Steroide sind im Organismus weit verbreitete, vielfältig substituierte Naturstoffe mit einem breiten biologischen Wirkungsspektrum. C/D-cis-verknüpfte Steroide kommen in der Natur zwar selten vor, sie sind aber in ihrer pharmakologischen Aktivität nicht weniger interessant. Total- bzw. Partialsynthesen sind bisher in der Literatur kaum beschrieben worden. Die Arbeit beschäftigt sich mit grundlegenden Reaktionen zur enantioselektiven Synthese eines 13,14-cis-verknüpften Steroids. Als Fernziel wurde C/D-cis-verknüpftes Östradiol ausgewählt mit der Option, das Substitutionsmuster dieses Steroids vielfältig variieren zu können. Der Syntheseplan beinhaltet zunächst den Aufbau des Kohlenstoffgerüsts im Sinne einer konvergenten Synthese, abschließende Cyclisierungen sollten dann das vollständige Molekül ergeben. Im Rahmen dieser Arbeit wurde zunächst das C/D-Ringfragment als Allylamin hergestellt. Das Amin ist dabei als chirales Auxiliar ausgelegt. Als A,B-Ringfragment wurden eine Reihe 4-Aryl-2,3-ungesättigte Carbonsäurefluoride synthetisiert. Im Verknüpfungsschritt, einer zwitterionischen Aza-Claisen-Umlagerung, konnten nun beide Fragmente mit hoher simpler, aber mäßiger induzierter Diastereoselektivität gekuppelt werden. Das Entfernen der Doppelbindung im Produkt erwies sich als erhebliche Hürde, die jedoch durch die Verwendung eines Enolethers genommen werden konnte. Das im Rahmen der Arbeit hergestellte Material sollte nun in wenigen Stufen zum C/D- cis-verknüpften Steroid umgesetzt werden können.

(De)colonization Through Topophilia: Marjorie Kinnan Rawlings’s Life and Work in Florida attempts to reveal the author’s intimate connection to and mental growth through her place, namely the Cross Creek environs, and its subsequent effect on her writing. In 1928, Marjorie Kinnan Rawlings and her first husband Charles Rawlings came to Cross Creek, Florida. They bought the shabby farmhouse on Cross Creek Road, trying to be both, writers and farmers. However, while Charles Rawlings was unable to write in the backwoods of the Florida Interior, Rawlings found her literary voice and entered a symbiotic, reciprocal relationship with the natural world of the Cracker frontier. Her biographical preconditions – a childhood spent in the rural area of Rock Creek, outside of Washington D. C. - and a father who had instilled in her a sense of place or topophilia, enabled her to overcome severe marriage tensions and the hostile climate women writers faced during the Depression era. Nature as a helping ally and as an “undomesticated”(1) space/place is a recurrent motif throughout most of Rawlings’s Florida literature. At a time when writing the American landscape/documentary and the extraction of the self from texts was the prevalent literary genre, Marjorie Kinnan Rawlings inscribed herself into her texts. However, she knew that the American public was not yet ready for a ‘feminist revolt’, but was receptive of the longtime ‘inaudible’ voices from America’s regions, especially with regard to urban poverty and a homeward yearning during the Depression years. Fusing with the dynamic eco-consciousness of her Cracker friends and neighbors, Rawlings wrote in the literary category of regionalism enabling her to pursue three of her major aims: an individuated self, a self that assimilated with the ‘master narratives’ of her time and the recognition of the Florida Cracker and Scrub region. The first part of this dissertation briefly introduces the largely unknown and underestimated