielelle. Eri kielten harjoituskorpusten yhteenlasketuksi kooksi tuli yli miljardi sanetta. Jokaisesta harjoituskorpuksesta luotiin 7 kielimallia kullekin kielelle. Kielimallit ovat kielten sanojen n-grammien (yhdestä kuuteen) listoja sekä listoja kielten koko-naisista sananmuodoista. Harjoituskorpuksista luotuja kielimalleja ja muutamia eri menetelmiä yhdistelemällä rakennettiin useita erilaisia kielentunnistimia, joiden suoriutumista vertailtiin keskenään laajojen testitunnistusten avulla. Testien suorittamista varten muodostettiin Europarl-korpuksesta noin 200 miljoonan sanan testikorpuksia kymmenelle eurooppalaiselle kielelle. Testitunnistuksia tehtiin automatisoidusti useita miljoonia. Näiden testien tulosten perusteella päädyttiin kielentunnistimeen, joka käyttää hyväkseen kaikkia kielimalleja sekä kahta eri menetelmää. Tutkielmassa kehitetyn kielentunnistimen suoriutumista vertailtiin vielä joidenkin artikkeleissa esiteltyjen kielentunnistimien suoriutumiseen ja lopuksi tutkielmassa esitellään mahdollisuuksia tunnistimen jatkokehittämiseen.

According to certain arguments, computation is observer-relative either in the sense that many physical systems implement many computations (Hilary Putnam), or in the sense that almost all physical systems implement all computations (John Searle). If sound, these arguments have a potentially devastating consequence for the computational theory of mind: if arbitrary physical systems can be seen to implement arbitrary computations, the notion of computation seems to lose all explanatory power as far as brains and minds are concerned. David Chalmers and B. Jack Copeland have attempted to counter these relativist arguments by placing certain constraints on the definition of implementation. In this thesis, I examine their proposals and find both wanting in some respects. During the course of this examination, I give a formal definition of the class of combinatorial-state automata , upon which Chalmers s account of implementation is based. I show that this definition implies two theorems (one an observation due to Curtis Brown) concerning the computational power of combinatorial-state automata, theorems which speak against founding the theory of implementation upon this formalism. Toward the end of the thesis, I sketch a definition of the implementation of Turing machines in dynamical systems, and offer this as an alternative to Chalmers s and Copeland s accounts of implementation. I demonstrate that the definition does not imply Searle s claim for the universal implementation of computations. However, the definition may support claims that are weaker than Searle s, yet still troubling to the computationalist. There remains a kernel of relativity in implementation at any rate, since the interpretation of physical systems seems itself to be an observer-relative matter, to some degree at least. This observation helps clarify the role the notion of computation can play in cognitive science. Specifically, I will argue that the notion should be conceived as an instrumental rather than as a fundamental or foundational one.

A straightforward computation of the list of the words (the `tail words' of the list) that are distributionally most similar to a given word (the `head word' of the list) leads to the question: How semantically similar to the head word are the tail words; that is: how similar are their meanings to its meaning? And can we do better? The experiment was done on nearly 18,000 most frequent nouns in a Finnish newsgroup corpus. These nouns are considered to be distributionally similar to the extent that they occur in the same direct dependency relations with the same nouns, adjectives and verbs. The extent of the similarity of their computational representations is quantified with the information radius. The semantic classification of head-tail pairs is intuitive; some tail words seem to be semantically similar to the head word, some do not. Each such pair is also associated with a number of further distributional variables. Individually, their overlap for the semantic classes is large, but the trained classification-tree models have some success in using combinations to predict the semantic class. The training data consists of a random sample of 400 head-tail pairs with the tail word ranked among the 20 distributionally most similar to the head word, excluding names. The models are then tested on a random sample of another 100 such pairs. The best success rates range from 70% to 92% of the test pairs, where a success means that the model predicted my intuitive semantic class of the pair. This seems somewhat promising when distributional similarity is used to capture semantically similar words. This analysis also includes a general discussion of several different similarity formulas, arranged in three groups: those that apply to sets with graded membership, those that apply to the members of a vector space, and those that apply to probability mass functions.

