The influence of feed form and energy concentration of the diet on growth performance and the development of the gastrointestinal tract (GIT) was studied in brown-egg laying pullets. Diets formed a 2 x 5 factorial with 2 feed forms (mash vs. crumbles) and 5 levels of energy differing in 50 kcal AMEn/kg. For the entire study (0 to 17 wk of age) feeding crumbles increased ADFI (52.9 vs. 49.7 g; P < 0.001) and ADG (12.7 vs. 11.6 g; P < 0.001) and improved feed conversion ratio (FCR; 4.18 vs. 4.27; P < 0.001). An increase in the energy content of the diet decreased ADFI linearly (P < 0.001) and improved FCR quadratically (P < 0.01) but energy intake (kcal AMEn/d) was not affected. BW uniformity was higher (P < 0.05) in pullets fed crumbles than in those fed mash but was not affected (P > 0.05) by energy content of the diet. At 5, 10, and 17 wk of age, the relative weight (RW, % BW) of the GIT and the gizzard, and gizzard digesta content were lower (P < 0.05 to P < 0.001) and gizzard pH was higher (P < 0.05 to P < 0.001) in pullets fed crumbles than in pullets fed mash. Energy concentration of the diet did not affect any of the GIT variables studied. In summary, feeding crumbles improved pullet performance and reduced the RW of the GIT and gizzard, and increased gizzard pH at all ages. An increase in the energy content of the diet improved FCR from 0 to 17 wk of age. The use of crumbles and the increase in the AMEn content of the diet might be used adventageously when the objetive is to increase the BW of the pullets. However, crumbles affected the development and weight of the organs of the GIT, which might have negative effects on feed intake and egg production at the beginning of the egg laying cycle.

Transplantations of fully allogeneic, autoimmune-resistant T-cell-depleted marrow (TCDM) plus syngeneic, autoimmune-prone TCDM into lethally irradiated BXSB mice were carried out to investigate the ability of the mixed bone marrow transplantation (BMT) to prevent development of autoimmune disease and, at the same time, to reconstitute fully the immunity functions of heavily irradiated BXSB recipients. Male BXSB mice were engrafted with mixed TCDM from both allogeneic, autoimmune-resistant BALB/c mice and syngeneic, autoimmune-prone BXSB mice. BMT with mixed TCDM from both resistant and susceptible strains of mice (mixed BMT) prolonged the median life span and inhibited development of glomerulonephritis in BXSB mice. BMT with mixed TCDM also prevented the formation of anti-DNA antibodies that is typically observed in male mice of this strain. Moreover, mixed BMT reconstituted primary antibody production in BXSB recipients, so that no annoying immunodeficiencies that are regularly observed in fully allogeneic chimeras were present in the recipient of the mixed TCDM. These findings indicate that transplanting allogeneic, autoimmune-resistant TCDM plus syngeneic, autoimmune-prone TCDM into lethally irradiated BXSB mice prevents development of autoimmune disease in this strain of mice. In addition, this dual BMT reconstitutes the immunity functions and avoids the immunodeficiencies that occur regularly in fully allogeneic chimeras after total-body irradiation.

When the heart fails, there is often a constellation of biochemical alterations of the β-adrenergic receptor (βAR) signaling system, leading to the loss of cardiac inotropic reserve. βAR down-regulation and functional uncoupling are mediated through enhanced activity of the βAR kinase (βARK1), the expression of which is increased in ischemic and failing myocardium. These changes are widely viewed as representing an adaptive mechanism, which protects the heart against chronic activation. In this study, we demonstrate, using in vivo intracoronary adenoviral-mediated gene delivery of a peptide inhibitor of βARK1 (βARKct), that the desensitization and down-regulation of βARs seen in the failing heart may actually be maladaptive. In a rabbit model of heart failure induced by myocardial infarction, which recapitulates the biochemical βAR abnormalities seen in human heart failure, delivery of the βARKct transgene at the time of myocardial infarction prevents the rise in βARK1 activity and expression and thereby maintains βAR density and signaling at normal levels. Rather than leading to deleterious effects, cardiac function is improved, and the development of heart failure is delayed. These results appear to challenge the notion that dampening of βAR signaling in the failing heart is protective, and they may lead to novel therapeutic strategies to treat heart disease via inhibition of βARK1 and preservation of myocardial βAR function.

