This work deals with a problem of mixed integer optimization model applied to production planning of a real world factory that aims for hydraulic hose production. To optimize production planning, a mathematic model of MILP Mixed Integer Linear Programming, so that, along with the Analytic Hierarchy process method, would be possible to create a hierarchical structure of the most import criteria for production planning, thus finding through a solving software the optimum hose attribution to its respective machine. The hybrid modeling of Analytic Hierarchy Process along with Linear Programming is the focus of this work. The results show that using this method we could unite factory reality and quantitative analysis and had success on improving performance of production planning efficiency regarding product delivery and optimization of the production flow

This study aimed to model a equation for the demand of automobiles and light commercial vehicles, based on the data from February 2007 to July 2014, through a multiple regression analysis. The literature review consists of an information collection of the history of automotive industry, and it has contributed to the understanding of the current crisis that affects this market, which consequence was a large reduction in sales. The model developed was evaluated by a residual analysis and also was used an adhesion test - F test - with a significance level of 5%. In addition, a coefficient of determination (R2) of 0.8159 was determined, indicating that 81.59% of the demand for automobiles and light commercial vehicles can be explained by the regression variables: interest rate, unemployment rate, broad consumer price index (CPI), gross domestic product (GDP) and tax on industrialized products (IPI). Finally, other ten samples, from August 2014 to May 2015, were tested in the model in order to validate its forecasting quality. Finally, a Monte Carlo Simulation was run in order to obtain a distribution of probabilities of future demands. It was observed that the actual demand in the period after the sample was in the range that was most likely to occur, and that the GDP and the CPI are the variable that have the greatest influence on the developed model

Based on a structured literature review, the ceramic tiles sector of Italy (benchmark) and Brazil (2nd world producer and consumer) are compared, under four strategic factors: normative, market, technology and strategic management, in order to identify critical risks for a national strategic sector. The document aims to propose guidelines for a strategic re-planning of the Brazilian ceramic tiles sector, making the Brazilian producers aware of the national market fragility (in spite of its recent remarkable evolution) and helping the policy makers to reflect on the need of reviewing the strategic planning methods and practice, of designing new targeted programs (based on coherence between operation and business strategies), of providing improved management to strengthen the sector against unfair competition by low-cost producers, enhancing the necessary infrastructure in technology, work, marketing and quality management. The analysis is limited to the single-firing production technology. The wide-coverage strategic analysis of the Brazilian ceramic tiles sector, very little studied until now in a scientific way, emphasizes the importance of applying research methodology and may be valuable to both scholars and practitioners. Additionally, it highlights the need of investments in innovation (product design and production technology) and the fundamental role of the sector organization, identifying different dimensions. It is possible to conclude that the recent Brazilian production growth is not due to a natural strengthening because of the hit of the sector and of correct enterprises strategy, but it seems the result of a temporary and favorable economic contingency.