This research is connected with an education development project for the four-year-long officer education program at the National Defence University. In this curriculum physics was studied in two alternative course plans namely scientific and general. Observations connected to the later one e.g. student feedback and learning outcome gave indications that action was needed to support the course. The reform work was focused on the production of aligned course related instructional material. The learning material project produced a customized textbook set for the students of the general basic physics course. The research adapts phases that are typical in Design Based Research (DBR). The research analyses the feature requirements for physics textbook aimed at a specific sector and frames supporting instructional material development, and summarizes the experiences gained in the learning material project when the selected frames have been applied. The quality of instructional material is an essential part of qualified teaching. The goal of instructional material customization is to increase the product's customer centric nature and to enhance its function as a support media for the learning process. Textbooks are still one of the core elements in physics teaching. The idea of a textbook will remain but the form and appearance may change according to the prevailing technology. The work deals with substance connected frames (demands of a physics textbook according to the PER-viewpoint, quality thinking in educational material development), frames of university pedagogy and instructional material production processes. A wide knowledge and understanding of different frames are useful in development work, if they are to be utilized to aid inspiration without limiting new reasoning and new kinds of models. Applying customization even in the frame utilization supports creative and situation aware design and diminishes the gap between theory and practice. Generally, physics teachers produce their own supplementary instructional material. Even though customization thinking is not unknown the threshold to produce an entire textbook might be high. Even though the observations here are from the general physics course at the NDU, the research gives tools also for development in other discipline related educational contexts. This research is an example of an instructional material development work together the questions it uncovers, and presents thoughts when textbook customization is rewarding. At the same time, the research aims to further creative customization thinking in instruction and development. Agriculture is an economic activity that heavily relies on the availability of natural resources. Through its role in food production agriculture is a major factor affecting public welfare and health, and its indirect contribution to gross domestic product and employment is significant. Agriculture also contributes to numerous ecosystem services through management of rural areas. However, the environmental impact of agriculture is considerable and reaches far beyond the agroecosystems. The questions related to farming for food production are, thus, manifold and of great public concern. Improving environmental performance of agriculture and sustainability of food production, sustainabilizing food production, calls for application of wide range of expertise knowledge. This study falls within the field of agro-ecology, with interphases to food systems and sustainability research and exploits the methods typical of industrial ecology. The research in these fields extends from multidisciplinary to interdisciplinary and transdisciplinary, a holistic approach being the key tenet. The methods of industrial ecology have been applied extensively to explore the interaction between human economic activity and resource use. Specifically, the material flow approach (MFA) has established its position through application of systematic environmental and economic accounting statistics. However, very few studies have applied MFA specifically to agriculture. The MFA approach was used in this thesis in such a context in Finland. The focus of this study is the ecological sustainability of primary production. The aim was to explore the possibilities of assessing ecological sustainability of agriculture by using two different approaches. In the first approach the MFA-methods from industrial ecology were applied to agriculture, whereas the other is based on the food consumption scenarios. The two approaches were used in order to capture some of the impacts of dietary changes and of changes in production mode on the environment. The methods were applied at levels ranging from national to sector and local levels. Through the supply-demand approach, the viewpoint changed between that of food production to that of food consumption. The main data sources were official statistics complemented with published research results and expertise appraisals. MFA approach was used to define the system boundaries, to quantify the material flows and to construct eco-efficiency indicators for agriculture. The results were further elaborated for an input-output model that was used to analyse the food flux in Finland and to determine its relationship to the economy-wide physical and monetary flows. The methods based on food consumption scenarios were applied at regional and local level for assessing feasibility and environmental impacts of relocalising food production. The approach was also used for quantification and source allocation of greenhouse gas (GHG) emissions of primary production. GHG assessment provided, thus, a means of crosschecking the results obtained by using the two different approaches. MFA data as such or expressed as eco-efficiency indicators, are useful in describing the overall development. However, the data are not sufficiently detailed for identifying the hot spots of environmental sustainability. Eco-efficiency indicators should not be bluntly used in environmental assessment: the carrying capacity of the nature, the potential exhaustion of non-renewable natural resources and the possible rebound effect need also to be accounted for when striving towards improved eco-efficiency. The