Los glicanos constituyen compuestos formados por residuos de carbohidratos que otorgan propiedades específicas a las macromoléculas a las cuales se unen, permitiendo su normal funcionamiento. La alteración en su formación ocasiona un fenotipo muy variable, de clínica multisistémica y afectación neurológica severa. Los Desórdenes Congénitos de Glicosilación (CDG) son defectos en la síntesis de N- u O-glicanos unidos a proteínas, glicoesfingolípidos o glicofosfatidilinositoles. La caracterización de más de 49 clases de CDG ha contribuido enormemente a comprender mecanismos de glicosilación humanos. El objetivo principal es profundizar en el conocimiento de la glicobiología humana, orientado a responder hipótesis de biología molecular en estas patologías. A partir de la sospecha clínica, por manifestaciones fenotípicas frecuentes (convulsiones, retraso psicomotor, hipoplasia cerebelo, coagulopatías), la metodología diagnóstica se basa en detectar cambios bioquímicos en las glicoproteínas. La más utilizada, es el análisis de transferrina, mediante diferentes metodologías (Western blot, IEF, HPLC y MALDI-TOFF MS). Sin embargo, actualmente existe un número creciente de pacientes con alteraciones de glicosilación aún por dilucidar (CDG-x). Hemos detectado alteraciones de N-glicosilación (n: 7) en pacientes con PMM2-CDG y CDG-IIx y alteraciones de O-glicosilación (n: 41) denominada EXT1/EXT2-CDG u Osteocondromatosis Múltiple (MO). MO está ocasionada por alteración en la biosíntesis del heparán sulfato proteoglicano, debido a mutaciones en los genes que codifican glicosiltranferasas responsables de su elongación. Nuestro programa interdisciplinario de CDG en Argentina, se espera contribuya a la formación y consolidación de la red “CDGnet América Latina” fomentando el intercambio científico, las capacidades tecnológicas y el conocimiento de los últimos avances en Desórdenes Congénitos de Glicosilación.

Alzheimer ist eine neurodegenerative Erkrankung deren molekulare Ursache, die Anhäufung von Amyloid-Plaques im Gehirn und die Bildung von Neurofibrillen in den Nervenzellen ist. Grund für diese molekularen Ursachen ist das Peptid Amylois beta, welches sich im Gehirn von Alzheimer Patienten anhäuft und zu Fibrillen aggregiert. In der Arbeit wurde untersucht, ob es Unterschiede bezüglich des proteolytischen Abbaus zwischen Abetafibrillen 1-40, 3-40 und pE3-40 gibt und ob die Fibrillen von unterschiedlichen Enzymen abgebaut werden können. Zudem wurden die Spaltprodukte, die bei diesem Abbau entstanden, miteinander verglichen und analysiert. Es wurde also untersucht, ob die Enzyme eine abbauende Wirkung auf aggregierte Aβ-Peptide haben und wie sich diese im Vergleich zu den gelösten Abeta-Peptiden verhalten. Die Untersuchungen fanden mit den Enzymen Neprilysin, Neprilysin 2, Insulysin, Matrix-Metalloproteinasen 2, 3, 9, 13, Plasmin, Plasma Kallikrein, Tissue Kallikrein und Meprin A statt und wurden Mittels MALDO-TOF analysiert.