Crown ethers have the ability of solubilizing inorganic salts in apolar solvents and to promote chemical reactions by phase-transfer catalysis. However, details on how crown ethers catalyze ionic S(N)2 reactions and control selectivity are not well understood. In this work, we have used high level theoretical calculations to shed light on the details of phase-transfer catalysis mechanism of KF reaction with alkyl halides promoted by 18-crown-6. A complete analysis of the of the model reaction between KF(18-crown-6) and ethyl bromide reveals that the calculations can accurately predict the product ratio and the overall kinetics. Our results point out the importance of the K* ion and of the crown ether ring in determining product selectivity. While the K* ion favors the S(N)2 over the E2 anti pathway, the crown ether ring favors the S(N)2 over E2 syn route. The combination effects lead to a predicted 94% for the S(N)2 pathway in excellent agreement with the experimental value of 92%. A detailed analysis of the overall mechanism of the reaction under phase-transfer conditions also reveals that the KBr product generated in the nucleophilic fluorination acts as an inhibitor of the 18-crown-6 catalyst and it is responsible for the observed slow reaction rate. Background: Antineoplastic phospholipids (ALPs) represent a promising class of drugs with a novel mode of action undergoes rapid turnover in the cell membrane of tumors, interfering with lipid signal transduction, inducing cell death. The aim of this study was to investigate the synthetic phosphoethanolamine (Pho-s) as a new anticancer agent. Materials and Methods: Cell viability and morphology were assessed by (3-(4,5-Dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide (MTT), Hoechst and rhodamine staining. Apoptosis was assessed by Annexin V and propidium iodide (PI) staining, caspase-3 activity, mitochondrial membrane potential (Delta m psi) and cell cycle analysis, combined with evaluation of tumor growth in Ehrlich Ascites Tumor (EAT) bearing mice. Results: We found that Pho-s 2.30 mg/ml induced cytotoxicity in all tumor cell lines studied without affecting normal cells. In vitro studies with EAT cells indicated that Pho-s induced apoptosis, demonstrated by an increase in Annexin-V positive cells, loss of mitochondrial potential (Delta m psi) and increased caspase-3 activity. It was also shown to increase the sub-G(1) apoptotic fraction and inhibit progression to the S phase of the cell cycle. Additionally, antitumor effects on the EAT-bearing mice showed that Pho-s, at a concentration of 35 and 70 mg/kg, inhibited tumor growth and increased the lifespan of animals without causing liver toxicity. Conclusion: These findings suggest that Pho-s is a potential anticancer candidate drug.

The recent discovery that peroxisome proliferator-activated receptor gamma (PPAR gamma) targeted anti-diabetic drugs function by inhibiting Cdk5-mediated phosphorylation of the receptor has provided a new viewpoint to evaluate and perhaps develop improved insulin-sensitizing agents. Herein we report the development of a novel thiazolidinedione that retains similar anti-diabetic efficacy as rosiglitazone in mice yet does not elicit weight gain or edema, common side effects associated with full PPAR gamma activation. Further characterization of this compound shows GQ-16 to be an effective inhibitor of Cdk5-mediated phosphorylation of PPAR gamma. The structure of GQ-16 bound to PPAR gamma demonstrates that the compound utilizes a binding mode distinct from other reported PPAR gamma ligands, although it does share some structural features with other partial agonists, such as MRL-24 and PA-082, that have similarly been reported to dissociate insulin sensitization from weight gain. Hydrogen/deuterium exchange studies reveal that GQ-16 strongly stabilizes the beta-sheet region of the receptor, presumably explaining the compound's efficacy in inhibiting Cdk5-mediated phosphorylation of Ser-273. Molecular dynamics simulations suggest that the partial agonist activity of GQ-16 results from the compound's weak ability to stabilize helix 12 in its active conformation. Our results suggest that the emerging model, whereby "ideal" PPAR gamma-based therapeutics stabilize the beta-sheet/Ser-273 region and inhibit Cdk5-mediated phosphorylation while minimally invoking adipogenesis and classical agonism, is indeed a valid framework to develop improved PPAR gamma modulators that retain antidiabetic actions while minimizing untoward effects.

E-mail: baccarin@ usp.br Blood-derived products are commonly administered to horses and humans to treat many musculoskeletal diseases, due to their potential antioxidant and anti-inflammatory effects. Nevertheless, antioxidant effects have never been shown upon horse synovial fluid cells in vitro. If proved, this could give a new perspective to justify the clinical application of blood-derived products. The aim of the present study was to investigate the antioxidant effects of two blood-derived products - plasma (unconditioned blood product - UBP) and a commercial blood preparation (conditioned blood product - CBP)(4) - upon stimulated equine synovial fluid cells. Healthy tarsocrural joints (60) were tapped to obtain synovial fluid cells; these cells were pooled, processed, stimulated with lipopolysaccharide (LPS) or phorbol 12-myristate 13-acetate (PMA), and evaluated by flow cytometry for the production of reactive oxygen species (ROS). Upon addition of any blood-derived product here used - UBP and CBP - there was a significant decrease in the oxidative burst of synovial fluid cells (P<0.05). There was no difference between UBP and CBP effects. In conclusion, treatment of stimulated equine synovial cells with either UBP or CBP efficiently restored their redox equilibrium.

