A series of 3(2H)-furanones, based on side-chain modifications of a parent 3(2H)-furanone, was synthesized in good yield. The parent compound was prepared by hydrogenolysis, and subsequent acid hydrolysis, of isoxazole derivatives. The isoxazole was prepared by a [3+2] 1,3-dipolar cycloaddition reaction between 3-butyn-2-ol and nitrile oxide.

Joseph Barbosa de Sáa (? – 1775), mais conhecido por seus escritos sobre a história do estado do Mato Grosso, Brasil, completou em 1769 um volumoso (408 fólios) e erudito manuscrito, intitulado “Dialogos geograficos, coronologicos, polliticos e naturais”, que nunca foi publicado na íntegra. Esse manuscrito está depositado na Biblioteca Pública do Porto (manuscrito no. 235), em Portugal. Dez capítulos desse manuscrito tratam dos produtos naturais do Brasil (acima de mil, quase a metade sendo animais), observados por Sáa ao longo da costa do Rio de Janeiro, em São Paulo, sul de Goiás e especialmente no Mato Grosso, sendo a primeira monografia sobre a história natural deste último estado. Esses capítulos são aqui transcritos e comentados.

Abstract Background Many important toxins and antibiotics are produced by non-ribosomal biosynthetic pathways. Microcystins are a chemically diverse family of potent peptide toxins and the end-products of a hybrid NRPS and PKS secondary metabolic pathway. They are produced by a variety of cyanobacteria and are responsible for the poisoning of humans as well as the deaths of wild and domestic animals around the world. The chemical diversity of the microcystin family is attributed to a number of genetic events that have resulted in the diversification of the pathway for microcystin assembly. Results Here, we show that independent evolutionary events affecting the substrate specificity of the microcystin biosynthetic pathway have resulted in convergence on a rare [D-Leu1] microcystin-LR chemical variant. We detected this rare microcystin variant from strains of the distantly related genera Microcystis, Nostoc, and Phormidium. Phylogenetic analysis performed using sequences of the catalytic domains within the mcy gene cluster demonstrated a clear recombination pattern in the adenylation domain phylogenetic tree. We found evidence for conversion of the gene encoding the McyA2 adenylation domain in strains of the genera Nostoc and Phormidium. However, point mutations affecting the substrate-binding sequence motifs of the McyA2 adenylation domain were associated with the change in substrate specificity in two strains of Microcystis. In addition to the main [D-Leu1] microcystin-LR variant, these two strains produced a new microcystin that was identified as [Met1] microcystin-LR. Conclusions Phylogenetic analysis demonstrated that both point mutations and gene conversion result in functional mcy gene clusters that produce the same rare [D-Leu1] variant of microcystin in strains of the genera Microcystis, Nostoc, and Phormidium. Engineering pathways to produce recombinant non-ribosomal peptides could provide new natural products or increase the activity of known compounds. Our results suggest that the replacement of entire adenylation domains could be a more successful strategy to obtain higher specificity in the modification of the non-ribosomal peptides than point mutations.

Abstract Background The current treatments for anxiety disorders and depression have multiple adverse effects in addition to a delayed onset of action, which has prompted efforts to find new substances with potential activity in these disorders. Citrus aurantium was chosen based on ethnopharmacological data because traditional medicine refers to the Citrus genus as useful in diminishing the symptoms of anxiety or insomnia, and C. aurantium has more recently been proposed as an adjuvant for antidepressants. In the present work, we investigated the biological activity underlying the anxiolytic and antidepressant effects of C. aurantium essential oil (EO), the putative mechanism of the anxiolytic-like effect, and the neurochemical changes in specific brain structures of mice after acute treatment. We also monitored the mice for possible signs of toxicity after a 14-day treatment. Methods The anxiolytic-like activity of the EO was investigated in a light/dark box, and the antidepressant activity was investigated in a forced swim test. Flumazenil, a competitive antagonist of benzodiazepine binding, and the selective 5-HT1A receptor antagonist WAY100635 were used in the experimental procedures to determine the mechanism of action of the EO. To exclude false positive results due to motor impairment, the mice were submitted to the rotarod test. Results The data suggest that the anxiolytic-like activity observed in the light/dark box procedure after acute (5 mg/kg) or 14-day repeated (1 mg/kg/day) dosing was mediated by the serotonergic system (5-HT1A receptors). Acute treatment with the EO showed no activity in the forced swim test, which is sensitive to antidepressants. A neurochemical evaluation showed no alterations in neurotransmitter levels in the cortex, the striatum, the pons, and the hypothalamus. Furthermore, no locomotor impairment or signs of toxicity or biochemical changes, except a reduction in cholesterol levels, were observed after treatment with the EO. Conclusion This work contributes to a better understanding of the biological activity of C. aurantium EO by characterizing the mechanism of action underlying its anxiolytic-like activity.