input-output model is suitable for nationwide economy analyses and it shows the distribution of monetary and material flows among the various sectors. Environmental impact can be captured only at a very general level in terms of total material requirement, gaseous emissions, energy consumption and agricultural land use. Improving environmental performance of food production requires more detailed and more local information. The approach based on food consumption scenarios can be applied at regional or local scales. Based on various diet options the method accounts for the feasibility of re-localising food production and environmental impacts of such re-localisation in terms of nutrient balances, gaseous emissions, agricultural energy consumption, agricultural land use and diversity of crop cultivation. The approach is applicable anywhere, but the calculation parameters need to be adjusted so as to comply with the specific circumstances. The food consumption scenario approach, thus, pays attention to the variability of production circumstances, and may provide some environmental information that is locally relevant. The approaches based on the input-output model and on food consumption scenarios represent small steps towards more holistic systemic thinking. However, neither one alone nor the two together provide sufficient information for sustainabilizing food production. Environmental performance of food production should be assessed together with the other criteria of sustainable food provisioning. This requires evaluation and integration of research results from many different disciplines in the context of a specified geographic area. Foodshed area that comprises both the rural hinterlands of food production and the population centres of food consumption is suggested to represent a suitable areal extent for such research. Finding a balance between the various aspects of sustainability is a matter of optimal trade-off. The balance cannot be universally determined, but the assessment methods and the actual measures depend on what the bottlenecks of sustainability are in the area concerned. These have to be agreed upon among the actors of the area

Recently it has been recognized that evolutionary aspects play a major role in conservation issues of a species. In this thesis I have combined evolutionary research with conservation studies to provide new insight into these fields. The study object of this thesis is the house sparrow, a species that has features that makes it interesting for this type of study. The house sparrow has been ubiquitous almost all over the world. Even though being still abundant, several countries have reported major declines. These declines have taken place in a relatively short time covering both urban and rural habitats. In Finland this species has declined by more than two thirds in just over two decades. In addition, as the house sparrow lives only in human inhabited areas it can also raise public awareness to conservation issues. I used both an extensive museum collection of house sparrows collected in 1980s from all over Finland as well as samples collected in 2009 from 12 of the previously collected localities. I used molecular techniques to study neutral genetic variation within and genetic differentiation between the study populations. This knowledge I then combined with data gathered on morphometric measurements. In addition I analyzed eight heavy metals from the livers of house sparrows that lived in either rural or urban areas in the 1980s and evaluated the role of heavy metal pollution as a possible cause of the declines. Even though dispersal of house sparrows is limited I found that just as the declines started in 1980s the house sparrows formed a genetically panmictic population on the scale of the whole Finland. When compared to Norway, where neutral genetic divergence has been found even with small geographic distances, I concluded that this difference would be due to contrasting landscapes. In Finland the landscape is rather homogeneous facilitating the movements of these birds and maintaining gene flow even with the low dispersal. To see whether the declines have had an effect on the neutral genetic variation of the populations I did a comparison between the historical and contemporary genetic data. I showed that even though genetic diversity has not decreased due to the drastic declines the populations have indeed become more differentiated from each other. This shows that even in a still quite abundant species the declines can have an effect on the genetic variation. It is shown that genetic diversity and differentiation may approach their new equilibriums at different rates. This emphasizes the importance of studying both of them and if the latter has increased it should be taken as a warning sign of a possible loss of genetic diversity in the future. One of the factors suggested to be responsible for the house sparrow declines is heavy metal pollution. When studying the livers of house sparrows from 1980s I discovered higher levels of heavy metal concentrations in urban than rural habitats, but the levels of the metals were comparatively low and based on that heavy metal pollution does not seem to be a direct cause for the declines in Finland. However, heavy metals are known to decrease the amount of insects in urban areas and thus in the cities heavy metals may have an indirect effect on house sparrows. Although neutral genetic variation is an important tool for conservation genetics it does not tell the whole story. Since neutral genetic variation is not affected by selection, information can be one-sided. It is possible that even neutral genetic differentiation is low, there can be substantial variation in additive genetic traits indicating local adaptation. Therefore I performed a comparison between neutral genetic differentiation and phenotypic differentiation. I discovered that two traits out of seven are likely to be under directional selection, whereas the others could be affected by random genetic drift. Bergmann s rule may be behind the observed directional selection in wing length and body mass. These results highlight the importance of estimating both neutral and adaptive genetic variation.