writer Marjorie Kinnan Rawlings, providing background information on her younger years, the relationship toward her family and other influential persons in her life. Furthermore, it takes a closer look at the literary category of regionalism and Rawlings’s use of ‘place’ in her writings. The second part is concerned with the ‘region’ itself, the state of Florida. It focuses on the natural peculiarities of the state’s Interior, the scrub and hammock land around her Cracker hamlet as well as the unique culture of the Florida Cracker. Part IV is concerned with the analysis of her four Florida books. The author is still widely related to the ever-popular novel The Yearling (1938). South Moon Under (1933) and Golden Apples (1935), her first two novels, have not been frequently republished and have subsequently fallen into oblivion. Cross Creek (1942), Rawlings’s last Florida book, however, has recently gained renewed popularity through its use in classes on nature writers and the non-fiction essay but it requires and is here re-evaluated as the author’s (relational) autobiography. The analysis through place is brought to completion in this work and seems to intentionally close the circle of Rawlings’s Florida writings. It exemplifies once more that detachment from place is impossible for Rawlings and that the intermingling of life and place in literature, is essential for the (re)creation of her identity. Cross Creek is therefore not only one of Rawlings’s greatest achievements; it is more importantly the key to understanding the author’s self and her fiction. Through the ‘natural’ interrelationship of place and self and by looking “mutually outward and inward,”(2) Marjorie Kinnan Rawlings finds her literary voice, a home and ‘a room of her own’ in which to write and come to consciousness. Biomedical analyses are becoming increasingly complex, with respect to both the type of the data to be produced and the procedures to be executed. This trend is expected to continue in the future. The development of information and protocol management systems that can sustain this challenge is therefore becoming an essential enabling factor for all actors in the field. The use of custom-built solutions that require the biology domain expert to acquire or procure software engineering expertise in the development of the laboratory infrastructure is not fully satisfactory because it incurs undesirable mutual knowledge dependencies between the two camps. We propose instead an infrastructure concept that enables the domain experts to express laboratory protocols using proper domain knowledge, free from the incidence and mediation of the software implementation artefacts. In the system that we propose this is made possible by basing the modelling language on an authoritative domain specific ontology and then using modern model-driven architecture technology to transform the user models in software artefacts ready for execution in a multi-agent based execution platform specialized for biomedical laboratories.

