The occurrence of chaotic instabilities is investigated in the swing motion of a dragline bucket during operation cycles. A dragline is a large, powerful, rotating multibody system utilised in the mining industry for removal of overburden. A simplified representative model of the dragline is developed in the form of a fundamental non-linear rotating multibody system with energy dissipation. An analytical predictive criterion for the onset of chaotic instability is then obtained in terms of critical system parameters using Melnikov's method. The model is shown to exhibit chaotic instability due to a harmonic slew torque for a range of amplitudes and frequencies. These chaotic instabilities could introduce irregularities into the motion of the dragline system, rendering the system difficult to control by the operator and/or would have undesirable effects on dragline productivity and fatigue lifetime. The sufficient analytical criterion for the onset of chaotic instability is shown to be a useful predictor of the phenomenon under steady and unsteady slewing conditions via comparisons with numerical results. In the English literature, facial approximation methods have been commonly classified into three types: Russian, American, or Combination. These categorizations are based on the protocols used, for example, whether methods use average soft-tissue depths (American methods) or require face muscle construction (Russian methods). However, literature searches outside the usual realm of English publications reveal key papers that demonstrate that the Russian category above has been founded on distorted views. In reality, Russian methods are based on limited face muscle construction, with heavy reliance on modified average soft-tissue depths. A closer inspection of the American method also reveals inconsistencies with the recognized classification scheme. This investigation thus demonstrates that all major methods of facial approximation depend on both face anatomy and average soft-tissue depths, rendering common method classification schemes redundant. The best way forward appears to be for practitioners to describe the methods they use (including the weight each one gives to average soft-tissue depths and deep face tissue construction) without placing them in any categorical classificatory group or giving them an ambiguous name. The state of this situation may need to be reviewed in the future in light of new research results and paradigms.

Documenting the history of settlement in Hawaii during the last few Centuries before European contact, is crucial to charting the evolution of the most complex chiefdom in Polynesia. It is precisely this period that Hawaii. and many Polynesian Societies, Underwent their most rapid changes in political, economic and social organisation. The last similar to 500 to 300 years in the C-14 calibration curve is problematic with wide fluctuations Often rendering large age spans that do not precisely date single events, especially troubling with a culture-historical record of similar to 1000 years duration. Here we present in extremely high precision Th-230 chronology for archaeologically constrained coral samples from a range of occupancy sites. Our high precision dates allow the time of site use to be clearly demonstrated. They also provide the first dates for habitation sites in Hawaii that clearly show contemporaneous occupation-the major problem in settlement pattern archaeology. We demonstrate that two sites were occupied within the same year. Our refined chronology, provides new and exciting oppurtunities for tracking sociopolitical and economic developments during the last few centuries-the crucial period in the evolution and transformation of Polynesian societies. Climate change is expected to affect the high latitudes first and most severely, rendering Antarctica one of the most significant baseline environments for the study of global climate change. The indirect effects of climate warming, including changes to the availability of key environmental resources, such as water and nutrients, are likely to have a greater impact upon continental Antarctic terrestrial ecosystems than the effects of fluctuations in temperature alone. To investigate the likely impacts of a wetter climate on Antarctic terrestrial communities a multiseason, manipulative field experiment was conducted in the floristically important Windmill Islands region of East Antarctica. Four cryptogamic communities (pure bryophyte, moribund bryophyte, crustose and fructicose lichen-dominated) received increased water and/or nutrient additions over two consecutive summer seasons. The increased water approximated an 18% increase in snow melt days (0.2 degrees C increase in temperature), while the nutrient addition of 3.5g Nm(-2) yr(-1) was within the range of soil N in the vicinity. A range of physiological and biochemical measurements were conducted in order to quantify the community response. While an overall increase in productivity in response to water and nutrient additions was observed, productivity appeared to respond more strongly to nutrient additions than to water additions. Pure bryophyte communities, and lichen communities dominated by the genus Usnea, showed stronger positive responses to nutrient additions, identifying some communities that may be better able to adapt and prosper under the ameliorating conditions associated with a warmer, wetter future climate. Under such a climate, productivity is overall likely to increase but some cryptogamic communities are likely to thrive more than others. Regeneration of moribund bryophytes appears likely only if a future moisture regime creates consistently moist conditions.