BACKGROUND: The purpose of this prospective study was to perform a head-to-head comparison of the two methods most frequently used for evaluation of carotid plaque characteristics: Multi-detector Computed Tomography Angiography (MDCTA) and black-blood 3 T-cardiovascular magnetic resonance (bb-CMR) with respect to their ability to identify symptomatic carotid plaques. METHODS: 22 stroke unit patients with unilateral symptomatic carotid disease and >50% stenosis by duplex ultrasound underwent MDCTA and bb-CMR (TOF, pre- and post-contrast fsT1w-, and fsT2w- sequences) within 15 days of symptom onset. Both symptomatic and contralateral asymptomatic sides were evaluated. By bb-CMR, plaque morphology, composition and prevalence of complicated AHA type VI lesions (AHA-LT6) were evaluated. By MDCTA, plaque type (non-calcified, mixed, calcified), plaque density in HU and presence of ulceration and/or thrombus were evaluated. Sensitivity (SE), specificity (SP), positive and negative predictive value (PPV, NPV) were calculated using a 2-by-2-table. RESULTS: To distinguish between symptomatic and asymptomatic plaques AHA-LT6 was the best CMR variable and presence / absence of plaque ulceration was the best CT variable, resulting in a SE, SP, PPV and NPV of 80%, 80%, 80% and 80% for AHA-LT6 as assessed by bb-CMR and 40%, 95%, 89% and 61% for plaque ulceration as assessed by MDCTA. The combined SE, SP, PPV and NPV of bb-CMR and MDCTA was 85%, 75%, 77% and 83%, respectively. CONCLUSIONS: Bb-CMR is superior to MDCTA at identifying symptomatic carotid plaques, while MDCTA offers high specificity at the cost of low sensitivity. Results were only slightly improved over bb-CMR alone when combining both techniques.

La insuficiencia pulmonar después de la reparación de la Tetralogía de Fallot ocasiona una dilatación del ventrículo derecho, IT y/o empeoramiento de la CF. El momento de la cirugía viene marcado por la presencia de la clínica y/o dilatación ventrículo derecho. El motivo del presente estudio es realizar una revisión de los pacientes sometidos a sustitución valvular pulmonar, un seguimiento clínico y ecocardiográfico. MÉTODOS Desde enero 2003 a enero 2009, 33 pacientes fueron sometidos a una cirugía de sustitución valvular pulmonar. El 41 % de los pacientes fueron mujeres y el 59 % varones. La edad media de los pacientes fue de 31,3 años, ( 18 - 63 a ). La edad media de la primera intervención fue a los 3,8 años ( 6 meses – 28 años ). Sólo un paciente no había sido sometido a ningún tipo de intervención en la infancia, realizándose una cirugía paliativa – correctora a los 28 años. La indicación de intervención quirúrgica vino marcada por la presencia de clínica en 11 pacientes ( 37 %) y por dilatación VD, ( única o asintomática) en 18 pacientes ( 62%). RESULTADOS No hubo mortalidad operatoria con una estancia media post IQ de 15,21 días ( 9- 27 días). Se analizan los resultados quirúrgicos de estos pacientes a tres niveles: Clínica y tolerancia al esfuerzo en postoperatorio inmediato y tardío. Eventos arrítmicos en el postoperatorio inmediato y seguimiento y Parámetros ecocardiográficos en postoperatorio inmediato y tardío. (Dimensiones cavidades derechas y función contráctil del VD ( TAPSE )).En el seguimiento al año de la intervención ningún paciente presentaba clínica de IP. El 67 % de los pacientes fueron intervenidos manteniéndose asintomáticos con un 37% ( 11 pacientes ) de eventos arrítmicos pre cirugía ( 9 ; 31 % pacientes) fueron sometidos a ablación precirugía y 3 pacientes ( 10 %) requirieron implante de un dispositivo DAI. Después de la cirugía el 86 % de los pacientes, 24 pacientes, se mantenían en CF I; 3 pacientes ( 10 %) continuaron presentando eventos arrítmicos y un solo paciente requirió implante de DAI. El remodelado del VD al año y medio de seguimiento presentó una reducción del 11 % respecto al diámetro teledistólico precirugía ( DTDV x 54,43 ( 41-70 mm), postcirugía ( DTDV x 44,29 ( 32-61), p ≤ 0,01; sin encontrar diferencias significativas en la reducción del dTS pre/postcirugía. La función del VD ( TAPSE pre IQ 16,35 ( 13-229;postcirugía inmediata 15,54 ( 11-23) y al año 17,5. El gradiente medio es inferior al 15 mmHg en el 84 % de los pacientes portadores de válvula biológica. 11 pacientes ≤ 11 mmHg ( 44 %); 10 pacientes ≤ 15 mm Hg ( 40 %). CONCLUSIONES En este estudio realizado en el Hospital Vall d’Hebron, la sustitución valvular pulmonar en pacientes afectos de una Tetralogía de Fallot mejora los diámetros ventriculares, la función contráctil y por lo tanto la capacidad funcional de los mismos. En los pacientes sometidos a Estudio electrofisiológicos que no presentaron inducción de eventos arrítmicos ventriculares, no han presentado episodios arrítmicos tras la cirugía de sustitución valvular pulmonar. Todos los pacientes se mantienen en Clase funcional I tras la cirugía y libres de reintervención con un seguimiento medio de 16,9 ( 5-33 ) meses. Los factores de riesgo para presentar una peor evolución son un retraso en la corrección inicial de la TOF y por lo tanto aparición de enfermedad pulmonar por hipertensión vascular subyacente, edad avanzada en el momento del PVR, deterioro funcional preoperatorio con Clase Funcional según NYHA ( III- IV), o bien aparición de eventos arrítmicos.

