Transmission of the protozoan parasite Giardia lamblia from one to another host individuum occurs through peroral ingestion of cysts which, following excystation in the small intestine, release two trophozoites each. Many studies have focused on the major surface antigen, VSP (for variant surface protein), which is responsible for the antigenic variability of the parasite. By using trophozoites of G. lamblia clone GS/M-83-H7 (expressing VSP H7) and the neonatal mouse model for experimental infections, we quantitatively assessed the process of antigenic variation of the parasite on the transcriptional level. In the present study, variant-specific regions identified on different GS/M-83-H7 vsp sequences served as targets for quantitative reverse transcription-PCR to monitor alterations in vsp mRNA levels during infection. Respective results demonstrated that antigenic switching of both the duodenal trophozoite and the cecal cyst populations was associated with a massive reduction in vsp H7 mRNA levels but not with a simultaneous increase in transcripts of any of the subvariant vsp genes analyzed. Most importantly, we also explored giardial variant-type formation and vsp mRNA levels after infection of mice with cysts. This infection mode led to an antigenic reset of the parasite in that a VSP H7-negative inoculum "converted" into a population of intestinal trophozoites that essentially consisted of the original VSP H7 type. This antigenic reset appears to be associated with excystation rather than with a selective process which favors expansion of a residual population of VSP H7 types within the antigenically diversified cyst inoculum. Based on these findings, the VSP H7 type has to be regarded as a predominant variant of G. lamblia clone GS/M-83-H7 which (re-)emerges during early-stage infection and may contribute to an optimal establishment of the parasite within the intestine of the experimental murine host.

BACKGROUND Researchers evaluating angiomodulating compounds as a part of scientific projects or pre-clinical studies are often confronted with limitations of applied animal models. The rough and insufficient early-stage compound assessment without reliable quantification of the vascular response counts, at least partially, to the low transition rate to clinics. OBJECTIVE To establish an advanced, rapid and cost-effective angiogenesis assay for the precise and sensitive assessment of angiomodulating compounds using zebrafish caudal fin regeneration. It should provide information regarding the angiogenic mechanisms involved and should include qualitative and quantitative data of drug effects in a non-biased and time-efficient way. APPROACH & RESULTS Basic vascular parameters (total regenerated area, vascular projection area, contour length, vessel area density) were extracted from in vivo fluorescence microscopy images using a stereological approach. Skeletonization of the vasculature by our custom-made software Skelios provided additional parameters including "graph energy" and "distance to farthest node". The latter gave important insights into the complexity, connectivity and maturation status of the regenerating vascular network. The employment of a reference point (vascular parameters prior amputation) is unique for the model and crucial for a proper assessment. Additionally, the assay provides exceptional possibilities for correlative microscopy by combining in vivo-imaging and morphological investigation of the area of interest. The 3-way correlative microscopy links the dynamic changes in vivo with their structural substrate at the subcellular level. CONCLUSIONS The improved zebrafish fin regeneration model with advanced quantitative analysis and optional 3-way correlative morphology is a promising in vivo angiogenesis assay, well-suitable for basic research and preclinical investigations.

Sporocarps of macrofungi have been recorded in two neighbouring pure stands of Norway spruce (Picea abies) of different age due to a wind-throw in the subalpine zone of the Alps. The still open young stand of 30 years with trees up to 6 m displayed 80 species, the mature closed forest stand 90 species. Species richness of mycorrhizal fungi is higher in the mature stand than in the younger one, however, with an almost doubled sporocarp production in the latter one. The opposite is found with saprotrophic fungi. Several fungi appeared only in one forest type confirming the concept of early stage versus late stage fungi. Wind-throws as irregular events in subalpine forests, create gaps and add considerably to the species diversity of macrofungi.

O mercado de geração distribuída de energia solar fotovoltaica apresenta-se em estágio inicial de desenvolvimento no Brasil. A recente conjuntura regulatória, entretanto, aponta para a mudança deste cenário: as últimas revisões da regulação aplicável trouxeram aumento na viabilidade econômica de projetos no setor. Isto, somado à presença abundante de recurso solar no Brasil e ao presente estágio de desenvolvimento da tecnologia fotovoltaica indicam um possível crescimento da penetração da fonte no país nos próximos anos. Nesse contexto, o presente estudo tem como objetivo analisar quais recursos e capacidades a empresa Axis Renováveis deve possuir para alcançar uma posição de vantagem competitiva, à luz do RBT. A empresa selecionada para a condução do estudo de caso único é uma start-up atuante no setor. Espera-se que, ao final do trabalho, as conclusões tenham implicação direta na estratégia da empresa e possam servir como ponto de partida para outras empresas do setor.