Finite replicative lifespan, or senescence, of mammalian cells in culture is a phenomenon that has generated much curiosity since its description. The obvious significance of senescence to organismal aging and the development of cancer has engendered a long-lasting and lively debate about its mechanisms. Recent discoveries concerning the phenotypes of telomerase knockout mice, the consequences of telomerase reexpression in somatic cells, and genes that regulate senescence have provided striking molecular insights but also have uncovered important new questions. The objective of this review is to reconcile old observations with new molecular details and to focus attention on the key remaining puzzles.

Two important features of amphibian metamorphosis are the sequential response of tissues to different concentrations of thyroid hormone (TH) and the development of the negative feedback loop between the pituitary and the thyroid gland that regulates TH synthesis by the thyroid gland. At the climax of metamorphosis in Xenopus laevis (when the TH level is highest), the ratio of the circulating precursor thyroxine (T4) to the active form 3,5,3′-triiodothyronine (T3) in the blood is many times higher than it is in tissues. This difference is because of the conversion of T4 to T3 in target cells of the tadpole catalyzed by the enzyme type II iodothyronine deiodinase (D2) and the local effect (cell autonomy) of this activity. Limb buds and tails express D2 early and late in metamorphosis, respectively, correlating with the time that these organs undergo TH-induced change. T3 is required to complete metamorphosis because the peak concentration of T4 that is reached at metamorphic climax cannot induce the final morphological changes. At the climax of metamorphosis, D2 expression is activated specifically in the anterior pituitary cells that express the genes for thyroid-stimulating hormone but not in the cells that express proopiomelanocortin. Physiological concentrations of T3 but not T4 can suppress thyrotropin subunit β gene expression. The timing and the remarkable specificity of D2 expression in the thyrotrophs of the anterior pituitary coupled with the requirement for locally synthesized T3 strongly support a role for D2 in the onset of the negative feedback loop at the climax of metamorphosis.

Glial cell line-derived neurotrophic factor (GDNF) promotes survival of midbrain dopaminergic neurons and motoneurons. Expression of GDNF mRNA in cerebellum raises the possibility that cells within this structure might also respond to GDNF. To examine potential trophic activities of GDNF, dissociated cultures of gestational day 18 rat cerebellum were grown for < or = 21 days in the presence of factor. GDNF increased Purkinje cell number without affecting the overall number of neurons or glial cells. A maximal response (50% above control) was elicited with GDNF at 1 pg/ml. Effects of GDNF on Purkinje cell differentiation were examined by scoring the morphologic maturation of cells in treated and control cultures. GDNF increased the proportion of Purkinje cells that displayed relatively mature morphologies, characterized by dendritic thickening and the development of spines and filopodial extensions. Morphologic maturation of the overall neuronal population was unaffected. In sum, our data indicate that GDNF is a potent survival and differentiation factor for Purkinje cells, the efferent neurons of cerebellar cortex. Together with its other actions, these findings raise the possibility that GDNF might be a critical trophic factor at multiple loci in neuronal circuits that control motor function.

The discovery that the epsilon 4 allele of the apolipoprotein E (apoE) gene is a putative risk factor for Alzheimer disease (AD) in the general population has highlighted the role of genetic influences in this extremely common and disabling illness. It has long been recognized that another genetic abnormality, trisomy 21 (Down syndrome), is associated with early and severe development of AD neuropathological lesions. It remains a challenge, however, to understand how these facts relate to the pathological changes in the brains of AD patients. We used computerized image analysis to examine the size distribution of one of the characteristic neuropathological lesions in AD, deposits of A beta peptide in senile plaques (SPs). Surprisingly, we find that a log-normal distribution fits the SP size distribution quite well, motivating a porous model of SP morphogenesis. We then analyzed SP size distribution curves in genotypically defined subgroups of AD patients. The data demonstrate that both apoE epsilon 4/AD and trisomy 21/AD lead to increased amyloid deposition, but by apparently different mechanisms. The size distribution curve is shifted toward larger plaques in trisomy 21/AD, probably reflecting increased A beta production. In apoE epsilon 4/AD, the size distribution is unchanged but the number of SP is increased compared to apoE epsilon 3, suggesting increased probability of SP initiation. These results demonstrate that subgroups of AD patients defined on the basis of molecular characteristics have quantitatively different neuropathological phenotypes.

