O presente relatório retrata e analisa o trabalho desenvolvido no âmbito da unidade curricular de Prática de Ensino Supervisionada (PES), do mestrado em Educação PréEscolar e Ensino do 1.º Ciclo do Ensino Básico da Escola Superior de Educação do Instituto Politécnico de Bragança. A prática pedagógica foi desenvolvida em dois contextos da cidade de Bragança, sendo numa primeira fase num Jardim de Infância de uma Instituição Particular de Solidariedade Social (IPSS), numa sala com catorze crianças de três e quatro anos de idade e, numa segunda fase, numa escola do 1.º Ciclo do Ensino Básico numa Instituição da rede pública, numa sala de 3.º ano com dezanove crianças de oito anos de idade. O principal objetivo deste relatório é dar a conhecer o trabalho realizado em ambos os contextos, descrevendo e refletindo sobre a prática realizada. Ao longo da PES procurámos desenvolver atividades que respondessem às necessidades e interesses das crianças, de forma a criar momentos de participação ativa, de partilha de saberes e de cooperação no âmbito das Orientações Curriculares para a Educação Pré-escolar e do Programa Nacional do 3.º ano do 1.º Ciclo do Ensino Básico. Era nossa intenção mostrar como o livro para a infância pode ser utilizado como promotor de experiências de ensino/aprendizagem ricas e diversificadas, tendo sempre como base a leitura. Nesta perspetiva optámos pela questão-problema: Como podemos fomentar nas crianças o gosto pela leitura e pelo livro? Neste âmbito formulámos os seguintes objetivos: (i) Promover o gosto pela leitura; (ii) Despertar o interesse dos alunos pela leitura literária, recorrendo à utilização do fantástico e do maravilhoso para a estimulação do imaginário na infância; (iii) Envolver a família na promoção leitora; (iv) Proporcionar um conjunto de atividades de forma a trabalhar três momentos fulcrais para a leitura: antes da leitura (pré-leitura), no decorrer da leitura (leitura) e no fim da leitura (pós leitura). A metodologia utilizada foi a investigação qualitativa de natureza interpretativa, baseando-nos na utilização de instrumentos de recolha de dados do tipo qualitativo, como a observação, as notas de campo e fotografias. Este tipo de investigação apresenta os resultados através de narrativas com descrições contextuais e citações dos participantes em experiências de ensino/aprendizagem significativas que estimularam o gosto pela leitura e pelo livro das crianças de ambos os contextos.

Relatório EPE - Relatório de estágio em Educação Pré-Escolar: O presente relatório de estágio em educação pré-escolar tem como intuito apresentar a descrição e a análise do percurso formativo da mestranda ao longo da prática pedagógica. O relatório integra os referenciais teóricos e legais adotados pela mesma, através de uma descrição reflexiva acerca da sua prática educativa, que lhe conferiu o desenvolvimento de competências e saberes profissionais. Neste sentido, o relatório irá abordar as conceções, os processos e os resultados obtidos pela mestranda ao longo da sua ação, onde o processo de ensino e de aprendizagem procurou focar-se no desenvolvimento de competências e capacidades das crianças, de acordo com uma perspetiva socioconstrutivista, suportada em vários modelos curriculares. Cabe ao educador proporcionar aprendizagens significativas às crianças com as quais desenvolve a sua prática devendo, por isso, intervir de forma fundamentada e em conformidade com as necessidades e interesses do grupo, através de uma postura observadora, dinâmica, flexível e proactiva adaptando as suas ações e reajustando as suas estratégias pedagógicas diferenciadas. Deste modo, a formanda procurou ser interveniente na construção de saberes pedagógicos, com uma postura investigadora da sua prática, sendo clarificado, ao longo de todo o documento, todo o seu percurso com o intuito de conhecer e compreender as competências profissionais desenvolvidas. Estando apresentadas as suas aprendizagens, bem como as suas reflexões, que potenciaram a evolução da formanda tanto a nível pessoal como profissional.

