This study investigates binder distribution in wet granulation and focuses on the nucleation zone, which is the area where the liquid binder and powder surface come into contact and form the initial nuclei. An equipment independent parameter, dimensionless spray flux Psi (a), is defined to characterise the most important process parameters in the nucleation process: solution flowrate, powder flux, and binder drop size. Ex-granulator experiments are used to study the relationship between dimensionless spray flux, process variables and the coverage of binder fluid on the powder surface. Lactose monohydrate powder on a variable speed riffler passed under a flat spray once only. Water and 7% HPC solution at two spray pressures were used as binders. Experiments with red dye and image analysis demonstrate that changes in dimensionless spray flux correlate with a measurable difference in powder surface coverage. Nucleation experiments show that spray flux controls the size and shape of the nuclei size distribution. At low Psi (a), the system operates in the drop controlled regime, where one drop forms one nucleus and the nuclei size distribution is narrow. At higher Psi (a), the powder surface cakes creating a broader size distribution. For controlled nucleation with the narrowest possible size distribution, it is recommended that the dimensionless spray flux be less than 0.1 to be in the drop-controlled regime. Despite evidence linking shrimp farming to several cases of environmental degradation, there remains a lack of ecologically meaningful information about the impacts of effluent on receiving waters. The aim of this study was to determine the biological impact of shrimp farm effluent, and to compare and distinguish its impacts from treated sewage effluent. Analyses included standard water quality/sediment parameters, as well as biological indicators including tissue nitrogen (N) content, stable isotope ratio of nitrogen (delta N-15) and amino acid composition of inhabitant seagrasses, mangroves and macroalgae. The study area consisted of two tidal creeks, one receiving effluent from a sewage treatment plant and the other from an intensive shrimp farm. The creeks discharged into the western side of Moreton Bay, a sub-tropical coastal embayment on the east coast of Australia. Characterization of water quality revealed significant differences between the creeks, and with unimpacted eastern Moreton Bay. The sewage creek had higher concentrations of dissolved nutrients (predominantly NO3-/NO2- and PO43-, compared to NH4+ in the shrimp creek). In contrast, the shrimp creek was more turbid and had higher phytoplankton productivity. Beyond 750 m from the creek mouths, water quality parameters were indistinguishable from eastern Moreton Bay values. Biological indicators detected significant impacts up to 4 km beyond the creek mouths (reference site). Elevated plant delta N-15 values ranged from 10.4-19.6 parts per thousand at the site of sewage discharge to 2.9-4.5 parts per thousand at the reference site. The free amino acid concentration and composition of seagrass and macroalgae was used to distinguish between the uptake of sewage and shrimp derived N. Proline (seagrass) and serine (macroalgae) were high in sewage impacted plants and glutamine (seagrass) and alanine (macroalgae) were high in plants impacted by shrimp effluent. The delta N-15 isotopic signatures and free amino acid composition of inhabitant flora indicated that sewage N extended further from the creek mouths than shrimp N. The combination of physical/chemical and biological indicators used in this study was effective in distinguishing the composition and subsequent impacts of aquaculture and sewage effluent on the receiving waters. Effluent water from shrimp ponds typically contains elevated concentrations of dissolved nutrients and suspended particulates compared to influent water. Attempts to improve effluent water quality using filter feeding bivalves and macroalgae to reduce nutrients have previously been hampered by the high concentration of clay particles typically found in untreated pond effluent. These particles inhibit feeding in bivalves and reduce photosynthesis in macroalgae by increasing effluent turbidity. In a small-scale laboratory study, the effectiveness of a three-stage effluent treatment system was investigated. In the first stage, reduction in particle