The treatment of advanced prostate cancer (PCa) remains a challenge. Identification of new molecular mechanisms that regulate PCa initiation and progression would provide targets for the development of new cancer treatments. The Foxm1 transcription factor is highly up-regulated in tumor cells, inflammatory cells, and cells of tumor microenvironment. However, its functions in different cell populations of PCa lesions are unknown. To determine the role of Foxm1 in tumor cells during PCa development, we generated two novel transgenic mouse models, one exhibiting Foxm1 gain-of-function and one exhibiting Foxm1 loss-of-function under control of the prostate epithelial-specific Probasin promoter. In the transgenic adenocarcinoma mouse prostate (TRAMP) model of PCa that uses SV40 large T antigen to induce PCa, loss of Foxm1 decreased tumor growth and metastasis. Decreased prostate tumorigenesis was associated with a decrease in tumor cell proliferation and the down-regulation of genes critical for cell proliferation and tumor metastasis, including Cdc25b, Cyclin B1, Plk-1, Lox, and Versican. In addition, tumor-associated angiogenesis was decreased, coinciding with reduced Vegf-A expression. The mRNA and protein levels of 11β-Hsd2, an enzyme playing an important role in tumor cell proliferation, were down-regulated in Foxm1-deficient PCa tumors in vivo and in Foxm1-depleted TRAMP C2 cells in vitro. Foxm1 bound to, and increased transcriptional activity of, the mouse 11β-Hsd2 promoter through the -892/-879 region, indicating that 11β-Hsd2 was a direct transcriptional target of Foxm1. Without TRAMP, overexpression of Foxm1 either alone or in combination with inhibition of a p19(ARF) tumor suppressor caused a robust epithelial hyperplasia, but was insufficient to induce progression from hyperplasia to PCa. Foxm1 expression in prostate epithelial cells is critical for prostate carcinogenesis, suggesting that inhibition of Foxm1 is a promising therapeutic approach for prostate cancer chemotherapy.

Logistic regression is included into the analysis techniques which are valid for observationalmethodology. However, its presence at the heart of thismethodology, and more specifically in physical activity and sports studies, is scarce. With a view to highlighting the possibilities this technique offers within the scope of observational methodology applied to physical activity and sports, an application of the logistic regression model is presented. The model is applied in the context of an observational design which aims to determine, from the analysis of use of the playing area, which football discipline (7 a side football, 9 a side football or 11 a side football) is best adapted to the child"s possibilities. A multiple logistic regression model can provide an effective prognosis regarding the probability of a move being successful (reaching the opposing goal area) depending on the sector in which the move commenced and the football discipline which is being played.

Athletes seem compelled to include some forms of altitude training in their preparation expecting additional performance gains compared to equivalent training at sea-level. For the general population, altitude training often only consists in spending weeks at altitude to enhance red blood cell production, hemoglobin mass and thus oxygen delivery to the muscles. Over the past two decades, intermittent hypoxic training (IHT), that is, a method where athletes live at or near sea-level but train in hypobaric hypoxia (HH, real altitude) or normobaric hypoxia (NH, simulated altitude) was shown to induce exclusive adaptations directly at the muscular level that may support performance improvements. Our work first demonstrated significant differences between exposure and exercise in HH vs. NH that may help disentangling hypoxia and hypobaria for athletes or mountaineers who use NH to prepare for altitude competitions or expeditions. Second, we produced a comprehensive review of the strikingly poor and controversial benefits of IHT for performance enhancement in team or racket sports. Using evidence of peripheral muscular adaptations with the recruitment of fast-twitch fibers playing a major role, we then developed and assessed the potential of a new training method in hypoxia based on the repetitions of "all-out" sprints interspersed with incomplete recovery periods, the so called "repeated sprint training in hypoxia" (RSH). We have consequently shown RSH to delay fatigue when sprints with incomplete recoveries are repeated until exhaustion both in cycling and cross-country ski double poling. We definitely outlined RSH as a promising training strategy and proposed new studies to judge the efficacy of RSH in team sports and determine the specific mechanisms that may enhance team game results. In conclusion, our work allowed updating the panorama over the contemporary hypoxic training possibilities. It provides an overview of the current scientific knowledge about intermittent hypoxic training and repeated sprint training in hypoxia (RSH). This will benefit athletes and teams in intermittent sports looking to include a hypoxic stimulus to their training to gain a specific competitive edge.

