Zielsetzung ist der synthetische Zugang zu metallorganischen Verbindungen, die Propylenoxid koordinativ polymerisieren und deren Aktivität sich durch gezielte Variation der sterischen oder elektronischen Eigenschaften kontrollieren läßt. Zur Bearbeitung dieser Aufgabe werden zwei verschiedene Klassen von Komplexen synthetisiert und charakterisiert: Vorwiegend durch Salzmetathese werden mehrere Yttriumamide der allgemeinen Formel Y(NRR´)3(THF)x erhalten. Dabei erfolgt eine Diskussion der Auswirkungen sterischer und elektronischer Variation der beiden Amido-Substituenten R und R´, die sich in der Bildung von „at“-Komplexen, Lösungsmitteladdukten und größeren Aggregaten äußern. Durch Alkaneliminierung wird eine umfassende Reihe von Aluminiumkomplexen methylen- und thioverbrückter Bisphenolatoliganden dargestellt. Die Verbindungen besitzen unter Ausschluß zusätzlicher Basen dimere oder mehrkernige Strukturen, deren Diskussion schwerpunktsmäßig anhand ihres Verhaltens in Lösung erfolgt. Die Aluminiumkomplexe werden in anschließenden Reaktionen zu Lösungsmitteladdukten und Aluminaten umgesetzt.Die Yttriumamide initiieren die Ringöffnungspolymerisation von Propylenoxid bei Temperaturen von 60 oder 80 °C. Die Polymerisation erfolgt nach einem koordinativen Mechanismus, eine zu niedrige Katalysatoreffizienz schließt jedoch die Yttriumamide als Vorläufer zu definierten „single site“-Katalysatoren aus. Mit der Kombination aus neutralen Aluminiumkomplexen mit den entsprechenden Aluminaten verläuft die Ringöffnungspolymerisation von Propylenoxid bei Raumtemperatur schnell und kontrolliert. Es läßt sich ein prinzipiell neuer Mechanismus belegen, bei dem die Polymerisation unter synergistischer Wirkung eines Aluminiumphenolato-Komplexes mit dem korrespondierenden „at“-Komplex erfolgt.

In der vorliegenden Arbeit konnte gezeigt werden, dass sowohl die durch TNFalpha-, als auch durch Röntgenstrahlen vermittelte Expression des Adhäsionsmoleküls E-Selektin in Endothelzellen durch kleine GTPasen der Rho-Proteinfamilie reguliert ist. Hemmung dieser kleinen G-Proteine z.B. durch HMG-CoA Reduktase-Inhibitoren (Statine) oder clostridiale Toxine führt zu verminderter Expression von E-Selektin in humanen Endothelzellen (HUVEC; EA.hy-926). Aus den in der Arbeit erhaltenen Ergebnissen kann außerdem die Schlussfolgerung gezogen werden, dass die Regulation der zytokininduzierten E-Selektin-Genexpression von der gamma-Strahlen-vermittelten Expression des E-Selektingens differiert. Für die strahleninduzierte endotheliale E-Selektin-Expression scheint beispielsweise der Transkriptionsfaktor AP-1 als ein weiterer Kontrollfaktor neben NF-kappaB zu fungieren. Des Weiteren konnte in der vorliegenden Arbeit gezeigt werden, dass eine gesteigerte E-Selektin-Proteinexpression mit erhöhter Tumorzelladhäsion und -transmigration an bzw. durch humane Endothelzellen korreliert. Hemmung der TNFalpha- bzw. gamma-Strahlen-induzierten Expression von E-Selektin durch Statine oder Retinsäuren ist ausreichend, sowohl Tumorzelladhäsion als auch Tumorzelldiapedese zu reduzieren. Zusammenfassend lässt sich festhalten, dass das Adhäsionsmolekül E-Selektin eine vielversprechende Zielstruktur ist, über deren kontrollierte Beeinflussung es auch in vivo möglich sein sollte, das Risiko der Metastasierung zu reduzieren. Mit Statinen und Retinsäurederivaten wurden somit Pharmaka identifiziert, die durch Hemmung der Rho-regulierten E-Selektin-Expression der Tumorzellmetastasierung vorbeugen könnten. Um diese Hypothese in zukünftigen tierexperimentelle Studien zu bestätigen, wurde im Rahmen dieser Arbeit mit der Generierung transgener Tiermodelle begonnen.

