Five different methods were critically examined to characterize the pore structure of the silica monoliths. The mesopore characterization was performed using: a) the classical BJH method of nitrogen sorption data, which showed overestimated values in the mesopore distribution and was improved by using the NLDFT method, b) the ISEC method implementing the PPM and PNM models, which were especially developed for monolithic silicas, that contrary to the particulate supports, demonstrate the two inflection points in the ISEC curve, enabling the calculation of pore connectivity, a measure for the mass transfer kinetics in the mesopore network, c) the mercury porosimetry using a new recommended mercury contact angle values. rnThe results of the characterization of mesopores of monolithic silica columns by the three methods indicated that all methods were useful with respect to the pore size distribution by volume, but only the ISEC method with implemented PPM and PNM models gave the average pore size and distribution based on the number average and the pore connectivity values.rnThe characterization of the flow-through pore was performed by two different methods: a) the mercury porosimetry, which was used not only for average flow-through pore value estimation, but also the assessment of entrapment. It was found that the mass transfer from the flow-through pores to mesopores was not hindered in case of small sized flow-through pores with a narrow distribution, b) the liquid penetration where the average flow-through pore values were obtained via existing equations and improved by the additional methods developed according to Hagen-Poiseuille rules. The result was that not the flow-through pore size influences the column bock pressure, but the surface area to volume ratio of silica skeleton is most decisive. Thus the monolith with lowest ratio values will be the most permeable. rnThe flow-through pore characterization results obtained by mercury porosimetry and liquid permeability were compared with the ones from imaging and image analysis. All named methods enable a reliable characterization of the flow-through pore diameters for the monolithic silica columns, but special care should be taken about the chosen theoretical model.rnThe measured pore characterization parameters were then linked with the mass transfer properties of monolithic silica columns. As indicated by the ISEC results, no restrictions in mass transfer resistance were noticed in mesopores due to their high connectivity. The mercury porosimetry results also gave evidence that no restrictions occur for mass transfer from flow-through pores to mesopores in the small scaled silica monoliths with narrow distribution. rnThe prediction of the optimum regimes of the pore structural parameters for the given target parameters in HPLC separations was performed. It was found that a low mass transfer resistance in the mesopore volume is achieved when the nominal diameter of the number average size distribution of the mesopores is appr. an order of magnitude larger that the molecular radius of the analyte. The effective diffusion coefficient of an analyte molecule in the mesopore volume is strongly dependent on the value of the nominal pore diameter of the number averaged pore size distribution. The mesopore size has to be adapted to the molecular size of the analyte, in particular for peptides and proteins. rnThe study on flow-through pores of silica monoliths demonstrated that the surface to volume of the skeletons ratio and external porosity are decisive for the column efficiency. The latter is independent from the flow-through pore diameter. The flow-through pore characteristics by direct and indirect approaches were assessed and theoretical column efficiency curves were derived. The study showed that next to the surface to volume ratio, the total porosity and its distribution of the flow-through pores and mesopores have a substantial effect on the column plate number, especially as the extent of adsorption increases. The column efficiency is increasing with decreasing flow through pore diameter, decreasing with external porosity, and increasing with total porosity. Though this tendency has a limit due to heterogeneity of the studied monolithic samples. We found that the maximum efficiency of the studied monolithic research columns could be reached at a skeleton diameter of ~ 0.5 µm. Furthermore when the intention is to maximize the column efficiency, more homogeneous monoliths should be prepared.rn

I crescenti volumi di traffico che interessano le pavimentazioni stradali causano sollecitazioni tensionali di notevole entità che provocano danni permanenti alla sovrastruttura. Tali danni ne riducono la vita utile e comportano elevati costi di manutenzione. Il conglomerato bituminoso è un materiale multifase composto da inerti, bitume e vuoti d'aria. Le proprietà fisiche e le prestazioni della miscela dipendono dalle caratteristiche dell'aggregato, del legante e dalla loro interazione. L’approccio tradizionalmente utilizzato per la modellazione numerica del conglomerato bituminoso si basa su uno studio macroscopico della sua risposta meccanica attraverso modelli costitutivi al continuo che, per loro natura, non considerano la mutua interazione tra le fasi eterogenee che lo compongono ed utilizzano schematizzazioni omogenee equivalenti. Nell’ottica di un’evoluzione di tali metodologie è necessario superare questa semplificazione, considerando il carattere discreto del sistema ed adottando un approccio di tipo microscopico, che consenta di rappresentare i reali processi fisico-meccanici dai quali dipende la risposta macroscopica d’insieme. Nel presente lavoro, dopo una rassegna generale dei principali metodi numerici tradizionalmente impiegati per lo studio del conglomerato bituminoso, viene approfondita la teoria degli Elementi Discreti Particellari (DEM-P), che schematizza il materiale granulare come un insieme di particelle indipendenti che interagiscono tra loro nei punti di reciproco contatto secondo appropriate leggi costitutive. Viene valutata l’influenza della forma e delle dimensioni dell’aggregato sulle caratteristiche macroscopiche (tensione deviatorica massima) e microscopiche (forze di contatto normali e tangenziali, numero di contatti, indice dei vuoti, porosità, addensamento, angolo di attrito interno) della miscela. Ciò è reso possibile dal confronto tra risultati numerici e sperimentali di test triassiali condotti su provini costituiti da tre diverse miscele formate da sfere ed elementi di forma generica.

