Cryolithological, ground ice and fossil bioindicator (pollen, diatoms, plant macrofossils, rhizopods, insects, mammal bones) records from Bol'shoy Lyakhovsky Island permafrost sequences (73°20'N, 141°30'E) document the environmental history in the region for the past c. 115 kyr. Vegetation similar to modern subarctic tundra communities prevailed during the Eemian/Early Weichselian transition with a climate warmer than the present. Sparse tundra-like vegetation and harsher climate conditions were predominant during the Early Weichselian. The Middle Weichselian deposits contain peat and peaty soil horizons with bioindicators documenting climate amelioration. Although dwarf willows grew in more protected places, tundra and steppe vegetation prevailed. Climate conditions became colder and drier c. 30 kyr BP. No sediments dated between c. 28.5 and 12.05 14C kyr BP were found, which may reflect active erosion during that time. Herb and shrubby vegetation were predominant 11.6-11.3 14C kyr BP. Summer temperatures were c. 4 °C higher than today. Typical arctic environments prevailed around 10.5 14C kyr BP. Shrub alder and dwarf birch tundra were predominant between c. 9 and 7.6 kyr BP. Reconstructed summer temperatures were at least 4 °C higher than present. However, insect remains reflect that steppe-like habitats existed until c. 8 kyr BP. After 7.6 kyr BP, shrubs gradually disappeared and the vegetation cover became similar to that of modern tundra. Pollen and beetles indicate a severe arctic environment c. 3.7 kyr BP. However, Betula nana, absent on the island today, was still present. Together with our previous study on Bol'shoy Lyakhovsky Island covering the period between about 200 and 115 kyr, a comprehensive terrestrial palaeoenvironmental data set from this area in western Beringia is now available for the past two glacial-interglacial cycles.

The impact of ocean acidification and increased water temperature on marine ecosystems, in particular those involving calcifying organisms, has been gradually recognised. We examined the individual and combined effects of increased pCO2 (180 ppmV CO2, 380 ppmV CO2 and 750 ppmV CO2 corresponding to past, present and future CO2 conditions, respectively) and temperature (13 °C and 18 °C) during the exponential growth phase of the coccolithophore E. huxleyi using batch culture experiments. We showed that cellular production rate of Particulate Organic Carbon (POC) increased from the present to the future CO2 treatments at 13 °C. A significant effect of pCO2 and of temperature on calcification was found, manifesting itself in a lower cellular production rate of Particulate Inorganic Carbon (PIC) as well as a lower PIC:POC ratio at future CO2 levels and at 18 °C. Coccosphere-sized particles showed a size reduction with both increasing temperature and CO2concentration. The influence of the different treatments on coccolith morphology was studied by categorizing SEM coccolith micrographs. The number of well-formed coccoliths decreased with increasing pCO2 while temperature did not have a significant impact on coccolith morphology. No interacting effects of pCO2 and temperature were observed on calcite production, coccolith morphology or on coccosphere size. Finally, our results suggest that ocean acidification might have a larger adverse impact on coccolithophorid calcification than surface water warming.

The present study investigates the combined effect of phosphorous limitation, elevated partial pressure of CO2 (pCO2) and temperature on a calcifying strain of Emiliania huxleyi (PML B92/11) by means of a fully controlled continuous culture facility. Two levels of phosphorous limitation were consecutively applied by renewal of culture media (N:P = 26) at dilution rates (D) of 0.3 d- and 0.1 d-1. CO2 and temperature conditions were 300, 550 and 900 µatm pCO2 at 14 °C and 900 µatm pCO2 at 18 °C. In general, the steady state cell density and particulate organic carbon (POC) production increased with pCO2, yielding significantly higher concentrations in cultures grown at 900 µatm pCO2 compared to 300 and 550 µatm pCO2. At 900 µatm pCO2, elevation of temperature as expected for a greenhouse ocean, further increased cell densities and POC concentrations. In contrast to POC concentration, C-quotas (pmol C cell-1) were similar at D = 0.3 d-1 in all cultures. At D = 0.1 d-1, a reduction of C-quotas by up to 15% was observed in the 900 µatm pCO2 at 18 °C culture. As a result of growth rate reduction, POC:PON:POP ratios deviated strongly from the Redfield ratio, primarily due to an increase in POC. Ratios of particulate inorganic and organic carbon (PIC:POC) ranged from 0.14 to 0.18 at D = 0.3 d-1, and from 0.11 to 0.17 at D = 0.1 d-1, with variations primarily induced by the changes in POC. At D = 0.1 d-1, cell volume was reduced by up to 22% in cultures grown at 900 µatm pCO2. Our results indicate that changes in pCO2, temperature and phosphorus supply affect cell density, POC concentration and size of E. huxleyi (PML B92/11) to varying degrees, and will likely impact bloom development as well as biogeochemical cycling in a greenhouse ocean.

