The creation, preservation, and degeneration of cis-regulatory elements controlling developmental gene expression are fundamental genome-level evolutionary processes about which little is known. In this study, critical differences in cis-regulatory elements controlling the expression of the sea urchin aboral ectoderm-specific spec genes were identified and explored. In genomes of species within the Strongylocentrotidae family, multiple copies of a repetitive sequence element termed RSR were present, but RSRs were not detected in genomes of species outside Strongylocentrotidae. RSRs are invariably associated with spec genes, and in Strongylocentrotus purpuratus, the spec2a RSR functioned as a transcriptional enhancer displaying greater activity than RSRs from the spec1 or spec2c paralogs. Single base-pair differences at two cis-regulatory elements within the spec2a RSR greatly increased the binding affinities of four transcription factors: SpCCAAT-binding factor at one element and SpOtx, SpGoosecoid, and SpGATA-E at another. The cis-regulatory elements to which SpCCAAT-binding factor, SpOtx, SpGoosecoid, and SpGATA-E bound were recent evolutionary acquisitions that could act either to activate or repress transcription, depending on the cell type. These elements were found in the spec2a RSR ortholog in Strongylocentrotus pallidus but not in the RSR orthologs of Strongylocentrotus droebachiensis or Hemicentrotus pulcherrimus. These results indicate that spec genes exhibit a dynamic pattern of cis-regulatory element evolution while stabilizing selection preserves their aboral ectoderm expression domain. ^

Heregulins constitute a family of growth factors belonging to the epidermal growth factor (EGF) family. Breast cancers that overexpress specific members of the EGF receptor family (EGFR, ErbB2, ErbB3, ErbB4) have increased metastatic potential, and Heregulin-β1 (HRGβ1), a ligand for ErbB3 and ErbB4, has also been shown to induce metastasis-related properties in breast cancer cells in vitro. The secreted form of the HRGβ1 is composed of five distinct structural domains, including the N-terminal domain, an immunoglobulin-like domain (IgG-like), a glycosylation domain, an EGF-like domain, and a β1-specific domain. Of these, the EGF-like domain is well characterized for its function in metastasis-related properties as well as its structure. However, the contributions of the other HRGβ1 domains in breast cancer metastasis remains unclear. ^ To investigate this, HRGβ1 proteins with targeted domain deletions were purified and subjected to assays for metastasis-related properties, including aggregation, invasion, activation of EGFR family members, and motility of breast cancer cells. These assays showed that retaining the EGF-like domain of HRGβ1 is important for activation of EGFRs. Interestingly, the HRGβ1 protein lacking the IgG-like domain (NGEB) led to a decrease in breast cancer cell motility, indicating the IgG-like domain modulates cell motility, an important step in cancer metastasis. ^ To understand the underlying mechanisms, I performed protein sequence and structural analysis of HRGβ1 and identified that the IgG-like domain of HRGβ1 shares sequence homology and three-dimensional structural similarity with the IgG-like domain of TRIO. TRIO is a cytoplasmic protein that directly associates with RhoA, a GTPase involved in cell reorganization and cell motility. Therefore, I hypothesized that HRGβ1 may translocate inside the breast cancer cells through receptor mediated endocytosis and bind to RhoA via its IgG-like domain. I show wild type HRGβ1 but not NGEB binds RhoA in vitro and in vivo, leading to RhoA activation. Inhibition of HRG-β1 internalization via endocytosis disrupted HRGβ1 binding to RhoA. Additionally, breast cancer cell motility induced by HRG-β1 is reduced after treatment with inhibitors to both endocytosis and RhoA function, similar to levels seen with NGEB treatment. ^ Thus, in addition to the well-known role of HRGβ1 as an extracellular stimulator of the EGFR family members, HRGβ1 also functions within the cell as a binding partner and activator of RhoA to modulate cancer cell motility. ^

A highly parallel and scalable Deblocking Filter (DF) hardware architecture for H.264/AVC and SVC video codecs is presented in this paper. The proposed architecture mainly consists on a coarse grain systolic array obtained by replicating a unique and homogeneous Functional Unit (FU), in which a whole Deblocking-Filter unit is implemented. The proposal is also based on a novel macroblock-level parallelization strategy of the filtering algorithm which improves the final performance by exploiting specific data dependences. This way communication overhead is reduced and a more intensive parallelism in comparison with the existing state-of-the-art solutions is obtained. Furthermore, the architecture is completely flexible, since the level of parallelism can be changed, according to the application requirements. The design has been implemented in a Virtex-5 FPGA, and it allows filtering 4CIF (704 × 576 pixels @30 fps) video sequences in real-time at frequencies lower than 10.16 Mhz.