It is currently widely accepted that the understanding of complex cell functions depends on an integrated network theoretical approach and not on an isolated view of the different molecular agents. Aim of this thesis was the examination of topological properties that mirror known biological aspects by depicting the human protein network with methods from graph- and network theory. The presented network is a partial human interactome of 9222 proteins and 36324 interactions, consisting of single interactions reliably extracted from peer-reviewed scientific publications. In general, one can focus on intra- or intermodular characteristics, where a functional module is defined as "a discrete entity whose function is separable from those of other modules". It is found that the presented human network is also scale-free and hierarchically organised, as shown for yeast networks before. The interactome also exhibits proteins with high betweenness and low connectivity which are biologically analyzed and interpreted here as shuttling proteins between organelles (e.g. ER to Golgi, internal ER protein translocation, peroxisomal import, nuclear pores import/export) for the first time. As an optimisation for finding proteins that connect modules, a new method is developed here based on proteins located between highly clustered regions, rather than regarding highly connected regions. As a proof of principle, the Mediator complex is found in first place, the prime example for a connector complex. Focusing on intramodular aspects, the measurement of k-clique communities discriminates overlapping modules very well. Twenty of the largest identified modules are analysed in detail and annotated to known biological structures (e.g. proteasome, the NFκB-, TGF-β complex). Additionally, two large and highly interconnected modules for signal transducer and transcription factor proteins are revealed, separated by known shuttling proteins. These proteins yield also the highest number of redundant shortcuts (by calculating the skeleton), exhibit the highest numbers of interactions and might constitute highly interconnected but spatially separated rich-clubs either for signal transduction or for transcription factors. This design principle allows manifold regulatory events for signal transduction and enables a high diversity of transcription events in the nucleus by a limited set of proteins. Altogether, biological aspects are mirrored by pure topological features, leading to a new view and to new methods that assist the annotation of proteins to biological functions, structures and subcellular localisations. As the human protein network is one of the most complex networks at all, these results will be fruitful for other fields of network theory and will help understanding complex network functions in general.

Im Rahmen dieser Arbeit konnten wichtige Schritte auf dem Weg zur asymmetrischen Totalsynthese des Cripowellin-Aglycons gemacht und wichtige Erkenntnisse gewonnen werden. Es gelang dabei, einen Teil des Grundgerüsts des Kerns mit dem entscheidenden stereogenen Zentrum optisch aktiv aufzubauen. Die Aza-Claisen-Umlagerung diente hierbei als Schlüsselschritt zum stereoselektiven Aufbau eines Epoxids. Für die Aza-Claisen-Umlagerung dienten ein Allylamin und ein Säurefluorid als Ausgangsverbindungen. Das Allylamin konnte ausgehend von Piperonylalkohol über sechs Stufen (Schutzgruppen-operationen, Bromierung, metallorganische Reaktionen, Reduktion) mit einem Auxliar versehen und mit einer Gesamtausbeute von 56 % synthetisiert werden. Die Synthese des Säurefluorids konnte im Rahmen dieser Arbeit entscheidend verbessert werden, insbesondere bezüglich der Reinheit. Die Aza-Claisen-Umlagerung zu einem Pentensäurederivat selbst konnte mit einer sehr befriedigenden Ausbeute von 91 % durchgeführt werden. Dabei entstanden zwei Diastereomere im Verhältnis 92:8. Kristallisationsversuche schlugen leider fehl, sodass der Konfigurationsbeweis noch aussteht. Dieses Pentensäurederivat konnte zunächst durch einen Reduktionsschritt und saure Abspaltung der Schutzgruppe und des Auxiliars in ein Benzylchlorid überführt werden, welches dann durch Umsetzung mit Ammoniak zu einem Benzazepinon reagierte, ein weiteres wichtiges Schlüsselintermediat in dieser Synthese. Mit diesem Benzazepinon wurden viele Versuche zur Doppelbindungsfunk-tionalisierung unternommen, so z. B. Dihydroxylierungen und Epoxidierungen. Auch wurde das Amid zum Amin reduziert und mit Pentensäure erneut funktionalisiert. Mit dem so gewonnenen Dien wurden erste Versuche unternommen, über eine Grubbs-Metathese das bicyclische System des Cripowellins aufzubauen, die jedoch scheiterten.

