Reactive oxygen species (ROS) are produced by aerobic metabolism and react with biomolecules, such as lipids, proteins and DNA. In high concentration, they lead to oxidative stress. Among ROS, singlet oxygen (1O2) is one of the main ROS involved in oxidative stress and is one of the most reactive forms of molecular oxygen. The exposure of some dyes, such as methylene blue (MB) to light (MB+VL), is able to generate 1O2 and it is the principle involved in photodynamic therapy (PDT). 1O2 e other ROS have caused toxic and carcinogenic effects and have been associated with ageing, neurodegenerative diseases and cancer. Oxidative DNA damage is mainly repaired by base excision repair (BER) pathway. However, recent studies have observed the involvement of nucleotide excision repair (NER) factors in the repair of this type of injury. One of these factors is the Xeroderma Pigmentosum Complementation Group A (XPA) protein, which acts with other proteins in DNA damage recognition and in the recruitment of other repair factors. Moreover, oxidative agents such as 1O2 can induce gene expression. In this context, this study aimed at evaluating the response of XPA-deficient cells after treatment with photosensitized MB. For this purpose, we analyzed the cell viability and occurrence of oxidative DNA damage in cells lines proficient and deficient in XPA after treatment with MB+VL, and evaluated the expression of this enzyme in proficient and complemented cells. Our results indicate an increased resistance to treatment of complemented cells and a higher level of oxidative damage in the deficient cell lines. Furthermore, the treatment was able to modulate the XPA expression up to 24 hours later. These results indicate a direct evidence for the involvement of NER enzymes in the repair of oxidative damage. Besides, a better understanding of the effects of PDT on the induction of gene expression could be provided

Shrimp farming is one of the activities that contribute most to the growth of global aquaculture. However, this business has undergone significant economic losses due to the onset of viral diseases such as Infectious Myonecrosis (IMN). The IMN is already widespread throughout Northeastern Brazil and affects other countries such as Indonesia, Thailand and China. The main symptom of disease is myonecrosis, which consists of necrosis of striated muscles of the abdomen and cephalothorax of shrimp. The IMN is caused by infectious myonecrosis virus (IMNV), a non-enveloped virus which has protrusions along its capsid. The viral genome consists of a single molecule of double-stranded RNA and has two Open Reading Frames (ORFs). The ORF1 encodes the major capsid protein (MCP) and a potential RNA binding protein (RBP). ORF2 encodes a probable RNA-dependent RNA polymerase (RdRp) and classifies IMNV in Totiviridae family. Thus, the objective of this research was study the IMNV complete genome and encoded proteins in order to develop a system differentiate virus isolates based on polymorphisms presence. The phylogenetic relationship among some totivirus was investigated and showed a new group to IMNV within Totiviridae family. Two new genomes were sequenced, analyzed and compared to two other genomes already deposited in GenBank. The new genomes were more similar to each other than those already described. Conserved and variable regions of the genome were identified through similarity graphs and alignments using the four IMNV sequences. This analyze allowed mapping of polymorphic sites and revealed that the most variable region of the genome is in the first half of ORF1, which coincides with the regions that possibly encode the viral protrusion, while the most stable regions of the genome were found in conserved domains of proteins that interact with RNA. Moreover, secondary structures were predicted for all proteins using various softwares and protein structural models were calculated using threading and ab initio modeling approaches. From these analyses was possible to observe that the IMNV proteins have motifs and shapes similar to proteins of other totiviruses and new possible protein functions have been proposed. The genome and proteins study was essential for development of a PCR-based detection system able to discriminate the four IMNV isolates based on the presence of polymorphic sites

