Introduction: Endometrial decidualization and associated extracellular matrix (ECM) remodeling are critical events to the establishment of the maternal-fetal interface and successful pregnancy. Here, we investigated the impact of type 1 diabetes on these processes during early embryonic development, in order to contribute to the understanding of the maternal factors associated to diabetic embryopathies. Methods: Alloxan-induced diabetic Swiss female mice were bred after different periods of time to determine the effects of diabetes progression on the development of gestational complications. Furthermore, the analyses focused on decidual development as well as mRNA expression, protein deposition and ultrastructural organization of decidual ECM. Results: Decreased number of implantation sites and decidual dimensions were observed in the group mated 90-110 days after diabetes induction (D), but not in the 50-70D group. Picrosirius staining showed augmentation in the fibrillar collagen network in the 90e110D group and, following immunohistochemical examination, that this was associated with increase in types I and V collagens and decrease in type III collagen and collagen-associated proteoglycans biglycan and lumican. qPCR, however, demonstrated that only type I collagen mRNA levels were increased in the diabetic group. Alterations in the molecular ratio among distinct collagen types and proteoglycans were associated with abnormal collagen fibrillogenesis, analyzed by transmission electron microscopy. Conclusions: Our results support the concept that the development of pregnancy complications is directly related with duration of diabetes (progression of the disease), and that this is a consequence of both systemic factors (i.e. disturbed maternal endocrine-metabolic profile) and uterine factors, including impaired decidualization and ECM remodeling

There is special interest in the incorporation of metallic nanoparticles in a surrounding dielectric matrix for obtaining composites with desirable characteristics such as for surface plasmon resonance, which can be used in photonics and sensing, and controlled surface electrical conductivity. We investigated nanocomposites produced through metallic ion implantation in insulating substrate, where the implanted metal self-assembles into nanoparticles. During the implantation, the excess of metal atom concentration above the solubility limit leads to nucleation and growth of metal nanoparticles, driven by the temperature and temperature gradients within the implanted sample including the beam-induced thermal characteristics. The nanoparticles nucleate near the maximum of the implantation depth profile (projected range), that can be estimated by computer simulation using the TRIDYN. This is a Monte Carlo simulation program based on the TRIM (Transport and Range of Ions in Matter) code that takes into account compositional changes in the substrate due to two factors: previously implanted dopant atoms, and sputtering of the substrate surface. Our study suggests that the nanoparticles form a bidimentional array buried few nanometers below the substrate surface. More specifically we have studied Au/PMMA (polymethylmethacrylate), Pt/PMMA, Ti/alumina and Au/alumina systems. Transmission electron microscopy of the implanted samples showed the metallic nanoparticles formed in the insulating matrix. The nanocomposites were characterized by measuring the resistivity of the composite layer as function of the dose implanted. These experimental results were compared with a model based on percolation theory, in which electron transport through the composite is explained by conduction through a random resistor network formed by the metallic nanoparticles. Excellent agreement was found between the experimental results and the predictions of the theory. It was possible to conclude, in all cases, that the conductivity process is due only to percolation (when the conducting elements are in geometric contact) and that the contribution from tunneling conduction is negligible.

The modern GPUs are well suited for intensive computational tasks and massive parallel computation. Sparse matrix multiplication and linear triangular solver are the most important and heavily used kernels in scientific computation, and several challenges in developing a high performance kernel with the two modules is investigated. The main interest it to solve linear systems derived from the elliptic equations with triangular elements. The resulting linear system has a symmetric positive definite matrix. The sparse matrix is stored in the compressed sparse row (CSR) format. It is proposed a CUDA algorithm to execute the matrix vector multiplication using directly the CSR format. A dependence tree algorithm is used to determine which variables the linear triangular solver can determine in parallel. To increase the number of the parallel threads, a coloring graph algorithm is implemented to reorder the mesh numbering in a pre-processing phase. The proposed method is compared with parallel and serial available libraries. The results show that the proposed method improves the computation cost of the matrix vector multiplication. The pre-processing associated with the triangular solver needs to be executed just once in the proposed method. The conjugate gradient method was implemented and showed similar convergence rate for all the compared methods. The proposed method showed significant smaller execution time.

