O presente relatório tem como objetivo a explanação e a discussão de forma concisa da atividade docente desenvolvida no ano letivo 2012/2013, na Escola Básica 2,3/S Cunha Rivara de Arraiolos com as turmas do 8.º C e 11.º D nas disciplinas de Ciências Naturais e Saúde respetivamente, sob a orientação da Doutora Marília Cid. Apresentam-se os fundamentos teóricos que sustentaram o delineamento de todas as metodologias e estratégias de ensino no sentido de promover situações de aprendizagem significativa aos alunos das turmas referidas. Este é também um testemunho da participação ativa no projeto de educação para a saúde em colaboração com as entidades promotoras de saúde no Concelho de Arraiolos no sentido de organizar atividades extracurriculares propiciadoras da aprendizagem nesta dimensão, com a dinamização de sessões de esclarecimento de Educação Sexual a várias turmas da escola e a produção de um website informativo e de apoio aos alunos do 3.º Ciclo do Ensino Básico e do Ensino Secundário sobre Educação Sexual. Foi apanágio deste trabalho a análise e reflexão também evidenciadas no trabalho de investigação sobre as atividades desenvolvidas no âmbito da Educação para a Saúde mas também em toda a atividade de lecionação. Esta análise e reflexão, bem como a pesquisa individual e o trabalho colaborativo permitiram o desenvolvimento progressivo das minhas capacidades, da qualidade do trabalho desenvolvido e o melhoramento da formação académica. /ABSTRACT: This report aims at explaining concisely and discussion of teaching developed in the academic year 2012/2013, at Escola Básica 2,3 / S Cunha Rivara in Arraiolos with classes C from 8th grade class and D from 11th , in the disciplines of Natural Sciences and Health respectively under the guidance of PhD Teacher Marilia Cid. Here we present the theoretical foundations that supported the design of all methodologies and teaching strategies to promote meaningful learning situations for students of classes above. This is also a testament to the active participation in the design of health education in collaboration with the health promoters in the County of Arraiolos to organize extracurricular activities conducive learning these dimensions, the organization of information sessions to various Sex Education school classes and the production of an informational website to support the students of the 3rd cycle and Secondary Education on Sexual Education. It was the hallmark of this work, analysis and reflection evidenced in research work on the activities within the Health Education but also of the entire teaching activity. This analysis and reflection as well as individual research and collaborative work allowed the progressive development of my abilities, quality of work and the improvement of academic training.

Background: Heart failure (HF) patients develop important changes in body composition, but only a small number of studies have evaluated the associations between these changes and functional class deterioration in a prospective manner. Objective: The aim of this study was to evaluate whether changes in bioimpedance parameters were associated with NYHA functional class deterioration over six months. Methods: A total of 275 chronic stable HF patients confirmed by echocardiography were recruited. Body composition measurements were obtained by whole body bioelectrical impedance with multiple frequency equipment (BodyStat QuadScan 4000). We evaluated functional class using the New York Heart Association (NYHA) classification at baseline and after six months. Results: According to our results, 66 (24%) subjects exhibited functional class deterioration, while 209 improved or exhibited no change. A greater proportion of patients exhibited higher extracellular water (> 5%), and these patients developed hypervolemia, according to location on the resistance/reactance graph. A 5% decrease in resistance/height was associated with functional class deterioration with an OR of 1.42 (95% CI 1.01-2.0, p = 0.04). Conclusions: Body composition assessment through bioelectrical impedance exhibited a valuable performance as a marker of functional class deterioration in stable HF patients.

In this work we demonstrate the efficiency of some dimeric [Ln4(H2O)6(β-GeW10O38)2]12− anions composed of lanthanide-stabilised dilacunary Keggin tungstogermanate fragments (ββ-Ln4, Ln = Dy, Ho, Er, Tm) as heterogeneous catalysts for the organic phase oxidation of aniline with hydrogen peroxide. The results obtained evidence total conversion of aniline at room temperature, as well as full selectivity towards nitrosobenzene, and the catalysts are able to retain both their activity and selectivity after several runs. Peroxopolyoxometalate intermediaries have been identified as the catalytically active species during the aniline-to-nitrosobenzene oxidation process.

Mastoparans are tetradecapeptides found to be the major component of vespid venoms. A mastoparan toxin isolated from the venom of Anterhynchium flavomarginatum micado has been crystallized and X-ray diffraction data collected to 2.7 Angstrom resolution using a synchrotron-radiation source. Crystals were determined to belong to the space group P6(2)22 (P6(4)22). This is the first mastoparan to be crystallized and will provide further insights into the conformational significance of mastoparan toxins with respect to their potency and activity in G-protein regulation.