concentration occurred through natural sedimentation. In the second stage, filtration by the Sydney rock oyster, Saccostrea commercialis (Iredale and Roughley), further reduced the concentration of suspended particulates, including inorganic particles, phytoplankton, bacteria, and their associated nutrients. In the final stage, the macroalga, Gracilaria edulis (Gmelin) Silva, absorbed dissolved nutrients. Pond effluent was collected from a commercial shrimp farm, taken to an indoor culture facility and was left to settle for 24 h. Subsamples of water were then transferred into laboratory tanks stocked with oysters and maintained for 24 h, and then transferred to tanks containing macroalgae for another 24 h. Total suspended solid (TSS), chlorophyll a, total nitrogen (N), total phosphorus (P), NH4+, NO3-, and PO43-, and bacterial numbers were compared before and after each treatment at: 0 h (initial); 24 h (after sedimentation); 48 h (after oyster filtration); 72 h (after macroalgal absorption). The combined effect of the sequential treatments resulted in significant reductions in the concentrations of all parameters measured. High rates of nutrient regeneration were observed in the control tanks, which did not contain oysters or macroalgae. Conversely, significant reductions in nutrients and suspended particulates after sedimentation and biological treatment were observed. Overall, improvements in water quality (final percentage of the initial concentration) were as follows: TSS (12%); total N (28%); total P (14%); NH4+ (76%); NO3- (30%); PO43-(35%); bacteria (30%); and chlorophyll a (0.7%). Despite the probability of considerable differences in sedimentation, filtration and nutrient uptake rates when scaled to farm size, these results demonstrate that integrated treatment has the potential to significantly improve water quality of shrimp farm effluent. Current shrimp pond management practices generally result in elevated concentrations of nutrients, suspended solids, bacteria and phytoplankton compared with the influent water. Concerns about adverse environmental impacts caused by discharging pond effluent directly into adjacent waterways have prompted the search for cost-effective methods of effluent treatment. One potential method of effluent treatment is the use of ponds or raceways stocked with plants or animals that act as natural biofilters by removing waste nutrients. In addition to improving effluent water quality prior to discharge, the use of natural biofilters provides a method for capturing otherwise wasted nutrients. This study examined the potential of the native oyster, Saccostrea commercialis (Iredale and Roughley) and macroalgae, Gracilaria edulis (Gmelin) Silva to improve effluent water quality from a commercial Penaeus japonicus (Bate) shrimp farm, A system of raceways was constructed to permit recirculation of the effluent through the oysters to maximize the filtration of bacteria, phytoplankton and total suspended solids. A series of experiments was conducted to test the ability of oysters and macroalgae to improve effluent water quality in a flow-through system compared with a recirculating system. In the flow-through system, oysters reduced the concentration of bacteria to 35% of the initial concentration, chlorophyll a to 39%, total particulates (2.28-35.2 mum) to 29%, total nitrogen to 66% and total phosphorus to 56%. Under the recirculating flow regime, the ability of the oysters to improve water quality was significantly enhanced. After four circuits, total bacterial numbers were reduced to 12%, chlorophyll a to 4%, and total suspended solids to 16%. Efforts to increase biofiltration by adding additional layers of oyster trays and macroalgae-filled mesh bags resulted in fouling of the lower layers causing the death of oysters and senescence of macroalgae. Supplementary laboratory experiments were designed to examine the effects of high effluent concentrations of suspended particulates on the growth and condition of oysters and macroalgae. The results demonstrated that high concentrations of particulates inhibited growth and reduced the condition of oysters and macroalgae. Allowing the effluent to settle before biofiltration improved growth and reduced signs of stress in the oysters and macroalgae. A settling time of 6 h reduced particulates to a level that prevented fouling of the oysters and macroalgae.

