Botryosphaeran, a new exopolysaccharide from the endophytic fungus Botryosphaeria rhodina MAMB-05, and algal laminarin were hydrolyzed by partially-fractionated enzymes of the beta-glucanolytic complex from Trichoderma harzianum Rifai. beta-Glucanase fractions (F-I and F-II) separated by gel permeation chromatography presented different modes of attack on botryosphaeran and laminarin. Botryosphaeran was hydrolyzed to the extent of 66% (F-I) and 98% (F-II) within 30 min, and its main hydrolysis products were gluco-oligosaccharides of DP >= 4, with lesser amounts of glucose, di- and tri-saccharides. The action of enzyme fractions I and II on laminarin resulted in 15% conversion to glucose, while the percentage of saccharification was radically different (70% for F-I and 25% for F-II). The different product arrays within the polysaccharide hydrolysates can be explained by the difference in the enzymes' specificities within each enzyme fraction, and the molecular structures of the polysaccharides and their complexity.

Endo-polygalacturonase (endo-PG), exo-polygalacturonase (exo-PG) and pectin liase (PL) were produced by solid-state fermentation of a mixture of orange bagasse and wheat bran (1:1) with the filamentous fungus Penicillium viridicatum RFC3. This substrate was prepared with two moisture contents, 70% and 80%, and each was fermented in two types of container, Erlenmeyer flask and polypropylene pack. When Erlenmeyer flasks were used, the medium containing 80% of initial moisture afforded higher PL production while neither exo- nor endo-PG production was influenced by substrate moisture. The highest enzyme activities obtained were 0.70 U mL(-1) for endo-PG, 8.90 U mL(-1) for exo-PG, and 41.30 U mL(-1) for PL. However, when the fermentation was done in polypropylene packs, higher production of all three enzymes was obtained at 70% moisture (0.7 and 8.33 U mL(-1) for endo- and exo-PG and 100 U mL(-1) for PL). An increase in the pH and decrease in the reducing sugar content of the medium was observed. The fungus was able to produce pectin esterase and other depolymerizing enzymes such as xylanase, CMCase, protease and amylase. (c) 2005 Elsevier Ltd. All rights reserved.

The thermophilic fungus Thermoascus aurantiacus 179-5 and the mesophilic Aureobasidium pullulans ER-16 were cultivated in corn-cob by solid state fermentation for P-glucosidase production. After fermentation both enzymes were purified. The beta-glucosidases produced by the strains A. pullulans and T aurantiacus were most active at pH 4.0-4.5 and 4.5, with apparent optimum temperatures at 80 and 75 degrees C, respectively. Surprisingly, the enzyme produced by the mesophilic A. pullulans was stable over a wider range of pH (4.5-9.5 against 4.5-6.5) and more thermostable (98% after 1 h at 75 degrees C against 98% after 1 h at 70 degrees C) than the enzyme from the thermophilic T. aurantiacus. The t((1/2)) at 80 degrees C were 90 and 30 min for A. pullulans and T. aurantiacus, respectively. beta-Glucosidase thermoinactivation followed first-order kinetics and the energies of denaturation were 414 and 537 kJ mol(-1) for T. aurantiacus and A. pullulans, respectively. The result showed that beta-glucosidase obtained from the mesophilic A. pullulans is more stable than that obtained from the thermophilic T. aurantiacus. (C) 2007 Elsevier Ltd. All rights reserved.

A cultura da seringueira (Hevea brasiliensis Muell. Agr.) vem se expandindo consideravelmente no noroeste do estado de São Paulo. O objetivo deste trabalho foi avaliar a fauna de ácaros em dois sistemas comuns de cultivo nessa região, um em monocultivo e outro em consórcio com gariroba (Syagrus oleracea (Mart.) Becc. O estudo foi conduzido entre maio de 2002 e abril de 2003, tomando-se amostras mensais de 150 folíolos de seringueira de cada plantação e 150 folíolos de gariroba. As faunas dos dois sistemas foram semelhantes; os números de espécies, gêneros e famílias, assim como as famílias mais diversas e mais abundantes foram semelhantes entre eles. Dezoito espécies ocorreram exclusivamente em plantas de gariroba. O número de ácaros sobre estas foi consideravelmente menor, mas a riqueza de espécies foi maior que em seringueiras. Dentre as espécies mais freqüentes e abundantes em seringueira, Eutetranychus banksi (McGregor) e Pronematus sp. foram as únicas também freqüentes em gariroba, porém bem menos abundantes. Nenhum dos predadores abundantes em seringueira (Zetzellia aff. yusti, Euseius citrifolius Denmark & Muma, Pronematus sp. e Spinibdella sp.) foi abundante em gariroba. O fungo Hirsutella thompsonii Fisher foi o único patógeno de ácaros encontrado neste estudo, atacando Calacarus heveae Feres. Aparentemente, na forma como cultivada na região, a gariroba não constitui um reservatório adequado dos ácaros predadores mais importantes em seringueira.