Within a larger program research work is being done on the history of settlement and landscape of the 'Siedlungskammer' Flögeln and the adjacent area. The 'Siedlungskammer' consists of an isolated pleistocene sand ground (Geest-island) surroundet by bogs. Starting from the edge of the Geest, near which large-scale archaeological excavations are carried out, three raised bog profiles were taken at 300, 500 and 4000 m off the prehistoric settlement. They were investigated by means of pollen analysis, and reflect in a decreasing way the activities of man on the Geestisland. Another pollen diagram from the nearby fen peat was worked out for comparison. At the same time it helped to date back a prehistoric sand path to the Roman period. The pollen diagrams cover the vegetational history without gaps from the early Atlantic period to modern times. The vegetation was decisively determined by the poor soils of this area. T'he pollen diagrams give evidence of the activity of settlers since the Neolithic age, with some gaps in the beginning, but later continuously from the middle of the Bronze age until the early migration period. The influence of the nearby settlement, which existed from the Birth of Christ to the 4/5th century, comes out distinctly. Among the cereals which were then cultivated here, there also was rye, at least in the 4/5th century, but most probably already during the Roman period. Besides that people cultivated barley, oats, and flax. The settlement break during the so-called dark ages between the 4/5th century and the time about 800 A.D. was confirmed by pollen analysis. During this time the area was once more covered by forests. The fluctuations of man's activities during the late Middle Ages and modern times, as they are made visible by pollen analysis, correspond to historically wellknown developments.

Here we test the hypothesis that calcification stabilizes the pH of the seawater proximate to the organisms, providing a level of acidification countering the detrimental basification that occurs during net photosynthesis. Such bioengineering provides a more stable pH environment for growth and fits the empirical evidence for changes in rates of calcification under different environmental conditions. Under this scenario, simulations suggest that the optimal production ratio of inorganic to organic particulate C (PIC : POCprod) will be lower (by approx. 20%) with ocean acidification and that overproduction of coccoliths in a future acidified ocean, where pH buffering is weaker, presents a risk to calcifying cells.

The global warming debate has sparked an unprecedented interest in temperature effects on coccolithophores. The calcification response to temperature changes reported in the literature, however, is ambiguous. The two main sources of this ambiguity are putatively differences in experimental setup and strain specificity. In this study we therefore compare three strains isolated in the North Pacific under identical experimental conditions. Three strains of Emiliania huxleyi type A were grown under non-limiting nutrient and light conditions, at 10, 15, 20 and 25 °C. All three strains displayed similar growth rate versus temperature relationships, with an optimum at 20–25 °C. Elemental production (particulate inorganic carbon (PIC), particulate organic carbon (POC), total particulate nitrogen (TPN)), coccolith mass, coccolith size, and width of the tube element cycle were positively correlated with temperature over the sub-optimum to optimum temperature range. The correlation between PIC production and coccolith mass/size supports the notion that coccolith mass can be used as a proxy for PIC production in sediment samples. Increasing PIC production was significantly positively correlated with the percentage of incomplete coccoliths in one strain only. Generally, coccoliths were heavier when PIC production was higher. This shows that incompleteness of coccoliths is not due to time shortage at high PIC production. Sub-optimal growth temperatures lead to an increase in the percentage of malformed coccoliths in a strain-specific fashion. Since in total only six strains have been tested thus far, it is presently difficult to say whether sub-optimal temperature is an important factor causing malformations in the field. The most important parameter in biogeochemical terms, the PIC : POC ratio, shows a minimum at optimum growth temperature in all investigated strains. This clarifies the ambiguous picture featuring in the literature, i.e. discrepancies between PIC : POC–temperature relationships reported in different studies using different strains and different experimental setups. In summary, global warming might cause a decline in coccolithophore's PIC contribution to the rain ratio, as well as improved fitness in some genotypes due to fewer coccolith malformations.

Research surrounding the transition from II to I millennium cal BC in Eastern Iberian Peninsula has a large and extensive tradition of investigation. However, the chances to do research on this historical process have been limited by the lack of a well contextualized and dated stratigraphic sequence. For this reason, recent studies in this topic have followed the periodic proposals which were developed in closer regions and areas, especially in the South East and North East of the Peninsula. The investigation perspective about the Late Bronze in Eastern Iberian has however now improved, with the development of several archaeological investigations, the increase in the number of sites being dated and more recent studies into the region helping to bring about this change. As such, it is now in the correct state to be able to propose a new periodization and delve into the changes which occurred in the transition between 1500 and 725 cal BC.

