Four magnetic carbon nanotube samples (CNTs: undoped, completely N-doped and two selectively N-doped) have been synthesized by chemical vapor deposition. The materials were tested in the catalytic wet peroxide oxidation (CWPO) of highly concentrated 4 nitrophenol solutions (4-NP, 5 g L-1). Relatively mild operating conditions were considered (atmospheric pressure, T = 50 ºC, pH = 3), using a catalyst load of 2.5 g L-1 and the stoichiometric amount of H2O2 needed for the complete mineralization of 4-NP. N doping was identified to influence considerably the CWPO performance of the materials. In particular, undoped CNTs, with a moderate hydrophobicity, favor the controllable and efficient decomposition of H2O2 into highly reactive hydroxyl radicals (HO•), thus showing high catalytic activity for 4-NP degradation. On the other hand, the completely N-doped catalyst, fully hydrophilic, favors a quick decomposition of H2O2 into non-reactive O2 and H2O species. The selectively N-doped amphiphilic catalysts, i.e. hybrid structures containing undoped sections followed by N-doped ones, provided intermediate results, namely: a higher N content favored H2O2 decomposition towards non-reactive H2O and O2 species, whilst a lower N content resulted in the formation of HO•, increasing 4-NP mineralization. Catalyst stability and reusability were also investigated by consecutive CWPO runs.

Novel magnetic carbon xerogels consisting of interconnected carbon microspheres with iron and/or cobalt microparticles embedded in their structure were developed by a simple route. As inferred from the characterization data, materials with distinctive properties may be directly obtained upon inclusion of iron and/or cobalt precursors during the sol-gel polymerization of resorcinol and formaldehyde, followed by thermal annealing. The unique properties of these magnetic carbon xerogels were explored in the catalytic wet peroxide oxidation (CWPO) of an antimicrobial agent typically found throughout the urban water cycle – sulfamethoxazole (SMX). A clear synergistic effect arises from the inclusion of cobalt and iron in carbon xerogels (CX/CoFe),the resulting magnetic material revealing a better performance in the CWPO of SMX at the ppb level(500 microg L−1) when compared to that of monometallic carbon xerogels containing only iron or cobalt.This effect was ascribed to the increased accessibility of highly active iron species promoted by the simultaneous incorporation of cobalt.The performance of the CWPO process in the presence of CX/CoFe was also evaluated in environmentally relevant water matrices, namely in drinking water and secondary treated wastewater, considered in addition to ultrapure water. It was found that the performance decreases when applied to more complex water and wastewater samples. Nevertheless, the ability of the CWPO technology for the elimination of SMX in secondary treated wastewater was unequivocally shown, with 96.8% of its initial content being removed after 6 h of reaction in the presence of CX/CoFe, at atmospheric pressure, room temperature(T = 25◦C), pH = 3, [H2O2]0= 500 mg L−1and catalyst load = 80 mg L−1. A similar performance (97.8% SMX removal) is obtained in 30 min when the reaction temperature is slightly increased up to 60◦C in an ultra-pure water matrix. Synthetic water containing humic acid, bicarbonate, sulphate or chloride, was also tested. The results suggest the scavenging effect of the different anions considered, as well as the negative impact of dissolved organic matter typically found in secondary treated wastewater, as simulated by the presence of humic acid.An in-situ magnetic separation procedure was applied for catalyst recovery and re-use during reusability cycles performed to mimic real-scale applications. CWPO runs performed with increased SMX concentration (10 mg L−1), under a water treatment process intensification approach, allowed to evalu-ate the mineralization levels obtained, the antimicrobial activity of the treated water, and to propose adegradation mechanism for the CWPO of SMX.

