The lyotropic liquid crystalline quaternary mixture made of potassium laurate (KL), potassium sulphate, 1-undecanol and water was investigated by experimental optical methods (optical microscopy and laser conoscopy). In a particular temperature and relative concentrations range, the three nematic phases (two uniaxial and one biaxial) were identified. The biaxial domain in the temperature/KL concentration surface is larger when compared to other lyotropic mixtures. Moreover, this new mixture gives nematic phases with higher birefringence than similar systems. The behavior of the symmetric tensor order parameter invariants sigma(3) and sigma(2) calculated from the measured optical birefringences supports that the uniaxial-to-biaxial transitions are of second order, described by a mean-field theory.

Lyotropic liquid crystalline quaternary mixtures of potassium laurate (KL), potassium sulphate (K2SO4)/alcohol (n-OH)/water, with the alcohols having different numbers of carbon atoms in the alkyl chain (n), from 1-octanol to 1-hexadecanol, were investigated by optical techniques (optical microscopy and laser conoscopy). The biaxial nematic phase domain is present in a window of values of n = n(KL) +/- 2, where n(KL) = 11 is the number of carbon atoms in the alkyl chain of KL. The biaxial phase domain became smaller and the uniaxial-to-biaxial phase transition temperatures shifted to relatively higher temperatures upon going from 1-nonanol to 1-tridecanol. Moreover, compared with other lyotropic mixtures these new mixtures present high birefringence values, which we expect to be related to the micellar shape anisotropy. Our results are interpreted assuming that alcohol molecules tend to segregate in the micelles in a way that depends on the relative value of n with respect to nKL. The larger the value of n, the more alcohol molecules tend to be located in the curved parts of the micelle, favoring the uniaxial nematic calamitic phase with respect to the biaxial and uniaxial discotic nematic phases.

The low efficiency of gene transfer is a recurrent problem in DNA vaccine development and gene therapy studies using non-viral vectors such as plasmid DNA (pDNA). This is mainly due to the fact that during their traffic to the target cell's nuclei, plasmid vectors must overcome a series of physical, enzymatic and diffusional barriers. The main objective of this work is the development of recombinant proteins specifically designed for pDNA delivery, which take advantage of molecular motors like dynein, for the transport of cargos from the periphery to the centrosome of mammalian cells. A DNA binding sequence was fused to the N-terminus of the recombinant human dynein light chain LC8. Expression studies indicated that the fusion protein was correctly expressed in soluble form using E. coli BL21(DE3) strain. As expected, gel permeation assays found the purified protein mainly present as dimers, the functional oligomeric state of LC8. Gel retardation assays and atomic force microscopy proved the ability of the fusion protein to interact and condense pDNA. Zeta potential measurements indicated that LC8 with DNA binding domain (LD4) has an enhanced capacity to interact and condense pDNA, generating positively charged complexes. Transfection of cultured HeLa cells confirmed the ability of the LD4 to facilitate pDNA uptake and indicate the involvement of the retrograde transport in the intracellular trafficking of pDNA: LD4 complexes. Finally, cytotoxicity studies demonstrated a very low toxicity of the fusion protein vector, indicating the potential for in vivo applications. The study presented here is part of an effort to develop new modular shuttle proteins able to take advantage of strategies used by viruses to infect mammalian cells, aiming to provide new tools for gene therapy and DNA vaccination studies. A hyaluronidase (CdtHya1) from Crotalus durissus terrificus snake venom (CdtV) was isolated and showed to exhibit a high activity on hyaluronan cleavage. However, surveys on this enzyme are still limited. This study aimed at its isolation, functional/structural characterization and the evaluation of its effect on the spreading of crotoxin and phospholipase A(2) (PLA(2)). The enzyme was purified through cation exchange, gel filtration and hydrophobic chromatography. After that, it was submitted to a reverse-phase fast protein liquid chromatography (RP-FPLC) and Edman degradation sequencing, which showed the first N-terminal 44 amino acid residues whose sequence evidenced identity with other snake venom hyaluronidases. CdtHya1 is a monomeric glycoprotein of 64.5 kDa estimated by SDS-PAGE under reducing conditions. It exhibited