The aim of this thesis was to investigate novel techniques to create complex hierarchical chemical patterns on silica surfaces with micro to nanometer sized features. These surfaces were used for a site-selective assembly of colloidal particles and oligonucleotides. To do so, functionalised alkoxysilanes (commercial and synthesised ones) were deposited onto planar silica surfaces. The functional groups can form reversible attractive interactions with the complementary surface layers of the opposing objects that need to be assembled. These interactions determine the final location and density of the objects onto the surface. Photolithographically patterned silica surfaces were modified with commercial silanes, in order to create hydrophilic and hydrophobic regions on the surface. Assembly of hydrophobic silica particles onto these surfaces was investigated and finally, pH and charge effects on the colloidal assembly were analysed. In the second part of this thesis the concept of novel, "smart" alkoxysilanes is introduced that allows parallel surface activation and patterning in a one-step irradiation process. These novel species bear a photoreactive head-group in a protected form. Surface layers made from these molecules can be irradiated through a mask to remove the protecting group from selected regions and thus generate lateral chemical patterns of active and inert regions on the substrate. The synthesis of an azide-reactive alkoxysilane was successfully accomplished. Silanisation conditions were carefully optimised as to guarantee a smooth surface layer, without formation of micellar clusters. NMR and DLS experiments corroborated the absence of clusters when using neither water nor NaOH as catalysts during hydrolysis, but only the organic solvent itself. Upon irradiation of the azide layer, the resulting nitrene may undergo a variety of reactions depending on the irradiation conditions. Contact angle measurements demonstrated that the irradiated surfaces were more hydrophilic than the non-irradiated azide layer and therefore the formation of an amine upon irradiation was postulated. Successful photoactivation could be demonstrated using condensation patterns, which showed a change in wettability on the wafer surface upon irradiation. Colloidal deposition with COOH functionalised particles further underlined the formation of more hydrophilic species. Orthogonal photoreactive silanes are described in the third part of this thesis. The advantage of orthogonal photosensitive silanes is the possibility of having a coexistence of chemical functionalities homogeneously distributed in the same layer, by using appropriate protecting groups. For this purpose, a 3',5'-dimethoxybenzoin protected carboxylic acid silane was successfully synthesised and the kinetics of its hydrolysis and condensation in solution were analysed in order to optimise the silanisation conditions. This compound was used together with a nitroveratryl protected amino silane to obtain bicomponent surface layers. The optimum conditions for an orthogonal deprotection of surfaces modified with this two groups were determined. A 2-step deprotection process through a mask generated a complex pattern on the substrate by activating two different chemistries at different sites. This was demonstrated by colloidal adsorption and fluorescence labelling of the resulting substrates. Moreover, two different single stranded oligodeoxynucleotides were immobilised onto the two different activated areas and then hybrid captured with their respective complementary, fluorescent labelled strand. Selective hybridisation could be shown, although non-selective adsorption issues need to be resolved, making this technique attractive for possible DNA microarrays.

Aufgaben der vorliegenden Untersuchungen waren die Etablierung von planaren Multilayern aus mensch­lichen Tumorzellen (WiDr und SiHa) und die Testung dieses Zellsystems als Bestrahlungsmodell solider Tumoren. Neben der konventionellen Röntgenbestrahlung (250 kV) wurde auch das Überle­ben nach Schwerionenbestrahlung (12C6+) und nach Behandlung mit dem Chemotherapeutikum Etoposid unter­sucht. Multilayer aus beiden Zelllinien zeigten ein geringeres Überleben nach Röntgen- und Schwerionenbestrah­lung als die entsprechenden Monolayer. Die hier beschriebene multizelluläre Sensitivierung steht aller­dings im Ge­gensatz zu der in der Literatur beschriebenen multizellulären Resistenz der Sphäroide, dem sog. Kontakteffekt. Nach durchflußzytometrischen Mes­sungen arretierten die bestrahlten SiHa-Zellen in der G2/M-Phase. Im Gegen­satz zum transienten Block der Monolayer verweilten die Multilayer in einem per­manenten Arrest. Im Vergleich zur Röntgenbe­strahlung verlän­gerte sich die Arrestzeit der Mono­layer nach Schwerionenbestrahlung im Bragg-Peak um 12-24 h. Auch waren mehr Zellen betroffen. Im Gegensatz dazu war kein Unterschied zwischen beiden Bestrahlungsmo­dalitäten bei den Multi­layern bis zum Ende des Beobachtungszeit­raumes zu verzeichnen. Nach Etoposid-Behandlung verhielten sich die Multilayer deutlich resistenter als die Monolayer. Somit zeigten Multilayer interessan­terweise nach Bestrahlung eine Sensitivierung und nach Etoposid-Behandlung eine Resistenz. Die Unterschiede im Überleben der beiden Kultivierungs­formen beruhen zum Großteil auf den Differenzen in der Zellzyk­lusverteilung. Besonders deutlich wurde dieser Zusam­men­hang zwischen Überleben und Zell­zyklusvertei­lung durch Wie­deraussaat- und Synchronisations-Experi­mente.