The interest in the use of evaporative light scattering detector (ELSD) for the analysis of different classes of natural products has grown over the years. This is because this detector has become an excellent alternative compared to other types of detectors, such as the refractive index detector and the ultraviolet (UV) detector. This review describes the basic principles of ELSD functioning and discusses the advantages and disadvantages in using an ELSD for the analysis of organic compounds. Additionaly, an overview, covering the last 23 years, of ELSD applications in natural products analysis (saponins, terpenes, carbohydrates, glycosides, alkaloids, steroids, flavonoids, peptides, polyketides, coumarins and iridoids) is presented and discussed.

This study describes the isolation and structural determination of two amides, isolated for the first time: N,4-dihydroxy-N-(2'-hydroxyethyl)-benzamide (0.019%) and N,4-dihydroxy-N-(2'-hydroxyethyl)-benzeneacetamide (0.023%). These amides, produced by the red macroalgae Bostrychia radicans, had their structures assigned by NMR spectral data and MS analyses. In addition, this chemical study led to the isolation of cholesterol, heptadecane, squalene, trans-phytol, neophytadiene, tetradecanoic and hexadecanoic acids, methyl hexadecanoate and methyl 9-octadecenoate, 4-(methoxymethyl)-phenol, 4-hydroxybenzaldehyde, methyl 4-hydroxybenzeneacetate, methyl 2-hydroxy-3-(4-hydroxyphenyl)-propanoate, hydroquinone, methyl 4-hydroxymandelate, methyl 4-hydroxybenzoate, 4-hydroxybenzeneacetic acid and (4-hydroxyphenyl)-oxo-acetaldehyde. This is the first report concerning these compounds in B. radicans, contributing by illustrating the chemical diversity within the Rhodomelaceae family.

Different species of Laurencia have proven to be a rich source of natural products yielding interesting bioactive halogenated secondary metabolites, such as terpenoids and acetogenins. It is shown that such compounds are accumulated in the spherical, reniform to claviform refractive inclusions called corps en cerise (CC), which are intensively osmiophilic and located mainly in the cortical cells of the thalli and also in trichoblast cells. Up to now, it was believed that CC were present only in these two kinds of cells. Recently, however, a species of Laurencia, L. marilzae, with CC in all cells of the thallus, i.e., cortical, medullary, including the pericentral and axial cells, as well as in the trichoblasts, was described from the Canary Islands, and subsequently also reported to Brazil and Mexico. Within the Laurencia complex, only Laurencia species produce CC. Since the species of Laurencia are targets of interest for the prospection of bioactive substances due to their potential antibacterial, antifungal, anticholinesterasic, antileishmanial, cytotoxic, and antioxidant activities, the present paper carries out a comparative analysis of the corps en cerise in several species of Laurencia from the Atlantic Ocean to obtain basic information that can support natural product bioprospection projects. Our results show that the number and size of the CC are constant within a species, independent of the geographical distribution, corroborating their use for taxonomical purposes to differentiate groups of species that present a lower number from those that have a higher number. In this regard, there was a tendency for the number of CC to be higher in some species of Laurencia from the Canary Islands. The presence of CC can also be used to distinguish species in which these organelles are present in all cells of the thallus from those in which CC are restricted to the cortical cells. Among the species analyzed, L. viridis displayed the most varied secondary metabolites composition, such as sesquiterpenes, diterpenes, triterpenes, all of which showed potent antiviral, cytotoxic, and antitumoral activities, including protein phosphatase type 2A (PP2A) inhibitory effects.

