ABSTRACT: This thesis report illustrates the applications and potentials of biogenic methane recovery in Nebraska’s agricultural and industrial sectors and as a means for increasing sustainable economic development in the state’s rural communities. As the nation moves toward a new green economy, biogenic methane recovery as a waste management strategy and renewable energy resource presents significant opportunities for Nebraska to be a national and world leader in agricultural and industrial innovation, advanced research and development of renewable energy technology, and generation of new product markets. Nebraska’s agricultural economy provides a distinct advantage to the state for supporting methane recovery operations that provide long-term economic and environmental partnerships among producers, industry, and communities. These opportunities will serve to protect Nebraska’s agricultural producers from volatile energy input markets and as well as creating new markets for Nebraska agricultural products. They will also serve to provide quality education and employment opportunities for Nebraska students and businesses. There are challenges and issues that remain for the state in order to take advantage of its resource potential. There is a need to produce a comprehensive Nebraska biogenic methane potential study and digital mapping system to identify high-potential producers, co-products, and markets. There is also a need to develop a web-based format of consolidated information specific to Nebraska to aid in connecting producers, service providers, educators, and policy-makers.

The aim of this article is to examine unbuilt residential projects designed by Vilanova Artigas. The formal and spatial conception of these projects is investigated through physical models. The object of this research project consists of the unbuilt residential projects designed by Vilanova Artigas in Sao Paulo that are available in FAUUSP's digital Library. The results indicate that physical models contribute to a better interpretation of unbuilt architectural design, both from the conceptual and aesthetic and from the functional and technical point of view. The original contribution lies in the object, i.e. the unbuilt projects, in the method, using physical models for analysis, and in the objective, viz. to establish a relationship between Artigas' built works and his unbuilt residential projects in order to better understand the design's spatial conception and its architectural approach.

This study evaluated bone responses to titanium implants in the presence of an inorganic graft material. The bilateral mandible incisors of 24 rabbits were surgically extracted and one of the exposed sockets, chosen at random, was filled with an inorganic xenogenic bone graft (Gen-ox (R)), whereas the remaining socket was left to heal naturally and served as a control. After 60 days, titanium implants were inserted in the specific areas, and on days 0, 30, 60, and 180 after the implant insertions, six animals of each group were killed. Digital periapical radiography of implant region was obtained and vertical bone height (VBH) and bone density (BD) were evaluated by digital analysis system. In the undecalcified tissue cuts, bone-to-implant contact (BIC) and bone area (BA) within the limits of the implant threads were evaluated and compared statistically by means of two-way ANOVA and Tukey's test (rho < 0.05). No significant differences were detected in VBH and BA, either between groups or between different experimental intervals. The BD was significantly higher in the experimental group than in the control group in all the intervals tested, but there were no significant differences by interval. The BIC was statistically lower in the control group on day 0; however, a significant increase was observed on days 60 and 180 (rho < 0.05). The use of an inorganic xenograft prior to insertion of a titanium implant did not interfere with the course of osseointegration.

O Modelo de Gestão do Sistema Integrado de Bibliote cas da Universidade de São Paulo (SIBi/USP) incorpora conceitos e ferramentas de ger enciamento que permeiam as organizações modernas para garantir melhores índice s de desempenho sistêmico e prestar serviços com qualidade e eficiência aos usuários. C omo parte do Modelo está a identificação e a descrição detalhada dos processos de trabalho do Sistema, assim como o estabelecimento de alguns indicadores de desempenho . A partir do trabalho inicial, foi elaborado um levantamento complementar para identif icação dos processos que abrangessem o conjunto do Sistema de forma ampla. O s dados foram dispostos em planilhas para melhor visualização, especialmente q uanto aos macro processos, processos, sub processos e atividades. Os processos foram sepa rados em essenciais, gerenciais e de apoio, além de elencadas as atividades pertinentes a cada um deles, bem como as instruções técnicas e fluxos de trabalho. Foram est abelecidos alguns indicadores, tendo por referência os da IFLA já estudados por outro Grupo de Trabalho. Daquele estudo quatro indicadores foram testados e validados pelo SIBi/US P por meio de aplicação piloto em algumas das Bibliotecas do Sistema e outros foram d efinidos no estudo atual. Com isso foi possível mapear os processos e as atividades desenv olvidas pelo conjunto de Bibliotecas sendo que cada Biblioteca, em função de sua especia lidade e especificidade pode adequar o seu mapeamento. A definição de um núcleo básico d e indicadores objetiva viabilizar a concretização da missão e dos objetivos em consonân cia com a política do SIBI/USP

