A bomba de cálcio de retículo sarcoplasmático é uma das proteínas mais extensivamente estudadas, capaz de interagir com várias espécies e compostos de vanádio. Combinando-se estudos de fluorescência com ensaios cinéticos de transporte e ligação de 45Ca à ATPase, avaliou-se o efeito de três complexos de vanádio na função bioenergética e estrutural de Ca2+-ATPase. Demonstrou-se que concentrações próximas dos valores de IC50 (para a hidrólise de ATP) de BMOV-V(IV) e de PDC-V(V) não inibem significativamente a acumulação de 45Ca por sarcovesículas. Por outro lado, o complexo PDC-V(V) mostrou ser capaz de estimular a ligação de 45Ca ao retículo sarcoplasmático, sugerindo que este complexos pode interagir com o domínio de ligação de cálcio à bomba. Vários estudos de fluorescência mostraram que BMOV-V(IV) e PDC-V(V) poderão ser capazes de induzir a conformação E2 (tal como soluções de “decavanadato” e de “monovanadato”) e E1 de Ca2+-ATPase (tal como soluções de “metavanadato”), respectivamente. Apesar de o composto HAIDA-V(IV) previlegiar a conformação E1 (devido à sua elevada afinidade para iões Ca2+), inibiu significativamente o transporte e a ligação de 45Ca, o que sugere que possa interagir com os locais de união de cálcio. Os resultados obtidos são consistentes com a formação de um aducto entre o composto de vanádio e a proteína, o que sugere um efeito na homeostasia intracelular de cálcio, nos sistemas de contracção muscular e, inclusive, nas vias de acção de insulina. Cada um dos três complexos promoveu respostas distintas na Ca2+-ATPase, sugerindo uma evidencia de actividades biológicas diversas em função da espécie química de V e do ambiente de coordenação.

A Ria Formosa é uma laguna costeira de características mesotidais, situada na costa Sul de Portugal, que apresenta pronunciadas variações sazonais de temperatura e radiação luminosa. As extensas zonas intertidais desta laguna são inundadas pela maré duas vezes por dia ficando, por isso, também sujeitas a flutuações semidiurnas de temperatura, pressão e radiação luminosa. Estas condições podem afectar os equilíbrios físico-químicos existentes no sedimento. Foi elaborado um esquema de amostragem de sedimentos em suspensão e de fundo em diversos locais da zona intertidal da Ria Formosa em duas escalas de tempo distintas: (a) uma escala sazonal, com o objectivo de estudar a influência das variações anuais de temperatura e os mecanismos por si desenvolvidos na composição elementar dos sedimentos; (b) ao longo de alguns minutos durante a inundação dos sedimentos pela água da maré, com objectivo de estudar o efeito da maré nos equilíbrios físico-químicos existentes no interior do sedimento. Nos sedimentos recolhidos sazonalmente foi medida a temperatura, o pH e o potencial redox, determinado o conteúdo em matéria orgânica e os teores de Al, Si, Mn, Fe, Cu e Cd na fracção sólida. Na água intersticial foram determinadas as concentrações de Mn(II), Fe, Cu e Cd totais dissolvidos. Nas amostras de água intersticial colhidas durante a inundação dos sedimentos foram determinadas as concentrações de Cl-, Mn(II), Fe(II), Fe(III), Cu e Cd totais dissolvidos. A variação do teor de cloretos na água intersticial indicou que a inundação dos sedimentos intertidais pela maré causa infiltração da água através da superfície do sedimento e movimentos de água intersticial no seu interior. Estes movimentos de fluídos ocorreram preferencialmente nas camadas de maior permeabilidade. As alterações ao equilíbrio dinâmico da água intersticial foram mais intensas nos primeiros 28 minutos de inundação, diminuindo ao longo do tempo. Os processos físicos acima referidos provocaram a exportação de Mn(II), Fe(III), Cu e Cd totais dissolvidos da água intersticial para a coluna de água e simultaneamente induziram a oxidação das espécies reduzidas de manganês e de ferro. O Mn(II) foi oxidado a Mn(IV) na camada superficial dos sedimentos e, também, exportado para a coluna de água. A oxidação do Fe(II) a Fe(III) ocorreu, por sua vez, no interior do sedimento numa curta escala de tempo (<7 min.), saindo para a coluna de água uma pequena porção de Fe(III). Estas alterações, provocadas pela inundação dos sedimentos intertidais, mascararam eventuais variações sazonais das concentrações de Mn e Fe na fracção intersticial dos sedimentos. Os perfis de Cu e Cd totais dissolvidos na água intersticial mostraram a existência de várias zonas de mobilização nos primeiros 4 cm de sedimento. A mobilização de Cu não apresentou uma variação sazonal enquanto que as concentrações de Cd mostraram um incremento no Verão. Neste período, o cálculo de fluxos difusivos indicou uma intensificação dos processos de transferência destes metais através da interface sedimento-água relativamente aos existentes no interior do sedimento. Os fluxos difusivos de Cu foram uma ordem de grandeza inferiores ao transporte advectivo associado à inundação pela maré, enquanto que para o Cd as estimativas dos fluxos difusivos e do transporte advectivo foram comparáveis. Em suma, os resultados obtidos mostraram que a distribuição destes elementos na água intersticial é reflexo do regime de transporte-reacção induzido pela inundação dos sedimentos intertidais, sendo para o Fe mais importantes os processos de reacção, enquanto que para o Mn, Cu e Cd os transportes advectivos são mais rápidos do que as reacções. Com base nestes resultados é proposto um modelo para os ciclos de Mn, Fe, Cu e Cd nas zonas intertidais de lagunas costeiras.