A modularidade pode ser entendida como um conjunto de conceitos que leva a novas possibilidades de arquitetura de produto, bem como novas maneiras de organização da produção, possibilitando a transferência de atividades ao longo da cadeia de suprimentos automotiva. Nesse contexto, o objetivo deste trabalho é investigar o emprego da modularidade no projeto de produto e na produção, considerando uma situação de relação específica entre uma montadora de veículos comerciais e fornecedores de motores, no âmbito do consórcio modular. É também analisada a relação entre a modularidade no projeto de produto e na produção. Para a consecução desses objetivos é adotada a abordagem metodológica de estudo de caso. Os resultados apresentam algumas influências que ocorrem entre o projeto do produto e a produção e vice-versa, uma parte delas esperadas, reforçando o caráter confirmatório dos dados coletados. Constata-se que o conceito de modularidade pode ser aplicado ao processo produtivo sem necessariamente que o produto tenha sido concebido em módulos, ou seja, a partir de um produto existente é possível montar seus componentes formando subconjuntos em uma lógica modular. Os dados confirmam ainda que a estrutura do produto de um veículo comercial favorece a sua divisão em módulos. Adicionalmente, a gestão da cadeia de suprimentos na montadora é simplificada em comparação ao sistema de produção convencional, pois parte das responsabilidades da montadora é transferida para os fornecedores.

Cada empresa dispone de un espacio web único e individual para poder promocionarse en internet. Mediante un buscador, permite a los usuarios acceder a los contenidos de cada empresa registrada en el sistema. Este buscador es abierto y cualquier usuario no registrado puede consultar la información acerca de productos y servicios ofertados, y disponer de toda la información relacionada con cada empresa. Las empresas registradas disponen de un sistema de información completo de fácil manejo e intuitivo que permite autogestionar todo el contenido de los productos y páginas web de cada empresa individualmente. También se incluye un sistema de gestión de contenidos que genera páginas web profesionales automáticamente, con posibilidad de edición de páginas. Por otra parte, los usuarios registrados podrán realizar: reservas de productos mediante un completo sistema de gestión de reservas, con especial atención al alojamiento, compras de productos mediante un completo sistema de compras, adaptado a la plataforma Paypal, clasificaciones de productos y páginas web del sistema, utilizando votaciones mediante rankings. La plataforma contiene un sistema de gestión de comentarios sobre productos y páginas web de empresas que permite seleccionar la visualización y la no visualización del contenido. Por último, los usuarios podrán compartir información sobre contenidos publicados en las páginas, mediante el uso de redes sociales como Twitter, Google+ y Facebook.

Combustion control is one of the key factors to obtain better performances and lower pollutant emissions for diesel, spark ignition and HCCI engines. An algorithm that allows estimating, as an example, the mean indicated torque for each cylinder, could be easily used in control strategies, in order to carry out cylinders trade-off, control the cycle to cycle variation, or detect misfires. A tool that allows evaluating the 50% of Mass Fraction Burned (MFB50), or the net Cumulative Heat Release (CHRNET), or the ROHR peak value (Rate of Heat Release), could be used to optimize spark advance or to detect knock in gasoline engines and to optimize injection pattern in diesel engines. Modern management systems are based on the control of the mean indicated torque produced by the engine: they need a real or virtual sensor in order to compare the measured value with the target one. Many studies have been performed in order to obtain an accurate and reliable over time torque estimation. The aim of this PhD activity was to develop two different algorithms: the first one is based on the instantaneous engine speed fluctuations measurement. The speed signal is picked up directly from the sensor facing the toothed wheel mounted on the engine for other control purposes. The engine speed fluctuation amplitudes depend on the combustion and on the amount of torque delivered by each cylinder. The second algorithm processes in-cylinder pressure signals in the angular domain. In this case a crankshaft encoder is not necessary, because the angular reference can be obtained using a standard sensor wheel. The results obtained with these two methodologies are compared in order to evaluate which one is suitable for on board applications, depending on the accuracy required.

