Urban centers significantly contribute to anthropogenic air pollution, although they cover only a minor fraction of the Earth's land surface. Since the worldwide degree of urbanization is steadily increasing, the anthropogenic contribution to air pollution from urban centers is expected to become more substantial in future air quality assessments. The main objective of this thesis was to obtain a more profound insight in the dispersion and the deposition of aerosol particles from 46 individual major population centers (MPCs) as well as the regional and global influence on the atmospheric distribution of several aerosol types. For the first time, this was assessed in one model framework, for which the global model EMAC was applied with different representations of aerosol particles. First, in an approach with passive tracers and a setup in which the results depend only on the source location and the size and the solubility of the tracers, several metrics and a regional climate classification were used to quantify the major outflow pathways, both vertically and horizontally, and to compare the balance between pollution export away from and pollution build-up around the source points. Then in a more comprehensive approach, the anthropogenic emissions of key trace species were changed at the MPC locations to determine the cumulative impact of the MPC emissions on the atmospheric aerosol burdens of black carbon, particulate organic matter, sulfate, and nitrate. Ten different mono-modal passive aerosol tracers were continuously released at the same constant rate at each emission point. The results clearly showed that on average about five times more mass is advected quasi-horizontally at low levels than exported into the upper troposphere. The strength of the low-level export is mainly determined by the location of the source, while the vertical transport is mainly governed by the lifting potential and the solubility of the tracers. Similar to insoluble gas phase tracers, the low-level export of aerosol tracers is strongest at middle and high latitudes, while the regions of strongest vertical export differ between aerosol (temperate winter dry) and gas phase (tropics) tracers. The emitted mass fraction that is kept around MPCs is largest in regions where aerosol tracers have short lifetimes; this mass is also critical for assessing the impact on humans. However, the number of people who live in a strongly polluted region around urban centers depends more on the population density than on the size of the area which is affected by strong air pollution. Another major result was that fine aerosol particles (diameters smaller than 2.5 micrometer) from MPCs undergo substantial long-range transport, with about half of the emitted mass being deposited beyond 1000 km away from the source. In contrast to this diluted remote deposition, there are areas around the MPCs which experience high deposition rates, especially in regions which are frequently affected by heavy precipitation or are situated in poorly ventilated locations. Moreover, most MPC aerosol emissions are removed over land surfaces. In particular, forests experience more deposition from MPC pollutants than other land ecosystems. In addition, it was found that the generic treatment of aerosols has no substantial influence on the major conclusions drawn in this thesis. Moreover, in the more comprehensive approach, it was found that emissions of black carbon, particulate organic matter, sulfur dioxide, and nitrogen oxides from MPCs influence the atmospheric burden of various aerosol types very differently, with impacts generally being larger for secondary species, sulfate and nitrate, than for primary species, black carbon and particulate organic matter. While the changes in the burdens of sulfate, black carbon, and particulate organic matter show an almost linear response for changes in the emission strength, the formation of nitrate was found to be contingent upon many more factors, e.g., the abundance of sulfuric acid, than only upon the strength of the nitrogen oxide emissions. The generic tracer experiments were further extended to conduct the first risk assessment to obtain the cumulative risk of contamination from multiple nuclear reactor accidents on the global scale. For this, many factors had to be taken into account: the probability of major accidents, the cumulative deposition field of the radionuclide cesium-137, and a threshold value that defines contamination. By collecting the necessary data and after accounting for uncertainties, it was found that the risk is highest in western Europe, the eastern US, and in Japan, where on average contamination by major accidents is expected about every 50 years.