The proximal femur is a high-diversity region of the human skeleton, especially at the anterior junction between head and neck, where various bony morphologies have been recognized since mid nineteenth century. Classical literature on this topic is chaotic and contradictory, making almost impossible the comparison of data from different researches. Starting from an extensive bibliographic review, the first standardized method to score these traits has been created. This method allows representing both the anatomical diversity of the region already described in literature and a part of variability not considered before, giving few and univocal definitions and allowing to collect comparable data. The method has been applied to three identified and five archaeological European skeletal collections, with the aim of investigating the distribution of these features by sex, age and side, in different places and time periods. It has also been applied to 3D digital reconstructions of femurs from CT scan files of coxo-femoral joints from fresh cadavers. In addition to the osseous traits described in the standardized method, the presence and frequency of some features known as herniation pits have been scored both on bones and on CT scans. The various osseous traits of the proximal femur are present at similar frequencies in skeletal samples from different countries and different historical periods, even if with clear local differentiation. Some of the features examined show significant trends related to their distribution by gender and age. Some hypotheses are proposed about the etiology of these morphologies and their possible implication with the acquisition of bipedalism in Humans. It is therefore highlighted the possible relation of some of these traits with the development of disorders of the hip joint. Moreover, it is not recommended the use of any of these features as a specific activity-related marker.

The Mediterranean Sea is expected to react faster to global change compared to the ocean and is already showing more pronounced warming and acidification rates. A study performed along the Italian western coast showed that porosity of the skeleton increases with temperature in the zooxanthellate (i.e. symbiotic with unicellular algae named zooxanthellae) solitary scleractinian Balanophyllia europaea while it does not vary with temperature in the solitary non-zooxanthellate Leptopsammia pruvoti. These results were confirmed by another study that indicated that the increase in porosity was accompanied by an increase of the fraction of the largest pores in the pore-space, perhaps due to an inhibition of the photosynthetic process at elevated temperatures, causing an attenuation of calcification. B. europaea, L. pruvoti and the colonial non-zooxanthellate Astroides calycularis, transplanted along a natural pH gradient, showed that high temperature exacerbated the negative effect of lowered pH on their mortality rates. The growth of the zooxanthellate species did not react to reduced pH, while the growth of the two non-zooxanthellate species was negatively affected. Reduced abundance of naturally occurring B. europaea, a mollusk, a calcifying and a non-calcifying macroalgae were observed along the gradient while no variation was seen in the abundance of a calcifying green alga. With decreasing pH, the mineralogy of the coral and mollusk did not change, while the two calcifying algae decreased the content of aragonite in favor of the less soluble calcium sulphates and whewellite (calcium oxalate), possibly as a mechanism of phenotypic plasticity. Increased values of porosity and macroporosity with CO2 were observed in B. europaea specimens, indicating reduces the resistance of its skeletons to mechanical stresses with increasing acidity. These findings, added to the negative effect of temperature on various biological parameters, generate concern on the sensitivity of this zooxanthellate species to the envisaged global climate change scenarios.

Bone remodelling is a fundamental mechanism for removing and replacing bone during adaptation of the skeleton to mechanical loads. Skeletal unloading leads to severe hypoxia (1%O2) in the bone microenvironment resulting in imbalanced bone remodelling that favours bone resorption. Hypoxia, in vivo, is a physiological condition for osteocytes, 5% O2 is more likely physiological for osteocytes than 20% O2, as osteocytes are embedded deep inside the mineralized bone matrix. Osteocytes are thought to be the mechanosensors of bone and have been shown to orchestrate bone formation and resorption. Oxygen-deprived osteocytes seem undergo apoptosis and actively stimulate osteoclasts. Hypoxia and oxidative stress increase 150-kDa oxygen-regulated protein (ORP 150) expression in different cell types. It is a novel endoplasmic-reticulum-associated chaperone induced by hypoxia/ischemia. It well known that ORP 150 plays an important role in the cellular adaptation to hypoxia, as anti-apoptotic factor, and seems to be involved in osteocytes differentiations. The aims of the present study are 1) to determine the cellular and molecular response of the osteocytes at two different conditions of oxygen deprivation, 1% and 5% of O2 compared to the atmospheric oxygen concentration at several time points. 2) To clarify the role of hypoxic osteocytes in bone homeostasis through the detection of releasing of soluble factors (RANKL, OPG, PGE2 and Sclerostin). 3) To detect the activation of osteoclast and osteoblast induced by condition media collected from hypoxic and normoxic osteocytes. The data obtained in this study shows that hypoxia compromises the viability of osteocytes and induces apoptosis. Unlike in other cells types, ORP 150 in MLO-Y4 does not seem to be regulated early during hypoxia. The release of soluble factors and the evaluation of osteoclast and osteoblast activation shows that osteocytes, grown under severe oxygen deprivation, play a role in the regulation of both bone resorption and bone formation.

