Cancer is a multistage process characterized by three stages: initiation, promotion and progression; and is one of the major killers worldwide. Oxidative stress acts as initiator in tumorigenesis; chronic inflammation promotes cancer; and apoptosis inactivation is an issue in cancer progression. In this study, it was investigated the antioxidant, antiinflammatory and antitumor properties of hexane, ether, chloroform, methanol and water extracts of five species of halophytes: A. macrostachyum, P. coronopus, J. acutus, C. edulis and A. halimus. Antioxidant activity was assessed by DPPH• and ABTS•+ methods, and the total phenolics content (TPC) was evaluated by the Folin-Ciocalteau method. The anti-inflammatory activity of the extracts was determined by the Griess method, and by evaluating the inhibition of NO production in LPS-stimulated RAW- 264.7 macrophages. The cytotoxic activity of the extracts against HepG2 and THP1 cell lines was estimated by the MTT assay, and the results obtained were further compared with the S17 non-tumor cell line. The induction of apoptosis of J. acutus ether extract was assessed by DAPI staining. The highest antioxidant activities was observed in C. edulis methanol and the J. acutus ether extracts against the DPPH• radical; and J. acutus ether and A. halimus ether extracts against the ABTS•+ radical. The methanol extracts of C. edulis and P. coronopus, and the ether extract of J. acutus revealed a high TPC. Generally the antioxidant activity had no correlation with the TPC. The A. halimus chloroform and P. coronopus hexane extracts demonstrated ability to reduce NO production in macrophages (> 50%), revealing their anti-inflammatory capacity. The ether extract of J. acutus showed high cytotoxicity against HepG2 cancer cells, with reduced cellular viability even at the lowest concentrations. This outcome was significantly lower than the obtained with the non-tumor cells (S17). This result was complemented by the induction of apoptosis.

Ocular pathologies are among the most debilitating medical conditions affecting all segments of the population. Traditional treatment options are often ineffective, and gene therapy has the potential to become an alternative approach for the treatment of several pathologies. Methacrylate polymers have been described as highly biocompatible and are successfully used in medical applications. Due to their cationic nature, these polymers can be used to form polyplexes with DNA for its delivery. This work aims to study the potential of PDMAEMA (poly(2-(N,N’-dimethylamino)ethyl methacrylate)) as a non viral gene delivery system to the retina. The first part of this work aimed to study the potential for gene delivery of a previously synthesized PDMAEMA polymer of high molecular weight (354kDa). In the second part, we synthesized by RAFT a PDMAEMA with a lower molecular weight (103.3kDa) and similarly, evaluated its ability to act as a gene delivery vehicle. PDMAEMA/DNA polyplexes were prepared at 5, 7.5, 10, 12.5 and 20 nitrogen/phosphorous (N/P) ratio for the 354kDa PDMAEMA and at 5 and 7.5 for the 103.3kDa PDMAEMA. Dynamic light scattering and zeta potential measurements confirmed the nanosize and positive charge of polyplexes for all ratios and for both polymers. Both high and low Mw PDMAEMA were able to efficiently complex and protect DNA from DNase I degradation. Their cytotoxicity was evaluated using a non-retinal cell line (HEK293) and a retinal pigment epithelium (RPE) cell line (D407). We have found that cytotoxicity of the free polymer is concentration and time dependent, as expected, and negligible for all the concentrations of the PDMAEMA-DNA polyplexes. Furthermore, for the concentrations to be used in vivo, the 354kDa PDMAEMA showed no signs of inflammation upon injection in the intravitreal space of C57BL/6 mice. The transfection efficiency, as evaluated by fluorescence microscopy and flow cytometry, showed that the D407 retinal cells were transfected by polyplexes of both high and low Mw PDMAEMA, but with varied efficiency, which was dependent on the N/P ratio. Althogether, these results suggest that PDMAEMA is a feasible candidate for non-viral gene delivery to the retina, and this work constitutes the basis of further studies to elucidate the bottleneck in transfection and further optimization of the material.