A presente dissertação é fruto da pesquisa que teve como objetivo investigar a participação docente, do Ensino Fundamental II, no processo de formação continuada e principalmente na construção da política curricular do município de Itatiba, o qual tem o ensino municipalizado do 1º ao 9º ano desde 1999. Vale ressaltar que o estudo discorreu entre a gestão municipal de 2001 a 2008 e a gestão de 2009 até 2012. A investigação se desenvolveu a partir das seguintes fontes: entrevista com o Secretário de Educação que atuou entre 2001 e 2008 e com a Secretária de 2009 a 2012; entrevista com duas supervisoras que participaram de todo o processo de 2001 a 2012; entrevista com duas diretoras que atuaram desde a municipalização até 2012; entrevista com quatro professores que se efetivaram após a municipalização (2001), atuam até hoje e participaram do processo de formação até 2012; documentos oficiais: Constituição Federal de 1988, LDB (9394/96), Diretrizes Curriculares Nacionais, FUNDEF (1997), FUNDEB (2007), Deliberação CEE nº73/2008, Lei da municipalização (3110/99), Lei do Sistema de Ensino do município de Itatiba (3485/2001), Projeto Político Pedagógico da SE de 2008, assim como os documentos curriculares do município: de 2001 a 2008 Orientações Curriculares e de 2009 a 2012 Currículo do Ensino Fundamental do 6º ao 9º de todas as disciplinas. É por meio de uma pesquisa qualitativa que esta dissertação se delineia. Desse modo, com entrevistas semiestruturadas bem como roteiros previamente elaborados, tivemos como critérios de escolha dos sujeitos: profissionais da educação que atuaram desde a municipalização até 2012, que participaram de todo o processo de formação continuada e da construção da política curricular. Nessa perspectiva, as entrevistas foram gravadas em áudio e, posteriormente, transcritas para o processo de análise. Dessa forma, o processo de análise consistiu na organização dos dados em categorias e subcategorias, estas estabelecidas pelo próprio conteúdo das entrevistas colhidas dos sujeitos, como também no entrecruzamento do conteúdo das entrevistas com os dados documentais das diferentes fontes utilizadas, que foram, por sua vez, analisados e discutidos à luz do referencial teórico que fundamentou a pesquisa. O estudo foi realizado com base em contribuições de autores que discutem o currículo (Veiga, Silva, Apple, Moreira, Sacristán), os saberes docentes (Tardif), a formação continuada de professores (Nóvoa, Hargreaves), a municipalização (Azanha, Arelaro).Os resultados do trabalho revelam, entre outras coisas, que a participação docente ocorreu no ponto de vista principalmente dos professores na gestão de 2009 a 2012, uma vez que esses se sentiram parte de todo o processo de discussão e construção.

The dynamic deformation upon stretching of Ni nanowires as those formed with mechanically controllable break junctions or with a scanning tunneling microscope is studied both experimentally and theoretically. Molecular dynamics simulations of the breaking process are performed. In addition, and in order to compare with experiments, we also compute the transport properties in the last stages before failure using the first-principles implementation of Landauer's formalism included in our transport package ALACANT.

