In the modern society, communications and digital transactions are becoming the norm rather than the exception. As we allow networked computing devices into our every-day actions, we build a digital lifestyle where networks and devices enrich our interactions. However, as we move our information towards a connected digital environment, privacy becomes extremely important as most of our personal information can be found in the network. This is especially relevant as we design and adopt next generation networks that provide ubiquitous access to services and content, increasing the impact and pervasiveness of existing networks. The environments that provide widespread connectivity and services usually rely on network protocols that have few privacy considerations, compromising user privacy. The presented work focuses on the network aspects of privacy, considering how network protocols threaten user privacy, especially on next generation networks scenarios. We target the identifiers that are present in each network protocol and support its designed function. By studying how the network identifiers can compromise user privacy, we explore how these threats can stem from the identifier itself and from relationships established between several protocol identifiers. Following the study focused on identifiers, we show that privacy in the network can be explored along two dimensions: a vertical dimension that establishes privacy relationships across several layers and protocols, reaching the user, and a horizontal dimension that highlights the threats exposed by individual protocols, usually confined to a single layer. With these concepts, we outline an integrated perspective on privacy in the network, embracing both vertical and horizontal interactions of privacy. This approach enables the discussion of several mechanisms to address privacy threats on individual layers, leading to architectural instantiations focused on user privacy. We also show how the different dimensions of privacy can provide insight into the relationships that exist in a layered network stack, providing a potential path towards designing and implementing future privacy-aware network architectures.

O trabalho que se segue estuda a natureza dos textos “criativos” produzidos por alunos de 12/13 anos, em situação de sala de aula. Para o efeito seleccionámos duas turmas do Ensino Básico – 3º ciclo – de perfis diferentes e acompanhámos os sessenta alunos que se enquadravam nessa faixa etária durante um ano lectivo, seguindo a sua evolução. Centrámos o nosso estudo no aluno, como produtor do texto, preocupado em fazer o seu melhor, desenvolvendo os temas a tratar com a originalidade de que era capaz e procurando a forma adequada ao assunto e à situação. Verificámos a sua preocupação em cumprir as expectativas do professor/receptor, mas também a sua vontade de entrar em negociações, numa tentativa de integrar as expectativas do docente nos seus próprios interesses pessoais. Para a sistematização das características globais dos textos produzidos pelos alunos utilizámos o modelo de análise textual de Beaugrande & Dressler, por nos parecer adaptar-se à situação de diálogo escolar, subjacente a qualquer produção de texto em sala de aula. O levantamento das características globais dos textos dos alunos tornou evidente as suas diferenças e os seus traços de individualidade. Por isso, achámos que este estudo não estaria completo sem a complementação duma segunda parte em que procurássemos encontrar os traços estilísticos desses mesmos textos, numa tentativa de analisar a presença de algumas características de estilo colectivas, produto das preocupações comuns aos alunos, assim como alguns traços estilísticos individuais, consequência de vivências particulares. Terminaremos fazendo uma síntese dos dados obtidos e reflectindo sobre as suas implicações na situação de ensino/aprendizagem.

This study aims to measure the effectiveness of four post-fire emergency techniques for reducing overland flow and soil erosion on the central-Portugal typical forest. The selection and development of these techniques was based on the review of the scientific background, but specially after checking throughout field rainfall simulation experiments which factors were the key for runoff and soil erosion on the specific case of high repellent soils. The forest residue mulch, a new treatment never tested before, was highly effective in reducing runoff and soil erosion in recently burnt eucalypt forest. The logging slash mulch had no obvious effect, but it was attributed to the small amounts of runoff and sediments that the untreated plots produced due to the extensive needle cast following a low severity fire. The hydromulch, a mixture of water, organic fibres, seeds, nutrients and a surfactant used in cutted slopes rehabilitation was also highly successful and was specially indicated for especially sensible areas. The utilization of polyacrylamides, a chemical agent with good performance in agricultural erosion, was not successful in post-fire runoff and soil erosion control, once that did not alter the most important key factor for soil erosion: the ground cover. The development of a new fibre optic turbidity sensor was a successful development on the soil erosion determination methodology, and its patent is being processed in the mean time.

