OBJETIVO: Analisar o perfil e a produção científica de pesquisadores da área da Pediatria contemplados com bolsas de produtividade em pesquisa pelo Conselho Nacional de Desenvolvimento Científico e Tecnológico. MÉTODOS: Foram analisados os currículos Lattes de 47 pesquisadores da Pediatria, com bolsas ativas no triênio 2010-2012. As variáveis analisadas foram: gênero, categoria do bolsista, instituição de origem, artigos nacionais e internacionais com o respectivo Qualis, publicações de livros e capítulos de livros, orientações de iniciação científica, mestrado e doutorado e produção de patente. RESULTADOS: Houve predominância do gênero masculino (66%) e de bolsistas na categoria 2 (51%) entre os pesquisadores da área da Pediatria. Os Estados de São Paulo, Rio Grande do Sul e Minas Gerais foram responsáveis por aproximadamente 90% dos pesquisadores. Os bolsistas em Pediatria publicaram no triênio estudado 1.174 artigos, sendo que 274 foram veiculados em periódicos com classificação Qualis B2. CONCLUSÕES: Os pesquisadores na área da Pediatria apresentam produção científica relevante do ponto de vista quantitativo. Entretanto, novos estudos são necessários para dimensionar o impacto do aumento da produção científica na área de Pediatria no País.

O entendimento das necessidades dos clientes e a utilização de tecnologia dos alimentos podem tornar uma ideia em um produto inovador. O objetivo deste trabalho foi estudar, por meio de uma pesquisa de mercado, a oportunidade de um novo produto e, posteriormente, elaborar uma salada de frutas frescas, utilizando técnicas de processamento mínimo com métodos combinados. A pesquisa de mercado foi realizada na região metropolitana de Belo Horizonte - MG para estudar o mercado potencial e avaliar a aceitação de uma salada de frutas pronta e embalada. No processo de desenvolvimento do produto, foram selecionadas três formulações, para avaliação sensorial, avaliadas a cada sete dias, durante 21 dias de armazenamento. Na análise de mercado, o produto salada de frutas foi aprovado, enquanto ideia, sendo considerada como ótima e original. Os clientes potenciais se apresentam como abertos a novidades de mercado e a novas marcas. Considera ainda como atributos de decisão de compra, em ordem de prioridade: preço, referência, necessidade, composição e ingredientes, valores nutricionais. No desenvolvimento da salada de frutas, todos os três tipos foram aceitos em análise sensorial com até 21 dias de armazenamento.

Starting from Evangelista Torricelli's pneumatic discoveries, we try to show that the crucial discoveries in pure science associated with the Scientific Revolution, although they may not have found immediate practical applications, have markedly impelled technological progress in the medium term - contrary to the diffused notion which states that the links between science and technology during the modern period have been tenuous, at best. Torricelli's ideas have turned into one of the privileged research areas for then newborn experimental program, actively participating in a process of reformulation in scientific research's epistemological base, which have culminated in steam power productive application.

Es preciso que Colombia se muestre ante la U.E como un socio estratégico para el desarrollo de sus intereses científicos y tecnológicos, lo cual es posible exhibiendo importantes resultados en el Séptimo Programa Marco (7PM), tanto en el número de proyectos avalados como en la excelencia contenida en los mismos. Esto conducirá a fortalecer la dinámica de colaboración internacional en CyT entre estos dos actores.

Na última década o fortalecimento da produção nacional de insumos em saúde tornou-se uma questão estratégica para o Governo Federal. Paralelamente à indução da pesquisa de novos medicamentos, vacinas e insumos diagnósticos, o Ministério da Saúde (MS), em conjunto com outros organismos governamentais, está investindo no desenvolvimento e na ampliação da pesquisa clínica. A exemplo do que vem sendo proposto e implementado para a área de pesquisa e desenvolvimento de insumos em saúde, uma das estratégias utilizadas é o estímulo à colaboração entre grupos de pesquisa clínica de diferentes instituições. Neste sentido, em 2005, com o apoio do MS se deu a criação da Rede Nacional de Pesquisa Clínica (RNPC). A presente dissertação parte do estudo empírico da Rede Nacional de Pesquisa Clínica, analisando primeiramente a política de editais de financiamento e a formação de grupos de pesquisa para em seguida observar as articulações políticas e técnicas em seu interior através dos relatos de dois pesquisadores envolvidos. O objetivo foi o de verificar se os arranjos constituídos pela RNPC possibilitam a ampla colaboração entre seus participantes por intermédio de recursos de informação e comunicação. A metodologia empregada dividiu-se em duas partes: a primeira destinada ao levantamento de dados da Plataforma Lattes, do Diretório dos Grupos de Pesquisa e das Chamadas Públicas em pesquisa clínica realizadas por CNPq e Finep A segunda parte do estudo ocupou-se em entrevistar dois coordenadores da RNPC de forma a analisar as ações dos pesquisadores, dentro deste arranjo. Os dados obtidos revelam que os editais de fomento lançados não têm atendido plenamente os objetivos desta política de formação da rede. É reconhecida, por parte dos pesquisadores, a colaboração como essencial para o desenvolvimento de estudos clínicos e as TIC\2019s são usadas amplamente nestas interações. Todavia, conclui-se que para que a rede cumpra seus objetivos sejam necessárias novas estratégias de financiamento capazes de estimular a cooperação para fortalecimento da pesquisa clínica, bem como o seguimento de uma política de informação que inclua a construção e implementação de indicadores mais adequados à colaboração em rede e à disseminação de seus resultados