Sistemas de gestão desenvolvidos para a web, a partir de metadados, permitem manutenção eficiente de grandes quantidades de informação. Um vocabulário controlado como o utilizado pelo Sistema Integrado de Bibliotecas da USP (SIBi/USP) necessita de atualização contínua realizada através de uma rede colaborativa com a participação de bibliotecários indexadores de todas as áreas do conhecimento. Este trabalho apresenta os resultados obtidos com o sistema de gestão desenvolvido pelo Grupo de Gerenciamento para a manutenção do Vocabulário Controlado do SIBi/USP. O fluxo deste sistema consiste em filtros de validação realizados pelos componentes do Grupo de Gerenciamento do Vocabulário. A metodologia de gestão do Vocabulário possui além deste sistema, uma política de governança. Os resultados obtidos nos seis anos desde a ativação do sistema de gestão pela Base de Sugestões consistiram em: 1192 inclusões de descritores, 240 alterações, 61 exclusões, totalizando 1493 operações. A gestão e o controle de qualidade do Vocabulário permitiram o aprimoramento do tratamento e da recuperação da informação no Banco de Dados Bibliográficos da USP – DEDALUS.

The University of São Paulo celebrates its Integrated Library System 30th anniversary with an exhibition, discussing the problems of retrieval, preservation and access to knowledge resulting from the exceptional changes ICTs produce in contemporary society. It opens up discussions on the main function of the ancient library institution, reinforces its relevance and reflects on technical tools and social practices that make information and basic raw material accessible, generating new forms of knowledge. About the future library, it´s a call for reflection on how the brilliant minds of the past projected into the future, which for us are the achievements of the present. The future has already started and expects each one to exercise inventiveness and determination to build it in a human and collaborative sense.

Concebida para rastrear dados de outros sites similares, ela se constitui também em uma útil ferramenta de pesquisa

Low level laser therapy is used as a treatment of several conditions, including inflammatory processes and wound healing. Possible changes in mechanical properties of cells, caused by illumination, are investigated with optical magnetic twisting cytometry (OMTC), which is a technique used to evaluate mechanical properties in cell culture. Ferromagnetic micro beads are bound to cell cytoskeleton, the beads are magnetized vertically and a horizontal twisting magnetic field is applied causing a torque that moves the beads and deforms the cell, the beads rotate and displace. Based on the lateral displacement of the beads, elastic shear and loss moduli are obtained. Samples of human bronchial epithelial cell culture were divided in two groups: one was illuminated with a 660 nm red laser, 30 mW power, 0.75 W/cm2 irradiance, during different time intervals, and the other one, the control group, was not illuminated. The values of the mechanical constants of the cells of the control group showed a tendency of increasing with the time out of the incubator. On the other hand, the illuminated group showed constancy on the behavior of both moduli, keeping the normal conditions of the cell culture. Those results indicate that illumination can induce cells to homeostasis, and OMTC is sensitive to observe departures from the steady conditions. Hence, OMTC is an important technique which can be used to aggregate knowledge on the light effect in cell cytoskeleton and even on the low level laser therapy mechanisms in inflammatory processes and/or wound healing.

A reassessment of the taxonomic status of Amblyomma cajennense based on the morphological analyses of ticks from the whole distribution area of the species resulted in the redescription of A. cajennense, the validation of 2 species which had been reduced to synonymy in the past, Amblyomma mixtum and Amblyomma sculptum, and the description and definition of 3 new species, Amblyomma tonelliae n. sp., Amblyomma interandinum n. sp., and Amblyomma patinoi n. sp. This study provides descriptions and redescriptions, scanning electron microscopic and stereomicroscopic images, updated synonymies, information on geographical distributions, and host associations for each of the 6 species. Amblyomma cajennense s.s. is found in the Amazonian region of South America, A. interandinum is reported from the northern part of the Inter-Andean valley of Peru, A. mixtum is present from Texas (U.S.A.) to western Ecuador, A. patinoi occurs in the Eastern Cordillera of Colombia, A. tonelliae is associated with the dry areas of the Chaco region which spans from central-northern Argentina to Bolivia and Paraguay, whereas A. sculptum is distributed from the humid areas of northern Argentina, to the contiguous regions of Bolivia and Paraguay and the coastal and central-western states of Brazil.