Both skeletal and cardiac muscles daily burn tremendous amounts of ATP to meet the energy requirements for contraction. So, it is not surprising that the maintenance of mitochondrial morphology, number, distribution and functionality in striated muscle are important for muscle homeostasis. In these tissues mitochondria present the added dimension of two populations, the intermyofibrillar (IMF) and the subsarcolemmal (SS) mitochondria, being IMF the most abundant one. In the present thesis, the molecular mechanisms harboured in mitochondria of striated muscles were studied using animal models, to better comprehend the role of mitochondrial plasticity in several pathophysiological conditions such as aging, diabetes mellitus and bladder cancer. The comparative analysis of IMF and SS populations isolated from heart evidenced a higher respiratory chain activity of mitochondria interspersed in the contractile apparatus. The higher susceptible of SS respiratory chain complexes subunits to carbonylation, but not to nitration, seems to justify the lower respiratory chain activity observed in this mitochondrial population. Our results showed that in heart from aged mice there is an accumulation of dysfunctional mitochondria. The age-related decrease of oxidative phosphorylation activity seems to be justified, at least partially, by the increased proneness of mitochondrial proteins as OXPHOS subunits and MnSOD to oxidative modifications. Moreover, a sedentary lifestyle seems to worsen the functional consequences of aging in heart by increasing mitochondrial proteins susceptibility to nitration. In skeletal muscle from rats with type 1 diabetes mellitus induced by streptozotocin administration, we verified the accumulation of dysfunctional mitochondria due, at least in part, to the impairment of PQC system. Indeed, the decreased activity of AAA proteases was accompanied by the accumulation of oxidatively modified mitochondrial proteins with impact in respiratory chain activity. The diminishing of mitochondria activity also underlies cancer-induced muscle wasting. Indeed, using a rat model of chemically induced urothelial carcinoma we verified that the loss of gastrocnemius mass was related to mitochondrial dysfunction due to, at least partially, the down-regulation of PQC system involving the mitochondrial proteases paraplegin and Lon. PQC impairment resulted in the accumulation of oxidatively modified mitochondrial proteins. In overall, regardless the pathophysiological stimuli that promote mitochondrial alterations, there are similarities in the pattern of disease-related mitochondrial plasticity. The diminished capacity for ATP production in striated muscle seems to be due to increased oxidative damage of mitochondrial proteins, namely subunits of respiratory chain complexes, metabolic proteins and MnSOD. Our data highlighted, for the first time, the impact of mitochondrial PQC system impairment in the accumulation of oxidized proteins, exacerbating the dysfunction of this organelle in striated muscle in several pathophysiological conditions.

Nos últimos anos, a Optoelectrónica tem sido estabelecida como um campo de investigação capaz de conduzir a novas soluções tecnológicas. As conquistas abundantes no campo da óptica e lasers, bem como em comunicações ópticas têm sido de grande importância e desencadearam uma série de inovações. Entre o grande número de componentes ópticos existentes, os componentes baseados em fibra óptica são principalmente relevantes devido à sua simplicidade e à elevada de transporte de dados da fibra óptica. Neste trabalho foi focado um destes componentes ópticos: as redes de difracção em fibra óptica, as quais têm propriedades ópticas de processamento únicas. Esta classe de componentes ópticos é extremamente atraente para o desenvolvimento de dispositivos de comunicações ópticas e sensores. O trabalho começou com uma análise teórica aplicada a redes em fibra e foram focados os métodos de fabricação de redes em fibra mais utilizados. A inscrição de redes em fibra também foi abordado neste trabalho, onde um sistema de inscrição automatizada foi implementada para a fibra óptica de sílica, e os resultados experimentais mostraram uma boa aproximação ao estudo de simulação. Também foi desenvolvido um sistema de inscrição de redes de Bragg em fibra óptica de plástico. Foi apresentado um estudo detalhado da modulação acústico-óptica em redes em fibra óptica de sílica e de plástico. Por meio de uma análise detalhada dos modos de excitação mecânica aplicadas ao modulador acústico-óptico, destacou-se que dois modos predominantes de excitação acústica pode ser estabelecidos na fibra óptica, dependendo da frequência acústica aplicada. Através dessa caracterização, foi possível desenvolver novas aplicações para comunicações ópticas. Estudos e implementação de diferentes dispositivos baseados em redes em fibra foram realizados, usando o efeito acústico-óptico e o processo de regeneração em fibra óptica para várias aplicações tais como rápido multiplexador óptico add-drop, atraso de grupo sintonizável de redes de Bragg, redes de Bragg com descolamento de fase sintonizáveis, método para a inscrição de redes de Bragg com perfis complexos, filtro sintonizável para equalização de ganho e filtros ópticos notch ajustáveis.