Die Analyse funktioneller Zusammenhänge zwischen Ernährung und Zahnmorphologie ist ein wichtiger Aspekt primatologischer und paläontologischer Forschung. Als überdauernder Teil des Verdauungssystems geben Zähne die bestmöglichen Hinweise auf die Ernährungsstrategien (ausgestorbener) Arten und eine Fülle weiterer Informationen. Aufgrund dessen ist es für die wissenschaftliche Arbeit von größter Bedeutung, die Zähne so detailliert und exakt wie möglich in ihrer gesamten Struktur zu erfassen. Bisher wurden zumeist zweidimensionale Parameter verwendet, um die komplexe Kronenmorphologie von Primatenmolaren vergleichend zu untersuchen. Die vorliegende Arbeit hatte das Ziel, Zähne verschiedener Arten von Altweltaffen mittels computerbasierter Methoden dreidimensional zu erfassen und neue Parameter zu definieren, mit denen die Form dieser Zähne objektiv erfasst und funktionell interpretiert werden kann. Mit einem Oberflächen-Scanner wurden die Gebisse einer Stichprobe von insgesamt 48 Primaten von fünf verschiedenen Arten eingescannt und mit Bildverarbeitungsmethoden so bearbeitet, dass dreidimensionale digitale Modelle einzelner Backenzähne zur Analyse vorlagen. Es wurden dabei sowohl Arten ausgewählt, die eine für ihre Gattung typische Ernährungsweise besitzen - also Frugivorie bei den Cercopithecinen und Folivorie bei den Colobinen - als auch solche, die eine davon abweichende Alimentation bevorzugen. Alle Altweltaffen haben sehr ähnliche Molaren. Colobinen haben jedoch höhere und spitzere Zahnhöcker, dünneren Zahnschmelz und scheinen ihre Zähne weniger stark abzukauen als die Meerkatzen. Diese Beobachtungen konnten mit Hilfe der neuen Parameter quantifiziert werden. Aus der 3D-Oberfläche und der Grundfläche der Zähne wurde ein Index gebildet, der die Stärke des Oberflächenreliefs angibt. Dieser Index hat bei Colobinen deutlich höhere Werte als bei Cercopithecinen, auch bei Zähnen, die schon stark abgekaut sind. Die Steilheit der Höcker und ihre Ausrichtung wurden außerdem gemessen. Auch diese Winkelmessungen bestätigten das Bild. Je höher der Blätteranteil an der Ernährung ist, desto höher sind die Indexwerte und umso steiler sind die Höcker. Besonders wichtig war es, dies auch für abgekaute Zähne zu bestätigen, die bisher nicht in funktionelle Analysen miteinbezogen wurden. Die Ausrichtung der Höckerseiten gibt Hinweise auf die Kaubewegung, die zum effizienten Zerkleinern der Nahrung notwendig ist. Die Ausrichtung der Höcker der Colobinen deutet darauf hin, dass diese Primaten flache, gleitende Kaubewegungen machen, bei denen die hohen Höcker aneinander vorbei scheren. Dies ist sinnvoll zum Zerschneiden von faserreicher Nahrung wie Blättern. Cercopithecinen scheinen ihre Backenzähne eher wie Mörser und Stößel zu verwenden, um Früchte und Samen zu zerquetschen und zu zermahlen. Je nachdem, was neben der hauptsächlichen Nahrung noch gekaut wird, unterscheiden sich die Arten graduell. Anders als bisher vermutet wurde, konnte gezeigt werden, dass Colobinen trotz des dünnen Zahnschmelzes ihre Zähne weniger stark abkauen und weniger Dentin freigelegt wird. Dies gibt eindeutige Hinweise auf die Unterschiede in der mechanischen Belastung, die während des Kauvorgangs auf die Zähne wirkt, und lässt sich gut mit der Ernährung der Arten in Zusammenhang bringen. Anhand dieser modellhaften Beobachtungen können in Zukunft ausgestorbene Arten hinsichtlich ihrer Ernährungsweise mit 3D-Techniken untersucht werden.

In der vorliegenden Arbeit wurden Derivate des Ionentransporters Valinomycin synthetisiert und als Sensorelemente in Modellmembransysteme inkorporiert. Als Modellmembranen wurden festkörpergestützte Lipiddoppelschichten (tBLMs, tethered bilayer lipid membranes) verwendet. rnValinomycin transportiert selektiv Kalium-Ionen durch Membranen, was durch einen Rückgang des Widerstandes über elektrochemische Messmethoden nachgewiesen werden kann. Es ist ein zyklisches Dodecadepsipeptid, das aus zwei verschiedenen Aminosäuren (L- und