O objetivo deste trabalho foi relatar o potencial da técnica de MALDI-MS para caracterizar espécies de lipídios presentes em um único embrião equino e estudar algumas estruturas lipídicas detectadas por dissociação induzida por colisão (CID). No espectro de modo íon positivo, pudemos observar espécies, principalmente, protonadas e sodiadas de esfingomielinas (SM), fosfatidileolinas (PC) e triacilgliceróis (TAG). No modo negativo, observamos fosfatidiletanolaminas (PE) e fosfatidilinositos (PI). Espectros de íons de lípidos com maior intensidade foram utilizados para demonstrar o potencial da informação estrutural por MALDI-MS/MS. O espectro no modo positivo de m/z (massa sobre carga) 760,6 (atribuída como PC34:1) apresentou características de fragmentos PC de m/z 184,1 (denominada cabeça polar de colina), além de perda neutral (NL) de m/z 183 (fosforilcolina). Para o íon de m/z 766,6 (atribuída como PE38:5), observou-se a NL de 140, característica do PE. Para o íon de m/z 808,7 (38,5 atribuído como PC), além do fragmento m/z 184,1 na NL de 183, foi possível observar a perda de trimetilamina (íon de m/z 749,6) e o ciclofosfano (íon de m/z 147,0). Finalmente, para o modo de íon negativo, foram isolados e fragmentados o íon de m/z 863,6 que foi atribuído como PI36:1, devido à presença de m/z 153 (fosfato de glicerol – H2O-H ), 223 (inositol fosfo - 2H2O-H) , 241 (fosfoinositol – H2O-H), 281 (ácido oleico) e 581,3 (lisofosfoinositol – H2O+H). Concluímos que a MALDI - MS permite a detecção de uma ampla gama de espécies de PC, SM, PE, PI e TAG lipídicas, bem como a caracterização rápida e confiante de estruturas lipídicas a partir de um único embrião equino.

Corrole and four of its isomers with subtle structural changes promoted by exchange of nitrogen and carbon atoms in the corrole ring have been studied by traveling wave ion mobility mass spectrometry and collision induced dissociation experiments. Significant differences in shapes and charge distributions for their protonated molecules were found to lead to contrasting gas phase mobilities, most particularly for corrorin, the most "confused" isomer. Accordingly, corrorin was predicted by B3LYP/6-31g(d,p) and collisional cross section calculations to display the most compact tri-dimensional structure, whereas NCC4 and corrole were found to be the most planar isomers. Better resolution between the corrole isomers was achieved using the more polarizable and massive CO2 as the drift gas. Sequential losses of HF molecules were found to dominate the dissociation chemistry of the protonated molecules of these corrole isomers, but their unique structures caused contrasting labilities towards CID, whereas NCC4 showed a peculiar and structurally diagnostic loss of NH3, allowing its prompt differentiation from the other isomers.

Analytic methods were applied and validated to measure residues of chlorfenvinphos, fipronil, and cypermethrin in meat and bovine fat, using the QuEChERS method and gas chromatography-mass spectrometry. For the meat, 2 g of sample, 4mL of acetonitrile, 1.6 g of MgSO4, and 0.4 g of NaCl were used in the liquid-liquid partition, while 80 mg of C18, 80 mg of primary and secondary amine and 150 mg of MgSO4 were employed in the dispersive solid-phase extraction. For the fat, 1 g of sample, 5 mL of hexane, 10 mL of water, 10 mL of acetonitrile, 4 g of MgSO4, and 0.5 g of NaCl were used in the liquid-liquid partition and 50 mg of primary and secondary amine and 150 mg of MgSO4 were used in the dispersive solid-phase extraction. The recovery percentages obtained for the pesticides in meat at different concentrations ranged from 81 to 129% with relative standard deviation below 27%. The corresponding results from the fat ranged from 70 to 123% with relative standard deviation below 25%. The methods showed sensitivity, precision, and accuracy according to EPA standards and quantification limits below the maximum residue limit established by European Union, except for chlorfenvinphos in the fat.