Nach einer kurzen Einführung in die Entwicklung der magnetischen Anwendungen, werden in Kapitel 2 und 3 die physikalischen Grundlagen der Messmethoden, insbesondere die Rastertunnelspektroskopie und Kerr-Magnetometrie, sowie der experimentelle Aufbau erläutert. Kapitel 4 beschäftigt sich mit den magnetischen Eigenschaften von quasi ein-dimensionalen ferromagnetischen Nanostreifen und Monolagen, die durch Selbstorganisation auf einem Wolfram(110)-Einkristall mit vizinaler und glatter Oberfläche präpariert werden. Hierbei wird die Temperaturabhängigkeit der magnetischen Größen, wie Remanenz, Sättigungsmagnetisierung und Suszeptibilität, sowie die Auswirkung einer Abdeckung des Systems auf die Domänenwandenergie und Anisotropie untersucht. Zusätzlich wird die Kopplung von parallelen Nanostreifen in Abhängigkeit des Streifenabstandes betrachtet. In Kapitel 5 werden das Wachstum und die Morphologie von Co-Monolagen auf W(110) untersucht. Der Übergang von pseudomorphem zu dicht gepacktem Wachstum in der Monolage wird mit Hilfe der Rastertunnelspektroskopie sichtbar gemacht, ebenso wie unterschiedliche Stapelfolgen in Tripellagen Co-Systemen. Atomar aufgelöste Rastertunnelmikroskopie erlaubt die genauen Atompositionen der Oberfläche zu bestimmen und mit theoretischen Wachstumsmodellen zu vergleichen. Auf die Untersuchung zwei-dimensionaler binärer Co-Fe und Fe-Mn Legierungen auf W(110) wird in Kapitel 6 eingegangen. Mit einer Präparationstemperatur von T=520 K ist es möglich, atomar geordnete Co-Fe Legierungsmonolagen wachsen zu lassen. Ein direkter Zusammenhang zwischen der Magnetisierung und der lokalen Zustandsdichte in Abhängigkeit der Legierungszusammensetzung wird gezeigt.

Tethered bilayer lipid membranes provide an efficient, stable and versatile platform for the investigation of integrated membrane proteins. However, the incorporation of large proteins, as well as of proteins with a large submembrane part is still a very critical issue and therefore, further optimisation of the system is necessary. The central element of a tBLM is a lipid bilayer. Its proximal leaflet is, at least to some extend, covalently attached to a solid support via a spacer group. The anchor lipid consists of three distinct parts, a lipid headgroup, a spacer group and an anchor. All parts together influence the final bilayer properties. In the frame of this work, the synthesis of new thiolipids for tBLMs on gold has been investigated. The aim was to obtain molecules with longer spacers in order to increase the submembrane space. The systems obtained have been characterized using SPR and EIS. The results obtained during this study are multiple. First, the synthesis of a previously synthesized architecture was successfully scaled up in an industrial lab using a new synthetic approach. The synthesis of large amounts is now feasible. Then, the synthesis of the new thiolipids was carried out taking into account the following requirements: the increase of the submembrane space by having longer ethyleneglycol spacers, the attachment of the molecules to a gold substrate via a thiol bond, and the tunability of the lateral mobility by changing the lipid headgroup. Three different synthetic strategies have been investigated. The polymeric approach did not prove to be successful, merely because of the broad molecular weight distribution. The synthesis of heterofunctionally protected oligoethyleneglycols allowed to obtain ethyleneglycol moieties with 6 and 8 units, but the tedious purification steps gave very low yields. Finally, the block by block synthesis using ethyleneglycol precursors proved to be an efficient and fast method to synthesize the target molecules. Indeed, these were obtained with very high yields, and the separation was very efficient. A whole family of new compounds was obtained, having 6, 8 and 14 ethyleneglycol units and with mono- or diphytanyl lipid headgroups. This new pathway is a very promising synthetic strategy that can be used further in the development of new compounds of the tether system. The formation of bilayers was investigated for the different thiolipids mainly by using EIS. The electrical properties of a bilayer define the quality of the membrane and allow the study of the functionality of proteins embedded in such a system. Despite multiple trials to improve the system using self assembly, Langmuir Blodgett transfer, and detergent mixed vesicles, the new polymer thiolipids did not show as high electrical properties as tBLMs reported in the literature. Nevertheless, it was possible to show that a bilayer could be obtained for the different spacer lengths. These bilayers could be formed using self assembly for the first monolayer, and two different methods for bilayer formation, namely vesicle fusion and solvent exchange. We could furthermore show functional incorporation of the ion carrier valinomycin: the selective transport of K+ ions could be demonstrated. For DPHL, it was even possible to show the functional incorporation of the ion channel gramicidin. The influence of the spacer length is translated into an increase of the spacer capacitance, which could correspond to an increase in the capacity of charge accumulation in the submembrane space. The different systems need to be further optimised to improve the electrical properties of the bilayer. Moreover, the incorporation of larger proteins, and proteins bearing submembrane parts needs to be investigated.