Worldwide, families Carangidae and Rachycentridae represent one of the groups most important commercial fish, used for food, and great potential for marine aquaculture. However, the genetic bases that can underpin the future cultivation of these species, cytogenetic between these aspects are very weak. The chromosomal patterns have provided basic data for the exploration of biotechnological processes aimed at handling chromosomal genetic improvement, such as induction of polyploidy, androgenesis and ginogenesis, as well as obtaining monosex stocks and interspecific hybridizations. This paper presents a comprehensive cytogenetic survey in 10 species, seven of the family Carangidae and the monotypic family Rachycentridae. Classical cytogenetic analysis and in situ mapping of multigene sequences were employed, and additionally for the genus Selene and morphotypes of Caranx lugubris, comparisons were made using geometric morphometrics. In general, conservative species exhibit a marked chromosome number (2n=48). Although present in large part, different karyotypic form, retain many characteristics typical of chromosomal Order Perciformes, the high number of elements monobrachyal, Ag-NORs/18S rDNA sites and heterochromatin simply reduced, preferably centromeric. The main mechanisms involved in karyotypic diversification are the pericentric inversions, with secondary action of centric fusions. In addition to physical mapping and chromosome detail for the species are presented and discussed patterns of intra-and interspecific diversity, cytotaxonomic markers. In vitro and in animal models, APE1, OGG1, and PARP-1 have been proposed as being involved with inflammatory response. In this work, we have investigated if the SNPs APE1 Asn148Glu, OGG1 Ser326Cys, and PARP-1 Val762Ala are associated to meningitis and also developed a system to enable the functional analysis of polymorphic proteins. Patients with bacterial meningitis (BM), aseptic meningitis (AM) and controls (non-infected) genotypes were investigated by PIRA-PCR or PCR-RFLP. DNA damages were detected in genomic DNA by Fpg treatment. IgG and IgA were measured from plasma and the cytokines and chemokines were measured from cerebrospinal fluid samples using Bio-Plex assays. The levels of NF-κB and c-Jun were measured in CSF by dot blot assays. A significant (P<0.05) increase in the frequency of APE1 148Glu allele in BM and AM patients was observed. A significant increase in the genotypes Asn/Asn in control group and Asn/Glu in BM group was also found. For the SNP OGG1 Ser326Cys, the genotype Cys/Cys was more frequent (P<0.05) in BM group. The frequency of PARP-1 Val/Val genotype was higher in control group (P<0.05). The occurrence of combined SNPs increased significantly in BM patients, indicating that these SNPs may be associated to the disease. Increasing in sensitive sites to Fpg was observed in carriers of APE1 148Glu allele or OGG1 326Cys allele, suggesting that SNPs affect DNA repair activity. Alterations in IgG production were observed in the presence of SNPs APE1Asn148Glu, OGG1Ser326Cys or PARP-1Val762Ala. Reductions in the levels ofIL-6, IL-1Ra, MCP-1/CCL2and IL-8/CXCL8 were observed in the presence of APE1148Glu allele in BM patients, however no differences were observed in the levels of NF-κB and c-Jun considering genotypes and analyzed groups. Using APE1 as model, a system to enable the analysis of cellular effects and functional characterization of polymorphic proteins was developed using strategies of cloning APE1 cDNA in pIRES2-EGFP vector, cellular transfection of the construction obtained, siRNA for endogenous APE1 and cellular cultures genotyping. In conclusion, we obtained evidences of an effect of SNPs in DNA repair genes on the regulation of immune response. This is a pioneering work in the field that shows association of BER variant enzymes with an infectious disease in human patients, suggesting that the SNPs analyzed may affect immune response and damage by oxidative stress level during brain infection. Considering these data, new approaches of functional characterization must be developed to better analysis and interactions of polymorphic proteins in response to this context