D-Valin) und  Hydroxysäuren (L Milchsäure und D Hydroxyisovaleriansäure) besteht. In dieser Arbeit wurde ein L Valin durch ein L-Lysin ausgetauscht, um eine freie Aminogruppe zum Anbinden verschiedenster Liganden zu erhalten. rnDie Synthese erfolgte in Lösung über einen linearen Decadepsipeptid-Precursor, hierbei wurde hauptsächlich mit Benzyl- und Boc-Schutzgruppen gearbeitet. An den Precursor wurden dann unterschiedlich modifizierte Lysin-Didepside gebunden und das daraus erhaltene lineare Dodecadepsipeptid zyklisiert.rnInsgesamt wurden sechs verschiedene Derivate synthetisiert, deren Ionentransportfähigkeit mit Hilfe eines angebundenen Liganden blockiert wurde. Diese Blockade kann entweder mechanisch durch Festhalten des Ionencarriers an der Oberfläche der Membran oder chemisch durch Einbringen einer Ladung erfolgen, da geladene Moleküle eine Membran nicht überwinden können. rnAcetyl-Lysin-Valinomycin wurde als Testsystem hergestellt, um zu zeigen, dass die Synthese einen funktionsfähigen Ionencarrier ergeben hatte. Im nächsten Schritt wurde Lysin-Valinomycin mit freier Aminogruppe synthetisiert, um es als pH-Sensor zu nutzen und damit zu überprüfen, ob das chemische Einbringen einer Ladung möglich ist. Es konnte ein pH abhängiger Kalium-Transport nachgewiesen werden, die Blockade der Ionentransportfähigkeit über eine eingebrachte Ladung ist somit möglich. rnAuf dem gleichen Konzept beruht Ferrocen-Valinomycin. Wird der Ferrocen-Ligand oxidiert, liegt eine positive Ladung vor und der Ionencarrier kann die Membran nicht mehr überwinden. Eine Reduktion macht diesen Prozess reversibel. Ferrocen-Valinomycin konnte innerhalb einer tBLM chemisch oxidiert und reduziert werden, dieses System kann somit als chemischer Redox-Sensor eingesetzt werden.rnEine mechanische Blockade liegt dem Biotin- und dem Sulfonamid-Valinomycin zugrunde. Dabei soll die Zugabe von Streptavidin bzw. BCA II (bovine Carboanhydrase) den Ionentransport durch die Membran stoppen. Beide Valinomycin-Derivate zeigten aber keine Ionentransportfähigkeit, eine Inkorporation in tBLMs konnte jedoch über SPR gezeigt werden. rnDie Synthese eines fluoreszenz-markierten (FITC) Valinomycins ergab zwar auch keinen transportfähigen Ionencarrier, aber mit diesem Derivat konnte der Diffusionskoeffizient von Valinomycin in sBLMs mit Hilfe von Fluorescence Recovery after photobleaching (FRAP) bestimmt werden.rn

Definiert konfigurierte mittelgroße ungesättigte Heterocyclen sind wertvolle Zwischenstufen in der Naturstoff- und Wirkstoffsynthese. Es konnte gezeigt werden, dass 2-alkinyl-substituierte Piperidine und Azepane in einer Aza-Keten-Claisen-Reaktion zu 10- und 11-gliedrigen Allenyllactamen umgelagert werden können. Ein 9-gliedriges Allenyllactam konnte nicht dargestellt werden (Ringspannung). Über eine sechs- bis sieben-stufige Reaktionssequenz konnten optisch aktive, geschützte Piperidinole aufgebaut werden. Es wurden Auxiliar kontrollierte Hetero-Diels-Alder-Reaktionen, diastereoselektive Reduktionen, Bestmann-Ohira Umlagerungen zu Alkinen und verschiedene Alkin-Funktionalisierungen erarbeitet. Eine Aza-Claisen-Umlagerung liefert schließlich optisch aktive Lactame deren absolute Konfiguration des Allensystems mittels NOE-NMR-Spektroskopie untersucht werden kann. Limitierungen und Möglichkeiten der Synthese werden eingehend diskutiert. Sowohl der stereochemische Verlauf der Reaktion als auch die Konformation der Produkte ermöglichen eine Fokussierung auf nachfolgende Naturstoffsynthesen vorzunehmen.

