Este trabalho buscou avaliar o comportamento e o desempenho sexual de suínos machos de linhas puras e cruzadas, criados com e sem a utilização de enriquecimento ambiental, na fase de crescimento. A pesquisa foi dividida em duas etapas, que compreenderam a fase de crescimento dos animais e o treinamento para coleta de sêmen. Na fase de crescimento, 128 machos foram alojados em ambientes enriquecidos ou estéreis. Utilizou-se como enriquecimento ambiental correntes suspensas, galão de cinco litros suspenso e um galão de 50 litros solto no piso. Esses objetos foram oferecidos de forma alternada e cada um ficou disponível na baia por um período de 30 dias. Na primeira etapa foram registrados o comportamento dos animais, os escores de lesão e a massa corporal. Após a fase de crescimento, foram escolhidos aleatoriamente 32 animais aprovados na seleção genética para serem avaliados durante o treinamento para coleta de sêmen. O treinamento ocorreu durante seis dias consecutivos e cada animal foi treinado por três vezes em dias alternados. Durante o treinamento para a coleta de sêmen, o comportamento animal, as relações humano-animal, o volume do ejaculado e os níveis de testosterona e cortisol foram registrados. Como respostas na fase de crescimento, verificou-se que, mesmo utilizando uma combinação de objetos, os suínos se habituaram rapidamente a eles e a frequência de manipulação diminuiu após o primeiro período para todos os objetos. Observamos que o ambiente enriquecido foi eficaz na redução dos comportamentos agonísticos e mordedura de cauda e orelha para os animais puros e cruzados, e isso consequentemente reduziu a quantidade e severidade de lesões de pele. Na fase de treinamento para coleta de sêmen, os resultados demonstraram que o comportamento sexual dos animais foi influenciado pelas linhas genéticas, sendo assim, observou-se que os machos de linha cruzada tiveram maior facilidade durante o treinamento para coleta de sêmen e apresentaram maior média do escore de libido, diferindo das linhas puras (P<0,001). Verificou-se que não houve diferença na média do escore de libido entre os tratamentos com e sem enriquecimento ambiental (P=0,276), porém, os tratamentos com enriquecimento tiveram o menor número de animais treinados. Dessa forma, os resultados indicam que o ambiente enriquecido com uma combinação de enriquecimentos pontuais (objetos) é uma estratégia eficaz para aumentar o comportamento exploratório e reduzir os comportamentos agonísticos e anormais na fase de crescimento. Mas, por outro lado, os animais criados em ambientes enriquecidos tiveram um pior desempenho sexual durante o treinamento para coleta de sêmen.

Com este estudo objetivou-se avaliar os níveis de estresse e a qualidade de carne de cordeiros mestiços Santa Inês x Dorper, submetidos a transporte de percurso curto (duas horas) e longo (seis horas) e em dois períodos de espera pré-abate (12 ou 24 horas). Foram utilizados 32 cordeiros com 127 ± 7 dias de idade e 30,4 ± 2,1 kg de peso vivo. Antes de serem submetidos aos períodos de transporte, os animais estavam alocados em baias coletivas e receberam ração concentrada (farelo de soja e milho, calcário calcítico e núcleo com monensina), bagaço úmido de citros e capim Napier (Pennisetum purpureum) e água ad libitum. Foram realizadas avaliações de reatividade dos animais no momento do embarque, desembarque e durante a contenção dos animais para as colheitas de sangue, urina e temperatura ocular por termografia infravermelho. Durante o período de espera pré-abate, realizou-se a colheita de dados comportamentais dos animais. Foram avaliadas as concentrações de cortisol no soro, na urina e as concentrações de haptoglobina no soro nos períodos que antecederam a insensibilização, assim como no momento do abate. Os animais apresentaram baixa reatividade durante todos os manejos. Houve diferença significativa no comportamento dos cordeiros durante os períodos de espera (P < 0,05) que durante as 12 horas apresentaram frequência de comportamentos que indicaram bem-estar favorável, enquanto que o período de transporte não afetou (P > 0,05). Os níveis de cortisol no soro mantiveram-se semelhantes da saída dos animais para o transporte até o final do período de espera (P > 0,05), enquanto houve oscilação dessas concentrações no cortisol na urina (P < 0,05), com pico no desembarque dos animais de duas horas de transporte e diminuição ao final do período de descanso. Os níveis de haptoglobina mantiveram-se semelhantes da colheita realizada antes do transporte, no embarque e no desembarque (P > 0,05) e diminuíram no final do período de espera pré-abate (P < 0,05). A temperatura ocular elevou-se no embarque e no desembarque dos animais, com diminuição da temperatura ao final do período de espera (P < 0,05). No momento do abate, foi observado aumento das concentrações de haptoglobina (P < 0,05), enquanto não houve alteração nas concentrações de cortisol no soro (P > 0,05). Animais que permaneceram por 24 horas de espera pré-abate apresentaram maior força de cisalhamento e menor luminosidade (L*) e intensidade de amarelo (b*). As variáveis comportamentais foram pouco afetadas pelos períodos de transporte e de espera pré-abate, porém o período de 12 horas de espera favoreceu a qualidade da carne