Perciformes are dominant in the marine environment, characterized as the largest and most diverse fish group. Some families, as Gerreidae, popularly known as silver jennies, carapebas, or mojarras have a high economic potential to marine fish farming, natural explotation and game fishing. Genetic information of these species are of fundamental importance for their management and production. Despite exist over 13,000 marine fish species described, only 2% were cytogenetically analyzed and less than 1% have some reproductive characteristics known. Induced breeding, cytogenetic characterization and cryopreservation of gametes, represent important areas in applied fish studies. In this project cytogenetic analyzes were performed to acess genetic aspects of Gerreidae species, distributed in coastal and estuarine regions of Northeast Brazil. Different methods for identifying chromosomal regions were employed using conventional techniques (Ag-NORs, C-banding), staining with base-specific fluorochromes (DAPI-CMA3), and physical mapping of ribosomal genes 18S and 5S rDNA, through hybridization in situ with fluorescent probes (FISH). The six species analyzed showed remarkable chromosome conservatism. The 18S and 5S ribosomal genes when analyzed in phylogenetic perspective demonstrate varied evolutionary dynamics, suggesting ocurrence of stasis process in some groups and greater dynamism in others. Double FISH with 18S and 5S probes showed both how efficient cytotaxonomic markers in the homogeneous karyotypes of this group of species. The karyotypic pattern identified in addition to the evolutionary aspects of karyotype, are suggestive of existence of low potential of post-zygotic barrier, prompting further research to prospect for artificial interspecific hybridization of these species of commercial importance

The general purpose of this work is to describe and analyse the financing phenomenon of crowdfunding and to investigate the relations among crowdfunders, project creators and crowdfunding websites. More specifically, it also intends to describe the profile differences between major crowdfunding platforms, such as Kickstarter and Indiegogo. The findings are supported by literature, gathered from different scientific research papers. In the empirical part, data about Kickstarter and Indiegogo was collected from their websites and also complemented with further data from other statistical websites. For finding out specific information, such as satisfaction of entrepreneurs from both platforms, a satisfaction survey was applied among 200 entrepreneurs from different countries. To identify the profile of users of the Kickstarter and of the Indiegogo platforms, a multivariate analysis was performed, using a Hierarchical Clusters Analysis for each platform under study. Descriptive analysis was used for exploring information about popularity of platforms, average cost and the most popular area of projects, profile of users and future opportunities of platforms. To assess differences between groups, association between variables, and answering to the research hypothesis, an inferential analysis it was applied. The results showed that the Kickstarter and Indiegogo are one of the most popular crowdfunding platforms. Both of them have thousands of users and they are generally satisfied. Each of them uses individual approach for crowdfunders. Despite this, they both could benefit from further improving their services. Furthermore, according the results it was possible to observe that there is a direct and positive relationship between the money needed for the projects and the money collected from the investors for the projects, per platform.

No decurso de um largo lapso de tempo, iniciado nos primórdios da presença humana no continente europeu, o território ocupado pelo concelho de Oeiras conservou testemunhos de sucessivas presenças humanas, as quais têm vindo a ser, nos últimos anos, objecto de um programa de pesquisas metódicas, conduzido pelo Centro de Estudos Arqueológicos do concelho de Oeiras, da Câmara Municipal de Oeiras. Deste modo, foram investigados diversos sítios de interesse arqueológico, do Paleolítico Inferior ao Período Romano, bem como de épocas ainda mais recentes, cujos resultados os transformam em elementos-chave para a cabal com preensão da evolução das sociedades humanas no espaço geográfico concelhio. Com efeito, os vestígios materiais que ali têm vindo a ser exumados, contribuem decisivamente para a caracterização económica, social e cultural das comunidades que se sucederam, ao longo dos milénios, na ocupação do solo oeirense, realidade que há pouco mais de uma década permanecia ainda quase totalmente por investigar.