Leuconostoc spp. are lactic acid bacteria (LAB) implicated in food spoilage, especially on refrigerated, modified atmosphere packaged (MAP) meats. The overall aim of this thesis was to learn more about Leuconostoc spp. as food spoilage organisms with a focus on commercial products where LAB spoilage is considered a problem and the main factor limiting shelf-life. Therefore, we aimed to identify Leuconostoc spp. involved in food spoilage, as well as to characterise the spoilage reactions they caused and their contamination sources during poultry meat processing. In addition, we examined the distribution of strains of Leuconostoc gasicomitatum in different food commodities. Finally, we analysed the genome content of L. gasicomitatum LMG 18811 with a special focus on metabolic pathways related to food spoilage. The findings show that Leuconostoc gelidum and L.gasicomitatum were responsible for the discoloration and off-odours developed in beef steaks. Together with Leuconostoc mesenteroides, these Leuconostoc spp., also cause spoilage of vegetable sausages. In contrast, we showed that Leuconostoc spp. are not important for the shelf-life or quality of non-marinated broiler products although, in marinated broiler fillet products, Leuconostoc spp., L.gasicomitatum in particular, are considered spoilage organisms. Furthermore, the findings of the contamination survey we carried out in a poultry processing plant indicated that spoilage Leuconostoc spp. are derived from the processing environment rather than from the broilers, and that air movement distributes psychrotrophic spoilage LAB, including leuconostocs, and has an important role in meat contamination during poultry processing. Pulsed-field gel electrophoresis (PFGE) based genotyping of L. gasicomitatum strains demonstrated that certain genotypes are common in various meat products. In contrast, genotypes associated with meat were not recovered in vegetable-based sources. This suggests that these two food categories either become contaminated with, or favour the growth of different genotypes. Furthermore, the results indicated that the meat processing environment contributes to L. gasicomitatum contamination as certain genotypes were repeatedly identified from products of the same processing plant. Finally, the sequenced and annotated genome of L.gasicomitatum LMG 18811 allowed us to identify the metabolic pathways and reactions resulting in food spoilage.

This thesis examines the media debate on pensions. The case analysed in the thesis is the debate that sharpened after the Finnish government made the decision to raise the retirement age. The analysed data consists of articles published in the printed media during one month after the decision was made on 24th of February in 2009. The aim of the study is to describe how the decision is argued about by different positions of speakers and how retirement is justified from the perspective of the individual. Furthermore, the purpose is to discover different ways of discussing the pensioner. The theoretical frame for this study is social constructivism, which understands reality as socially constructed with language. From this perspective, media texts can be seen as one form of shaping reality. The data is analysed by using different methods. Thematisation is used to discover the key topics, and quantification is used to examine the prevalence of different arguments. The method in which the speaker’s ways of speaking is analysed in different participant categories I call “a speaker position analysis”. The debate around the decision to raise the retirement age highlight the power struggle both between the government and the opposition as well as the government and employee unions. One thing all discussants agree is the need to raise the retirement age. From the individual's perspective, retirement is justified mostly with hard working conditions and inadequacy of health. The pensioner's image is appearing gloomy in most discourses. Prevailing discourses are seeing a pensioner either sick and tired or someone who is not good for work and has lost his dignity. The debate around the decision is intertwined around the concepts of welfare state and individual's well-being. In the postmodern society, human preferences are individualised. Welfare state means different things to different people, as well as the individual's subjective perception of well-being is unique. These two aspects are the ones which raise the tension in the analysed media debate.