In order to understand the influence of alkyl side chains on the gas-phase reactivity of 1,4-naphthoquinone derivatives, some 2-hydroxy-1,4-naphthoquinone derivatives have been prepared and studied by electrospray ionization tandem mass spectrometry in combination with computational quantum chemistry calculations. Protonation and deprotonation sites were suggested on the basis of gas-phase basicity, proton affinity, gas-phase acidity (?Gacid), atomic charges and frontier orbital analyses. The nature of the intramolecular interaction as well as of the hydrogen bond in the systems was investigated by the atoms-in-molecules theory and the natural bond orbital analysis. The results were compared with data published for lapachol (2-hydroxy-3-(3-methyl-2-butenyl)-1,4-naphthoquinone). For the protonated molecules, water elimination was verified to occur at lower proportion when compared with side chain elimination, as evidenced in earlier studies on lapachol. The side chain at position C(3) was found to play important roles in the fragmentation mechanisms of these compounds. Copyright (c) 2012 John Wiley & Sons, Ltd.

Sulphur plays an essential role in plants and is one of the main nutrients in several metabolic processes. It has four stable isotopes (32S, 33S, 34S, and 36S) with a natural abundance of 95.00, 0.76, 4.22, and 0.014 in atom %, respectively. A method for isotopic determination of S by isotope-ratio mass spectrometry (IRMS) in soil samples is proposed. The procedure involves the oxidation of organic S to sulphate (S-SO4(2-)), which was determined by dry combustion with alkaline oxidizing agents. The total S-SO4(2-) concentration was determined by turbidimetry and the results showed that the conversion process was adequate. To produce gaseous SO2 gas, BaSO4 was thermally decomposed in a vacuum system at 900 ºC in the presence of NaPO3. The isotope determination of S (atom % 34S atoms) was carried out by isotope ratio mass spectrometry (IRMS). In this work, the labeled material (K2(34)SO4) was used to validate the method of isotopic determination of S; the results were precise and accurate, showing the viability of the proposed method.

Spontane-Desorption-Massenspektrometrie zur Charakterisierung von gemischten, selbstorganisierten Schichten zur Metallabscheidung und zur Beobachtung von chemischen Reaktionen in dünnsten Filmen Stephan Krämer Ein Ziel dieser Arbeit war es, selbstorganisierte Schichten aus steifen benzolhaltigen Thiolen herzustellen und zu charakterisieren. Diese selbstorganisierten Schichten sollten als optimale Substrate zur Abscheidung von Metallen durch CVD dienen.In einem ersten Schritt wurden Schichten aus Biphenylthiol (BT) und Biphenyldithiol (BDT) auf Edelmetalloberflächen hergestellt. Die Abhängigkeit der Eigenschaften der Schicht von dem verwendeten Substrat und von der Dauer der Selbstorganisation wurde mit der Spontane-Desorption-Massenspektrometrie untersucht. Die Untersuchung der Schichtdicke erfolgte mit Oberflächenplasmonen-Spektroskopie und die Frage der Struktur der Schichten wurde versucht, mit Hilfe der Fourier-Transform-Infrarot-Spektroskopie zu klären. Nach der Charakterisierung der reinen Schichten wurden binäre Mischungen aus BT und BDT hergestellt und auf Goldoberflächen abgeschieden. Die so hergestellten binären Schichten wurden als Substrate zur Abscheidung von Gold benutzt. Dazu wurde mit Hilfe der CVD-Technik Gold auf den Filmen abgeschieden. Im nächsten Schritt wurden die einfacheren Halogen-substituierten Phenylthiole sowohl als reine Schichten als auch als binäre Mischungen untersucht. Ein weiterer Schwerpunkt stellte die Untersuchungen zur Abscheidung von Metallen auf selbstorganisierten Schichten durch CVD dar. Neben der schon vorgestellten Abscheidung von Gold wurde die Abscheidung von Palladium und von Kupfer untersucht. Im letzten Teil dieser Arbeit wurden der Verlauf einer chemischen Reaktion in einem ultradünnen Polymerfilm beobachtet. Dazu wurden die Vernetzungsreaktion und die Hydrolyse des Copolymer P[tBMA1-co-DMIMA0,11] untersucht.

