Apocynin is the most employed inhibitor of NADPH oxidase (NOX), a multienzymatic complex capable of catalyzing the one-electron reduction of molecular oxygen to the superoxide anion. Despite controversies about its selectivity, apocynin has been used as one of the most promising drugs in experimental models of inflammatory and neurodegenerative diseases. Here, we aimed to study the chemical and biophysical properties of apocynin. The oxidation potential was determined by cyclic voltammetry (Epa = 0.76V), the hydrophobicity index was calculated (logP = 0.83) and the molar absorption coefficient was determined (ε275nm = 1.1 × 104 M-1 cm-1). Apocynin was a weak free radical scavenger (as measured using the DPPH, peroxyl radical and nitric oxide assays) when compared to protocatechuic acid, used here as a reference antioxidant. On the other hand, apocynin was more effective than protocatechuic acid as scavenger of the non-radical species hypochlorous acid. Apocynin reacted promptly with the non-radical reactive species H2O2 only in the presence of peroxidase. This finding is relevant, since it represents a new pathway for depleting H2O2 in cellular experimental models, besides the direct inhibition of NADPH oxidase. This could be relevant for its application as an inhibitor of NOX4, since this isoform produces H 2O2 and not superoxide anion. The binding parameters calculated by fluorescence quenching showed that apocynin binds to human serum albumin (HSA) with a binding affinity of 2.19 × 104 M -1. The association did not alter the secondary and tertiary structure of HSA, as verified by synchronous fluorescence and circular dichroism. The displacement of fluorescent probes suggested that apocynin binds to site I and site II of HSA. Considering the current biomedical applications of this phytochemical, the dissemination of these chemical and biophysical properties can be very helpful for scientists and physicians interested in the use of apocynin.

A new reaction mode of 6,7-bis(methylsulfanyl)-1,4-dihydro-1,4-methanonaphthalene-5,8-dione 1 with the hard nucleophiles sodium benzene- or methane-sulfinate and cyanide, in DMSO, at room temperature, leads to the unexpected hydroquinonoid products 3a-c. All the data are in agreement with a mechanistic pathway involving the initial attack of the hard nucleophile onto the hard carbonyl group, followed by a symbiotic re-attack of the oxygen on the incoming group. In the case of soft nucleophiles, reaction on the olefinic carbon of the enedione system is preferential.

This article presents some results of a study investigating the use of the simulator VLAB, developed by Carnegie Mellon University, to increase the capacity of graduate students in Chemistry in the preparation of experimental procedures. The activity was conducted in two stages, first the students were asked a manuscript describing in detail the procedure and equipment used in the technique of dissolution and delivering it to the teacher. Later in the computer lab, each student performed the same procedure using the simulator. The activity has been proposed to determine if there could be accuracy gains of the description of the process when performed by the simulator. One of the main positive aspects obtained using the simulator students was the recognition of error, important in that the great majority recognized omitted or confused any process step. This reflection showed the students that in addition to testing a procedure using the simulator facilitates the organization of thought and helps arrange it logically. With the use of simulators, students are encouraged to take risks, to explore, to experience a new way, and a real lab usually offers much less room for risk, exploitation and failure.

Climate change and its consequences seem to be increasingly evident in our daily lives. However, is it possible for students to identify a relationship between these large-scale events and the chemistry taught in the classroom? The aim of the present work is to demonstrate that chemistry can assist in elucidating important environmental issues. Simple experiments are used to demonstrate the mechanism of cloud formation, as well as the influence of anthropogenic and natural emissions on the precipitation process. The experiments presented show the way in which particles of soluble salts commonly found in the environment can absorb water in the atmosphere and influence cloud formation.

Silica gel having a particle size between 0.2 and 0.05 mm and a specific surface area, S BET = 473 m 2 g -1, was chemically modified with benzimidazole. Adsorption isotherms of CuX 2 (X = Cl, Br or ClO 4) from ethanol and acetone solutions were studied at 298 K. The metal is bonded to the surface through the free nitrogen atom of the attached benzimidazole. The average number of ligands co-ordinated to the central metal ion was shown to depend on the solid surface loading by the solute. At low loading the electronic and ESR spectral parameters indicated that the copper ion is in a distorted-tetragonal symmetry field.