In the recent years, consumers became more aware and sensible in respect to environment and food safety matters. They are more and more interested in organic agriculture and markets and tend to prefer ‘organic’ products more than their traditional counterparts. To increase the quality and reduce the cost of production in organic and low-input agriculture, the 6FP-European “QLIF” project investigated the use of natural products such as bio-inoculants. They are mostly composed by arbuscular mycorrhizal fungi and other microorganisms, so-called “plant probiotic” microorganisms (PPM), because they help keeping an high yield, even under abiotic and biotic stressful conditions. Italian laws (DLgs 217, 2006) have recently included them as “special fertilizers”. This thesis focuses on the use of special fertilizers when growing tomatoes with organic methods in open field conditions, and the effects they induce on yield, quality and microbial rhizospheric communities. The primary objective was to achieve a better understanding of how plant-probiotic micro-flora management could buffer future reduction of external inputs, while keeping tomato fruit yield, quality and system sustainability. We studied microbial rhizospheric communities with statistical, molecular and histological methods. This work have demonstrated that long-lasting introduction of inoculum positively affected micorrhizal colonization and resistance against pathogens. Instead repeated introduction of compost negatively affected tomato quality, likely because it destabilized the ripening process, leading to over-ripening and increasing the amount of not-marketable product. Instead. After two years without any significant difference, the third year extreme combinations of inoculum and compost inputs (low inoculum with high amounts of compost, or vice versa) increased mycorrhizal colonization. As a result, in order to reduce production costs, we recommend using only inoculum rather than compost. Secondly, this thesis analyses how mycorrhizal colonization varies in respect to different tomato cultivars and experimental field locations. We found statistically significant differences between locations and between arbuscular colonization patterns per variety. To confirm these histological findings, we started a set of molecular experiments. The thesis discusses preliminary results and recommends their continuation and refinement to gather the complete results.

Die Schwämme (Porifera) sind eine reiche Quelle bioaktiver Naturstoffe. Viele dieser Naturstoffe besitzen das Potential, als Pharmazeutika, molekulare Sonden usw. eingesetzt oder weiterentwickelt zu werden. Die Beschaffung dieser Naturstoffe in ausreichenden Mengen stellt jedoch eines der größten Probleme bei der Testung und Produktion vielversprechender Substanzen dar. Der Transfer von DNA in Schwammzellen bzw. in komplette Organismen wäre ein vielversprechender Ansatz, dieses Problem zu lösen. Das Ziel dieser Arbeit war es deshalb, die Funktion und Struktur homologer Promotoren zu untersuchen und eine Methode des Gentransfers in Schwammzellen auszuarbeiten. Zu diesem Zweck wurde zusätzlich zu der bereits vorhandenen 5'-flankierenden Region des conventional PKC-Gens aus Geodia cydonium eine genomische Bibliothek von Suberites domuncula konstruiert, um diese mit Hilfe des DNA-Homologiescreenings nach den 5'-flankierenden Regionen des cPKC- und des SNZ (SnooZe)-Gens (SD_SNZG) zu durchsuchen. Die Klonierung und Sequenzierung sowohl des 5'-Bereichs als auch die Charakterisierung der Exon-Intron Struktur beider Gene wurde erfolgreich durchgeführt. In der 5'-Region des SNZ-Gens konnte dabei ein weiteres Gen (SD_SNO; SNZ proximal Open Reading Frame) identifiziert werden, das in einer 'Kopf-an-Kopf' Anordnung zu SD_SNZG orientiert ist. Sowohl SD_SNZG als auch SD_SNO wurden hochkonservierten Genfamilien zugeordnet, deren Vorkommen in Metazoen hier erstmals beschrieben wird.Funktionelle Studien mit Hilfe der Reportergene Luciferase und Enhanced Green Fluorescent Protein (EGFP) im heterologen System der NIH 3T3 Zellen wiesen sowohl dem cPKC-Promotor aus G. cydonium als auch dem SNZ-Promotor aus S. domuncula eine starke Promotoraktivität im Verhältnis zum SV40-Promotor nach. Die Aktivität des cPKC-Promotors aus S. domuncula dagegen war relativ schwach. Darüber hinaus konnte geklärt werden, daß die 5'-flankierende Region des SNZ-Gens bidirektionale Promotoraktivität aufweist und daß der G. cydonium cPKC-Promotor keine TATA-Box besitzt, sondern eine GC-Box für die basale Funktion benötigt.Als geeignete Methode zur Transfektion von Zellen des Schwamms S. domuncula erwies sich der ballistische Gentransfer mit Hilfe der Gene Gun. Homologe Promotoren konnten die sichtbare Expression des Reportergens EGFP jedoch nicht bewirken. Nur der virale CMV-Promotor erwies sich als hierfür geeignet.