The response of Arabidopsis to stress caused by mechanical wounding was chosen as a model to compare the performances of high resolution quadrupole-time-of-flight (Q-TOF) and single stage Orbitrap (Exactive Plus) mass spectrometers in untargeted metabolomics. Both instruments were coupled to ultra-high pressure liquid chromatography (UHPLC) systems set under identical conditions. The experiment was divided in two steps: the first analyses involved sixteen unwounded plants, half of which were spiked with pure standards that are not present in Arabidopsis. The second analyses compared the metabolomes of mechanically wounded plants to unwounded plants. Data from both systems were extracted using the same feature detection software and submitted to unsupervised and supervised multivariate analysis methods. Both mass spectrometers were compared in terms of number and identity of detected features, capacity to discriminate between samples, repeatability and sensitivity. Although analytical variability was lower for the UHPLC-Q-TOF, generally the results for the two detectors were quite similar, both of them proving to be highly efficient at detecting even subtle differences between plant groups. Overall, sensitivity was found to be comparable, although the Exactive Plus Orbitrap provided slightly lower detection limits for specific compounds. Finally, to evaluate the potential of the two mass spectrometers for the identification of unknown markers, mass and spectral accuracies were calculated on selected identified compounds. While both instruments showed excellent mass accuracy (<2.5ppm for all measured compounds), better spectral accuracy was recorded on the Q-TOF. Taken together, our results demonstrate that comparable performances can be obtained at acquisition frequencies compatible with UHPLC on Q-TOF and Exactive Plus MS, which may thus be equivalently used for plant metabolomics.

AIM: Antidoping procedures are expected to greatly benefit from untargeted metabolomic approaches through the discovery of new biomarkers of prohibited substances abuse. RESULTS: Endogenous steroid metabolites were monitored in urine samples from a controlled elimination study of testosterone undecanoate after ingestion. A platform coupling ultra-high pressure LC with high-resolution quadrupole TOF MS was used and high between-subject metabolic variability was successfully handled using a multiblock data analysis strategy. Links between specific subsets of metabolites and influential genetic polymorphisms of the UGT2B17 enzyme were highlighted. CONCLUSION: This exploratory metabolomic strategy constitutes a first step toward a better understanding of the underlying patterns driving the high interindividual variability of steroid metabolism. Promising biomarkers were selected for further targeted study.

Capillary zone electrophoresis (CZE) with UV detection has been widely used for the determination of carbohydrate-deficient transferrin (CDT), an indirect marker of the chronic alcohol consumption (≥60-80g/day). A commercially available method (CEofix? CDT kit), containing a bilayer anionic coating, allows for the analysis of CDT with a high resolution between transferrin (Tf) glycoforms with reduced protein adsorption onto the capillary wall. Although widely used in routine analysis, this procedure presents some limitations in terms of selectivity and sensitivity which may be overcome with mass spectrometry (MS). However, the available method is not MS-compatible due to the non-volatile coating as well as the phosphate and borate buffers present in the background electrolyte (BGE). This study firstly consisted in developing MS-compatible separation conditions, i.e., coating and BGE compositions. Numerous cationic, neutral, and anionic coatings were evaluated in combination with BGEs covering a broad range of pH values. A bilayer coating composed of a cationic layer of 10% polybrene (m/v) and an anionic layer of 10% dextran sulfate (m/v) combined with a BGE composed of 20mM ammonium acetate at pH 8.5 provided the best results in terms of glycoforms' resolution, efficiency, adsorption reduction, migration times' repeatability, and coating stability. The method was then transferred to CZE-MS after investigations of the electrospray ionization (ESI) source, equipped with a sheath-flow interface, and the time-of-flight (TOF/MS) parameters. A successful MS detection of tetrasialo-Tf was obtained during infusion, while the experiments highlighted the challenges and issues encountered with intact glycoprotein analysis by CZE-ESI-MS.

