The isolation of thionein (T) from tissues has not been reported heretofore. T contains 20 cysteinyl residues that react with 7-fluorobenz-2-oxa-1,3-diazole-4-sulfonamide to form fluorescent adducts. In metallothionein (MT) the cysteinyl residues, which are bound to zinc, do not react. However, they do react in the presence of a chelating agent such as EDTA. The resultant difference in chemical reactivity provides a means to measure T in the absence of EDTA, (MT + T) in its presence, and, of course, MT by difference. The 7-fluorobenz-2-oxa-1,3-diazole-4-sulfonamide derivative of T can be isolated from tissue homogenates by HPLC and quantified fluorimetrically with a detection limit in the femtomolar range and a linear response over 3 orders of magnitude. Analysis of liver, kidney, and brain of rats reveals almost as much T as MT. Moreover, in contrast to earlier views, MT in tissue extracts appears to be less stable than T. The existence of T in tissues under normal physiological conditions has important implications for its function both in zinc metabolism and the redox balance of the cell.

Neste trabalho, anticorpos anti-IgGh foram conjugados às nanopartículas de prata (NPAg) para detectar imunoglobulina G humana (IgGh). Um imunoensaio colorimétrico baseado na diminuição da agregação devido ao aumento da repulsão eletrostática após a interação ligante-alvo. A agregação é induzida pela variação da força iônica e uma mudança da coloração da suspensão coloidal de amarelo para vermelho pode ser observada. Na presença de IgGh, a agregação é inibida e a coloração da suspensão coloidal não se altera. As nanopartículas foram obtidas por meio de cinco procedimentos diferentes e caracterizadas por espectroscopia UV-Vis, espalhamento dinâmico de luz, difração de raios-X e microscopia eletrônica. Glicose e borohidreto de sódio foram utilizados como agentes redutores, enquanto CTAB e β-ciclodextrina foram utilizados como estabilizantes. Citrato de sódio foi utilizado como agente redutor e/ou estabilizante. Nanoesferas de carbono foram obtidas por tratamento hidrotérmico de uma solução aquosa de glicose e também foram utilizadas no preparo das nanopartículas. As nanopartículas foram funcionalizadas com ácido mercaptossuccínico e a conjugação ocorreu devido à interação entre grupos aminas e grupos carboxílicos ionizados, presentes no anticorpo e agente de acoplamento, respectivamente. A estabilidade dos conjugados e o efeito da adição de IgGh foram avaliados para todos os sistemas preparados. As nanopartículas de prata preparadas com borohidreto de sódio e citrato de sódio foram selecionadas para serem aplicadas no desenvolvimento do imunoensaio e as condições experimentais foram avaliadas. Em condições ótimas, observou-se uma correlação linear entre a diminuição da agregação do sistema (NPAg-anti-IgGh) e a concentração de IgGh (0 a 200 ng mL-1). O limite de detecção foi estimado em 25 ng mL-1. O método colorimétrico apresentou boa seletividade para a detecção de IgGh. Além disso, foi obtido um resultado satisfatório ao aplicar o método para determinação do fator IX de coagulação. Foi desenvolvido também um método para determinação de ATP baseado na agregação de nanopartículas de ouro. Aptâmeros foram utilizados como elemento de reconhecimento. Em princípio, o método pode ser aplicável à determinação de outros analitos, por meio da substituição do aptâmero utilizado neste trabalho pelo oligonucleotídeo específico para o alvo de interesse.