Many viruses including HIV, hepatitis C and hepatitis B, have an outer lipid envelope which maintains inserted viral peptides in the “correct” functional conformation and orientation. Disruption of the lipid envelope by most solvents destroys infectivity and often results in a loss of antigenicity. This communication outlines a novel approach to viral inactivation by specific solvent delipidation which modifies the whole virion rendering it non-infective, but antigenic. Duck hepatitis B virus (DHBV) was delipidated using a diisopropylether (DIPE) and butanol mixture and residual infectivity tested by inoculation into day-old ducks. Delipidation completely inactivated the DHBV (p < 0.001). Delipidated DHBV was then used to vaccinate ducks. Three doses of delipidated DHBV induced anti-DHBs antibody production and prevented high dose challenge infection in five out of six ducks. In comparison, five of six ducks vaccinated with undelipidated DHBV and four of four ducks vaccinated with glutaraldehyde inactivated DHBV were unprotected (p < 0.05). Although this solvent system completely inactivated DHBV, viral antigens were retained in an appropriate form to induce immunity. Delipidation of enveloped viruses with specific organic solvents has potential as the basis for development of vaccines.

This study evaluates the pro-inflammatory response to the thermoplastic biopolymer poly(3-hydroxybutyrate-co-3-hydroxyvalerate) (PHBV) through the analysis of cellular responses in vitro. The murine macrophage RAW264.7 cell line was cultured on solvent cast PHBV films, which was found to induce pro-inflammatory activity that required direct contact between the material and the macrophages. The identity of the pro-inflammatory stimulus was determined by culturing bone marrow-derived macrophages from bacterial lipopolysaccharide (LPS) hyporesponsive C3H/HeJ mice and CpG non-responsive TLR9-/- mice on PHBV. The lack of a pro-inflammatory response by the C3H/HeJ cells indicates that the pro-inflammatory agent present within PHBV is predominately LPS while the TLR9-/- macrophages confirmed that CpG-containing bacterial DNA is unlikely to contribute to the activity. A series of purification procedures was evaluated and one procedure was developed that utilized hydrogen peroxide treatment in solution. The optimized purification was found to substantially reduce the pro-inflammatory response to PHBV without adversely affecting either the molecular structure or molecular weight of the material thereby rendering it more amenable for use as a biomaterial in vivo. Morphology, occlusal surface topography, macrowear, and microwear features of parrotfish pharyngeal teeth were investigated to relate microstructural characteristics to the function of the pharyngeal mill using scanning electron microscopy of whole and sectioned pharyngeal jaws and teeth. Pharyngeal tooth migration is anterior in the lower jaw (fifth ceratobranchial) and posterior in the upper jaw (paired third pharyngobranchials), making the interaction of occlusal surfaces and wear-generating forces complex. The extent of wear can be used to define three regions through which teeth migrate: a region containing newly erupted teeth showing little or no wear; a midregion in which the apical enameloid is swiftly worn; and a region containing teeth with only basal enameloid remaining, which shows low to moderate wear. The shape of the occlusal surface alters as the teeth progress along the pharyngeal jaw, generating conditions that appear suited to the reduction of coral particles. It is likely that the interaction between these particles and algal cells during the process of the rendering of the former is responsible for the rupture of the latter, with the consequent liberation of cell contents from which parrotfish obtain their nutrients.