In this work several techniques to monitor the performance of optical networks were developed. These techniques are dedicated either to the measurement of the data signal parameters (optical signal to noise ratio and dispersion) or to the detection of physical failures on the network infrastructure. The optical signal to noise ratio of the transmitted signal was successfully monitored using methods based on the presence of Bragg gratings imprinted on high birefringent fibres that allowed the distinction of the signal from the noise due to its polarization properties. The monitoring of the signal group-velocity dispersion was also possible. In this case, a method based on the analysis of the electric spectrum of the signal was applied. It was experimentally demonstrated that this technique is applicable on both amplitude and phase modulated signals. It was also developed a technique to monitor the physical infrastructure of an optical access network. Once again, the application of Bragg gratings (this time imprinted on standard single mode fibres) was the basis of the developed method.

Durante a sua vida Frederico de Freitas abordou praticamente todos os géneros musicais, demonstrando uma imensa e notória versatilidade tanto na música erudita como na não erudita. O repertório para canto e piano representa uma parte substancial da sua obra vocal e, apesar de menos divulgado, não pode deixar de se considerar de grande importância. É precisamente neste género que mais se evidencia a versatilidade do seu estilo composicional. Este repertório abrange, assim, os géneros erudito e ligeiro, ou popular, pelo que as suas canções refletem a facilidade com que se movia de um meio para outro. Enquanto investigador, os estudos que realizou sobre música medieval portuguesa, o número elevadíssimo de recolhas de melodias tradicionais portuguesas que fez ao longo da vida e as investigações que realizou sobre o fado deram origem, paralelamente à composição de canções eruditas, a harmonizações de melodias medievais, a arranjos de temas na música tradicional e à composição de fados. Do ponto de vista interpretativo, a diversidade de estilos presentes nas canções de Frederico de Freitas exige do cantor lírico uma versatilidade e capacidade de se adaptar, ainda que mantendo a matiz da sua formação erudita, ao estilo próprio de cada canção. Com vista a uma interpretação estilística e vocalmente adequada, e por representarem um universo tão diferenciado, esta tese de doutoramento centra a sua atenção na problemática da vocalidade das suas canções.

In this work physical and behavioral models for a bulk Reflective Semiconductor Optical Amplifier (RSOA) modulator in Radio over Fiber (RoF) links are proposed. The transmission performance of the RSOA modulator is predicted under broadband signal drive. At first, the simplified physical model for the RSOA modulator in RoF links is proposed, which is based on the rate equation and traveling-wave equations with several assumptions. The model is implemented with the Symbolically Defined Devices (SDD) in Advanced Design System (ADS) and validated with experimental results. Detailed analysis regarding optical gain, harmonic and intermodulation distortions, and transmission performance is performed. The distribution of the carrier and Amplified Spontaneous Emission (ASE) is also demonstrated. Behavioral modeling of the RSOA modulator is to enable us to investigate the nonlinear distortion of the RSOA modulator from another perspective in system level. The Amplitude-to-Amplitude Conversion (AM-AM) and Amplitude-to-Phase Conversion (AM-PM) distortions of the RSOA modulator are demonstrated based on an Artificial Neural Network (ANN) and a generalized polynomial model. Another behavioral model based on Xparameters was obtained from the physical model. Compensation of the nonlinearity of the RSOA modulator is carried out based on a memory polynomial model. The nonlinear distortion of the RSOA modulator is reduced successfully. The improvement of the 3rd order intermodulation distortion is up to 17 dB. The Error Vector Magnitude (EVM) is improved from 6.1% to 2.0%. In the last part of this work, the performance of Fibre Optic Networks for Distributed and Extendible Heterogeneous Radio Architectures and Service Provisioning (FUTON) systems, which is the four-channel virtual Multiple Input Multiple Output (MIMO), is predicted by using the developed physical model. Based on Subcarrier Multiplexing (SCM) techniques, four-channel signals with 100 MHz bandwidth per channel are generated and used to drive the RSOA modulator. The transmission performance of the RSOA modulator under the broadband multi channels is depicted with the figure of merit, EVM under di erent adrature Amplitude Modulation (QAM) level of 64 and 254 for various number of Orthogonal Frequency Division Multiplexing (OFDM) subcarriers of 64, 512, 1024 and 2048.

