Potassium fertilization is very important to alfalfa crop in terms of yield, quality and persistence of forage, especially on soils naturally poor K. Thus, to assess the effects of K fertilization in alfalfa production and nutritional status, was carried out an experiment in a greenhouse using samples of a Dystrophic Oxisol medium texture (LV) (0.6 mmol(c) dm(-3) K) and a Dystrophic Ultisol sandy/medium texture (PVA) (2.2 mmol(c) dm(-3) K). A completely randomized design in a factorial arrangement 6 x 2 (six K rates and two soils) was used, with four replications. The K rates used were: 0, 25, 50, 100, 150 and 200 mg kg(-1) K. Potassium fertilization increased K content in soil and shoots. Dry matter production was increased with the K addition. However, in the PVA, this occurred only in the second cut. In LV, potassium fertilization increased N concentration in alfalfa shoots in both cuts. Plants with K concentration around 10 g kg(-1) had typical symptoms of this nutrient deficiency. The K critical levels of K in soil and shoots were 1.8 mmolc dm(-3) and 16.7 g kg(-1), respectively.

We present a new method to quantify substructures in clusters of galaxies, based on the analysis of the intensity of structures. This analysis is done in a residual image that is the result of the subtraction of a surface brightness model, obtained by fitting a two-dimensional analytical model (beta-model or Sersic profile) with elliptical symmetry, from the X-ray image. Our method is applied to 34 clusters observed by the Chandra Space Telescope that are in the redshift range z is an element of [0.02, 0.2] and have a signal-to-noise ratio (S/N) greater than 100. We present the calibration of the method and the relations between the substructure level with physical quantities, such as the mass, X-ray luminosity, temperature, and cluster redshift. We use our method to separate the clusters in two sub-samples of high-and low-substructure levels. We conclude, using Monte Carlo simulations, that the method recuperates very well the true amount of substructure for small angular core radii clusters (with respect to the whole image size) and good S/N observations. We find no evidence of correlation between the substructure level and physical properties of the clusters such as gas temperature, X-ray luminosity, and redshift; however, analysis suggest a trend between the substructure level and cluster mass. The scaling relations for the two sub-samples (high-and low-substructure level clusters) are different (they present an offset, i. e., given a fixed mass or temperature, low-substructure clusters tend to be more X-ray luminous), which is an important result for cosmological tests using the mass-luminosity relation to obtain the cluster mass function, since they rely on the assumption that clusters do not present different scaling relations according to their dynamical state.

In questo elaborato abbiamo analizzato un campione di 22 galssie early-type. Utilizzando una tecnica di cross-correlazione, abbiamo ottenuto profili radiali di rotazione e di dis- persione di velocitá. Questi dati ci hanno permesso di investigare molte delle proprietá dinamiche delle nostre galassie. Abbiamo ottenuto indizi sull’anisotropia orbitale e stimato le masse e il rapporto M/L del campione. Le masse misurate variano da 1010 a 1012 M , mentre i valori degli M/L, per cui abbiamo trovato una dipendenza del tipo Log M/L ∝ 0.28 Log L , sono dell’ordine dell’unitá. Abbiamo anche riprodotto le famose relazioni di scala e abbi- amo utlizzato un set di dati sugli indici di Lick/IDS per ricercare relazioni tra le proprietá chimiche e quelle dinamiche. In particolare, abbiamo riscontrato una correlazione tra molti degli indici dipendenti dalla metallicitá e la profonditá della buca di potenziale. Tali indici sembrano correlare anche con il M/L. La rotazione e la forma del profilo di dispersione di velocitá sembrano essere ininfluenti sulle proprietá chimiche. In ultima analisi, abbiamo considerato le implicazioni delle nostre misure riguardo la natura della popolazione stellare e dell’emissione X delle nostre galassie. L’indice di colore e il M/L sembrano indicare che la popolazione stellare delle nostre galassie é dominata da stelle appartenenti alle classi spettrali late-G e early-K. Sembra inoltre esserci una correlazione tra l’emissione X degli elementi del nostro campione e la profonditá della loro buca di potenziale.

The aim of this work was to study the dense cloud structures and to obtain the mass distribution of the dense cores (CMF) within the NGC6357 complex, from observations of the dust continuum at 450 and 850~$\mu$m of a 30 $\times$ 30 arcmin$^2$ region containing the H\textsc{ii} regions, G353.2+0.9 and G353.1+0.6.

Pancreatic beta-cell-restricted knockout of the insulin receptor results in hyperglycemia due to impaired insulin secretion, suggesting that this cell is an important target of insulin action. The present studies were undertaken in beta-cell insulin receptor knockout (betaIRKO) mice to define the mechanisms underlying the defect in insulin secretion. On the basis of responses to intraperitoneal glucose, approximately 7-mo-old betaIRKO mice were either diabetic (25%) or normally glucose tolerant (75%). Total insulin content was profoundly reduced in pancreata of mutant mice compared with controls. Both groups also exhibited reduced beta-cell mass and islet number. However, insulin mRNA and protein were similar in islets of diabetic and normoglycemic betaIRKO mice compared with controls. Insulin secretion in response to insulin secretagogues from the isolated perfused pancreas was markedly reduced in the diabetic betaIRKOs and to a lesser degree in the nondiabetic betaIRKO group. Pancreatic islets of nondiabetic betaIRKO animals also exhibited defects in glyceraldehyde- and KCl-stimulated insulin release that were milder than in the diabetic animals. Gene expression analysis of islets revealed a modest reduction of GLUT2 and glucokinase gene expression in both the nondiabetic and diabetic mutants. Taken together, these data indicate that loss of functional receptors for insulin in beta-cells leads primarily to profound defects in postnatal beta-cell growth. In addition, altered glucose sensing may also contribute to defective insulin secretion in mutant animals that develop diabetes.