This dissertation presents a comparative study of three factories in Cork Harbour area, Sunbeam Wolsey (1927-90), Irish Steel (1939-2001) and the Ford Marina Plant (1917-84). All three factories were significant industrial employers in both a domestic (Irish) and a local (Cork) context and are broadly representative of the Irish manufacturing industry that was developed under the policies of tariff protection introduced in the 1930s and gradually phased out between the late 1950s and the mid-1980s. Sunbeam Wolsey was a textile and clothing concern located on the north side of Cork City that possessed a borderline monopoly within its economic sector and was among the largest private employers of female labour in twentieth century Ireland. Irish Steel was the country’s only steel mill, located on Haulbowline island, a brief ferry-ride from the seaside town of Cobh, and was unusual in being one of the few manufacturing concerns operated as a nationalised industry under the auspices of the state. The Ford Marina plant predated the introduction of protectionism by more than a decade and began as the centre of the Ford empire’s tractor manufacturing business, before switching to the production of private motor vehicles for the Irish market in 1932. All three industries were closed or sold off when the state withdrew support, either in the form of tariff protection (Ford, Sunbeam) or direct funding (Irish Steel). While devoting much attention to the three firms, the central concern of this dissertation is not the companies themselves (though the economic history portion of the dissertation is substantial), but the workers they employed, examining the lives of these individuals both as members of the Irish working class, and, more specifically, as employees of the three factories under consideration. The project can be best described as a comparative factory study, comparing and contrasting the three workforces, focusing primarily on industrial relation and the experience of work. This dissertation utilises both documentary evidence and a significant quantity of oral testimony, breaking new ground by making the workplace the central focus of its investigation. The principal aims of the study are: 1. To document the lives of those who worked in these factories, capturing through oral testimony their subjective experiences of social class and factory life, as well as differences among narrators in terms of gender and status. In achieving this aim, the study will provide a broader social context for its detailed analysis of work and industrial relations in each firm. 2. To analyse the three workplaces and determine how and why each developed such distinct systems of industrial relations at the factory level, as well as to compare and contrast these systems. 3. To examine the nature of work in each factory and to determine how work and industrial relations in each firm developed over time, relating these changes both to internal and external factors. Additionally, the project will provide a comparative analysis of these changes.

O presente relatório surge como um complemento à minha Prática de Ensino Supervisionada, realizada no ano lectivo de 2009/201O, visando essencialmente a reflexão e documentação do trabalho por mim desenvolvido ao longo deste ano. A minha Prática de Ensino Supervisionada debruçou-se sobre a disciplina de Educação Física, e decorreu na Escola EB 2,3 Conde de Vilalva, onde leccionei a disciplina a uma turma de 8° ano, e na Escola Secundária André de Gouveia, tendo leccionado a disciplina a um 12° ano. Durante este mesmo ano lectivo, leccionei ainda entre 8 intervenções a uma turma do 1° Ciclo de Ensino Básico, 1° ano; e 9 intervenções a uma turma do 2° Ciclo de Ensino Básico, o 5° ano. ABSTRACT: This document comes as a complement to my Supervised Teaching Practice, held in the 2009/201O academic year, aiming primarily to reflection and documentation of the work developed throughout this year. My Supervised Teaching Practice was focused on the discipline of Physical Education, and held at EB 2,3 Conde de Vilalva where I taught a class in 8th grade, and in high school André de Gouveia, where I taught the discipline to a class in 12th grade. During that same academic year, I taught between 8 and 1O interventions with a class of 1st cycle of education, 1st grade, and with a class of 2nd cycle of education, a 5th grade.

The notion of academic performance renders account of the results achieved by the students during their school education.  Internally, there are two opposed phenomena:  school success and school failure.  Sociology of education has contributed to the discussion of both notions revealing their social nature closely related to the institutional and socio-cultural contexts in which education is developed.  This paper is the result of a research project conducted for our doctoral thesis.  Its purpose is to contribute to enlighten the scopes of this discussion, by analyzing the school development of a group of working class women from Mendoza. We were interested, in the first place, in knowing the reasons why they quit school when they were young girls or adolescents.  Subsequently, we have approached the opportunities they had to access the system again during their adulthood, the obstacles they had to face and the resources to be able to complete the medium level.

After initial efforts in the late 1980s, the interest in thermochemiluminescence (TCL) as an effective detection technique has gradually faded due to some drawbacks, such as the high temperatures required to trigger the light emission and the relatively low intensities, which determined a poor sensitivity. Recent advances made with the adoption of variably functionalized 1,2-dioxetanes as innovative luminophores, have proved to be a promising approach for the development of reagentless and ultrasensitive detection methods exploitable in biosensors by using TCL compounds as labels, as either single molecules or included in modified nanoparticles. In this PhD Thesis, a novel class of N-substituted acridine-containing 1,2-dioxetanes was designed, synthesized, and characterized as universal TCL probes endowed with optimal emission-triggering temperatures and higher detectability particularly useful in bioanalytical assays. The different decorations introduced by the insertion of both electron donating (EDGs) and electron withdrawing groups (EWGs) at the 2- and 7-positions of acridine fluorophore was found to profoundly affect the photophysical properties and the activation parameters of the final 1,2-dioxetane products. Challenges in the synthesis of 1,2-dioxetanes were tackled with the recourse to continuous flow photochemistry to achieve the target parent compound in high yields, short reaction time, and easy scalability. Computational studies were also carried out to predict the olefins reactivity in the crucial photooxygenation reaction as well as the final products stability. The preliminary application of TCL prototype molecule has been performed in HaCaT cell lines showing the ability of these molecules to be detected in real biological samples and cell-based assays. Finally, attempts on the characterization of 1,2-dioxetanes in different environments (solid state, optical glue and nanosystems) and the development of bioconjugated TCL probes will be also presented and discussed.