An impaired glutathione (GSH) synthesis was observed in several multifactorial diseases, including schizophrenia and myocardial infarction. Genetic studies revealed an association between schizophrenia and a GAG trinucleotide repeat (TNR) polymorphism in the catalytic subunit (GCLC) of the glutamate cysteine ligase (GCL). Disease-associated genotypes of this polymorphism correlated with a decrease in GCLC protein expression, GCL activity and GSH content. To clarify consequences of a decreased GCL activity at the proteome level, three schizophrenia patients and three controls have been selected based on the GCLC GAG TNR polymorphism. Fibroblast cultures were obtained by skin biopsy and were challenged with tert-butylhydroquinone (t-BHQ), a substance known to induce oxidative stress. Proteome changes were analyzed by two dimensional gel electrophoresis (2-DE) and results revealed 10 spots that were upregulated in patients following t-BHQ treatment, but not in controls. Nine corresponding proteins could be identified by MALDI mass spectrometry and these proteins are involved in various cellular functions, including energy metabolism, oxidative stress response, and cytoskeletal reorganization. In conclusion, skin fibroblasts of subjects with an impaired GSH synthesis showed an altered proteome reaction in response to oxidative stress. Furthermore, the study corroborates the use of fibroblasts as an additional mean to study vulnerability factors of psychiatric diseases.

En aquest treball s’investiga la reactivitat de tres complexos de Pt, cisplatí, complex 1, [Pt(dmba)(aza-N1)(dmso)], complex 2, i [Pt(dmba)(N9-9AA)(PPh3)]+, complex 3; els quals presenten activitat antitumoral, amb diferents proteïnes i oligonucleòtids mitjançant espectrometria de masses (ESI-TOF MS). ). L’estudi del cisplatí, 1, droga molt coneguda i emprada avui en dia pel tractament de molts tipus de càncer, permet comparar la reactivitat dels nous complexos d’estudi, 2 i 3, els quals presenten un IC50 més baix que el cisplatí en algunes línies de cèl·lules tumorals.

We perform a structural and optical characterization of InAs1¿xNx epilayers grown by molecular beam epitaxy on InAs substrates x 2.2% . High-resolution x-ray diffraction HRXRD is used to obtain information about the crystal quality and the strain state of the samples and to determine the N content of the films. The composition of two of the samples investigated is also obtained with time-of-flight secondary ion mass spectroscopy ToF-SIMS measurements. The combined analysis of the HRXRD and ToF-SIMS data suggests that the lattice parameter of InAsN might significantly deviate from Vegard"s law. Raman scattering and far-infrared reflectivity measurements have been carried out to investigate the incorporation of N into the InAsN alloy. N-related local vibrational modes are detected in the samples with higher N content. The origin of the observed features is discussed. We study the compositional dependence of the room-temperature band gap energy of the InAsN alloy. For this purpose, photoluminescence and optical absorption measurements are presented. The results are analyzed in terms of the band-anticrossing BAC model. We find that the room-temperature coupling parameter for InAsN within the BAC model is CNM=2.0 0.1 eV.