The aim of this paper is to underline the fact that, in most contemporary philosophical debates (and somehow, issues in feminist theory are not an exception), a well-known term is playing an important role: experience. We need a cartography of the uses of experience, if we want to know what we are talking about and also if we intend to analyze and to rethink the last 25 years of ¿women studies¿, ¿gender studies¿ or ¿subaltern studies¿.

The hallmark of social insects is their caste system: reproduction is primarily monopolized by queens, whereas workers specialize in the other tasks required for colony growth and survival. Pheromones produced by reining queens have long been believed to be the prime factor inhibiting the differentiation of new reproductive individuals. However, there has been very little progress in the chemical identification of such inhibitory pheromones. Here we report the identification of a volatile inhibitory pheromone produced by female neotenics (secondary queens) that acts directly on target individuals to suppress the differentiation of new female neotenics and identify n-butyl-n-butyrate and 2-methyl-1-butanol as the active components of the inhibitory pheromone. An artificial pheromone blend consisting of these two compounds had a strong inhibitory effect similar to live neotenics. Surprisingly, the same two volatiles are also emitted by eggs, playing a role both as an attractant to workers and an inhibitor of reproductive differentiation. This dual production of an inhibitory pheromone by female reproductives and eggs probably reflects the recruitment of an attractant pheromone as an inhibitory pheromone and may provide a mechanism ensuring honest signaling of reproductive status with a tight coupling between fertility and inhibitory power. Identification of a volatile pheromone regulating caste differentiation in a termite provides insights into the functioning of social insect colonies and opens important avenues for elucidating the developmental pathways leading to reproductive and nonreproductive castes.

More than 80 % of vascular plants in the world form symbioses with arbuscular mycorrhizal fungi (AMF). AMF supply plants with nutrients such as phosphate and nitrogen, and can also help the plants to take up water. Hence, the symbiosis can greatly influence the growth and the defence of plants. By modifying plant productivity and diversity, AMF are considered as keystone species in ecosystems, playing a role that ultimately affects many food webs. This is why mycorrhizal symbioses have been investigated for several decades by many research groups.¦However, a large part of the scientific research done on AMF symbiosis has focused on the interaction between one plant and one fungus. This situation is far from realistic, as in natural ecosystems, many different fungal strains and species are co-existing and interacting in a belowground network. The main goal of this PhD was to investigate first, the interaction occurring among different co-existing AMF depending on their genetic relatedness and second, the outcome of the interaction and their effects on associated species.¦We found that AMF genetic relatedness partly explains the interaction among AMF, and this was in agreement with theories made for completely different species. Briefly, we demonstrated that AMF isolates of the same species coexisted more easily when they were closely-related, whereas AMF from different species were more in competition in this case of high relatedness. We also demonstrated that coexistence and competition among AMF can mediate plant growth as well as herbivore behaviour, opening new insights in our understanding of AMF effects on ecosystem functioning.¦Overall, the results of the different experiments of this PhD highlight the necessity of using multiple AMF to understand their interactions. Even so, we demonstrated here that simple species richness is not enough to understand these interactions and genetic relatedness among the co-existing AMF is a parameter that must be taken into account.¦-¦Sur Terre, plus de 80 % des plantes vasculaires forment des symbioses avec des champignons endomycorhiziens à arbuscules (CEA). Ces CEA permettent aux plantes d'acquérir plus facilement des nutriments tels que des phosphates, des nitrates, ou simplement de l'eau. Ainsi, cette symbiose peut avoir un effet important à la fois sur la croissance mais aussi sur la défense des plantes. En modulant la productivité et la diversité des plantes, les CEA sont donc des espèces clefs dans l'écosystème. Leur présence peut avoir des répercussions sur l'ensemble des réseaux trophiques. C'est pourquoi de nombreuses équipes de recherches étudient ces symbioses mycorhizienes depuis plusieurs décennies.¦La plupart des études concernant ces symbioses se sont focalisées sur l'action d'une espèce de CEA sur une espèce de plante. Malheureusement, cette situation ne correspond pas à ce que l'on peut retrouver dans la nature, où de nombreuses souches et de nombreuses espèces de CEA coexistent et interagissent dans un réseau mycélien souterrain. Le principal but de cette thèse était d'étudier, premièrement les interactions entre les différent CEA en fonction de leur apparentement génétique, et deuxièmement, d'étudier l'effet de ces interactions fongiques sur l'écologie des espèces associées.¦Au cours des différentes expériences de cette thèse, nous avons démontré que l'apparentement génétique entre les CEA expliquait une part non négligeable de leurs interactions. En résumé, plus l'apparentement génétique entre des souches de CEA d'une même espèce sera grand, plus ces souches seront capables de coexister. En revanche, s'il s'agit d'espèces différentes de CEA, plus elles seront apparentées, plus la compétition sera grande entre elles. Nous avons également démontré que la coexistence et la compétition entre différents CEA peut modifier à la fois la croissance des plantes mais aussi le comportement de leur prédateurs, ce qui ouvre de nouvelles perspectives sur notre compréhension des effets des CEA dans le fonctionnement des écosystèmes.¦Globalement, les résultats de nos différentes expériences mettent en évidence la nécessité d'utiliser plusieurs souches ou espèces de CEA pour mieux comprendre leurs interactions. Quand bien même, nos expériences démontrent que le simple recensement du nombre d'espèces de CEA n'est pas suffisant pour comprendre les interactions et que l'apparentement génétique des CEA coexistants est un paramètre qui doit être pris en compte.

