以克拉维酸产生菌棒状链霉菌Streptomyces clavuligerus CCRC11518(ATCC 27064)III50为出发菌株, 首先比较各种物理和化学诱变剂处理对其克拉维酸生物合成的影响, 确定了亚硝基胍为棒状链霉菌诱变育种的诱变剂及其处理剂量: 2mg/ml、40min. 经浓度为2mg/ml的亚硝基胍处理40min后, 采用新颖理性化筛选方法, 通过逐步筛选自身代谢产物抗性突变株、克拉维酸抗性突变株和链霉素抗性突变株, 最终得到一株克拉维酸高产菌VI118(效价633μg/ml), 其克拉维酸效价是出发菌株(效价377μg/ml)的167.9%. 该高产突变株在琼脂斜面培养基上连续传接10代, 克拉维酸效价保持稳定. 通过单因子和多因子摇瓶正交试验, 对高产菌株VI118的发酵条件进行了研究, 确定最佳发酵条件: 甘油60g, 水解植物蛋白 60g, KH2PO4 0.5 g, 玉米浆 7.5g, MnSO4•H2O 0.34g, MgSO4•7H2O 0.99g, FeSO4•7H2O 0.56g, 蒸馏1000ml, pH 7.0, 发酵培养基装量20ml/250ml三角瓶, 接种量10%, 培养温度28ºC, 220r/min摇床培养72h后测定效价. 在最佳发酵条件下克拉维酸效价达到651μg/ml, 同时把初始发酵培养基的昂贵成分替换为廉价的工业原料. 通过摇瓶分批补料试验, 得到最佳补料物质和补料方式:在上述最佳发酵条件下, 分别在发酵培养48h、56h、64h、72h时补加4ml无菌水, 80h发酵结束, 克拉维酸效价达到905μg/ml. 在不增加原料成本的情况下通过摇瓶补料方式克拉维酸效价为未补料的139.0%, 总产量为未补料的264%. By a novel rational screening method, mutant Streptomyces clavuligerus CCRC11518(ATCC 27064)III50(titres 377μg/ml), as the clavulanic acid-producing parent strain, was treated by NTG (2mg/ml) for 40min, and the self-generated metabolites resistant mark, the clavulanic acid resistant mark and the streptomycin resistant mark were added step by step. Finally, the mutant VI118(titres 633μg/ml)with the three marks was obtained. The clavulanic acid productivity of this mutant was increased by 167.9% compared with the parent strain. After reproducing 10 generations on the agar medium slant, the productivity of this mutant was stable. The optimum fermentation conditions were established as followings: glycerol 60g, acid hydrolyzed vegetable protein 60g, KH2PO4 0.5g, corn steep liquor 7.5g, MnSO4•H2O 0.34g, MgSO4•7H2O 0.99g, FeSO4•7H2O 0.56g, distilled water 1 liter, pH 7.0, 20ml in 250ml shake-flask, inoculation 10%(v/v), fermentation temperature 28ºC, rotation speed 220 r/min, time 72h. The clavulanic acid productivity was 651μg/ml, while used the low-priced industrial raw materials. After studying on fed-batch in the shake-flask, the optimum fed-batch manner was obtained: under optimum fermentation conditions, at 48h, 56h, 64h and 72h, adding 4ml distilled water into each flask, fermentation ending at 80h. The clavulanic acid productivity was increased by 139% compared with no fed-batch, meanwhile the total yield was increased by 264%.