Optical networks are under constant evolution. The growing demand for dynamism require devices that can accommodate different types of traffic. Thus the study of transparent optical networks arises. This approach makes optical networks more "elegant" , due to a more efficient use of network resources. In this thesis, the author proposes devices that intend to form alternative approaches both in the state of art of these same technologies both in the fitting of this technologies in transparent optical networks. Given that full transparency is difficult to achieve with current technology (perhaps with more developed optical computing this is possible), the author proposes techniques with different levels of transparency. On the topic of performance of optical networks, the author proposes two techniques for monitoring chromatic dispersion with different levels of transparency. In Chapter 3 the proposed technique seems to make more sense for long-haul optical transmission links and high transmission rates, not only due to its moderate complexity but also to its potential moderate/high cost. However it is proposed to several modulation formats, particularly those that have a protruding clock component. In Chapter 4 the transparency level was not tested for various modulation formats, however some transparency is achieved by not adding any electrical device after the receiver (other than an analog-digital converter). This allows that this technique can operate at high transmission rates in excess of 100 Gbit / s, if electro-optical asynchronous sampling is used before the optical receiver. Thus a low cost and low bandwidth photo-detector can be used. In chapter 5 is demonstrated a technique for simultaneously monitoring multiple impairments of the optical network by generating novel performance analysis diagrams and by use of artificial neural networks. In chapter 6 the author demonstrates an all-optical technique for controlling the optical state of polarization and an example of how all-optical signal processing can fully cooperate with optical performance monitoring.

Portugal has strong musical traditions, which have been perpetrated by decades through folkloristic activities. In folk groups from Alto Minho (north of Portugal), folk singing is mostly performed by cantadeiras, amateur female solo singers who learn this style orally. Their vocal characteristics are distinctive when compared with other regions of the country; however, deep understanding of these vocal practices is still missing. The present work aims at studying Alto Minho cantadeira’s vocal performance in a multidimensional perspective, envisioning social, cultural and physiological understanding of this musical style. Thus, qualitative and quantitative data analyses were carried out, to: (i) describe current performance practices, (ii) explore existent perceptions about most relevant voice features in this region, (iii) investigate physiological and acoustic properties of this style, and (iv) compare this style of singing with other non-classical singing styles of other countries. Dataset gathered involved: 78 groups whose members were telephone interviewed, 13 directors who were asked to fill in a questionnaire on performance practices, 1 cantadeira in a pilot study, 16 cantadeiras in preliminary voice recordings, 77 folk group members in listening tests, and 10 cantadeiras in multichannel recordings, including audio, ELG, air flow and intra-oral pressure signals. Data were analysed through thematic content analysis, descriptive and inferential statistics, hierarchical principal components, and multivariate linear regression models. Most representative voices have a high pitched and loud voice, with a bright timbre, predominance of chest register without excessive effort, and good text intelligibility with regional accent. High representativeness levels were obtained by few cantadeiras; these sing with high levels of subglottal pressure and vocal fold contact quotient, predominance of high spectrum energy and vocal loudness, corroborating indications of prevalence of pressed phonation. These vocal characteristics resemble belting in musical theatre and share similarities with country (USA) and ojikanje (Croatia) singing. Strategies that may contribute to the preservation of this type of singing and the vocal health of current cantadeiras are discussed, pointing at the direction of continuous education among folk groups, following practices that are already adopted elsewhere in Europe.