Die Bildung von lokalen Rezidiven wird bei Glioblastomen vor allem durch das stark infiltrierende Wachstum gefördert. Die Rolle der angewendeten Therapieverfahren bei der Induktion der Zellmotilität ist noch weitgehend unklar. Im Rahmen dieser Dissertation wurde daher in vitro die Wirkung der Photonen- und Schwerionenstrahlung auf die Migration von humanen Glioblastomzelllinien sowie auf EGFR-gekoppelte, migrationsregulierende Signalmoleküle untersucht. Gezeigt werden konnte, dass die EGF-induzierte Stimulierung des EGFR über den PI3K und MAPK Signalweg an der Regulation der Zellmigration beteiligt ist. Hinsichtlich des Verhaltens nach Bestrahlung wurden Zelllinien- und Strahlen-spezifische Unterschiede beobachtet. Die Photonenstrahlung führte in U87 Zellen zu einer Aktivierung des EGFR sowie zur Steigerung der Migration nach klinisch relevanten Dosen. Versuche mit einem EGFR spezifischen Inhibitor bestätigten die funktionelle Verknüpfung von Strahlen-induzierter Aktivierung des EGFR und Strahlen-induzierter Migrationssteigerung. Demgegenüber wurden nach Bestrahlung mit Kohlenstoffionen eine Hemmung der Zellmigration sowie keine gesteigerte Aktivität des EGFR festgestellt. Die erhaltenen in vitro Ergebnisse geben Hinweise auf ein in Glioblastomen mögliches erhöhtes Risiko einer Rezidivbildung nach einer konventionellen Radiotherapie mit Photonen. Bei der modernen Schwerionentherapie kann dieses Risiko aufgrund der Strahlen-vermittelten Migrationshemmung weitestgehend ausgeschlossen werden. Sollte sich die Strahlen-induzierte Migrationssteigerung in vivo bestätigen, wäre es sinnvoll den Einsatz von Migrationsinhibitoren als Begleittherapie zur Bestrahlung zu testen.

Since historical times, coastal areas throughout the eastern Mediterranean are exposed to tsunami hazard. For many decades the knowledge about palaeotsunamis was solely based on historical accounts. However, results from timeline analyses reveal different characteristics affecting the quality of the dataset (i.e. distribution of data, temporal thinning backward of events, local periodization phenomena) that emphasize the fragmentary character of the historical data. As an increasing number of geo-scientific studies give convincing examples of well dated tsunami signatures not reported in catalogues, the non-existing record is a major problem to palaeotsunami research. While the compilation of historical data allows a first approach in the identification of areas vulnerable to tsunamis, it must not be regarded as reliable for hazard assessment. Considering the increasing economic significance of coastal regions (e.g. for mass tourism) and the constantly growing coastal population, our knowledge on the local, regional and supraregional tsunami hazard along Mediterranean coasts has to be improved. For setting up a reliable tsunami risk assessment and developing risk mitigation strategies, it is of major importance (i) to identify areas under risk and (ii) to estimate the intensity and frequency of potential events. This approach is most promising when based on the analysis of palaeotsunami research seeking to detect areas of high palaeotsunami hazard, to calculate recurrence intervals and to document palaeotsunami destructiveness in terms of wave run-up, inundation and long-term coastal change. Within the past few years, geo-scientific studies on palaeotsunami events provided convincing evidence that throughout the Mediterranean ancient harbours were subject to strong tsunami-related disturbance or destruction. Constructed to protect ships from storm and wave activity, harbours provide especially sheltered and quiescent environments and thus turned out to be valuable geo-archives for tsunamigenic high-energy impacts on coastal areas. Directly exposed to the Hellenic Trench and extensive local fault systems, coastal areas in the Ionian Sea and the Gulf of Corinth hold a considerably high risk for tsunami events, respectively.Geo-scientific and geoarcheaological studies carried out in the environs of the ancient harbours of Krane (Cefalonia Island), Lechaion (Corinth, Gulf of Corinth) and Kyllini (western Peloponnese) comprised on-shore and near-shore vibracoring and subsequent sedimentological, geochemical and microfossil analyses of the recovered sediments. Geophysical methods like electrical resistivity tomography and ground penetrating radar were applied in order to detect subsurface structures and to verify stratigraphical patterns derived from vibracores over long distances. The overall geochronological framework of each study area is based on radiocarbon dating of biogenic material and age determination of diagnostic ceramic fragments. Results presented within this study provide distinct evidence of multiple palaeotsunami landfalls for the investigated areas. Tsunami signatures encountered in the environs of Krane, Lechaion and Kyllini include (i) coarse-grained allochthonous marine sediments intersecting silt-dominated quiescent harbour deposits and/or shallow marine environments, (ii) disturbed microfaunal assemblages and/or (iii) distinct geochemical fingerprints as well as (iv) geo-archaeological destruction layers and (v) extensive units of beachrock-type calcarenitic tsunamites. For Krane, geochronological data yielded termini ad or post quem (maximum ages) for tsunami event generations dated to 4150 ± 60 cal BC, ~ 3200 ± 110 cal BC, ~ 650 ± 110 cal BC, and ~ 930 ± 40 cal AD, respectively. Results for Lechaion suggest that the harbour was hit by strong tsunami impacts in the 8th-6th century BC, the 1st-2nd century AD and in the 6th century AD. At Kyllini, the harbour site was affected by tsunami impact in between the late 7th and early 4th cent. BC and between the 4th and 6th cent. AD. In case of Lechaion and Kyllini, the final destruction of the harbour facilities also seems to be related to the tsunami impact. Comparing the tsunami signals obtained for each study areas with geo-scientific data from palaeotsunami events from other sites indicates that the investigated harbour sites represent excellent geo-archives for supra-regional mega-tsunamis.