Protein modification via enzymatic cross-linking is an attractive way for altering food structure so as to create products with increased quality and nutritional value. These modifications are expected to affect not only the structure and physico-chemical properties of proteins but also their physiological characteristics, such as digestibility in the GI-tract and allergenicity. Protein cross-linking enzymes such as transglutaminases are currently commercially available, but also other types of cross-linking enzymes are being explored intensively. In this study, enzymatic cross-linking of β-casein, the most abundant bovine milk protein, was studied. Enzymatic cross-linking reactions were performed by fungal Trichoderma reesei tyrosinase (TrTyr) and the performance of the enzyme was compared to that of transglutaminase from Streptoverticillium mobaraense (Tgase). Enzymatic cross-linking reactions were followed by different analytical techniques, such as size exclusion chromatography -Ultra violet/Visible multi angle light scattering (SEC-UV/Vis-MALLS), phosphorus nuclear magnetic resonance spectroscopy (31P-NMR), atomic force (AFM) and matrix-assisted laser desorption/ionisation-time of flight mass spectrometry (MALDI-TOF MS). The research results showed that in both cases cross-linking of β-casein resulted in the formation of high molecular mass (MM ca. 1 350 kg mol-1), disk-shaped nanoparticles when the highest enzyme dosage and longest incubation times were used. According to SEC-UV/Vis-MALLS data, commercial β-casein was cross-linked almost completely when TrTyr and Tgase were used as cross-linking enzymes. In the case of TrTyr, high degree of cross-linking was confirmed by 31P-NMR where it was shown that 91 % of the tyrosine side-chains were involved in the cross-linking. The impact of enzymatic cross-linking of β-casein on in vitro digestibility by pepsin was followed by various analytical techniques. The research results demonstrated that enzymatically cross-linked β-casein was stable under the acidic conditions present in the stomach. Furthermore, it was found that cross-linked β-casein was more resistant to pepsin digestion when compared to that of non modified β-casein. The effects of enzymatic cross-linking of β-casein on allergenicity were also studied by different biochemical test methods. On the basis of the research results, enzymatic cross-linking decreased allergenicity of native β-casein by 14 % when cross-linked by TrTyr and by 6 % after treatment by Tgase. It can be concluded that in addition to the basic understanding of the reaction mechanism of TrTyr on protein matrix, the research results obtained in this study can have high impact on various applications like food, cosmetic, medical, textile and packing sectors.

Tutkimukseni tarkoituksena on tutkia kaikkein varhaisimpia Uuden testamentin kanonisia ja apokryfisia käsikirjoituksia sekä niiden kirjureita. Huomion keskipisteenä ovat kyseisten käsikirjoitusten ulkoiset piirteet, kuten esimerkiksi kirjoitusmateriaali, muoto, koko ja käsiala. Näiden avulla vertaan kanonisia käsikirjoituksia apokryfisiin ja pyrin selvittämään, erosivatko näiden kopiointimenetelmät toisistaan. Yksi keskeisimmistä kysymyksistä on se, voidaanko ulkoisten piirteiden avulla päätellä jotain käsikirjoitusten asemasta ja arvostuksesta. Tutkimuksen aluksi esittelen tutkimusaineistoni käsikirjoitukset ja tekstit. Päädyin vertaamaan kanonisia evankeliumeita kaikkiin 100- ja 200-luvuilta löydettyihin apokryfisiin käsikirjoituksiin, joihin sisältyy esimerkiksi Tuomaan ja Pietarin evankeliumit sekä Hermaan paimen. Kaikki tämän ajanjakson kristilliset käsikirjoitukset ovat löytyneet Egyptistä, minkä vuoksi tutkimukseni keskittyy Egyptiin ja sen kirjureihin. Ennen varsinaista käsikirjoitusten analyysiä käsittelen käsikirjoitusten löytöpaikkoja, kaanonin sekä antiikin kirjureiden historiaa. Ensimmäiseksi käsittelen käsikirjoitusten materiaalia, eli papyrusta ja pergamenttia. Tässä suhteessa kanoniset käsikirjoitukset eivät eroa apokryfisista, vaan aineistot ovat