Vortex-induced motion (VIM) is a specific way for naming the vortex-induced vibration (VIV) acting on floating units. The VIM phenomenon can occur in monocolumn production, storage and offloading system (MPSO) and spar platforms, structures presenting aspect ratio lower than 4 and unity mass ratio, i.e., structural mass equal to the displaced fluid mass. These platforms can experience motion amplitudes of approximately their characteristic diameters, and therefore, the fatigue life of mooring lines and risers can be greatly affected. Two degrees-of-freedom VIV model tests based on cylinders with low aspect ratio and small mass ratio have been carried out at the recirculating water channel facility available at NDF-EPUSP in order to better understand this hydro-elastic phenomenon. The tests have considered three circular cylinders of mass ratio equal to one and different aspect ratios, respectively L/D = 1.0, 1.7, and 2.0, as well as a fourth cylinder of mass ratio equal to 2.62 and aspect ratio of 2.0. The Reynolds number covered the range from 10 000 to 50 000, corresponding to reduced velocities from 1 to approximately 12. The results of amplitude and frequency in the transverse and in-line directions were analyzed by means of the Hilbert-Huang transform method (HHT) and then compared to those obtained from works found in the literature. The comparisons have shown similar maxima amplitudes for all aspect ratios and small mass ratio, featuring a decrease as the aspect ratio decreases. Moreover, some changes in the Strouhal number have been indirectly observed as a consequence of the decrease in the aspect ratio. In conclusion, it is shown that comparing results of small-scale platforms with those from bare cylinders, all of them presenting low aspect ratio and small mass ratio, the laboratory experiments may well be used in practical investigation, including those concerning the VIM phenomenon acting on platforms. Abstract Background Vaccination of neonates is generally difficult due to the immaturity of the immune system and consequent higher susceptibility to tolerance induction. Genetic immunization has been described as an alternative to trigger a stronger immune response in neonates, including significant Th1 polarization. In this investigation we analysed the potential use of a genetic vaccine containing the heat shock protein (hsp65) from Mycobacterium leprae (pVAXhsp65) against tuberculosis (TB) in neonate mice. Aspects as antigen production, genomic integration and immunogenicity were evaluated. Methods Hsp65 message and genomic integration were evaluated by RT-PCR and Southern blot, respectively. Immunogenicity of pVAXhsp65 alone or combined with BCG was analysed by specific induction of antibodies and cytokines, both quantified by ELISA. Results This DNA vaccine was transcribed by muscular cells of neonate mice without integration into the cellular genome. Even though this vaccine was not strongly immunogenic when entirely administered (three doses) during early animal's life, it was not tolerogenic. In addition, pVAXhsp65 and BCG were equally able to prime newborn mice for a strong and mixed immune response (Th1 + Th2) to pVAXhsp65 boosters administered later, at the adult life. Conclusion These results suggest that pVAXhsp65 can be safely used as a priming stimulus in neonate animals in prime-boost similar strategies to control TB. However, priming with BCG or pVAXhsp65, directed the ensuing immune response triggered by an heterologous or homologous booster, to a mixed Th1/Th2 pattern of response. Measures as introduction of IL-12 or GM-CSF genes in the vaccine construct or even IL-4 neutralization, are probably required to increase the priming towards Th1 polarization to ensure control of tuberculosis infection.

Este artigo busca analisar o princípio da insignificância em matéria penal, e refletir acerca da melhor forma de operacionalizá-lo dogmaticamente. Após uma introdução que trata brevemente do contexto do surgimento desse princípio, o texto apresenta as principais concepções utilizadas pela doutrina e pela jurisprudência nacionais para aplicá-lo, com o objetivo de refletir sobre as vantagens e desvantagens sistemáticas e político-criminais inscritas em cada uma delas. Nesse plano, o artigo problematiza a principal formulação defendida pela doutrina nacional - segundo a qual a insignificância constitui espécie de cláusula de exclusão da "tipicidade material" da conduta analisada -, sugerindo que essa concepção pode estar na base de sérios equívocos cometidos por nossos tribunais no momento de definir se uma conduta é ou não penalmente insignificante. Ao final, o texto aponta os contornos gerais de uma formulação dogmática mais adequada para esse princípio, visando suprir as diversas dificuldades advindas da adoção da concepção majoritária sobre a matéria no Brasil.