The intestinal bacterial metabolites of ginsenosides are responsible for the main pharmacological activities of ginseng. The purpose of this study was to find whether these metabolites influence hepatic metabolic enzymes and to predict the potential for ginseng-prescription drug interactions. Utilizing the probe reaction of CYP3A activity, testosterone 6beta-hydroxylation, the effects of derivatives of 20(S)-protopanaxadiol and 20(S)-protopanaxatriol families on CYP3A activity in rat liver microsomes were assayed. Our results showed that ginsenosides from the 20(S)-protopanaxadiol and 20(S)-protopanaxatriol family including Rb-1, Rb-2, Rc, Compound-K, Re, and Rg(1), had no inhibitory effect, whereas Rg(2), 20(S)-panaxatriol and 20(S)-protopanaxatriol exhibited competitive inhibitory activity against CVP3A activity in these microsomes with the inhibition constants (K) of 86.4+/-0.8mum, 1.7+/-0.1mum, and 3.2+/-0.2 mum, respectively. This finding demonstrates that differences in their chemical structure might influence the effects of ginsenosides on CYP3A activity and that ginseng-derived products might have potential for significant ginseng-drug interactions.

Metabonomics, the study of metabolites and their roles in various disease states, is a novel methodology arising from the post-genomics era. This methodology has been applied in many fields, including work in cardiovascular research and drug toxicology. In this study, metabonomics method was employed to the diagnosis of Type 2 diabetes mellitus (DM2) based on serum lipid metabolites. The results suggested that serum fatty acid profiles determined by capillary gas chromatography combined with pattern recognition analysis of the data might provide an effective approach to the discrimination of Type 2 diabetic patients from healthy controls. And the applications of pattern recognition methods have improved the sensitivity and specificity greatly. (C) 2004 Elsevier B.V. All rights reserved.

Toward the development of an in vitro cultivation of marine sponge cells for sustainable production of bioactive metabolites, the attachment characteristics of marine sponge cells of Hymeniacidon perleve on three types of microcarriers, Hillex, Cytodex 3, and glass beads, were studied. Mixed cell population and enriched cell fractions of specific cell types by Ficoll gradient centrifugation (6%/8%/15%/20%) were also assessed. Cell attachment ratio (defined as the ratio of cells attached on microcarrier to the total number of cells in the culture) on glass beads is much higher than that on Cytodex 3 and Hillex for both mixed cell population and cell fraction at Ficoll 15-20% interface. The highest attachment ratio of 41% was obtained for the cell fraction at Ficoll 15-20% interface on glass beads, which was significantly higher than that of a mixed cell population (18%). The attachment kinetics on glass beads indicated that the attachment was completed within 1 h. Cell attachment ratio decreases with increase in cell-to-microcarrier ratio (3-30 cells/bead) and pH (7.6-9.0). The addition of serum and BSA (bovine serum albumin) reduced the cell attachment on glass beads.

Marine sponges (Porifera) possess an extraordinary diversity of bioactive metabolites for new drug discovery and development. In vitro cultivation of sponge cells in a bioreactor system is very attractive for the sustainable production of sponge-derived bioactive metabolites; however, it is still a challenging task. The recent establishment of sponge primmorphs, multicellular aggregates from dissociated mixed-cell population (MCP), has been widely acknowledged to hold great promise for cultivation in vitro. Here we present a new method to establish an in vitro sponge primmorph culture from archaeocyte-dominant cell population (ADCP) enriched by a Ficoll gradient, rather than a mixed-cell population (MCP). Our rationale is based upon the totipotency (the ability of a cell to differentiate into other cell types) of archaeocyte cells and the different biological functions of various sponge cell types. A sponge, Hymeniacidon perleve collected from the China Yellow Sea was used as a model system for this investigation. Distinct dynamics of primmorph formation were observed while significant increases in DNA synthesis, cell proliferation (up to threefold), and cell growth (up to fourfold) were achieved. Furthermore, a time-dependent spiculogenesis was clearly demonstrated in our longterm culture, indicating high metabolic activity of primmorphs from the ADCP. This new method represents an important step forward to advance sponge cell culture in vitro that may lead to commercial exploitation of sponge-derived drugs. (C) 2003 Wiley Periodicals, Inc.

