野大豆和锦鸡儿,同属豆科植物。前者是一年生自交植物,后者是多年生异交植物;一个是大豆的种质资源,另一个是固沙植物;一个具有耐盐适应,另一个能够抗旱。本文首先结合同工酶分析结果,通过随机扩增多态性DNA(RAPD]标记研究了锦鸡儿和野大豆群体的分子生态学特征。然后用限制性内切酶消化了单态的野大豆群体扩增产物以提高RAPD标记检测野大豆DNA多态性的能力。并且根据锦鸡儿群体的RAPD谱估算了每位点上的等位基因频率,分别用Shannon信息指数和Nei指数估测了各群体的遗传变异。最后,在以上研究的基础上结合我们实验室有关锦鸡儿和野大豆的形态,种子蛋白或同工酶方面的分析得到以下结论: 1]在本文的实验条件下,RAPD标记的重复性很好,使有关锦鸡儿和野大豆群体分子生态学的研究结果有了可靠的基础。 2]RAPD标记的显性特征使实验中确定显、隐性等位基因频率困难较大。用估测的显、隐性等位基因频率通过Shannon信息指数估算群体遗传多样性可能更适合于异交植物。 3)用限制性内切酶消化随机扩增产物后,能够提高RAPD标记检测野大豆群体DNA多态性的能力。4]按RAPD多态位点比率排列毛乌素沙地锦鸡儿各群体为:硬粱群体<沙丘群体<硬粱覆沙群体<毛条群体<滩地覆沙群体<软梁覆沙群体。按遗传多样性排列各群体为:硬粱群体<硬梁覆沙群体<沙丘群体<软梁覆沙群体<毛条群体<滩地覆沙群体。反映了RAPD多态位点比率与遗传多样性在检测群体DNA多态性能力上的异同。 5]毛乌素沙地锦鸡儿群体间 存在着强大的基因流,其高度异交性与生态过渡带可能是一致的。 6]根据同工酶、种子蛋白的分析结果,硬粱覆沙群体的基因多样性在毛鸟素沙地锦鸡儿各群体中是最高的。然而,根据DNA多态性的研究结果,硬粱覆沙群体的基因多样性在各个群体中仅大于硬梁群体。反映了锦鸡尔表型分化与基因型分化的差异。7]就本文的研究结果来看,野大豆群体以其高水平的遗传多样性和发育变通性适应着多变的盐环境。8]无论从DNA多态位点比率还是群体的基因多样性来看,异交植物锦鸡儿群体都具有比自交植物野大豆群体较高的水平。除了盐适应的野大豆群体外,一般来讲,锦鸡儿群体间的基因流要明显地大干野大豆的群体间的基因流。

当前分子生物学的方法以惊人的速度渗透到生命科学研究的各个领域。植物对不断变化的环境逐步适应的过程中,积累了丰富的遗传多样性。与此同时,人类活动空间的不断扩大已经严重威胁到其他生命的生存和繁衍,越来越多的物种以越来越快的速度在我们还没有来得及认识它们时就已经永远地消失了。加快物种鉴定和保护的步伐就必须发展更多能充分揭示物种遗传多样性的实验技术,从具有丰富遗传多样性的野生资源中寻找到更多能够服务于人类可持续发展的基因资源。本文以杨树杂交后代过氧化物同工酶和RAPD分析为基础,论证了我们改进的RAPD方法用于遗传分析的可行性。在前期工作的基础上,进一步测定了野大豆自然群体的耐盐性变异,并且用微卫星和RAPD分析的方法研究分子标记与DXA变异、植株耐盐性之间的关系。对四个可能与抗盐性有关的RAPD片段进行克隆、测序,并进行序列比较。由此得出以下结论: 1、在本文的实验条件下,杨树同工酶和RAPD分析均表明,RAPD标记在亲本及其杂交后代中性状比例符合孟德尔遗传规律,尽管有时也会出现遗传负载等机制引起的基因分布扭曲现象。 2、初步研究了个体发育阶段和环境条件对植株耐盐性的影响。结果表明,植物耐盐性不仅仅与外界的盐度有关,而且受发育阶段和其它环境条件(如,温度)的影响。但也发现了某些个体在各种条件下都具有较高的耐盐性,而且,不易受到其它环境条件的影响。 3、微卫星标记的结果表明,10对引物中的8对引物共检测到时17个等位基因,平均每对引物2.125个等位基因。本文的实验条件下,双核苷酸和三核苷酸的引物对扩增产物都没有出现“ghosts"条带或“打滑”现象。 4、有4个RAPD标记可能与野大豆群体的耐盐性有关,分别是OPCO8460bp、OPCO8213bp、OPCO2690bp、以及OPCO5270bp。测序结果与GenBank中的序列作同源性比较,结果显示,OPCO2_(690bp)与小麦、松树等植物的吉普赛性的逆转录转座子的部分区域(24--53)有很高的同源性(86-89%)。此外,OPCO2690bp与栽培大豆胞质谷氨酰胺合成酶(gs15)基因的启动子有高达95%的同源性。 5、本文实验条件下,RAPD扩增产物在限制性内切酶消化后,消化产物的多态性未见增大,也没有发现与耐盐性相关的多态位点。 6、野大豆自然群体DNA变异的研究中也可以应用SWAPP方法。