Nerve-related complications have been frequently reported in dental procedures, and a very frequent type of occurrence involves the inferior alveolar nerve (IAN). The nerve injury in humans often results in persistent pain accompanied by allodynia and hyperalgesia. In this investigation, we used an experimental IAN injury in rats, which was induced by a Crile hemostatic clamp, to evaluate the effects of laser therapy on nerve repair. We also studied the nociceptive behavior (von Frey hair test) before and after the injury and the behavioral effects of treatment with laser therapy (emitting a wavelength of 904 nm, output power of 70 Wpk, a spot area of *0.1 cm2, frequency of 9500 Hz, pulse time 60 ns and an energy density of 6 J/cm2). As neurotrophins are essential for the process of nerve regeneration, we used immunoblotting techniques to preliminarily examine the effects of laser therapy on the expression of nerve growth factor (NGF) and brainderived neurotrophic factor (BDNF). The injured animals treated with laser exhibited an improved nociceptive behavior. In irradiated animals, there was an enhanced expression of NGF (53%) and a decreased BDNF expression (40%) after laser therapy. These results indicate that BDNF plays a locally crucial role in pain-related behavior development after IAN injury, increasing after lesions (in parallel to the installation of pain behavior) and decreasing with laser therapy (in parallel to the improvement of pain behavior). On the other hand, NGF probably contributes to the repair of nerve tissue, in addition to improving the pain-related behavior.

Bifidobacteria constitute up to 3% of the total microbiota and represent one of the most important healthpromoting bacterial groups of the human intestinal microflora. The presence of Bifidobacterium in the human gastrointestinal tract has been directly related to several health-promoting activities; however, to date, no information about the specific mechanisms of interaction with the host is available. The first health-promoting activities studied in these job was the oxalate-degrading activity. Oxalic acid occurs extensively in nature and plays diverse roles, especially in pathological processes. Due to its highly oxidizing effects, hyper absorption or abnormal synthesis of oxalate can cause serious acute disorders in mammals and be lethal in extreme cases. Intestinal oxalate-degrading bacteria could therefore be pivotal in maintaining oxalate homeostasis, reducing the risk of kidney stone development. In this study, the oxalate-degrading activity of 14 bifidobacterial strains was measured by a capillary electrophoresis technique. The oxc gene, encoding oxalyl-CoA decarboxylase, a key enzyme in oxalate catabolism, was isolated by probing a genomic library of B. animalis subsp. lactis BI07, which was one of the most active strains in the preliminary screening. The genetic and transcriptional organization of oxc flanking regions was determined, unravelling the presence of other two independently transcribed open reading frames, potentially responsible for B. animalis subsp. lactis ability to degrade oxalate. Transcriptional analysis, using real-time quantitative reverse transcription PCR, revealed that these genes were highly induced in cells first adapted to subinhibitory concentrations of oxalate and then exposed to pH 4.5. Acidic conditions were also a prerequisite for a significant oxalate degradation rate, which dramatically increased in oxalate pre-adapted cells, as demonstrated in fermentation experiments with different pH-controlled batch cultures. These findings provide new insights in the characterization of oxalate-degrading probiotic bacteria and may support the use of B. animalis subsp. lactis as a promising adjunct for the prophylaxis and management of oxalate-related kidney disease. In order to provide some insight into the molecular mechanisms involved in the interaction with the host, in the second part of the job, we investigated whether Bifidobacterium was able to capture human plasminogen on the cell surface. The binding of human plasminogen to Bifidobacterium was dependent on lysine residues of surface protein receptors. By using a proteomic approach, we identified six putative plasminogen-binding proteins in the cell wall fraction of three strain of Bifidobacterium. The data suggest that plasminogen binding to Bifidobactrium is due to the concerted action of a number of proteins located on the bacterial cell surface, some of which are highly conserved cytoplasmic proteins which have other essential cellular functions. Our findings represent a step forward in understanding the mechanisms involved in the Bifidobacterium-host interaction. In these job w studied a new approach based on to MALDI-TOF MS to measure the interaction between entire bacterial cells and host molecular target. MALDI-TOF (Matrix Assisted Laser Desorption Ionization-Time of Flight)—mass spectrometry has been applied, for the first time, in the investigation of whole Bifidobacterium cells-host target proteins interaction. In particular, by means of this technique, a dose dependent human plasminogen-binding activity has been shown for Bifidobacterium. The involvement of lysine binding sites on the bacterial cell surface has been proved. The obtained result was found to be consistent with that from well-established standard methodologies, thus the proposed MALDI-TOF approach has the potential to enter as a fast alternative method in the field of biorecognition studies involving in bacterial cells and proteins of human origin.