A iodação do sal de mesa é considerada o caminho mais eficiente para controlar os Distúrbios por Deficiência de Iodo. Em países tropicais, o elemento pode ser adicionado na forma de KIO3. Para garantir que os níveis ideais do ânion estejam disponíveis ao consumidor, o controle de qualidade do sal consiste numa estratégia fundamental. Sistemas em fluxo com multicomutação representam uma alternativa versátil para o desenvolvimento de procedimentos simples, rápidos e limpos, minimizando o consumo de reagentes e a geração de resíduos. Nesse contexto, um procedimento analítico utilizando sistema com multicomutação e detecção espectrofotométrica foi desenvolvido para a determinação de iodato em sal de mesa. A reação empregada foi baseada na formação de um composto roxo (540 nm) entre iodato (IO3-) e p-aminofenol (PAP) em meio ácido. O tempo de residência da zona de amostra no percurso analítico foi explorado a fim de favorecer a reação lenta e a frequência de amostragem para a melhoria do desempenho analítico. Foram selecionados 2 pulsos para inserção de amostra, 3 pulsos para reagente (PAP 0,25% (m/v) em HCl 0,025 mol L-1), 7 ciclos de amostragem, 200 pulsos de carregador (água), bolha de ar de 1 s (40 µL), reator de 70 cm (3 mm d.i.) e parada de fluxo de 480 s. Resposta linear foi observada entre 2,28x10-5 e 3,65x10-4 mol L-1, descrita pela equação A = 0,2443 + 2030 C, r = 0,997. Limite de detecção (99,7% de confiança), coeficiente de variação (n = 20) e frequência de amostragem foram estimados em 8,2x10-6 mol L-1, 0,42% e 70 determinações por hora, respectivamente. Houve consumo de 1,05 mg de PAP e geração de 0,70 mL de resíduos por determinação. As principais espécies concomitantes presentes na amostra não interferiram na determinação de iodato em concentrações até 8 vezes maiores que as usualmente encontradas. Estudos de adição e recuperação de iodato foram realizados pelo procedimento proposto, obtendo porcentagens de recuperação entre 88 e 104%. O procedimento analítico desenvolvido apresenta sensibilidade adequada para a determinação de iodato em amostra de sal de mesa e elevada frequência de amostragem quando comparado com procedimentos descritos na literatura

A method for quantitative mineralogical analysis by ATR-FTIR has been developed. The method relies on the use of the main band of calcite as a reference for the normalization of the IR spectrum of a mineral sample. In this way, the molar absorptivity coefficient in the Lambert–Beer law and the components of a mixture in mole percentage can be calculated. The GAMS equation modeling environment and the NLP solver CONOPT (©ARKI Consulting and Development) were used to correlate the experimental data in the samples considered. Mixtures of different minerals and gypsum were used in order to measure the minimum band intensity that must be considered for calculations and the detection limit. Accordingly, bands of intensity lower than 0.01 were discarded. The detection limit for gypsum was about 7% (mol/total mole). Good agreement was obtained when this FTIR method was applied to ceramic tiles previously analyzed by X-ray diffraction (XRD) or mineral mixtures prepared in the lab.

A chemical sensor based on a coated long-period grating has been prepared and characterized. Designer coatings based on polydimethylsiloxane were prepared by the incorporation of diphenylsiloxane and titanium cross-linker in order to provide enhanced sensitivity for a variety of key environmental pollutants and optimal refractive index of the coating. Upon microextraction of the analyte into the polymer matrix, an increase in the refractive index of the coating resulted in a change in the attenuation spectrum of the long-period grating. The grating was interrogated using ring-down detection as a means to amplify the optical loss and to gain stability against misalignment and power fluctuations. Chemical differentiation of cyclohexane and xylene was achieved and a detection limit of 300 ppm of xylene vapour was realized.

