Este trabalho tem como tema central autobiografias escritas por indivíduos com autismo. A partir dos enredos que conduzem as narrativas, pretende-se mostrar como elas afetam e contribuem para visões existentes sobre autismo. O traço relevante e singular deste estudo é sua proposta de sublinhar a narrativa em primeira pessoa, as maneiras pelas quais eles se identificam e redefinem as noções existentes sobre autismo como categoria psiquiátrica. O objetivo é compreender as diferentes formas como os autistas se adaptam, negociam, resistem ou até mesmo criam novas normas para lidar com sua condição. Como ferramenta teórica, foram tomadas as ideias do filósofo canadense Ian Hacking, sobre tipos humanos e efeito looping, para analisar como as pessoas classificadas ou autoclassificadas ressignificam, através de suas experiências, as tipificações sobre autismo. As narrativas selecionadas não só permitem que autistas compartilhem suas experiências com o mundo, mas também ampliem os sentidos que atribuímos ao autismo como experiência e como diagnóstico.

Based on numerous pharmacological studies that have revealed an interaction between cannabinoid and opioid systems at the molecular, neurochemical, and behavioral levels, a new series of hybrid molecules has been prepared by coupling the molecular features of two well-known drugs, ie, rimonabant and fentanyl. The new compounds have been tested for their affinity and functionality regarding CB1 and CB2 cannabinoid and mu opioid receptors. In [S-35]-GTP.S (guanosine 5'-O-[gamma-thio] triphosphate) binding assays from the post-mortem human frontal cortex, they proved to be CB1 cannabinoid antagonists and mu opioid antagonists. Interestingly, in vivo, the new compounds exhibited a significant dual antagonist action on the endocannabinoid and opioid systems.

Papillomaviruses (PVs) are widespread pathogens. However, the extent of PV infections in bats remains largely unknown. This work represents the first comprehensive study of PVs in Iberian bats. We identified four novel PVs in the mucosa of free-ranging Eptesicus serotinus (EserPV1, EserPV2, and EserPV3) and Rhinolophus ferrumequinum (RferPV1) individuals and analyzed their phylogenetic relationships within the viral family. We further assessed their prevalence in different populations of E. serotinus and its close relative E. isabellinus. Although it is frequent to read that PVs co-evolve with their host, that PVs are highly species-specific, and that PVs do not usually recombine, our results suggest otherwise. First, strict virus-host co-evolution is rejected by the existence of five, distantly related bat PV lineages and by the lack of congruence between bats and bat PVs phylogenies. Second, the ability of EserPV2 and EserPV3 to infect two different bat species (E. serotinus and E. isabellinus) argues against strict host specificity. Finally, the description of a second noncoding region in the RferPV1 genome reinforces the view of an increased susceptibility to recombination in the E2-L2 genomic region. These findings prompt the question of whether the prevailing paradigms regarding PVs evolution should be reconsidered.

Purpose. To review the proposed pathogenic mechanisms of age macular degeneration (AMD), as well as the role of antioxidants (AOX) and omega-3 fatty acids (omega-3) supplements in AMD prevention. Materials and Methods. Current knowledge on the cellular/molecular mechanisms of AMD and the epidemiologic/experimental studies on the effects of AOX and omega-3 were addressed all together with the scientific evidence and the personal opinion of professionals involved in the Retina Group of the OFTARED (Spain). Results. High dietary intakes of omega-3 and macular pigments lutein/zeaxanthin are associated with lower risk of prevalence and incidence in AMD. The Age-Related Eye Disease study (AREDS) showed a beneficial effect of high doses of vitamins C, E, beta-carotene, and zinc/copper in reducing the rate of progression to advanced AMD in patients with intermediate AMD or with one-sided late AMD. The AREDS-2 study has shown that lutein and zeaxanthin may substitute beta-carotene because of its potential relationship with increased lung cancer incidence. Conclusion. Research has proved that elder people with poor diets, especially with low AOX and omega-3 micronutrients intake and subsequently having low plasmatic levels, are more prone to developing AMD. Micronutrient supplementation enhances antioxidant defense and healthy eyes and might prevent/retard/modify AMD.