The effects of UVB radiation on the different developmental stages of the carrageenan-producing red alga Iridaea cordata were evaluated considering: (1) carpospore and discoid germling mortality; (2) growth rates and morphology of young tetrasporophytes; and (3) growth rates and pigment content of field-collected plant fragments. Unialgal cultures were submitted to 0.17, 0.5, or 0.83 W m(-2) of UVB radiation for 3 h per day. The general culture conditions were as follows: 12 h light/12 h dark cycles; irradiance of 55 mu mol photon. per square meter per second; temperature of 9 +/- 1 degrees C; and seawater enriched with Provasoli solution. All UVB irradiation treatments were harmful to carpospores (0.17 W m(-2) = 40.9 +/- 6.9%, 0.5 W m(-2) = 59.8 +/- 13.4%, 0.83 W m(-2) = 49 +/- 17.4% mortality in 3 days). Even though the mortality of all discoid germlings exposed to UVB radiation was unchanged when compared to the control, those germlings exposed to 0.5 and 0.83 W m(-2) treatments became paler and had smaller diameters than those cultivated under control treatment. Decreases in growth rates were observed in young tetrasporophytes, mainly in 0.5 and 0.83 W m(-2) treatments. Similar effects were only observed in fragments of adult plants cultivated at 0.83 W m(-2). Additionally, UVB radiation caused morphological changes in fragments of adult plants in the first week, while the young individuals only displayed this pattern during the third week. The verified morphological alterations in I. cordata could be interpreted as a defense against UVB by reducing the area exposed to radiation. However, a high level of radiation appears to produce irreparable damage, especially under long-term exposure. Our results suggest that the sensitivity to ultraviolet radiation decreases with increased algal age and that the various developmental stages have different responses when exposed to the same doses of UVB radiation.

Vegetables are critical for human health as they are a source of multiple vitamins including vitamin E (VTE). In plants, the synthesis of VTE compounds, tocopherol and tocotrienol, derives from precursors of the shikimate and methylerythritol phosphate pathways. Quantitative trait loci (QTL) for alpha-tocopherol content in ripe fruit have previously been determined in an Solanum pennellii tomato introgression line population. In this work, variations of tocopherol isoforms (alpha, beta, gamma, and delta) in ripe fruits of these lines were studied. In parallel all tomato genes structurally associated with VTE biosynthesis were identified and mapped. Previously identified VTE QTL on chromosomes 6 and 9 were confirmed whilst novel ones were identified on chromosomes 7 and 8. Integrated analysis at the metabolic, genetic and genomic levels allowed us to propose 16 candidate loci putatively affecting tocopherol content in tomato. A comparative analysis revealed polymorphisms at nucleotide and amino acid levels between Solanum lycopersicum and S. pennellii candidate alleles. Moreover, evolutionary analyses showed the presence of codons evolving under both neutral and positive selection, which may explain the phenotypic differences between species. These data represent an important step in understanding the genetic determinants of VTE natural variation in tomato fruit and as such in the ability to improve the content of this important nutriceutical.

With the aim of determining the genetic basis of metabolic regulation in tomato fruit, we constructed a detailed physical map of genomic regions spanning previously described metabolic quantitative trait loci of a Solanum pennellii introgression line population. Two genomic libraries from S. pennellii were screened with 104 colocated markers from five selected genomic regions, and a total of 614 bacterial artificial chromosome (BAC)/cosmids were identified as seed clones. Integration of sequence data with the genetic and physical maps of Solanum lycopersicum facilitated the anchoring of 374 of these BAC/cosmid clones. The analysis of this information resulted in a genome-wide map of a nondomesticated plant species and covers 10% of the physical distance of the selected regions corresponding to approximately 1% of the wild tomato genome. Comparative analyses revealed that S. pennellii and domesticated tomato genomes can be considered as largely colinear. A total of 1,238,705 bp from both BAC/cosmid ends and nine large insert clones were sequenced, annotated, and functionally categorized. The sequence data allowed the evaluation of the level of polymorphism between the wild and cultivated tomato species. An exhaustive microsynteny analysis allowed us to estimate the divergence date of S. pennellii and S. lycopersicum at 2.7 million years ago. The combined results serve as a reference for comparative studies both at the macrosyntenic and microsyntenic levels. They also provide a valuable tool for fine-mapping of quantitative trait loci in tomato. Furthermore, they will contribute to a deeper understanding of the regulatory factors underpinning metabolism and hence defining crop chemical composition.