maximum activity in the presence of 0.2 M NaCl, at 37 degrees C, pH 5.5 and a specificity to hyaluronan higher than that to chondroitin-4-sulphate, chondroitin-6-sulphate or dermatan. Divalent cations (Ca2+ and Mg2+) and 1 M NaCl significantly reduced the enzyme activity. The specific activity of CdtHya1 was 5066 turbidity reducing units (TRU)/mg, against 145 TRU/mg for the soluble venom, representing a 34.9-fold purification. The pure enzyme increased the diffusion of crotoxin and PLA (2) through mice tissues. CdtHya1 (32 TRU/40 mu L) potentiated crotoxin action, as evidenced by mice death, and it decreased the oedema caused by subplantar injections of buffer, crotoxin or PLA(2), thus evidencing the relevance of hyaluronidase in the crotalic envenoming. Sulphur plays an essential role in plants and is one of the main nutrients in several metabolic processes. It has four stable isotopes (32S, 33S, 34S, and 36S) with a natural abundance of 95.00, 0.76, 4.22, and 0.014 in atom %, respectively. A method for isotopic determination of S by isotope-ratio mass spectrometry (IRMS) in soil samples is proposed. The procedure involves the oxidation of organic S to sulphate (S-SO4(2-)), which was determined by dry combustion with alkaline oxidizing agents. The total S-SO4(2-) concentration was determined by turbidimetry and the results showed that the conversion process was adequate. To produce gaseous SO2 gas, BaSO4 was thermally decomposed in a vacuum system at 900 ºC in the presence of NaPO3. The isotope determination of S (atom % 34S atoms) was carried out by isotope ratio mass spectrometry (IRMS). In this work, the labeled material (K2(34)SO4) was used to validate the method of isotopic determination of S; the results were precise and accurate, showing the viability of the proposed method.

A survey of the experiences of owners of diabetic dogs was carried out by distributing questionnaires to veterinary clinics in São Paulo, Brazil. A total of 93 dog owners were surveyed over a 4 month period and descriptive statistical analysis, frequency distribution and correlations were performed. In most cases the clinical condition of the dogs had improved following treatment and owners were largely satisfied with veterinary intervention. Injection of neutral protamine hagedorm twice daily was the most commonly prescribed treatment and respondents found this procedure easy to perform, although it did pose some difficulties when the owners were away from their animals. Cost was considered an important factor at the initiation of treatment programs. The information gleaned from this survey, in providing useful insights into the attitudes and experiences of owners of diabetic animals, will contribute to improvements in the treatment and management of such patients.

Polycyclic aromatic hydrocarbons are chemicals produced by both human activities and natural sources and they have been present in the biosphere since millions of years. For this reason microorganisms should have developed, during the world history, the capacity of metabolized them under different electron acceptors and redox conditions. The deep understanding of these natural attenuation processes and of microbial degradation pathways has a main importance in the cleanup of contaminated areas. Anaerobic degradation of aromatic hydrocarbons is often presumed to be slow and of a minor ecological significance compared with the aerobic processes; however anaerobic bioremediation may play a key role in the transformation of organic pollutants when oxygen demand exceeds supply in natural environments. Under such conditions, anoxic and anaerobic degradation mediated by denitrifying or sulphate-reducing bacteria can become a key pathway for the contaminated lands clean up. Actually not much is known about anaerobic bioremediation processes. Anaerobic biodegrading techniques may be really interesting for the future, because they give the possibility of treating contaminated soil directly in their natural status, decreasing the costs concerning the oxygen supply, which usually are the highest ones, and about soil excavations and transports in appropriate sites for a further disposal. The aim of this dissertation work is to characterize the conditions favouring the anaerobic degradation of polycyclic aromatic hydrocarbons. Special focus will be given to the assessment of the various AEA efficiency, the characterization of degradation performance and rates under different redox conditions as well as toxicity monitoring. A comparison with aerobic and anaerobic degradation concerning the same contaminated material is also made to estimate the different biodegradation times.