Duas partes substantivas dividem o presente relatório: i) o estudo de uma prática teatral específica, sustentada numa pedagogia de natureza genética e consubstanciado na cocriação de um projeto teatral coletivo; ii) o estudo de um vocabulário específico, consubstanciado na redação de verbetes a partir da obra de alguns mestres de teatro contemporâneo e da minha própria experimentação. Através da prática teatral baseada no princípio geral da autenticidade pude chegar à compreensão e visão concreta de que o teatro é uma estética, uma técnica e uma ética ligadas entre si de forma interdependente, que um ator só aprende a amar o teatro quando chega a ser capaz de se autorrevelar e que um ator só se autorrevela quando é capaz de se entregar sem reservas à cocriação com os outros e com o público. Pelo estudo e redação dos verbetes compreendemos que o teatro é uma arte que acontece sempre no presente e que tanto a alma quanto a estética teatral subsistem na capacidade de o ator estar totalmente presente em cena, de tal modo que o inebriamento dionisíaco acontece apenas sob esta condição e, sem esta condição, ou quando esta condição se rompe, quebra-se o equilíbrio entre o dionisíaco e a racionalidade apolínea que acompanha a atuação.

Aggregation and fibrillation of proteins have a great importance in medicine and industry. Misfolding and aggregation are the basis of many neurodegenerative diseases like Alzheimer and Parkinson. Osmolytes are molecules that can accumulate within cells and act as protective agents and they can inclusively act as protein stabilizers when cells are exposed to stress conditions. Osmolytes can also act as protein stabilizers in vitro. In this work, two different proteins were studied, the ribosomal protein from Thermus thermophilus and the mouse prion protein. The existence of an unstructured N-terminal on the prion protein does not affect its stability. The effect of the osmolyte sucrose on the fibrillation and stabilization of these two proteins was studied through kinectic and equilibrium measurements. It was shown that sucrose is able to compact the native structure of S6 protein in fibrillization conditions. Sucrose affects also folding and unfolding kinetic of S6 protein, delaying unfolding and increasing folding rate constants. The mechanism of stabilization by sucrose is non-specific because it is distributed for all protein structure, as it was demonstrated by a protein engineering approach. Sucrose delays the process of formation and elongation of S6 and prion protein from mouse. This delay is the result of the compaction of the native structure refered above. However, cellular toxicity studies have shown that fibrils formed in the presence of sucrose are more toxic to neuronal cells.