Variation in the growth, survival and change in total biomass (termed biomass increase) of different families of juvenile Penaeus japonicus was investigated over a range of temperatures in controlled laboratory experiments. In the first experiment, the effects of temperature on six families of juveniles were examined over a broad range of temperatures (24 to 30 degreesC). In the second experiment, the effects of temperature on six more families of juveniles were examined over a narrower range of temperatures (27.5 to 31.2 degreesC). Over the broad temperature range, mean growth and biomass increase were highest at 27 degreesC and mean survival was highest at 24 degreesC. Mean growth was lowest at 24 degreesC, whilst survival and biomass increase were lowest at 30 degreesC. However, there was a significant interaction between family and temperature, with some families tolerating a broader range of temperatures than others. As a result, the ranking of families in relation to growth, survival and biomass increase changed at each temperature. This effect was more pronounced for survival than for growth. Over the narrower range, temperature significantly affected growth, survival and biomass increase, but there was no significant interaction between family and temperature. Growth, survival and biomass increase were significantly lower at 31.2 than at 27.5 and 29.2 degreesC. These results suggest that if grow-out conditions for P. japonicus vary by more than a few degrees, interactions between family and temperature could affect the efficiency of selection. The results also suggest that the family x temperature interaction may have a more pronounced effect on survival than on growth. Crown Copyright (C) 2002 Published by Elsevier Science B.V. There is considerable anecdotal evidence from industry that poor wetting and liquid distribution can lead to broad granule size distributions in mixer granulators. Current scale-up scenarios lead to poor liquid distribution and a wider product size distribution. There are two issues to consider when scaling up: the size and nature of the spray zone and the powder flow patterns as a function of granulator scale. Short, nucleation-only experiments in a 25L PMA Fielder mixer using lactose powder with water and HPC solutions demonstrated the existence of different nucleation regimes depending on the spray flux Psi(a)-from drop-controlled nucleation to caking. In the drop-controlled regime at low Psi(a) values. each drop forms a single nucleus and the nuclei distribution is controlled by the spray droplet size distribution. As Psi(a) increases, the distribution broadens rapidly as the droplets overlap and coalesce in the spray zone. The results are in excellent agreement with previous experiments and confirm that for drop-controlled nucleation. Psi(a) should be less than 0.1. Granulator flow studies showed that there are two powder flow regimes-bumping and roping. The powder flow goes through a transition from bumping to roping as impeller speed is increased. The roping regime gives good bed turn over and stable flow patterns. This regime is recommended for good liquid distribution and nucleation. Powder surface velocities as a function of impeller speed were measured using high-speed video equipment and MetaMorph image analysis software, Powder surface velocities were 0.2 to 1 ms(-1)-an order of magnitude lower than the impeller tip speed. Assuming geometrically similar granulators, impeller speed should be set to maintain constant Froude number during scale-up rather than constant tip speed to ensure operation in the roping regime. Objectives: (a) To compare the magnitude of gluteus medius and tensor fascia lata activation between a group of subjects with clinical unilateral hip osteoarthritis and a group of healthy older adults. (b) To compare the magnitude of activation of the gluteus medius and tensor fascia lata between sides in a group of subjects with clinical unilateral hip osteoarthritis and a group of healthy older adults. Methods: 19 subjects with clinical unilateral hip osteoarthritis and 19 healthy controls were investigated. The subjects performed a stepping task during which recordings were obtained using surface electromyograms from the hip abductors, and kinetic data were obtained from a dual force platform. Results: Subjects with clinical hip osteoarthritis had higher gluteus medius activation than the healthy older adults (p=0.037). In addition, there were no differences in the magnitude of gluteus medius activation between the sides (p=0.733). There was no difference in the force platform data between the groups (p=0.078). Conclusions: The increased magnitude of gluteus medius activation in the group with hip osteoarthritis is evidence of a muscular dysfunction associated with hip disease. This has implications for the progressive nature of the disease and for its conservative management.

The pre-ingestive selection of microphytobenthic algae by the cockle Cerastoderma edule was studied in comparison with diets containing the pelagic diatom Phaeodactylum tricomutum. Treatments with the different diets covered a range of seston concentrations and organic content similar to field conditions. Rejection rates of C. edule exposed to the different treatments were significantly correlated with the concentration of total particulate matter. No significant differences in total rejection rates were found between pelagic and benthic diets. Organic rejection rate was significantly correlated with particulate organic matter of the treatments and no significant differences were found between both diets. Selection efficiency was significantly correlated with particulate organic matter concentration in both diets and no significant differences were found between the diets. Analysis of the pseudofeces composition by flow cytometry from cockles exposed to a mixed diet of microphytobenthic algae and P. tricornutum, showed a preferential ingestion of the pelagic diatom. Benthic species, such as small pennates and Navicula sp., were preferentially ingested in comparison to larger microphytobenthic species. The largest microphytobenthic species, Cylindrotheca sp., was significantly rejected. In general, C. edule is an opportunistic filter feeder that takes advantage of both pelagic and benthic algal cells.