Ewing sarcoma (EWS) and CIC-DUX4 sarcoma (CDS) are pediatric fusion gene-driven tumors of mesenchymal origin characterized by an extremely stable genome and limited clinical solutions. Post-transcriptional regulatory mechanisms are crucial for understanding the development of this class of tumors. RNA binding proteins (RBPs) play a crucial role in the aggressiveness of these tumors. Numerous RBP families are dysregulated in cancer, including IGF2BPs. Among these, IGF2BP3 is a negative prognostic factor in EWS because it promotes cell growth, chemoresistence, and induces the metastatic process. Based on preliminary RNA sequencing data from clinical samples of EWS vs CDS patients, three major axes that are more expressed in CDS have been identified, two of which are dissected in this PhD work. The first involves the transcription factor HMGA2, IGF2BP2-3, and IGF2; the other involves the ephrin receptor system, particularly EphA2. EphA2 is involved in numerous cellular functions during embryonic stages, and its increased expression in adult tissues is often associated with pathological conditions. In tumors, its role is controversial because it can be associated with both pro- and anti-tumoral mechanisms. In EWS, it has been shown to play a role in promoting cell migration and neoangiogenesis. Our study has confirmed that the HMGA2/IGF2BPs/IGF2 axis contributes to CDS malignancy, and Akt hyperactivation has a strong impact on migration. Using loss/gain of function models for EphA2, we confirmed that it is a substrate of Akt, and Akt hyperactivation in CDS triggers ligand-independent activation of EphA2 through phosphorylation of S897. Moreover, the combination of Trabectedin and NVP/BEZ235 partially inhibits Akt/mTOR activation, resulting in reduced tumor growth in vivo. Inhibition of EphA2 through ALWII 41_27 significantly reduces migration in vitro. The project aim is the identification of target molecules in CDS that can distinguish it from EWS and thus develop new targeted therapeutic strategies.

Phospholipases A2 (PLA2) are key enzymes for production of lipid mediators. We previously demonstrated that a snake venom sPLA2 named MT-III leads to prostaglandin (PG)E2 biosynthesis in macrophages by inducing the expression of cyclooxygenase-2 (COX-2). Herein, we explored the molecular mechanisms and signaling pathways leading to these MT-III-induced effects. Results demonstrated that MT-III induced activation of the transcription factor NF-κB in isolated macrophages. By using NF-κB selective inhibitors, the involvement of this factor in MT-III-induced COX-2 expression and PGE2 production was demonstrated. Moreover, MT-III-induced COX-2 protein expression and PGE2 release were attenuated by pretreatment of macrophages with SB202190, and Ly294002, and H-7-dihydro compounds, indicating the involvement of p38MAPK, PI3K, and PKC pathways, respectively. Consistent with this, MT-III triggered early phosphorylation of p38MAPK, PI3K, and PKC. Furthermore, SB202190, H-7-dihydro, but not Ly294002 treatment, abrogated activation of NF-κB induced by MT-III. Altogether, these results show for the first time that the induction of COX-2 protein expression and PGE2 release, which occur via NF-κB activation induced by the sPLA2-MT-III in macrophages, are modulated by p38MAPK and PKC, but not by PI3K signaling proteins.

This study aimed at evaluating whether human papillomavirus (HPV) groups and E6/E7 mRNA of HPV 16, 18, 31, 33, and 45 are prognostic of cervical intraepithelial neoplasia (CIN) 2 outcome in women with a cervical smear showing a low-grade squamous intraepithelial lesion (LSIL). This cohort study included women with biopsy-confirmed CIN 2 who were followed up for 12 months, with cervical smear and colposcopy performed every three months. Women with a negative or low-risk HPV status showed 100% CIN 2 regression. The CIN 2 regression rates at the 12-month follow-up were 69.4% for women with alpha-9 HPV versus 91.7% for other HPV species or HPV-negative status (P < 0.05). For women with HPV 16, the CIN 2 regression rate at the 12-month follow-up was 61.4% versus 89.5% for other HPV types or HPV-negative status (P < 0.05). The CIN 2 regression rate was 68.3% for women who tested positive for HPV E6/E7 mRNA versus 82.0% for the negative results, but this difference was not statistically significant. The expectant management for women with biopsy-confirmed CIN 2 and previous cytological tests showing LSIL exhibited a very high rate of spontaneous regression. HPV 16 is associated with a higher CIN 2 progression rate than other HPV infections. HPV E6/E7 mRNA is not a prognostic marker of the CIN 2 clinical outcome, although this analysis cannot be considered conclusive. Given the small sample size, this study could be considered a pilot for future larger studies on the role of predictive markers of CIN 2 evolution.