Cell cycle and differentiation are two highly coordinated processes during organ development. Recent studies have demonstrated that core cell cycle regulators also play cell cycle-independent functions in post-mitotic neurons, and are essential for the maintenance of neuronal homeostasis. CDC25 phosphatases are well-established CDK activators and their activity is mainly associated to proliferating tissues. The expression and activity of mammalian CDC25s has been reported in adult brains. However, their physiological relevance and the potential substrates in a non-proliferative context have never been addressed. string (stg) encodes the Drosophila CDC25 homolog. Previous studies from our group showed that stg is expressed in photoreceptors (PRs) and in lamina neurons, which are two differentiated cell types that compose the fly visual system. The aims of this work are to uncover the function of stg and to identify its potential neuronal substrates, using the Drosophila visual system as a model. To gain insight into the function of stg in a non-dividing context we used the GAL4/UAS system to promote downregulation of stg in PR-neurons, through the use of an RNAi transgene. The defects caused by stg loss-of-function were evaluated in the developing eye imaginal disc by immunofluorescence, and during adult stages by scanning electron microscopy. This genetic approach was combined with a specific proteomic method, two-dimensional difference gel electrophoresis (2D-DIGE), to identify the potential substrates in PR-cells. Our results showed that stg downregulation in PRs affects the well-patterned retina organization, inducing the loss of apical maintenance of PR-nuclei on the eye disc, and ommatidia disorganization. We also detected an abnormal accumulation of cytoskeletal proteins and a disruption of the axon structure. As a consequence, the projection of PR-axons into the lamina and medulla neuropils of the optic lobe was impaired. Upon stg downregulation, we also detected that PR-cells accumulate Cyclin B. Although the rough eye phenotype observed upon stg downregulation suggests neurodegeneration, we did not detect neuronal death during larval stages, suggesting that it likely occurs during pupal stages or during adulthood. By 2D-DIGE, we identified seven proteins which were differentially expressed upon stg downregulation, and are potential neuronal substrates of Stg. Altogether, our observations suggest that Stg phosphatase plays an essential role in the Drosophila visual system neurons, regulating several cell components and processes in order to ensure their homeostasis.

Diplodia corticola is regarded as the most virulent fungus involved in cork oak decline, being able to infect not only Quercus species (mainly Q. suber and Q. ilex), but also grapevines (Vitis vinifera) and eucalypts (Eucalyptus sp.). This endophytic fungus is also a pathogen whose virulence usually manifests with the onset of plant stress. Considering that the infection normally culminates in host death, there is a growing ecologic and socio-economic concern about D. corticola propagation. The molecular mechanisms of infection are hitherto largely unknown. Accordingly, the aim of this study was to unveil potential virulence effectors implicated in D. corticola infection. This knowledge is fundamental to outline the molecular framework that permits the fungal invasion and proliferation in plant hosts, causing disease. Since the effectors deployed are mostly proteins, we adopted a proteomic approach. We performed in planta pathogenicity tests to select two D. corticola strains with distinct virulence degrees for our studies. Like other filamentous fungi D. corticola secretes protein at low concentrations in vitro in the presence of high levels of polysaccharides, two characteristics that hamper the fungal secretome analysis. Therefore, we first compared several methods of extracellular protein extraction to assess their performance and compatibility with 1D and 2D electrophoretic separation. TCA-Acetone and TCA-phenol protein precipitation were the most efficient methods and the former was adopted for further studies. The proteins were extracted and separated by 2D-PAGE, proteins were digested with trypsin and the resulting peptides were further analysed by MS/MS. Their identification was performed by de novo sequencing and/or MASCOT search. We were able to identify 80 extracellular and 162 intracellular proteins, a milestone for the Botryosphaeriaceae family that contains only one member with the proteome characterized. We also performed an extensive comparative 2D gel analysis to highlight the differentially expressed proteins during the host mimicry. Moreover, we compared the protein profiles of the two strains with different degrees of virulence. In short, we characterized for the first time the secretome and proteome of D. corticola. The obtained results contribute to the elucidation of some aspects of the biology of the fungus. The avirulent strain contains an assortment of proteins that facilitate the adaptation to diverse substrates and the identified proteins suggest that the fungus degrades the host tissues through Fenton reactions. On the other hand, the virulent strain seems to have adapted its secretome to the host characteristics. Furthermore, the results indicate that this strain metabolizes aminobutyric acid, a molecule that might be the triggering factor of the transition from a latent to a pathogenic state. Lastly, the secretome includes potential pathogenicity effectors, such as deuterolysin (peptidase M35) and cerato-platanin, proteins that might play an active role in the phytopathogenic lifestyle of the fungus. Overall, our results suggest that D. corticola has a hemibiotrophic lifestyle, switching from a biotrophic to a necrotrophic interaction after plant physiologic disturbances.This understanding is essential for further development of effective plant protection measures.

