The cyanobacteria are known to be a rich source of metabolites with a variety of biological activities in different biological systems. In the present work, the bioactivity of aqueous and organic (methanolic and hexane) crude extracts of cyanobacteria isolated from estuarine ecosystems was studied using different bioassays. The assessment of DNA damage on the SOS gene repair region of mutant PQ37 strain of Escherichia coli was performed. Antiviral activity was evaluated against influenza virus, HRV-2, CVB3 and HSV-1 viruses using crystal violet dye uptake on HeLa, MDCK and GMK cell lines. Cytotoxicity evaluation was performed with L929 fibroblasts by MTT assay. Of a total of 18 cyanobacterial isolates studied, only the crude methanolic extract of LEGE 06078 proved to be genotoxic (IF > 1.5) in a dose-dependent manner and other four were putative candidates to induce DNA damage. Furthermore, the crude aqueous extract of LEGE 07085 showed anti- herpes type 1 activity (IC50 = 174.10 μg dry extract mL−1) while not presenting any cytotoxic activity against GMK cell lines. Of the 54 cyanobacterial extracts tested, only the crude methanolic and hexane ones showed impair on metabolic activity of L929 fibroblasts after long exposure (48–72 h). The inhibition of HSV-1 and the strong cytotoxicity against L929 cells observed emphasizes the importance of evaluating the impact of those estuarine cyanobacteria on aquatic ecosystem and on human health. The data also point out their potential application in HSV-1 treatment and pharmacological interest.

RESUMO - Nos últimos vinte anos tem-se assistido a uma crescente consciencialização de que os nossos estilos de vida são insustentáveis aos níveis económico, social e ambiental, o que tem repercussões na nossa saúde e bem-estar. Do crescimento populacional à pobreza e inequidade geradas pelo modelo de “crescimento económico” actual, à perda de biodiversidade e disrupção dos ecossistemas naturais, ao desmesurado crescimento urbano, à poluição e acumulação de desperdícios, às alterações climáticas, ao isolamento individual e à diminuição do capital social na sociedade do consumo: a necessidade de desenvolvimento sustentável e gerador de bem-estar nunca foi tão grande e evidente. Ao longo dos últimos anos têm surgido comunidades intencionais que se organizam segundo princípios de sustentabilidade, como um fenómeno de contra-cultura – as Ecoaldeias (Ecovillages). No entanto, os benefícios para a saúde e bem-estar deste tipo de comunidades não são ainda claros, sendo a experiência de investigação nesta área escassa. O estudo aqui proposto visa conhecer, a título exploratório, os níveis de bem-estar subjectivo em comunidades intencionais que vivem segundo princípios de sustentabilidade em Portugal, se estes níveis são melhores que na população em geral, e quais os factores percebidos que o influenciam. Para tal, terá componentes quantitativas e qualitativas e irá basear-se num questionário auto-administrado aos residentes das Ecoaldeias portuguesas, que inclui o Índice de Bem-estar Pessoal - uma escala de medição do Bem-estar subjectivo validada para a população portuguesa. As suas conclusões poderão contribuir para o desenvolvimento de abordagens mais elaboradas, capazes de edificar uma infra-estrutura teórica para o sistema de conceitos em foco, tão necessária quer a investigações com maior potencial explicativo, quer a decisões com melhor fundamento. ------------ ABSTRACT - Over the past twenty years there has been a growing awareness that the way we live is unsustainable at the economic, social and environmental level, which has impact in our health and wellbeing. From the population growth to poverty and inequity generated by the current model of economic growth, to biodiversity loss and disruption of natural ecosystems, to disproportionate urban growth, to pollution and waste accumulation, to climate change and the individual isolation social loss capital in the consumption society: the need for a development that is sustainable and generates wellbeing has never been greater and more evident. Over the last years intentional communities who live according to principles of sustainability have emerged, has a phenomenon of counter-culture - the ecovillages. The health and wellbeing benefits of this type of communities are not clear, as the investigation in this area is little. The aim of this exploratory study is to know the levels of subjective wellbeing of such communities, in Portugal, if these levels are different from the general population and what are the main perceived contributing factors. This study will have a qualitative and quantitative component and will be based in the application of a self-administered questionnaire that includes the Subjective Wellbeing Index, a measurement scale of subjective wellbeing, validated for the Portuguese population. Its findings may contribute to the development of more elaborate approaches that allow to build a theoretical framework for the system of concepts focused, needed both for further investigations with more explanatory potential, as for more grounded decision-making, to tackle the challenges of sustainable development.