Com o objetivo de avaliar o efeito do veranico ocorrido na estação úmida sobre o metabolismo de nitrogênio em cafeeiros em sistema a pleno sol e associados com abacateiro (Persea americana Mill.) e ingazeiro (Inga edulis Mart.), foi realizado este experimento. O estudo foi conduzido em propriedade situada no município de Barra do Choça, BA, composto por plantas de café (Coffea arabica L.), variedade Catuaí, sendo caracterizados dois campos experimentais (sistema sombreado x sistema a pleno sol). As avaliações foram realizadas em janeiro (período de veranico) e março (final da estação úmida), em cinco repetições por campo experimental. Os dados foram submetidos ao teste t por meio do programa SAEG, versão 9.1. Maior teor de NO3 - foi verificado no sistema a pleno sol, tanto no período de veranico como no final da estação úmida. Em março foi observado maior acúmulo de N-orgânico no terço superior do cafeeiro a pleno sol e no terço médio do cafeeiro sombreado. Maiores teores de nitrogênio total e a tendência de maior atividade enzimática da redutase do nitrato foram verificados nos sistemas arborizados, quando avaliados dentro do mesmo terço do cafeeiro nas duas estações.

Em função da dificuldade de propagação de Passiflora setacea DC., técnicas de cultura de tecidos tornam-se alternativas viáveis. Objetivou-se estudar a germinação in vitro e determinar o melhor meio de cultivo na sua micropropagação. O presente trabalho constou de duas etapas, na primeira as sementes (sem escarificação, escarificadas na extremidade ou na parte mediana) na foram colocadas em % MS e 30 g L-1 de sacarose, distribuído em tubos de ensaio e suplementado com zero, 20 ou 40 mg L-1 de GA3. O pH do meio foi ajustado para 5,8 antes da adição de 6,0 mg L-1 de ágar. Após a inoculação das sementes nos meios de cultura, elas foram mantidas em sala de crescimento com luminosidade de 35 µmol.m-2.s-1, temperatura de 26 ± 1ºC e fotoperíodo de 16 horas. Assim que germinaram, as plântulas foram transferidas para tubos contendo meio MS, onde constituíram um segundo experimento, a fim de se testarem meios combinados com concentrações de sacarose. O delineamento experimental utilizado foi inteiramente casualizado, em esquema fatorial 3 x 3, quatro repetições e 15 sementes por parcela no primeiro experimento e 4 x 4, com quatro repetições e 3 plantas por parcela no segundo experimento. Melhores resultados na micropropagação foram obtidos em meio de cultivo MSM, associado a 28,51 e 28,74 g L-1 de sacarose. Maior índice de velocidade de germinação foi verificado com uso de 20 mg L-1 de GA3, associado à escarificação na ponta da semente.

Foram avaliadas as mudanças físicas, químicas e fisiológicas ocorridas durante o desenvolvimento do maracujá doce, da antese até o completo amadurecimento na planta. Os frutos apresentaram formato ligeiramente oblongo, padrão de desenvolvimento sigmoidal simples e padrão climatérico para a respiração. O desenvolvimento do fruto foi dividido em três fases: divisão celular até 4,70 dias após a antese (daa), expansão celular de 4,70 daa até 28,94 daa e maturação, de 28,94 daa a 91 daa. Na primeira fase, há pouco incremento nas dimensões do fruto, altas taxas respiratórias, crescimento acelerado da espessura do pericarpo e coloração do pericarpo verde-claro. Na fase seguinte, há intenso desenvolvimento das dimensões do fruto, ganho acelerado de massa da matéria fresca e coloração do pericarpo verde-intenso. Na terceira fase, há tendência à estabilização das dimensões, desenvolvimento de polpa acentuado, o teor de sólidos solúveis aumenta, os teores de vitamina C e acidez titulável diminuem. O pico do climatério foi registrado aos 63 daa. No último dia de avaliação, aos 91 daa, a polpa (suco + sementes) representava 24,46% da massa da matéria fresca total do fruto, o pericarpo respondia por 74,10% e as sementes isoladas, por 3,14%.