Marine algae are one of the major sources of biologic compounds. In extracellular matrix of these organisms there are sulfated polysaccharides that functions as structural components and provides protection against dehydration. The fraction 1.0 (F1.0) rich in sulfated galactans obtained from red seaweed Hypnea musciformis was physicochemical characterized and evaluated for pharmacologic activity through antioxidant activity, cytotoxic action on erythrocytes, anticoagulant, stimulatory action under antithrombotic heparan sulfate synthesis and their effects on cell proliferation and cycle cell progression. The main components of F1.0 were carbohydrates (49.70 ± 0.10%) and sulfate (44.59 ± 0.015%), presenting phenolic compounds (4.79 ± 0.016%) and low protein contamination (0.92 ± 0.001%). Fraction 1.0 showed polidisperse profile and signs in infrared analysis in 1262, 1074 and 930, 900 and 850 attributed to sulfate esters S=O bond, presence of a 3,6- anidrogalactose C-O bond, non-sulfated β-D-galactose and a C-O-SO4 bond in galactose C4, respectively. The fraction rich in sulfated galactans exhibited strong antioxidant action under lipid peroxidation assay with IC50 of 0.003 mg/mL. Besides the inhibition of hemolysis induced by H2O2 in erythrocytes treated with F1.0, this fraction did not promote significant cytotoxity under erythrocytes membranes. F1.0 exhibited low anticoagulant activity causing moderate direct inhibition of enzimatic activity of thrombin. This fraction promoted stimulation around of 4.6 times on this synthesis of heparan sulfate (HS) by rabbit aortic endothelial cells (RAEC) in culture when was compared with non treated cells. The fraction of this algae displayed antiproliferative action under RAEC cells causing incresing on cell number on S fase, blocking the cycle cell progression. Thus F1.0 presented cytostatic and no cytotoxic action under this cell lineage. These results suggest that F1.0 from H. musciformis have antioxidant potential which is a great effect for a compound used as food and in food industry which could be an alternative to food industry to prevent quality decay of lipid containing food due to lipid peroxidation. These polysaccharides prevent the lipid peroxidation once the fraction in study exhibited strong inhibitory action of this process. Furthermore that F1.0 present strong antithrombotic action promoting the stimulation of antithrombotic HS synthesis by endothelial cells, being important for thrombosis preventing, by its inhibitory action under reactive oxygen species (ROS) in some in vitro methods, being involved in promotion of hypercoagulability state.

O objectivo principal deste trabalho foi estudar as propriedades e comportamento de polioxotungstatos (POM) do tipo Keggin com interesse em catálise oxidativa. Os estudos efectuados centraram-se no comportamento electroquímico dos aniões em meio não aquoso, na estabilidade das suas estruturas em diferentes meios, na sua capacidade de catalisar a oxidação de diferentes substratos orgânicos e na sua eficácia em oxidar, de forma selectiva, um modelo não fenólico da lenhina. Efectuou-se, igualmente, o estudo estrutural de diversos aniões pela técnica de EXAFS. Neste trabalho, prepararam-se e caracterizaram-se alguns dos sais de tetra-n-butilamónio (TBA) dos polioxotungstatos estudados: compostos de Keggin, TBAx[XW12O40], lacunares, TBAxHy[XW11O39], mono-substituídos, TBAxHy[XW11M(H2O)O39]·nH2O, com X= P e Si e M = CoII, NiII, ZnII, CuIl, RuIII, MnIII e FeIII e os compostos com mistura de átomos adenda TBAx[XW11VO40]·nH2O (X = P e Si). Seleccionou-se este conjunto de polioxotungstatos de forma a efectuar-se um estudo comparativo da influência da natureza do heteroátomo central (P e Si) e dos metais de transição M nas propriedades estudadas. O conjunto de sais de TBA dos silicotungstatos estudados mostraram ser isoestruturais, apresentando a mesma estrutura dos fosfotungstatos análogos. O comportamento electroquímico dos polioxotungstatos foi estudado em soluções de acetonitrilo por voltametria cíclica e electrólise a potencial controlado. Verificou-se a ocorrência de vários processos mono-electrónicos de oxi-redução, reversíveis ou quasi-reversíveis, associados aos átomos de WVI/V e a alguns dos metais de transição. Os metais em estado de oxidação +3 reduziram-se mais facilmente do que os átomos de WVI. O metal CuII apresentou um comportamento diferente dos outros metais de transição. Este metal, na estrutura do POM, reduziu-se a CuI, proporcionando a observação do anião [PW11CuIO39]6- pela primeira vez. A redução posterior do CuI conduziu à formação de Cu0, que se depositou na superfície do eléctrodo. A re-oxidação do cobre a CuII conduziu à reconstituição da estrutura do POM, nas condições estudadas. Constatou-se que a ocorrência de protões na fórmula molecular dos POMs