Ser/Arg-rich proteins (SR proteins) are essential splicing factors that commit pre-messenger RNAs to splicing and also modulate 5' splice site choice in the presence or absence of functional U1 small nuclear ribonucleoproteins (snRNPs). Here, we perturbed the U1 snRNP in HeLa cell nuclear extract by detaching the U1-specific A protein using a 2'-O-methyl oligonucleotide (L2) complementary to its binding site in U1 RNA. In this extract, the standard adenovirus substrate is spliced normally, but excess amounts of SR proteins do not exclusively switch splicing from the normal 5' splice site to a proximal site (site 125 within the adenovirus intron), suggesting that modulation of 5' splice site choice exerted by SR proteins requires integrity of the U1 snRNP. The observation that splicing does not necessarily follow U1 binding indicates that interactions between the U1 snRNP and components assembled on the 3' splice site via SR proteins may also be critical for 5' splice site selection. Accordingly, we found that SR proteins promote the binding of the U2 snRNP to the branch site and stabilize the complex formed on a 3'-half substrate in the presence or absence of functional U1 snRNPs. A novel U2/U6/3'-half substrate crosslink was also detected and promoted by SR proteins. Our results suggest that SR proteins in collaboration with the U1 snRNP function in two distinct steps to modulate 5' splice site selection.

A dedicated mission to investigate exoplanetary atmospheres represents a major milestone in our quest to understand our place in the universe by placing our Solar System in context and by addressing the suitability of planets for the presence of life. EChO—the Exoplanet Characterisation Observatory—is a mission concept specifically geared for this purpose. EChO will provide simultaneous, multi-wavelength spectroscopic observations on a stable platform that will allow very long exposures. The use of passive cooling, few moving parts and well established technology gives a low-risk and potentially long-lived mission. EChO will build on observations by Hubble, Spitzer and ground-based telescopes, which discovered the first molecules and atoms in exoplanetary atmospheres. However, EChO’s configuration and specifications are designed to study a number of systems in a consistent manner that will eliminate the ambiguities affecting prior observations. EChO will simultaneously observe a broad enough spectral region—from the visible to the mid-infrared—to constrain from one single spectrum the temperature structure of the atmosphere, the abundances of the major carbon and oxygen bearing species, the expected photochemically-produced species and magnetospheric signatures. The spectral range and resolution are tailored to separate bands belonging to up to 30 molecules and retrieve the composition and temperature structure of planetary atmospheres. The target list for EChO includes planets ranging from Jupiter-sized with equilibrium temperatures T_ eq up to 2,000 K, to those of a few Earth masses, with T _eq \u223c 300 K. The list will include planets with no Solar System analog, such as the recently discovered planets GJ1214b, whose density lies between that of terrestrial and gaseous planets, or the rocky-iron planet 55 Cnc e, with day-side temperature close to 3,000 K. As the number of detected exoplanets is growing rapidly each year, and the mass and radius of those detected steadily decreases, the target list will be constantly adjusted to include the most interesting systems. We have baselined a dispersive spectrograph design covering continuously the 0.4–16 μm spectral range in 6 channels (1 in the visible, 5 in the InfraRed), which allows the spectral resolution to be adapted from several tens to several hundreds, depending on the target brightness. The instrument will be mounted behind a 1.5 m class telescope, passively cooled to 50 K, with the instrument structure and optics passively cooled to \u223c45 K. EChO will be placed in a grand halo orbit around L2. This orbit, in combination with an optimised thermal shield design, provides a highly stable thermal environment and a high degree of visibility of the sky to observe repeatedly several tens of targets over the year. Both the baseline and alternative designs have been evaluated and no critical items with Technology Readiness Level (TRL) less than 4–5 have been identified. We have also undertaken a first-order cost and development plan analysis and find that EChO is easily compatible with the ESA M-class mission framework.

No país existem inúmeras estruturas e obras civis que estão em operação a dezenas de anos e necessitam de monitoramento periódico devido a sua importância. Por este motivo, a dissertação apresenta um caso de um túnel antigo com problema de queda de bloco e visa instigar novas pesquisas e aumentar o conhecimento sobre o tema. Foram realizadas inspeções em campo em alguns túneis não revestidos da Estrada de Ferro Vitória-Minas (EFVM), bem como os ensaios em laboratório e in situ realizados nas amostras e no maciço rochoso para caracterizar o problema. Para o estudo foi escolhido o túnel Monte Seco Linha 1 e Linha 2 nos quais foram realizadas sondagens rotativas inclinadas e orientadas próximas ao eixo para investigação dos planos de descontinuidade. Os conceitos da Teoria dos Blocos-Chave foram aplicados às famílias de descontinuidades encontradas nos Túneis Monte Seco L1 e L2 para identificar os possíveis blocos instáveis formados pelas escavações. Para obtenção dos parâmetros geotécnicos de resistência e deformabilidade foram realizados ensaios de compressão uniaxial instrumentados com strain gages. A resistência a tração foi obtida através de Ensaio de Compressão Diametral (ECD). No ensaio de campo foi utilizado o Martelo de Schmidt para avaliação da rocha in situ. Através da análise dos dados foi possível distinguir setores cuja ocorrência de queda de blocos são maiores e a classe do maciço rochoso de acordo com a proposta de Bieniawski.