A generic bio-inspired adaptive architecture for image compression suitable to be implemented in embedded systems is presented. The architecture allows the system to be tuned during its calibration phase. An evolutionary algorithm is responsible of making the system evolve towards the required performance. A prototype has been implemented in a Xilinx Virtex-5 FPGA featuring an adaptive wavelet transform core directed at improving image compression for specific types of images. An Evolution Strategy has been chosen as the search algorithm and its typical genetic operators adapted to allow for a hardware friendly implementation. HW/SW partitioning issues are also considered after a high level description of the algorithm is profiled which validates the proposed resource allocation in the device fabric. To check the robustness of the system and its adaptation capabilities, different types of images have been selected as validation patterns. A direct application of such a system is its deployment in an unknown environment during design time, letting the calibration phase adjust the system parameters so that it performs efcient image compression. Also, this prototype implementation may serve as an accelerator for the automatic design of evolved transform coefficients which are later on synthesized and implemented in a non-adaptive system in the final implementation device, whether it is a HW or SW based computing device. The architecture has been built in a modular way so that it can be easily extended to adapt other types of image processing cores. Details on this pluggable component point of view are also given in the paper.

The art of construction is a risky activity that directly affects the life and physical integrity of persons. Since the approval of Law 31/1995, of November 8, Prevention of Occupational Risks was the first legislation that established the current basis in all sectors and then transposed into Spanish law Directive 92/57/CEE called Royal Decree 1627/1997 of October 24, on minimum safety and health dispositions in construction works, measures have been proposed to develop a mixed body of scientific literature composed of researchers and professionals in the field of occupational safety and health, but even today there is still no clear and firm proposal, showing a lack of awareness in the occupational risk prevention and, therefore, a consolidation of the culture of prevention in society. Therefore, the technicians, who make up the building process, can incur in very high responsibilities, such as: Author of the project, Coordinator of Safety and Health during the preparation of the project and during the execution of works, Site Management: Site Manager. This involves the immediate creation of a general training in prevention for all architects starting when still studying, as well as specific training, appropriate and complementary to all the architects that will be devoted to the specialty of occupational safety and health in construction works. That is, first, we must make the responsible bodies aware of the urgent need to integrate risk prevention in the curricula of architecture and later in the continuing education of the profession. It is necessary that our teaching must conform to the laws on safety and health, due to the fact that the law recognizes our academic degrees and professional qualifications to perform functions in that area

The concepts of holon and holarchy were first applied in the manufacturing world to develop Holonic Manufacturing Systems. Since then, they have been used in many fields and have proved to be applicable concepts for developing applications in any business area. Resulting applications are based on conceptual holonic constructions. Like any model, a holarchy needs to be validated under real circumstances. Such validation assures the quality of the holarchy before it is implemented. In general, validation research tends to target: 1) the specific types of holons handled in each proposal and/or the selected development paradigms; and 2) algorithm performance rather than architecture quality. This paper proposes and evaluates a methodology that focuses on the quality of the architecture. This methodology is able to validate any holonic architecture built to meet trade requirements. Moreover, this is a general-purpose methodology. Therefore, the methodology would be valid for any domain and would not be invalidated by holon types and/or implementation paradigms emerging, changing or falling into disuse. For this purpose, we consider holonic architectures as conceptual models, using the pure holon and holarchy concepts and passing up not only any specific implementation paradigm but also any set of specific holon types.

Current bias estimation algorithms for air traffic control (ATC) surveillance are focused on radar sensors, but the integration of new sensors (especially automatic dependent surveillance-broadcast and wide area multilateration) demands the extension of traditional procedures. This study describes a generic architecture for bias estimation applicable to multisensor multitarget surveillance systems. It consists on first performing bias estimations using measurements from each target, of a subset of sensors, assumed to be reliable, forming track bias estimations. All track bias estimations are combined to obtain, for each of those sensors, the corresponding sensor bias. Then, sensor bias terms are corrected, to subsequently calculate the target or sensor-target pair specific biases. Once these target-specific biases are corrected, the process is repeated recursively for other sets of less reliable sensors, assuming bias corrected measures from previous iterations are unbiased. This study describes the architecture and outlines the methodology for the estimation and the bias estimation design processes. Then the approach is validated through simulation, and compared with previous methods in the literature. Finally, the study describes the application of the methodology to the design of the bias estimation procedures for a modern ATC surveillance application, specifically for off-line assessment of ATC surveillance performance.