Lo vendemmia meccanica incontra ancora resistenze legate al timore di peggiorare la qualità del prodotto e di avere elevate perdite di raccolta. In questo contesto sono state effettuate quattro prove sperimentali, finalizzate a definire le interazioni macchina, pianta e prodotto raccolto e a valutare nuove possibilità di regolazione delle vendemmiatrici e di gestione del prodotto raccolto. Le prime due sono state realizzate con vendemmiatrici a scuotimento orizzontale e verticale. L’obiettivo è stato quello di individuare l’influenza della frequenza del battitore sull’efficienza di raccolta e sulla qualità del prodotto e di verificare il maltrattamento provocato dagli organi di intercettazione e trasporto della vendemmiatrice. I risultati hanno dimostrato l’importanza della corretta regolazione del battitore delle vendemmiatrici a scuotimento orizzontale che operano direttamente sulla fascia produttiva del vigneto. Questa regolazione risulta più semplice sulle macchine a scuotimento verticale che agiscono indirettamente sui fili di sostegno delle doppie cortine. La misura delle sollecitazioni all’interno della macchina ha evidenziato valori anche elevati, pericolosi per l’integrità del prodotto raccolto, legati alla differente costruzione degli organi d’intercettazione e trasporto. La terza prova ha valutato l’efficacia di due nuovi accessori per le vendemmiatrici: la regolazione dell’ampiezza del battitore e un sensore per misurare in continuo il grado di ammostamento provocato. I risultati hanno dimostrato la loro validità per migliorare le prestazioni operative delle vendemmiatrici e per fornire agli operatori uno strumento di controllo in tempo reale sulla qualità della raccolta. Infine, considerando che le vendemmiatrici producono sempre un ammostamento dell’uva, abbiamo verificato un sistema innovativo che permette di anticipare la protezione del mosto libero già durante il trasporto dal campo. Il sistema si è dimostrato semplice, efficace ed economico. Queste esperienze hanno dimostrano che la vendemmia meccanica, se correttamente gestita, permette di ottenere ottimi risultati sotto il profilo qualitativo, tecnologico ed economico.

The needs of customers to improve machinery in recent years have driven tractor manufacturers to reduce product life and development costs. The most significant efforts have concentrated on the attempt to decrease the costs of the experimental testing sector. The validation of the tractor prototypes are presently performed with a replication of a particularly unfavourable condition a defined number of times. These laboratory tests do not always faithfully reproduce the real use of the tractor. Therefore, field tests are also carried out to evaluate the prototype during real use, but it is difficult to perform such tests for a period of time long enough to reproduce tractor life usage. In this context, accelerated tests have been introduced in the automotive sector, producing a certain damage to the structure in a reduced amount of time. The goal of this paper is to define a methodology for the realization of accelerated structural tests on a tractor, through the reproduction of real customer tractor usage. A market analysis was performed on a 80 kW power tractor and a series of measures were then taken to simulate the real use of the tractor. Subsequently, the rainflow matrixes of the signals were extrapolated and used to estimate the tractor loadings for 10 years of tractor life. Finally these loadings were reproduced on testing grounds with special road pavements. The results obtained highlight the possibility of reproducing field loadings during road driving on proving grounds (PGs), but the use of two field operations is also necessary. The global acceleration factor obtained in this first step of the methodology is equal to three.