FIR spectroscopy is an alternative way of collecting spectra of many inorganic pigments and corrosion products found on art objects, which is not normally observed in the MIR region. Most FIR spectra are traditionally collected in transmission mode but as a real novelty it is now also possible to record FIR spectra in ATR (Attenuated Total Reflectance) mode. In FIR transmission we employ polyethylene (PE) for preparation of pellets by embedding the sample in PE. Unfortunately, the preparation requires heating of the PE in order to produces at transparent pellet. This will affect compounds with low melting points, especially those with structurally incorporated water. Another option in FIR transmission is the use of thin films. We test the use of polyethylene thin film (PETF), both commercial and laboratory-made PETF. ATR collection of samples is possible in both the MIR and FIR region on solid, powdery or liquid samples. Changing from the MIR to the FIR region is easy as it simply requires the change of detector and beamsplitter (which can be performed within a few minutes). No preparation of the sample is necessary, which is a huge advantage over the PE transmission method. The most obvious difference, when comparing transmission with ATR, is the distortion of band shape (which appears asymmetrical in the lower wavenumber region) and intensity differences. However, the biggest difference can be the shift of strong absorbing bands moving to lower wavenumbers in ATR mode. The sometimes huge band shift necessitates the collection of standard library spectra in both FIR transmission and ATR modes, provided these two methods of collecting are to be employed for analyses of unknown samples. Standard samples of 150 pigment and corrosion compounds are thus collected in both FIR transmission and ATR mode in order to build up a digital library of spectra for comparison with unknown samples. XRD, XRF and Raman spectroscopy assists us in confirming the purity or impurity of our standard samples. 24 didactic test tables, with known pigment and binder painted on the surface of a limestone tablet, are used for testing the established library and different ways of collecting in ATR and transmission mode. In ATR, micro samples are scratched from the surface and examined in both the MIR and FIR region. Additionally, direct surface contact of the didactic tablets with the ATR crystal are tested together with water enhanced surface contact. In FIR transmission we compare the powder from our test tablet on the laboratory PETF and embedded in PE. We also compare the PE pellets collected using a 4x beam condenser, focusing the IR beam area from 8 mm to 2 mm. A few samples collected from a mural painting in a Nepalese temple, corrosion products collected from archaeological Chinese bronze objects and samples from a mural paintings in an Italian abbey, are examined by ATR or transmission spectroscopy.