The present work is part of a larger project aimed at obtaining compounds of industrial interest from renewable sources. The work is particularly aimed to investigate the reactivity of 2,5-bis-hydroxymethylfuran (BHMF), an important building block of organic nature easily obtainable from biomass, with acid catalysis. Through the study of the reactivity of BHMF in water, in the presence of an heterogeneous acid catalyst (Amberlyst 15), has been developed a new synthetic method for the preparation of α-6-hydroxy-6-methyl-4-enyl-2H-pyran-3-one a derivative whose molecular skeleton is similar to that of natural products which are used in pharmaceutical chemistry. The product is obtained in milder conditions and with better selectivity with respect to the strongly oxidizing conditions with which it is prepared in the literature starting from different precursors containing furan ring.

The work described in this thesis deals with the development of the asymmetric organocatalytic conjugate addition reaction of 1,3-dicarbonyl compounds to ortho-quinone methides. Due to their instability, these synthetically appealing intermediates have not been fully exploited in catalytic asymmetric settings. In this work, the instability of ortho-quinone methides is overcome by their generation in situ under mild basic conditions, starting from the corresponding sulfonyl derivatives. The bifunctional catalysts used are able to activate both substrates for the reaction, by means of a synergic action of the two catalytic sites, inducing at the same time high enantioselection in the addition step. The reaction leads to the generation of a 2-alkylphenolic framework, featuring a chiral centre at the benzylic position. In particular, the employment of acetylacetone and Meldrum acid as nucleophiles has allowed the obtainment of 4H-chromenes and chroman-2-ones in good yields and generally excellent enantioselectivities. These compounds are synthetic precursors of several natural products, some of which showing interesting biological activity, and of some active pharmaceutical ingredients used in commercial drugs.

Der Suche nach neuen Wirkstoffen für den chemischen Pflanzenschutz kommt insbesondere vor dem Hintergrund der steigenden Weltbevölkerung und weniger zur Verfügung stehenden kulturfähigen Ackerflächen eine stetig wachsende Bedeutung zu. Ziel dieser Arbeit war die Synthese von cyclischen Peptiden und Depsipeptiden, die aufgrund ihrer biologischen Aktivität als potentielle Insektizide für den chemischen Pflanzenschutz in Frage kommen. Darüber hinaus sollten von Kohlenhydraten abgeleitete Katalysatoren zur enantioselektiven Cyanhydrinsynthese entwickelt werden, um einen leichten Zugang zu den Bausteinen der Depsipeptide zu ermöglichen. Als vielversprechender Naturstoff mit insektiziden Eigenschaften gilt das cyclische Pentapeptid Cycloaspeptid E, dessen Totalsynthese in 10 Stufen mit einer Gesamtausbeute von 25% erreicht wurde, sodass die Verbindung für biologische Tests bereitgestellt werden konnte. Zusätzlich gelang die Kristallisation der Verbindung, was eine Röntgenstrukturanalyse ermöglichte. Ein Derivat von Cycloaspeptid E sollte 2-Aminonicotinsäure anstelle von Anthranilsäure enthalten. Die Synthese dieser Verbindung wurde auf drei Wegen versucht. Dabei zeigte sich, dass es bei einer zur Totalsynthese des Naturstoffs analogen Strategie zur quantitativen Bildung eines Diketopiperazins kommt. Auf den anderen Routen ließ sich entweder ein Kupplungsschritt nicht realisieren, oder die Verbindung erwies sich unter den gewählten Bedingungen als instabil. Die Darstellung eines 2-Aminonicotinsäure-Derivats von Cycloaspeptid E bleibt daher weiterhin ein ungelöstes Problem, das weiterer Forschung bedarf. Verticilid A1 ist ein cyclisches Depsipeptid, das aufgrund seiner Bindungsfähigkeit an den Ryanodinrezeptor von Insekten, als Leitstruktur für die Suche nach neuen Insektiziden von Interesse ist. Um zu untersuchen, wie wichtig die Esterbindungen im Molekül für die biologische Aktivität sind, sollte das entsprechende Amid-Derivat und das Cyclodepsipeptid mit nur zwei statt vier Esterbindungen hergestellt werden. Hierbei zeigte sich, dass eine zur Darstellung von Verticilid A1 analoge Syntheseroute zu einer ausgeprägten Epimerisierung führt. Eine lineare Synthese der Derivate endet in der Bildung des Diketopiperazins. Weiterhin wurden zwei neue, zueinander pseudoenantiomere Vanadium(IV)-Katalysatoren auf Basis von D-Glucose einerseits und L-Xylose andererseits dargestellt. Diese lassen sich in fünf bzw. 14 Stufen synthetisieren und liefern in der enantioselektiven Katalyse von Mandelsäurenitril Enantiomerenüberschüsse von 89% bzw. 91% bei hohen Ausbeuten. Zusammenfassend lässt sich feststellen, dass im Rahmen dieser Arbeit die Totalsynthese von Cycloaspeptid E erfolgreich durchgeführt wurde, und die Syntheseversuche von weiteren cyclischen Peptiden wichtige Erkenntnisse für weitere Synthesen lieferten. Mit den beiden hergestellten Vanadium(IV)-Komplexen wurden zwei potente, auf Kohlenhydraten basierende Katalysatoren für die enantioselektive Synthese von Cyanhydrinen entwickelt.