influenciou o seu comportamento electroquímico. Para os compostos que apresentam protões, a redução dos átomos de tungsténio ocorreu a potenciais menos negativos do que para aqueles que não apresentam protões na sua fórmula. Para os primeiros observou-se a transferência global de um maior número de electrões no mesmo intervalo de potencial, originando soluções fortemente azuladas. Quando os catiões tetra-n-butilamónio foram substituídos por catiões de menor dimensão, como Li+ e Na+, ocorreu a formação de pares iónicos com os polianiões [PW12O40]3- e [SiW11VO40]5-, originando um aumento do potencial de redução. Não houve evidência da formação de pares iónicos com os catiões TBA+. Este foi o primeiro estudo sistemático do comportamento electroquímico dos aniões lacunares e mono-substituídos em meio não aquoso. Estudou-se a estrutura dos polioxotungstatos em sais de TBA e em soluções de acetonitrilo. A aplicação da técnica de EXAFS ao estudo deste tipo de compostos em solução foi realizada pela primeira vez. Pela análise estrutural nos sólidos verificou-se que, a natureza do metal de transição M e do átomo central X, na estrutura do POM, influenciam o tamanho dos vários octaedros que o constituem. Não se observaram diferenças significativas nas estruturas dos polianiões em solução. A estabilidade da estrutura dos polioxometalatos na presença de um excesso de H2O2, em soluções de acetonitrilo/H2O foi analisada por espectroscopia de absorção de EXAFS, RMN de 31P, IV e espectrofotometria de absorção no UV-Vis. De uma forma geral, os POMs em que o átomo central da estrutura é o Si apresentaram maior estabilidade do que os POMs correspondentes com átomo de P no centro. Em solução de acetonitrilo, na ausência de H2O2, todos os aniões mostraram ser estáveis durante vários dias. Em solução, na presença de H2O2 em excesso (H2O2/POM = 1300), o anião lacunar [PW11O39]7- não é estável, transformando-se no anião de Venturello, {PO4[W(O)(O2)2]4}3-, após a formação de [PW12O40]3-, como produto intermediário. Em relação aos aniões substituídos [PW11M(H2O)O39]p-, M = MnIII, RuIII, FeIII, CoII e ZnII, verificou-se o seguinte na presença de H2O2: os aniões com MnIII e CoII transformaram-se no anião de Keggin, [PW12O40]3-. Os aniões de RuIII e FeIII mantiveram as suas estruturas e o anião de ZnII decompôs-se em {HPO4[W(O)(O2)2]2}2- e fosfato. Para estes casos de não estabilidade estrutural, o processo de decomposição foi mais rápido na presença de maiores conteúdos de água. Pela análise de EXAFS, na presença de um menor excesso de H2O2 (H2O2/POM = 70) e apenas 8% de parte aquosa, verificou-se que os aniões substituídos por MnIII mantiveram a sua estrutura, embora o ligando H2O, coordenado ao Mn, tivesse sido substituído por um grupo oxo no polianião [SiW11Mn(H2O)O39]5-, e por um grupo peroxo no polianião [PW11Mn(H2O)O39]4-. O anião com RuIII, nestas condições, também mostrou substituir o seu ligando H2O por um grupo peroxo ou hidrogenoperoxo. Os polioxotungstatos mono-substituídos e lacunares mostraram ser catalisadores eficientes para a oxidação de cis-cicloocteno, geraniol e ciclooctano com H2O2. A maior novidade deste trabalho residiu na actividade catalítica apresentada pelos silicotungstatos estudados, contrariando o que é referido na literatura. Outro aspecto inovador foi o elevado valor de conversão obtido para a oxidação de ciclooctano. Este substrato foi oxidado com 74% de conversão, após 2h de reacção e com 80% de selectividade para o hidroperóxido de ciclooctilo, na presença do anião [PW11Fe(H2O)O39]4-. Os restantes produtos de reacção foram o ciclooctanol e a ciclooctanona. Os silicotungstatos apresentaram maior selectividade para o hidroperóxido de ciclooctilo do que os fosfotungstatos. O geraniol foi completamente oxidado após 3h de reacção, com 82% de selectividade para o 2,3-epoxigeraniol, na presença do anião [PW11Mn(H2O)O39]4-. O cis-cicloocteno foi oxidado ao seu epóxido, com 92% de conversão ao fim de 5h de reacção, na presença do anião lacunar [PW11O39]7-. O estudo da capacidade oxidativa do anião [SiW11VO40]5- foi analisada utilizando-se um modelo não fenólico da lenhina, a anisoína. Estudaram-se as condições favoráveis à obtenção de uma reacção selectiva para o anisilo, de forma a poder estudar-se a cinética da reacção. A estequiometria da reacção mostrou ser de 1:2 anisoína/POM. As ordens de reacção foram determinadas pelo método das velocidades iniciais e, a partir destes resultados, conheceu-se que o POM não estava envolvido no passo que limita a velocidade da reacção, sendo esta limitada pela transformação da anisoína. O estudo realizado sobre o efeito isotópico sugeriu que o passo que limitou a velocidade de reacção correspondeu à enolação da anisoína. Desta forma, observou-se pela primeira vez, que o POM oxida um modelo não fenólico da lenhina por via de enolação.