Applikationsfertige Zytostatikazubereitungen werden heute unter der Verantwortung eines Apothekers in zentralisierten Herstellungsbereichen hergestellt. Weil die Verordnung der Chemotherapie ein großes Fehlerrisiko birgt, ist konsequentes Verordnungsmonitoring ein wesentlicher Teilprozess der zentralen Zytostatikazubereitung. rnDie aktuelle Umsetzung und die Ergebnisse des Verordnungsmonitorings in den Universitätskliniken Deutschlands wurden im Rahmen dieser Arbeit in einer prospektiven Erhebung erfasst. Als häufigste Verordnungsirrtümer wurden Dosisberechnungsfehler (48%), welche als von hoher Relevanz (78%) für die Patientensicherheit angesehen wurden, genannt. Die Inzidenz der Verordnungsfehler betrug durchschnittlich 0,77% bei rund 1950 Verordnungen pro Tag. Das konsequente Verordnungsmonitoring von pharmazeutischer Seite erfolgt höchst effizient und leistet einen hohen Beitrag zur Patienten- und Arzneimitteltherapiesicherheit in der Onkologie.rnFür die Herstellung der applikationsfertiger Zytostatika-Zubereitungen sind fundierte Kenntnisse zu deren physikalisch-chemischen Stabilität erforderlich. Zu neu zugelassenen Zytostatika und insbesondere Biologicals, stehen häufig noch keine Daten zur Stabilität der applikationsfertigen Lösungen zur Verfügung. Die Bestimmung der physikalisch-chemischen Stabilität war daher Gegenstand dieser Arbeit. Die applikationsfertigen Infusionslösungen der Purin-Analoga Nelarabin und Clofarabin (RP-HPLC), sowie des monoklonalen Antiköpers Trastuzumab (SEC, UV-Spektroskopie, SDS-Page), erwiesen sich über einen Zeitraum von mindestens 28 Tagen als stabil. Die Stabilität zweier Camptothecin-Derivate (Topotecan und Irinotecan) beladen auf DC Beads™, wie auch die Ladungskapazität und Kompatibilität mit Kontrastmitteln, wurde ebenfalls bewiesen. rn

Introduzione. Le cellule mesenchimali derivate dal tessuto adiposo (hASC) rappresentano un importante strumento per la terapia cellulare, in quanto derivano da un tessuto adulto abbondante e facilmente reperibile. Con il dispositivo medico Lipogems l’isolamento di tali cellule è eseguito esclusivamente mediante sollecitazioni meccaniche. Il prodotto ottenuto è quindi minimamente manipolato e subito utilizzabile. Ad oggi, il condizionamento pro-differenziativo delle staminali è per lo più attuato mediante molecole di sintesi. Tuttavia, altri fattori possono modulare la fisiologia cellulare, come gli stimoli fisici e molecole naturali. Onde elettromagnetiche hanno indotto in modelli cellulari staminali l’espressione di alcuni marcatori di differenziamento e, in cellule adulte, una riprogrammazione, mentre estratti embrionali di Zebrafish sono risultati antiproliferativi sia in vitro che in vivo. Metodi. La ricerca di nuove strategie differenziative sia di natura fisica che molecolare, nel particolare onde acustiche ed estratti embrionali di Zebrafish, è stata condotta utilizzando come modello cellulare le hASC isolate con Lipogems. Onde acustiche sono state somministrate mediante l’utilizzo di due apparati di trasduzione, un generatore di onde meccaniche e il Cell Exciter . I trattamenti con gli estratti embrionali sono stati effettuati utilizzando diverse concentrazioni e diversi tempi sperimentali. Gli effetti sull’espressione dei marcatori di staminalità e differenziamento relativi ai trattamenti sono stati saggiati in RT-PCR quantitativa relativa e/o in qPCR. Per i trattamenti di tipo molecolare è stata valutata anche la proliferazione. Risultati e conclusioni. La meta-analisi dei dati delle colture di controllo mostra la stabilità d’espressione genica del modello. I trattamenti con i suoni inducono variazioni dell’espressione genica, suggerendo un ruolo regolatorio di tali stimoli, in particolare del processo di commitment cardiovascolare. Due degli estratti embrionali di Zebrafish testati inibiscono la proliferazione alle 72 ore dalla somministrazione. L’analisi d’espressione associata ai trattamenti antiproliferativi suggerisce che tale effetto abbia basi molecolari simili ai processi di differenziamento.