第一部分:生物安全信息的建设 生物安全( biosafety)是指经过遗传修饰的生物释放到自然界以后对人类健康和生态环境的影响。随着生物技术的发展,转基因生物的大量涌现,其安全性问题越来越引起人们的关注。本文主要介绍了作者在生物安全信息建设的一些工作,建立了生物安全主页和生物安全文献数据库。对主页和数据库的结构和主要内容进行了详细介绍,生物安全主页介绍了全球转基因生物的发展情况,由此带来的一些潜在风险,国内外对生物安全管理的法规和管理机制,以及对风险评估的技术准则和风险的评价指标体系。生物安全文献数据库收录了600多篇有关的文献。 第二部分:转基因棉花根际土壤微生物分子生态学初探 应用分子生物学方法对转基因棉花和对照非转基因棉花根际土壤微生物的多样性进行了一个生长季的跟踪研究,直接从土壤中提取微生物的总DNA,用PCR方法扩增出原核生物核糖体小亚基16S基因和真核生物ITS片段,并对它们进行了克隆和序列测定,共获得80条16S基因全序列和50条ITS段序列,经过分析它们分别属于7个大类的原核微生物和5个大类的真核生物共计33个属。其中,有44个克隆是传统方法所不能分析的不能被培养的微生物种类。应用邻近法分别绘制了系统关系树状图。 这是首次应用全序列分析的方法研究棉花根际土壤微生物的种类组成。为国际核酸序列数据库贡献130条核酸序列,其中有50条是16S rDNA的全序列。在最后对分子生物学方法在微生物生态学中的应用进行了简短讨论。