Acute myeloid leukaemia (AML) is a cancer of the haematopoietic system, which can in many cases only be cured by haematopoietic stem cell transplantation (HSCT) and donor lymphocyte infusion (DLI) (Burnett et al., 2011). This therapy is associated with the beneficial graft-versus-leukaemia (GvL) effect mediated by transplanted donor T and NK cells that either recognise mismatch HLA molecules or polymorphic peptides, so-called minor histocompatibility antigens, leukaemia-associated or leukaemia-specific antigens in the patient and thus eliminate remaining leukaemic blasts. Nevertheless, the mature donor-derived cells often trigger graft-versus-host disease (GvHD), leading to severe damages in patients’ epithelial tissue, mainly skin, liver and intestine (Bleakley & Riddell, 2004). Therefore, approaches for the selective mediation of strong GvL effects are needed, also in order to prevent relapse after transplantation. One promising opportunity is the in vitro generation of AML-reactive CD4+ T cells for adoptive transfer. CD4+ T cells are advantageous compared to CD8+ T cells, as HLA class II molecules are under non-inflammatory conditions only expressed on haematopoietic cells; a fact that would minimise GvHD (Klein & Sato, 2000). In this study, naive CD4+ T cells were isolated from healthy donors and were successfully stimulated against primary AML blasts in mini-mixed lymphocyte/leukaemia cell cultures (mini-MLLC) in eight patient/donor pairs. After three to seven weekly restimulations, T cells were shown to produce TH1 type cytokines and to be partially of monoclonal origin according to their TCR Vβ chain usage. Furthermore, they exhibited lytic activity towards AML blasts, which was mediated by the release of granzymes A and B and perforin. The patient/donor pairs used in this study were fully HLA-class I matched, except for one pair, and also matched for HLA-DR and -DQ, whereas -DP was mismatched in one or both alleles, reflecting the actual donor selection procedure in the clinic (Begovich et al., 1992). Antibody blocking experiments suggested that the generated CD4+ T cells were directed against the HLA-DP mismatches, which could be confirmed by the recognition of donor-derived lymphoblastoid cell lines (LCLs) electroporated with the mismatched DP alleles. Under non-inflammatory conditions primary fibroblasts did not express HLA-DP and were thus not recognised, supporting the idea of a safer application of CD4+ T cells regarding induction of GvHD. For the assessment of the biological significance of these T cells, they were adoptively transferred into NSG mice engrafted with human AML blasts, where they migrated to the bone marrow and lymphoid tissue and succeeded in eliminating the leukaemic burden after only one week. Therefore, AML-reactive CD4+ T cells expanded from the naive compartment by in vitro stimulation with primary leukaemia blasts appear to be a potent tool for DLI in HSCT patients and promise to mediate specific GvL effects without causing GvHD.