The composition of the atmosphere is frequently perturbed by the emission of gaseous and particulate matter from natural as well as anthropogenic sources. While the impact of trace gases on the radiative forcing of the climate is relatively well understood the role of aerosol is far more uncertain. Therefore, the study of the vertical distribution of particulate matter in the atmosphere and its chemical composition contribute valuable information to bridge this gap of knowledge. The chemical composition of aerosol reveals information on properties such as radiative behavior and hygroscopicity and therefore cloud condensation or ice nucleus potential. rnThis thesis focuses on aerosol pollution plumes observed in 2008 during the POLARCAT (Polar Study using Aircraft, Remote Sensing, Surface Measurements and Models, of Climate, Chemistry, Aerosols, and Transport) campaign over Greenland in June/July and CONCERT (Contrail and Cirrus Experiment) campaign over Central and Western Europe in October/November. Measurements were performed with an Aerodyne compact time-of-flight aerosol mass spectrometer (AMS) capable of online size-resolved chemical characterization of non-refractory submicron particles. In addition, the origins of pollution plumes were determined by means of modeling tools. The characterized pollution episodes originated from a large variety of sources and were encountered at distinct altitudes. They included pure natural emissions from two volcanic eruptions in 2008. By the time of detection over Western Europe between 10 and 12 km altitude the plume was about 3 months old and composed to 71 % of particulate sulfate and 21 % of carbonaceous compounds. Also, biomass burning (BB) plumes were observed over Greenland between 4 and 7 km altitude (free troposphere) originating from Canada and East Siberia. The long-range transport took roughly one and two weeks, respectively. The aerosol was composed of 78 % organic matter and 22 % particulate sulfate. Some Canadian and all Siberian BB plumes were mixed with anthropogenic emissions from fossil fuel combustion (FF) in North America and East Asia. It was found that the contribution of particulate sulfate increased with growing influences from anthropogenic activity and Asia reaching up to 37 % after more than two weeks of transport time. The most exclusively anthropogenic emission source probed in the upper troposphere was engine exhaust from commercial aircraft liners over Germany. However, in-situ characterization of this aerosol type during aircraft chasing was not possible. All long-range transport aerosol was found to have an O:C ratio close to or greater than 1 implying that low-volatility oxygenated organic aerosol was present in each case despite the variety of origins and the large range in age from 3 to 100 days. This leads to the conclusion that organic particulate matter reaches a final and uniform state of oxygenation after at least 3 days in the free troposphere. rnExcept for aircraft exhaust all emission sources mentioned above are surface-bound and thus rely on different types of vertical transport mechanisms, such as direct high altitude injection in the case of a volcanic eruption, or severe BB, or uplift by convection, to reach higher altitudes where particles can travel long distances before removal mainly caused by cloud scavenging. A lifetime for North American mixed BB and FF aerosol of 7 to 11 days was derived. This in consequence means that emission from surface point sources, e.g. volcanoes, or regions, e.g. East Asia, do not only have a relevant impact on the immediate surroundings but rather on a hemispheric scale including such climate sensitive zones as the tropopause or the Arctic.

All currently available human skeletal remains from the Wadi Howar (Eastern Sahara, Sudan) were employed in an anthropological study. The study’s first aim was to describe this unique 5th to 2nd millennium BCE material, which comprised representatives of all three prehistoric occupation phases of the region. Detecting diachronic differences in robusticity, occupational stress levels and health within the spatially, temporally and culturally heterogeneous sample was its second objective. The study’s third goal was to reveal metric and non-metric affinities between the different parts of the series and between the Wadi Howar material and other relevant prehistoric as well as modern African populations. rnThe reconstruction and comprehensive osteological analysis of 23 as yet unpublished individuals, the bulk of the Wadi Howar series, constituted the first stage of the study. The analyses focused on each individual’s in situ position, state of preservation, sex, age at death, living height, living weight, physique, biological ancestry, epigenetic traits, robusticity, occupational stress markers, health and metric as well as morphological characteristics. Building on the results of these efforts and the re-examination of the rest of the material, the Wadi Howar series as a whole, altogether 32 individuals, could be described. rnA wide variety of robusticity, occupational stress and health variables was evaluated. The pre-Leiterband (hunter-gatherer-fisher/hunter-gatherer-fisher-herder) and