La polisemia es un fenómeno poco estudiado debido, aparentemente, a su dificultad. Es precisamente esa dificultad la causa de la problemática de la polisemia en la adquisición y en el acceso léxico en la enseñanza de una LE/L2. En el presente trabajo se muestra una visión teórica global así como un intento de aproximación experimental para demostrar que existe un dominio deficiente de la polisemia en alumnos de ELE con nivel avanzado y que probablemente se deba a los manuales y al pobre registro polisémico que poseen los profesores de ELE. Para ello se describe la complejidad de la polisemia y, mediante unas encuestas realizadas a un grupo de alumnos de ELE y a un grupo de profesores de ELE, se presenta el análisis de errores del dominio de la polisemia en ambos grupos

Modern compilers present a great and ever increasing number of options which can modify the features and behavior of a compiled program. Many of these options are often wasted due to the required comprehensive knowledge about both the underlying architecture and the internal processes of the compiler. In this context, it is usual, not having a single design goal but a more complex set of objectives. In addition, the dependencies between different goals are difficult to be a priori inferred. This paper proposes a strategy for tuning the compilation of any given application. This is accomplished by using an automatic variation of the compilation options by means of multi-objective optimization and evolutionary computation commanded by the NSGA-II algorithm. This allows finding compilation options that simultaneously optimize different objectives. The advantages of our proposal are illustrated by means of a case study based on the well-known Apache web server. Our strategy has demonstrated an ability to find improvements up to 7.5% and up to 27% in context switches and L2 cache misses, respectively, and also discovers the most important bottlenecks involved in the application performance.

Although it is known that the Spanish current Educative System promotes using the Communicate Approach to teach foreign languages in schools, other recently designed approaches are also used to help students improve their skills when communicating in a foreign language. One of these approaches is Content and Language Integrated Learning, also known as CLIL, which is used to teach content courses using the English language as the language of instruction. This approach improves the students’ skills in English as the same time as they learn content from other areas. The goal of this thesis is to present a research project carried out at the University of Alicante during the academic year 2011-2012. With this research we obtained results that provide quantitative and qualitative data which explains how the use of the CLIL methodology affects the English level of students in the “Didactics of the English Language in Preschool Education” course in Preschool Education Teacher Undergraduate Program as students acquire the contents of the course.

Although frequently discarded and despised in the 20th century, translation now seems to find wider acceptance within the Second Language Teaching (SLT) field. However, it still has a long way to go before recovering its due place in the L2 classroom. The aim of this paper is to suggest a number of translation (and interpreting)-based activities covering the different competence levels, thus showing that communicative content and translation can perfectly go hand in hand so that old, unjustified prejudices can be superseded once and for all.

Con el fin de mejorar la metodología en la enseñanza de español como lengua extranjera a hablantes de polaco como L1, el presente estudio pretende analizar las complicaciones que la noción de aspectualidad puede desencadenar a la hora de adquirir el español como L2. De la misma manera, y puesto que el aspecto se manifiesta de forma diferente en los sistemas verbales de las lenguas eslavas y romances y, por consiguiente, crea un esquema de división temporal dispar, es también objeto de estudio la problemática que el sistema verbal polaco puede presentar al traductor en el momento de trasladar el contenido aspectual y temporal de las estructuras verbales a la lengua meta, en este caso, el español.

In un paese come l’Italia sempre più investito dal fenomeno del multiculturalismo come conseguenza dell’immigrazione, la lingua è elemento di scambio culturale e linguistico, determinante nella negoziazione delle identità diverse che si incontrano. L’apprendimento e, di conseguenza, la conoscenza della lingua del paese ospitante, costituiscono per l’immigrato il primo fattore di integrazione nonché vettore della propria cultura e dei propri valori, difficilmente trasmissibili ai parlanti nativi nella lingua d’origine. Essendo dunque la questione della lingua per gli immigrati, una prospettiva di indagine di estrema importanza e attualità, ho deciso di farne l’oggetto della mia tesi di fine percorso di studi universitario. Partirò con un’analisi della nascita dell’italiano come lingua di studio e dunque come lingua seconda, sino ad arrivare ai risultati ottenuti dalla linguistica e dalla glottodidattica. Analizzerò quelli che erano i destinatari e i docenti dell’italiano L2 di un tempo, e quelli che invece sono oggi i principali protagonisti dello studio della nostra lingua e i nuovi insegnanti, mettendo in luce le diversità nei metodi e negli scopi. Presenterò i fenomeni e i fattori coinvolti nell’italiano L2 degli immigrati e quelli derivanti dall’incontro delle lingue d’origine di quest’ultimi con la lingua italiana. Non mancherà infatti la riflessione sugli altri attori in gioco, noi italiani, in quanto società ospitante, che nella nostra messa in scena quotidiana, attraverso i comportamenti linguistici, esterniamo il nostro sentimento nei riguardi degli immigrati, sia esso di inclusione o di esclusione. Infine farò un excursus sulla letteratura dell’immigrazione, la quale rappresenta la concretizzazione di quanto verrà detto sulla questione della lingua, analizzando infatti attraverso le varie fasi storiche, proprio le diverse visioni degli scrittori e quindi dei protagonisti delle loro opere in merito alla propria identità linguistica.