One of the challenges to biomedical engineers proposed by researchers in neuroscience is brain machine interaction. The nervous system communicates by interpreting electrochemical signals, and implantable circuits make decisions in order to interact with the biological environment. It is well known that Parkinson’s disease is related to a deficit of dopamine (DA). Different methods has been employed to control dopamine concentration like magnetic or electrical stimulators or drugs. In this work was automatically controlled the neurotransmitter concentration since this is not currently employed. To do that, four systems were designed and developed: deep brain stimulation (DBS), transmagnetic stimulation (TMS), Infusion Pump Control (IPC) for drug delivery, and fast scan cyclic voltammetry (FSCV) (sensing circuits which detect varying concentrations of neurotransmitters like dopamine caused by these stimulations). Some softwares also were developed for data display and analysis in synchronously with current events in the experiments. This allowed the use of infusion pumps and their flexibility is such that DBS or TMS can be used in single mode and other stimulation techniques and combinations like lights, sounds, etc. The developed system allows to control automatically the concentration of DA. The resolution of the system is around 0.4 µmol/L with time correction of concentration adjustable between 1 and 90 seconds. The system allows controlling DA concentrations between 1 and 10 µmol/L, with an error about +/- 0.8 µmol/L. Although designed to control DA concentration, the system can be used to control, the concentration of other substances. It is proposed to continue the closed loop development with FSCV and DBS (or TMS, or infusion) using parkinsonian animals models.

Seeds from legumes including the Glycine max are known to be a rich source of protease inhibitors. The soybean Kunitz trypsin inhibitor (SKTI) has been well characterised and has been found to exhibit many biological activities. However its effects on inflammatory diseases have not been studied to date. In this study, SKTI was purified from a commercial soy fraction, enriched with this inhibitor, using anion exchange chromatography Resource Q column. The purified protein was able to inhibit human neutrophil elastase (HNE) and bovine trypsin. . Purified SKTI inhibited HNE with an IC50 value of 8 µg (0.3 nM). At this concentration SKTI showed neither cytotoxic nor haemolytic effects on human blood cell populations. SKTI showed no deleterious effects on organs, blood cells or the hepatic enzymes alanine amine transferase (ALT) and aspartate amino transferase (AST) in mice model of acute systemic toxicity. Human neutrophils incubated with SKTI released less HNE than control neutrophils when stimulated with PAF or fMLP (83.1% and 70% respectively). These results showed that SKTI affected both pathways of elastase release by PAF and fMLP stimuli, suggesting that SKTI is an antagonist of PAF/fMLP receptors. In an in vivo mouse model of acute lung injury, induced by LPS from E. coli, SKTI significantly suppressed the inflammatory effects caused by elastase in a dose dependent manner. Histological sections stained by hematoxylin/eosin confirmed this reduction in inflammation process. These results showed that SKTI could be used as a potential pharmacological agent for the therapy of many inflammatory diseases

Processed meat products are of worldwide importance and, because of their intrinsic factors as well as the processing methods, they are highly prone to fungal and mycotoxin contamination. Ochratoxin A (OTA) is the most significant mycotoxin in processed meat products. Penicillium nordicum is considered to be responsible for OTA contamination of meat products, as it is highly adapted to salt and protein-rich matrices and is moderately psycrotrophic. However, another OTA-producing fungus, Aspergillus westerdijkiae, adapted to carbon-rich matrices such as cereals and coffee beans, has been recently associated with high levels of OTA in meat products. Several Lactic Acid Bacteria (LAB) and yeasts have been tested as biocontrol agents against P. nordicum growth and OTA production in meat products, with promising results, but none of the studies have considered A. westerdijkiae. The aim of this work was to evaluate in vitro the effect of a commercial starter culture used in sausage fermentation and four yeasts isolated from dry-cured sausage on these two OTA-producing fungi, both in terms of fungal growth and of OTA production, using different meat-based culture media as model systems. The mechanisms underlying the observed effect were also studied. For this purpose, C. krusei, C. zeylanoides, R. mucilaginosa, R. glutinis, a mix of these yeasts and the starter culture were co-inoculated with P. nordicum and A. westerdijkiae in industrial sausage, traditional sausage, and ham-based media, under conditions of water activity, salt concentration and temperature that mimic real conditions at beginning and end of sausage curing process. Fungal growth was determined by measuring colony diameter, and OTA production was quantified by HPLC-FLD after extraction with methanol. Yeasts where found to inhibit significantly the growth of both fungi. P. nordicum was unable to produce detectable OTA in both sausage-based media under any condition. In ham, yeasts reduced OTA production, while the starter culture significantly increased it. Unexpectedly, OTA production by A. westerdijkiae was significantly stimulated in all media tested by all microorganisms. Matrix has a significant effect on OTA production by P. nordicum, but not by A. westerdijkiae, for which only temperature showed to have effect. By testing the mechanisms of action by which starter culture and C. zeylanoides influenced fungal responses, we were able to determine that direct contact and simultaneous growth of test organisms were the mechanisms more significantly involved in the responses. In conclusion, ochratoxigenic fungi do not all respond to antagonistic microorganisms in the same way. The use of biocontrol agents with the intent of reducing fungal growth and mycotoxin production by one fungus can have unexpected effects on others, thus leading to unforeseen safety problems. Further experiments are recommended to properly understand the reasons behind the different effects of microorganisms, to ensure their safe as biocontrol agents.