The purpose of this study was to compare the effects of Small-Sided Games (SSG) vs. Interval Training (IT) in soccer training on aerobic fitness and physical enjoyment in youth elite soccer players during the last 8 weeks of the season. Seventeen U-16 male soccer players (age = 15.5 +/- 0.6 years, and 8.5 years of experience) of a Spanish First Division club academy were randomized to 2 different groups for 6 weeks: SSG group (n = 9) and IT group (n = 8). In addition to the usual technical and tactical sessions and competitive games, the SSG group performed 11 sessions with different SSGs, whereas the IT group performed the same number of sessions of IT. Players were tested before and after the 6-week training intervention with a continuous maximal multistage running field test and the counter movement jump test (CMJ). At the end of the study, players answered the physical activity enjoyment scale (PACES). During the study, heart rate (HR) and session perceived effort (sRPE) were assessed. SSGs were as effective as IT in maintaining the aerobic fitness in elite young soccer players during the last weeks of the season. Players in the SSG group declared a greater physical enjoyment than IT (P = 0.006; ES = 1.86 +/- 1.07). Coaches could use SSG training during the last weeks of the season as an option without fear of losing aerobic fitness while promoting high physical enjoyment.

Objetiva-se reconstruir o sentido e o alcance do princípio do ne bis in idem, estudando-se as interferências recíprocas do direito penal e do direito administrativo sancionador, com ênfase na concorrência normativa entre tais manifestações do ius puniendi do Estado, seus desdobramentos e os riscos que representam para a liberdade humana, especificamente em face da interdição de duplicidade ou multiplicidade punitiva encartada no princípio em comento. Estrutura-se o texto em três pilares: a primeira parte cuida dos aspectos mais universais do princípio do ne bis in idem, percorrendo tanto seu traçado histórico como seu reconhecimento internacional; a segunda parte examina a consistente experiência jurídica europeia, analisando os marcos teóricos e práticos relacionados à matéria; finalmente, a terceira parte atinge o âmago da investigação, enfocando teoricamente o princípio do ne bis in idem, de modo a renovar sua interpretação no plano nacional, redimensionando as convergências entre o direito penal e o direito administrativo sancionador, a unicidade da (re)ação repressiva do Estado e as possibilidades de enfrentamento das disfunções desse princípio no direito brasileiro. Demonstra-se que a acumulação de sanções de caráter punitivo, de natureza penal e/ou administrativa sancionadora, quando presentes os pressupostos de identidade de sujeito, de fatos e de fundamentos, é vedada pelo espectro de proteção do princípio do ne bis in idem. Postula-se, ainda, esclarecer se, nas situações de exacerbação punitiva com fins semelhantes ou confluentes, deverá sempre prevalecer a aplicação da lei penal. Espera-se, ademais, formular propostas para a regulamentação de conflitos nos casos de concorrência normativa entre o direito penal e o direito administrativo sancionador. Evidencia-se, enfim, que o objetivo principal da investigação é a plena compreensão do princípio do ne bis in idem, refletindo-se a respeito da ilegitimidade da acumulação de sanções penais e sanções administrativas, tão somente pelo fundamento de que não é possível a desvinculação das regras de independência entre a competência jurisdicional e a atribuição sancionadora da administração ou em razão de supostas indiferenças ontológicas entre os ilícitos penal e administrativo.