The plants are often exposed to variations in environmental conditions that may trigger metabolic disturbances leading to a consequent loss in productivity of crops. These stressful conditions usually induce an accumulation of reactive oxygen species (ROS) in the cell, a condition known how oxidative stress. Among these species, hydrogen peroxide (H2O2) is an important molecule involved in numerous signaling mechanisms. The present study aimed to understand the relationship between the different enzymatic mechanisms of elimination of H2O2 by catalase (CAT) and ascorbate peroxidase (APX) in leaf tissues of seedlings of the species Vigna unguiculata L. Walp, under conditions of oxidative stress induced by application of CAT inhibitor, 3-amino-1,2,4-triazole (3-AT), and H2O2 itself on the roots. Three experiments were conducted. The first experiment was performed applying the compound 3-AT (5 mM) during the time (hours). In the second experiment, seedlings were exposed to different concentrations of H2O2 (2.5, 5.0, 7.5, 10 mM) for 48 h. The third strategy included the pre-treatment with H2O2 (2.5 mM) for 24 h, followed by subsequent treatment with the inhibitor 3-AT and recovery control condition. Treatment with 3-AT causes a strong inhibition of CAT activity in leaf tissues accompanied by an increase of activity of APX. However a decrease in oxidative damage to lipids is not observed as indicated by TBARS. It was observed that activity of APX is directly linked to the content of peroxide. Inductions in the activities of CAT and APX were observed mainly in the seedlings treated with 2.5 mM H2O2. This can be associated with a decrease in oxidative damage to lipids. In contrast, one same tendency was not observed in treatments with higher concentrations of this ROS. These results suggest that the concentration of 2.5 mM H2O2 can induce responses antioxidants later in seedling cowpea. This concentration when applied as pre-treatment for 24 h promoted an induction systems removers CAT and APX, both in activity and in terms of gene expression. However this increment was not observed in the recovered plants and the plants subsequently subjected to 3-AT. Additionally, the pretreatment was not sufficient to attenuate the inhibition of CAT activity and oxidative damage to lipids caused by the subsequent application of this inhibitor. The results showed that the application of 3-AT and H2O2 in the root systems of seedlings of cowpea promote changes in the parameters analyzed in leaf tissues that indicate a direct response to the presence of these factors or systemic signaling mecanisms. H2O2 appears to activate the responses of two antioxidant systems in this study thar does not promote greater protection in case of additional treatment with 3-AT. This demonstrates the importance of the CAT system. In this work, complete results indicate that there is a difference between the signaling and the effects caused by exposure to H2O2 and by treatment with 3-AT

The β-proteobacterium Chromobacterium violaceum is a Gram-negative, free-living, saprophytic and opportunistic pathogen that inhabits tropical and subtropical ecosystems among them, in soil and water of the Amazon. It has great biotechnological potential, and because of this potential, its genome was completely sequenced in 2003. Genome analysis showed that this bacterium has several genes with functions related to the ability to survive under different kinds of environmental stresses. In order to understand the physiological response of C. violaceum under oxidative stress, we applied the tool of shotgun proteomics. Thus, colonies of C. violaceum ATCC 12472 were grown in the presence and absence of 8 mM H2O2 for two hours, total proteins were extracted from bacteria, subjected to SDS-PAGE, stained and hydrolysed. The tryptic peptides generated were subjected to a linear-liquid chromatography (LC) followed by mass spectrometer (LTQ-XL-Orbitrap) to obtain quantitative and qualitative data. A shotgun proteomics allows to compare directly in complex samples, differential expression of proteins and found that in C. Violaceum, 131 proteins are expressed exclusively in the control condition, 177 proteins began to be expressed under oxidative stress and 1175 proteins have expression in both conditions. The results showed that, under the condition of oxidative stress, this