Marine algae are one of the major sources of biologic compounds. In extracellular matrix of these organisms there are sulfated polysaccharides that functions as structural components and provides protection against dehydration. The fraction 1.0 (F1.0) rich in sulfated galactans obtained from red seaweed Hypnea musciformis was physicochemical characterized and evaluated for pharmacologic activity through antioxidant activity, cytotoxic action on erythrocytes, anticoagulant, stimulatory action under antithrombotic heparan sulfate synthesis and their effects on cell proliferation and cycle cell progression. The main components of F1.0 were carbohydrates (49.70 ± 0.10%) and sulfate (44.59 ± 0.015%), presenting phenolic compounds (4.79 ± 0.016%) and low protein contamination (0.92 ± 0.001%). Fraction 1.0 showed polidisperse profile and signs in infrared analysis in 1262, 1074 and 930, 900 and 850 attributed to sulfate esters S=O bond, presence of a 3,6- anidrogalactose C-O bond, non-sulfated β-D-galactose and a C-O-SO4 bond in galactose C4, respectively. The fraction rich in sulfated galactans exhibited strong antioxidant action under lipid peroxidation assay with IC50 of 0.003 mg/mL. Besides the inhibition of hemolysis induced by H2O2 in erythrocytes treated with F1.0, this fraction did not promote significant cytotoxity under erythrocytes membranes. F1.0 exhibited low anticoagulant activity causing moderate direct inhibition of enzimatic activity of thrombin. This fraction promoted stimulation around of 4.6 times on this synthesis of heparan sulfate (HS) by rabbit aortic endothelial cells (RAEC) in culture when was compared with non treated cells. The fraction of this algae displayed antiproliferative action under RAEC cells causing incresing on cell number on S fase, blocking the cycle cell progression. Thus F1.0 presented cytostatic and no cytotoxic action under this cell lineage. These results suggest that F1.0 from H. musciformis have antioxidant potential which is a great effect for a compound used as food and in food industry which could be an alternative to food industry to prevent quality decay of lipid containing food due to lipid peroxidation. These polysaccharides prevent the lipid peroxidation once the fraction in study exhibited strong inhibitory action of this process. Furthermore that F1.0 present strong antithrombotic action promoting the stimulation of antithrombotic HS synthesis by endothelial cells, being important for thrombosis preventing, by its inhibitory action under reactive oxygen species (ROS) in some in vitro methods, being involved in promotion of hypercoagulability state.