Dans ce mémoire, intitulé « Les dépotoirs dans le schème d’établissement iroquoien: exemple du site Mailhot-Curran», je m’attèle à définir les modes de rejets des déchets dans le contexte d’un schème d’établissement sédentaire semi-permanent au Sylvicole supérieur. Les dépotoirs des sites archéologiques ne semblent pas obtenir toute l’attention analytique qui leur est due et l’étude des modes de rejets a longtemps été négligée. Pourtant, les amas de déchets sont récurrents sur les sites du Nord-Est américain et ce depuis le Sylvicole moyen. L’étude approfondie de l’organisation d’un site iroquoien se doit de passer par la compréhension des modes d’élimination des déchets, puisque la gestion des rejets est généralement le reflet de la structure du site et du type de peuplement. Les dépotoirs peuvent fournir des informations précieuses sur les zones d’activité, la fonction du site ou encore la durée d’occupation. Le village de Mailhot-Curran témoigne de plusieurs concentrations d’objets. Je souhaite établir si ces concentrations de vestiges peuvent être qualifiées de dépotoirs et si oui, peut-on en distinguer différents types? D’autre part, je m’intéresse à l’aspect social qui encadre la mise en place de dépotoirs. Bien que les dépotoirs soient des indices associés aux premières traces de sédentarité saisonnière, leur présence est précisément liée au phénomène de sédentarisation annuelle. Il s’agit également de définir le geste culturel, l’assainissement d’un village étant un schéma culturel très répandu. De ce geste de rejet découle forcément le choix d’un emplacement, ainsi que le concept de «flux de déplacement» des déchets. Il s’agit de déterminer le lien entretenu entre les aires d’activités et les aires de dépôts primaires et secondaires. Pour ce faire, il est primordial d’évaluer la densité, l’hétérogénéité et le contenu des dépotoirs. Suite à la définition de l’ensemble de ces concepts appliqués au site Mailhot-Curran, je souhaite, à travers une étude comparative, répondre à la question suivante : Les dépotoirs peuvent-ils refléter une pratique culturelle commune aux villages iroquoiens de la région de Saint-Anicet?

Studies on the fate of organohalogen contaminants (OHCs) in wild top predator mammals in the Arctic have often been a challenge due to important knowledge deficiencies in the life history of the sampled animals. The present study investigated the influence of age, dietary and trans-generational factors on the fate of major lipophilic chlorinated and brominated OHCs in adipose tissue of a potential surrogate captive species for the polar bear (Ursus maritimus), the sledge dog (Canis familiaris) in West Greenland. Adult female sledge dogs (P) and their sexually-mature (F1) and/or pre-weaning pups (F1-MLK) were divided into an exposed group (EXP) fed blubber from a Greenland minke whale (Balaenoptera acutorostrata) and a control group (CON) given commercially available pork fat. Large dietary treatment-related differences in summed and individual congener/compound adipose tissue concentrations of polybrominated diphenyl ethers (PBDEs), hexachlorobenzene (HCB), chlordanes (CHLs) and polychlorinated biphenyls (PCBs) were found between the EXP and CON groups for all the sledge dog cohorts. However, among the F1-MLK, F1 and P dogs in both of the EXP and CON groups, little or no difference existed in PBDE, HCB, CHL and PCB concentrations, suggesting higher state of equilibrium in adipose tissue concentrations from a very early stage of life. In contrast, the distribution pattern (proportions to the summed concentrations) of OHC classes, and the major congeners/ compounds constituting those classes, varied on a dietary group- and/or cohort-dependent manner. The present captive sledge dog study demonstrated the importance of the confounding effects of diet composition, mother-pup association (maternal transfer), reproductive status (nursing), and to a lesser extent age in the fate of OHCs in adipose tissue of a large top carnivore mammal.

Stable oxygen and carbon isotope (d18O and d13C) values measured in foraminiferal calcite are one of the primary tools used in paleoceanography. Diagenetic recrystallization of foraminiferal calcite can act to reset primary isotopic values, but its effects are typically poorly quantified. Here we test the impact of early stage diagenesis on stable isotope records generated from a suite of drill sites in the equatorial Pacific Ocean recovered during Ocean Drilling Program Leg 199 and Integrated Ocean Drilling Program Expedition 320. Our selected sites form paleowater and burial depth transects, with excellent stratigraphic control allowing us to confidently correlate our records. We observe large intersite differences in the preservation state of benthic foraminiferal calcite, implying very different recrystallization histories, but negligible intersite offsets in benthic d18O and d13C values. We infer that diagenetic alteration of benthic foraminiferal calcite (in sedimentary oozes) must predominantly occur at shallow burial depths (<100 m) where offsets in both the temperature and isotopic composition of waters in which the foraminifera calcified and pore waters in which diagenesis occurs are small. Our results suggest that even extensive recrystallization of benthic foraminiferal calcite results in minimal shifts from primary d18O and d13C values. This finding supports the long-held suspicion that diagenetic alteration of foraminiferal calcite is less problematic in benthic than in planktic foraminifera and that in deep-sea sediments routinely employed for paleoceanographic studies benthic foraminifera are robust recorders of stable isotope values in the fossil record.