El objetivo del trabajo es determinar si el uso de un grupo de verbos es propio del español de Argentina o si, por el contrario, se extiende a otros países hispanohablantes. Para ello, se analizan el proceso de derivación verbal, la semántica y el carácter neológico de las voces.

Despite the increase of animal and plant introductions worldwide and the strong augmentation of the reptile trade, few invasive snake populations have been studied. Dice snakes (Natrix tessellata) were introduced to the shores of Lake Geneva (Switzerland) in the early 1920s, and are now well established. This region of introduction was previously inhabited by Viperine snakes (N. maura). Ever since these two species have been under monitoring (which began in 1996) the Viperine snake population has shown drastic decline. We examine here the possibility of trophic competition by analysing diet composition, prey size and trophic niche overlap. Spatial distribution is also assessed in order to address the question of spatial competitive exclusion. We found very similar diets, and thus a high trophic niche overlap, indicating no partitioning of the trophic resource. No arguments in favour of spatial competitive exclusion were found. Our study suggests that trophic competition may occur between the two natricines and that it may give an explanation for the drastic decline of the Viperine snake in this area. Other pathways potentially playing a role in the exclusion of the Viperine snake are discussed.

En los años 30 del siglo XX, mientras las primeras vanguardias artísticas recorren Europa, María Zambrano y Martin Heidegger denuncian la pérdida del lugar del arte. El arte, para Zambrano, ya no da a ver, sólo se ve como arte. Por ello, perdió la tierra, está desterrado. Para Heidegger, por su parte, muchas de las obras de arte del siglo XX no pueden establecer un mundo y producir la tierra. En ese destierro del arte nos situaremos para interrogar el papel de la música en el pensamiento de ambos filósofos

Este proyecto es un estudio que pretende realizar una aplicación dirigida a centros deportivos, implementando a través de BIOMAX el acceso a las instalaciones utilizando tarjetas de proximidad. Además la aplicación permitirá gestionar de forma eficiente a sus socios e instalaciones. De esta manera se consigue automatizar - mejorando en tiempo y calidad -una tarea imprescindible de realizar, control de accesos de forma sencilla y con un mantenimiento fácil por parte de los usuarios que la utilicen. Para ello se utilizará un dispositivo KIMALDI que nos permitirá gestionar el control de accesos