Esta obra trata da sucessão cultural registada na Estremadura portuguesa desde a emergência das sociedades complexas do Calcolítico até à chegada dos Romanos, correspondendo a um lapso de tempo entre os finais do IV milénio e os finais do século II a.C. Embora corresponda apenas a intervalo temporal de aproximadamente três mil anos, é o que, no registo material da nossa Pré-História e Proto-História, se afigura mais rico e diversificado de informação, com o desenvolvimento e fixação de regionalismos culturais, que na Estremadura cunharam identidades próprias, as quais persistiram nalguns casos até época recente, no quotidiano dos seus habitantes. A percepção geral desta evolução, bem como as suas determinantes, é o primeiro, e talvez mais importante objectivo desta obra, a par de outros a seguir enunciados: – a génese dos povoados fortificados calcolíticos, em resultado da crescente intensificação económica e da especialização das produções – a Revolução dos Produtos Secundários (RPS), que decorreu ao longo de boa parte do III milénio a.C. – a par do crescimento demográfico, que determinou, por seu turno, a competição inter-grupos, com a consequente necessidade de fortificação; – a monumentalização / fortificação de alguns dos sítios habitados como expressão da coesão social da respectiva comunidade, acompanhada da emergência de diferenciações inter e intra-comunitárias, indício de diferenciação social, em crescente afirmação, decorrente do processo de desenvolvimento económico complexo, característico do Calcolítico; – as arquitecturas defensivas do III milénio a.C., como expressão pública indissociável da monumentalização acima referida: exemplos mais importantes no território estremenho, distribuição geográfica, características principais, semelhanças e diferenças; neste âmbito, importa conhecer as diversas teorias explicativas para o seu surgimento, desde o modelo difusionista e orientalista vigente em Portugal (dos anos 40 aos anos 70), passando pelo modelo indigenista (anos 80), até às formas difusionistas mitigadas, de expressão regional, dos finais da década de 80 em diante, e principais argumentos invocados; – a desarticulação do modelo de sociedade calcolítica, caracterizada pela concentração da população em sítios fortificados ou pelo menos implantados predominantemente em locais altos e defensáveis; – os moldes em que se processou a acentuação das influências mediterrâneas no decurso do Calcolítico (em especial na metade meridional do território): a generalização do comércio transregional calcolítico e a intensificação e especialização das produções, no quadro da Revolução dos Produtos Secundários (RPS), exemplificada pela exploração de jazidas cupríferas, como veículo de difusão de novas técnicas (metalurgia), matérias-primas exógenas (marfim) e artefactos ideotécnicos de características até então desconhecidas (generalização do culto da divindade feminina e correspondentes expressões simbólicas, algumas de âmbito estritamente regional), acompanhada da difusão, de Sul para Norte, de novas arquitecturas funerárias (tholoi); – sobre o Campaniforme, fenómeno cultural com identidade própria da fase média e tardia do Calcolítico estremenho, serão discutidas as características e cronologia da sua emergência, na Estremadura (um dos pólos mais importantes, a nível europeu) no quadro da sociedade calcolítica pré-existente: tipo de povoamento e de necrópoles, bem como as relações estabelecidas com as comunidades de tradição cultural mais antiga; o faseamento interno do “fenómeno”, com base nas diferenças identificadas no registo material (em particular a tipologia das cerâmicas); e principais tipos artefactuais que o integram. O campaniforme deverá ser entendido como uma expressão material específica, associada a um novo tipo de povoamento, que resultou do decréscimo do interesse oferecido pelos sítios fortificados edificados no início do Calcolítico. Neste sentido, corresponde a período de transição para a Idade do Bronze: existem argumentos, com base no registo arqueológico (jóias de ouro, artefactos de prestígio) que ilustram o incremento do processo de diferenciação social, então verificado, ao contrário do que uma abordagem mais superficial, com base simplesmente no reordenamento demográfico, faria supor; – o registo arqueológico do Bronze Pleno configura a acentuação dos regionalismos, apesar de similitudes do sistema de povoamento face ao período imediatamente anterior, o que indicia realidades socioeconómicas comparáveis. Importa, assim, conhecer as principais características dos escassos povoados identificados, bem como a organização social a ele subjacente, a partir dos testemunhos arqueológicos conhecidos, incluindo os de carácter funerário; – segue-se o Bronze Final, período dominado pela plena afirmação do comércio transregional atlântico-mediterrâneo, favorecido pela própria realidade geográfica do território português. Devem