Das Ziel der vorliegenden Arbeit ist die Untersuchung der räumlichen und zeitlichen Aspekte der heterogenen Dynamik in Modellglasbildnern. Dabei wird vor allem die langsame alpha-Relaxationsdynamik oberhalb des Glasüberganges Tg untersucht. Die nukleare Magnetresonanz zeigt ihre einmalige Vielseitigkeit bei der Untersuchung molekularer Dynamik, wenn die angewandten Techniken und Experimente durch Simulationen unterstützt werden. Die räumliche Aspekt dynamischer Heterogenitäten wird untersucht durch ein reduziertes vierdimensionales Spindiffusionsexperiment (4D3CP), ein Experiment, das Reorientierungsraten örtlich korreliert. Eine Simulation dieses Experimentes an einem System harter Kugeln liefert wertvolle Informationen über die Auswertemethode des 4D3CP Experiments. Glycerol und o-terphenyl werden durch das 4D3CP Experiment untersucht. Die erhaltenen Resultate werden mit bereits publizierten Daten des polymeren Glasbildners PVAc verglichen. Während PVAc und o-terphenyl eine Längenskale von 3.7 nm bzw. 2.9 nm aufweisen, ist die Längenskale von Glycerol signifikant kleiner bei 1.1 nm. Ein neues Experiment, welches sensitiv auf Translationsbewegung reagiert, wird vorgestellt. Durch Verwendung eines pi-Impulszuges kann eine separate Evolution unter dem Hamiltonian der dipolaren Kopplung und der chemischen Verschiebungsanisotropie erreicht werden.

In this investigation I look at patents and software agents as a way to study broader relation between law and science (the latter term broadly understood as inclusive of science and technology). The overall premise framing the entire discussion, my basic thesis, is that this relation, between law and science, cannot be understood without taking into account a number of intervening factors identifying which makes it necessary to approach the question from the standpoint of fields and disciplines other than law and science themselves.