Oxidative DNA-Schäden, wie 7,8-Dihydro-8-oxoguanin (8-oxoG), werden kontinuierlich in allen Zellen durch endogene und exogene Noxen gebildet. Ohne eine effektive Reparatur können DNA-Schäden nach erfolgter Replikation als Mutationen fixiert werden und somit die Kanzerogenese initiieren.rnUntersuchungsgegenstand dieser Arbeit war die Reparatur, vorrangig von oxidativen DNA-Schäden, in humanen Lymphozyten. Dabei sollte ebenfalls überprüft werden, inwiefern eine Aktivierung dieser Immunzellen, die u.a. zu einer Initiierung der Proliferation führt, modulierend auf die DNA-Reparatur wirkt. Für diese Untersuchungen wurden primäre Lymphozyten aus Buffy Coats isoliert. Eine Aktivierung von T Lymphozyten, welche physiologisch Antigen-vermittelt über den T-Zell-Rezeptor verläuft, wurde durch eine ex vivo Stimulation mit Phytohämagglutinin (PHA) nachgeahmt. Die Induktion oxidativer DNA-Basenmodifikationen erfolgte mit Hilfe des Photosensibilisators Acridinorange in Kombination mit sichtbarem Licht. Das Schadensausmaß sowie die Reparatur wurden mittels der Alkalischen Elution unter Nutzung der Reparaturendonuklease Fpg bestimmt.rnDie Ergebnisse zeigten, dass global keine Reparatur induzierter oxidativer DNA-Schäden in primären Lymphozyten stattfindet. Eine Aktivierung der Lymphozyten mittels PHA führte hingegen zu einer deutlichen Reduktion der induzierten DNA-Schäden innerhalb einer 24-stündigen Reparaturzeit. Diese verbesserte Reparatur konnte auf eine Steigerung der Transkription und somit eine erhöhte Proteinmenge von OGG1, welches die Reparatur von 8-oxoG DNA-Glykosylase initiiert, zurückgeführt werden. Weiterführende mechanistische Untersuchungen deuten darauf hin, dass der transkriptionellen Regulation von OGG1 eine Aktivierung der JNK-Signalkaskade zugrunde liegt. Als ein verantwortlicher Transkriptionsfaktor konnte NF-YA identifiziert werden. Dessen erhöhte Bindung am OGG1-Promotor in Folge einer PHA-Stimulation konnte durch eine JNK-Hemmung reduziert werden.rnDie Ergebnisse dieser Arbeit zeigen, dass eine Aktivierung von Lymphozyten, welche die Proliferation initiiert und dadurch mit dem Risiko für die Entstehung von Mutationen und malignen Entartungen verknüpft ist, gleichzeitig eine transkriptionelle Hochregulation von OGG1 bewirkt, die die Reparatur oxidativer DNA-Schäden sicherstellt. Die Fähigkeit zur Steigerung der DNA-Reparatur unter den gezeigten Bedingungen bietet den proliferierenden Zellen einen Schutzmechanismus zur Erhaltung ihrer genomischen Stabilität.rn

Glioblastoma multiforme (GBM) is the most common and most aggressive astrocytic tumor of the central nervous system (CNS) in adults. The standard treatment consisting of surgery, followed by a combinatorial radio- and chemotherapy, is only palliative and prolongs patient median survival to 12 to 15 months. The tumor subpopulation of stem cell-like glioma-initiating cells (GICs) shows resistance against radiation as well as chemotherapy, and has been suggested to be responsible for relapses of more aggressive tumors after therapy. The efficacy of immunotherapies, which exploit the immune system to specifically recognize and eliminate malignant cells, is limited due to strong immunosuppressive activities of the GICs and the generation of a specialized protective microenvironment. The molecular mechanisms underlying the therapy resistance of GICs are largely unknown. rnThe first aim of this study was to identify immune evasion mechanisms in GICs triggered by radiation. A model was used in which patient-derived GICs were treated in vitro with fractionated ionizing radiation (2.5 Gy in 7 consecutive passages) to select for a more radio-resistant phenotype. In the model cell line 1080, this selection process resulted in increased proliferative but diminished migratory capacities in comparison to untreated control GICs. Furthermore, radio-selected GICs downregulated various proteins involved in antigen processing and presentation, resulting in decreased expression of MHC class I molecules on the cellular surface and diminished recognition potential by cytotoxic CD8+ T cells. Thus, sub-lethal fractionated radiation can promote immune evasion and hamper the success of adjuvant immunotherapy. Among several immune-associated proteins, interferon-induced transmembrane protein 3 (IFITM3) was found to be upregulated in radio-selected GICs. While high expression of IFITM3 was associated with a worse overall survival of GBM patients (TCGA database) and increased proliferation and migration of differentiated glioma cell lines, a strong contribution of IFITM3 to proliferation in vitro as well as tumor growth and invasiveness in