Stable oxygen and carbon isotope and sedimentological-paleontological investigations supported by accelerator mass spectrometry 14C datings were carried out on cores from north of 85°N in the eastern central Arctic Ocean. Significant changes in accumulation rates, provenance of ice-rafted debris (IRD), and planktic productivity over the past 80,000 years are documented. During peak glacials, i.e., oxygen isotope stages 4 and 2, the Arctic Ocean was covered by sea ice with decreased seasonal variation, limiting planktic productivity and bulk sedimentation rates. In early stage 3 and during Termination I, major deglaciations of the circum-Arctic regions caused lowered salinities and poor oxygenation of central Arctic surface waters. A meltwater spike and an associated IRD peak dated to ~14-12 14C ka can be traced over the southern Eurasian Basin of the Arctic Ocean. This event was associated with the early and rapid deglaciation of the marine-based Barents Sea Ice Sheet. A separate Termination Ib meltwater event is most conspicuous in the central Arctic and is associated with characteristic dolomitic carbonate IRD. This lithology suggests an origin of glacial ice from northern Canada and northern Greenland where lower Paleozoic platform carbonates crop extensively out.

A reconnaissance study of alkenone stratigraphy for the past 35 m.y. in the northern South China Sea (SCS) using sediments from Sites 1147 and 1148 of Ocean Drilling Program (ODP) Leg 184 has been completed. Alkenones were not detected in sediment samples older than ~31 Ma. However, C37:2 appeared in the sedimentary record between ~8 and 31 Ma and both C37:2 and C37:3 were present between 0 and 8 Ma. These changes in alkenone occurrences may signal a response to global-scale Neogene cooling as well as to monsoon intensification and sea level changes over time as a result of Himalayan uplift and the opening of the SCS. Alternatively, they may be related to an evolutionary record of the development of temperature control on alkenone production in coccolithophores. The Uk'37 index for 0-8 Ma produces sea-surface temperatures (SST) of 19°-26°C, which are in the range of previously determined glacial-interglacial values for the northern SCS. Before the late Pleistocene (~1.2 Ma), the SST range is between 23° and 26°C with less variation. This change in variability may signify the early stage of intensified winter monsoons where cold wind and waters from the north may not yet have had a significant effect on SST or it may be the evolutionary link between the early development of unsaturated alkenones in coccolithophores and modern temperature control of alkenone production. We believe a long-term alkenone record is useful for further understanding of global-scale neogene cooling, the development of the East Asian monsoon system, and the evolutionary development of temperature control on alkenone unsaturation. Our data indicate that a high-resolution Uk'37 record for at least the last ~8 Ma is feasible for the northern SCS.

Of all human cancers, HNSCC is the most distressing affecting pain, disfigurement, speech and the basic survival functions of breathing and swallowing. Mortality rates have not significantly changed in the last 40 years despite advances in radiotherapy and surgical treatment. Molecular markers are currently being identified that can determine prognosis preoperatively by routine tumour biopsy Leading to improved management of HNSCC patients. The approach could help decide which early stage patient should have adjuvant neck dissection and radiotherapy, and whether Later stage patients with operable lesions would benefit from resection and reconstructive surgery or adopt a conservative approach to patients with poor prognosis regardless of treatment. Kallikrein 6 (hK6, also known as protease M/zyme/neurosin) is a member of the human kallikrein gene family. We have previously cloned the cDNA for this gene by differential display and shown the overexpression of the mRNA in breast and ovarian primary tumour tissues and cell lines. To thoroughly characterise the expression of this kallikrein in ovarian cancer, we have developed a novel monoclonal antibody specific to hK6 and employed it in immunohistochemistry with a wide range of ovarian tumour samples. The expression was found elevated in 67 of 80 cases of ovarian tumour samples and there was a significant difference in the expression levels between normal and benign ovarian tissues and the borderline and invasive tumours (P<0.001). There was no difference of expression level between different subtypes of tumours. More significantly, high level of kallikrein 6 expression was found in many early-stage and low-grade tumours, and elevated hK6 proteins were found in benign epithelia coexisting with borderline and invasive tissues, suggesting that overexpression of hK6 is an early phenomenon in the development of ovarian cancer. Quantitative real-time reverse transcription-polymerase chain reactions also showed elevated kallikrein 6 mRNA expression in ovarian tumours. Genomic Southern analysis of 19 ovarian tumour samples suggested that gene amplification is one mechanism for the overexpression of hK6 in ovarian cancer.

Despite the importance of peritubular myoid (PM) cells in the histogenesis of the fetal testis, understanding the origin and function of these cells has been hampered by the lack of suitable markers. The current study was aimed at identifying molecular markers for PM cells during the early stages of testis development in the mouse embryo. Expression of candidate marker genes was tested by section in situ hybridisation, in some instances followed by immunofluorescent detection of protein products. Collagen type-1, inhibin beta A, caldesmon 1 and tropomyosin 1 were found to be expressed by early-stage PM cells. These markers were also expressed in subsets of interstitial cells, most likely reflecting their common embryological provenance from migrating mesonephric cells. Although not strictly specific for PM cells, these markers are likely to be useful in studying the biology of early PM cells in the fetal testis.