Lymphangioleiomyomatosis (LAM) is a rare lung-metastasizing neoplasm caused by the proliferation of smooth muscle-like cells that commonly carry loss-of-function mutations in either the tuberous sclerosis complex 1 or 2 (TSC1 or TSC2) genes. While allosteric inhibition of the mechanistic target of rapamycin (mTOR) has shown substantial clinical benefit, complementary therapies are required to improve response and/or to treat specific patients. However, there is a lack of LAM biomarkers that could potentially be used to monitor the disease and to develop other targeted therapies. We hypothesized that the mediators of cancer metastasis to lung, particularly in breast cancer, also play a relevant role in LAM. Analyses across independent breast cancer datasets revealed associations between low TSC1/2 expression, altered mTOR complex 1 (mTORC1) pathway signaling, and metastasis to lung. Subsequently, immunohistochemical analyses of 23 LAM lesions revealed positivity in all cases for the lung metastasis mediators fascin 1 (FSCN1) and inhibitor of DNA binding 1 (ID1). Moreover, assessment of breast cancer stem or luminal progenitor cell biomarkers showed positivity in most LAM tissue for the aldehyde dehydrogenase 1 (ALDH1), integrin-beta 3 (ITGB3/CD61), and/or the sex-determining region Y-box 9 (SOX9) proteins. The immunohistochemical analyses also provided evidence of heterogeneity between and within LAM cases. The analysis of Tsc2-deficient cells revealed relative over-expression of FSCN1 and ID1; however, Tsc2-deficient cells did not show higher sensitivity to ID1-based cancer inhibitors. Collectively, the results of this study reveal novel LAM biomarkers linked to breast cancer metastasis to lung and to cell stemness, which in turn might guide the assessment of additional or complementary therapeutic opportunities for LAM.

Age of onset (AO) of Huntington disease (HD) is mainly determined by the length of the CAG repeat expansion (CAGexp) in exon 1 of the HTT gene. Additional genetic variation has been suggested to contribute to AO, although the mechanism by which it could affect AO is presently unknown. The aim of this study is to explore the contribution of candidate genetic factors to HD AO in order to gain insight into the pathogenic mechanisms underlying this disorder. For that purpose, two AO definitions were used: the earliest age with unequivocal signs of HD (earliest AO or eAO), and the first motor symptoms age (motor AO or mAO). Multiple linear regression analyses were performed between genetic variation within 20 candidate genes and eAO or mAO, using DNA and clinical information of 253 HD patients from REGISTRY project. Gene expression analyses were carried out by RT-qPCR with an independent sample of 35 HD patients from Basque Country Hospitals. We found suggestive association signals between HD eAO and/or mAO and genetic variation within the E2F2, ATF7IP, GRIN2A, GRIN2B, LINC01559, HIP1 and GRIK2 genes. Among them, the most significant was the association between eAO and rs2742976, mapping to the promoter region of E2F2 transcription factor. Furthermore, rs2742976 T allele patient carriers exhibited significantly lower lymphocyte E2F2 gene expression, suggesting a possible implication of E2F2-dependent transcriptional activity in HD pathogenesis. Thus, E2F2 emerges as a new potential HD AO modifier factor.

Este estudo trata do surgimento de inovações no que se refere ao campo das identidades pessoais atreladas ao processo de envelhecimento. Identificamos, na contemporaneidade, o surgimento de condutas, imagens, hábitos e crenças que alteram significativamente as concepções tradicionalmente associadas ao envelhecimento. Adotamos como hipótese o entendimento da terceira idade - termo que vem sendo utilizado para identificar esta inovação - como uma nova identidade etária que vem se somar à infância, adolescência, idade adulta e velhice na composição do curso de vida contemporâneo. A compreensão desta nova identidade etária inclui a delimitação de seu percurso histórico, a sua diferenciação em relação à identidades pessoais vigente na contemporaneidade e a delimitação de suas características específicas. A partir dos indícios oferecidos pelo caso específico da terceira idade, refletimos acerca das tendências de indefinição e fragmentação que, paradoxalmente, atingem o curso de vida contemporâneo e provocam a redefinição e a reorganização das diferentes idades.