RESUMO - A diabetes mellitus é considerada um flagelo à escala mundial, não apenas por atingir milhões de pessoas, mas também porque se tem verificado um aumento da sua prevalência e, consequente-mente, das suas complicações crónicas, que se traduzem no aumento dos gastos em saúde e, sobretudo, em sofrimento das pessoas com diabetes, seus familiares e comunidade. A prevenção das complicações crónicas da diabetes reside, sobretudo, na adopção de comporta-mentos saudáveis que, no dia-a-dia, se traduzem em competência no autocuidado. Assim, será importante conhecer em que medida o sentido de coerência – característica pessoal entendida como influenciadora das estratégias utilizadas face aos acontecimentos originadores de stress, com repercussões na saúde mental, bem como na saúde física – pode determinar a escolha de comportamentos de saúde e, por tal, possa considerar-se como um aspecto sobre o qual os pro-fissionais de saúde poderão investir em prole do seu fortalecimento e, consequentemente, garantir a facilitação do processo de adopção de comportamentos saudáveis. De acordo com o estudo transversal exploratório retrospectivo proposto ambiciona-se conhecer a relação entre o nível de sentido de coerência e o nível de autocuidado e de controlo metabólico, das pessoas com diabetes de tipo 2. Para tal investigação reconhece-se a pertinência da aplica-ção da escala de avaliação do sentido de coerência – Questionário Orientação para Viver (Nunes, 1999), da escala de avaliação do autocuidado com a diabetes – Escala de Avaliação das Activida-des de Autocuidado com a Diabetes (Bastos, Severo e Lopes, 2007) e do Formulário de Controlo Metabólico na Diabetes (European Society of Cardiology e European Association for the Study of Diabetes cit. por Direcção-Geral da Saúde, 2007), numa amostra constituída por 391 pessoas com diabetes de tipo 2, seleccionadas por amostragem aleatória estratificada proporcional, de entre os utentes inscritos nas 33 instituições de Cuidados de Saúde Primários, do distrito de Viana do Castelo --------------------------- ABSTRACT - Diabetes mellitus is considered a global scourge, not only for reaching millions of people, but also because there has been an increase in its prevalence and, consequently, its chronic complications, which translate into increased spending on health and especially in the suffering of people with diabetes, their families and community. The prevention of chronic complications of diabetes includes adoption healthy behaviors that on the day to day, translate into competence in self-care. Therefore it is important to know how far the sense of coherence – understood as influential personal characteristic of the strategies used by the events originators of stress, affecting the mental health and physical health – may determine the choice of health behaviors and, as such, can be considered as an aspect about which health pro-fessionals will invest in strengthening their offspring and thus ensure the facilitation of the process of adopting healthy behaviors. According to the proposed retrospective exploratory cross-sectional study aims to discover the relationship between the level of sense of coherence and level of self-care and metabolic control on people with type 2 diabetes. For this research recognizes the appropriateness of applying the rating scale of sense of coherence – Questionário Orientação para Viver (Nunes, 1999), the rating scale of self-care in diabetes – Escala de Avaliação das Actividades de Autocuidado com a Di-abetes (Bastos, Severo and Lopes, 2007) and the Formulário de Controlo Metabólico na Diabetes (European Society of Cardiology and European Association for the Study of Diabetes cit. por Di-recção-Geral da Saúde, 2007), a sample of 391 people with type 2 diabetes, selected by propor-tional stratified random sampling from among registered users in 33 institutions of Primary Health Care, on district of Viana do Castelo.