L’utilisation de lentilles cornéennes peut servir à améliorer le profil d’administration d’un principe actif dans les yeux. Avec une efficacité d’administration de 5% par l’utilisation de gouttes, on comprend rapidement que l’administration oculaire doit être améliorée. Cette faible administration a donné naissance à plusieurs tentatives visant à fabriquer des lentilles cornéennes médicamentées. Cependant, à cause de multiples raisons, aucune de ces tentatives n’a actuellement été mise sur le marché. Nous proposons dans cette étude, une possible amélioration des systèmes établis par le développement d’une lentille cornéenne à base de 2-(hydroxyéthyle)méthacrylate (HEMA), dans laquelle des microgels, à base de poly N-isopropylacrylamide (pNIPAM) thermosensible encapsulant un principe actif, seront incorporé. Nous avons donc débuté par développer une méthode analytique sensible par HPCL-MS/MS capable de quantifier plusieurs molécules à la fois. La méthode résultante a été validée selon les différents critères de la FDA et l’ICH en démontrant des limites de quantifications et de détections suffisamment basses, autant dans des fluides simulés que dans les tissus d’yeux de lapins. La méthode a été validée pour sept médicaments ophtalmiques : Pilocarpine, lidocaïne, proparacaïne, atropine, acétonide de triamcinolone, timolol et prednisolone. Nous avons ensuite fait la synthèse des microgels chargés négativement à base de NIPAM et d’acide méthacrylique (MAA). Nous avons encapsulé une molécule modèle dans des particules ayant une taille entre 200 et 600 nm dépendant de la composition ainsi qu’un potentiel zêta variant en fonction de la température. L’encapsulation de la rhodamine 6G (R6G) dans les microgels a été possible jusqu’à un chargement (DL%) de 38%. L’utilisation des isothermes de Langmuir a permis de montrer que l’encapsulation était principalement le résultat d’interactions électrostatiques entre les MAA et la R6G. Des cinétiques de libérations ont été effectuées à partir d’hydrogels d’acrylamide chargés en microgels encapsulant la R6G. Il a été trouvé que la libération des hydrogels chargés en microgels s’effectuait majoritairement selon l’affinité au microgel et sur une période d’environ 4-24 heures. La libération à partir de ces systèmes a été comparée à des formules d’hydrogels contenant des liposomes ou des nanogels de chitosan. Ces trois derniers (liposomes, microgels et nanogels) ont présenté des résultats prometteurs pour différentes applications avec différents profils de libérations. Enfin, nous avons transposé le modèle développé avec les gels d’acrylamide pour fabriquer des lentilles de contact de 260 à 340 µm d’épaisseur à base de pHEMA contenant les microgels avec une molécule encapsulée devant être administrée dans les yeux. Nous avons modifié la composition de l’hydrogel en incorporant un polymère linéaire, la polyvinylpyrrolidone (PVP). L’obtention d’hydrogels partiellement interpénétrés améliore la rétention d’eau dans les lentilles cornéennes. L’encapsulation dans les microgels chargés négativement a donné de meilleurs rendements avec la lidocaïne et cette dernière a été libérée de la lentille de pHEMA en totalité en approximativement 2 heures qu’elle soit ou non encapsulée dans des microgels. Ainsi dans cette étude pilote, l’impact des microgels n’a pas pu être déterminé et, de ce fait, nécessitera des études approfondies sur la structure et les propriétés de la lentille qui a été développée. En utilisant des modèles de libération plus représentatifs de la physiologie de l’œil, nous pourrions conclure avec plus de certitude concernant l’efficacité d’un tel système d’administration et s’il est possible de l’optimiser.

Paleoproductivity, nutrient burial, and carbon cycling were investigated across the Eocene/Oligocene (E/O) boundary (begin to end; 36.9-32.7 Ma at ~40 kyr resolution, timescale of Shackleton et al. (1999, doi:10.1098/rsta.1999.0407) at Ocean Drilling Program Site 925 on the Ceara Rise in the western equatorial Atlantic (3040 m present water depth; 748.26-850.70 mbsf). Downcore bulk sediment records of biogenic barium, total reactive phosphorus, biogenic silica, and calcium carbonate are interpreted to represent export production, net nutrient burial, biogenic opal production, and inorganic carbon burial, respectively. The global positive excursion in d13C subsequent to the E/O boundary is recorded at Site 925. Export production appears to have been externally forced by orbital parameters at eccentricity frequencies during the study interval, based on spectral analysis of the biogenic barium and reactive phosphorus records. Biogenic silica production or preservation increased after the Eocene/Oligocene boundary to a higher baseline, although overall productivity and nutrient burial did not increase, based on barium and reactive phosphorus records. Thus, although absolute production did not increase at this site, a shift in relative abundance of siliceous versus carbonate productivity may have resulted in a change in relative organic carbon burial. This may have contributed to the positive excursion in global oceanic d13C subsequent to the Eocene/Oligocene boundary, although the silica maximum persists after the carbon isotope excursion ends.