The history of the quinine synthesis can be used as a case study to emphasize that science is influenced by social and historical processes. The first efforts toward the synthesis of this substance, which until recently was the only treatment for malaria, were by Perkin in 1856 when, trying to obtain quinine,,. he synthesized mauveine. Since then, the quest for the total synthesis of quinine involved several characters in a web of controversies. A major step in this process was made in 1918 by Rabe and Kindler, who proposed the synthesis of quinine from quinotoxine. Twenty-six years later, after obtaining the total synthesis of quinotoxine, Woodward and Doering announced the total synthesis of quinine. However, the lack of experimental details about Rabe and Kindler's process, associated with Woodward and Doering's failure to reproduce it, raised a series of doubts about the synthesis. Stork and colleagues questioned the veracity of the experimental data and even the scientific reputation of the involved researchers. Doubts remained alive until 2008, when Williams and Smith reported, not without reservations, the reproducibility of Rabe and Kindler's protocol. The scientific knowledge as a social and historical development, its legitimating process, and the absence of neutrality in science constitute aspects that can be discussed from this case study, providing significant contributions to science education, in particular, to the initial or continued training of chemistry teachers.

The ligand 1,2-bis(1H-benzimidazol-2-yl)-1,2-ethanediol, 1, and its methylated derivative 2 are readily synthesized from tartaric acid, and act as chiral, facially coordinating tridentate ligands, forming complexes of composition ML2 with octahedral transition metals. The copper(II) complexes show distorted 4 + 2 coordination with benzimidazoles occupying the equatorial sites and alcohol functions weakly binding in the axial sites. Nickel(II) complexes in three different states of protonation show regular octahedral geometry with the alcohols mutually cis. Deprotonation of the coordinated alcohol produces little structural change but the monodeprotonated complex forms a hydrogen bonded dimer. Magnetic measurements show the hydrogen bonded bridge to offer a pathway for weak antiferromagnetic coupling. UV-Visible spectroscopy shows the ligand to have a field intermediate between water and pyridine. The diastereoselectivity of complexation depends on the geometry: nickel(II) shows a weak preference for the homochiral complex, whereas copper(II) forms almost exclusively homochiral complexes.

This paper describes the syntheses and characterization of two new copper(II) diphosphonates: [NH3(CH2)2NH3]2[Cu2(hedp)2]·H2O (1) and [NH3CH(CH3)CH2NH3]2[Cu2(hedp)2] (2) (hedp = 1-hydroxyethylidenediphosphonate). Both compounds exhibit similar one-dimensional linear chain structures. The symmetrical {Cu2(hedp)2} dimers are connected by edge-shared {CuO5} square pyramids and form infinite chains. The Cu(II) ions are alternately bridged by O–P–O groups and O atoms. The Cu–O–Cu angles are 95.8(1) and 96.1(1)° for 1 and 2, respectively. Their magnetic properties show moderately strong antiferromagnetic interactions in both compounds.

One of the minor products from the previously described peripheral -methylation of a magnesium()-20-methyl--pyrrocorphinate is a C-19-methylated 19,20-seco-corphinoid derivative which, on complexation with nickel() acetate, recyclizes to a nickel()-tetradehydro-corrinate.

A sigmatropic methyl shift from the angular position C-1 in ring to the position C-20 between rings and constitutes the crucial step in syntheses leading to a 20-methyl-isobacteriochlorin and to 20-methyl-pyrrocorphins which served as substrates in the investigation presented in the accompanying communication.

After the development of the viral-based prostate cancer vaccine, Ad5-PSA, much research has been orientated to help enhance the induced immune response by combining the vaccine with physical and chemical modulating agents, more specifically the polymers polyethylenimine (PEI), chitosan, and chitosan coated with CD3 complex antibodies; all previously shown to stimulate an immune response as isolated gene carriers. To compare the vaccine-induced immune responses between the naked vaccine and the polymer-vaccine combinations, a mouse model using the ovalbumin- specific Ad-OVA vaccine was tested using intracellular cytokine staining (ICS), tetramer staining, and cytotoxic T-cell lymphocyte assays to measure the activation of CD8+ T-cells, interferon gamma proteins (INFƒ×), and the induced cytotoxicity to ovalbumin. The Ad-OVA vaccine combined with both chitosan and chitosan with CD3 complex antibodies, both natural polymers, were found to induce similar immune responses to the naked vaccine while the vaccine combined with the synthetic polymer, PEI, diminished the immune response.