This study takes on the issue of political and socio-economic conditions for the hydrogen economy as part of a future low carbon society in Europe. It is subdivided into two parts. A first part reviews the current EU policy framework in view of its impact on hydrogen and fuel cell development. In the second part an analysis of the regional dynamics and possible hydrogen and fuel cell clusters is carried out. The current EU policy framework does not hinder hydrogen development. Yet it does not constitute a strong push factor either. EU energy policies have the strongest impact on hydrogen and fuel cell development even though their potential is still underexploited. Regulatory policies have a weak but positive impact on hydrogen. EU spending policies show some inconsistencies. Regions with a high activity level in HFC also are generally innovative regions. Moreover, the article points out certain industrial clusters that favours some regions' conditions for taking part in the HFC development. However, existing hydrogen infrastructure seems to play a minor role for region's engagement. An overall well-functioning regional innovation system is important in the formative phase of an HFC innovation system, but that further research is needed before qualified policy implications can be drawn. Looking ahead the current policy framework at EU level does not set clear long term signals and lacks incentives that are strong enough to facilitate high investment in and deployment of sustainable energy technologies. The likely overall effect thus seems to be too weak to enable the EU hydrogen and fuel cell deployment strategy. According to our analysis an enhanced EU policy framework pushing for sustainability in general and the development of hydrogen and fuel cells in particular requires the following: 1) A strong EU energy policy with credible long term targets; 2) better coordination of EU policies: Europe needs a common understanding of key taxation concepts (green taxation, internalisation of externalities) and a common approach for the market introduction of new energy technologies; 3) an EU cluster policy as an attempt to better coordinate and support of European regions in their efforts to further develop HFC and to set up the respective infrastructure.

A multiproxy record including benthic foraminifera, diatoms and XRF data of a marine sediment core from a SW Greenland fjord provides a detailed reconstruction of the oceanographic and climatic variations of the region during the last 4400 cal. years. The lower part of our record represents the final termination of the Holocene Thermal Maximum. After the onset of the 'Neoglaciation' at approximately 3.2 ka cal. BP, the fjord system was subject to a number of marked hydrographical changes that were closely linked to the general climatic and oceanographic development of the Labrador Sea and the North Atlantic region. Our data show that increased advection of Atlantic water (Irminger Sea Water) from the West Greenland Current into the Labrador Sea was a typical feature of Northeast Atlantic cooling episodes such as the 'Little Ice Age' and the 'European Dark Ages', while the advection of Irminger Sea Water decreased significantly during warm episodes such as the 'Mediaeval Warm Period' and the 'Roman Warm Period'.Whereas the 'Mediaeval Warm Period' was characterized by relatively cool climate as suggested by low meltwater production, the preceding 'Dark Ages' display higher meltwater runoff and consequently warmer climate. When compared with European climate, these regional climate anomalies indicate persisting patterns of advection of colder, respectively warmer air masses in the study region during these periods and thus a long-term seesaw climate pattern between West Greenland and Europe.

A reconstruction of Holocene sea ice conditions in the Fram Strait provides insight into the palaeoenvironmental and palaeoceanographic development of this climate sensitive area during the past 8,500 years BP. Organic geochemical analyses of sediment cores from eastern and western Fram Strait enable the identification of variations in the ice coverage that can be linked to changes in the oceanic (and atmospheric) circulation system. By means of the sea ice proxy IP25, phytoplankton derived biomarkers and ice rafted detritus (IRD) increasing sea ice occurrences are traced along the western continental margin of Spitsbergen throughout the Holocene, which supports previous palaeoenvironmental reconstructions that document a general cooling. A further significant ice advance during the Neoglacial is accompanied by distinct sea ice fluctuations, which point to short-term perturbations in either the Atlantic Water advection or Arctic Water outflow at this site. At the continental shelf of East Greenland, the general Holocene cooling, however, seems to be less pronounced and sea ice conditions remained rather stable. Here, a major Neoglacial increase in sea ice coverage did not occur before 1,000 years BP. Phytoplankton-IP25 indices ("PIP25-Index") are used for more explicit sea ice estimates and display a Mid Holocene shift from a minor sea ice coverage to stable ice margin conditions in eastern Fram Strait, while the inner East Greenland shelf experienced less severe to marginal sea ice occurrences throughout the entire Holocene.