Iron is an essential element for many cellular functions, including the immune response against intracellular pathogens. In this study, we aimed evaluate the effect of iron on IRP2, IFN-γ, TNF-α, IL-6, IL-10, MIG and IP10 expression in PBMC and assess the effect of the spleen parasite load on the expression of these genes in the spleen of L. infantum naturally infected dogs. Blood sample from 7 DTH+ donor was collected and PBMC was obtained. The cells were cultivated in absence (iron chelator desferroximane, DFO 10 μM supplemented media) or in presence of iron (hemin 6 mM) for 1 h, followed by stimulation with Leishmania infatum antigen for 4 h. 44 dog spleen samples were obtained and parasite load in this organ was determinate by qPCR. Gene expression was analyzed by qPCR and cytokine production quantified by flow cytometry. In antigen stimulated cells, genes involved in immune response are significantly more expressed in presence of iron. T CD4+ and TCD8+ lymphocytes produces IFN-γ, TNF-α and IL-10 possibly in iron dependent pathway. Monocytes antigen stimulated reduced TNF-α, IL-6 and IL-10 production in presence of iron. We found spleen of infected dogs IRP2 expression increases according to parasite load in that organ, while an inverse profile was found for IFN-γ, TNF-α e IL-10 expression. These results suggest that T lymphocytes depends on iron to produce IFN-γ, TNF-α and IL-10, while iron seems to inhibit cytokine production in monocytes. So, we propose an immunoregulatory mechanism carried out by iron during L. infantum infection in humans and dogs

Sugarcane (Saccharum spp.) is a plant from Poaceae family that has an impressive ability to accumulate sucrose in the stalk, making it a significant component of the economy of many countries. About 100 countries produce sugarcane in an area of 22 million hectares worldwide. For this reason, many studies have been done using sugarcane as a plant model in order to improve production. A change in gravity may be one kind of abiotic stress, since it generates rapid responses after stimulation. In this work we decided to investigate the possible morphophysiological, biochemical and molecular changes resulting from microgravity. Here, we present the contributions of an experiment where sugarcane plants were submitted to microgravity flight using a vehicle VSB-30, a sounding rocket developed by Aeronautics and Space Institute teams, in cooperation with the German Space Agency. Sugarcane plants with 10 days older were submitted to a period of six minutes of microgravity using the VSB-30 rocket. The morphophysiological analyses of roots and leaves showed that plants submitted to the flight showed changes in the conduction tissues, irregular pattern of arrangement of vascular bundles and thickening of the cell walls, among other anatomical changes that indicate that the morphology of the plants was substantially influenced by gravitational stimulation, besides the accumulation of hydrogen peroxide, an important signaling molecule in stress conditions. We carried out RNA extraction and sequencing using Illumina platform. Plants subjected to microgravity also showed changes in enzyme activity. It was observed an increased in superoxide dismutase activity in leaves and a decreased in its activity in roots as well as for ascorbate peroxidase activity. Thus, it was concluded that the changes in gravity were perceived by plants, and that microgravity environment triggered changes associated with a reactive oxygen specie signaling process. This work has helped the understanding of how the gravity affects the structural organization of the plants, by comparing the anatomy of plants subjected to microgravity and plants grown in 1g gravity

Seaweeds are a major source of biologically active compounds . In the extracellular matrix of these organisms are sulfated polysaccharides that functions as structural components preventing it against dehydration. The fraction 0.9 (FucB) rich in sulfated fucans obtained from brown seaweed Dictyota menstrualis was chemical characterized and evaluated for pharmacological activity by testing anticoagulant activity, stimulatory action on the synthesis of an antithrombotic heparan sulfate, antioxidant activity and its effects in cell proliferation. The main components were FucB carbohydrates (49.80 ± 0.10 %) and sulfate (42.30 ± 0.015 %), with phenolic compounds ( 3.86 ± 0.016 %) and low protein contamination ( 0.58 ± 0.001 % ) . FucB showed polydisperse profile and analysis of signals in the infrared at 1262, 1074 and 930 cm -1 and 840 assigned to S = O bonds sulfate esters , CO bond presence of 3,6- anhydrogalactose , β -D- galactose non- sulfated sulfate and the axial position of fucose C4 , respectively. FucB exhibited moderate anticoagulant activity , the polysaccharides prolonged time (aPTT ) 200 ug ( > 90s ) partial thromboplastin FucB no effect on prothrombin time (PT), which corresponds to the extrinsic pathway of coagulation was observed. This stimulation promoted fraction of about 3.6 times the synthesis of heparan sulfate (HS) by endothelial cells of the rabbit aorta ( RAEC ) in culture compared with cells not treated with FucB . This has also been shown to compete for the binding site with heparin. The rich fraction sulfated fucans exhibited strong antioxidant activity assays on total antioxidant (109.7 and 89.5 % compared with BHT and ascorbic acid standards ) , reducing power ( 71 % compared to ascorbic acid ) and ferric chelation ( 71 , comparing with 5 % ascorbic acid). The fraction of algae showed cytostatic activity on the RAEC cells revealed that the increase of the synthesis of heparan sulfate is not related to proliferation. FucB showed antiproliferative action on cell lines modified as Hela and Hep G2 by MTT assay . These results suggest that FucB Dictyota menstrualis have anticoagulant , antithrombotic , antioxidant potential as well as a possible antitumor action, promoting the stimulation of the synthesis of antithrombotic HS by endothelial cells and is useful in the prevention of thrombosis, also due to its inhibitory action on species reactive oxygen ( ROS ) in some in vitro systems , being involved in promoting a hypercoagulable state