Plant cell cultures have been suggested as a feasible technology for the production of a myriad of plant-derived metabolites. However, commercial application of plant cell culture has met limited success with only a handful of metabolites produced at the pilot- and commercial-scales. To improve the production of secondary metabolites in plant cell cultures, efforts have been devoted predominantly to the optimization of biosynthetic pathways by both process and genetic engineering approaches. Given that secondary metabolism includes-the synthesis. metabolism and catabolism of endogenous compounds by the specialized proteins, this review intends to draw attention to the manipulation and optimization of post-biosynthetic events that follow the formation of core metabolite structures in biosynthetic pathways. These post-biosynthetic events-the chemical and enzymatic modifications, transport, storage/secretion and catabolism/degradation have been largely unexplored in the past. Potential areas are identified where further research is needed to answer fundamental questions that have implications for advanced bioprocess design. Anthocyanin production by plant cell cultures is used as a case study for this discussion, as it presents a good example of compounds for which there are extensive research publications but still no commercial bioprocess. It is perceived that research on post-biosynthetic processes may lead to future opportunities for significant advances in commercial plant cell cultures. (C) 2002 Elsevier Science Inc. To study the biotransformation of arctigenin, arctigenin was anaerobically incubated with Eubacterium sp. ARC-2 of human intestinal bacteria in vitro. Arctigenin formed a molecular ion [M-H](-) in negative ion mode. The arctigenin and its metabolites were investigated directly by the electrospray ionization tandem mass spectrometry ion trap and Fourier transform ion cyclotron resonance. Arctigenin was transformed to 4',4 ''-dihydroxylenterolactone by E sp. ARC-2 through 3 types of demethylation products.

The toxicological effects of realgar after intragastrical administration (1 g/kg body weight) were investigated over a 21 day period in male Wistar rats using metabonomic analysis of H-1 NMR spectra of urine, serum and liver tissue aqueous extracts. Liver and kidney histopathology examination and serum clinical chemistry analyses were also performed. H-1 NMR spectra and pattern recognition analyses from realgar treated animals showed increased excretion of urinary Kreb's cycle intermediates, increased levels of ketone bodies in urine and serum, and decreased levels of hepatic glucose and glycogen, as well as hypoglycemia and hyperlipoidemia, suggesting the Perturbation of energy metabolism. Elevated levels of choline containing metabolites and betaine in serum and liver tissue aqueous extracts and increased serum creatine indicated altered transmethylation. Decreased urinary levels of trimethylamine-N-oxide, phenylacetylglycine and hippurate suggested the effects on the gut microflora environment by realgar.

The fast analysis of ranitidine is of clinical importance in understanding its efficiency and a patient's treatment history. In this paper, a novel determination method for ranitidine based on capillary electrophoresis-electrochemiluminescence detection is described. The conditions affecting separation and detection were investigated in detail. End-column detection of ranitidine in 5 mM Ru(bpy)(3)(2+) solution at applied voltage of 1.20 V was performed. Favorable ECL intensity with higher column efficiency was achieved by electrokinetic injection for 10 s at 10 kV. The R.S.D. values of ECL intensity and migration time were 6.38 and 1.84% for 10(-4) M and 6.01 and 0.60% for 10(-5) M, respectively. A detection limit of 7 x 10(-8) M (S/N = 3) was achieved. The proposed method was applied satisfactorily to the determination of ranitidine in urine in 6 min.

Metabolic profiles caused by rare earth complex were investigated using NMR and ICP-MS techniques. Male and female Wistar rats were treated orally with Changle (A kind of rare earth complex applied in agriculture to raise the production of crops) at dose of 2, 5 and 20 mg (.) kg(-1) body weight/day respectively for 90 d. Urine and serum samples are collected on 90 d. The relative concentrations of important endogenous metabolites in urine and serum are determined from H-1 NMR spectra and the contents of the four rare earth elements ( La, Ce, Pr and Nd) constituting Changle in the serum samples are measured by ICP-MS technique. Changle-induced renal and liver damage in rats is found based on the increase in the amounts of the amino acids, trimethylamine N-oxide, N, N-dimethyglycine, dimethylamine, succinate, aketoglutarate and ethanol as well as rare earth concentrations. The similarities and differentiations are found in the alteration patterns of metabolites and rare earth concentrations in serum.