Ziel war Herstellung und Charakterisierung festkörperunterstützter Membransysteme aus Glykolipopolymeren auf Goldoberflächen, mit elektrischen Eigenschaften, die denen der Schwarzfilmmembranen entsprechen. Um diese Eigenschaften mit impedanzspektroskopischen Techniken messen zu können, ist die Präparation der Membranen auf Goldfilmen notwendig. Eine direkte Verankerung der Glykolipopolymermoleküle (GLP) auf der Goldoberfläche ist nicht möglich, daher werden die untersuchten GLP mit Hilfe von photoreaktiven Molekülen kovalent auf der Goldoberfläche immobilisiert. Untersuchten Abstandhalter waren Thio-Polyethylenglykol und eine Mischung zweier Alkanthiole, bestehend aus 1-Thiohexanol und Bis-(Aminododecyl)-disulfid. Thio-PEG-Monoschichten zeigten sehr unterschiedliche Schichtdicken, weil die Moleküle auf der Oberfläche sehr unterschiedliche Konformationen annehmen können, die eine regelmässige Anordnung erschweren. Langmuir-Blodgett Übertrag sowie die durchgeführte photochemische Fixierung der GLP-Moleküle auf Thio-PEG Schichten führte zu Eigenschaften, die auf den Aufbau einer Lipidmonolage hinweisen. Diese stellte jedoch im Bereich der Lipidmoleküle keine geschlossene Schicht dar. Es kommen als Abstandhaltermoleküle für die Photofunktionalisierung nur Moleküle in Frage, die aufgrund ihrer Eigenschaften eine regelmässige Anordnung auf der Goldoberfläche anstreben. Diese Voraussetzung erfüllt am besten die Gruppe der Alkanthiole. Terminal funktionalisierte Alkanthiole müssen jedoch mit nicht oder anderweitig funktionalisierten Alkanthiolen verdünnt assembliert werden, um weitergehende Funktionalisierungen einzugehen. Die untersuchten GLP lassen sich aufgrund ihrer amphiphilen Struktur an der Wasser-Luft Grenzfläche vororientieren. In allen Fällen gelingt auch der Langmuir-Blodgett Übertrag der komprimierten Schicht, sowie, nach der für die Anbindung notwendigen Trocknung, die photochemisch kovalente Fixierung der Monoschicht durch Bestrahlung mit UV-Licht. Damit konnte erstmals die photochemisch kovalente Fixierung von GLPs auf der Goldoberfläche gezeigt werden. Untersuchungen erfolgten an einem Copolymer sowie zwei unterschiedlichen Homopolymermolekülen. Der unterschiedliche molekulare Aufbau der GLP spiegelt sich in ihrem Verhalten an der Wasser-Luft Grenzfläche sowie in den Eigenschaften der gebildeten Monoschichten wieder. Die Copolymer-Schichten zeigten sehr unterschiedliche Schichtdicken. Auch die EIS-Daten sind schlecht reproduzierbar. Dies ist auf die molekulare Struktur des Copolymers zurückzuführen. Gänzlich unterschiedlich verhalten sich die Homopolymere. Aufgrund ihrer Struktur lassen sie sich zu dichten Schichten komprimieren. Messungen der Fluoreszenzerholung nach Photobleichung (FRAP) zeigen homogene aber nicht fluide Schichten. Die photochemisch kovalente Fixierung des Moleküls auf der Goldoberfläche konnte durch SPR- sowie EIS-Messungen nachgewiesen werden. Die EIS-Messungen zeigen Werte, die sich in Bereichen der idealen Modellmembran bewegen. Der erfolgreiche Einbau von Valinomycin konnte bestätigt werden. FRAP Untersuchungen zeigten die Bildung homogener Schichten. Diese sind jedoch im Bereich der proximalen Lipidschicht nicht fluide. Um die Fluidität in Anlehnung an die Eigenschaften der natürlichen Membranen zu erhöhen, wurden die photochemisch kovalent fixierten Anker-Glykolipopolymer-Moleküle durch Mischung mit freien Lipiden lateral verdünnt. Auch die kovalente Fixierung der GLP-Bausteine innerhalb gemischten Schichten konnte erfolgreich demonstriert werden. Die Schichten zeigten sich jedoch, mit Ausnahme der Schichten aus 50mol% Homopolymer und 50mol% Lipid, inhomogen. Nach Photobleichung durch den Laserblitz kam es nur bei den 50mol%:50mol% -Schichten (Ho: Lipid) zur Erholung der Fluoreszenz, was auf das Vorliegen von beweglichen Lipidmolekülen innerhalb der Membran schliessen lässt. Der Versuch der Inkorporation von Valinomycin gelang ebenfalls. Alle genannten Ergebnisse deuten darauf hin, dass die molekulare Architektur der hergestellten Schichten durch die unterschiedlichen Längendimensionen des Homopolymer-Moleküls einerseits, sowie des Lipids andererseits nicht für alle Mischungsverhältnisse ausreichend stabil ist. Die für die kovalente Fixierung erforderliche Trocknung der Schicht führt zu einer deutlichen Verminderung des Wassergehaltes des Systems und einer daraus resultierenden starken Destabilisierung der aufgebauten Schichten. Insgesamt gesehen stellt somit die photochemische Fixierung der glykosidischen Homopolymer-Membranen ein vielversprechendes Modellsystem dar.

Many studies on the morphology, molecular orientation, device performance, substrate nature and growth parameter dependence have been carried out since the proposal of Sexithiophene (6T) for organic electronics [ ] However, these studies were mostly performed on films thicker than 20nm and without specifically addressing the relationship between morphology and molecular orientation within the nano and micro structures of ultrathin films of 0-3 monolayers. In 2004, the observation that in OFETs only the first few monolayers at the interface in contact with the gate insulator contribute to the charge transport [ ], underlined the importance to study submonolayer films and their evolution up to a few monolayers of thickness with appropriate experimental techniques. We present here a detailed Non-contact Atomic Force Microscopy and Scanning Tunneling Microscopy study on various substrates aiming at the investigation of growth mechanisms. Most reported similar studies are performed on ideal metals in UHV. However it is important to investigate the details of organic film growth on less ideal and even technological surfaces and device testpatterns. The present work addresses the growth of ultra thin organic films in-situ and quasi real-time by NC-AFM. An organic effusion cell is installed to evaporate the organic material directly onto the SPM sample scanning stage.