Os sistemas de climatização, hoje em dia, são implementados em vários edifícios, cujo objetivo é obter conforto térmico. Este trabalho cingiu-se a edifícios hoteleiros, por dois motivos principais, o facto de haver uma exigência de conforto térmico por parte dos hóspedes instalados nas unidades hoteleiras e por a região algarvia ser caracteristicamente turística. Foram alvo de estudo quatro unidades hoteleiras: dois hotéis (uma unidade de 3 estrelas e uma unidade de 4 estrelas) situados no município de Faro e, dois hotéis (uma unidade de 3 estrelas e uma unidade de 4 estrelas) no município de Albufeira. Para a obtenção dos dados, foi efetuado um inquérito aos hóspedes das quatro unidades hoteleiras, realizadas medições de ruído nos quartos em situação desfavorável e favorável à propagação de ruído e verificou-se quais os equipamentos que transmitiam vibração para a estrutura do edifício.

This work describes the synthesis of nanosized metal sulfides and respective SiO2 and/or TiO2 composites in high yield via a straightforward process, under ambient conditions (temperature and pressure), by adding to aqueous metals a nutrient solution containing biologically generated sulfide from sulfate-reducing bacteria (SRB). The nanoparticles‘ (NPs) morphological properties were shown not to be markedly altered by the SRB growth media composition neither by the presence of bacterial cells. We further extended the work carried out, using the effluent of a bioremediation system previously established. The process results in the synthesis of added value products obtained from metal rich effluents, such as Acid Mine Drainage (AMD), when associated with the bioremediation process. Precipitation of metals using sulfide allows for the possibility of selective recovery, as different metal sulfides possess different solubilities. We have evaluated the selective precipitation of CuS, ZnS and FeS as nanosized metal sulfides. Again, we have also tested the precipitation of these metal sulfides in the presence of support structures, such as SiO2. Studies were carried out using both artificial and real solutions in a continuous bioremediation system. We found that this method allowed for a highly selective precipitation of copper and a lower selectivity in the precipitation of zinc and iron, though all metals were efficiently removed (>93% removal). This research has also demonstrated the potential of ZnS-TiO2 nanocomposites as catalysts in the photodegradation of organic pollutants using the cationic dye, Safranin-T, as a model contaminant. The influence of the catalyst amount, initial pH and dye concentration were also evaluated. Finally, the efficiency of the precipitates as catalysts in sunlight mediated photodegradation was investigated, using different volumes of dye-contaminated water (150 mL and 10 L). This work demonstrates that all tested composites have the potential to be used as photocatalysts for the degradation of Safranin-T.