A análise de calos que apresentem características embriogênicas é importante para posterior regeneração, in vitro, de espécies com características agronômicas desejáveis, como o maracujazeiro nativo Passiflora gibertii. Diante do exposto, objetivou-se, com este trabalho, analisar a indução de calos oriundos de explantes foliares de Passiflora gibertiiN. E. Brown, bem como caracterizá-los, morfológica e ultraestruturalmente. Para obtenção de calos, folhas cotiledonares foram inoculadas, em meio de cultura, suplementado com picloram e 2,4-D, combinados com cinetina. Após 30 dias em meio de cultura, no escuro, os calos obtidos foram preparados para a visualização em microscopia eletrônica (transmissão e varredura) e microscopia de luz. Os resultados permitem afirmar que a adição de picloram e cinetina ao meio de cultura promove maior formação de calos em explantes foliares de P. gibertii que 2,4-D e cinetina. O regulador 2,4-D proporciona a obtenção de calos com células de formato isodiamétrico, pequenas e com pequeno espaço intercelular, sistema celular organizado e predominância de mitocôndrias de formato arredondado. Já com a utilização do regulador de crescimento picloram, observa-se a predominância de células grandes e de formato alongado, de espaços intercelulares, de sistema celular desorganizado e de mitocôndrias de formato alongado.

A aquacultura desempenha, actualmente, um papel fundamental no abastecimento de proteína animal de elevada qualidade. Nesta dissertação foi elaborado um projecto de uma unidade de piscicultura semi-intensiva de enguia, robalo, dourada e linguado e em regime extensivo a ostra. A unidade está localizada na Ria de Aveiro, na propriedade Ilha do Poço, com uma área total de 54,3 hectares. A execução de um projecto de aquacultura é um trabalho multidisciplinar que necessita a mobilização de diferentes recursos e conhecimentos. Os projectos de aquicultura estão directamente relacionados com o local de instalação da unidade e podem ser negativamente afectados pela poluição, por factores económicos locais ou por factores sociais que coloquem em risco o sucesso desta actividade económica. O principal objectivo desta dissertação foi o estudo de crescimento das espécies, taxa de alimentação, qualidade da água à entrada e saída da aquicultura, o consumo de oxigénio pelas espécies e o oxigénio necessário para oxidar a amónia em nitrato, o estudo de arejamento pelo vento, a necessidade de arejamento mecânico nos tanques de engorda e por fim a análise de investimento do projecto. Os valores obtidos para o tempo de crescimento das espécies até atingir o peso comercial foram, para a enguia 2,25anos, para a dourada 1,5 anos, para o robalo 2 anos, para o linguado 2,25 anos e para a ostra 1ano. Os índices de conversão alimentar (ICA) obtidos para as espécies foram os seguintes: 1 para a enguia, 1,24 para o robalo, 1,2 para a dourada e 1,58 para o linguado. Relativamente à concentração de amónia nos tanques de engorda, a espécie robalo apresenta o maior valor (0,3mg/L) por tanque. O arejamento natural dos tanques apenas é conseguido se o vento tiver uma velocidade de 20m/s. Por essa razão, e para garantir o arejamento requerido, serão necessários pelo menos 46 arejadores mecânicos com potência de 6kW a funcionar 24/24 horas. Para a taxa ponderada obteve-se um valor de 11,80%. O valor obtido para o VAL foi de 2.036.862 € e para a TIR, de 27,95%. O tempo necessário para que o investidor efectue o reembolso do capital investido no projecto é de 5,5 anos. O índice de rentabilidade tem um valor de 1,83 o que significa que o projecto é rentável.