bacterium changes its metabolism by increasing the expression of proteins capable of combating oxidative stress and decreasing the expression of proteins related processes bacterial growth and catabolism (transcription, translation, carbon metabolism and fatty acids). A tool with of proteomics as an approach of integrative biology provided an overview of the metabolic pathways involved in the response of C. violaceum to oxidative stress, as well as significantly amplified understanding physiological response to environmental stress. Biochemical and "in silico" assays with the hypothetical ORF CV_0868 found that this is part of an operon. Phylogenetic analysis of superoxide dismutase, protein belonging to the operon also showed that the gene is duplicated in genome of C. violaceum and the second copy was acquired through a horizontal transfer event. Possibly, not only the SOD gene but also all genes comprising this operon were obtained in the same manner. It was concluded that C. violaceum has complex, efficient and versatile mechanisms in oxidative stress response

The main objective of this study was to obtain an effective catalyst for removal of diazo dye - Sudan IV by Catalytic Wet Peroxide Oxidation (CWPO). For this purpose liquid phase treatment was used to increase the basicity of activated carbon surface favoring the adsorption of organic pollutants. Modified activated carbon catalysts were used in different types of experiments: 1) decomposition of H2O2 in aquatic media, 2) decomposition of H2O2 in organic media, 3) adsorption of Sudan IV, 4) Sudan IV removal by CWPO. As the result of all of these experiments the most effective catalyst was obtained and discussed. It was not observed removal of Sudan IV from biphasic system by CWPO. The obtained results in some cases show a slight increase in concentration of Sudan IV, which may be ascribed to experimental errors. Different factors could be the reason of those errors. For example, the high volatility properties of organic media used in experiments should be taken into account during experiments. Under settled reaction temperature the decrease of cyclohexane volume during experiment could give rise in Sudan IV concentration. The initial concentration of model diazo dye also should be reviewed more detailed for CWPO experiments. Despite of these experimental errors the behavior of our catalysts in different media was observed.

En la presente investigación se evaluó la aplicación de los tratamientos de electrocoagulación (EC), Fenton y foto-Fenton para remover compuestos fenólicos presentes en el agua residual de una refinería del petróleo, y cumplir con el nivel regulatorio de descarga establecido por la Agencia de Protección Ambiental (EPA, por sus siglas en inglés). En el proceso de electrocoagulación se evaluaron las variables de tiempo de tratamiento, densidad de corriente, tipo de electrodo, tipo de configuración, posición de los electrodos, pH y agitación, mediante un diseño experimental. Al agua electrocoagulada se le aplicó el tratamiento Fenton, en el cual se evaluaron las variables de concentración de H2O2, Fe2+, y el tiempo de tratamiento. Con el fin de mejorar los porcentajes de mineralización del agua electrocoagulada se aplicó el tratamiento foto-Fenton con luz ultravioleta, en el cual se evaluaron los efectos de las concentraciones de H2O2, Fe2+ y aireación. Conclusiones y contribuciones: Para el tratamiento de electrocoagulación, las mejores condiciones de operación se obtienen bajo las siguientes condiciones: 40 mAcm-2, tiempo de tratamiento de 20 min, pH 7, agitación de 155 rpm, con electrodos orientados horizontalmente en configuración monopolar en paralelo, usando electrodos de hierro; bajo estas condiciones se remueve 52.4% de fenoles totales y 42.3% de COT. La aplicación al agua residual electrocoagulada del tratamiento Fenton, conduce a la degradación de los compuestos fenólicos remanentes del tratamiento de EC a concentraciones menores a 1 ppm y 57.6 % de remoción de COT, siendo las condiciones siguientes las mejores obtenidas: concentración de 19.8 mgL-1 de Fe2+ y 612 mgL-1 de H2O2 en un tiempo de reacción de 15 min a pH 3. Los mejores resultados del tratamiento foto-Fenton aplicado al agua electrocoagulada se obtienen con 306 mgL-1 de H2O2, 19.8 mgL-1 de Fe2+, y con aplicación de aire, alcanzando una remoción de 88% de COT.