Il tema della ricerca ha riguardato preliminarmente la definizione di farmaco descritta nel d.lgs. n. 219 del 2006 (Codice dei farmaci per uso umano). Oltre al danno prodotto da farmaci, la ricerca ha approfondito anche la tutela ex ante ed ex post riguardante la produzione di dispositivi medici (Direttiva della Comunità Economica Europea n. 42 del 1993 e Direttiva della Comunità Economica Europea n. 374 del 1985). E’ stato necessario soffermarsi sull’analisi del concetto di precauzione per il quale nell’ambito di attività produttive, come quelle che cagionano inquinamento ambientale, o “pericolose per la salute umana” come quelle riguardanti la produzione di alimenti e farmaci, è necessario eliminare i rischi non conosciuti nella produzione di questi ultimi al fine di garantire una tutela completa della salute. L’analisi della Direttiva della Comunità Economica Europea n. 374 del 1985 nei suoi aspetti innovativi ha riguardato l’esame dei casi di danno da farmaco (Trib. Roma, 20 Giugno 2002, Trib. Roma 27 Giugno 1987, Trib. Milano 19 Novembre 1987, Cassazione Civile n. 6241 del 1987): profilo critico è quello riguardante la prova liberatoria, mentre l'art. 2050 prevede che «si debbano adottare tutte le misure necessarie per evitare il danno», l'art. 118, lett. e), c. cons.) prevede una serie di casi di esonero di responsabilità del produttore (tra cui il rischio di sviluppo). Dall'analisi emerge poi la necessità da parte del produttore di continuo utilizzo del duty to warn (Art. 117 del Codice del Consumo lett. A e B ): esso consiste nel dovere continuo di informazione del produttore tramite i suoi rappresentanti e il bugiardino presente nelle confezioni dei farmaci. Tale dovere è ancor più importante nel caso della farmacogenetica, infatti, al fine di evitare reazioni avverse nel bugiardino di alcuni farmaci verrà prescritta la necessità di effettuare un test genetico prima dell’assunzione.

“Difetto di Informazione e danno da prodotto” è un tema di grande interesse ed attualità, in un momento storico in cui si è riacceso il dibattito sulla disciplina della responsabilità per danni da prodotto difettoso. Complice è “il difetto di informazione”, da sempre rimasto ai margini della casistica giurisprudenziale, ma che improvvisamente “ruba la scena” imponendosi all'attenzione della Cassazione (nn. 6007/2007 e 20985/2007) e rivelando nodi interpretativi di forte impatto pratico nelle cause di responsabilità del produttore. Di qui l’esigenza di approfondire la complessa tematica degli information defects, sotto il profilo della nozione di “difetto” e della sua prova, nonché dell’incidenza di eventuali carenze informative sulla responsabilità del produttore. Muovendo dall’analisi della Direttiva e della sua attuazione italiana, il lavoro individua i punti nodali di tale disciplina ed i vari limiti posti – sia a livello interpretativo che applicativo - alla tutela del danneggiato, suscettibili di renderla meno “appetibile” di quanto potrebbe sembrare. Affronta, quindi, criticamente le questioni trattate nelle due sentenze di legittimità del 2007 in ordine sia alla distribuzione tra le parti dell'onere probatorio, sia alla rilevanza delle informazioni fornite dal produttore. Peculiare interrogativo cui si cerca di dare risposta è se, ed in che misura, l’“informazione” consenta al produttore di andare esente da responsabilità. Il lavoro passa ad esplorare i nuovi scenari offerti dalla interazione tra la disciplina della responsabilità del produttore e quella sulla sicurezza generale dei prodotti, la cui coesistenza all’interno del medesimo corpo di norme pone nuovi interrogativi, quali la sussistenza della responsabilità del produttore di un bene “conforme” ed il ruolo dell’informazione nell’ipotesi di prodotti non difettosi e/o “conformi”. La ricerca affronta tali interrogativi con sguardo critico e provocatorio sollevando il dubbio se la “conformità alle norme armonizzate” non rischi di diventare una “nuova” clausola di esonero della responsabilità del produttore.