valorizar-se os testemunhos materiais desse período e as respectivas balizas cronológicas: Assim, deverão os leitores ficar familiarizados com as produções de carácter atlântico, como as armas, objectos utilitários e respectivas tipologias e com as de cunho mediterrâneo (com destaque para objectos de indumentária e de carácter cultual, embora estes últimos quase se desconheçam na área estremenha), cujo comércio e difusão foi suportado pela existência de solidariedades económicas transregionais, baseadas em prováveis pactos formalmente estabelecidos entre comunidades vizinhas. Os respectivos territórios, de norte a sul do País, apresentar-se-iam cada vez melhor delimitados; o mesmo deverá ter-se verificado na Estremadura. A caracterização da respectiva economia será, por isso, objecto da análise e discussão; embora de base agro-pastoril (com importância evidente na Estremadura dadas as características dos solos e a quase inexistência de minérios de cobre ou de estanho), a produção de peças metálicas de bronze assumiu importância crescente, como se conclui pelas ocorrências conhecidas. O reforço e a consolidação das elites então verificada, eram necessários para a boa gestão de grandes povoados muralhados que despontam no Bronze Final; na Estremadura, embora os testemunhos de tais centros demográficos não sejam particularmente evidentes, no fim da Idade do Bronze desponta um vigoroso povoamento de altura; seria a partir desses locais que as elites da época, de cunho guerreiro, administrariam territórios bem delimitados. Também a existência de outros testemunhos arqueológicos são concorrentes para a percepção da realidade social: as jóias auríferas, tornadas então relativamente frequentes, deixam transparecer influências ora atlânticas ora mediterrâneas, por vezes reunidas numa única peça (técnicas e tipologias decorativas), expressivas das correntes culturais que, então, se faziam sentir na Estremadura; também as armas, são testemunho da afirmação das elites guerreiras, encontrando-se representadas por exemplares cujas principais características devem ser conhecidas. As diversas práticas funerárias, apesar de escassamente representadas, revelam influências continentais (cremação e campos de urnas, já fora da área estremenha, mas dela próxima: caso dos campos de urnas de Tanchoal e de Meijão, Alpiarça) e mediterrâneas (inumações na tholos da Roça do Casal do Meio, Sesimbra), que traduzem um mosaico cultural complexo, reforçando a ideia de se tratar de região receptora de influxos culturais de diversas áreas geográficas em simultâneo: é, no essencial, a comprensão global desta realidade, a um tempo económica, social e cultural, coroando um longo processo de diferenciação social, por um lado e, por outro, de intensificação económica e interacção cultural, que lhe está subjacente, que deverá ter-se presente. Por último, segue-se o estudo e caracterização das principais estações e materiais da Idade do Ferro, de início (I Idade do Ferro) profundamente marcadas pela presença, directa ou indirecta, de colonizadores fenícios; depois, pelos comerciantes de origem púnica (II Idade do Ferro) e, enfim, pelos exércitos itálicos. Trata-se, em suma, de processo de características próprias, sempre determinado pelas influências mediterrâneas, largamente dominantes face às originárias do interior peninsular, as quais cunharam uma realidade cultural com características próprias, que persistiu no decurso da dominação romana.

heterogeneous catalyst such as a silicoaluminophosphate, molecular sieve with AEL (Aluminophosphate eleven) structure such as SAPO-11, was synthesized through the hydrothermal method starting from silica, pseudoboehmite, orthophosphoric acid (85%) and water, in the presence of a di-isopropylamine organic template. For the preparation of SAPO-11 in a dry basis it was used as reactants: DIPA; H3PO4; SiO4; Pseudoboehmite and distilled water. The crystallization process occurred when the reactive hydrogel was charged into a vessel and autoclaved at 200ºC for a period of 72 hours under autogeneous pressure. The obtained material was washed, dried and calcined to remove the molecular sieves of DIPA. The samples were characterized by X-ray diffraction (XRD), scanning electron microscopy (SEM), infrared spectroscopy (FT-IR), nitrogen adsorption (BET) and thermal analysis (TG/DTG). The acidic properties were determined using adsorption of nbutylamine followed by programmed thermodessorption. This method revealed that SAPO-11 shows an acidity that ranges from weak to moderate. However, a small quantity of strong acid sites could be detected there. The deactivation of the catalysts was conducted by artificial coking followed by the cracking of the n-hexane in a fixed bed with a continuous flow micro-reactor coupled on line to a gas chromatograph. The main products obtained were: ethane, propane, isobutene, n-butane, n-pentane and isopentane. The