Imunoglobulinas na fase crônica da doença de Chagas

De um grupo de 112 casos crônicos de Doença de Chagas recebidos, foi possível selecionar 32 casos dotados de dados clínicos e testes de diagnóstico completos que permitiram uma avaliação da resposta imunitária humoral em face de quadros clínicos e cardiológicos bem definidos. Observou-se um ligeiro aumento dos níveis de IgG, sem que houvesse significância destes resultados quando feita a comparação com um grupo de habitantes da região de Bambuí, cujos testes, empregados normalmente para diagnóstico da Doença de Chagas, foram negativos. Este grupo, que foi considerado como o "normal" da região, não apresentava nenhuma sintomatologia e, tanto quanto foi possível observar, gozava bom estado de saúde. Os níveis de IgM e IgA apresentavam-se dentro da faixa de normalidade, enquanto os títulos de anticorpos heterófilos, ficaram entre 1/56 e 1/112. Na Correlação que se tentou estabelecer entre anticorpos heterófilos e cardiopatias chagásicas constatou-se não existir, aparentemente, relação entre concentração e cardiopatia.

Novo procedimento de xenodiagnóstico na forma crônica da doença de Chagas

Xenodiagnósticos feitos com Triatoma infestans durante 24 horas consecutivas, a intervalos de duas horas, demonstraram o Trypanosoma cruzi em 9 de 10 pacientes suspeitos de infecção chagástica crônica.

Hiperbilirrubinemia em portadores de miocardiopatia crônica chagasica descompensada

Analisando a histopatologia hepática em 20 indivíduos que faleceram com miocardiopatia crônica chagásica descompesada, 10 com hiperbilirrubinemia e icterícia e 10 sem icterícia, foi possível verificar-se que: a diferença entre os dois grupos e mais quantitativa que qualitativa, sendo que o grau e extensão da necrose congestiva parece ser o fator mais importante nos casos ictéricos: a presença de infarto pulmonar ocorreu de maneira semelhante nos dois grupos e não pareceu ter papel fundamental na patogenese da icterícia.

São descritas as principais etapas de uma técnica simples e sensível para a pesquisa, na mesma placa, do antígeno, do anticorpo e de imunocomplexos específicos baseada no "Enzyme-Linked Immunosorbent Assay" (ELISA). A técnica se mostrou de grande utilidade na pesquisa do antígeno, do anticorpo e de imunocomplexos específicos no soro de pacientes com esquistossomose hépato-intestinal crônica.

Traficando identidades: la construcción del ethos intermedio en Crónica da Rua 513.2 de João Paulo Borges Coelho

En Crónica da Rua 513.2 (2006), Borges Coelho construye un nuevo lugar de escritura en la literatura mozambiqueña. Se trata del espacio del ethos intermedio, constituyendo la indicianidad el culminar de esta configuración. En este estudio es analizado, a partir de una metodología interdisciplinar, el conjunto de estrategias utilizadas por el narrador que cristaliza la constitución de este ethos: la onomástica y el espacio-temporalidad. Estos aspectos serán estudiados en el conjunto de la novela y en el caso específico del personaje de origen indio, Valgy, añadiéndose el estudio de la voz y los grados de alteridad que emergen del texto

Productivity growth and competition in Spanish manufacturing firms: What has happened in recent years?

This paper addresses the issue of the relationship between productivity and market competition. In comparison to the economies of other European countries, the Spanish economy has been growing, while productivity growth has stagnated. Here we provide empirical evidence about the relationship between productivity and market competition from Spanish manufacturing firms at firm level between 1994 and 2004. Correcting for selection bias, our study pays special attention to the patterns of productivity growth between openness and non-openness firms. When market competition increases the effect on firms operating in domestic markets is positive but when the level of competition is high incentives to invest in innovation and productivity gains disappear. The empirical relationship between competition and productivity is an inverted U-shape, where productivity growth is highest at intermediate levels of competition. The productivity growth of firms operating in international markets is higher than that of non-openness firms, but when market competition rises they moderate their productivity growth. Our empirical results suggest that the correct competition policy in the Spanish economy should remove the barriers to competition in internal markets in order to increase the incentives for manufacturing firms to invest in innovation and productivity growth.

Isolamento de amostras do Trypanosoma cruzi por xenodiagnóstico e hemocultura de pacientes na fase crônica da doença de Chagas

Cinquenta e nove pacientes chagásicos crônicos foram submetidos a xenodiagnósticos e hemocultura concomitantes para isolamento de amostras de Trypanosoma cruzi. O xenodiagnóstico foi composto de 40 ninfas de Panstrongylus megistrus, Triatoma infestans e Dipetalogylus megistus, Triatoma infestans e Dipetalogaster maximus num total de 120 triatomíneos. Os insetos foram dissecados em grupo de 10 por espécie e o conteúdo intestinal, agrupado, examinado após prévia trituração e homogeneização. O material negativo ao microscópio foi semeado em meio LIT e examinado após 20 dias. Vinte e nove pacientes foram parasitologicamente comprovados, sendo 15 apenas no xenodiagnóstico, quatro apenas com a hemocultura e 10 por ambos os métodos. Discutem-se as dificuldades para a comprovação parasitológica dos pacientes chagásicos crônicos, o valor da utilização simultânea de diferentes espécies de triatomíneos no xenodiagnóstico e a hemocultura, numa associação positiva favorável ao aumento da sensibilidade para o diagnóstico da doença de Chagas. A positividade de 49,2% obtida neste grupo de pacientes visualiza abordagens do tipo ensaio clínico-terapêutico e/ou epidemiológico (tipo caso-controle) com a finalidade de investigar uma possível associação entre amostras do T. cruzi e diferentes formas clínicas da doença de Chagas.