Gilthead seabream is the most important farmed species in the Mediterranean, and knowledge on how common farming practices impact its quality is limited. As such, this Thesis aimed to evaluate how gilthead seabream flesh quality is affected by some of these practices. In Chapter 2, the influence of nutritional factors was evaluated, specifically the high replacement of traditional marine-derived ingredients, both fishmeal and fish oil, with vegetable sources. We have seen that the vegetable-based diets tested did not greatly impact seabream flesh quality, although some alterations were seen in the fatty acid profile of the muscle. However, and despite having caused no alterations in flesh texture, vegetable ingredients reduced the amount of sulphated glycosaminoglycans in the extracellular matrix, affected muscle pH and reduced the activity of proteolytic enzymes. Throughout this Thesis, we measured for the first time the activity of proteolytic enzymes in seabream muscle, and cathepsin B was found to play a pivotal role in post-mortem muscle degradation. In Chapter 3, we evaluated the effect of harvesting and slaughter stress on seabream quality, and contrary to what is seen in most farmed species, our results show that gilthead seabream muscle structure is highly resistant to changes caused by stressful events. Nonetheless, considering that welfare is an increasingly important quality criterion, the use of a zero-withdrawal anaesthetic as a rested harvest technique or even slaughter method could prove valuable to the industry. In Chapter 4, we used maslinic acid as a dietary supplement, to modulate the muscle’s energetic status pre-mortem. As a finishing strategy, maslinic acid failed to increase levels of glycogen and ATP in the muscle. However, supplementation resulted in higher muscle fibre diameter and lower cathepsin B activity, and maslinic acid is likely to be useful to promote growth in this species. In general our Thesis has generated new knowledge to a major challenge facing the aquaculture industry, which is to find a compromise between the trends towards intensive rearing and consumer demand for healthy, high quality seafood being ethically acceptable and having a low impact on the environment.

In ecotoxicology a major focus is in the aquatic environment, not only because it presents a great economic value to man but it is an ecosystem widely affected by the growing anthropogenic pollution. Most of the studies performed relate to adverse effects in development, reproductive or endocrine disruption but little is known about the possible effects in bone formation and skeletal development. In this study, we set out to evaluate the effects of 8 aquatic pollutants on the skeletal development using an in vivo system, the zebrafish larvae aged 20 days post-fertilization, through chronic exposure. Several endpoints were considered such as the cumulative mortality, total length, occurrence of skeletal deformities and marker gene expression. We were able to establish LD50 values for some pollutants, like 3-methylcholanthrene, lindane, diclofenac, cobalt and vanadate and found that the total length was not affected by any of the pollutants tested. Cobalt was the most harmful chemical to affect hatching time, severely affecting the ability of the zebrafish embryos to hatch and overall the number of deformities increased upon exposure to tested chemicals but no patterns of deformities were identified. We also propose that 3-methylcholanthrene has an osteogenic effect, affecting osteoblast and osteoclast function and that op levels can act as a mediator of 3-methylcholanthrene toxic stress to the osteoblast. In turn we found naphthalene to probably have a chondrogenic effect. Our results provided new insights into the potential osteotoxicity of environmental pollutants. Future studies should aim at confirming these preliminary data and at determining mechanisms of osteotoxicity.

Bone morphogenetic proteins (BMPs) are multifunctional growth factors belonging to the transforming growth factor β (TGFβ) superfamily with a central role in bone formation and mineralization. BMP2, a founding member of this family, has demonstrated remarkable osteogenic properties and is clinically used to promote bone repair and fracture healing. Lack of basic data on factors regulating BMP2 expression and activity have hampered a better understanding of its role in bone formation and bone-related diseases. The objective of this work was to collect new functional data and determine spatiotemporal expression patterns in a fish system aiming towards a better understanding of BMP2 function and regulation. Transcriptional and post-transcriptional regulation of gilthead seabream BMP2 gene was inferred from luciferase reporter systems. Several bone- and cartilage-related transcription factors (e.g. RUNX3, MEF2c, SOX9 and ETS1) were found to regulate BMP2 transcription, while microRNA 20a was shown to affect stability of the BMP2 transcript and thus the mineralogenic capacity of fish bone-derived host cells. The regulation of BMP2 activity through an interaction with the matrix Gla protein (MGP) was investigated in vitro using BMP responsive elements (BRE) coupled to luciferase reporter gene. Although we demonstrated the functionality of the experimental system in a fish cell line and the activation of BMP signaling pathway by seabream BMP2, no conclusive evidence could be collected on a possible interaction beween MGP and BMP2. The evolutionary relationship among the members of BMP2/4/16 subfamily was inferred from taxonomic and phylogenetic analyses. BMP16 diverged prior to BMP2 and BMP4 and should be the result of an ancient genome duplication that occurred early in vertebrate evolution. Structural and functional data suggested that all three proteins are effectors of the BMP signaling pathway, but expression data revealed different spatiotemporal patterns in teleost fish suggesting distinct mechanisms of regulation. In this work, through the collection of novel data, we provide additional insight into the regulation, the structure and the phylogenetic relationship of BMP2 and its closely related family members.