Many studies indicate that smoking is one of risk factors influencing the accumulation of visceral fat (VFA). The mechanism by which smoking contributes to the accumulation of visceral fat is not yet fully understood, but it is assumed that smoking increases the level of plasma cortisol, causes imbalance between male and female sex hormones in women, and decreases testosterone levels in men (Chiolero et al., 2008). The objection of this study was to evaluate the effect of smoking on the accumulation of VFA in the population of the Czech Republic. The research included 1,412 individuals of both genders divided into categories of smokers vs nonsmokers and physically active vs. physically inactive. VFA was measured by the device InBody 720 and information about physical activity and smoking was collected via a questionnaire. We supposed that smokers would have a higher percentage of VFA than non-smokers. The results showed that smokers had a higher amount of VFA, but the difference was not statistically significant. In contrast, statistically significant differences were found in the waist-to-hip ratio (WHR) and total % body fat. Physically active individuals had a much more favourable results than physically inactive individuals. Remarkably, the amount of VFA, WHR and % body fat tended to be smaller in smoking men than in non-smoking men, but higher in smoking women than in non-smoking women.

Introdução: O hipopituitarismo é caracterizado por insuficiência da secreção hormonal hipófisária. A clínica é variável e depende da etiologia, evolução temporal e hormonas envolvi- das. Caso: Criança do sexo masculino com 2 anos, trazida à urgência por alteração súbita da consciência. No período neonatal apresentou quadro de hipoglicemia, trombocitopenia, icterícia e sépsis sem agente identificado. Objetivou-se crescimento regular no P10-25, desenvolvimento psicomotor adequado e estrabismo divergente. Ao exame objectivo apresentava-se subfebril e com Escala de Coma de Glasgow 10. Foi constatada hipoglicemia grave (26mg/dL) sendo realizado de imediato estudo endocrinológico e metabólico que mostrou cortisol baixo e défices de ACTH e GH; posteriormente foi confirmado défice de TSH. Iniciou terapêutica de substituição com hidrocortisona e levotiroxina. A neuroimagem mostrou alterações estruturais com hipoplasia da neuro-hipófise. Conclusão: Este diagnóstico raro exige elevado grau de suspeição. A ocorrência progressiva dos défices hormonais obriga à avaliação clínica e laboratorial regular.

L’acceptabilité d’un décès lors d’une simulation médicale reste débattue mais il existe peu de données sur la perspective des apprenants. Des médecins résidents ont effectué une pratique de code et ont rempli un questionnaire pré et post-simulation. Ils ont été exposés à deux scénarios où un bébé naissait sans signe de vie: 1. Nouveau-né ne répondant pas aux manœuvres de réanimation (DCD); 2. Nouveau-né s’améliorant avec une réanimation adéquate (REA). Les performances étaient évaluées à l’aide de la grille standardisée du Programme de Réanimation Néonatale. Le stress objectif (cortisol salivaire) et subjectif a été mesuré après le code. La rétroaction («feedback»), individuelle et en groupe, fut analysée à l’aide de méthodologies qualitatives. 59/62 apprenants ont répondu au questionnaire et 42 ont participé à la simulation. Tous les résidents trouvent les simulations bénéfiques et souhaitent y être exposés davantage. Le type et l’ordre des scénarios n’ont pas eu d’impact sur la performance. Un seul résident a interrompu les manœuvres de réanimation après 10 minutes d’asystolie, tel que recommandé, et 31% ont poursuivi après 20 minutes. Les participants trouvaient le scénario DCD plus stressant. Les niveaux de cortisol salivaire ont augmenté après les simulations (p<0.001) et ce, pour les deux scénarios. Cette augmentation était indépendante du scénario (p=0.06) et n’était pas associée à la performance. Les réponses à la question « Comment a été votre expérience? », ont permis d’identifier deux thèmes: 1. Le mannequin ne meurt pas en simulation médicale; 2. Le décès lors de la simulation signifie une réanimation inadéquate. Le décès lors d’une pratique de code est stressant, mais n’interfère pas avec la performance des soignants. Les apprenants trouvent cet exercice acceptable et bénéfique à leur pratique future.