The Emperor Has New Clothes: Empirical Tests of Mainstream Theories of Economic Growth

Modern macroeconomic theory utilises optimal control techniques to model the maximisation of individual well-being using a lifetime utility function. Agents face choices over current and future consumption (with resultant implied savings decisions) seeking to maximise the present value of current plus future well-being. However, such inter-temporal welfare-maximising assumptions remain empirically untested. In the work presented here we test whether welfare was in (historical) fact maximised in the US between 1870-2000 and find empirical support for the optimising basis of growth theory, but only once a comprehensive view of what constitutes a country’s wealth or capital is taken into account.

Evaluation of the xenodiagnosis of chronic Chagas patients infected ten years or over in an area where transmission has been interrupted - Iguatama and Pains, west Minas Gerais State, Brazil

To evaluate the results of xenodiagnosis in chronic Chagas patients infected for ten years or over in an area where transmission has been stemmed as well as the performance of these tests applied one or more times to determine the presence of the paraiste in serum-positive patients for Trypanosoma cruzi infection, 570 xenodiagnosis were performed in 246 patients by exoposing each pacient to 40 Triatoma infestans nymphs of 3 rd/4th stage once, twice or three times, at 30 days intervals. The 570 xenodiagnosis showed overall positive results in 50.7% with a peak 78% in patients under 20 years of age, and 60.5% in those over 60. Of the l58 patients who underwent three xenodiagnosis, 51 (32.3%) had three positive tests, 48 (30.3%) had all negative results, and the remainder had alternating positve and negative findings. There was no difference in number of positive results between the 1st, 2nd and 2rd tests; however, the 1st and 2nd trials added up to 53.2% and the sum total of all three trials was 57.7%.

Vitamin D has a direct immunomodulatory effect on CD8+ T cells of patients with early multiple sclerosis and healthy control subjects.

Little is known on a putative effect of vitamin D on CD8+ T cells. Yet, these cells are involved in the immmunopathogenesis of MS. We assessed the cytokine profile of EBV-specific CD8+ T cells of 10 early MS patients and 10 healthy control subjects with or without 1,25(OH)(2)D(3) and found that, with 1,25(OH)(2)D(3), these cells secreted less IFN-γ and TNF-α and more IL-5 and TGF-β. CD4+ T cell depletion or even culture with CD8+ T cells only did not abolish the immunomodulatory effect of 1,25(OH)(2)D(3) on CD8+ T cells, suggesting that 1,25(OH)(2)D(3) can act directly on CD8+ T cells.

Ectodysplasin has a dual role in ectodermal organogenesis: inhibition of Bmp activity and induction of Shh expression.

Ectodermal organogenesis is regulated by inductive and reciprocal signalling cascades that involve multiple signal molecules in several conserved families. Ectodysplasin-A (Eda), a tumour necrosis factor-like signalling molecule, and its receptor Edar are required for the development of a number of ectodermal organs in vertebrates. In mice, lack of Eda leads to failure in primary hair placode formation and missing or abnormally shaped teeth, whereas mice overexpressing Eda are characterized by enlarged hair placodes and supernumerary teeth and mammary glands. Here, we report two signalling outcomes of the Eda pathway: suppression of bone morphogenetic protein (Bmp) activity and upregulation of sonic hedgehog (Shh) signalling. Recombinant Eda counteracted Bmp4 activity in developing teeth and, importantly, inhibition of BMP activity by exogenous noggin partially restored primary hair placode formation in Eda-deficient skin in vitro, indicating that suppression of Bmp activity was compromised in the absence of Eda. The downstream effects of the Eda pathway are likely to be mediated by transcription factor nuclear factor-kappaB (NF-kappaB), but the transcriptional targets of Edar have remained unknown. Using a quantitative approach, we show in cultured embryonic skin that Eda induced the expression of two Bmp inhibitors, Ccn2/Ctgf (CCN family protein 2/connective tissue growth factor) and follistatin. Moreover, our data indicate that Shh is a likely transcriptional target of Edar, but, unlike noggin, recombinant Shh was unable to rescue primary hair placode formation in Eda-deficient skin explants.

Characterization of a Plasmodium falciparum mutant that has deleted the majority of the gametocyte-specific Pf11-1 locus

We identified a gametocyte-specific protein of Plasmodium falciparum called Pf11-1 and provide experimental evidence that this molecule is involved in the emergence of gametes of the infected erythrocyte (gametogenesis). A mutant parasite clone, which has deleted over 90% of the PF11-1 gene locus, was an important control to establish the gametocyte-specific expression of the Pf11-1. Molecular analysis of the Pf11-1 deletion indicates that it is presumably due a chromosome breakage with subsequent "healing" by the addition of telomeric heptanucleotides. Moreover, similar DNA rearrangements are observed in most of the laboratory isolates during asexual propagation in vitro.

c-Jun N-terminal kinase has a key role in Alzheimer disease synaptic dysfunction in vivo.

Altered synaptic function is considered one of the first features of Alzheimer disease (AD). Currently, no treatment is available to prevent the dysfunction of excitatory synapses in AD. Identification of the key modulators of synaptopathy is of particular significance in the treatment of AD. We here characterized the pathways leading to synaptopathy in TgCRND8 mice and showed that c-Jun N-terminal kinase (JNK) is activated at the spine prior to the onset of cognitive impairment. The specific inhibition of JNK, with its specific inhibiting peptide D-JNKI1, prevented synaptic dysfunction in TgCRND8 mice. D-JNKI1 avoided both the loss of postsynaptic proteins and glutamate receptors from the postsynaptic density and the reduction in size of excitatory synapses, reverting their dysfunction. This set of data reveals that JNK is a key signaling pathway in AD synaptic injury and that its specific inhibition offers an innovative therapeutic strategy to prevent spine degeneration in AD.