One of the objectives of Leg 55 was to investigate the Tertiary history of sedimentation and environment on the Emperor Seamounts after their volcanic activity. For the three first sites, 430, 431, and 432, drilled on Ojin, Nintoku, and Yömei Seamounts, the Neogene sedimentary deposits are not well represented and are not typical pelagic sediments. Except for two holes (430A and 432), where we found calcareous oozes, the sediments are heterogeneous sands, gravels, and pebbly mudstones with a wide range in grain size and composition. Two phenomena characterize these deposits: the inheritance of volcaniclastic material and its alteration, and the authigenesis of secondary minerals including silicates, phosphates, and ferromanganese oxides formed under volcanic influence in a marine environment.

A 10Be/9Be-based chronostratigraphy has been determined for ODP 181, Site 1121 sediment core, recovered from the foot of the Campbell Plateau, Southwest Pacific Ocean. This core was drilled through the Campbell 'skin drift' in ca. 4500 m water depth on the mid-western margin of the extensive Campbell Nodule Field, beneath the flow of the major cold-water Deep Western Boundary Current (DWBC). In the absence of detailed biostratigraphy, beryllium isotopes have provided essential time information to allow palaeo-environmental interpretation to be undertaken on the upper 7 m of the core. Measured 10Be/9Be ratios of sediment, and of ferromanganese nodules entrapped in the sediment, decrease systematically with depth in the core, in accordance with radioactive decay. However, the 10Be/9Be data diverge from ca. 3 m below the seafloor (mbsf) to the top of the core, giving rise to several possible geochronological models. The preferred model assumes that the measured 10Be/9Be ratios of the nodule rims reflect initial 10Be/9Be ratios equivalent to contemporary seawater, and that these can be used to derive the true age of the sediment where the nodules occur. The nodule rim ages can be then used to interpret the sediment 10Be/9Be data, which indicate an overall age to ca. 7 mbsf of ca. 17.5 Ma. The derived chronology is consistent with diatom biostratigraphy, which indicates an age of 2.2-3.6 Ma at 1 mbsf. Calculated sedimentation rates range from 8 to 95 cm m.y.**-1, with an overall rate to 7 mbsf of ca. 39 cm m.y.**-1. The lowest rates generally coincide with the occurrence of entrapped nodules, and reflect periods of increased bottom current flow causing net sediment loss. Growth rates of individual nodules decrease towards the top of the sediment core, similar to the observed decrease in growth rate from core to rim of seafloor nodules from the Campbell Nodule Field. This may be related to an overall increase in the vigour of the DWBC from ca. 10 Ma to the present.

X-ray fluorescence (XRF) scanning of sediment cores from the Lomonosov Ridge and the Morris Jesup Rise reveals a distinct pattern of Ca intensity peaks through Marine Isotope Stages (MIS) 1 to 7. Downcore of MIS 7, the Ca signal is more irregular and near the detection limit. Virtually all major peaks in Ca coincide with a high abundance of calcareous microfossils; this is particularly conspicuous in the cores from the central Arctic Ocean. However, the recorded Ca signal is generally caused by a combination of biogenic and detrital carbonate, and in areas influenced by input from the Canadian Arctic, detrital carbonates may effectively mask the foraminiferal carbonates. Despite this, there is a strong correlation between XRF-detected Ca content and foraminiferal abundance. We propose that in the Arctic Ocean north of Greenland a common palaeoceanographic mechanism is controlling Ca-rich ice-rafted debris (IRD) and foraminiferal abundance. Previous studies have shown that glacial periods are characterized by foraminfer-barren sediments. This implies that the Ca-rich IRD intervals with abundant foraminifera were most likely deposited during interglacial periods when glaciers left in the Canadian Arctic Archipelago were still active and delivered a large amount of icebergs. At the same time, conditions were favourable for planktic foraminifera, resulting in a strong covariance between these proxies. Therefore, we suggest that the XRF scanner's capability to efficiently map Ca concentrations in sediment cores makes it possible to systematically examine large numbers of cores from different regions to investigate the palaeoceanographic reasons for the calcareous microfossils' spatial and temporal variability.