In ihrer dualen Funktion als Monophenolhydroxylase (EC 1.14.18.1) und Diphenoloxidase (EC 1.10.3.1) ist die Tyrosinase das Schlüsselenzym der Melanogenese, der Synthese des Melanins, und übernimmt damit quer durch alle Organismenreiche Aufgaben von der Pigmentierung bis hin zu einer Beteiligung an der Immunantwort. Sie zählt, zusammen mit den Catecholoxidasen und Hämocyaninen, zu den Typ-3-Kupfer-Proteinen, die sich durch ein Aktives Zentrum auszeichnen, das in der Lage ist, Sauerstoff und phenolische Substrate reversibel zwischen zwei Kupfer-Ionen zu binden. Bisher konnte weder die Funktion der pflanzlichen Tyrosinase genau identifiziert, noch die Struktur eines solchen Enzyms aufgeklärt werden. Mit dem späteren Ziel, durch eine röntgenkristallographische Analyse die zugrunde liegende strukturelle Ursache der zusätzlichen Monophenolhydroxylase-Aktivität von Tyrosinasen gegenüber reinen Catecholoxidasen ermitteln zu können, wurde in dieser Arbeit ein bakterielles Expressionssystem entwickelt, das zur Herstellung einer rekombinanten Tyrosinase oder Polyphenoloxidase (PPO) aus Spinacia oleracea (Spinat) für die Kristallisation verwendet werden kann. Das rekombinante Protein wurde in Form von Inclusion Bodies isoliert, anhand einer Affinitätschromatographie aufgereinigt und in anschließende Rückfaltungsexperimente eingesetzt. In einer parallelen Versuchsreihe konnte Spinat, aufgrund seiner hohen Tyrosinaseaktivität, als geeignetes Objekt für die Isolation des nativen Enzyms identifiziert werden. Im Anschluss an eine Thylakoidpräparation, Solubilisierung der Thylakoidmembranen und Fällung des Proteins mit Ammoniumsulfat, wurden Experimente zur weiteren Anreicherung der Tyrosinase-Aktivität über eine Anionenaustausch-Chromatographie und zur Etablierung einiger nachfolgender Aufreinigungsschritte durchgeführt.

Membrane lipid rafts are detergent-resistant microdomains containing glycosphingolipids, cholesterol and glycosylphosphatidylinositol-linked proteins; they seem to be actively involved in many cellular processes including signal transduction, apoptosis, cell adhesion and migration. Lipid rafts may represent important functional platforms where redox signals are produced and transmitted in response to various agonists or stimuli. In addition, a new concept is emerging that could be used to define the interactions or amplification of both redox signalling and lipid raft-associated signalling. This concept is characterized by redox-mediated feed forward amplification in lipid platforms. It is proposed that lipid rafts are formed in response to various stimuli; for instance, NAD(P)H oxidase (Nox) subunits are aggregated or recruited in these platforms, increasing Nox activity. Superoxide and hydrogen peroxide generation could induce various regulatory activities, such as the induction of glucose transport activity and proliferation in leukaemia cells. The aim of our study is to probe: i) the involvement of lipid rafts in the modulation of the glucose transporter Glut1 in human acute leukemia cells; ii) the involvement of plasma membrane caveolae/lipid rafts in VEGF-mediated redox signaling via Nox activation in human leukemic cells; iii) the role of p66shc, an adaptor protein, in VEGF signaling and ROS production in endothelial cells (ECs); iv) the role of Sindecan-2, a transmembrane heparan sulphate proteoglycan, in VEGF signaling and physiological response in ECs and v) the antioxidant and pro-apoptotic activities of simple dietary phenolic acids, i. e. caffeic, syringic and protocatechuic acids in leukemia cells, characterized by a very high ROS content. Our results suggest that the role played by NAD(P)H oxidase-derived ROS in the regulation of glucose uptake, proliferation and migration of leukaemia and endothelial cells could likely occur through the control of lipid raft-associated signalling.