the Leiterband (herder-gatherer) data of over a third of these variables differed statistically significantly or in tendency from each other. The Leiterband sub-sample was characterised by higher enamel hypoplasia frequencies, lower mean ages at death and less pronounced expressions of occupational stress traits. This pattern was interpreted as evidence that the adoption and intensification of animal husbandry did probably not constitute reactions to worsening conditions. Apart from that, the relevant observations, noteworthy tendencies and significant differences were explained as results of a broader spectrum of pre-Leiterband subsistence activities and the negative side effects of the increasingly specialised herder-gatherer economy of the Leiterband phase. rnUsing only the data which could actually be collected from it, multiple, separate, individualised discriminant function analyses were carried out for each Wadi Howar skeleton to determine which prehistoric and which modern comparative sample it was most similar to. The results of all individual analyses were then summarised and examined as a whole. Thus it became possible to draw conclusions about the affinities the Wadi Howar material shared with prehistoric as well as modern populations and to answer questions concerning the diachronic links between the Wadi Howar’s prehistoric populations. When the Wadi Howar remains were positioned in the context of the selected prehistoric (Jebel Sahaba/Tushka, A-Group, Malian Sahara) and modern comparative samples (Southern Sudan, Chad, Mandinka, Somalis, Haya) in this fashion three main findings emerged. Firstly, the series as a whole displayed very strong affinities with the prehistoric sample from the Malian Sahara (Hassi el Abiod, Kobadi, Erg Ine Sakane, etc.) and the modern material from Southern Sudan and, to a lesser extent, Chad. Secondly, the pre-Leiterband and the Leiterband sub-sample were closer to the prehistoric Malian as well as the modern Southern Sudanese material than they were to each other. Thirdly, the group of pre-Leiterband individuals approached the Late Pleistocene sample from Jebel Sahaba/Tushka under certain circumstances. A theory offering explanations for these findings was developed. According to this theory, the entire prehistoric population of the Wadi Howar belonged to a Saharo-Nilotic population complex. The Jebel Sahaba/Tushka population constituted an old Nilotic and the early population of the Malian Sahara a younger Saharan part of this complex. The pre-Leiterband groups probably colonised the Wadi Howar from the east, either during or soon after the original Saharo-Nilotic expansion. Unlike the pre-Leiterband groups, the Leiterband people originated somewhere west of the Wadi Howar. They entered the region in the context of a later, secondary Saharo-Nilotic expansion. In the process, the incoming Leiterband groups absorbed many members of the Wadi Howar’s older pre-Leiterband population. The increasing aridification of the Wadi Howar region ultimately forced its prehistoric inhabitants to abandon the wadi. Most of them migrated south and west. They, or groups closely related to them, probably were the ancestors of the majority of the Nilo-Saharan-speaking pastoralists of modern-day Southern Sudan and Eastern Chad.

Die technische Silikatproduktion erfordert in der Regel hohe Temperaturen und extreme pH-Werte. In der Natur hingegen haben insbesondere Kieselschwämme die außergewöhnliche Fähigkeit, ihr Silikatskelett, das aus einzelnen sogenannten Spiculae besteht, enzymatisch mittels des Proteins Silicatein zu synthetisieren. rnIm Inneren der Spiculae, im zentralen Kanal, befindet sich das Axialfilament, welches hauptsächlich aus Silicatein-α aufgebaut ist. Mittels Antikörperfärbungen und Elektronenmikroskopischen Analysen konnte festgestellt werden, dass Silicatein in mit Kieselsäure-gefüllten Zellorganellen (silicasomes) nachzuweisen ist. Mittels dieser Vakuolen kann das Enzym und die Kieselsäure aus der Zelle zu den Spiculae im extrazellulären Raum befördert werden, wo diese ihre endgültige Länge und Dicke erreichen. Zum ersten Mal konnte nachgewiesen werden, dass rekombinant hergestelltes Silicatein-α sowohl als Siliciumdioxid-Polymerase als auch Siliciumdioxid-Esterase wirkt. Mittels Massenspektroskopie konnte die enzymatische Polymerisation von Kieselsäure nachverfolgt werden. Durch Spaltung der Esterbindung des künstlichen Substrates Bis(p-aminophenoxy)-dimethylsilan war es möglich kinetische Parameter der Siliciumdioxid-Esterase-Aktivität des rekombinanten Silicateins zu ermitteln.rnZu den größten biogenen Silikatstukuren auf der Erde gehören die Kieselnadeln der Schwammklasse Hexactinellida. Nadelextrakte aus den Schwammklassen Demospongien (S. domuncula) und Hexactinellida (M. chuni) wurden miteinander verglichen um die potentielle Existenz von Silicatein oder Silicatein-ähnliche Molekülen und die dazu gehörige proteolytischen Aktivität nachzuweisen. Biochemische Analysen zeigten, dass das 27 kDA große isolierte Polypeptid in Monoraphis mehrere gemeinsame Merkmale mit den Silicateinen der Demospongien teilt. Dazu gehören die Größe und die Proteinase-Aktivität. rnUm