Calix[4]arenes with urea functions attached to the p-positions of the phenolic units usually form dimers in apolar solvents. Tetraureas functionalized by pyridyl and carboxyl groups form dimers only with bis- or tetraloop tetraureas. This heterodimerization was used for the synthesis of a bis-[3]catenane. Tetraureas functionalized with sulfide functions were synthesized for the preparation of monolayers from the dimeric capsules containing electrochemically active guests on gold. Bis-tetraureacalix[4]arenes singly-linked via their wide rim by rigid spacers were synthesized and their self-assembly to polymers in apolar solvents was proved by the 1H NMR spectroscopy and AFM studies. Dimerization of the first example of the tetraurea calix[4]arenes bridged in 1,3-positions at the narrow rim was proved by 1H NMR spectroscopy. Calix[8]arenes functionalized by urea, amido or naphthalimido groups at their p-positions self-assemble to columnar structures by hydrogen bonding or by π-π-stacking.

Nature leads, we follow. But nanotechnologists are in hot pursuit, in designing controllable structures that can mimic naturally occurring and artificially synthesized materials on a common platform. The supramolecular chemistry concerns the investigation of nature principles to produce fascinating complexed and functional molecular assemblies, as well as the utilization of these principles to generate novel devices and materials, potentially useful for sensing, catalysis, transport and other applications in medical or engineering science. The work presented in this thesis is a compilation of different synthetic methods to achieve inorganic-organic hybrid nanomaterials. Silicatein, a protein enzyme, which acts both as a catalyst and template for the formation of silica needles in marine sponges, has been used for the biosynthesis of semiconductor metal oxides on surfaces. Silicatein was immobilized on gold (111) surfaces using alkane thiol, as well as on a novel self-assembly of NTA on top of a “cushion” of reactive ester polymer has been successfully employed to make functionalised surfaces. The immobilization of silicatein on surfaces was monitored by surface plasmon spectroscopy, atomic force microscopy and confocal laser scanning microscopy. Surface bound silicatein retains its biocatalytic activity, which was demonstrated by monitoring its hydrocatalytic activity to catalyse the synthesis of biosilica, biotitania, and biozirconia. The synthesis of semiconductor metal oxides was characterized using scanning electron microscopy. This hydrolytic biocatalyst is used to synthesize the gold nanoparticles. The gold nanoparticles are formed by reduction of tetrachloroaurate, AuCl4-, by the action of sulfhydryl groups hidden below the surface groups of the protein. The resulting gold nanoparticles which are stabilized by surface bound silicatein further aggregate to form Au nanocrystals. The shape of the nanocrystals obtained by using recombinant silicatein is controlled through chiral induction by the protein during the nucleation of the nanocrystals. As an extension of this work, TiO2 nanowires were functionalized using polymeric ligand which incorporates the nitrilotriacetic acid (NTA) linker in the back bone to immobilize His-tagged silicatein onto the TiO2 nanowires. The surface bound protein not only retains its original hydrolytic properties, but also acts as a reductant for AuCl4- in the synthesis of hybrid TiO2/silicatein/Au nanocomposites. Functionalized, monocrystalline rutile TiO2 nanorods were prepared from TiCl4 in aqueous solution in the presence of dopamine. The surface bound organic ligand controls the morphology as well as the crystallinity and the phase selection of TiO2. The surface amine groups can be tailored further with functional molecules such as dyes. As an example, this surface functionality is used for the covalent binding of a fluorescent dye,4-chloro-7- nitrobenzylurazene (NBD) to the TiO2 nanorods. The polymeric ligands have been used successfully for the in-situ and post-functionalization of TiO2 nanoparticles. Besides to chelating dopamine anchor group the multifunctional ligand system presented here incorporates a modifier molecule which allows the binding of functional molecules (here the dyes pyrene, NBD, and Texas Red) as well as additional entities which allow tailoring the solubility of inorganic nanocrystals in different solvents. A novel method for the surface functionalization of fullerene-type MoS2 nanoparticles and subsequently binding these nanoparticles onto TiO2 nanowires has been reported using polymeric ligands. The procedure involves the complexation of IF-MoS2 with a combination of Ni2+ via an umbrella-type nitrilotriacetic acid (NTA) and anchoring them to the sidewalls of TiO2 nanowires utilizing the hydroxyl groups of dopamine present in the main contents of polymeric ligand. A convenient method for the synthesis of Au/CdS nanocomposites has been presented, which were achieved through the novel method of thiol functionalization of gold colloids. The thermodynamically most stable phase of ZrO2 (cubic) has been obtained at much lower temperature (180°C). These nanoparticles are highly blue fluorescent, with a high surface area.