Pretendemos, com este estudo, analisar o modo como a escolarização das crianças de etnia cigana e o seu desenvolvimento cognitivo são afetados, entre outros, pelo desfavorecimento sociocultural de que são alvo, justificar a resistência destes alunos a todo o tipo de aprendizagens formais e apurar acerca das representações sociais que envolvem estas comunidades. A escolarização desta minoria, cuja dificuldade é diretamente proporcional ao grau de etnicidade do grupo em estudo, tem vindo a gerar uma crescente preocupação por parte dos vários governos, do que resultou, recentemente, um pedido dos Estados-Membros, à União Europeia, para que elaborasse um Plano Estratégico de Integração das Comunidades Ciganas, onde a Educação é um dos eixos de intervenção. Sendo o professor um elemento primordial no processo de ensino-aprendizagem, cabe-lhe a ele e à instituição onde presta funções, enquanto responsáveis por garantir um ensino de equidade e qualidade, aberto à multi e à interculturalidade, recorrer às práticas pedagógicas e às medidas mais adequadas com vista à estimulação das capacidades cognitivas destes alunos e à efetiva inclusão da diversidade na comunidade escolar. Dos resultados obtidos, as representações da classe docente, face a esta franja social, estão ainda muito marcadas pelo preconceito, pelo estereótipo e pela segregação, embora as responsabilidades sejam bilaterais. Como acontece com qualquer aluno, para que se atinja sucesso na escolarização, torna-se premente uma maior sensibilização das famílias para a utilidade das aprendizagens e uma consequente coresponsabilização pelo percurso dos seus educandos. Surge ainda a questão da legitimidade de avaliar o perfil cognitivo destes alunos, tão específicos do ponto de vista da sua identidade histórica e cultural, pelos mesmos parâmetros dos alunos da sociedade maioritária. Paralelamente, extra escola, há todo um trabalho de concertação ao nível da integração social desta minoria que, quando bem conseguido, se refletirá em termos educativos mas que exige a articulação e o esforço conjunto de várias instâncias, nem sempre dispostas a investir nestas comunidades.

Ao longo das minhas vivências pessoais e percurso académico fui-me deparando com injustiças para com aqueles que num “golpe de azar “ nasceram com algum problema que os fazia necessitar de estratégias pedagógicas especiais. A igualdade não é sinónima de justiça. E tratar com justiça é tratar cada um segundo as suas circunstâncias. Esta perspectiva fez-me ter vontade de me especializar em Educação Especial para estar melhor preparada, de forma a contribuir para atenuar as necessidades educativas de cada criança. Escolhi o tema Perturbação do Espectro do Autismo por ser um desafio. Neste sentido optei por verificar a importância da Expressão Plástica na socialização de crianças com Perturbações do Espectro do Autismo porque a arte é a forma como o ser humano apreende a realidade e é interessante ver como ela pode ou não influenciar o indivíduo na sua socialização, principalmente, quando ele é portador autismo . O meu trabalho terá como objetivo aprofundar os conhecimentos sobre a influência da Expressão Plástica no Autismo, nomeadamente, na socialização de crianças. A interação social das crianças autistas em casa é limitada, faltando-lhes iniciativa e sensibilidade para com os outros. Evitam o contato social refugiando-se nelas mesmas. Será a Expressão Plástica um meio importante para a estimulação e inclusão das crianças autistas? Esta área, através de métodos e técnicas específicas, ajuda estas crianças nas suas limitações e na interação social permitindo-lhes adquirir novos conhecimentos?