We envision that dynamic multiband transmissions taking advantage of the receiver diversity (even for collocated antennas with different polarization or radiation pattern) will create a new paradigm for these links guaranteeing high quality and reliability. However, there are many challenges to face regarding the use of broadband reception where several out of band (with respect to multiband transmission) strong interferers, but still within the acquisition band, may limit dramatically the expected performance. In this paper we address this problem introducing a specific capability of the communication system that is able to mitigate these interferences using analog beamforming principles. Indeed, Higher Order Crossing (HOCs) joint statistics of the Single Input ? Multiple Output (SIMO) system are shown to effectively determine the angle on arrival of the wavefront even operating over highly distorted signals.

This paper proposes a methodology for developing a speech into sign language translation system considering a user-centered strategy. This method-ology consists of four main steps: analysis of technical and user requirements, data collection, technology adaptation to the new domain, and finally, evalua-tion of the system. The two most demanding tasks are the sign generation and the translation rules generation. Many other aspects can be updated automatical-ly from a parallel corpus that includes sentences (in Spanish and LSE: Lengua de Signos Española) related to the application domain. In this paper, we explain how to apply this methodology in order to develop two translation systems in two specific domains: bus transport information and hotel reception.

One medium-term strategy for helping in the management of complexity is the introduction of a conceptual complexity component in the very centre of university curricula. In very few areas is the growth of complexity as evident as in the information technologies (ITs), the focus of the work presented in the current paper. We have therefore developed an integrated way of tackling the specific field of information technologies by means of an approach,to complexity. The content of this paper describes the guidelines of our research effort, placing an emphasis on informatics. Concepts of complexity based on the system metaphor have been substantially drawn upon in this exercise and are thus presented in some detail. Also described is a didactic experiment conducted by the author and designed to provide a new and integrating approach to University curricula for future professionals. The students' "discovery" of complexity is the focal point of the experiment. The findings of this effort are encouraging and call for the continuation and expansion of this experiment.

Software Product Line Engineering has significant advantages in family-based software development. The common and variable structure for all products of a family is defined through a Product-Line Architecture (PLA) that consists of a common set of reusable components and connectors which can be configured to build the different products. The design of PLA requires solutions for capturing such configuration (variability). The Flexible-PLA Model is a solution that supports the specification of external variability of the PLA configuration, as well as internal variability of components. However, a complete support for product-line development requires translating architecture specifications into code. This complex task needs automation to avoid human error. Since Model-Driven Development allows automatic code generation from models, this paper presents a solution to automatically generate AspectJ code from Flexible-PLA models previously configured to derive specific products. This solution is supported by a modeling framework and validated in a software factory.

There is an increasing tendency of turning the current power grid, essentially unaware of variations in electricity demand and scattered energy sources, into something capable of bringing a degree of intelligence by using tools strongly related to information and communication technologies, thus turning into the so-called Smart Grid. In fact, it could be considered that the Smart Grid is an extensive smart system that spreads throughout any area where power is required, providing a significant optimization in energy generation, storage and consumption. However, the information that must be treated to accomplish these tasks is challenging both in terms of complexity (semantic features, distributed systems, suitable hardware) and quantity (consumption data, generation data, forecasting functionalities, service reporting), since the different energy beneficiaries are prone to be heterogeneous, as the nature of their own activities is. This paper presents a proposal on how to deal with these issues by using a semantic middleware architecture that integrates different components focused on specific tasks, and how it is used to handle information at every level and satisfy end user requests.

Business information has become a critical asset for companies and it has even more value when obtained and exploited in real time. This paper analyses how to integrate this information into an existing banking Enterprise Architecture, following an event-driven approach, and entails the study of three main issues: the definition of business events, the specification of a reference architecture, which identifies the specific integration points, and the description of a governance approach to manage the new elements. All the proposed solutions have been validated with a proof-of-concept test bed in an open source environment. It is based on a case study of the banking sector that allows an operational validation to be carried out, as well as ensuring compliance with non-functional requirements. We have focused these requirements on performance.