Aunque hace más de 50 años que se describió que la glutamato descarboxilasa (GAD) lleva a cabo la descarboxilación del glutamato para producir GABA, y en animales ha sido muy estudiada debido al papel del GABA como neurotransmisor, la información disponible sobre las GADs de plantas es aún limitada, conociéndose sólo algunos aspectos de la regulación por calcio de su actividad enzimática o de expresión de algunos de los genes de su familia génica. El GABA es un metabolito que tradicionalmente se ha asociado a estrés, pero su papel en plantas todavía no está claro. En las últimas dos décadas los resultados experimentales obtenidos sobre la GAD y el GABA, destacando las alteraciones fenotípicas mostradas por plantas tratadas con GABA y por plantas transgénicas para GAD, han generado preguntas interesantes sobre el posible papel de este metabolito y la enzima en señalización en plantas. En plantas, son varios los papeles que se han propuesto para el metabolismo del GABA tales como su participación como componente del metabolismo del carbono y del nitrógeno (Fait y col., 2008), protección frente especies reactivas de oxigeno (Liu y col., 2011), regulación de la expresión génica incluyendo la regulación de genes implicados en la síntesis de hormonas (Khatiresan y col., 1997; Shi y col., 2010; Lancien y Roberts, 2006) y señalización a larga distancia (Beuve y col., 2004) y en gradiente guiando el crecimiento del tubo polínico (Palanivelu y col., 2013). Nuestro grupo de investigación ha sugerido un papel novedoso para la producción de GABA durante la xilogénesis en pino (Molina-Rueda y col., 2010, 2015). En base a estos antecedentes, los objetivos planteados para este trabajo han sido: la asignación de posibles funciones a las GADs de Populus en condiciones normales de crecimiento y en estrés abióticos, estudiar la adquisición del dominio de unión a calmodulina (CaMBD) de las GADs de plantas vasculares y analizar el efecto del GABA y del glutamato en las raíces de Populus. Las conclusiones que se derivan de los resultados de este trabajo se detallan a continuación. El dominio de unión a calmodulina de la GAD de plantas esta conservado en GADs de plantas consideradas ancestros de plantas vasculares y ausente en plantas no vasculares, lo que sitúa juntos en la evolución los eventos de adquisición del dominio de unión a CaM y el desarrollo del tejido vascular de plantas. Los resultados similares de la localización de GABA en xilema y una expresión GAD asociada a la formación de madera de reacción tanto en pino como en chopo apuntan a un papel relevante de la producción de GABA durante la xilogénesis en leñosas. La familia génica GAD posee seis genes codificando todos ellos para proteínas aparentemente funcionales y susceptibles de ser reguladas por calcio. Esta familia génica ha sufrido duplicaciones y eventos de especialización durante la evolución de Populus. Este trabajo ha posibilitado la asociación entre papeles específicos y los diferentes genes de esta familia. Objective: We evaluated the protective activity of an extract from a by-product such as olive stones, through its ability to inhibit H2O2 induced apoptosis in the SH-SY5Y human neuroblastoma cell line. Material and methods: To such end, 20,000 cells/well were cultivated and differentiation with retinoic acid was initiated. Once the cells were differentiated, apoptosis was induced with and without H2O2 extract. Finally, cDNA extraction was performed, and pro-apoptotic genes Bax and anti-apoptotic genes Bcl-2 were analyzed. Quantification of the gene expression was performed using the GAPDH gene marker. Results: Cell viability with the extract is 97.6% (SD 5.7) with 10 mg/l and 62.8% (SD 1.2) to 50 mg/l, using 10 mg/l for the biomarker assay. The retinoic acid differentiated SH-S cell line (10 µM) shows a clear apoptosis when treated with H2O2 150 µM, with a Bax/Bcl-2 ratio of 3.75 (SD 0.80) in contrast to the differentiated control cells subjected to H2O2 and with extract, which have the same ratio of 1.02 (SD 0.01-0.03). Conclusion: The olive stone extract shows anti-apoptotic activity in the provoked cell death of SH-SY5Y human neuroblastoma cells in their normal state, defending them from oxidative stress which produces a significant increase in the apoptotic gene ratio in contrast to anti-apoptotic genes (Bax/Bcl-2).