Millions of blood products are transfused every year; many lives are thus directly concerned by transfusion. The three main labile blood products used in transfusion are erythrocyte concentrates, platelet concentrates and fresh frozen plasma. Each of these products has to be stored according to its particular components. However, during storage, modifications or degradation of those components may occur, and are known as storage lesions. Thus, biomarker discovery of in vivo blood aging as well as in vitro labile blood products storage lesions is of high interest for the transfusion medicine community. Pre-analytical issues are of major importance in analyzing the various blood products during storage conditions as well as according to various protocols that are currently used in blood banks for their preparations. This paper will review key elements that have to be taken into account in the context of proteomic-based biomarker discovery applied to blood banking.

Two methods of differential isotopic coding of carboxylic groups have been developed to date. The first approach uses d0- or d3-methanol to convert carboxyl groups into the corresponding methyl esters. The second relies on the incorporation of two 18O atoms into the C-terminal carboxylic group during tryptic digestion of proteins in H(2)18O. However, both methods have limitations such as chromatographic separation of 1H and 2H derivatives or overlap of isotopic distributions of light and heavy forms due to small mass shifts. Here we present a new tagging approach based on the specific incorporation of sulfanilic acid into carboxylic groups. The reagent was synthesized in a heavy form (13C phenyl ring), showing no chromatographic shift and an optimal isotopic separation with a 6 Da mass shift. Moreover, sulfanilic acid allows for simplified fragmentation in matrix-assisted laser desorption/ionization (MALDI) due the charge fixation of the sulfonate group at the C-terminus of the peptide. The derivatization is simple, specific and minimizes the number of sample treatment steps that can strongly alter the sample composition. The quantification is reproducible within an order of magnitude and can be analyzed either by electrospray ionization (ESI) or MALDI. Finally, the method is able to specifically identify the C-terminal peptide of a protein by using GluC as the proteolytic enzyme.

Expression of water soluble proteins of fresh pork Longissimus thoracis from 4 pure breed pigs (Duroc, Large White, Landrace, and Piétrain) was studied to identify candidate protein markers for meat quality. Surface-enhanced laser desorption/ionisation time-of-flight mass spectrometry (SELDI-TOF-MS) was used to obtain the soluble protein profiles of Longissimus thoracis muscles. The pure breeds showed differences among the studied meat quality traits (pHu, drip loss, androstenone, marbling, intramuscular fat, texture, and moisture), but no significant differences were detected in sensory analysis. Associations between protein peaks obtained with SELDI-TOF-MS and meat quality traits, mainly water holding capacity, texture and skatole were observed. Of these peaks, a total of 10 peaks from CM10 array and 6 peaks from Q10 array were candidate soluble protein markers for pork loin quality. The developed models explained a limited proportion of the variability, however they point out interesting relationships between protein expression and meat quality