a xenograft model could not be observed. rnMultiple sclerosis (MS) is the most common autoimmune disease of the CNS in young adults of the Western World, which leads to progressive disability in genetically susceptible individuals, possibly triggered by environmental factors. It is assumed that self-reactive, myelin-specific T helper cell 1 (Th1) and Th17 cells, which have escaped the control mechanisms of the immune system, are critical in the pathogenesis of the human disease and its animal model experimental autoimmune encephalomyelitis (EAE). It was observed that in vitro differentiated interleukin 17 (IL-17) producing Th17 cells co-expressed the Th1-phenotypic cytokine Interferon-gamma (IFN-γ) in combination with the two respective lineage-associated transcription factors RORγt and T-bet after re-isolation from the CNS of diseased mice. Pathogenic molecular mechanisms that render a CD4+ T cell encephalitogenic have scarcely been investigated up to date. rnIn the second part of the thesis, whole transcriptional changes occurring in in vitro differentiated Th17 cells in the course of EAE were analyzed. Evaluation of signaling networks revealed an overrepresentation of genes involved in communication between the innate and adaptive immune system and metabolic alterations including cholesterol biosynthesis. The transcription factors Cebpa, Fos, Klf4, Nfatc1 and Spi1, associated with thymocyte development and naïve T cells were upregulated in encephalitogenic CNS-isolated CD4+ T cells, proposing a contribution to T cell plasticity. Correlation of the murine T-cell gene expression dataset to putative MS risk genes, which were selected based on their proximity (± 500 kb; ensembl database, release 75) to the MS risk single nucleotide polymorphisms (SNPs) proposed by the most recent multiple sclerosis GWAS in 2011, revealed that 67.3% of the MS risk genes were differentially expressed in EAE. Expression patterns of Bach2, Il2ra, Irf8, Mertk, Odf3b, Plek, Rgs1, Slc30a7, and Thada were confirmed in independent experiments, suggesting a contribution to T cell pathogenicity. Functional analysis of Nfatc1 revealed that Nfatc1-deficient CD4+ T cells were restrained in their ability to induce clinical signs of EAE. Nfatc1-deficiency allowed proper T cell activation, but diminished their potential to fully differentiate into Th17 cells and to express high amounts of lineage cytokines. As the inducible Nfatc1/αA transcript is distinct from the other family members, it could represent an interesting target for therapeutic intervention in MS.rn

Die Funktionsweise der Laserionenquellenfalle LIST, sowie deren Implementierung bei der Forschungseinrichtung ISOLDE am CERN als neue Standard-Ionenquelle und die ermittelten Spezifikationen Effizienz und Selektivität werden vorgestellt.rnrnDurch die Implementierung der LIST bei ISOLDE konnte on-line mit Hilfe von Radionukliden ein Minimalwert zur Unterdrückung von Kontaminationen durch die LIST bestimmt werden. Die erfolgreiche Unterdrückung von Francium-Kontamination ermöglichte es, neue Messdaten für den mittleren Ladungsradius und die Hyperfeinstruktur für Po-217 zu erzeugen.rnrnUm die Funktionalität der LIST bei ISOLDE hinsichtlich der Ionisationseffizienz gegenüber anderen Ionenquellen einzuordnen, wurden in Mainz am RISIKO-Massenseparator mit der bereits existierenden Standard-Ionenquelle RILIS und der LIST die Effizienzen bestimmt und miteinander verglichen. Es konnte gezeigt werden, dass die LIST im Modus hoher Ionisationseffizienz eine vergleichbare Effizienz aufweist wie die RILIS. Im Modus zur Produktion eines hochreinen Ionenstrahls ist die Ionisationseffizienz gegenüber der RILIS reduziert.rnrnDa die Bestimmung der Selektivität im On-line-Betrieb aufwendig und zeitintensiv ist, wurde die Reinheit des Ionenstrahls am RISIKO-Massenseparator mittels Laufzeitmessungen der Ionen off-line bestimmt und analysiert. Die Zeitstrukturen der RILIS ermöglichen einerseits Rückschlüsse auf die Reinheit des Ionenstrahls zu ziehen, andererseits konnte auch die Ionisation außerhalb des Atomisators, in dem überwiegend die resonante Ionisation stattfindet, nachgewiesen werden. Durch diesen Nachweis kann der Effizienzverlust während der Produktion eines hochreinen Ionenstrahls erklärt werden. Zudem bietet er einen Ansatz für weitere Entwicklungsarbeiten der LIST zur Steigerung der Effizienz.rnrnEine Übertragung der Messergebnisse zur Zeitstruktur der RILIS auf die LIST eröffnet neue Möglichkeiten zur Steigerung deren Selektivität im massenselektiven Mode. Dieser wurde anhand von Simulationen überprüft und mit Messungen an Kalium experimentell quantifiziert.