In der vorliegenden Arbeit konnte mit Hilfe von neu synthetisierten Monomeren und Polymeren Kristallisationstemplate für die Kalziumkarbonatkristallisation dargestellt werden. Hierzu wurde zunächst die Phasenseparation des Monomeren und Polymeren mit der nukleationsfördernden Stearinsäure detailliert untersucht und gezeigt, dass die Monomer und Polymer-Mischsysteme auf Ca(HCO3)2-Lsg. perfekt phasenseparieren. Weiter konnte diese entmischte Struktur mit Hilfe von UV-Bestrahlung in der Monoschicht polymerisiert und damit fixiert werden. Des weiteren konnte gezeigt werden, dass die Kristallisation unter den monomeren und polymeren Bereichen vollständig inhibiert wird und ausschließlich unter den Stearinsäuredomänen von statten geht. Das Problem der Kristallisation in die dritte Dimension, also ins Volumen, konnte durch die Zugabe von Polyacrylsäure zur Subphase kontrolliert werden, so dass nun eine Abbildung der strukturierten Monoschicht durch Kalkkristallisation möglich ist. Im zweiten Teil der Arbeit konnte ein amphiphiles Itaconat synthetisiert werden, welches auf Grund seiner Carbonsäurefunktion ebenfalls nukleationsfördernd wirkt. Auch hier war es möglich die Monoschicht zu polymerisieren. Weiter konnte erneut gezeigt werden, dass es möglich ist mit Hilfe von Polyacrylsäurezusatz die interne Struktur der Membran als Templat für die Kristallisation zu nutzen, so dass auch dieses System zur gezielten Kalkkristallisation genutzt werden kann.

Antimicrobial peptides (AMPs) are an important component of the innate immune system of the plants. Plant defensins are a large family of antimicrobial peptides with several interesting features, such as small dimension, high stability and broad spectrum of action. The discovery of new molecules and the study of their mechanism of action allow to consider them attractive for biotechnological applications. In this PhD thesis a defensin from Prunus persica (PpDFN1) and four novel DEFensin Like (DEFL) peptides from Vitis vinifera have been studied. In order to characterize the antimicrobial activity of these molecules, the recombinant mature peptides have been expressed in Escherichia coli and purified to homogeneity by chromatography techniques. PpDFN1 is able to inhibit the growth of B. cinerea, P. expansum and M. laxa with different intensity. The recombinant peptide is capable of membrane permeabilization as demonstrated by SYTOX green fluorescence uptake in treated mycelia. Its interaction with membranes containing sphingolipid species has been shown by artificial lipid monolayers. Furthermore, PpDFN1 displays stronger interaction with monolayers composed by lipids extracted from sensitive fungi with the highest interaction against P. expansum, the most sensitive fungi to PpDFN1 action. DEFL 13, a defensin from grapevine, resulted the strongest antibotrytis peptides. It is electrostatically attracted to the fungal membranes as shown by the antagonist effect of the cations and is able to membrane permeabilization in B. cinerea hyphae. DEFL 13 is internalized in fungal cells and leads to fungal death by activation of some signaling pathways as demonstrated by screening of a mutant collection of B. cinerea

Ein neu entwickeltes globales Atmosphärenchemie- und Zirkulationsmodell (ECHAM5/MESSy1) wurde verwendet um die Chemie und den Transport von Ozonvorläufersubstanzen zu untersuchen, mit dem Schwerpunkt auf Nichtmethankohlenwasserstoffen. Zu diesem Zweck wurde das Modell durch den Vergleich der Ergebnisse mit Messungen verschiedenen Ursprungs umfangreich evaluiert. Die Analyse zeigt, daß das Modell die Verteilung von Ozon realistisch vorhersagt, und zwar sowohl die Menge als auch den Jahresgang. An der Tropopause gibt das Modell den Austausch zwischen Stratosphäre und Troposphäre ohne vorgeschriebene Flüsse oder Konzentrationen richtig wieder. Das Modell simuliert die Ozonvorläufersubstanzen mit verschiedener Qualität im Vergleich zu den Messungen. Obwohl die Alkane vom Modell gut wiedergeben werden, ergibt sich einige Abweichungen für die Alkene. Von den oxidierten Substanzen wird Formaldehyd (HCHO) richtig wiedergegeben, während die Korrelationen zwischen Beobachtungen und Modellergebnissen für Methanol (CH3OH) und Aceton (CH3COCH3) weitaus schlechter ausfallen. Um die Qualität des Modells im Bezug auf oxidierte Substanzen zu verbessern, wurden einige Sensitivitätsstudien durchgeführt. Diese Substanzen werden durch Emissionen/Deposition von/in den Ozean beeinflußt, und die Kenntnis über den Gasaustausch mit dem Ozean ist mit großen Unsicherheiten behaftet. Um die Ergebnisse des Modells ECHAM5/MESSy1 zu verbessern wurde das neue Submodell AIRSEA entwickelt und in die MESSy-Struktur integriert. Dieses Submodell berücksichtigt den Gasaustausch zwischen Ozean und Atmosphäre einschließlich der oxidierten Substanzen. AIRSEA, welches Informationen über die Flüssigphasenkonzentration des Gases im Oberflächenwasser des Ozeans benötigt wurde ausgiebig getestet. Die Anwendung des neuen Submodells verbessert geringfügig die Modellergebnisse für Aceton und Methanol, obwohl die Verwendung einer vorgeschriebenen Flüssigphasenkonzentration stark den Erfolg der Methode einschränkt, da Meßergebnisse nicht in ausreichendem Maße zu Verfügung stehen. Diese Arbeit vermittelt neue Einsichten über organische Substanzen. Sie stellt die Wichtigkeit der Kopplung zwischen Ozean und Atmosphäre für die Budgets vieler Gase heraus.