Although androgens are commonly seen as male sex hormones, it has been established over the years that in both sexes, androgens also respond to social challenges. To explain the socially driven changes in androgens, two theoretical models have been proposed: the biosocial model and the challenge hypothesis. These models are typically seen as partly overlapping; however, they generate different predictions that are clarified here. In humans, sports competition and nonmetabolic competitive tasks have been used in the laboratory setting, as a proxy for agonistic interactions in animals. The results reviewed here show that the testosterone (T) response to competition in humans is highly variable – the studies present postcompetition T levels and changes in T that depend on the contest outcome and that cannot be predicted by the current theoretical models. These conflicting results bring to the foreground the importance of considering cognitive factors that could moderate the androgen response to competition. Among these variables, we elect cognitive appraisal and its components as a key candidate modulating factor. It is known that T also modulates the cognitive processes that are relevant to performance in competition. In this article, we reviewed the evidence arising from studies investigating the effect of administering exogenous T and compare those results with the findings from studies that measured endogenous T levels. Finally, we summarized the importance of also considering the interaction between androgens and other hormones, such as cortisol, when investigating the social modulation of T, as proposed by the dual-hormone hypothesis.

social stressors typically elicit two distinct behavioural responses in vertebrates: an active response (i.e., "fight or flight") or behavioural inhibition (i.e., freezing). Here, we report an interesting exception to this dichotomy in a Caribbean cleaner fish, which interacts with a wide variety of reef fish clients, including predatory species. Cleaning gobies appraise predatory clients as potential threat and become stressed in their presence, as evidenced by their higher cortisol levels when exposed to predatory rather than to non-predatory clients. Nevertheless, cleaning gobies neither flee nor freeze in response to dangerous clients but instead approach predators faster (both in captivity and in the wild), and interact longer with these clients than with non-predatory clients (in the wild). We hypothesise that cleaners interrupt the potentially harmful physiological consequences elicited by predatory clients by becoming increasingly proactive and by reducing the time elapsed between client approach and the start of the interaction process. The activation of a stress response may therefore also be responsible for the longer cleaning service provided by these cleaners to predatory clients in the wild. Future experimental studies may reveal similar patterns in other social vertebrate species when, for instance, individuals approach an opponent for reconciliation after a conflict.

Background: This study investigated whether patients suffering from post-stroke depressive disorder had a similar disturbance in central serotonergic function to that described in non-brain injured depressed patients. Methods: Twenty-three depressed patients (nine major, 14 minor) and 38 non-depressed patients were examined 4-8 weeks post-stroke with a structured interview, rating scales and MRI brain scans. Patients were administered 30 mg D-fenfluramine orally and plasma prolactin and D-fenfluramine concentrations were measured for 6 h post-dose. Results: The prolactin response was significantly blunted in major depression compared to minor depression and non-depressed patients as measured by both delta prolactin and area under the prolactin versus time curve. There was no significant relationship between prolactin response and lesion lateralization or any of the measured clinical characteristics. Limitations: The major limitation of the study is the relatively small number in each depressive group. Conclusions: Patients suffering from major depression in the post-stroke period have a blunted prolactin response to D-fenfluramine. This indicates a serotonergic abnormality consistent with that found in major depression where neurological disease is not present. (C) 2003 Elsevier B.V. All rights reserved.

Primary objective: The study aimed to examine the changes in water distribution in the soft tissue during systemic steroid activity. Research design: A three-way cross-over, randomized, placebo-controlled, double-blind trial was used, including 4 weeks of fluticasone propionate pMDI 200 mug b.i.d. delivered via Babyhaler(R), budesonide pressurized metered dose inhaler (pMDI) 200 mug b.i.d. delivered via Nebuchamber(R) and placebo. Spacers were primed before use. In total, 40 children aged 1-3 years, with mild intermittent asthma were included. Twenty-five of the children completed all three treatments. At the end of each treatment period body impedance and skin ultrasonography were measured. Methods and procedures: We measured changes in water content of the soft tissues by two methods. Skin ultrasonography was used to detect small changes in dermal water content, and bioelectrical impedance was used to assess body water content and distribution. Main outcomes and results: We found an increase in skin density of the shin from fluticasone as measured by ultrasonography (p = 0.01). There was a tendency for a consistent elevation of impedance parameters from active treatments compared to placebo although overall this effect was not statistically significant (0.1< p <0.2). However, sub-analyses indicated a significant effect on whole-body and leg impedance from budesonide treatment (p <0.05). Conclusion: Decreased growth during inhaled steroid treatment seems to partly reflect generalized changes in body water.