Hybrid materials with enhanced properties can now be obtained by combining nanomaterials such as carbon nanotubes and metallic nanoparticles, where the main challenge is to control fabrication conditions. In this study, we demonstrate that platinum nanoparticles (PtNps) can be electrogenerated within layer-by-layer (LbL) films of polyamidoamine (PAMAM) dendrimers and single-walled carbon nanotubes (SWCNTs), which serve as stabilizing matrices. The advantages of the possible control through electrogeneration were demonstrated with a homogeneous distribution of PtNps over the entire surface of the PAMAM/SWCNT LbL films, whose electroactive sites could be mapped using magnetic force microscopy. The Pt-containing films were used as catalysts for hydrogen peroxide reduction, with a decrease in the reduction potential of 60 mV compared to a Pt film deposited onto bare ITO. By analyzing the mechanisms responsible for hydrogen peroxide reduction, we ascribed the enhanced catalytic activity to synergistic effects between platinum and carbon in the LbL films, which are promising for sensing and fuel cell applications.

[EN]A natural generalization of the classical Moore-Penrose inverse is presented. The so-called S-Moore-Penrose inverse of a m x n complex matrix A, denoted by As, is defined for any linear subspace S of the matrix vector space Cnxm. The S-Moore-Penrose inverse As is characterized using either the singular value decomposition or (for the nonsingular square case) the orthogonal complements with respect to the Frobenius inner product. These results are applied to the preconditioning of linear systems based on Frobenius norm minimization and to the linearly constrained linear least squares problem.

The increasing use of Fiber Reinforced methods for strengthening existing brick masonry walls and columns, especially for the rehabilitation of historical buildings, has generated considerable research interest in understanding the failure mechanism in such systems. This dissertation is aimed to provide a basic understanding of the behavior of solid brick masonry walls unwrapped and wrapped with Fiber Reinforced Cementitious Matrix Composites. This is a new type of composite material, commonly known as FRCM, featuring a cementitious inorganic matrix (binder) instead of the more common epoxy one. The influence of the FRCM-reinforcement on the load-carrying capacity and strain distribution during compression test will be investigated using a full-field optical technique known as Digital Image Correlation. Compression test were carried on 6 clay bricks columns and on 7 clay brick walls in three different configuration, casted using bricks scaled respect the first one with a ratio 1:2, in order to determinate the effects of FRCM reinforcement. The goal of the experimental program is to understand how the behavior of brick masonry will be improved by the FRCM-wrapping. The results indicate that there is an arching action zone represented in the form of a parabola with a varying shape according to the used configuration. The area under the parabolas is considered as ineffectively confined. The effectively confined area is assumed to occur within the region where the arching action had been fully developed.

ZusammenfassungSchwerpunkt dieser Arbeit war die Verfahrensentwicklung zur Ultraspurenbestimmung der Platingruppenelemente (PGE) in Umwelt- und geologischen Proben unter Verwendung der massenspektrometrischen Isotopenverdünnungsanalyse mit anschließender Bestimmung an einem Quadrupol ICP-MS (ICP-QMSIVA). Geeignete Separationstechniken in der Probenaufbereitung, um die PGE von der Matrix der untersuchten Proben zu trennen, stellten eine richtige und präzise Bestimmung der Ultraspuren an einem Quadrupol ICP-MS sicher.Das Verfahren konnte anhand von geologischen Referenzmaterialien aus Kanada sichergestellt werden. Gerade die Wiederholungsbestimmungen der verschiedenen Referenzmaterialien unter Verwendung des ICP-QMSIVA Verfahrens sind beispiellos und in dieser Form noch nicht in der Literatur beschrieben. Durch systematische Messungen konnten Richtigkeit und Präzision des Verfahrens bestätigt werden und die Inhomogenität des Referenzmaterials UMT-1 bezüglich Pt bewiesen werden. Das in dieser Arbeit entwickelte Verfahren zur Ultraspurenbestimmung der PGE mit ICP-QMSIVA wurde im Rahmen des Projektes 'Production and certification of a road dust reference material for platinum, palladium and rhodium (PGEs) in automative catalytic converters (PACEPAC)' der Europäischen Union zur Zertifizierung von zwei Referenzmaterialien für Umweltproben eingesetzt. Hierbei wurde bei der Zertifizierung ('intercomparison round') eine sehr gute Übereinstimmung der Ergebnisse mit dem gewichteten Mittelwert der Ergebnisse für Pd und Pt mit den übrigen teilnehmenden Laboratorien festgestellt. Die mit der hier entwickelten Methode erhaltenen Ergebnisse wurden ohne Ausnahme für alle gemessenen Elemente zur Zertifizierung herangezogen. Damit leistete die vorliegende Arbeit einen erheblichen Beitrag zum erfolgreichen Abschluß dieses Projekts. Den Erwartungen hinsichtlich Richtigkeit und Reproduzierbarkeit des entwickelten Verfahrens wurde somit voll entsprochen. Erneut konnte die große Bedeutung der Isotopenverdünnungstechnik für die Zertifizierung von Referenzmaterialien aufgezeigt werden, da mit dieser Technik bei sachgerechtem Einsatz Ergebnisse hoher Richtigkeit erzielt werden. Durch vergleichende Messungen mit der NiS-Dokimasie und NAA, die in einer Kooperation mit dem Kernchemischen Institut der Universität Mainz durchgeführt wurden, und dem hier verwendeten Verfahren, konnten übereinstimmende Daten, hinsichtlich der Abnahme der Konzentrationen von Pd und Pt in Abhängigkeit von der Entfernung zu einer Autobahn, erzielt werden. Diese Arbeit und die Forschungsergebnisse, die mit der anerkannten NAA erzielt wurden, zeigen, daß die PGE durch Katalysatoren von Automobilen überwiegend metallisch emittiert werden. Der anthropogene Eintrag der PGE in die Umwelt kann mit dem ICP-QMSIVA Verfahren weiterhin sehr gut verfolgt werden.