中国是受沙漠化影响最严重的国家之一。梭梭(Haloxylon ammodendronBunge)是藜科梭梭属多年生小乔木或灌木,作为荒漠地区主要的建群种和优良固沙植物,梭梭在维持荒漠生态系统的结构与功能、防止土地沙漠化、改善小气候方面具有十分重要的作用。近年来,由于不合理地开发利用,大面积的天然梭梭林衰退甚至死亡。然而,我们对于梭梭种群的空间分布和遗传结构所知甚少。本文拟通过对自然梭梭种群的空间格局、遗传结构、遗传结构的空间分布、以及种子种群与地面植被种群的遗传比较的研究,探讨种群的空间结构特征及成因,评价种群遗传多样性水平,了解遗传多样性在种群内、种群间的分布情况以及在空间上的分布格局,预测种群的遗传潜能,并为制定合理的保护、利用策略提供理论依据。 1.对古尔班通古特沙漠东南部地区处于不同演替阶段的四个梭梭种群的分布格局和种群动态进行了考察。结果表明,不同样地中梭梭种群的幼苗(<0.5m)和幼树( 0,5-lm)均呈聚集分布,样地A、B、D中的成熟个体(>1m)也呈聚集分布,样地c中的成熟个体呈随机分布,从整个种群来看,所有梭梭种群均呈聚集分布。种群的龄级结构图表明样地A和c中的梭梭种群表现为稳定型结构,可进行持续更新,虽然样地D中的种群幼苗和幼树数量较少,但这种现象是暂时的,种群仍具有持续更新的能力,样地B中的种群表现为衰退型结构。 2.利用空间自相关分析检测上述四个种群的空间遗传结构,结果表明,四个种群中的成株均无显著的空间自相关。虽然梭梭的种子散布能力有限,但花粉流有可能是空间遗传结构不显著的原因之一;另一个可能的解释是在种群更新和发展过程中,由于种内竞争的增强而发生了自疏现象。 3.利用ISSR标记对新疆和内蒙古境内共9个种群的遗传结构进行研究。8个引物共扩增出219条带,其中184条(84%)具多态性,种群的遗传多样性水平较高,通过AMOVA分析表明大部分遗传多样性分布在种群内,区域间、种群间的遗传变异均很小。梭梭种群较高的遗传多样性水平可能源于对异质、高胁迫环境的长期适应,而种群间遗传分异低的主要原因是种群间存在强大的基因流。 4.对5个梭梭种群的地面植被和种子库的遗传结构进行了比较分析,并调查了其中3个种群地面植被和种子库的空间遗传结构。结果表明,在所有种群中,地面植被和种子库的遗传多样性水平相似,而地面植被的遗传分异高于种子库的遗传分异:二者在遗传结构的空间分布上也不相同,地面植被种群无显著的空间自相关,而种子库种群在第一距离级上( 0-10 m)呈显著正相关。研究结果表明梭梭种群的种子库有能力保持种群的遗传多样性并减弱种群的遗传分化。 5.研究结果建议,由于现存梭梭种群和其潜在种群(种子种群)仍保持了较高的遗传多样性,种群间的基因交换并没有受到阻碍,因此对梭梭的保护可以与生态恢复工作同时进行。在具体工作中,应充分考虑种群的遗传结构特征及其空间格局,选取适合的种群或种子资源进行恢复与重建,并保持一定水平的种群大小,才能实现种群的稳定发展以及在高胁迫环境中的长期存活。 荒漠生态系统植被结构简单,环境压力大,对荒漠地区主要建群种和优势种植物的空间分布和遗传结构进行研究对于探讨在胁迫环境中各种进化力量(如选择、突变、迁移、漂变)的作用机制有重要意义,并为荒漠生态系统的恢复与重建工作提供了有价值的信息。在本文中,对荒漠植物在较大地理范围内的遗传结构研究、通过空间自相关分析进行种群的空间遗传结构研究、地面植被种群与种子库种群在遗传结构、空间遗传结构上的比较研究在国内均为首例。这些研究结果为进一步深入探讨荒漠植物生态适应与进化的分子机制奠定了基础。

Although new empirical evidence shows that sympatric speciation has occurred in some species, there are few indisputable model organisms for this process of speciation. The two subspecies (Gymnocypris eckloni eckloni and G. e. scoliostomus) of the schizothoracine Gymnocypris fish species complex from a small glacier lake in the Tibetan Plateau, Lake Sunmcuo, fit several of the key characteristics of the sympatric speciation model. We used combined mitochondrial control region sequences and the cytochrome b gene (1894 bp) to address the phylogenetics and population genetics of 232 specimens of G. e. eckloni and G. e. scoliostomus, as well as all of its closely related sister species. We found that: (i) a total of four old lineages were uncovered in the widespread G. e. eckloni, of which only one was shown to be shared with all G. e. scoliostomus individuals and (ii) the new subspecies (G. e. scoliostomus) evolved in Lake Sunmcuo from the ancestral G. e. eckloni population within approximately 0.057 Ma. These two taxa of the species complex are morphologically distinct, and reproductive isolation is further suggested. Ecological disruptive selection based on morphological traits (e.g. mouth cleft characters) and food utilization may be a mechanism of incipient speciation of two sympatric populations within Lake Sunmcuo. This study provides the first genetic evidence for sympatric speciation in the schizothoracine fish.

Genetic diversity of the plankton community in Lake Xiliang was depicted by polymerase chain reaction-denaturing gradient gel electrophoresis (PCR-DGGE) fingerprinting. Seventy-seven bands (33 of 16S rDNA and 44 of 18S rDNA) were detected, sixty-two planktonic taxa were identified in six sample stations in November 2007. The most common taxa were Ceratium hirundinella, Bdelloidea, Keratella cochlearis, Polyarthra trigla, and copepod nauplii. Based on environmental factors, taxonomic composition, and PCR-DGGE fingerprinting, unweighted pair-group method using arithmetic averages clustering and principal components analysis were used to analyze habitat similarities. There was distinct spatial heterogeneity in Lake Xiliang, and the genetic diversity of the plankton community was closely related to taxonomic composition and environmental factors.