The objective of this thesis is to study the properties of resistive switching effect based on bistable resistive memory which is fabricated in the form of Al2O3/polymer diodes and to contribute to the elucidation of resistive switching mechanisms. Resistive memories were characterized using a variety of electrical techniques, including current-voltage measurements, small-signal impedance, and electrical noise based techniques. All the measurements were carried out over a large temperature range. Fast voltage ramps were used to elucidate the dynamic response of the memory to rapid varying electric fields. The temperature dependence of the current provided insight into the role of trapped charges in resistive switching. The analysis of fast current fluctuations using electric noise techniques contributed to the elucidation of the kinetics involved in filament formation/rupture, the filament size and correspondent current capabilities. The results reported in this thesis provide insight into a number of issues namely: (i) The fundamental limitations on the speed of operation of a bi-layer resistive memory are the time and voltage dependences of the switch-on mechanism. (ii) The results explain the wide spread in switching times reported in the literature and the apparently anomalous behaviour of the high conductance state namely the disappearance of the negative differential resistance region at high voltage scan rates which is commonly attributed to a “dead time” phenomenon which had remained elusive since it was first reported in the ‘60s. (iii) Assuming that the current is filamentary, Comsol simulations were performed and used to explain the observed dynamic properties of the current-voltage characteristics. Furthermore, the simulations suggest that filaments can interact with each other. (iv) The current-voltage characteristics have been studied as a function of temperature. The findings indicate that creation and annihilation of filaments is controlled by filling and neutralizing traps localized at the oxide/polymer interface. (v) Resistive switching was also studied in small-molecule OLEDs. It was shown that the degradation that leads to a loss of light output during operation is caused by the presence of a resistive switching layer. A diagnostic tool that predicts premature failure of OLEDs was devised and proposed. Resistive switching is a property of oxides. These layers can grow in a number of devices including, organic light emitting diodes (OLEDs), spin-valve transistors and photovoltaic devices fabricated in different types of material. Under strong electric fields the oxides can undergo dielectric breakdown and become resistive switching layers. Resistive switching strongly modifies the charge injection causing a number of deleterious effects and eventually device failure. In this respect the findings in this thesis are relevant to understand reliability issues in devices across a very broad field.

The identification of genes involved in signaling and regulatory pathways, and matrix formation is paramount to the better understanding of the complex mechanisms of bone formation and mineralization, and critical to the successful development of therapies for human skeletal disorders. To achieve this objective, in vitro cell systems derived from skeletal tissues and able to mineralize their extracellular matrix have been used to identify genes differentially expressed during mineralization and possibly new markers of bone and cartilage homeostasis. Using cell systems of fish origin and techniques such as suppression subtractive hybridization and microarray hybridization, three genes never associated with mechanisms of calcification were identified: the calcium binding protein S100-like, the short-chain dehydrogenase/reductase sdr-like and the betaine homocysteine S-methyltransferase bhmt3. Analysis of the spatial-temporal expression of these 3 genes by qPCR and in situ hybridization revealed: (1) the up-regulation of sdr-like transcript during in vitro mineralization of gilthead seabream cell lines and its specificity for calcified tissues and differentiating osteoblasts; (2) the up-regulation of S100-like and the down-regulation of bhmt3 during in vitro mineralization and the central role of both genes in cartilaginous tissues undergoing endo/perichondral mineralization in juvenile fish. While expression of S100-like and bhmt3 was restricted to calcified tissues, sdr-like transcript was also detected in soft tissues, in particular in tissues of the gastrointestinal tract. Functional analysis of gene promoters revealed the transcriptional regulation of the 3 genes by known regulators of osteoblast and chondrocyte differentiation/mineralization: RUNX2 and RAR (sdr-like), ETS1 (s100-like; bhmt3), SP1 and MEF2c (bhmt3). The evolutionary relationship of the different orthologs and paralogs identified within the scope of this work was also inferred from taxonomic and phylogenetic analyses and revealed novel protein subfamilies (S100-like and Sdr-like) and the explosive diversity of Bhmt family in particular fish groups (Neoteleostei). Altogether our results contribute with new data on SDR, S100 and BHMT proteins, evidencing for the first time the role for these three proteins in mechanisms of mineralization in fish and emphasized their potential as markers of mineralizing cartilage and bone in developing fish.