Im Rahmen der Entwicklung einer Matrix für Fibroblasten zurAnwendung als dermales Äquivalent für den Aufbau einesin-vitro Testsystems für Wundauflagen wurden zunächstGelatine- und Agarfolien mit einer Streichanlage imLabormaßstab hergestellt. Keimdichtigkeit,Wasserdampfdurchlässigkeit, Elastizität, Wundverklebung,Dicke, Gewicht und Wassergehalt der Folien und zusätzlichdie Adsorption von Fibronectin an die Folienoberflächewurden bestimmt.Auf Basis einer 10 %-igen Gelatinelösung und durch Anwendungeines wasserlöslichen Carbodiimids (EDAC) konnten vernetzteGelatineschäume hergestellt werden. Untersuchungen derphysikalischen, chemischen und biologischen Eigenschaftender Gelatine-schäume dienten der Charakterisierung derMuster. Die Gelatineschäume wurden mit humanen Fibroblastenbeimpft und die zeitliche Entwicklung der Fibroblasten durchAnwendung der MTT Methode ermittelt. Zusätzlich wurde mit Hilfe des Antikörpers gegen Ki-67 die Proliferation derFibroblasten an Gefrierschnitten der Gelatineschäume untersucht.Die mit EDAC vernetzten und mit humanen Fibroblastenbeimpften Gelatineschäume dienten als dermales Äquivalentfür den Aufbau eines in-vitro Testsystems. Zur Untersuchungder Wechselwirkung zwischen verschiedenen Materialien undden Fibroblasten auf den Gelatineschäumen wurdenorientierende Versuche mit dem in-vitro Testsystem durchgeführt.

Food technologies today mean reducing agricultural food waste, improvement of food security, enhancement of food sensory properties, enlargement of food market and food economies. Food technologists must be high-skilled technicians with good scientific knowledge of food hygiene, food chemistry, industrial technologies and food engineering, sensory evaluation experience and analytical chemistry. Their role is to apply the modern vision of science in the field of human nutrition, rising up knowledge in food science. The present PhD project starts with the aim of studying and improving frozen fruits quality. Freezing process in very powerful in preserve initial raw material characteristics, but pre-treatment before the freezing process are necessary to improve quality, in particular to improve texture and enzymatic activity of frozen foods. Osmotic Dehydration (OD) and Vacuum Impregnation (VI), are useful techniques to modify fruits and vegetables composition and prepare them to freezing process. These techniques permit to introduce cryo-protective agent into the food matrices, without significant changes of the original structure, but cause a slight leaching of important intrinsic compounds. Phenolic and polyphenolic compounds for example in apples and nectarines treated with hypertonic solutions are slightly decreased, but the effect of concentration due to water removal driven out from the osmotic gradient, cause a final content of phenolic compounds similar to that of the raw material. In many experiment, a very important change in fruit composition regard the aroma profile. This occur in strawberries osmo-dehydrated under vacuum condition or under atmospheric pressure condition. The increment of some volatiles, probably due to fermentative metabolism induced by the osmotic stress of hypertonic treatment, induce a sensory profile modification of frozen fruits, that in some way result in a better acceptability of consumer, that prefer treated frozen fruits to untreated frozen fruits. Among different processes used, a very interesting result was obtained with the application of a osmotic pre-treatment driven out at refrigerated temperature for long time. The final quality of frozen strawberries was very high and a peculiar increment of phenolic profile was detected. This interesting phenomenon was probably due to induction of phenolic biological synthesis (for example as reaction to osmotic stress), or to hydrolysis of polymeric phenolic compounds. Aside this investigation in the cryo-stabilization and dehydrofreezing of fruits, deeper investigation in VI techniques were carried out, as studies of changes in vacuum impregnated prickly pear texture, and in use of VI and ultrasound (US) in aroma enrichment of fruit pieces. Moreover, to develop sensory evaluation tools and analytical chemistry determination (of volatiles and phenolic compounds), some researches were bring off and published in these fields. Specifically dealing with off-flavour development during storage of boiled potato, and capillary zonal electrophoresis (CZE) and high performance liquid chromatography (HPLC) determination of phenolic compounds.