Es wurde ein für bodengebundene Feldmessungen geeignetes System zur digital-holographischen Abbildung luftgetragener Objekte entwickelt und konstruiert. Es ist, abhängig von der Tiefenposition, geeignet zur direkten Bestimmung der Größe luftgetragener Objekte oberhalb von ca. 20 µm, sowie ihrer Form bei Größen oberhalb von ca. 100µm bis in den Millimeterbereich. Die Entwicklung umfaßte zusätzlich einen Algorithmus zur automatisierten Verbesserung der Hologrammqualität und zur semiautomatischen Entfernungsbestimmung großer Objekte entwickelt. Eine Möglichkeit zur intrinsischen Effizienzsteigerung der Bestimmung der Tiefenposition durch die Berechnung winkelgemittelter Profile wurde vorgestellt. Es wurde weiterhin ein Verfahren entwickelt, das mithilfe eines iterativen Ansatzes für isolierte Objekte die Rückgewinnung der Phaseninformation und damit die Beseitigung des Zwillingsbildes erlaubt. Weiterhin wurden mithilfe von Simulationen die Auswirkungen verschiedener Beschränkungen der digitalen Holographie wie der endlichen Pixelgröße untersucht und diskutiert. Die geeignete Darstellung der dreidimensionalen Ortsinformation stellt in der digitalen Holographie ein besonderes Problem dar, da das dreidimensionale Lichtfeld nicht physikalisch rekonstruiert wird. Es wurde ein Verfahren entwickelt und implementiert, das durch Konstruktion einer stereoskopischen Repräsentation des numerisch rekonstruierten Meßvolumens eine quasi-dreidimensionale, vergrößerte Betrachtung erlaubt. Es wurden ausgewählte, während Feldversuchen auf dem Jungfraujoch aufgenommene digitale Hologramme rekonstruiert. Dabei ergab sich teilweise ein sehr hoher Anteil an irregulären Kristallformen, insbesondere infolge massiver Bereifung. Es wurden auch in Zeiträumen mit formal eisuntersättigten Bedingungen Objekte bis hinunter in den Bereich ≤20µm beobachtet. Weiterhin konnte in Anwendung der hier entwickelten Theorie des ”Phasenrandeffektes“ ein Objekt von nur ca. 40µm Größe als Eisplättchen identifiziert werden. Größter Nachteil digitaler Holographie gegenüber herkömmlichen photographisch abbildenden Verfahren ist die Notwendigkeit der aufwendigen numerischen Rekonstruktion. Es ergibt sich ein hoher rechnerischer Aufwand zum Erreichen eines einer Photographie vergleichbaren Ergebnisses. Andererseits weist die digitale Holographie Alleinstellungsmerkmale auf. Der Zugang zur dreidimensionalen Ortsinformation kann der lokalen Untersuchung der relativen Objektabstände dienen. Allerdings zeigte sich, dass die Gegebenheiten der digitalen Holographie die Beobachtung hinreichend großer Mengen von Objekten auf der Grundlage einzelner Hologramm gegenwärtig erschweren. Es wurde demonstriert, dass vollständige Objektgrenzen auch dann rekonstruiert werden konnten, wenn ein Objekt sich teilweise oder ganz außerhalb des geometrischen Meßvolumens befand. Weiterhin wurde die zunächst in Simulationen demonstrierte Sub-Bildelementrekonstruktion auf reale Hologramme angewandt. Dabei konnte gezeigt werden, dass z.T. quasi-punktförmige Objekte mit Sub-Pixelgenauigkeit lokalisiert, aber auch bei ausgedehnten Objekten zusätzliche Informationen gewonnen werden konnten. Schließlich wurden auf rekonstruierten Eiskristallen Interferenzmuster beobachtet und teilweise zeitlich verfolgt. Gegenwärtig erscheinen sowohl kristallinterne Reflexion als auch die Existenz einer (quasi-)flüssigen Schicht als Erklärung möglich, wobei teilweise in Richtung der letztgenannten Möglichkeit argumentiert werden konnte. Als Ergebnis der Arbeit steht jetzt ein System zur Verfügung, das ein neues Meßinstrument und umfangreiche Algorithmen umfaßt. S. M. F. Raupach, H.-J. Vössing, J. Curtius und S. Der Tumorstoffwechsel ist charakterisiert durch eine erhöhte Glykolyserate und die Anreicherung von Laktat auch in Anwesenheit von Sauerstoff. In früheren Arbeiten dieser Arbeitsgruppe konnte gezeigt werden, dass ein hoher Laktatgehalt in Primärtumoren signifikant mit einem gesteigerten Metastasierungsverhalten und einer schlechteren Patientenprognose verknüpft ist. Ein wichtiges Verfahren zur Bestimmung des Metabolitstatus in kryokonservierten Tumorschnitten ist die induzierte, metabolische Biolumineszenz. Im Rahmen dieser Arbeit wurde ein neues, digitales Kamerasystem für Biolumineszenzmessungen etabliert. Außerdem wurde der für die Laktat- und Glukosemessungen benötigte Enzymmix optimiert, so dass eine bessere Lichtausbeute und eine Kosteneinsparung von etwa 50% erzielt werden konnte. Durch die Einführung von Kontrollmessungen und die Verwendung eines neu entwickelten ImageJ-PlugIns für eine halbautomatisierte Auswertung, konnte ein zuverlässiges Messsystem etabliert werden, das Ergebnisse mit einer verbesserten Reproduzierbarkeit liefert. rnDieses neue Messsytem wurde in einer Studie an zehn humanen xenotransplantierten Plattenepithelkarzinomen des Kopf-Hals-Bereiches erfolgreich eingesetzt. Es konnte gezeigt werden, dass eine signifikante, positive Korrelation zwischen dem Laktatgehalt in soliden Primärtumoren und der Resistenz gegenüber einer fraktionierten Strahlentherapie besteht. rnIn einer weiterführenden Studie wurde im gleichen Xenograftmodell der Einfluss einer fraktionierten Strahlentherapie auf den Tumorstoffwechsel untersucht. Der Vergleich von zwei humanen Plattenepithelkarzinomlinien des Kopf-Hals-Bereichs im Verlauf einer fraktionierten Strahlentherapie zeigt deutliche Unterschiede zwischen strahlenresistenten und strahlensensiblen Tumoren. In der strahlenempfindlichen Tumorlinie UT-SCC 14 treten nach Bestrahlung mit drei Fraktionen in der Expression glykolyse-assoziierter Gene und nach zehn Fraktionen im ATP- und Laktatgehalt signifikante Veränderungen auf. Im Gegensatz dazu wird bei der strahlenresistenten Linie UT-SCC 5 lediglich eine Absenkung des Laktatgehaltes nach zehn Fraktionen beobachtet, nicht jedoch des ATP-Gehalts. Die mRNA-Expression wird in UT-SCC 5 nicht durch eine fraktionierte Bestrahlung verändert. Diese Ergebnisse liefern erste Hinweise, dass das mRNA-Expressionslevel und der Metabolitgehalt frühe Marker für das Ansprechen auf eine Strahlentherapie sein können.rn