In the absence of an external frame of reference-i.e., in background independent theories such as general relativity-physical degrees of freedom must describe relations between systems. Using a simple model, we investigate how such a relational quantum theory naturally arises by promoting reference systems to the status of dynamical entities. Our goal is twofold. First, we demonstrate using elementary quantum theory how any quantum mechanical experiment admits a purely relational description at a fundamental. Second, we describe how the original non-relational theory approximately emerges from the fully relational theory when reference systems become semi-classical. Our technique is motivated by a Bayesian approach to quantum mechanics, and relies on the noiseless subsystem method of quantum information science used to protect quantum states against undesired noise. The relational theory naturally predicts a fundamental decoherence mechanism, so an arrow of time emerges from a time-symmetric theory. Moreover, our model circumvents the problem of the collapse of the wave packet as the probability interpretation is only ever applied to diagonal density operators. Finally, the physical states of the relational theory can be described in terms of spin networks introduced by Penrose as a combinatorial description of geometry, and widely studied in the loop formulation of quantum gravity. Thus, our simple bottom-up approach (starting from the semiclassical limit to derive the fully relational quantum theory) may offer interesting insights on the low energy limit of quantum gravity.

A direct quadrupole ICP-MS technique has been developed for the analysis of the rare earth elements and yttrium in natural waters. The method has been validated by comparison of the results obtained for the river water reference material SLRS-4 with literature values. The detection limit of the technique was investigated by analysis of serial dilutions of SLRS-4 and revealed that single elements can be quantified at single-digit fg/g concentrations. A coherent normalised rare earth pattern was retained at concentrations two orders of magnitude below natural concentrations for SLRS-4, demonstrating the excellent inter-element accuracy and precision of the method. The technique was applied to the analysis of a diluted mid-salinity estuarine sample, which also displayed a coherent normalised rare earth element pattern, yielding the expected distinctive marine characteristics. We use a functional integral formalism developed earlier for the pure Luttinger liquid (LL) to find an exact representation for the electron Green function of the LL in the presence of a single backscattering impurity in the low-temperature limit. This allows us to reproduce results (well known from the bosonization techniques) for the suppression of the electron local density of states (LDOS) at the position of the impurity and for the Friedel oscillations at finite temperature. In addition, we have extracted from the exact representation an analytic dependence of LDOS on the distance from the impurity and shown how it crosses over to that for the pure LL.

We examine the existence and stability of discrete spatial solitons in coupled nonlinear lasing cavities (waveguide resonators), addressing the case of active defocusing media, where the gain exceeds damping in the low-amplitude limit. A new family of stable localized structures is found: these are bright and gray cavity solitons representing the connections between homogeneous and inhomogeneous states. Solitons of this type can be controlled by discrete diffraction and are stable when the bistability of homogenous states is absent. © 2012 Optical Society of America.

Fabrication and characterization of a UVinscribed fiber Bragg grating (FBG) with a micro-slot liquid core is presented. Femtosecond (fs) laser patterning/chemical etching technique was employed to engrave a micro-slot with dimensions of 5.74 μm(h) × 125 μm(w) × 1388.72 μm(l) across the whole grating. The device has been evaluated for refractive index (RI) and temperature sensitivities and exhibited distinctive thermal response and RI sensitivity beyond the detection limit of reported fiber gratings. This structure has not just been RI sensitive, but also maintained the robustness comparing with the bare core FBGs and long-period gratings with the partial cladding etched off. © 2012 Optical Society of America.