This thesis is developed in the contest of Ritmare project WP1, which main objective is the development of a sustainable fishery through the identification of populations boundaries in commercially important species in Italian Seas. Three main objectives are discussed in order to help reach the main purpose of identification of stock boundaries in Parapenaeus longirostris: 1 -Development of a representative sampling design for Italian seas; 2 -Evaluation of 2b-RAD protocol; 3 -Investigation of populations through biological data analysis. First of all we defined and accomplished a sampling design which properly represents all Italian seas. Then we used information and data about nursery areas distribution, abundance of populations and importance of P. longirostris in local fishery, to develop an experimental design that prioritize the most important areas to maximize the results with actual project funds. We introduced for the first time the use of 2b-RAD on this species, a genotyping method based on sequencing the uniform fragments produced by type IIB restriction endonucleases. Thanks to this method we were able to move from genetics to the more complex genomics. In order to proceed with 2b-RAD we performed several tests to identify the best DNA extraction kit and protocol and finally we were able to extract 192 high quality DNA extracts ready to be processed. We tested 2b-RAD with five samples and after high-throughput sequencing of libraries we used the software “Stacks” to analyze the sequences. We obtained positive results identifying a great number of SNP markers among the five samples. To guarantee a multidisciplinary approach we used the biological data associated to the collected samples to investigate differences between geographical samples. Such approach assures continuity with other project, for instance STOCKMED, which utilize a combination of molecular and biological analysis as well.

Due to its practical importance and inherent complexity, the optimisation of distribution networks for supplying drinking water has been the subject of extensive study for the past 30 years. The optimization is governed by sizing the pipes in the water distribution network (WDN) and / or optimises specific parts of the network such as pumps, tanks etc. or try to analyse and optimise the reliability of a WDN. In this thesis, the author has analysed two different WDNs (Anytown City and Cabrera city networks), trying to solve and optimise a multi-objective optimisation problem (MOOP). The main two objectives in both cases were the minimisation of Energy Cost (€) or Energy consumption (kWh), along with the total Number of pump switches (TNps) during a day. For this purpose, a decision support system generator for Multi-objective optimisation used. Its name is GANetXL and has been developed by the Center of Water System in the University of Exeter. GANetXL, works by calling the EPANET hydraulic solver, each time a hydraulic analysis has been fulfilled. The main algorithm used, was a second-generation algorithm for multi-objective optimisation called NSGA_II that gave us the Pareto fronts of each configuration. The first experiment that has been carried out was the network of Anytown city. It is a big network with a pump station of four fixed speed parallel pumps that are boosting the water dynamics. The main intervention was to change these pumps to new Variable speed driven pumps (VSDPs), by installing inverters capable to diverse their velocity during the day. Hence, it’s been achieved great Energy and cost savings along with minimisation in the number of pump switches. The results of the research are thoroughly illustrated in chapter 7, with comments and a variety of graphs and different configurations. The second experiment was about the network of Cabrera city. The smaller WDN had a unique FS pump in the system. The problem was the same as far as the optimisation process was concerned, thus, the minimisation of the energy consumption and in parallel the minimisation of TNps. The same optimisation tool has been used (GANetXL).The main scope was to carry out several and different experiments regarding a vast variety of configurations, using different pump (but this time keeping the FS mode), different tank levels, different pipe diameters and different emitters coefficient. All these different modes came up with a large number of results that were compared in the chapter 8. Concluding, it should be said that the optimisation of WDNs is a very interested field that has a vast space of options to deal with. This includes a large number of algorithms to choose from, different techniques and configurations to be made and different support system generators. The researcher has to be ready to “roam” between these choices, till a satisfactory result will convince him/her that has reached a good optimisation point.