Purpose: The exact role of individual T cell-subsets in the development of rejection is not clearly defined. Given their distinct phenotypes, effector functions and trafficking patterns, naïve (CD45RBhiCD44lo) and memory (CD45RBloCD44hi) T cells may play distinct roles in anti-donor immunity after transplantation. Furthermore, only the CD4+CD45RBlo population contains CD4+CD25+ T cells, a subset with suppressive functions playing a major role in the maintenance of peripheral tolerance. The aim of this work was to study the contribution of these individual subsets in alloresponses via the direct and indirect pathways using a murine experimental model. Methods and materials: Purified naïve or memory CD4+ T cells were adoptively transferred into lymphopenic mice undergoing a skin allograft. Donor to recipient MHC combinations were chosen in order to study the direct and the indirect pathways of allorecognition separately. Graft survival and in vivo expansion, effector function and trafficking of the transferred T cells was assessed at different time points after transplantation. Results: We found that the cross-reactive CD4+CD45RBlo memory T-cell pool was heterogeneous and contained cells with regulatory potentials, both in the CD4+CD25+ and CD4+CD25-populations. CD4+ T cells capable of inducing strong primary alloreactive responses in vitro and rejection of a first allograft in vivo were mainly contained within the CD45RBhi naïve CD4+ T-cell compartment. CD4+CD45RBlo T cells proliferated less abundantly to allogeneic stimulation than their naïve counterparts both in vitro and in vivo, and allowed prolonged allograft survival even after the depletion of the CD4+CD25+ subset. Interestingly, CD4+CD25-CD45RBlo T cells were capable of prolonging allograft survival, mainly when the indirect pathway was the only mechanism of allorecognition. The indirect pathway response, which was shown to drive true chronic rejection and contribute to chronic allograft dysfunction, was predominantly mediated by naïve CD4+ T cells. Conclusion: This work provides new insights into the mechanisms that drive allograft rejection and should help develop new clinical immunosuppressive protocols. In particular, our results highlight the importance of selectively targeting individual T-cell subsets to prevent graft rejection but at the same time maintain immune protective responses to common pathogens.

Segons Castelo (1999, 2009) i Ardá i Casal (2003) la fase ofensiva del futbol es divideix en la fase de progressió, en la qual es busca progressar a porteria, i en la fase de manteniment o emergència, en la qual s’assegura la possessió de la pilota. L’objectiu és analitzar si quan el davanter centre rep la pilota d’esquena a porteria, es manté amb més eficàcia la possessió jugant en progressió o en emergència durant els 5” posteriors a la recepció de la pilota per part del davanter. L’estudi és Descriptiu d’Observació Directa, en aquest s’observen i s’analitzen, a través de vídeos, 10 partits de la fase de grups de la fase final de la Eurocopa 2012, en concret els partits són dels grups A i B. Els resultats confirmen la hipòtesi inicial, ja que es manté amb més eficàcia la pilota quan aquesta es juga en emergència (71%) que no pas en progressió (54%).

We present a framework for modeling right-hand gestures in bowed-string instrument playing, applied to violin. Nearly non-intrusive sensing techniques allow for accurate acquisition of relevant timbre-related bowing gesture parameter cues. We model the temporal contour of bow transversal velocity, bow pressing force, and bow-bridge distance as sequences of short segments, in particular B´ezier cubic curve segments. Considering different articulations, dynamics, andcontexts, a number of note classes is defined. Gesture parameter contours of a performance database are analyzed at note-level by following a predefined grammar that dictatescharacteristics of curve segment sequences for each of the classes into consideration. Based on dynamic programming, gesture parameter contour analysis provides an optimal curve parameter vector for each note. The informationpresent in such parameter vector is enough for reconstructing original gesture parameter contours with significant fidelity. From the resulting representation vectors, weconstruct a statistical model based on Gaussian mixtures, suitable for both analysis and synthesis of bowing gesture parameter contours. We show the potential of the modelby synthesizing bowing gesture parameter contours from an annotated input score. Finally, we point out promising applicationsand developments.