A temática sobre redes organizacionais tem ocupado um lugar importante no campo de estudos da ciência social aplicada à administração dado o grau de interesse dos pesquisadores compreenderem o fenômeno da coopetição nos mercados e sua relação entre os atores na economia. A análise proposta neste estudo apresenta o sistema de consorciamento de municípios tendo como ponto central da rede o CIS/AMUNPAR em torno das políticas públicas de saúde. A proposta foi de entender quais foram os ganhos/conquistas que o consórcio obteve a partir dos conceitos da teoria de redes. Dado esse contexto, buscou-se revisitar nesta pesquisa a memória social do CIS/AMUNPAR caracterizado como uma rede de cooperação intergovernamental, sendo apoiada pelo SUS como parte integrante das políticas públicas de saúde no período de democratização e municipalização do setor de saúde no Brasil. Os Consórcios Intermunicipais de Saúde assumiram a responsabilidade de atendimento das especialidades no Estado, deixando de serem apenas um mero coadjuvante e passando a ser um ator social principal na prestação de serviços de saúde no país. A partir do processo de redemocratização política do país, os consórcios foram criados objetivando a aproximação dos municípios com os usuários do sistema SUS. Trata-se portanto, de uma pesquisa metodologicamente calcada na abordagem qualitativa, exploratória e descritiva, valendo-se da história oral e análise documental. Os procedimentos de análise dos dados foram baseados no modelo analítico geral e bibliografia fundamentada. Os dados permitiram compreender que o CIS/AMUNPAR passa por uma fragmentação na parceria estabelecida na rede em sua gênese e que precisa unir esforços para planejar suas ações em conjunto, assim como se caracteriza uma rede cooperativa efetiva. A pesquisa ainda revelou que isoladamente os municípios não conseguiriam oferecer toda a assistência necessária e requerida pela União a partir do processo de descentralização da saúde e que a estratégia mais adequada para a região do noroeste do Paraná seria na forma de consorciamento dos 28 municípios que compreendiam esta região.

O principal objetivo deste estudo foi identificar e analisar qual o impacto da tecnologia da informação (TI) na satisfação das pessoas em uma empresa pública. Este estudo pretendeu abordar o assunto de forma a concentrar-se na investigação, com um estudo de caso em uma empresa pública de saneamento ambiental da região do grande ABC paulista, o SEMASA, com o intuito de avaliar as mudanças nas rotinas operacionais dos diversos departamentos da empresa e o impacto causado pela TI na satisfação das pessoas envolvidas. Com base na literatura sobre trabalho e sociedade na era da globalização e empresas públicas, foram analisados inicialmente os motivos que podem contribuir para as pessoas buscarem ingressar na carreira pública. Esse estudo verificou a importância da adoção de ferramentas de tecnologia nos processos produtivos e na prestação de serviços das organizações. Verificou também, que estimular os funcionários de empresas públicas a criarem mecanismos para facilitarem seus trabalhos, através de ferramentas de TI atualizadas, pode ser benéfico, ao mesmo tempo em que pode proporcionar condições destes adquirirem novas capacitações profissionais com a aquisição de novos conhecimentos tecnológicos. Os resultados obtidos apontaram para um significativo aumento na satisfação das pessoas envolvidas sendo estas estimuladas a estarem em constante aperfeiçoamento, através da realização de novos treinamentos e cursos de tecnologia.(AU)

Este trabalho discutiu o desempenho do Centro Nacional de Pesquisa em Energia e Materiais CNPEM, que deu início às suas atividades como Associação Brasileira de Tecnologia de Luz Síncrotron - ABTLuS, Organização Social (OS), entidade da sociedade civil, sem fins lucrativos, disciplinada pela Lei 9.637 de 15 de maio de 1998, para gerir organismos públicos mediante a assinatura de contrato de gestão baseado em metas a serem cumpridas. O modelo nasceu concomitantemente com a criação do Ministério da Administração e Reforma do Estado MARE, na gestão do ministro Luiz Carlos Bresser Pereira, no governo do presidente Fernando Henrique Cardoso - FHC, tendo como princípio básico a prestação de serviços sociais, nas áreas de cultura, educação, pesquisa científica, proteção e preservação do meio ambiente e saúde, na expectativa de que ele pudesse prestar melhores serviços do que as empresas estatais e as entidades da administração direta. Hoje, dez anos se passaram e o CNPEM continua prestando serviços na área de pesquisa científica ao CNPq e ao MCT, apesar de não haver apoio para a criação de novas OS na esfera do governo federal, o que indicou ameaças ao seu ciclo de vida. A pesquisa desenvolveu-se através de estudo descritivo-exploratório, interpretando dados por intermédio do processo de hermenêutica para interpretação de dados retirados dos documentos institucionais, com conclusão favorável à continuidade do modelo, tendo em vista que a pesquisa demonstra que ele é legal, legítimo e competente no cumprimento das metas e missão, bem como está em plena forma física, sem ameaças ao seu ciclo de vida, demonstrando que tem muito a oferecer para a ciência brasileira e, como não poderia deixar de ser, a milhares de usuários espalhados pelo Brasil e o mundo.