tämän suhteen identtisiä. Tämän jälkeen huomio kiinnittyy käsikirjoitusten muotoon, eli siihen kirjoitettiinko käsikirjoitukset koodeksiin vai kääröön. Analyysin edetessä paljastuu, että apokryfisissa käsikirjoituksissa käytettiin useammin käärömuoto kuin kanonisissa käsikirjoituksissa. Voidaan esittää, että kristityt kirjoittivat koodeksiin kaikkein arvostetuimmat tekstit, joten käärömuoto voi kertoa käsikirjoituksen sisältävän tekstin alemmasta arvostuksesta. Tutkimuksen seuraavissa osioissa käsitellään koodeksien kokoa, marginaaleja, palstoja sekä käsialan kokoa. Lukujen perusteella voidaan sanoa, että kanoniset koodeksit muodostavat hieman yhtenäisemmän linjan kuin apokryfiset koodeksit. Kanonisten koodeksien kirjureiden toimintatavat ovat siis olleet hieman yhtenäisempiä. Käsikirjoitusten käsialan tarkempi analyysi paljastaa kuitenkin, että niin kanoniset kuin apokryfiset käsikirjoitukset kirjoitettiin suhteellisen hyvällä käsialalla. Tutkimuksen viimeisessä luvussa käy ilmi, että apokryfisten koodeksien epäyhtenäisyys johtuu osittain niiden erilaisista käyttötarkoituksista. Apokryfisia koodekseja valmistettiin enemmän kristittyjen henkilökohtaiseen käyttöön verrattuna kanonisiin koodekseihin. Tästä huolimatta suurin osa käsikirjoituksista oli seurakuntien liturgisessa käytössä. Toisin sanoen niin kanonisia evankeliumeita kuin apokryfisia tekstejä luettiin ääneen seurakuntien kokoontumisissa, mikä kertoo niiden arvosta.

Migraine is the common cause of chronic episodic headache, affecting 12%-15% of the Caucasian population (41 million Europeans and some half a million Finns), and causes considerable loss of quality of life to its sufferers, as well as being linked to increased risk for a wide range of conditions, from depression to stroke. Migraine is the 19th most severe disease in terms of disability-adjusted life years, and 9th among women. It is characterized by attacks of headache accompanied by sensitivity to external stimuli lasting 4-72 hours, and in a third of cases by neurological aura symptoms, such as loss of vision, speech or muscle function. The underlying pathophysiology, including what triggers migraine attacks and why they occur in the first place, is largely unknown. The aim of this study was to identify genetic factors associated with the hereditary susceptibility to migraine, in order to gain a better understanding of migraine mechanisms. In this thesis, we report the results of genetic linkage and association analyses on a Finnish migraine patient collection as well as migraineurs from Australia, Denmark, Germany, Iceland and the Netherlands. Altogether we studied genetic information of nearly 7,000 migraine patients and over 50,000 population-matched controls. We also developed a new migraine analysis method called the trait component analysis, which is based on individual patient responses instead of the clinical diagnosis. Using this method, we detected a number of new genetic loci for migraine, including on chromosome 17p13 (HLOD 4.65) and 10q22-q23 (female-specific HLOD 7.68) with significant evidence of linkage, along with five other loci (2p12, 8q12, 4q28-q31, 18q12-q22, and Xp22) detected with suggestive evidence of linkage. The 10q22-q23 locus was the first genetic finding in migraine to show linkage to the same locus and markers in multiple populations, with consistent detection in six different scans. Traditionally, ion channels have been thought to play a role in migraine susceptibility, but we were able to exclude any significant role for common variants in a candidate gene study of 155 ion transport genes. This was followed up by the first genome-wide association study in migraine, conducted on 2,748 migraine patients and 10,747 matched controls followed by a replication in 3,209 patients and 40,062 controls. In this study, we found interesting results with genome-wide significance, providing targets for future genetic and functional studies. Overall, we found several promising genetic loci for migraine providing a promising base for future studies in migraine.