Desmosomen sind hoch organisierte adhäsive interzelluläre Verbindungen, die benachbarte Zellen durch Verankerung mit den Intermediärfilamenten des Zytoskeletts miteinander verknüpfen und so Zellen und Geweben Stabilität verleihen. Die Adhäsionsmoleküle der Desmosomen sind die desmosomalen Cadherine. Diese transmembranen Glykoproteine gehen im Interzellulärraum Verbindungen mit den desmosomalen Cadherinen der Nachbarzelle ein und sind im zytoplasmatischen Bereich Anheftungspunkte für weitere an der Desmosomenbildung beteiligte Proteine. Ziel dieser Arbeit war die Untersuchung der Rolle von Desmoglein 2 (Dsg 2), einem in allen Epithelien exprimierten desmosomalen Cadherin. Da der konstitutive knock out von Dsg 2 embryonal letal ist, wurde im Rahmen dieser Doktorarbeit eine transgene Maus generiert, in der die Reduktion von Dsg 2 temporär regulierbar war (konditionaler knock down). Dazu wurde der Mechanismus der RNA Interferenz genutzt, wodurch Sequenz-spezifische, post-transkriptionelle Regulation von Genen möglich ist. Unter Verwendung eines über Cre/lox-induzierbaren Vektors wurden transgene Mäuse generiert, welche nach Induktion Dsg 2 shRNA exprimieren, die in der Zelle in siRNA umgewandelt wird und zum Abbau der Dsg 2 mRNA führt. Durch Verpaarung der generierten Dsg 2 knock down Maus mit der über Tamoxifen induzierbaren Cre Deleter knock in Mauslinie Rosa26CreERT2 konnte deutliche Reduktion der Dsg 2 Proteinmenge in Leber, Darm und Herz erreicht werden. In Immunfärbungen der Leber wurde zudem eine reduzierte Desmosomenbildung durch Expression der Dsg 2 shRNA detektiert. Die für diese Versuche generierte und getestete Rosa26CreERT2 Mauslinie ermöglichte jedoch nicht in allen Zellen eines Gewebes die komplette Aktivierung der Cre Rekombinase und damit die Expression der shRNA. Dadurch entstanden mosaikartige Wildtyp/knock down-Gewebe, in denen noch ausreichend Desmosomen gebildet wurden, um die Gewebestabilität und -struktur zu erhalten. Für eine funktionale Untersuchung von Dsg 2 in Zusammenhang mit der chronisch entzündlichen Darmerkrankung Colitis ulcerosa wurden die Dsg 2 knock down Mäuse mit Darm-spezifischen, induzierbaren Cre Deleter Mäusen (VillinCreERT2) verpaart. Nach Aktivierung der Cre Rekombinase mittels Tamoxifen wurde in bitransgenen Tieren über Gabe von Azoxymethan (AOM) und Dextransodiumsulfat (DSS) Colitis ulcerosa induziert. Diese entzündliche Erkrankung des Darms ist mit der Induktion von Darmtumoren assoziiert. Bereits nach einmaliger Induktion mit AOM/DSS wurde in der ersten endoskopischen Untersuchung eine starke Entzündung des Darmgewebes und die Ausbildung von flächig wachsenden Tumoren in den Dsg 2 knock down Tieren hervorgerufen. Es ist anzunehmen, dass durch knock down von Dsg 2, und die damit verbundene verminderte Desmosomenbildung und Zelladhäsion, Infiltration von Bakterien durch die epitheliale Barriere des Darms möglich war, und so die Entzündungsreaktion in der Darmmukosa verstärkte. In Zusammenhang mit Verlust der epithelialen Festigkeit durch verringerte