In this thesis, we investigated the evaporation of sessile microdroplets on different solid substrates. Three major aspects were studied: the influence of surface hydrophilicity and heterogeneity on the evaporation dynamics for an insoluble solid substrate, the influence of external process parameters and intrinsic material properties on microstructuring of soluble polymer substrates and the influence of an increased area to volume ratio in a microfluidic capillary, when evaporation is hindered. In the first part, the evaporation dynamics of pure sessile water drops on smooth self-assembled monolayers (SAMs) of thiols or disulfides on gold on mica was studied. With increasing surface hydrophilicity the drop stayed pinned longer. Thus, the total evaporation time of a given initial drop volume was shorter, since the drop surface, through which the evaporation occurs, stays longer large. Usually, for a single drop the volume decreased linearly with t1.5, t being the evaporation time, for a diffusion-controlled evaporation process. However, when we measured the total evaporation time, ttot, for multiple droplets with different initial volumes, V0, we found a scaling of the form V0 = attotb. The more hydrophilic the substrate was, the more showed the scaling exponent a tendency to an increased value up to 1.6. This can be attributed to an increasing evaporation rate through a thin water layer in the vicinity of the drop. Under the assumption of a constant temperature at the substrate surface a cooling of the droplet and thus a decreased evaporation rate could be excluded as a reason for the different scaling exponent by simulations performed by F. Schönfeld at the IMM, Mainz. In contrast, for a hairy surface, made of dialkyldisulfide SAMs with different chain lengths and a 1:1 mixture of hydrophilic and hydrophobic end groups (hydroxy versus methyl group), the scaling exponent was found to be ~ 1.4. It increased to ~ 1.5 with increasing hydrophilicity. A reason for this observation can only be speculated: in the case of longer hydrophobic alkyl chains the formation of an air layer between substrate and surface might be favorable. Thus, the heat transport to the substrate might be reduced, leading to a stronger cooling and thus decreased evaporation rate. In the second part, the microstructuring of polystyrene surfaces by drops of toluene, a good solvent, was investigated. For this a novel deposition technique was developed, with which the drop can be deposited with a syringe. The polymer substrate is lying on a motorized table, which picks up the pendant drop by an upward motion until a liquid bridge is formed. A consecutive downward motion of the table after a variable delay, i.e. the contact time between drop and polymer, leads to the deposition of the droplet, which can evaporate. The resulting microstructure is investigated in dependence of the processes parameters, i.e. the approach and the retraction speed of the substrate and the delay between them, and in dependence of the intrinsic material properties, i.e. the molar mass and the type of the polymer/solvent system. The principal equivalence with the microstructuring by the ink-jet technique was demonstrated. For a high approach and retraction speed of 9 mm/s and no delay between them, a concave microtopology was observed. In agreement with the literature, this can be explained by a flow of solvent and the dissolved polymer to the rim of the pinned droplet, where polymer is accumulated. This effect is analogue to the well-known formation of ring-like stains after the evaporation of coffee drops (coffee-stain effect). With decreasing retraction speed down to 10 µm/s the resulting surface topology changes from concave to convex. This can be explained with the increasing dissolution of polymer into the solvent drop prior to the evaporation. If the polymer concentration is high enough, gelation occurs instead of a flow to the rim and the shape of the convex droplet is received. With increasing delay time from below 0 ms to 1s the depth of the concave microwells decreases from 4.6 µm to 3.2 µm. However, a convex surface topology could not be obtained, since for longer delay times the polymer sticks to the tip of the syringe. Thus, by changing the delay time a fine-tuning of the concave structure is accomplished, while by changing the retraction speed a principal change of the microtopolgy can be achieved. We attribute this to an additional flow inside the liquid bridge, which enhanced polymer dissolution. Even if the pendant drop is evaporating about 30 µm above the polymer surface without any contact (non-contact mode), concave structures were observed. Rim heights as high as 33 µm could be generated for exposure times of 20 min. The concave structure exclusively lay above the flat polymer surface outside the structure even after drying. This shows that toluene is taken up permanently. The increasing rim height, rh, with increasing exposure time to the solvent vapor obeys a diffusion law of rh = rh0  tn, with n in the range of 0.46 ~ 0.65. This hints at a non-Fickian swelling process. A detailed analysis showed that the rim height of the concave structure is modulated, unlike for the drop deposition. This is due to the local stress relaxation, which was initiated by the increasing toluene concentration in the extruded polymer surface. By altering the intrinsic material parameters i.e. the polymer molar mass and the polymer/solvent combination, several types of microstructures could be formed. With increasing molar mass from 20.9 kDa to 1.44 MDa the resulting microstructure changed from convex, to a structure with a dimple in the center, to concave, to finally an irregular structure. This observation can be explained if one assumes that the microstructuring is dominated by two opposing effects, a decreasing solubility with increasing polymer molar mass, but an