As defined by the European Union, “ ’Nanomaterial’ (NM) means a natural, incidental or manufactured material containing particles, in an unbound state or as an aggregate or agglomerate, where, for 50 % or more of the particles in the number size distribution, one or more external dimensions is in the size range 1 nm-100 nm ” (2011/696/UE). Given their peculiar physico-chemical features, nanostructured materials are largely used in many industrial fields (e.g. cosmetics, electronics, agriculture, biomedical) and their applications have astonishingly increased in the last fifteen years. Nanostructured materials are endowed with very large specific surface area that, besides making them very useful in many industrial processes, renders them very reactive towards the biological systems and, hence, potentially endowed with significant hazard for human health. For these reasons, in recent years, many studies have been focused on the identification of toxic properties of nanostructured materials, investigating, in particular, the mechanisms behind their toxic effects as well as their determinants of toxicity. This thesis investigates two types of nanostructured TiO2 materials, TiO2 nanoparticles (NP), which are yearly produced in tonnage quantities, and TiO2 nanofibres (NF), a relatively novel nanomaterial. Moreover, several preparations of MultiWalled Carbon Nanotubes (MWCNT), another nanomaterial widely present in many products, are also investigated.- Although many in vitro and in vivo studies have characterized the toxic properties of these materials, the identification of their determinants of toxicity is still incomplete. The aim of this thesis is to identify the structural determinants of toxicity, using several in vitro models. Specific fields of investigation have been a) the role of shape and the aspect ratio in the determination of biological effects of TiO2 nanofibres of different length; b) the synergistic effect of LPS and TiO2 NP on the expression of inflammatory markers and the role played therein by TLR-4; c) the role of functionalization and agglomeration in the biological effects of MWCNT. As far as biological effects elicited by TiO2 NF are concerned, the first part of the thesis demonstrates that long TiO2 nanofibres caused frustrated phagocytosis, cytotoxicity, hemolysis, oxidative stress and epithelial barrier perturbation. All these effects were mitigated by fibre shortening through ball-milling. However, short TiO2 NF exhibited enhanced ability to activate acute pro-inflammatory effects in macrophages, an effect dependent on phagocytosis. Therefore, aspect ratio reduction mitigated toxic effects, while enhanced macrophage activation, likely rendering the NF more prone to phagocytosis. These results suggest that, under in vivo conditions, short NF will be associated with acute inflammatory reaction, but will undergo a relatively rapid clearance, while long NF, although associated with a relatively smaller acute activation of innate immunity cells, are not expected to be removed efficiently and, therefore, may be associated to chronic inflammatory responses. As far as the relationship between the effects of TiO2 NP and LPS, investigated in the second part of the thesis, are concerned, TiO2 NP markedly enhanced macrophage activation by LPS through a TLR-4-dependent intracellular pathway. The adsorption of LPS onto the surface of TiO2 NP led to the formation of a specific bio-corona, suggesting that, when bound to TiO2 NP, LPS exerts a much more powerful pro-inflammatory effect. These data suggest that the inflammatory changes observed upon exposure to TiO2 NP may be due, at least in part, to their capability to bind LPS and, possibly, other TLR agonists, thus enhancing their biological activities. Finally, the last part of the thesis demonstrates that surface functionalization of MWCNT with amino or carboxylic groups mitigates the toxic effects of MWCNT in terms of macrophage activation and capability to perturb epithelial barriers. Interestingly, surface chemistry (in particular surface charge) influenced the protein adsorption onto the MWCNT surface, allowing to the formation of different protein coronae and the tendency to form agglomerates of different size. In particular functionalization a) changed the amount and the type of proteins adsorbed to MWCNT and b) enhanced the tendency of MWCNT to form large agglomerates. These data suggest that the different biological behavior of functionalized and pristine MWCNT may be due, at least in part, to the different tendency to form large agglomerates, which is significantly influenced by their different capability to interact with proteins contained in biological fluids. All together, these data demonstrate that the interaction between physico-chemical properties of nanostructured materials and the environment (cells + biological fluids) in which these materials are present is of pivotal importance for the understanding of the biological effects of NM. In particular, bio-persistence and the capability to elicit an effective inflammatory response are attributable to the interaction between NM and macrophages. However, the interaction NM-cells is heavily influenced by the formation at the nano-bio interface of specific bio-coronae that confer a novel biological identity to the nanostructured materials, setting the basis for their specific biological activities.