Con le "Imagini degli dei degli antichi", pubblicate a Venezia nel 1556 e poi in più edizioni arricchite e illustrate, l’impegnato gentiluomo estense Vincenzo Cartari realizza il primo, fortunatissimo manuale mitografico italiano in lingua volgare, diffuso e tradotto in tutta l’Europa moderna. Cartari rimodula, secondo accenti divulgativi ma fedeli, fonti latine tradizionali: come le ricche "Genealogie deorum gentilium" di Giovanni Boccaccio, l’appena precedente "De deis gentium varia et multiplex historia" di Lilio Gregorio Giraldi, i curiosi "Fasti" ovidiani, da lui stesso commentati e tradotti. Soprattutto, però, introduce il patrimonio millenario di favole ed esegesi classiche, di aperture egiziane, mediorientali, sassoni, a una chiave di lettura inedita, agile e vitalissima: l’ecfrasi. Le divinità e i loro cortei di creature minori, aneddoti leggendari e attributi identificativi si susseguono secondo un taglio iconico e selettivo. Sfilano, in trionfi intrisi di raffinato petrarchismo neoplatonico e di emblematica picta poesis rinascimentale, soltanto gli aspetti figurabili e distintivi dei personaggi mitici: perché siano «raccontate interamente» tutte le cose attinenti alle figure antiche, «con le imagini quasi di tutti i dei, e le ragioni perché fossero così dipinti». Così, le "Imagini" incontrano il favore di lettori colti e cortigiani eleganti, di pittori e ceramisti, di poeti e artigiani. Allestiscono una sorta di «manuale d’uso» pronto all’inchiostro del poeta o al pennello dell’artista, una suggestiva raccolta di «libretti figurativi» ripresi tanto dalla maniera di Paolo Veronese o di Giorgio Vasari, quanto dal classicismo dei Carracci e di Nicolas Poussin. Si rivelano, infine, summa erudita capace di attirare appunti e revisioni: l’antiquario padovano Lorenzo Pignoria, nel 1615 e di nuovo nel 1626, vi aggiunge appendici archeologiche e comparatistiche, interessate al remoto regno dei faraoni quanto agli esotici idoli orientali e dei Nuovi Mondi.

The aging process is characterized by the progressive fitness decline experienced at all the levels of physiological organization, from single molecules up to the whole organism. Studies confirmed inflammaging, a chronic low-level inflammation, as a deeply intertwined partner of the aging process, which may provide the “common soil” upon which age-related diseases develop and flourish. Thus, albeit inflammation per se represents a physiological process, it can rapidly become detrimental if it goes out of control causing an excess of local and systemic inflammatory response, a striking risk factor for the elderly population. Developing interventions to counteract the establishment of this state is thus a top priority. Diet, among other factors, represents a good candidate to regulate inflammation. Building on top of this consideration, the EU project NU-AGE is now trying to assess if a Mediterranean diet, fortified for the elderly population needs, may help in modulating inflammaging. To do so, NU-AGE enrolled a total of 1250 subjects, half of which followed a 1-year long diet, and characterized them by mean of the most advanced –omics and non –omics analyses. The aim of this thesis was the development of a solid data management pipeline able to efficiently cope with the results of these assays, which are now flowing inside a centralized database, ready to be used to test the most disparate scientific hypotheses. At the same time, the work hereby described encompasses the data analysis of the GEHA project, which was focused on identifying the genetic determinants of longevity, with a particular focus on developing and applying a method for detecting epistatic interactions in human mtDNA. Eventually, in an effort to propel the adoption of NGS technologies in everyday pipeline, we developed a NGS variant calling pipeline devoted to solve all the sequencing-related issues of the mtDNA.

Nowadays microalgae are studied, and a number of species already mass-cultivated, for their application in many fields: food and feed, chemicals, pharmaceutical, phytoremediation and renewable energy. Phytoremediation, in particular, can become a valid integrated process in many algae biomass production systems. This thesis is focused on the physiological and biochemical effects of different environmental factors, mainly macronutrients, lights and temperature on microalgae. Microalgal species have been selected on the basis of their potential in biotechnologies, and nitrogen occurs in all chapters due to its importance in physiological and applicative fields. There are 5 chapters, ready or in preparation to be submitted, with different specific matters: (i) to measure the kinetic parameters and the nutrient removal efficiencies for a selected and local strain of microalgae; (ii) to study the biochemical pathways of the microalga D. communis in presence of nitrate and ammonium; (iii) to improve the growth and the removal efficiency of a specific green microalga in mixotrophic conditions; (iv) to optimize the productivity of some microalgae with low growth-rate conditions through phytohormones and other biostimulants; and (v) to apply the phyto-removal of ammonium in an effluent from anaerobic digestion. From the results it is possible to understand how a physiological point of view is necessary to provide and optimize already existing biotechnologies and applications with microalgae.