A (GT)(n) enriched partial genomic library of bighead carp (Aristichthys nobilis) was constructed by employing the (fast isolation by AFLP of sequences containing repeats) FIASCO protocol. Sixteen loci exhibited polymorphism with two to seven alleles/locus (mean 3.263) in a test population and the observed heterozygosity ranging from 0.100 to 0.690 (mean 0.392). Eleven of the 16 bighead carp microsatellites were found to be also polymorphic in silver carp. These polymorphic loci should provide sufficient level of genetic diversity to evaluate population structure of bighead carp.

Largemouth bronze gudgeon (Coreius guichenoti) is a medium-sized fish endemic from the upper Yangtze River of China and its survival is threatened by the construction of the Three Gorges Dam. This study reports 20 new polymorphic microsatellites from a repeat-enriched genomic library with a mean number allele of 5.2, and observed and expected heterozygosities ranging from 0.035 to 1, and from 0.13 to 0.917, respectively. In a cross-species amplification test, nine of the 37 tested loci were found to be also polymorphic in a congeneric species, brass gudgeon (C. heterodon). In addition, other four loci from common carp (Cyprinus carpio) were also polymorphic in C. guichenoti. Out of these 24 polymorphic microsatellites, only three loci significantly deviated from Hardy-Weinberg equilibrium in the sampled population (P < 0.0025), and all pairwise tests for linkage disequilibrium among loci were nonsignificant after applying sequential Bonferroni correction (P > 0.0026). These novel microsatellites provide sufficient levels of polymorphism for studies on population genetics and conservation in C. guichenoti and its related species.

Fifteen polymorphic microsatellite loci were developed for the Chinese soft-shelled turtle (Pelodiscus sinensis) from the (GT)(n) microsatellite-enriched genomic library, using the fast isolation by amplified fragment length polymorphism of sequences containing repeats protocol. The polymorphism of all 15 loci ranged from two to seven alleles with observed heterozygosities ranging from 0.03 to 0.98 (mean 0.43) in one population of 40 individuals. These novel loci will be helpful for understanding the population structure at genetic level and marker-assisted breeding of this vulnerable species.

Wuchang bream (Megalobrama amblycephala) is an economically important fish in China. From a (GT)(13)-enriched genomic library, 20 microsatellites were developed. Nine of these 20 loci were polymorphic in a test population with allele numbers ranging from two to four, and the observed and expected heterozygosities ranging from 0.2609 to 0.7826 and from 0.3739 to 0.7546, respectively. In the cross-species amplifications, six of these nine loci were also polymorphic in white amur bream (Parabramis pekinensis). These polymorphic microsatellite loci are potentially useful for population genetics of Wuchang bream and its closely related species.

The rice field eel (Monopterus albus) is a fish of economic importance in China and some Asian countries. From a (GT)(n)-enriched genomic library, 30 microsatellites were developed by employing the fast isolation by AFLP of sequences containing repeats (FIASCO) protocol. Thirteen loci exhibited polymorphism with two to 13 alleles (mean 7.9 alleles/locus) in a test population and observed heterozygosity ranged from 0.3125 to 0.9688 (mean 0.7140). These loci should provide sufficient level of genetic variation to study the fine-scale population structure and reproductive ecology of the species.

Eighteen microsatellite markers were isolated and characterized using an enrichment protocol in the Chinese mandarin fish Siniperca chuatsi (Basilewsky), a commercially important piscivorous fish in China. Out of 48 pairs of primers designed, 18 loci exhibited polymorphism with three to six alleles (mean 4.4 alleles/locus) and average observed heterozygosity ranged from 0.633 to 0.833 (mean 0.748) in a test population from Dongting Lake of China. Except for two loci, all other 16 loci were in the Hardy-Weinberg equilibrium. These markers would be useful for such studies as population genetics, ecology and selective breeding of the Chinese mandarin fish in future.