Es wurden drei Ansätze zur Totalsynthese von Fungerin verfolgt, dessen charakteristisches Strukturmerkmal ein N-methylierter in 4,5-Position disubstituierter Imidazolkern ist. Zunächst wurde ein Syntheseweg eingeschlagen, bei dem die Bildung des Imidazolrings nach Marckwald erfolgte. Das hierfür benötigte α-Aminoketon wurde in einer konvergenten Synthesesequenz aus zwei Bausteinen zusammengesetzt. Die anschließende Ringschlussreaktion mit Kaliumthiocyanat lieferte ein Thioimidazolderivat, welches erfolgreich zum angestrebten Zielmolekül entschwefelt werden konnte. Die Gesamtausbeute betrug 8,1 % über sieben Stufen. In einem zweiten Syntheseweg wurde ein in 4- und 5-Position orthogonal geschütztes Imidazolderivat synthetisiert, um eine höhere Flexibilität bei geplanten Strukturvariationen der Seitenketten zu erreichen. Nach sequentieller Entschützung und Funktionalisierung sollten verschiedene Substituenten angebracht werden. Die Bildung des Imidazolkerns erfolge über eine Kondensationsreaktion von Methylamin mit einem N-formylierten α-Aminoketon, welches über eine Claisen-Kondensation erhalten wurde. Die zur Einführung der C5-Seitenkette geplante Grignard- bzw. Schlosser-Fouquet-Kupplung erwies sich als nicht zuverlässig reproduzierbar. In der Folge wurde in einer dritten Synthesesequenz ein Imidazolderivat mit zwei unterschiedlichen Anknüpfungspunkten in 4- und 5-Position synthetisiert. Dadurch war es möglich über Julia-Kocienski Olefinierungen verschiedene Seitenketten in 5-Position anzubringen. In 4-Position erfolgte die Einführung über Heck-Kupplungen. Insgesamt konnten so, neben Fungerin, noch sieben weitere Fungerinderivate erhalten werden.

In traditional medicine, numerous plant preparations are used to treat inflammation both topically and systemically. Several anti-inflammatory plant extracts and a few natural product-based monosubstances have even found their way into the clinic. Unfortunately, a number of plant secondary metabolites have been shown to trigger detrimental pro-allergic immune reactions and are therefore considered to be toxic. In the phytotherapy research literature, numerous plants are also claimed to exert immunostimulatory effects. However, while the concepts of plant-derived anti-inflammatory agents and allergens are well established, the widespread notion of immunostimulatory plant natural products and their potential therapeutic use is rather obscure, often with the idea that the product is some sort of "tonic" for the immune system without actually specifying the mechanisms. In this commentary it is argued that the paradigm of oral plant immunostimulants lacks clinical evidence and may therefore be a myth, which has originated primarily from in vitro studies with plant extracts. The fact that no conclusive data on orally administered immunostimulants can be found in the scientific literature inevitably prompts us to challenge this paradigm.

It is intriguing that during human cultural evolution man has detected plant natural products that appear to target key protein receptors of important physiological systems rather selectively. Plants containing such secondary metabolites usually belong to unique chemotaxa, induce potent pharmacological effects and have typically been used for recreational and medicinal purposes or as poisons. Cannabis sativa L. has a long history as a medicinal plant and was fundamental in the discovery of the endocannabinoid system. The major psychoactive Cannabis constituent Delta(9)-tetrahydrocannabinol (Delta(9)-THC) potently activates the G-protein-coupled cannabinoid receptor CB(1) and also modulates the cannabinoid receptor CB(2). In the last few years, several other non-cannabinoid plant constituents have been reported to bind to and functionally interact with CB receptors. Moreover, certain plant natural products, from both Cannabis and other plants, also target other proteins of the endocannabinoid system, such as hydrolytic enzymes that control endocannabinoid levels. In this commentary we summarize and critically discuss recent findings.