Zellkontakte kam es zur Hyperproliferation der Darmmukosa, die sich in Ausbildung von flächigen Tumoren äußerte. In weiteren Experimenten müssen nun die Tumore und das entzündete Gewebe der Colitis-induzierten Mäuse mittels Immunfluoreszenz untersucht werden, um Veränderungen in der Desmosomenformation in situ detektieren zu können. Des Weiteren sind Verpaarungen der Dsg 2 knock down Maus mit anderen Cre Rekombinase exprimierenden Mauslinien möglich, um den Einfluss von Dsg 2 auch in anderen Geweben, beispielsweise im Herzen, zu untersuchen. Die hier vorgelegte Arbeit zeigt also erstmalig den ursächlichen Zusammenhang zwischen Dsg 2 und dem Auftreten von Colitis-assoziierten Tumoren in einem konditionalen RNAi-vermittelten knock down Tiermodell. Die Etablierung dieser Maus ist somit das erste konditionale Mausmodell, welches die bei vielen Krebspatienten gefundenen flachzellig wachsenden Tumore in vivo rekapituliert. Vorausschauend kann man sagen, dass mit Hilfe des im Rahmen dieser Doktorarbeit entwickelten Tiermodells wichtige Erkenntnisses über die Pathologie von Darmtumoren erbracht werden können, die unser Verständnis der Colitis-induzierten Tumorentstehung verbessern.

Throughout the world, pressures on water resources are increasing, mainly as a result of human activity. Because of their accessibility, groundwater and surface water are the most used reservoirs. The evaluation of the water quality requires the identification of the interconnections among the water reservoirs, natural landscape features, human activities and aquatic health. This study focuses on the estimation of the water pollution linked to two different environmental issues: salt water intrusion and acid mine drainage related to the exploitation of natural resources. Effects of salt water intrusion occurring in the shallow aquifer north of Ravenna (Italy) was analysed through the study of ion- exchange occurring in the area and its variance throughout the year, applying a depth-specific sampling method. In the study area were identified ion exchange, calcite and dolomite precipitation, and gypsum dissolution and sulphate reduction as the main processes controlling the groundwater composition. High concentrations of arsenic detected only at specific depth indicate its connexion with the organic matter. Acid mine drainage effects related to the tin extraction in the Bolivian Altiplano was studied, on water and sediment matrix. Water contamination results strictly dependent on the seasonal variation, on pH and redox conditions. During the dry season the strong evaporation and scarce water flow lead to low pH values, high concentrations of heavy metals in surface waters and precipitation of secondary minerals along the river, which could be released in oxidizing conditions as demonstrated through the sequential extraction analysis. The increase of the water flow during the wet season lead to an increase of pH values and a decrease in heavy metal concentrations, due to dilution effect and, as e.g. for the iron, to precipitation.