mixed metal oxides constitute an important class of catalytic materials widely investigated in different fields of applications. Studies of rare earth nickelates have been carried by several researchers in order to investigate the structural stability afforded by oxide formed and the existence of catalytic properties at room temperature. So, this study aims synthesize the nanosized catalyst of nickelate of lanthanum doped with strontium (La(1-x)SrxNiO4-d; x = 0,2 and 0,3), through the Pechini method and your characterization for subsequent application in the desulfurization of thiophene reaction. The precursor solutions were calcined at 300ºC/2h for pyrolysis of polyester and later calcinations occurred at temperatures of 500 - 1000°C. The resulting powders were characterized by thermogravimetric analysis (TG / DTG), surface area for adsorption of N2 by BET method, X-ray diffraction (XRD), scanning electron microscopy (HR_SEM) and spectrometry dispersive energy (EDS). The results of XRD had show that the perovskites obtained consist of two phases (LSN and NiO) and from 700ºC have crystalline structure. The results of SEM evidenced the obtainment of nanometric powders. The results of BET show that the powders have surface area within the range used in catalysis (5-50m2/g). The characterization of active sites was performed by reaction of desulfurization of thiophene at room temperature and 200ºC, the relation F/W equal to 0,7 mol h-1mcat -1. The products of the reaction were separated by gas chromatography and identified by the selective detection PFPD sulfur. All samples had presented conversion above 95%

O concelho de Montemor-o-Novo é rico em vestígios arqueológicos de várias cronologias, pelo que a época romana não é excepção. Durante muito tempo pouco se conhecia deste período cronológico, particularmente a forma do seu povoamento. Durante os anos 60 do século XX, foram identificadas e escavadas algumas estruturas deste período, nomeadamente a Fonte do Prior, o Curral dos Cães e o Cabeço do Ceivo. No entanto, foi durante as obras da Barragem dos Minutos que começamos a ter uma nova perceção de como seria o povoamento romano neste território. Sítios como estes têm vindo, ao longo do tempo, a ser colocados de parte quando se falava sobre a romanização dos campos por se considerar que essa ocupação era exclusiva das luxuosas villae e por se pensar que estes sítios fariam parte das villae. Neste trabalho iremos ver o resultado das intervenções realizados na Barragem dos Minutos, bem como as realizadas no regolfo da Barragem de Alqueva com o objectivo de tentar perceber estas novas realidades do povoamento romano; Abstract: The Montemor-o-Novo municipality is rich in archaeological remains of various chronologies, so the Roman era is no exception. But for a long time little was known of this chronological period and especially the shape of your settlement. During the 60s of the twentieth century, they have been identified and excavated some structures of this period, namely the Fonte do Prior, the Curral dos Cães and the Cabeço Ceivo. But it was during the construction of the dam of Minutos we started to have a new perception of how would the Roman settlement in this territory. Sites like these have been, over time, to be put aside when talking about the romanization of the fields in Roman times as it was considered that this occupation was exclusive of villae luxurious and think that these sites would be part of the villae. In this work we will see the result of the work done at the dam of the Minutos, as well as the work carried out in the surroundings of the Alqueva Dam and try to understand these new realities of the Roman settlement.

This dissertation focuses on the theme of urban revitalization in historical centers. It starts from a survey of international and national concepts and experiences about the subject, in special, the historical center of Ribeira, in Natal, Brazil. It gives more attention to local management and it is dedicated to the study of objectives and actions developed, basically, by two segments that manage works in the district. Companhia Docas do Rio Grande do Norte aims at the economical development of the state and it implies the enlargement of capacity of loading and unloading of the harbor of Natal and the Natal city counsil aims at urban preservation and revitalization. It was analysed speeches ans projects related to Ribeira and it was discovered that Natal has done very little to revitalize its historical center in the last two dacades. It analyses what seemed, or at least, should be a conflict of interests between incompatible projects, revealing both conciliatory proceedings and connivance between the participants in decision making. The conclusion directs attentions to the threat to historical building in the city and the importance of enhancing discussion about the future of our historical sites