Im Rahmen dieser Arbeit wurde die zeitaufgelöste Photoemissions Elektronenmikroskopie (TR-PEEM) für die in-situ Untersuchung ultraschneller dynamischer Prozesse in dünnen mikrostrukturierten magnetischen Schichten während eines rasch verändernden externen Magnetfelds entwickelt. Das Experiment basiert auf der Nutzung des XMCD-Kontrasts (X-ray magnetic circular dichroism) mit Hilfe des zirkularpolarisierten Lichts von Synchrotronstrahlungsquellen (Elektronenspeicherringen BESSY II (Berlin) und ESRF (Grenoble)) für die dynamische Darstellung der magnetischen Domänen während ultraschneller Magnetisierungsvorgänge. Die hier entwickelte Methode wurde als erfolgreiche Kombination aus einer hohen Orts- und Zeitauflösung (weniger als 55 nm bzw. 15 ps) realisiert. Mit der hier beschriebenen Methode konnte nachgewiesen werden, dass die Magnetisierungsdynamik in großen Permalloy-Mikrostrukturen (40 µm x 80 µm und 20 µm x 80 µm, 40 nm dick) durch inkohärente Drehung der Magnetisierung und mit der Bildung von zeitlich abhängigen Übergangsdomänen einher geht, die den Ummagnetisierungsvorgang blockieren. Es wurden neue markante Differenzen zwischen der magnetischen Response einer vorgegebenen Dünnfilm-Mikrostruktur auf ein gepulstes externes Magnetfeld im Vergleich zu dem quasi-statischen Fall gefunden. Dies betrifft die Erscheinung von transienten raumzeitlichen Domänenmustern und besonderen Detailstrukturen in diesen Mustern, welche im quasi-statischen Fall nicht auftreten. Es wurden Beispiele solcher Domänenmuster in Permalloy-Mikrostrukturen verschiedener Formen und Größen untersucht und diskutiert. Insbesondere wurde die schnelle Verbreiterung von Domänenwänden infolge des präzessionalen Magnetisierungsvorgangs, die Ausbildung von transienten Domänenwänden und transienten Vortizes sowie die Erscheinung einer gestreiften Domänenphase aufgrund der inkohärenten Drehung der Magnetisierung diskutiert. Ferner wurde die Methode für die Untersuchung von stehenden Spinwellen auf ultradünnen (16 µm x 32 µm groß und 10 nm dick) Permalloy-Mikrostrukturen herangezogen. In einer zum periodischen Anregungsfeld senkrecht orientierten rechteckigen Mikrostruktur wurde ein induziertes magnetisches Moment gefunden. Dieses Phänomen wurde als „selbstfangende“ Spinwellenmode interpretiert. Es wurde gezeigt, dass sich eine erzwungene Normalmode durch Verschiebung einer 180°-Néelwand stabilisiert. Wird das System knapp unterhalb seiner Resonanzfrequenz angeregt, passt sich die Magnetisierungsverteilung derart an, dass ein möglichst großer Teil der durch das Anregungsfeld eingebrachten Energie im System verbleibt. Über einem bestimmten Grenzwert verursacht die Spinwellenmode nahe der Resonanzfrequenz eine effektive Kraft senkrecht zur 180°-Néel-Wand. Diese entsteht im Zentrum der Mikrostruktur und wird durch die streufeldinduzierte Kraft kompensiert. Als zusätzliche Möglichkeit wurden die Streufelder von magnetischen Mikrostrukturen während der dynamischen Prozesse quantitativ bestimmt und das genaue zeitliche Profil des Streufelds untersucht. Es wurde gezeigt, dass das zeitaufgelöste Photoemissions Elektronenmikroskop als ultraschnelles oberflächensensitives Magnetometer eingesetzt werden kann.

The weathering of Fe-bearing minerals under extraterrestrial conditions was investigated by Mössbauer (MB) spectroscopy to gain insights into the role of water on the planet Mars. The NASA Mars Exploration Rovers Spirit and Opportunity each carry a miniaturized Mössbauer spectrometer MIMOS II for the in situ investigation of Martian soils and rocks as part of their payload. The MER flight instruments had to be modified in order to work over the Martian diurnal temperature range (180 K – 290 K) and within the unique electronic environment of the rovers. The modification required special calibration procedures. The integration time necessary to obtain a good quality Mössbauer spectrum with the MIMOS II flight instruments was reduced by 30 % through the design of a new collimator. The in situ investigation of rocks along the rover Spirit's traverse in Gusev crater revealed weakly altered olivine basalt on the plains and pervasively altered basalt in the Columbia Hills. Correlation plots of primary Fe-bearing minerals identified by MB spectroscopy such as olivine versus secondary Fe-bearing phases such as nanophase Fe oxides showed that olivine is the mineral which is primarily involved in weathering reactions. This argues for a reduced availability of water. Identification of the Fe-oxyhydroxide goethite in the Columbia Hills is unequivocal evidence for aqueous weathering processes in the Columbia Hills. Experiments in which mineral powders were exposed to components of the Martian atmosphere showed that interaction with the atmosphere alone, in the absence of liquid water, is sufficient to oxidize Martian surface materials. The fine-grained dust suspended in the Martian atmosphere may have been altered solely by gas-solid reactions. Fresh and altered specimens of Martian meteorites were investigated with MIMOS II. The study of Martian meteorites in the lab helped to identify in Bounce Rock the first rock on Mars which is similar in composition to basaltic shergottites, a subgroup of the Martian meteorites. The field of astrobiology includes the study of the origin, evolution and distribution of life in the universe. Water is a prerequisite for life. The MER Mössbauer spectrometers identified aqueous minerals such as jarosite and goethite. The identification of jarosite was crucial to evaluate the habitability of Opportunity's landing site at Meridiani Planum during the formation of the sedimentary outcrop rocks, because jarosite puts strong constrains on pH levels. The identification of olivine in rocks and soils on the Gusev crater plains provide evidence for the sparsity of water under current conditions on Mars. Ratios of Fe2+/Fe3+ were obtained with Mössbauer spectroscopy from basaltic glass samples which were exposed at a deep sea hydrothermal vent. The ratios were used as a measure of potential energy for use by a microbial community. Samples from Mars analogue field sites on Earth exhibiting morphological biosignatures were also investigated.