A satisfação das necessidades de apoio dos pais com filhos com incapacidades, cujo nascimento implica alterações no equilíbrio da estrutura familiar, é essencial não só para o bem-estar dos pais, mas para o desenvolvimento dos filhos. Foi sob este argumento que surgiu o interesse pelo estudo das redes de suporte, como estratégia de coping, no apoio às necessidades das famílias. O objetivo deste estudo consistiu em avaliar as necessidades de apoio de 226 pais de crianças, jovens e adultos com incapacidade que participaram no 1º nível das Oficinas de Pais – Grupo de Apoio Emocional (GAE), designadamente: (1) os tipos de apoio identificados pelos pais como alvo de maior necessidade; (2) as redes de suporte; (3) a relação entre as necessidades de apoio e as características dos pais/família e filhos com incapacidades. Pretendíamos também avaliar em que medida a oficina produziu mudanças nas suas necessidades de apoio. Para tal, administrámos, antes e após o GAE, a Escala de Funções de Apoio e a Escala de Apoio Social, (Dunst, Trivette, & Deal, 1988). Os resultados mostraram que as necessidades parecem relacionar-se com: (i) apoios nos aspetos práticos do quotidiano; (ii) apoio emocional; (iii) apoio dos serviços e dos profissionais de saúde e de educação. É ao nível do apoio emocional que os pais revelam maiores necessidades de ajuda. Os pais de crianças mais novas reportam maior necessidade de ajuda, nos aspetos práticos do quotidiano e dos serviços/profissionais de saúde e de educação. Adicionalmente, o grau de necessidade aumentou no final da formação.

Purpose: This work aims at further developing and testing the psychometric properties of the Cultural Intelligence Scale (Ang & Van Dyne, 2006) in an Erasmus Mundus Students and Alumni Population, including reliability. Design Methodology: The study included 626 participants from 109 different countries that emcompasses 6 continents. Exploratory and Confirmatory Factor Analysis procedures were carried out in order to test the scale in a multicultural scale of Erasmus Mundus Students. Reliability was assessed using Cronbach Alpha. Results: The scale presents excellent psychometric properties with alpha values that range from .84 to .90. Exploratory and Confirmatory Factor Analyses demonstrated that the original model of the scale presents an exceptionally good fit. Limitations: The present study was conducted using a convenience sample and online questionnaires that limit its conclusions when we consider the globality of the Erasmus Mundus Students. Research/Practical Implications: This study presents evidence that Ang and Van Dyne’s scale is an adequate measure instrument to assess intercultural intelligence in a multicultural setting of students and alumni. Originality/Value: Multicultural samples and studies are becoming more and more present and relevant; the study of intercultural competences and habilities is becoming increasingly important, and in this task, solid psychometric instruments are of paramount importance. This study presents evidence that Ang and Van Dyne’s (2006) scale is a fairly recent and parsimonious instrument with excellent psychometric properties properties.

From June to July 1999 an outbreak of acute respiratory illness occurred in the town of Iporanga. Out of a total of 4,837 inhabitants, 324 cases were notified to the Regional Surveillance Service. Influenza virus was isolated from 57.1% of the collected samples and 100% seroconversion to influenza A (H1N1) was obtained in 20 paired sera tested. The isolates were related to the A/Bayern/07/95 strain (H1N1). The percentages of cases notified during the outbreak were 28.4%, 29.0%, 20.7%, 6.2% and 15.7% in the age groups of 0-4, 5-9, 10-14, 15-19 and older than 20 years, respectively. The highest proportion of positives was observed among children younger than 14 years and no cases were notified in people older than 65 years, none of whom had been recently vaccinated against influenza. These findings suggest a significant vaccine protection against A/Bayern/7/95, the H1 component included in the 1997-98 influenza vaccine for elderly people. This viral strain is antigenically and genetically related to A/Beijing/262/95, the H1 component of the 1999 vaccine. Vaccines containing A/Beijing/262/95 (H1N1) stimulated post-immunization hemagglutination inhibition antibodies equivalent in frequency and titre to both A/Beijing/262/95-like and A/Bayern/7/95-like viruses. Thus, this investigation demonstrates the effectiveness of vaccination against influenza virus in the elderly.

Serotype, mating type and ploidy of 84 strains of Cryptococcus neoformans isolated from 61 AIDS and 23 non-AIDS patients admitted in a tertiary teaching hospital in São Paulo, Brazil were examined. Among 61 strains isolated from AIDS patients, 60 strains were var. grubii (serotype A). Only one strain was var. gattii (serotype B). No var. neoformans (serotype D) was found. Among 23 strains isolated from non-AIDS patients, 15 were var. grubii (serotype A) and the remaining 8 were var. gattii, all of which were serotype B. Seventy-three of the 75 serotype A strains were the heterothallic alpha type (MATalpha) and the remaining 2 were untypable (asexual). Most of the MATalpha strains (69/73) were haploid and the remaining 4 strains were diploid. Similarly, both of the 2 asexual strains among the 75 serotype A strains were haploid. There were no alpha-mating type (MATalpha) strains among the 84 isolates. All of the 8 var. gattii strains were serotype B and haploid. Among a total of 84 strains tested, neither serotype AD nor serotype D were found. Neither triploid nor tetraploid were found. These results suggest that the serological, sexual and ploidy characteristics in C. neoformans strains isolated from AIDS patients in São Paulo were rather simple, whereas strains isolated from non-AIDS patients presented serotype A and B with predominance of serotype A.