Aims: The ATP-binding cassette transporters, ABCA1 and ABCG1, are LXR-target genes that play an important role in reverse cholesterol transport. We examined the effects of inhibitors of the cholesterol absorption (ezetimibe) and synthesis (statins) on expression of these transporters in HepG2 cells and peripheral blood mononuclear cells (PBMCs) of individuals with primary (and nonfamilial) hypercholesterolemia (HC). Materials & methods: A total of 48 HC individuals were treated with atorvastatin (10 mg/day/4 weeks) and 23 were treated with ezetimibe (10 mg/day/4 weeks), followed by simvastatin (10 mg/day/8 weeks) and simvastatin plus ezetimibe (10 mg of each/day/4 weeks). Gene expression was examined in statin- or ezetimibe-treated and control HepG2 cells as well as PBMCs using real-time PCR. Results: In PBMCs, statins and ezetimibe downregulated ABCA1 and ABCG1 mRNA expression but did not modulate NR1H2 (LxR-beta) and NR1H3 (LXR-alpha) levels. Positive correlations of ABCA1 with ABCG1 and of NR1H2 with NR1H3 expressions were found in all phases of the treatments. In HepG2 cells, ABCA1 mRNA levels remained unaltered while ABCG1 expression was increased by statin (1.0-10.0 mu M) or ezetimibe (5.0 mu M) treatments. Atorvastatin upregulated NR1H2 and NR1H3 only at 10.0 mu M, meanwhile ezetimibe (1.0-5.0 mu M) downregulated NR1H2 but did not change NR1H3 expression. Conclusion: Our findings reveal that lipid-lowering drugs downregulate ABCA1 and ABCG1 mRNA expression in PBMCs of HC individuals and exhibit differential effects on HepG2 cells. Moreover, they indicate that the ABCA1 and ABCG1 transcript levels were not correlated directly to LXR mRNA expression in both cell models treated with lipid-lowering drugs.

Cysteine metabolism exhibits atypical features in Leishmania parasites. The nucleotide sequence annotated as LmjF32.2640 encodes a cysteine desulfhydrase, which specifically catalyzes the breakdown of cysteine into pyruvate, NH(3) and H(2)S. Like in other pathogens, this capacity might be associated with regulatory mechanisms to control the intracellular level of cysteine, a highly toxic albeit essential amino acid, in addition to generate pyruvate for energy production. Besides, our results provide the first insight into the biochemical properties of Leishmania major serine acetyltransferase (SAT), which is likely involved in the two routes for de novo synthesis of cysteine in this pathogen. When compared with other members of SAT family, the N-terminal region of L. major homologue is uniquely extended, and seems to be essential for proper protein folding. Furthermore, unlike plant and bacterial enzymes, the carboxy-terminal-C(10) sequence stretch of L major SAT appears not to be implicated in forming a tight bi-enzyme complex with cysteine synthase. Cell adhesion molecules (CAMs) are surface receptors present in eukaryotic cells that mediate cell-cell or cell-extracellular matrix interactions. Vascular endothelium stimulation in vitro that lead to the upregulation of CAMs was reported for the pathogenic spirochaetes, including rLIC10365 of Leptospira interrogans. In this study, we report the cloning of LIC10507, LIC10508, LIC10509 genes of L interrogans using Escherichia coli as a host system. The rational for selecting these sequences is due to their location in L. interrogans serovar Copenhageni genome that has a potential involvement in pathogenesis. The genes encode for predicted lipoproteins with no assigned functions. The purified recombinant proteins were capable to promote the upregulation of intercellular adhesion molecule 1 (ICAM-1) and E-selectin on monolayers of human umbilical vein endothelial cells (HUVECS). In addition, the coding sequences are expressed in the renal tubules of animal during bacterial experimental infection. The proteins are probably located at the outer membrane of the bacteria since they are detected in detergent-phase of L interrogans Triton X-114 extract. Altogether our data suggest a possible involvement of these proteins during bacterial infection and provide new insights into the role of this region in the pathogenesis of Leptospira. In this paper we describe and evaluate a geometric mass-preserving redistancing procedure for the level set function on general structured grids. The proposed algorithm is adapted from a recent finite element-based method and preserves the mass by means of a localized mass correction. A salient feature of the scheme is the absence of adjustable parameters. The algorithm is tested in two and three spatial dimensions and compared with the widely used partial differential equation (PDE)-based redistancing method using structured Cartesian grids. Through the use of quantitative error measures of interest in level set methods, we show that the overall performance of the proposed geometric procedure is better than PDE-based reinitialization schemes, since it is more robust with comparable accuracy. We also show that the algorithm is well-suited for the highly stretched curvilinear grids used in CFD simulations. Copyright (C) 2010 John Wiley & Sons, Ltd.