die Frage zu klären, ob das axiale Filament selbst zur Formbildung der Skelettelemente beiträgt, wurde ein neues mildes Extraktionsverfahren eingeführt. Dieses Verfahren ermöglichte die Solubilisierung des nativen Silicateins aus den Spiculae. Die isolierten Silicateine lagen als Monomere (24 kDa) vor, die Dimere durch nicht-kovalente Bindungen ausbildeten. Darüber hinaus konnten durch PAGE-Gelelektrophorese Tetramere (95 kDa) und Hexamere (135 kDa) nachgewiesen werden. Die Monomere zeigten eine beträchtliche proteolytische Aktivität, die sich während der Polymerisationsphase des Proteins weiter erhöhte. Mit Hilfe der Lichtmikroskopie und Elektronenmikroskopie (TEM) konnte die Assemblierung der Proteine zu filamentartigen Strukturen gezeigt werden. Die Selbstorganisation der Silicatein-α-Monomeren scheint eine Basis für Form- und Musterbildung der wachsenden Nadeln zu bilden.rn Um die Rolle des kürzlich entdeckten Proteins Silintaphin-1, ein starker Interaktionspartner des Silicatein-α, während der Biosilifizierung zu klären, wurden Assemblierungs-Experimente mit den rekombinanten Proteinen in vitro durchgeführt. Zusätzlich wurde deren Effekt auf die Biosilikatsynthese untersucht. Elektronenmikroskopische Analysen ergaben, dass rekombinantes Silicatein-α zufällig verteilte Aggregate bildet, während die Koinkubation beider Proteine (molekulares Verhältnis 4:1) über fraktal artige Strukturen zu Filamenten führt. Auch die enzymatische Aktivität der Silicatein-α-vermittelte Biosilikatsynthese erhöhte sich in Gegenwart von Silintaphin-1 um das 5,3-fache. rn

In dieser Arbeit wurde eine neue Methode zur asymmetrischen Substitution der K-Regionen von Pyren entwickelt, auf welcher das Design und die Synthese von neuartigen, Pyren-basierten funktionalen Materialien beruht. Eine Vielzahl von Substitutionsmustern konnte erfolgreich realisiert werden um die Eigenschaften entsprechend dem Verwendungszweck anzupassen. Der polyzyklische aromatische Kohlenwasserstoff (PAK) Pyren setzt sich aus vier Benzolringen in Form einer planaren Raute mit zwei gegenüberliegenden K-Regionen zusammen. Der synthetische Schlüsselschritt dieser Arbeit ist die chemische Transformation der einen K-Region zu einem α-Diketon und der darauffolgenden selektiven Bromierung der zweiten K-Region. Dieser asymmetrisch funktionalisierte Baustein zeichnet sich durch zwei funktionelle Gruppen mit orthogonaler Reaktivität aus und erweitert dadurch das Arsenal der etablierten Pyren Chemie um eine vielseitig einsetzbare Methode. Aufbauend auf diesem synthetischen Zugang wurden fünf wesentliche Konzepte auf dem Weg zu neuen, von Pyren abgeleiteten Materialen verfolgt: (i) Asymmterische Substitution mit elektronenziehenden versus -schiebenden Gruppen. (ii) Darstellung von Pyrenocyaninen durch Anbindung von Pyren mit einer der K-Regionen an das Phthalocyanin Gerüst zur Ausdehnung des π-Systems. (iii) Einführung von Thiophen an die K-Region um halbleitende Eigenschaften zu erhalten. (iv) Symmetrische Annullierung von PAKs wie Benzodithiophen und Phenanthren an beide K Regionen für cove-reiche und dadurch nicht-planare Strukturen. (v) Verwendung des K-Region-funktionalisierten Pyrens als Synthesebaustein für das Peri-Pentacen. Neben der Synthese wurde die Selbstorganisation in der Festphase und an der flüssig/fest Grenzfläche mittels zweidimensionaler Weitwinkel-Röntgenstreuung (2D WAXS) bzw. Rastertunnelmikroskopie (STM) untersucht. Die halbleitenden Eigenschaften wurden in organischen Feld-Effekt Transistoren (OFETs) charakterisiert.

Anguzykline sind eine große Gruppe von Naturstoffen. Ihnen ist gemein, dass sie eine Benz[a]anthracen-Struktur besitzen oder dass sie in der Biosynthese aus einer Verbindung mit einem solchem Grundgerüst hervorgegangen sind. Viele Vertreter der Anguzykline sind bioaktive Substanzen mit insbesondere antibiotischer Wirkung. In dieser Arbeit wurde eine flexible, modulare Synthese für Anguzykline erarbeitet. Eine Schlüsselreaktion stellte die intramolekulare [2+2+2]-Zykloaddition aus Triinen zu Phthaliden dar. Die in dieser Umwandlung eingesetzten Triine enthalten eine Diin- und eine Monoinkomponente, die über eine Esterbindung miteinander verknüpft sind. Die intramolekulare [2+2+2]-Zyklotrimerisierung wurde an zwei verschiedenen Strukturen der Triinsysteme untersucht: Zum einen Triine, in denen Nona-2,8-diinsäuren mit 1-Aryl-propargylalkoholen verknüpft waren und zum anderen Substrate, in denen die Propargylsäureester von 1-Aryl-nona-2,8-diin-1-olen gebildet wurden. Für die Umsetzung wurden zwei verschiedene Katalysatoren verwendet. Neben dem Wilkinsonkatalysator wurde der Rutheniumkatalysator [Cp*RuCl(cod)] eingesetzt. Beide Katalysatoren sind für die Reaktion geeignet und ergänzen sich hinsichtlich der verschiedenen Triinsysteme. Die Phthalide dienten als Intermediate für den Aufbau eines Tetrahydrobenz[a]anthrachinon-Grundgerüst. Zum Aufbau dieser Struktur wurde eine Synthesesequenz zur Gerüstumlagerung vorgestellt und die Synthesemethode auf verschiedene Vertreter der Anguzyklin-Familie angewendet. Besonderes Augenmerk wurde auf die Synthese des Urdamycinon B gelegt, in dem das Grundgerüst C-glykosidisch mit einem Olivosesubstituenten verknüpft ist.