Innerhalb dieser Dissertation wurde zwischen den Jahren 2002 und 2005 mit Hilfe von Barberfallen die Laufkäferfauna der Auwälder am nördlichen Oberrhein zwischen Mainz und Bingen erfasst. Dabei dienten verschiedene Rheininseln und ufernahe Festlandgebiete als Probeflächen. Fünf der typischen Bewohner dieser Flächen (Agonum afrum, Nebria brevicollis, Oxypselaphus obscurus, Platynus assimilis, Pterostichus anthracinus) dienten weiterhin als Modellarten für die Untersuchung der genetischen Diversität zwischen den einzelnen Populationen mittels RAPD-Analysen. Alles in allem konnten im Untersuchungsgebiet über 20.000 Individuen aus 101 Carabidenarten gefangen werden. Die häufigsten Vertreter waren Platynus assimilis, Pterostichus melanarius und Agonum afrum. Hohe Diversitäts- und Dominanzindices auf allen Flächen sprechen für die Dynamik des Lebensraumes und somit die Intaktheit der untersuchten Auwälder. Einen weiteren Hinweis auf die ständig wechselnden Lebensbedingungen durch immer wiederkehrende Überflutungen zeigt das Auftreten verschiedener ökologischer Gruppen. Überall dominierten deutlich die Arten, die mit gewissen Störungen des Habitates auskommen oder durch ihr hohes Ausbreitungspotential davor fliehen können. Das sind die Imaginalüberwinterer, makropteren, hygrophilen und kleinen Spezies. Auch das Geschlechterverhältnis weist auf deutliche Anzeichen für regelmäßige Beeinträchtigungen der Flächen hin. Knapp die Hälfte der beobachteten Arten im Untersuchungsgebiet steht auf einer der Roten Listen von Deutschland, Rheinland-Pfalz oder Hessen. Somit besteht für das gesamte Gebiet ein hoher Schutzbedarf. Das Hauptaugenmerk dieser Arbeit lag bei den Einflüssen der Hochwasserstände auf die Artenzusammensetzungen und die genetischen Diversitäten der Laufkäferpopulationen. Deshalb untersuchte man auch die Wirkung derjenigen Faktoren, die ihrerseits unmittelbar von den Extremwasserständen beeinflusst werden. Hier sind vor allem der Auentyp (Weichholz/Hartholz) und die Lage der Flächen auf Insel oder Festland zu nennen, die die deutlichsten Unterschiede in Artendiversität und Genetik der einzelnen Populationen zeigten. Aber auch weitere Faktoren, wie Wasserstandsdynamik, Auwaldbreite, Entfernung vom Fluss und Lage zum Damm weisen Zusammenhänge mit bestimmten ökologischen Gruppen auf. Lediglich die Habitatgröße scheint keinen Einfluss auf die Diversitäten zu nehmen. Abschließend konnten auch für das Jahr 2003, in dem extrem heiße und trockene Bedingungen herrschten, negative Effekte auf die Laufkäfergemeinschaften gezeigt werden.