increasing surface tension gradient leading to instabilities of Marangoni-type. Thus, a polymer with a low molar mass close or below the entanglement limit is subject to a high dissolution rate, which leads to fast gelation compared to the evaporation rate. This way a coffee-rim like effect is eliminated early and a convex structure results. For high molar masses the low dissolution rate and the low polymer diffusion might lead to increased surface tension gradients and a typical local pile-up of polymer is found. For intermediate polymer masses around 200 kDa, the dissolution and evaporation rate are comparable and the typical concave microtopology is found. This interpretation was supported by a quantitative estimation of the diffusion coefficient and the evaporation rate. For a different polymer/solvent system, polyethylmethacrylate (PEMA)/ethylacetate (EA), exclusively concave structures were found. Following the statements above this can be interpreted with a lower dissolution rate. At low molar masses the concentration of PEMA in EA most likely never reaches the gelation point. Thus, a concave instead of a convex structure occurs. At the end of this section, the optically properties of such microstructures for a potential application as microlenses are studied with laser scanning confocal microscopy. In the third part, the droplet was confined into a glass microcapillary to avoid evaporation. Since here, due to an increased area to volume ratio, the surface properties of the liquid and the solid walls became important, the influence of the surface hydrophilicity of the wall on the interfacial tension between two immiscible liquid slugs was investigated. For this a novel method for measuring the interfacial tension between the two liquids within the capillary was developed. This technique was demonstrated by measuring the interfacial tensions between slugs of pure water and standard solvents. For toluene, n-hexane and chloroform 36.2, 50.9 and 34.2 mN/m were measured at 20°C, which is in a good agreement with data from the literature. For a slug of hexane in contact with a slug of pure water containing ethanol in a concentration range between 0 and 70 (v/v %), a difference of up to 6 mN/m was found, when compared to commercial ring tensiometry. This discrepancy is still under debate.

Tethered bilayer lipid membranes (tBLMs) are a promising model system for the natural cell membrane. They consist of a lipid bilayer that is covalently coupled to a solid support via a spacer group. In this study, we developed a suitable approach to increase the submembrane space in tBLMs. The challenge is to create a membrane with a lower lipid density in order to increase the membrane fluidity, but to avoid defects that might appear due to an increase in the lateral space within the tethered monolayers. Therefore, various synthetic strategies and different monolayer preparation techniques were examined. Synthetical attempts to achieve a large ion reservoir were made in two directions: increasing the spacer length of the tether lipids and increasing the lateral distribution of the lipids in the monolayer. The first resulted in the synthesis of a small library of tether lipids (DPTT, DPHT and DPOT) characterized by 1H and 13C NMR, FD-MS, ATR, DSC and TGA. The synthetic strategy for their preparation includes synthesis of precursor with a double bond anchor that can be easily modified for different substrates (e.g. metal and metaloxide). Here, the double bond was modified into a thiol group suitable for gold surface. Another approach towards the preparation of homogeneous monolayers with decreased two-dimensional packing density was the synthesis of two novel anchor lipids: DPHDL and DDPTT. DPHDL is “self-diluted” tether lipid containing two lipoic anchor moieties. DDPTT has an extended lipophylic part that should lead to the preparation of diluted, leakage free proximal layers that will facilitate the completion of the bilayer. Our tool-box of tether lipids was completed with two fluorescent labeled lipid precursors with respectively one and two phytanyl chains in the hydrophobic region and a dansyl group as a fluorophore. The use of such fluorescently marked lipids is supposed to give additional information for the lipid distribution on the air-water interface. The Langmuir film balance was used to investigate the monolayer properties of four of the synthesized thiolated anchor lipids. The packing density and mixing behaviour were examined. The results have shown that mixing anchor with free lipids can homogeneously dilute the anchor lipid monolayers. Moreover, an increase in the hydrophylicity (PEG chain length) of the anchor lipids leads to a higher packing density. A decrease in the temperature results in a similar trend. However, increasing the number of phytanyl chains per lipid molecule is shown to decrease the packing density. LB-monolayers based on pure and mixed lipids in different ratio and transfer pressure were tested to form tBLMs with diluted inner layers. A combination of the LB-monolayer transfer with the solvent exchange method accomplished successfully the formation of tBLMs based on pure DPOT. Some preliminary investigations of the electrical sealing properties and protein incorporation of self-assembled DPOT and DDPTT-based tBLMs were conducted. The bilayer formation performed by solvent exchange resulted in membranes with high resistances and low capacitances. The appearance of space beneath the membrane is clearly visible in the impedance spectra expressed by a second RC element. The latter brings the conclusion that the longer spacer in DPOT and the bigger lateral space between the DDPTT molecules in the investigated systems essentially influence the electrical parameters of the membrane. Finally, we could show the functional incorporation of the small ion carrier valinomycin in both types of membranes.