Os traumatismos crânio-encefálicos (TCE) são a causa mais frequente de lesão cerebral em jovens adultos, integrando o campo de intervenção mais frequente na prática neuropsicológica (Portellano, 2005), sendo o lobo frontal uma das estruturas cerebrais mais vulneráveis ao impacto que ocorre no cérebro durante um TCE. Lesões nesta estrutura cerebral estão frequentemente associadas a sequelas em diferentes funções cognitivas, alterações a nível do comportamento emocional e social e que podem alterar gravemente o estilo de vida do doente. Está bem documentado na literatura a relação entre o reconhecimento visual de emoções e o lobo frontal (ver, por exemplo Green, Turner, & Thompson, 2004; Croker & McDonald, 2005; Radice-Neumanet al., 2007). Os estudos indicam uma marcada dificuldade por partes destes sujeitos no reconhecimento visual de emoções básicas e uma associação entre esta dificuldade e as sequelas comportamentais (ver, por exemplo, Radice-Neumanet al., 2007). Relativamente à valência das emoções, investigações prévias indicam que existe uma maior dificuldade no reconhecimento das emoções negativas, comparativamente às emoções positivas, sendo consensual que a alegria é a emoção básica mais bem reconhecida no grupo clínico de sujeitos com TCE, quando comparados com grupos não-clínicos. Pretendemos com a presente investigação dar continuidade ao estudo sobre o processamento de emoções e verificar se existe um compromisso no reconhecimento visual de emoções básicas em doentes com TCE frontais. Adicionalmente, procuramos investigar se existe uma associação entre o reconhecimento de emoções e o desempenho dos sujeitos em provas que avaliam o funcionamento executivo. Participaram no estudo 17 sujeitos com TCE recente no lobo frontal, e/ou em regiões subcorticais com conexão aos lobos frontais, recrutados nas unidades de Neurotraumatologia, Neuropsicologia e de Neurocirurgia do Hospital de S. José, em Lisboa. Este grupo foi emparelhado a um grupo de controlo constituído por 17 sujeitos saudáveis. A ambos os grupos foi aplicada uma entrevista clínica para recolha de dados sócio-demográficos e clínicos, testes neuropsicológicos para avaliação das funções executivas e cinco sub-testes adaptados da Florida Affect Battery (FAB) para avaliar o processamento de emoções. No geral, os resultados revelam um desempenho estatisticamente inferior no grupo TCE nas provas de avaliação do reconhecimento visual de emoções, quando comparado com o grupo de controlo, sobretudo para as emoções “triste” e “zangada”. Contudo, não existe compromisso no reconhecimento da emoção “alegria”. Relativamente às funções executivas, os resultados do estudo indicam que existe correlação entre a maioria das componentes avaliadas e o reconhecimento de emoções. Em suma, o nosso estudo indica a existência de um compromisso no reconhecimento visual de emoções no grupo clínico com TCE, sendo a dificuldade superior no reconhecimento de emoções negativas.