A novel screening platform for potential retroviral fusion inhibitors on the basis of fully functional membrane‐anchored coiled coil lipopeptide receptors has been established. The work comprises the scrutiny of lateral organization of functional lipids in phase separated bilayers and an in‐depth investigation of the biophysical properties of lipopeptide‐based receptors. Lateral sorting of lipids was detected by the recognition of streptavidin of biotinylated lipids in phase separated bilayers and by nanoscopic patterns in mixed fluorocarbon / hydrocarbon lipid bilayers, employing temperature controlled atomic force microscopy (AFM) as a versatile characterization method. Particular features of fluorocarbon bilayers were additionally investigated in great detail by means of ellipsometry and ATR‐IR spectroscopy. Lipopeptide‐receptors were synthesized on the basis of a robust and reliable in situ coupling reaction by coupling terminal cysteine modified receptor‐peptides to a maleimide functionalized lipid bilayer. Receptor functionality of the lipopeptides was visualized by specific binding of vesicles and nanoparticles tracked by a multiplicity of characterization methods, such as AFM, ellipsometry, CLSM and fluorescence spectroscopy. Finally, in situ coupling of viral peptides, originating from the fusion protein of HIV resulted in a mimic of the pre‐hairpin intermediate of gp41. Structural analysis of N36‐lipopepides by means of CD‐spectroscopy in combination with FT‐IR spectroscopy revealed a coiled coil assembly of lipopeptides, which render the aggregates fully functional receptors for potent fusion inhibitors. Thereby, reversible inhibitor binding of T20 and the corresponding C‐ peptides was detected by AFM and ellipsometry, rendering coiled coil lipopeptides a new promising technique for screening of retroviral fusion inhibitors.

Il progetto di ricerca si situa nell’ambito dell’informatica giudiziaria settore che studia i sistemi informativi implementati negli uffici giudiziari allo scopo di migliorare l’efficienza del servizio, fornire una leva per la riduzione dei lunghi tempi processuali, al fine ultimo di garantire al meglio i diritti riconosciuti ai cittadini e accrescere la competitività del Paese. Oggetto di studio specifico del progetto di ricerca è l’utilizzo delle ICT nel processo penale. Si tratta di una realtà meno studiata rispetto al processo civile, eppure la crisi di efficienza del processo non è meno sentita in tale area: l’arretrato da smaltire al 30 giugno del 2011 è stato quantificato in 3,4 milioni di processi penali, e il tempo medio di definizione degli stessi è di quattro anni e nove mesi. Guardare al processo penale con gli occhi della progettazione dei sistemi informativi è vedere un fluire ininterrotto di informazioni che include realtà collocate a monte e a valle del processo stesso: dalla trasmissione della notizia di reato alla esecuzione della pena. In questa prospettiva diventa evidente l’importanza di una corretta gestione delle informazioni: la quantità, l’accuratezza, la rapidità di accesso alle stesse sono fattori così cruciali per il processo penale che l’efficienza del sistema informativo e la qualità della giustizia erogata sono fortemente interrelate. Il progetto di ricerca è orientato a individuare quali siano le condizioni in cui l’efficienza può essere effettivamente raggiunta e, soprattutto, a verificare quali siano le scelte tecnologiche che possono preservare, o anche potenziare, i principi e le garanzie del processo penale. Nel processo penale, infatti, sono coinvolti diritti fondamentali dell’individuo quali la libertà personale, la dignità, la riservatezza, diritti fondamentali che vengono tutelati attraverso un ampia gamma di diritti processuali quali la presunzione di innocenza, il diritto di difesa, il diritto al contraddittorio, la finalità di rieducazione della pena.

Chiroptical spectroscopies play a fundamental role in pharmaceutical analysis for the stereochemical characterisation of bioactive molecules, due to the close relationship between chirality and optical activity and the increasing evidence of stereoselectivity in the pharmacological and toxicological profiles of chiral drugs. The correlation between chiroptical properties and absolute stereochemistry, however, requires the development of accurate and reliable theoretical models. The present thesis will report the application of theoretical chiroptical spectroscopies in the field of drug analysis, with particular emphasis on the huge influence of conformational flexibility and solvation on chiroptical properties and on the main computational strategies available to describe their effects by means of electronic circular dichroism (ECD) spectroscopy and time-dependent density functional theory (TD-DFT) calculations. The combination of experimental chiroptical spectroscopies with state-of-the-art computational methods proved to be very efficient at predicting the absolute configuration of a wide range of bioactive molecules (fluorinated 2-arylpropionic acids, β-lactam derivatives, difenoconazole, fenoterol, mycoleptones, austdiol). The results obtained for the investigated systems showed that great care must be taken in describing the molecular system in the most accurate fashion, since chiroptical properties are very sensitive to small electronic and conformational perturbations. In the future, the improvement of theoretical models and methods, such as ab initio molecular dynamics, will benefit pharmaceutical analysis in the investigation of non-trivial effects on the chiroptical properties of solvated systems and in the characterisation of the stereochemistry of complex chiral drugs.