The breccia-hosted epithermal Au-Ag deposit of Rosia Montana is located 7 kin northeast of Abrud, in the northern part of the South Apuseni Mountains, Romania. Estimated total reserves of 214.91 million metric toils (Mt) of ore at 1.46 g/t An and 6.9 g/t Ag (10.1 Moz of An and 47.6 Moz of Ag) make Rosia Montana one of the largest gold deposits in Europe. At this location, Miocene calc-alkaline magmatic and hydrothermal activity was associated with local extensional tectonics within a strike-slip regime related to the indentation of the Adriatic microplate into the European plate during the Carpathian orogenesis. The host rocks of the magmatic complex consist of pre-Mesozoic metamorphosed continental crust covered by Cretaceous turbiditic sediment (flysch). Magmatic activity at Rosia Montana and its surroundings occurred in several pulses and lasted about 7 m.y, Rosia Montana is a breccia-hosted epithermal system related to strong phreatomagmatic activity due to the shallow emplacement of the Montana dacite. The Montana dacite intruded Miocene volcaniclastic material (volcaniclastic breccias) and crops out at Cetate and Carnic Hills. Current mining is focused primarily on the Cetate open pit, which was mapped in detail, leading to the recognition of three distinct breccia bodies: the dacite breccia with a dominantly hydrothermal matrix, the gray polymict breccia with a greater proportion of sand-sized matrix support, and the black polymict breccia, which reached to the surface, contains carbonized tree trunks and has a dominantly barren elastic matrix. The hydrothermal alteration is pervasive. Adularia alteration with a phyllic overprint is ubiquitous; silicification and argillic alteration occur locally. Mineralization consists of quartz, adularia, carbonates (commonly Mn-rich), pyrite, Fe-poor sphalerite, galena, chalcopyrite, tetrahedrite, and native gold and occurs as disseminations, as well as in veins and filling vugs within the Montana dacite and the different breccias. The age of mineralization (12.85 +/- 0.07 Ma) was determined by Ar-40- Ar-39 dating on hydrothermal adularia crystals from vugs in the dacite breccia in the Cetate open pit. Microthermometric measurements of fluid inclusions in quartz phenocrysts from the Montana dacite revealed two fluid types that are absent from the hydrothermal breccia and must have been trapped at depth prior to dacite dome emplacement: brine inclusions (32-55 -wt % NaCl equiv, homogenizing at T-h > 460 degrees C) and intermediate density fluids (4.9-15.6 wt % NaCl equiv, T, between 345 degrees-430 degrees C). Secondary aqueous fluid inclusion assemblages in the phenocrysts have salinities of 0.2 to 2.2 wt percent NaCl equiv and T-h of 200 degrees to 280 degrees C. Fluid inclusion assemblages in hydrothermal quartz from breccias and veins have salinities of 0.2 to 3.4 wt percent NaCl equiv and T-h, from 200 degrees to 270 degrees C. The oxygen isotope composition of several zones of an ore-related epithermal quartz crystal indicate a very constant delta O-18 of 4.5 to 5.0 per mil for the mineralizing fluid, despite significant salinity and temperature variation over time. Following microthermometry, selected fluid inclusion assemblages were analyzed by laser ablation-inductively coupled-plasma mass spectrometry (LA-ICMS). Despite systematic differences in salinity between phenocryst-hosted fluids trapped at depth and fluids from quartz in the epithermal breccias, all fluids have overlapping major and trace cation ratios, including identical Na/K/Rb/Sr/Cs/Ba. Consistent with the constant near-magmatic oxygen isotope composition of the hydrothermal fluids, these data strongly indicate a common magmatic component of these chemically conservative solutes in all fluids. Cu, Pb, Zn, and Mn show variations in concentration relative to the relatively non-reactive alkalis, reflecting the precipitation of sulfide minerals together with An in the epithermal breccia, and possibly of Cu in an inferred subjacent porphyry environment. The magmatic-hydrothermal processes responsible for epithermal Au-Ag mineralization at Rosia Montana are, however, not directly related to the formation of the spatially associated porphyry Cu-Au deposit of Rosia Poieni, which occurred lout 3 m.y. later.

PURPOSE: To prospectively compare various parameters of vessels imaged at 3 T by using time-of-flight (TOF) and T2-prepared magnetic resonance (MR) angiography in a rabbit model of hind limb ischemia. MATERIALS AND METHODS: Experiments were approved by the institutional animal care and use committee. Endovascular occlusion of the left superficial femoral artery was induced in 14 New Zealand white rabbits. After 2 weeks, MR angiography and conventional (x-ray) angiography were performed. Vessel sharpness was evaluated visually in the ischemic and nonischemic limbs, and the presence of small collateral vessels was evaluated in the ischemic limbs. Vessel sharpness was also quantified by evaluating the magnitude of signal intensity change at the vessel borders. RESULTS: The sharpness of vessels in the nonischemic limbs was similar between the TOF and the T2-prepared images. In the ischemic limbs, however, T2-prepared imaging, as compared with TOF imaging, generated higher vessel sharpness in arteries with diminished blood flow (mean vessel sharpness: 44% vs 30% for popliteal arteries, 45% vs 28% for saphenous arteries; P < .001 for both comparisons) and enabled better detection of small collateral vessels (93% vs 36% of vessels, P < .001). CONCLUSION: T2-prepared imaging can facilitate high-spatial-resolution MR angiography of small vessels with low blood flow and thus has potential as a tool for noninvasive evaluation of arteriogenic therapies, without use of contrast material.