Hyperactivity of the sympathetic and noradrenergic systems is thought to be a feature of post-traumatic stress disorder (PTSD). Assessment of noradrenergic receptor function can be undertaken by measuring the growth hormone (GH) response to the alpha(2)-agonist clonidine. The aim of this study was to examine whether subjects with combat-related PTSD (with or without co-morbid depression) have a blunted growth hormone response to clonidine, compared to a combat-exposed control group. Twenty-three Vietnam veterans suffering from PTSD alone, 27 suffering from PTSD and co-morbid depression, and 32 veteran controls with no psychiatric illness were administered 1.5 mug/kg clonidine i.v. Plasma growth hormone was measured every 20 min for 120 min. The growth hormone response to clonidine was significantly blunted in the non-depressed PTSD group compared to both the depressed PTSD group and the control group as measured by peak growth hormone, delta growth hormone and AUC growth hormone. Subjects with PTSD and no co-morbid depressive illness show a blunted growth hormone response to clonidine. This suggests that post-synaptic alpha(2)-receptors are subsensitive. This finding is consistent with other studies showing increased noradrenergic activity in PTSD. (C) 2003 Elsevier Ltd. All rights reserved.

Background: Renal transplant recipients were noted to appear cushingoid while on low doses of steroid as part of a triple therapy immunosuppression of cyclosporin A (CsA), prednisolone, and azathioprine. Methods: The study group comprised adult renal transplant recipients with stable graft function who had received their renal allograft a minimum of 1 year previously (43 studies undertaken in 22 men and 20 women) with median daily prednisone dose of 7 mg (range 3-10). The control group was healthy nontransplant subjects [median dose 10 mg (10-30)]. Prednisolone bioavailability was measured using a limited 6-hour area under the curve (AUC), with prednisolone measured using specific HPLC assay. Results: The median prednisolone AUC/mg dose for all transplant recipients was significantly greater than the control group by approximately 50% (316 nmol(.)h/L/mg prednisolone versus 218). AUC was significantly higher in female recipients (median 415 versus 297 for men) and in recipients receiving cyclospotin (348 versus 285). The highest AUC was in women on estrogen supplements who were receiving cyclosporin (median 595). A significantly higher proportion of patients on triple therapy had steroid side effects compared with those on steroid and azathioprine (17/27 versus 4/15), more women than men had side effects (14/16 versus 7/22), and the AUC/mg prednisone was greater in those with side effects than without (median 377 versus 288 nmol-h/L/mg). Discussion: The results are consistent with the hypothesis that CsA increases the bioavailability of prednisolone, most likely through inhibition of beta-glycoprotein. The increased exposure to steroid increased the side-effect profile of steroids in the majority of patients. Because the major contributor to AUC is the maximum postdose concentration, it may be possible to use single-point monitoring (2 hours postdose) for routine clinical studies.

Chronic fatigue syndrome (CFS) is characterized by idiopathic fatigue of greater than 6 months' duration with postexertional exacerbation and many other symptoms. A trend toward relative hypocortisolism is described in CFS. Twin and family studies indicate a substantial genetic etiologic component to CFS. Recently, severe corticosteroid-binding globulin (CBG) gene mutations have been associated with CFS in isolated kindreds. Human leukocyte elastase, an enzyme important in CBG catabolism at inflammatory sites, is reported to be elevated in CFS. We hypothesized that CBG gene polymorphisms may act as a genetic risk factor for CFS. A total of 248 patients with CFS defined by Centers for Disease Control criteria, and 248 controls were recruited. Sequencing and restriction enzyme testing of the CBG gene coding region allowed detection of severe CBG gene mutations and a common exon 3 polymorphism (c.825G --> T, Ala-Ser(224)). Plasma CBG levels were measured in 125 CFS patients and 198 controls by radioimmunoassay. Total and free (calculated and measured) cortisol levels were ascertained in single samples between 8-10 a.m. The age of onset (mid 30s) and gender ratio (2.2:1, female:male) of the patients were similar to those reported in U.S. epidemiologic studies. A trend toward a preponderance of serine(224) homozygosity among the CFS patients was noted, compared with controls (chi(2) = 5.31, P = 0.07). Immunoreactive-CBG (IR-CBG) levels were higher in Serine/Alanine (Ser/Ala) than Ala/Ala subjects and higher again in Ser/Ser subjects, this effect was strongest in controls; Ser/Ser: 46.1 +/- 1.8 (n = 31, P = 0.03) vs. Ser/Ala: 42.4 +/- 1.0 (n = 56, P = 0.05) vs. Ala/Ala: 40.8 +/- 1.7 mug/mL (n = 21). Despite higher CBG levels, there was a nonsignificant trend toward lower total and free plasma cortisol in serine allele positive patients, total cortisol: Ser/Ser: 13.3 +/- 1.4 (n = 34) vs. Ser/Ala: 14.0 +/- 0.7 (n = 66) vs. Ala/Ala: 15.4 +/- 1.0 (n = 23). Homozygosity for the serine allele of the CBG gene may predispose to CFS, perhaps due to an effect on hypothalamic-pituitary-adrenal axis function related to altered CBG-cortisol transport function or immune-cortisol interactions.