Hämocyanine sind große, multimere Sauerstofftransport- proteine, die frei gelöst in der Hämolymphe von Arthropoden und Mollusken vorkommen.Zur Charakterisierung verschiedener Arthropoden-hämocyanine wurden deren molare Massen bestimmt. Die mit einer Vielwinkel-Laser-Lichtstreuapparatur ermittelten Molekulargewichte zeigten eine grosse Schwankungsbreite. Dies konnte auf Ungenauigkeiten der zur Berechnung der Molekulargewichte verwendeten spezifischen Extinktions- koeffizienten und Brechungsindex-Inkremente zurückgeführt werden.Mit der Methode der Massenspektrometrie (MALDI-TOF) bestimmte Molekulargewichte einzelner Untereinheiten des Hämocyanins der Vogelspinne Eurypelma californicum zeigten eine sehr gute Übereinstimmung mit aus der Sequenz errechneten Werten.Für das 24-mere Spinnenhämocyanin von Eurypelma californicum wurde die Stabilität gegenüber GdnHCl und der Temperatur auf den verschiedenen strukturellen Ebenen des Proteins untersucht.Viele Stabilitätsuntersuchungen werden an kleinen Proteinen durchgeführt, deren Entfaltung kooperativerfolgt. Bei größeren Proteinen mit unterschiedlichen strukturellen Bereichen (Domänen) ist der Entfaltungs-prozess weitaus komplexer. Ziel war es, durch die Denaturierung des Spinnen-Hämocyanins Erkenntnisse über die Stabilität und Entfaltung der verschiedenen strukturellen Ebenen eines so großen Proteinkomplexes zu gewinnen.Ein wichtiges Charakteristikum für die Interpretation der Entfaltungsexperimente ist die starke Löschung der Tryptophanfluoreszenz im oxygenierten Spinnen-Hämocyanin. Die Löschung kann vollständig durch Förster-Transfer erklärt werden kann. Sie bleibt auf die einzelnen Untereinheiten beschränkt und stellt somit ein reines O2-Beladungssignal dar.Unter Einwirkung von GdnHCl dissoziiert das native, 24-mere Spinnen-Hämocyanin ohne die Entstehung langlebiger Inter- mediate. Die Untereinheiten werden durch das Oligomer stabilisiert. Die Entfaltung eines Monomers, der Unter- einheit e, folgt einer Hierarchie der verschiedenen strukturellen Ebenen des Moleküls. Die Entfaltung beginnt zunächst von außen mit der Auflockerung der Tertiärstruktur. Der Kern von Domäne II mit dem aktiven Zentrum weist hingegen eine besondere Stabilität auf.Die ausgeprägte Hitzestabilität des Eurypelma-Hämocyanins hängt vom Oligomerisierungsgrad, dem verwendeten Puffer und dessen Ausgangs-pH-Wert ab und spiegelt offensichtlich die extremen Lebensbedingungen im Habitat wider.

Perfluoroalkylated substances are a group of chemicals that have been largely employed during the last 60 years in several applications, widely spreading and accumulating in the environment due to their extreme resistance to degradation. As a consequence, they have been found also in various types of food as well as in drinking water, proving that they can easily reach humans through the diet. The available information concerning their adverse effects on health has recently increased the interest towards these contaminants and highlighted the importance of investigating all the potential sources of human exposure, among which diet was proved to be the most relevant. This need has been underlined by the European Union through Recommendation 2010/161/EU: in this document, Member States were called to monitor their presence of in food, producing accurate estimations of human exposure. The purpose of the research presented in this thesis, which is the result of a partnership between an Italian and a French laboratory, was to develop reliable tools for the analysis of these pollutants in food, to be used for generating data on potentially contaminated matrices. An efficient method based on liquid chromatography-mass spectrometry for the detection of 16 different perfluorinated compounds in milk has been validated in accordance with current European regulation guidelines (2002/657/EC) and is currently under evaluation for ISO 17025 accreditation. The proposed technique was applied to cow, powder and human breast milk samples from Italy and France to produce a preliminary monitoring on the presence of these contaminants. In accordance with the above mentioned European Recommendation, this project led also to the development of a promising technique for the quantification of some precursors of these substances in fish. This method showed extremely satisfying performances in terms of linearity and limits of detection, and will be useful for future surveys.