A presente dissertação tem como objetivo principal contribuir para um maior conhecimento e compreensão das propriedades catalíticas de sistemas baseados em metais de transição com estado de oxidação elevado. O trabalho desenvolvido englobou a síntese e a caracterização de novos compostos de molibdénio, tendo como ponto de partida a síntese de cis-dioxocomplexos do tipo MoO2Cl2Ln (n = 1 ou 2, conforme ligandos bidentados ou monodentados, respetivamente) a partir dos precursores MoO2Cl2, MoO3 e Na2MoO4 com ligandos como dimetilformamida (DMF), tris(benzimidazolil)metano (HC(bim)3). Os compostos MoO2Cl2(DMF)2 (Capítulo2) e [MoO2Cl(HC(bim)3]Y (Y = Cl, BF4 e PF6) (Capítulo 3) foram testados como precursores catalíticos na epoxidação de olefinas, em fase heterogénea. O primeiro foi imobilizado no suporte MCM-41 enquanto os últimos foram heterogeneizados com recurso a líquidos iónicos - [BMIM]Y e [BMPy]Y (BIM = 1-n-butil-3-metilimidazólio, BMPy = 1-n-butil-3-metilpiridina; Y = BF4 ou PF6), como estratégia alternativa à heterogeneização dos catalisadores homogéneos. Por outro lado, aprofundou-se também a epoxidação de olefinas em fase homogénea e, para isso, sintetizaram-se os compostos Mo(CO)3(HC(pz)3) (HC(pz)3 = tris(1-pirazoli)metano), Mo(CO)3(HC(3,5-Me2pz)3 (HC(3,5-Me2pz)3 = tris(3,5-dimetil-1-pirazolil)metano) (Capítulo 4) e Mo(CO)4(BPM) (BPM = bis(pirazolil)metano) (Capítulo 5) a partir do material de partida Mo(CO)6. A descarbonilação oxidativa destes compostos, com TBHP, originaram estruturas cristalinas do tipo dioxo--oxo, [{MoO2(HC(pz)3)}2(2-O)][Mo6O19] e [{MoO2(HC(3,5-Me2pz)3)}2(2-O)][Mo6O19], e oxodiperoxo, MoO(O2)2(BPM), respetivamente. Obtiveram-se os mesmos produtos in situ durante as respetivas reações catalíticas. Todos os compostos foram utilizados como catalisadores ou pré-catalisadores na epoxidação de olefinas, com bons resultados. Por fim e, tendo em conta o crescente interesse em materiais híbridos orgânicos-inorgânicos baseados em óxidos de molibdénio, sintetizou-se o composto MoO2Cl2(pypzEA) (pypzEA = etil[(3-piridin-2-il)-1H-pirazol-1-il]acetato) o qual, ao reagir com água, originou o material híbrido [Mo2O6(HpypzA)] (HpypzA = [3-(piridin-2-il)-1H-pirazol-1-il]acetato) (Capítulo 6). A estrutura deste último composto foi resolvida por difração de monocristal e de raio-X de pós, em conjunto com a informação proveniente de técnicas como FT-IR, RMN 13C CP/MAS e análise elementar. Este material híbrido apresenta um bom desempenho catalítico quando o TBHP é usado como oxidante e continua estável e a comportar-se como um catalisador heterogéneo mesmo após sofrer testes de reciclagem e de filtração. Ainda durante este estudo constatou-se que a reação oxidativa do material de partida, MoO2Cl2(pypzEA), com TBHP, originou o oxo(diperoxo)complexo MoO(O2)2(pypzEA) e o complexo octamérico [Mo8O24(pypzEA)4] obtido previamente pela descarbonilação oxidativa do Mo(CO)4(pypzEA). Todos os catalisadores ou precursores de espécies ativas foram analisados por técnicas de caracterização comuns (AE, FT-IR, Raman, ATR, 1H RMN) bem como por técnicas de caracterização do estado sólido (EXAFS, DRX de pós e de cristal único, 13C e 29Si RMN CP MAS). Os compostos com propriedades catalíticas e pré-catalíticas promissoras foram estudados nas reações catalíticas de epoxidação de diversas olefinas, nomeadamente, cis-cicloocteno, 1-octeno, trans-2-octeno, -pineno, (R)-(+)-limoneno, estireno, norborneno, entre outras.

Bone morphogenetic proteins (BMPs) are multifunctional growth factors belonging to the transforming growth factor β (TGFβ) superfamily with a central role in bone formation and mineralization. BMP2, a founding member of this family, has demonstrated remarkable osteogenic properties and is clinically used to promote bone repair and fracture healing. Lack of basic data on factors regulating BMP2 expression and activity have hampered a better understanding of its role in bone formation and bone-related diseases. The objective of this work was to collect new functional data and determine spatiotemporal expression patterns in a fish system aiming towards a better understanding of BMP2 function and regulation. Transcriptional and post-transcriptional regulation of gilthead seabream BMP2 gene was inferred from luciferase reporter systems. Several bone- and cartilage-related transcription factors (e.g. RUNX3, MEF2c, SOX9 and ETS1) were found to regulate BMP2 transcription, while microRNA 20a was shown to affect stability of the BMP2 transcript and thus the mineralogenic capacity of fish bone-derived host cells. The regulation of BMP2 activity through an interaction with the matrix Gla protein (MGP) was investigated in vitro using BMP responsive elements (BRE) coupled to luciferase reporter gene. Although we demonstrated the functionality of the experimental system in a fish cell line and the activation of BMP signaling pathway by seabream BMP2, no conclusive evidence could be collected on a possible interaction beween MGP and BMP2. The evolutionary relationship among the members of BMP2/4/16 subfamily was inferred from taxonomic and phylogenetic analyses. BMP16 diverged prior to BMP2 and BMP4 and should be the result of an ancient genome duplication that occurred early in vertebrate evolution. Structural and functional data suggested that all three proteins are effectors of the BMP signaling pathway, but expression data revealed different spatiotemporal patterns in teleost fish suggesting distinct mechanisms of regulation. In this work, through the collection of novel data, we provide additional insight into the regulation, the structure and the phylogenetic relationship of BMP2 and its closely related family members.