High resolution H-1 nuclear magnetic resonance ( NMR) spectroscopy has been employed to assess long-term toxicological effects of ChangLe (a kind of rare earth complex applied in agriculture). Male Wistar rats were administrated orally with ChangLe at doses of 0, 0.1, 0.2, 2.0, 10 and 20 mg/kg body weight daily, respectively, for 6 months. Urine was collected at-day 30, 60, go and serum samples were taken after 6 months. Many low-molecular weight metabolites were identified by H-1 NMR spectra of rat urine. A decrease in citrate and an increase in ketone bodies, creatinine, DMA, DMG, TMAO, and taurine in the urine of the rats. receiving high doses were found by H-1 NMR spectra. These may mean that high-dosage of ChangLe impairs the specific region of liver and kidney, such as renal tubule and mitochondria. The decrease in citrate and the increase in succinate and alpha-ketoglutarate were attributed to a combination of the inhibition of certain citric acid enzymes, renal tubular acidosis and the abnormal fatty acid catabolism. The information of the renal capillary necrosis could be derived from the increase in DMIA, DMG and TMAO. The increase in taurine was due to hepatic mitochondria dysfunction. The conclusions were supported by the results of biochemical measure. merits and enzymatic assay.

A method of capillary HPLC-high-resolution MS was developed for the trace analysis of ATP, GTP, dATP and dGTP Dimethylhexylamine (DMHA) was used as ion-pairing agent for the HPLC retention and separation of the nucleotides and positive ion electrospray time-of-flight MS was used for the detection. The application of capillary HPLC allowed minimal usage of DMHA while providing excellent peak retention and resolution, which significantly reduced the ion suppression in electrospray ionization-MS analysis and thus increased the sensitivity. Adduct ions of nucleotides and DMHA were used as quantitative ions in order to achieve the best sensitivity. DMHA concentration at 5 mM in the aqueous mobile phase at pH 7 was found to be the optimal conditions for the C Is capillary column. The method was applied to determine ATP level in cultured C6 glioma cells that were treated with toxic concentrations of Zn. The results showed that the cellular ATP level decreased from 2.7 pmol/cell (<10% cell death) in average control cell samples to 0.36 pmol/cell as the concentration of Zn increased to 120 mg/l (>35% cell death) in culture medium.

The high-field nuclear magnetic resonance (NMR) spectra can be used for the rapid multicomponent analysis in small amounts of biological fluids. In this paper, the effect of La (NO3)(3) on the rats' metabolism in urine was investigated by H-1 NMR analysis. The experimental groups of wistar rats were injected intraperitoneally with La(NO3)(3) at doses of 0.2, 2.0, 10 and 20mg/kg body weight. The remarkable variation of low molecular weight metabolites in urine has been identified by H-1 NMR spectra, in which dimethylamine, N, N-dimethylglycine, urea, alpha -ketoglutarate, trimethylamine N-oxide, succinate, citrate and amino acids have been suggested as NMR markers for renal damage and ethanol, lactate, taurine as the markers for liver damage. This work may assess its possible use in the early detection of biochemical changes associated with Rare Earth induced kidney and liver dysfunction.

Male Wistar rats were administrated orally with La(NO3)(3) at doses of 0. 05, 0. 2, 2. 0, 10 and 20 mg/kg body weight. Urine was collected over a 24 h period after dosing. Resonances for a large number of low molecular weight metabolites were assigned in a high resolution H-1 NMR spectra of rat urine. The variation of some low molecular weight metabolites in urine provided a sensitive measurement of Rare Earth induced renal and liver lesions, in which DMA, DMG, urea, Kg, TMAO, succinate, citrate and amino acids have been suggested as NMR markers for renal damage and ethanol, lactate, taurine as the markers for liver damage. The method could be applicable to study of the toxicological effects of other compounds and drugs.