Untersucht wird die Proteinzusammensetzung verschiedener Weinsorten der Anbaugebiete Rheinhessen, Rheingau und Pfalz. Es erfolgt erstmalig die Identifizierung der Proteine eines Rotweins (Portugieser 2005 aus der Pfalz). Hierzu werden die Proteine mittels Dialyse und Gefriertrocknung konzentriert, auf einer SDS-PAGE aufgetrennt und mittels ESI-Q-TOF-Massenspektrometrie identifiziert. Von den identifizierten Proteinen des Rotweins stammen zwölf aus der Weinbeere und sechs werden im Laufe der Weinbereitung durch die Hefe ein-gebracht. Der Großteil der über die Weinbeere in den Wein gelangten Proteine ist der Gruppe der Pathogenese bezogenen Proteine zuzuordnen. Ein Vergleich der Proteinzusammensetzung verschiedener Rotweine, Weißweine und Rosé-weine zeigt, dass ein gemeinsames Proteinspektrum in allen Weinsorten enthalten ist, es je-doch auch Unterschiede hinsichtlich der Proteinzusammensetzung und Konzentration der ein-zelnen Proteine zwischen den Rebsorten, insbesondere roten und weißen, gibt. In Portugieser Rotwein des Jahrgangs 2005 aus der Pfalz sowie in Dornfelder Rotwein der Jahrgänge 2002, 2003, 2004 und 2005 kann das als Allergen beschriebene Lipid Transfer Pro-tein (Isoform 4) nachgewiesen werden. In den untersuchten Weißweinsorten Riesling, Sau-vignon Blanc, Morio Muskat sowie Gewürztraminer ist dieses nicht bzw. nur in sehr geringen Mengen enthalten. Ebenfalls nur in Rotwein wird eine Klasse IV Endochitinase identifiziert, die als Allergen in jungem Rotwein bereits beschrieben wurde. Außerdem bedingen Chitin-asen zusammen mit den Thaumatin-ähnlichen Proteinen die Proteininstabilität. Thaumatin-ähnliche Proteine stellen in allen Weinsorten den größten Anteil der Proteine dar. Viele der Weinproteine liegen in glykosylierter Form vor, was durch die Perjodsäurefärbung und den Lektinblot gezeigt werden kann. Mit der Detektion von Thaumatin-ähnlichen Proteinen, Lipid Transfer Proteinen sowie einer Endochitinase werden potentielle Allergene im Wein nachgewiesen sowie strukturell mit be-kannten Allergenen verglichen. Die Kenntnis der Proteinzusammensetzung im Wein bildet die Grundlage für weiterführende Untersuchungen zur Weinstabilität und zur Allergenität von Weinproteinen.

This thesis reports an integrated analytical approach for the study of physicochemical and biological properties of new synthetic bile acid (BA) analogues agonists of FXR and TGR5 receptors. Structure-activity data were compared with those previous obtained using the same experimental protocols on synthetic and natural occurring BA. The new synthetic BA analogues are classified in different groups according also to their potency as a FXR and TGR5 agonists: unconjugated and steroid modified BA and side chain modified BA including taurine or glycine conjugates and pseudo-conjugates (sulphonate and sulphate analogues). In order to investigate the relationship between structure and activity the synthetic analogues where admitted to a physicochemical characterization and to a preliminary screening for their pharmacokinetic and metabolism using a bile fistula rat model. Sensitive and accurate analytical methods have been developed for the quali-quantitative analysis of BA in biological fluids and sample used for physicochemical studies. Combined High Performance Liquid Chromatography Electrospray tandem mass spectrometry with efficient chromatographic separation of all studied BA and their metabolites have been optimized and validated. Analytical strategies for the identification of the BA and their minor metabolites have been developed. Taurine and glycine conjugates were identified in MS/MS by monitoring the specific ion transitions in multiple reaction monitoring (MRM) mode while all other metabolites (sulphate, glucuronic acid, dehydroxylated, decarboxylated or oxo) were monitored in a selected-ion reaction (SIR) mode with a negative ESI interface by the following ions. Accurate and precise data where achieved regarding the main physicochemical properties including solubility, detergency, lipophilicity and albumin binding . These studies have shown that minor structural modification greatly affect the pharmacokinetics and metabolism of the new analogues in respect to the natural BA and on turn their site of action, particularly where their receptor are located in the enterohepatic circulation.