Adoptive T cell therapy using antigen-specific T lymphocytes is a powerful immunotherapeutic approach against cancer. Nevertheless, many T cells against tumor-antigens exhibit only weak anti-tumoral response. To overcome this barrier it is necessary to improve the potency and anti-tumoral efficacy of these T cells. Activation and activity of T cells are tightly controlled to inhibit unwanted T cell responses and to reduce the risk of autoimmunity. Both are regulated by extrinsic signals and intrinsic mechanisms which suppress T cell activation. The intrinsic mechanisms include the expression of phosphatases that counteract the activation-inducing kinases. Modifying the expression of these phosphatases allows the targeted modulation of T cell reactivity. MicroRNAs (miRNAs) are regulatory small noncoding RNA molecules that control gene expression by targeting messenger RNAs in a sequence specific manner. Gene-specific silencing plays a key role in diverse biological processes, such as development, differentiation, and functionality. miR181a has been shown to be highly expressed in immature T cells that recognize low-affinity antigens.rnThe present study successfully shows that ectopic expression of miR181a is able to enhance the sensitivity of both murine and human T cells. In CD4+ T helper cells as well as in CD8+ cytotoxic T cells the overexpression of miR181a leads to downregulation of multiple phosphatases involved in the T cell receptor signaling pathway. Overexpression of miR181a in human T cells achieves a co-stimulatory independent activation and has an anti-apoptotic effect on CD4+ T helper cells. Additionally, increasing the amount of miR181a enhances the cytolytic activity of murine CD8+ TCRtg T cells in an antigen-specific manner.rnTo test miR181a overexpressing T cells in vivo, a mouse tumor model using a B cell lymphoma cell line (A20-HA) expressing the Influenza hemagglutinin (Infl.-HA) antigen was established. The expression of model antigens in tumor cell lines enables targeted elimination of tumors using TCRtg T cells. The transfer of miR181a overexpressing Infl.-HA TCRtg CD8+ T cells alone has no positive effect neither on tumor control nor on survival of A20-HA tumor-bearing mice. In contrast, the co-transfer of miR181a overexpressing Infl.-HA TCRtg CD8+ and CD4+ T cells leads to improved tumor control and prolongs survival of A20-HA tumor-bearing mice. This effect is characterized by higher amounts of effector T cells and the expansion of Infl.-HA TCRtg CD8+ T cells.rnAll effects were achieved by changes in expression of several genes including molecules involved in T cell differentiation, activation, and regulation, cytotoxic effector molecules, and receptors important for the homing process of T cells in miR181a overexpressing T cells. The present study demonstrates that miR181a is able to enhance the anti-tumoral response of antigen-specific T cells and is a promising candidate for improving adoptive cell therapy.
Il diritto d'asilo in Austria: il ruolo dell'interprete nelle richieste di protezione internazionale

Meine Diplomarbeit beschäftigt sich mit den Themen des Asylverfahrens in Österreich sowie mit der Rolle von DolmetscherInnen im Asylbereich; sie besteht aus der Übersetzung von Teilen einiger Lernmodule des Trainingshandbuchs, das 2015 von UNHCR Österreich herausgegeben wurde, sowie aus meinen Kommentaren. In den ersten Kapiteln werden die Voraussetzungen für einen Asylantrag, die Richtlinie der Europäischen Union und die Bundesgesetze bezüglich des Asylrechts sowie die Rolle der UNHCR erläutert. Es werden dann spezifisch das Asylverfahren in Österreich, die Phasen der Einvernahme mit den Behörden und die Rolle der DolmetscherInnen - mit Blick auf die Auswahl von DolmetscherInnen und die dazu erforderlichen Kompetenzen - analysiert. Darauffolgend werden die Themen von der Haftung der DolmetscherInnen und dem Risiko von einer Traumatisierung für die Sprachmittler sowie das Dolmetschen für vulnerable AntragstellerInnen angesprochen. Zuletzt vertieft meine Arbeit die ethischen Prinzipien für das Dolmetschen im Asylverfahren und analysiert, anhand von konkreten Beispielen, die Probleme, die sich aus den berufsethischen Anforderungen an DolmetscherInnen ergeben. Schließlich wird die Komplexität des Dolmetschens im Asylbereich hervorgehoben.