Shellac is the purified product of the natural polymer Lac. Shellac types, from different origins and with different ages, all purified by the solvent extraction process were compared in this study. Their physicochemical properties acid value, glass transition temperatures, color numbers and molecular sizes were determined. Metoprolol tartrate pellets were coated by air suspension coating with these different grades of shellac. Two coating levels 20% w/w and 25% w/w were applied and then subjected to in vitro dissolution testing. Enteric resistance was achieved for all tested brands for the two coating levels. At pH 6.8, 7.2 and 7.4, significant variations were obvious between the brands. rnMoreover the molecular size of shellac has a pronounced effect in that shellac types with larger molecular size show a higher and faster release than others, while the one with the smaller molecular size show the opposite effect on the release of metoprolol.rnIn this study commercially available ready for use aqueous shellac solutions (SSB AQUAGOLD), which are based on shellac SSB 57 (Dewaxed Orange Shellac, Bysakhi-Ber type refined in a solvent extraction process), with different manufacturing dates were used. rnTo improve the enteric coating properties of films from aqueous shellac solutions, different aqueous polymeric solutions of hydroxypropyl methylcellulose (HPMC), hydroxypropyl cellulose (HPC), carboyxmethyl cellulose (CMC), gum arabic and polysaccharides (Pullulan®) were used. These water soluble polymers will act as pore formers to enhance drug release from pellets coated with the combination of shellac and these polymers. The influence of these polymers on the gloss of the shellac films, mechanical properties of the films and drug release from metoprolol tartrate pellets were studied.rnThe potential of ethanol to alter the rate of drug release from shellac coated pellets was assessed by using a modified in vitro dose dumping in alcohol (DDA) method and the test concluded that shellac coated dosage forms can be co-administered with alcohol beverages containing ≤ 5% with no effect of alcohol on the shellac coat.rnPellets coated with shellac sodium salts, showed higher release rates than pellets coated with shellac as ammonium salt forms. rn

Gli ultimi anni hanno visto importanti cambiamenti positivi nella cooperazione interstatale in Asia centrale. Crescenti minacce come il terrorismo internazionale, l'estremismo religioso e politico, il traffico di droga, ecc, causati dagli interessi geopolitici e geo-economici delle potenze mondiali, hanno contribuito alla formazione di una politica estera più coordinata e coerente degli Stati della regione. Questo processo si manifesta nella partecipazione attiva dell’istituzionalizzazione della Shanghai Organizzazione del Commonwealth (SOC), Conferenza sulle misure di costruzione d’interazione e fiducia in Asia (CICA) e Organizzazione del Trattato di Sicurezza Collettiva (CSTO). Problemi moderni di sicurezza in Asia centrale dovrebbero essere risolti in dei nuovi modi, non convenzionali. Le nuove minacce alla sicurezza richiedono modi non standard per risolvere il problema. Considerate le differenze fondamentali dell'Unione europea e degli Stati dell'Asia centrale nei valori e dal punto di vista in materia di sicurezza. I paesi dell'Asia centrale non sono pronti per l'integrazione politica. Nonostante questo, nell’Asia centrale sono state adottate misure per contrastare le minacce non convenzionali. L’esperienza europea di unire gli sforzi della regione per garantire la sicurezza interna, può essere utilizzata dai paesi dell'Asia centrale, soprattutto, in primo luogo sulla formazione del quadro istituzionale e giuridico per la cooperazione operativa delle forze dell'ordine per le seguenti aree: • prevenzione del traffico di droga attraverso gli Stati dell'Asia centrale; • lotta contro nuove forme di terrorismo ed estremismo; • limitare la dimensione della migrazione clandestina; • migliorare la protezione giuridica dei cittadini. Fino a poco tempo fa, questi temi hanno ricevuto poca attenzione, sia nella teoria sia nella pratica, poiché i problemi di sicurezza della società erano principalmente ridotti per evitare il pericolo di guerra.