In 2001, an autochthonous case of dual viremia, resulting from naturally acquired dengue virus DEN-1 and DEN-2 infections was detected during the dengue outbreak that occurred in Barretos, a city with about 105,000 inhabitants in the North region of São Paulo State. Serotype identification was based on virus isolation to C6/36 mosquito cells culture and immunofluorescence assays using type-specific monoclonal antibodies. The double infection was also confirmed by reverse transcriptase polymerase chain reaction (RT-PCR). Comparative analysis of the 240-nucleotide sequences of E/NS1 gene junction region between the genome of DEN-1 and DEN-2 isolates of the corresponding reference Nauru and PR 159S1 strains, respectively, showed some nucleotide differences, mainly silent mutations in the third codon position. Results of maximum likelihood phylogenetic analysis of E/NS1 gene sequences indicated that both genotypes of DEN-1 and DEN-2 viruses recovered from double infection in Barretos belonged to genotypes I and III, respectively.

Strain typing is a critical tool for molecular epidemiological analysis and can provide important information about the spread of dengue viruses. Here, we performed a molecular characterization of DEN-2 viruses isolated in Brazil during 1990-2000 from geographically and temporally distinct areas in order to investigate the genetic distribution of this serotype circulating in the country. Restriction site-specific polymerase chain reaction (RSS)-PCR presented the same pattern for all 52 Brazilian samples, showing the circulation of just one DEN-2 variant. Phylogenetic analysis using progressive pairwise alignments from 240-nucleotide sequences of the E/NS1 junction in 15 isolates showed that they belong to genotype III (Jamaica genotype).

The genomic sequences of the Envelope-Non-Structural protein 1 junction region (E/NS1) of 84 DEN-1 and 22 DEN-2 isolates from Brazil were determined. Most of these strains were isolated in the period from 1995 to 2001 in endemic and regions of recent dengue transmission in São Paulo State. Sequence data for DEN-1 and DEN-2 utilized in phylogenetic and split decomposition analyses also include sequences deposited in GenBank from different regions of Brazil and of the world. Phylogenetic analyses were done using both maximum likelihood and Bayesian approaches. Results for both DEN-1 and DEN-2 data are ambiguous, and support for most tree bipartitions are generally poor, suggesting that E/NS1 region does not contain enough information for recovering phylogenetic relationships among DEN-1 and DEN-2 sequences used in this study. The network graph generated in the split decomposition analysis of DEN-1 does not show evidence of grouping sequences according to country, region and clades. While the network for DEN-2 also shows ambiguities among DEN-2 sequences, it suggests that Brazilian sequences may belong to distinct subtypes of genotype III.

The aim of this study was to evaluate the genotypic resistance profiles of HIV-1 in children failing highly active antiretroviral therapy (HAART). Forty-one children (median age = 67 months) receiving HAART were submitted to genotypic testing when virological failure was detected. cDNA was extracted from PBMCs and amplified by nested PCR for the reverse transcriptase and protease regions of the pol gene. Drug resistance genotypes were determined from DNA sequencing. According to the genotypic analysis, 12/36 (33.3%) and 6/36 (16.6%) children showed resistance and possible resistance, respectively, to ZDV; 5/36 (14%) and 4/36 (11.1%), respectively, showed resistance and possible resistance to ddI; 4/36 (11.1%) showed resistance to 3TC and D4T; and 3/36 (8.3%) showed resistance to Abacavir. A high percentage (54%) of children exhibited mutations conferring resistance to NNRTI class drugs. Respective rates of resistance and possible resistance to PIs were: RTV (12.2%, 7.3%); APV (2.4%, 12.1%); SQV(0%, 12.1%); IDV (14.6%, 4.9%), NFV (22%, 4.9%), LPV/RTV (2.4%, 12.1%). Overall, 37/41 (90%) children exhibited virus with mutations related to drug resistance, while 9% exhibited resistance to all three antiretroviral drug classes.