Due to the high price of natural oil and harmful effects of its usage, as the increase in emission of greenhouse gases, the industry focused in searching of sustainable types of the raw materials for production of chemicals. Ethanol, produced by fermentation of sugars, is one of the more interesting renewable materials for chemical manufacturing. There are numerous applications for the conversion of ethanol into commodity chemicals. In particular, the production of 1,3-butadiene whose primary source is ethanol using multifunctional catalysts is attractive. With the 25% of world rubber manufacturers utilizing 1,3-butadiene, there is an exigent need for its sustainable production. In this research, the conversion of ethanol in one-step process to 1,3-butadiene was studied. According to the literature, the mechanisms which were proposed to explain the way ethanol transforms into butadiene require to have both acid and basic sites. But still, there are a lot of debate on this topic. Thus, the aim of this research work is a better understanding of the reaction pathways with all the possible intermediates and products which lead to the formation of butadiene from ethanol. The particular interests represent the catalysts, based on different ratio Mg/Si in comparison to bare magnesia and silica oxides, in order to identify a good combination of acid/basic sites for the adsorption and conversion of ethanol. Usage of spectroscopictechniques are important to extract information that could be helpful for understanding the processes on the molecular level. The diffuse reflectance infrared spectroscopy coupled to mass spectrometry (DRIFT-MS) was used to study the surface composition of the catalysts during the adsorption of ethanol and its transformation during the temperature program. Whereas, mass spectrometry was used to monitor the desorbed products. The set of studied materials include MgO, Mg/Si=0.1, Mg/Si=2, Mg/Si=3, Mg/Si=9 and SiO2 which were also characterized by means of surface area measurements.

In this study the Aerodyne Aerosol Mass Spectrometer (AMS) was used during three laboratory measurement campaigns, FROST1, FROST2 and ACI-03. The FROST campaigns took place at the Leipzig Aerosol Cloud Interaction Simulator (LACIS) at the IfT in Leipzig and the ACI-03 campaign was conducted at the AIDA facility at the Karlsruhe Institute of Technology (KIT). In all three campaigns, the effect of coatings on mineral dust ice nuclei (IN) was investigated. During the FROST campaigns, Arizona Test Dust (ATD) particles of 200, 300 and 400 nm diameter were coated with thin coatings (< 7 nm) of sulphuric acid. At these very thin coatings, the AMS was operated close to its detection limits. Up to now it was not possible to accurately determine AMS detection limits during regular measurements. Therefore, the mathematical tools to analyse the detection limits of the AMS have been improved in this work. It is now possible to calculate detection limits of the AMS under operating conditions, without losing precious time by sampling through a particle filter. The instrument was characterised in more detail to enable correct quantification of the sulphate loadings on the ATD particle surfaces. Correction factors for the instrument inlet transmission, the collection efficiency, and the relative ionisation efficiency have been determined. With these corrections it was possible to quantify the sulphate mass per particle on the ATD after the condensation of sulphuric acid on its surface. The AMS results have been combined with the ice nucleus counter results. This revealed that the IN-efficiency of ATD is reduced when it is coated with sulphuric acid. The reason for this reduction is a chemical reaction of sulphuric acid with the particle's surface. These reactions are increasingly taking place when the aerosol is humidified or heated after the coating with sulphuric acid. A detailed analysis of the solubility and the evaporation temperature of the surface reaction products revealed that most likely aluminium sulphate is produced in these reactions.