Retinitis pigmentosa (RP) point mutations in both the intradiscal (ID) and transmembrane domains of rhodopsin cause partial or complete misfolding of rhodopsin, resulting in loss of 11-cis-retinal binding. Previous work has shown that misfolding is caused by the formation of a disulfide bond in the ID domain different from the native Cys-110–Cys-187 disulfide bond in native rhodopsin. Here we report on direct identification of the abnormal disulfide bond in misfolded RP mutants in the transmembrane domain by mass spectrometric analysis. This disulfide bond is between Cys-185 and Cys-187, the same as previously identified in misfolded RP mutations in the ID domain. The strategy described here should be generally applicable to identification of disulfide bonds in other integral membrane proteins.

We address two issues in the determination of particulate carbon and nitrogen in suspended matter of aquatic environments. One is the adsorption of dissolved organic matter on filters, leading to overestimate particulate matter. The second is the material loss during filtration due to fragile algal cells breaking up. Examples from both laboratory cultures and natural samples are presented. We recommend using stacked filters in order to estimate thefirst and filtering different volumes of water in order to evaluate the second.

The research described in this PhD thesis focuses on proteomics approaches to study the effect of oxidation on the modification status and protein-protein interactions of PTEN, a redox-sensitive phosphatase involved in a number of cellular processes including metabolism, apoptosis, cell proliferation, and survival. While direct evidence of a redox regulation of PTEN and its downstream signaling has been reported, the effect of cellular oxidative stress or direct PTEN oxidation on PTEN structure and interactome is still poorly defined. In a first study, GST-tagged PTEN was directly oxidized over a range of hypochlorous acid (HOCl) concentration, assayed for phosphatase activity, and oxidative post-translational modifications (oxPTMs) were quantified using LC-MS/MS-based label-free methods. In a second study, GSTtagged PTEN was prepared in a reduced and reversibly H2O2-oxidized form, immobilized on a resin support and incubated with HCT116 cell lysate to capture PTEN interacting proteins, which were analyzed by LC-MS/MS and comparatively quantified using label-free methods. In parallel experiments, HCT116 cells transfected with a GFP-tagged PTEN were treated with H2O2 and PTENinteracting proteins immunoprecipitated using standard methods. Several high abundance HOCl-induced oxPTMs were mapped, including those taking place at amino acids known to be important for PTEN phosphatase activity and protein-protein interactions, such as Met35, Tyr155, Tyr240 and Tyr315. A PTEN redox interactome was also characterized, which identified a number of PTEN-interacting proteins that vary with the reversible inactivation of PTEN caused by H2O2 oxidation. These included new PTEN interactors as well as the redox proteins peroxiredoxin-1 (Prdx1) and thioredoxin (Trx), which are known to be involved in the recycling of PTEN active site following H2O2-induced reversible inactivation. The results suggest that the oxidative modification of PTEN causes functional alterations in PTEN structure and interactome, with fundamental implications for the PTEN signaling role in many cellular processes, such as those involved in the pathophysiology of disease and ageing.

We present a study of the star-forming properties of a stellar mass-selected sample of galaxies in the GOODS (Great Observatories Origins Deep Survey) NICMOS Survey (GNS), based on deep Hubble Space Telescope (HST) imaging of the GOODS North and South fields. Using a stellar mass-selected sample, combined with HST/ACS and Spitzer data to measure both ultraviolet (UV) and infrared-derived star formation rates (SFRs), we investigate the star forming properties of a complete sample of ∼1300 galaxies down to log M_*= 9.5 at redshifts 1.5 < z < 3. Eight per cent of the sample is made up of massive galaxies with M_*≥ 10^11 M_⊙. We derive optical colours, dust extinctions and UV and infrared SFR to determine how the SFR changes as a function of both stellar mass and time. Our results show that SFR increases at higher stellar mass such that massive galaxies nearly double their stellar mass from star formation alone over the redshift range studied, but the average value of SFR for a given stellar mass remains constant over this ∼2 Gyr period. Furthermore, we find no strong evolution in the SFR for our sample as a function of mass over our redshift range of interest; in particular we do not find a decline in the SFR among massive galaxies, as is seen at z < 1. The most massive galaxies in our sample (log M_*≥ 11) have high average SFRs with values SFR_UV, corr= 103 ± 75 M_⊙ yr^−1, and yet exhibit red rest-frame (U−B) colours at all redshifts. We conclude that the majority of these red high-redshift massive galaxies are red due to dust extinction. We find that A_2800 increases with stellar mass, and show that between 45 and 85 per cent of massive galaxies harbour dusty star formation. These results show that even just a few Gyr after the first galaxies appear, there are strong relations between the global physical properties of galaxies, driven by stellar mass or another underlying feature of galaxies strongly related to the stellar mass.