Die chronisch-kritische Beinischämie (CLI) ist die schwerste Form der peripher-arteriellen Verschlusskrankheit und geht mit einem erhöhten Risiko für letale kardiovaskuläre Ereignisse einher. Die meisten Patienten überleben aber die ersten Jahre nach Diagnosestellung und sind für den Erhalt ihrer Selbständigkeit auf eine nachhaltige Verbesserung der Beindurchblutung angewiesen. Dieser CME-Artikel fasst die chirurgischen und endovaskulären Optionen zur Revaskularisierung zusammen und geht auf deren Nachhaltigkeit bei CLI ein. Grenzen der verfügbaren Evidenz werden aufgezeigt. Sie beruhen vor allem auf dem Fehlen einer einheitlichen Definition des Behandlungserfolges. Ein klinisch orientierter Lösungsvorschlag wird diskutiert, der helfen könnte, die verschiedenen Behandlungsindikationen zu schärfen. Da sich aber nur die wenigsten Patienten gleich gut für verschiedene Verfahren eignen, werden CLI-Patienten auch in Zukunft am besten in einem interdisziplinär arbeitenden Team betreut sein.

Rund ein Drittel der weltweiten Todesfälle sind auf Herz-Kreislauf-Krankheiten zurückzuführen. In den industrialisierten Ländern sind sie die häufigste Todesursache. Den größten Anteil hat dabei die koronare Herzkrankheit, gefolgt vom Schlaganfall und der Herzinsuffizienz. In westlichen Industrienationen dürfte jede zweite Person im Lauf ihres Lebens an Herz-Kreislauf-Krankheiten erkranken. Da das Risiko mit zunehmendem Alter zunimmt, wird die Anzahl von Herz-Kreislauf- Erkrankungen aufgrund der demografischen Entwicklung in Zukunft weiter ansteigen. In diesem Abschnitt betrachten wir zuerst die epidemiologische Bedeutung der Herz-Kreislauf-Krankheiten und schauen hier insbesondere auf die globale Bedeutung, die geografischen Unterschiede sowie die sich derzeit entwickelnden zeitlichen Trends. Anschließend erörtern wir, welche Risikofaktoren zur Entstehung von Herz-Kreislauf-Krankheiten beitragen und mit welchen präventiven Maßnahmen diese Erkrankungen zu verhindern sind. Objective: Description of a cat with ischemic muscle necrosis that suffered from cardiopulmonary arrest due to hyperkalemia. Pathogenesis, clinical signs and therapy of ischemic muscle necrosis are discussed and possible causes, symptoms and treatment of hyperkalemia are shown. Material and methods: case report of a four-year-old male castrated domestic shorthair cat. Results: The cat was successfully resuscitated and hyperkalemia was treated with different treatment modalities. Conclusion: Ischemic muscle necrosis can lead to severe live-threatening hyperkalemia which has to be anticipated, monitored and treated adequately. Aggressive fluid therapy might be responsible for a higher risk of hyperkalemia in predisposed cases. Clinical relevance: Potassium concentrations and acid-base disturbances must be closely monitored in patients with ischemic muscle necrosis

Die gegenwärtige Situation der deutschen Geschichtswissenschaft ist durch eine ungewöhnliche Offenheit gekennzeichnet. Die Krise der tradierten Großdeutungen schafft Raum für neue, konkurrierende Sinnkonstruktionen. Hinzu kommt eine Erweiterung der methodologischen Vielfalt. Vor allem die Zeitgeschichte ist von der Konkurrenz neuartiger 'Erinnerungsspezialisten' betroffen, die meist jenseits der professionalisierten Geschichtswissenschaft stehen. Auf diese Herausforderungen muss auch die Neugestaltung der Studiengänge reagieren. Generell sind die Chancen, die eine größere Pluralität bietet, zu begrüßen, doch ist auch das Risiko der Orientierungslosigkeit nicht zu unterschätzen. Für eine sinnvolle Neustrukturierung der Studiengänge sollte das Prinzip der Leistungsauswahl eine stärkere Bedeutung erhalten. Inhaltlich könnte das Thema einer "demokratisierten Geschichte der Deutschen in europäischer Perspektive" eine mögliche Richtung weisen.