Primitive kohlige Chondrite sind Meteorite, die seit ihrer Entstehung im frühen Sonnensystem kaum verändert wurden und dadurch einen Einblick in Prozesse geben, die zur Bildung und Veränderung der ersten festen Materie führten. Solche Prozesse können anhand von Bruchstücken dieser Meteorite detailliert im Labor studiert werden, sodass Rückschlüsse auf die Entwicklung unseres Sonnensystems im frühen Stadium getroffen werden können. Ca-, Al-reiche Einschlüsse (CAIs) aus chondritischen Meteoriten sind die ersten Festkörper des Sonnensystems und enthalten viele refraktäre Metallnuggets (RMNs), welche hauptsächlich aus den Elementen Os, Ir, Ru, Mo und Pt bestehen. Nach weit verbreiteter Ansicht sind diese Nuggets wahrscheinlich im Gleichgewicht mit dem solaren Nebel kondensiert, bereits früher oder gleichzeitig mit Oxiden und Silikaten. Die exakten Mechanismen, die zu ihren heute beobachteten Eigenschaften führten, sind allerdings unklar. Um frühere Arbeiten fortzuführen, wurde eine hohe Anzahl RMNs in vier unterschiedlichen Typen von Meteoriten detailliert studiert, darunter solche aus dem nahezu unveränderten Acfer 094, Allende (CV3ox), Leoville (CV3red) und Murchison (CM2). Die RMNs wurden in-situ, assoziiert mit ihren Wirtsmineralen und auch in Säurerückständen gefunden, deren Präparationsprozedur in dieser Arbeit speziell für RMNs durch eine zusätzliche Dichtetrennung verbessert wurde.rnDie Ergebnisse decken eine Reihe von Ungereimtheiten zwischen den beobachteten RMN-Eigenschaften und einer Kondensationsherkunft auf, sowohl für Kondensation in solarer Umgebung, als auch für Kondensation aus Material von Supernovae oder roten Riesen, für die die Kondensationssequenzen refraktärer Metalle speziell für diesen Vergleich berechnet wurden. Stattdessen wurden in dieser Arbeit neue Einblicke in die RMN-Entstehung und die Entwicklung der ersten Festkörper (CAIs) durch eine Kombination aus experimentellen, isotopischen, strukturellen und petrologischen Studien an RMNs gewonnen. Viele der beobachteten Eigenschaften sind mit Ausfällung der RMN aus einer CAI-Schmelze vereinbar. Ein solches Szenario wird durch entsprechende Untersuchungen an synthetisch hergestellten, mit refraktären Metallen im Gleichgewicht stehenden CAI-Schmelzen bestätigt. Es folgt aus den Ergebnissen, dass die Mehrzahl der RMNs isotopisch solar ist und alle untersuchten RMNs innerhalb von CAIs bei rascher Abkühlung (um bis zu 1000 °C/40 sek.) einer CAI-Schmelze gebildet wurden. rn

ZusammenfassungrnAlle bekannten Exemplare der Amphibien-Gruppe Microsauria des mitteleuropäischen Rotliegend wurden untersucht sowie – für die kladistische Untersuchung – ihre nächsten Verwandten. Im Saar-Nahe-Becken sind vier Arten aus zwei Microsauria-Gattungen nachzuweisen: Batropetes niederkirchensis n. sp., Batropetes palatinus n. sp., Batropetes appelensis n. sp. und Altenglanerpeton schroederi n. sp.. Aus dem Döhlen-Becken bei Dresden stammt Batropetes fritschi.rnDie Rekonstruktionen legen nahe, dass die in Seeablagerungen gefundenen Batropetes-Arten kleine, terrestrisch lebende Microsaurier waren, die sich von Insekten oder deren Larven ernährten. Ihre mittlere Gesamtlänge beträgt fast 8 cm, ihr Rumpf ist auffallend kurz mit je nach Art 17 oder 19 Präsakralwirbeln. Sie besitzen große von Wülsten umgebene Augenöffnungen, ein großes Pinealforamen, drei längs angeordnete Grübchen auf den Frontalia, dreispitzige Zähne, einen zahnlosen Gaumen und vier kurze kräftige Extremitäten. Becken- und Schultergürtel sind komplett verknöchert, außer den bei Batropetes appelensis und Batropetes fritschi knorpeligen Coracoid-Anteilen. Die Batropetes-Arten unterscheiden sich bezüglich Form, Proportionen bzw. Verknöcherungsgrad einiger Knochen im cranialen und postcranialen Skelett. rnDie kladistischen Untersuchungen ergeben für die Arten der monophyletische Gattung Batropetes, dass Batropetes appelensis den übrigen Arten gegenübersteht und Batropetes palatinus mit Batropetes fritschi eine Schwestergruppe zu Batropetes niederkirchensis bildet. Mit den etwas jüngeren texanischen Gattungen Carrolla und Quasicaecilia