Summary : Four distinct olfactory subsystems compose the mouse olfactory system, the main olfactory epithelium (MOE), the septal organ of Masera (SO), the vomeronasal organ (VNO) and the Grueneberg ganglion (GG). They are implicated in the sensory modalities of the animal and they evolved to analyse and discriminate molecules carrying chemical messages, such as odorants and pheromones. In this thesis, the VNO, principally implicated in pheromonal communications as well as the GG, which had no function attributed until this work, were investigated from their morphology to their physiological functions, using an array of biochemical and physiological methods. First, the roles of a particular protein, the CNGA4 ion channel, were investigated in the VNO. In the MOE, CNGA4 is expressed as a modulatory channel subunit implicated in odour discrimination and adaptation. Interestingly, this calcium channel is the unique member of the cyclic nucleotide-gated (CNG) family to be expressed in the VNO and up to this work its functions remained unknown. Using a combination of transgenic and knockout mice, as well as histological and physiological approaches, we have characterized CNGA4 expression in the VNO. A strong expression in immature neurons was found as well as in the microvilli of mature neurons (putative site of chemodetection). Interestingly and confirming its dual localisation, the genetic invalidation of the CNGA4 channel has, as consequences, a strong impairment in vomeronasal maturation as well as deficit in pheromone sensing. Thus the CNGA4 channel appears to be a multifunctional protein in the mouse VNO playing essential role(s) in this organ. During the second part of the work, the morphology of the most recently described olfactory subsystem, the Grueneberg ganglion, was investigated in detail. Interestingly we found that glial cells and ciliated neurons compose this olfactory ganglion. This particular morphological aspect was similar to the olfactory AWC neurons from C. elegans which was used for further comparisons. Thus as for AWC neurons, we found that GG neurons are sensitive to temperature changes and are able to detect highly volatile molecules. Indeed, the presence of alarm pheromones (APs) secreted by stressed mice, elicit strong cellular responses, as well as a GG dependent behavioural changes. Investigations on the signaling elements present in GG neurons revealed that, as for AWC neurons, or pGC-D expressing neurons from the MOE, proteins participating in a cGMP pathway were found in GG neurons such as pGC-G and CNGA3 channels. These two proteins might be implicated in chemosensing as well as in thermosensing, two apparent properties of this organ. In this thesis, the multisensory modalities of two mouse olfactory subsystems were described and are related to a high degree of complexity required for the animal to sense its environment

BACKGROUND: Chronic endoplasmic reticulum (ER) stress contributes to the apoptotic cell death in the myocardium, thereby playing a critical role in the development of cardiomyopathy. ER stress has been reported to be induced after high-fat diet feeding in mice and also after saturated fatty acid treatment in vitro. Therefore, since several studies have shown that peroxisome proliferator-activated receptor (PPAR)β/δ inhibits ER stress, the main goal of this study consisted in investigating whether activation of this nuclear receptor was able to prevent lipid-induced ER stress in cardiac cells. METHODS AND RESULTS: Wild-type and transgenic mice with reduced PPARβ/δ expression were fed a standard diet or a high-fat diet for two months. For in vitro studies, a cardiomyocyte cell line of human origin, AC16, was treated with palmitate and the PPARβ/δ agonist GW501516. Our results demonstrate that palmitate induced ER stress in AC16 cells, a fact which was prevented after PPARβ/δ activation with GW501516. Interestingly, the effect of GW501516 on ER stress occurred in an AMPK-independent manner. The most striking result of this study is that GW501516 treatment also upregulated the protein levels of beclin 1 and LC3II, two well-known markers of autophagy. In accordance with this, feeding on a high-fat diet or suppression of PPARβ/δ in knockout mice induced ER stress in the heart. Moreover, PPARβ/δ knockout mice also displayed a reduction in autophagic markers. CONCLUSION: Our data indicate that PPARβ/δ activation might be useful to prevent the harmful effects of ER stress induced by saturated fatty acids in the heart by inducing autophagy.