Die pulmonale Applikation von Arzneimitteln hat in den vergangenen Jahren zu- rnnehmend an Bedeutung gewonnen und wird vor allem in der lokalen Therapie bei Asthma, COPD und Mukoviszidose genutzt. Als neues innovatives Inhalationsgerät befindet sich der Respimat® seit 2004 auf dem Arzneimittelmarkt und ist bisher nur für Erwachsene zugelassen. Daher wurden im Rahmen dieser Arbeit die Anwendbarkeit und die Modifikation des Respimats® bei Kindern untersucht. Zusammenfassend zeigen die Ergebnisse, dass Kinder ab einem Alter von fünf Jahren fähig sind suffizient mit dem Respimat® zu inhalieren und gute Werte für die estimierte Intrathorakaldosis zu erreichen. Besonders Kinder von 9-12 Jahren und Patienten mit DPI- oder Autohaler®-Inhalationserfahrung erreichten aufgrund eines hohen Atemflusses eine niedrigere estimierte Intrathorakaldosis als es das Potenzial des Respimats® erlaubt hätte. Durch den Einbau eines höheren internen Widerstands in den Orginalrespimat® wurde der Atemfluss signifikant verlangsamt und die Intrathorakaldosis erhöht. Des Weiteren wurden in der vorliegenden Arbeit die physikalisch-chemische Kompatibilität und die aerodynamischen Eigenschaften von Mischinhalationslösung/-suspensionen untersucht. In der Praxis mischen Muko-viszidose-Patienten oftmals mehrere Fertigarzneimittel und inhalieren diese simultan. Dadurch wird der Zeitaufwand der täglichen Inhalation reduziert und die Compliance sowie die Lebensqualität verbessert. Die 4er Kombination aus Colistimethat mit konservierungsmittelfreier Salbutamolsulfat, Ipratropiumbromidinhalationslösungen und Fluticason-17-propionatinhalationssuspension erwies sich als physikalisch-chemisch kompatibel. Die entwickelte mikrobiologische Wertbestimmungsmethode erwies sich als robust, präzise, gut reproduzierbar und erfüllte alle Anforderungen des statistischen Modells. Weiterhin wurden die aerodynamischen Eigenschaften einer praxisrelevanten 3er Mischinhalationssuspension aus Salbutamolsulfat, Ipratropiumbromid und Fluticason-17-propionat im Vergleich zu deren konsekutiven Vernebelung mittels Kaskadenimpaktion betrachtet. Die Charakteristik des erzeugten Aerosols änderte sich für simultan vernebeltes Salbutamolsulfat und Ipratropiumbromid nicht, wohingegen Fluticason-17-propionat bei simultaner Vernebelung eine Reduktion des MMADs, der delivered dose und der recovery aufwies. Dennoch ist die inhalative Glukokortikoiddosis ausreichend hoch um eine effektive antiinflammatorische Wirkung zu gewährleisten. Daher kann die simultane Inhalation der 3er Kombination empfohlen werden und somit die Compliance und die Lebensqualität von Mukoviszidosepatienten erhöht werden.rn

Air quality represents a key issue in the so-called pollution “hot spots”: environments in which anthropogenic sources are concentrated and dispersion of pollutants is limited. One of these environments, the Po Valley, normally experiences exceedances of PM10 and PM2.5 concentration limits, especially in winter when the ventilation of the lower layers of the atmosphere is reduced. This thesis provides a highlight of the chemical properties of particulate matter and fog droplets in the Po Valley during the cold season, when fog occurrence is very frequent. Fog-particles interactions were investigated with the aim to determine their impact on the regional air quality. Size-segregated aerosol samples were collected in Bologna, urban site, and San Pietro Capofiume (SPC), rural site, during two campaigns (November 2011; February 2013) in the frame of Supersito project. The comparison between particles size-distribution and chemical composition in both sites showed the relevant contribution of the regional background and secondary processes in determining the Po Valley aerosol concentration. Occurrence of fog in November 2011 campaign in SPC allowed to investigate the role of fog formation and fog chemistry in the formation, processing and deposition of PM10. Nucleation scavenging was investigated with relation to the size and the chemical composition of particles. We found that PM1 concentration is reduced up to 60% because of fog scavenging. Furthermore, aqueous-phase secondary aerosol formation mechanisms were investigated through time-resolved measurements. In SPC fog samples have been systematically collected and analysed since the nineties; a 20 years long database has been assembled. This thesis reports for the first time the results of this long time series of measurements, showing a decrease of sulphate and nitrate concentration and an increase of pH that reached values close to neutrality. A detailed discussion about the occurred changes in fog water composition over two decades is presented.