MMP-2 and MMP-14 process extracellular matrix proteins,cytokines, growth factors and adhesion molecules to generatefragments with enhanced or reduced biological activity.In this study, a vectorsystem was developed for theconditional expression of MMP-2 and MMP-14 in the liver oftransgenic mice. For this vectorsystem the murine albuminpromotor was chosen together with the cre/lox system toachieve an inducible MMP-expression in the liver.Only one of the MMP-14 transgenic lines expressed highamounts of active MMP-14 protein after recombination of thelox-P sites. In these mice MMP-14 was able to activate MMP-2and MMP-13 in vivo. However, none of the livers of MMP-14overexpressing mice showed no differences in liverweight,amount of extracellular matrixproteins and rate ofproliferation, apoptosis and tumor-induction when comparedto the liver of wildtype mice.On the other hand overexpression of MMP-2 was embryoniclethal in all MMP-2 transgenic lines. After crossing theMMP-2 transgenic mice with cre deleter mice, a cre mediatedrecombination could be shown at day 6.5 post coitum (pc).Some of the double transgenic embryos of one of thetransgenic lines had severe deformations of the head,especially of the telencephalon and the mesencephalon.It could be shown in this study that disregulation of MMP-2in early embryonic development is lethal but anoverexpression of MMP-14 has no influence on the embryonicdevelopment or the homeostasis of the adult liver.With this conditional vectorsystem it is to possible studythe influnce of MMP-2 and MMP-14 on fibrogenesis,regeneration and tumorgenesis in the liver of mice.

RAGE mediates diverse physiological and pathological effects by binding a variety of ligands. Despite incomplete understanding of RAGE-mediated disorders soluble RAGE (sRAGE) has been identified as a potential biomarker for RAGE-related diseases and possibly represents a hopeful pharmaceutical against RAGE-mediated disorders. Nevertheless, the source of sRAGE remains poorly investigated. Currently sRAGE is thought to be derived exclusively from alternative splicing of mRNA. In this thesis it was investigated whether sRAGE can also be released as a result of ectodomain shedding of full-length RAGE. Using cells overexpressing RAGE as a model system, it was demonstrated clearly that RAGE undergoes ectodomain shedding in both constitutive and regulated manner. Several stimuli including PMA, AMPA, calcium and chelerythrine stimulated the release of sRAGE into cell culture medium. Moreover, possible mechanisms that regulate ectodomain shedding of RAGE were investigated and it was found that shedding of RAGE is likely independent from PKC and MAPK pathways. By using gain of function and loss of function approaches MMP9 but not ADAM10, ADAM17 or MT1-MMP was characterized as the metalloproteinase that mediates shedding of RAGE. Furthermore, it was shown that cytoplasmic domain of RAGE is not essential for shedding of RAGE. In addition, the potential cleavage site of RAGE by MMP9 was investigated and a lack of sequence specificity for the RAGE processing proteinase was demonstrated by mutation analysis. Finally the physiopathological significance of shedding of RAGE is discussed. In conclusion, for the first time ectodomain shedding of human RAGE and the underlying regulatory mechanisms were investigated. The data open a new field for modulation of RAGE shedding as a novel intervention approach against RAGE-mediated diseases.