Reading strategies vary across languages according to orthographic depth - the complexity of the grapheme in relation to phoneme conversion rules - notably at the level of eye movement patterns. We recently demonstrated that a group of early bilinguals, who learned both languages equally under the age of seven, presented a first fixation location (FFL) closer to the beginning of words when reading in German as compared with French. Since German is known to be orthographically more transparent than French, this suggested that different strategies were being engaged depending on the orthographic depth of the used language. Opaque languages induce a global reading strategy, and transparent languages force a local/serial strategy. Thus, pseudo-words were processed using a local strategy in both languages, suggesting that the link between word forms and their lexical representation may also play a role in selecting a specific strategy. In order to test whether corresponding effects appear in late bilinguals with low proficiency in their second language (L2), we present a new study in which we recorded eye movements while two groups of late German-French and French-German bilinguals read aloud isolated French and German words and pseudo-words. Since, a transparent reading strategy is local and serial, with a high number of fixations per stimuli, and the level of the bilingual participants' L2 is low, the impact of language opacity should be observed in L1. We therefore predicted a global reading strategy if the bilinguals' L1 was French (FFL close to the middle of the stimuli with fewer fixations per stimuli) and a local and serial reading strategy if it was German. Thus, the L2 of each group, as well as pseudo-words, should also require a local and serial reading strategy. Our results confirmed these hypotheses, suggesting that global word processing is only achieved by bilinguals with an opaque L1 when reading in an opaque language; the low level in the L2 gives way to a local and serial reading strategy. These findings stress the fact that reading behavior is influenced not only by the linguistic mode but also by top-down factors, such as readers' proficiency.

The development of the ecosystem approach and models for the management of ocean marine resources requires easy access to standard validated datasets of historical catch data for the main exploited species. They are used to measure the impact of biomass removal by fisheries and to evaluate the models skills, while the use of standard dataset facilitates models inter-comparison. North Atlantic albacore tuna is exploited all year round by longline and in summer and autumn by surface fisheries and fishery statistics compiled by the International Commission for the Conservation of Atlantic Tunas (ICCAT). Catch and effort with geographical coordinates at monthly spatial resolution of 1° or 5° squares were extracted for this species with a careful definition of fisheries and data screening. In total, thirteen fisheries were defined for the period 1956-2010, with fishing gears longline, troll, mid-water trawl and bait fishing. However, the spatialized catch effort data available in ICCAT database represent a fraction of the entire total catch. Length frequencies of catch were also extracted according to the definition of fisheries above for the period 1956-2010 with a quarterly temporal resolution and spatial resolutions varying from 1°x 1° to 10°x 20°. The resolution used to measure the fish also varies with size-bins of 1, 2 or 5 cm (Fork Length). The screening of data allowed detecting inconsistencies with a relatively large number of samples larger than 150 cm while all studies on the growth of albacore suggest that fish rarely grow up over 130 cm. Therefore, a threshold value of 130 cm has been arbitrarily fixed and all length frequency data above this value removed from the original data set.

Hydrogenetic ferromanganese crusts were dredged from four seamounts in the western Pacific, OSM7, OSM2, Lomilik, and Lemkein, aligned in a NW-SE direction parallel to Pacific Plate movement. The crusts consist of four well-defined layers with distinct textural and geochemical properties. The topmost layer 1 is relatively enriched in Mn, Co, Ni, and Mo compared to the underlying layer 2, which is relatively enriched in Al, Ti, K, and Rb and Cu, Zn, and excess Ba. Textural and geochemical properties of layer 2 suggest growth conditions under high biogenic and detrital flux. Such conditions are met in the equatorial Pacific (i.e., between the Intertropical Convergence Zone (ITCZ) and equatorial high-productivity zone). Layer 2 likely formed when each seamount was beneath the equatorial Pacific along its back track path. On the other hand, layer 1 probably started to grow after seamounts moved northwest from the ITCZ. This interpretation is consistent with the thickness of layer 1 across the four crusts, which increases to the northwest. Ages of the layer 1-layer 2 boundary in each crust, a potential proxy for northern margin of the ITCZ, also increase to the northwest at 17, 11, 8, and 5 Ma for OSM7, OSM2, Lomilik, and Lemkein, respectively. Assuming Pacific Plate motion of 0.3°/Myr, the seamounts were located at 12°N, 11°N, 9°N, and 8°N at the time of boundary formation. This result suggests that the north edge of the ITCZ has shifted south since the middle Miocene in the western Pacific, which agrees with information from the eastern Pacific.