Sugar esters are substances which possess surfactant, antifungical and bactericidal actions and can be obtained through two renewable sources of raw materials: sugars and vegetable oils. Their excellent biodegradability, allied to lhe fact that they are non toxic, insipid, inodorous, biocompatible, no-ionic, digestible and because they can resist to adverse conditions of temperature, pH and salinity, explain lhe crescent use of these substances in several sections of lhe industry. The objective of this thesis was to synthesize and characterize surfactants and polymers containing sugar branched in their structures, through enzymatic transesterification of vinyl esters and sugars, using alkaline protease from Bacillus subtilis as catalyst, in organic medium (DMF).Three types of sugars were used: L-arabinose, D-glucose and sucrose and two types of vinyl esters: vinyl laurate and vinyl adipate. Aiming to reach high conversions from substrates to products for a possible future large scale industrial production, a serie of variables was optimized, through Design of Experiments (DOE), using Response Surface Methodology (RSM).The investigated variables were: (1) enzyme concentration; (2) molar reason of substrates; (3) water/solvent rale; (4) temperature and (5) time. We obtained six distinct sugar esters: 5-0-lauroyl L-arabinose, 6-0-lauroyl D-glucose, 1'-O-lauroyl sucrose, 5-0-vinyladipoyl L-arabinose, 6-0-vinyladipoyl D-glucose and 1 '-O-vinyladipoyl sucrose, being lhe last three polymerizable. The progress of lhe reaction was monitored by HPLC analysis, through lhe decrease of sugar concentration in comparison to lhe blank. Qualitative analysis by TLC confirmed lhe formation of lhe products. In lhe purification step, two methodologies were adopted: (1) chromatographic column and (2) extraction with hot acetone. The acylation position and lhe chemical structure were determined by 13C-RMN. The polymerization of lhe three vinyl sugar esters was possible, through chemical catalysis, using H2O2 and K2S2O8 as initiators, at 60°C, for 24 hours. IR spectra of lhe monomers and respective polymers were compared revealing lhe disappearance of lhe vinyl group in lhe polymer spectra. The molar weights of lhe polymers were determined by GPC and presented lhe following results: poly (5-0-vinyladipoyl L-arabinose): Mw = 7.2 X 104; PD = 2.48; poly (6-0-vinyladipoyl D-glucose): Mw = 2.7 X 103; PD = 1.75 and poly (1'-O-vinyladipoyl sucrose): Mw = 4.2 X 104; PD = 6.57. The six sugar esters were submitted to superficial tension tests for determination of the critical micelle concentrations (CMC), which varied from 122 to 167 ppm. Finally, a study of applicability of these sugar esters, as lubricants for completion fluids of petroleum wells was' accomplished through comparative analysis of lhe efficiency of these sugar esters, in relation to three commercial lubricants. The products synthesized in this thesis presented equivalent or superior action to lhe tested commercial products

This project describes a methodology optimization that would allow for a more efficient microwave assisted digestion process for petroleum samples. With the possible chance to vary various factors at once to see if any one factor was significant enough in the answers, experimental planning was used. Microwave assisted digestion allows, through the application of potency, an increasing number of collisions between the HNO3 and H2O2 molecules, favoring sample opening for complex matrixes. For this, a 24 factorial experimental planning was used, varying potency, time and the volumes for HNO3 65% and H2O2 30%. To achieve the desired answers, several elements were monitored (C, Cu, Cr, Fe, Ni, Zn and V) through Inductively coupled plasma atomic emission spectroscopy (ICP-OES). With this initial study it was noticed that the HNO3 was not a significant factor for any of the statistical studies for any of the analytes and the other 3 factors and their interactions showed statistical significance. A Box Behnken experimental planning was used taking in consideration 3 factors: H2O2 volume, time (min) and Potency (W), Nitric Acid kept at 4mL for a mass of 0,1g of petroleum. The results were extremely satisfying showing higher efficiency in the digestion process and taking in a responsibility between the answers for each analyte and the carbon monitoring was achieved in the following conditions: 7mL of H2O2, 700 Watts of potency and a reaction time of 7 minutes with 4mL de HNO3 for a mass of 0,1g of petroleum. The optimized digestion process was applied to four different petroleum samples and the analytes determined by ICP-OES