Inspired by the idea of safe citizenship this article queries the possibilities of safety in an age of securitization. It challenges the cosmopolitan worldview and its iteration of a global cosmopolitan citizen. It champions an account of affective citizenship, narration and attends to the trauma of exile. It offers an account of exile before suggesting an institutional design premised on politicization. This design, it is argued, facilitates moments of storytelling fostering individual empowerment. This unorthodox rendering of agency allows the traumatized exile to negotiate the world as it is, not as it could be, as a potential ‘safe’ citizen.

Adjuvants are often composed of different constituents that can be divided into two groups based on their primary activity: the delivery system which carries and presents the vaccine antigen to antigen-presenting cells, and the immunostimulator that activates and modulates the ensuing immune response. Herein, we have investigated the importance of the delivery system and in particular its physical characteristics by comparing the delivery properties of two lipids which differ only in the degree of saturation of the acyl chains, rendering the liposomes either rigid (DDA, dimethyldioctadecylammonium) or highly fluid (DODA, dimethyldioleoylammonium) at physiological temperature. We show that these delivery systems are remarkably different in their ability to prime a Th1-directed immune response with the rigid DDA-based liposomes inducing a response more than 100 times higher compared to that obtained with the fluid DODA-based liposomes. Upon injection with a vaccine antigen, DDA-based liposomes form a vaccine depot that results in a continuous attraction of antigen-presenting cells that engulf a high amount of adjuvant and are subsequently efficiently activated as measured by an elevated expression of the co-stimulatory molecules CD40 and CD86. In contrast, the fluid DODA-based liposomes are more rapidly removed from the site of injection resulting in a lower up-regulation of co-stimulatory CD40 and CD86 molecules on adjuvant-positive antigen-presenting cells. Additionally, the vaccine antigen is readily dissociated from the DODA-based liposomes leading to a population of antigen-presenting cells that are antigen-positive but adjuvant-negative and consequently are not activated. These studies demonstrate the importance of studying in vivo characteristics of the vaccine components and furthermore show that physicochemical properties of the delivery system have a major impact on the vaccine-induced immune response. © 2012 Elsevier B.V. Blurred edges appear sharper in motion than when they are stationary. We proposed a model of this motion sharpening that invokes a local, nonlinear contrast transducer function (Hammett et al, 1998 Vision Research 38 2099-2108). Response saturation in the transducer compresses or 'clips' the input spatial waveform, rendering the edges as sharper. To explain the increasing distortion of drifting edges at higher speeds, the degree of nonlinearity must increase with speed or temporal frequency. A dynamic contrast gain control before the transducer can account for both the speed dependence and approximate contrast invariance of motion sharpening (Hammett et al, 2003 Vision Research, in press). We show here that this model also predicts perceived sharpening of briefly flashed and flickering edges, and we show that the model can account fairly well for experimental data from all three modes of presentation (motion, flash, and flicker). At moderate durations and lower temporal frequencies the gain control attenuates the input signal, thus protecting it from later compression by the transducer. The gain control is somewhat sluggish, and so it suffers both a slow onset, and loss of power at high temporal frequencies. Consequently, brief presentations and high temporal frequencies of drift and flicker are less protected from distortion, and show greater perceptual sharpening.

The inclusion of high-level scripting functionality in state-of-the-art rendering APIs indicates a movement toward data-driven methodologies for structuring next generation rendering pipelines. A similar theme can be seen in the use of composition languages to deploy component software using selection and configuration of collaborating component implementations. In this paper we introduce the Fluid framework, which places particular emphasis on the use of high-level data manipulations in order to develop component based software that is flexible, extensible, and expressive. We introduce a data-driven, object oriented programming methodology to component based software development, and demonstrate how a rendering system with a similar focus on abstract manipulations can be incorporated, in order to develop a visualization application for geospatial data. In particular we describe a novel SAS script integration layer that provides access to vertex and fragment programs, producing a very controllable, responsive rendering system. The proposed system is very similar to developments speculatively planned for DirectX 10, but uses open standards and has cross platform applicability. © The Eurographics Association 2007.