bildet Batropetes die monophyletische Familie Brachystelechidae. rnAltenglanerpeton schroederi, nur durch ein schlecht erhaltenes Exemplar überliefert, besitzt einen massiv gebauten Schädel, kaum länger als breit mit annähernd dreieckigem Umriss, ohne Anzeichen für Seitenlinien, mit kleiner rundlicher Orbita, sehr breiter Interorbitalregion und bis weit vor die Orbita reichendem Jugale. Sein langer Rumpf umfasst mindestens 30 lepospondyle Präsakralwirbel. Wegen der unvollkommenen Entwicklung der Gliedmaßen kann diese Art nicht auf vier Beinen gelaufen sein und lebte möglicherweise in der Laubschicht wühlend oder in Sümpfen. Altenglanerpeton ist keiner bestimmten Microsauria-Familie zuzuordnen und steht den Pantylidae, Gymnarthridae, Goniorhynchidae und Ostodolepidae am nächsten, wobei zu letzteren die meisten morphologischen Ähnlichkeiten bestehen.rnAußerdem sind mit isolierten Einzelknochen, insbesondere Wirbeln, Microsauria und andere Gruppen lepospondyler Amphibien ohne Bestimmung der Gattung und Art aus wenigen stratigraphischen Niveaus des Saar-Nahe-Beckens überliefert. rn

Indolizines and pyrroles are considered as “privileged” structures since their skeletons were found in many biologically active natural products and they possess a wide range of pharmaceutical properties. Syntheses of these small drug-like molecules are very important in medicinal chemistry. However, most existent methodologies are usually limited to specific substitution patterns or require impractical starting materials or expensive catalysts. Therefore, developing new methodologies for the synthesis of indolizines and pyrroles from commercially available or readily accessible sources is highly desirable.rnIn this PhD thesis, several methods has been described for the synthesis of indolizines and pyrroles. In the first part, indolizines carrying substituents in positions 1-3 were synthesized via a formal [3+2]-cycloaddition of pyridinium ylides and nitroalkenes. Pyridinium salts were prepared by N-alkylation of pyridines with cyanohydrin triflates which could be prepared from corresponding aldehydes via a Strecker reaction followed by O-triflylation. Nitroalkenes were simply prepared from the corresponding aldehydes and nitroalkanes in a nitroaldol condensation. Overall, this modular approach allows to construct the indolizine framework with various substitution patterns starting from a pyridine, two different aldehydes and a nitroalkane. In contrast to reported methods, the produced indolizines do not have to contain an electron-withdrawing group.rnIt has also been found that nitrile-stabilized 2-alkylpyridinium ylides cyclize to unstable 2-aminoindolizines via an intramolecular 5-exo-dig cyclization. Using an in situ acetylation of the amino group, N-protected 2-aminoindolizines could be synthesized. As a less common substitution pattern, indolizines carrying substituents in positions 5–8 were synthesized from enones and 2-(1H-pyrrol-1-yl)nitriles obtained from α-aminonitriles using a modified Paal-Knorr pyrrole synthesis. The decoration of the pyridine unit in the indolizine skeleton has been achieved by a one-pot conjugate addition/cycloaromatization sequence.rnIn the second part of the thesis, the diversity-oriented synthesis of pyrroles from 3,5-diaryl substituted 2H-pyrrole-2-carbonitriles (cyanopyrrolines) obtained in a cyclocondensation of enones with aminoacetonitrile hydrochloride is being discussed. 2,4-Di-, 2,3,5-trisubstituted pyrroles, pyrrole-2-carbonitriles and 2,2’-bipyrroles were synthesized in a one- or two-step protocol. While the microwave-assisted thermal elimination of HCN from cyanopyrrolines gave 2,4-disubstituted pyrroles, DDQ-oxidation of the same intermediates furnished pyrrole-2-carbonitriles. Furthermore, 2,3,5-trisubstituted pyrroles were obtained via a C-2-alkylation of the deprotonated cyanopyrrolines followed by the elimination of HCN. Finally, it has also been found that tetraaryl substituted 2,2’-bipyrroles could be synthesized by the oxidative dimerization of cyanopyrrolines using copper (II) acetate at 100 °C.rn