El objetivo de este trabajo es estudiar el primer tropiezo histórico de las tropas de Napoleón Bonaparte, en Santo Domingo, y los paralelismos entre la Guerra de Independencia española y la Reconquista dominicana. Para ello se han analizado los testimonios de los personajes implicados, con el fin de indagar el ideario que inspiró su acción militar. Además probaremos que, contra las ideas defendidas por algunos historiadores, tras la paz de Basilea los dominicanos siguieron sintiéndose ligados a España, como demostraron en la Guerra de Reconquista contra el gobierno francés. Sólo Cuba, Puerto Rico y Santo Domingo participaron de este sentimiento, mientras el resto de Hispanoamérica comenzaba a clamar por la independencia.

Este trabajo presenta la reelaboración de un modelo de producción de textos escritos, publicado por el Grupo Didactext en 2003. Se sitúa en un marco sociocognitivo, lingüístico y didáctico, y está concebido desde la interacción de tres dimensiones simbolizadas por círculos concéntricos recurrentes. El primer círculo corresponde al ámbito cultural: las diversas esferas de la praxis humana en las que está inmersa toda actividad de composición escrita. El segundo se refiere a los contextos de producción, de los que forman parte el contexto social, el situacional, el físico, la audiencia y el medio de composición. El tercer círculo corresponde al individuo, en el que se tiene en cuenta el papel de la memoria en la producción de un texto desde el enfoque sociocultural, la motivación, las emociones y las estrategias cognitivas y metacognitivas, dentro de las cuales se conciben seis unidades funcionales que actúan en concurrencia: acceso al conocimiento, planificación, redacción, revisión y reescritura, edición, y presentación oral. La orientación didáctica se interesa por la enseñanza y el aprendizaje de la escritura académica en las aulas, así como por la investigación de la escritura en contextos de educación.

Nuclear import of HIV-1 preintegration complexes (PICs) allows the virus to infect nondividing cells. Integrase (IN), the PIC-associated viral enzyme responsible for the integration of the viral genome into the host cell DNA, displays karyophilic properties and has been proposed to participate to the nuclear import of the PIC. Styrylquinolines (SQs) have been shown to block viral replication at nontoxic concentrations and to inhibit IN 3'-processing activity in vitro by competing with the DNA substrate binding. However, several lines of evidence suggested that SQs could have a postentry, preintegrative antiviral effect in infected cells. To gain new insights on the mechanism of their antiviral activity, SQs were assayed for their ability to affect nuclear import of HIV-1 IN and compared with the effect of a specific strand transfer inhibitor. Using an in vitro transport assay, we have previously shown that IN import is a saturable mechanism, thus showing that a limiting cellular factor is involved in this process. We now demonstrate that SQs specifically and efficiently inhibit in vitro nuclear import of IN without affecting other import pathways, whereas a specific strand transfer inhibitor does not affect IN import. These data suggest that SQs not only inhibit IN-DNA interaction but would also inhibit the interaction between IN and the cellular factor required for its nuclear import.

The karyophilic properties of the human immunodeficiency virus, type I (HIV-1) pre-integration complex (PIC) allow the virus to infect non-dividing cells. To better understand the mechanisms responsible for nuclear translocation of the PIC, we investigated nuclear import of HIV-1 integrase (IN), a PIC-associated viral enzyme involved in the integration of the viral genome in the host cell DNA. Accumulation of HIV-1 IN into nuclei of digitonin-permeabilized cells does not result from passive diffusion but rather from an active transport that occurs through the nuclear pore complexes. HIV-1 IN is imported by a saturable mechanism, implying that a limiting cellular factor is responsible for this process. Although IN has been previously proposed to contain classical basic nuclear localization signals, we found that nuclear accumulation of IN does not involve karyopherins alpha, beta1, and beta2-mediated pathways. Neither the non-hydrolyzable GTP analog, guanosine 5'-O-(thiotriphosphate), nor the GTP hydrolysis-deficient Ran mutant, RanQ69L, significantly affects nuclear import of IN, which depends instead on ATP hydrolysis. Therefore these results support the idea that IN import is not mediated by members of the karyopherin beta family. More generally, in vitro nuclear import of IN does not require addition of cytosolic factors, suggesting that cellular factor(s) involved in this active but atypical pathway process probably remain associated with the nuclear compartment or the nuclear pore complexes from permeabilized cells.

Le contexte de ce projet de recherche est celui de la stabilité des barrages-poids et aborde le besoin d’évaluation de la résistance de l’interface béton-rocher. Puisqu’il est techniquement difficile d’évaluer si l’interface est liée ou non, la cohésion réelle et sa contribution à la résistance au cisaillement sont souvent négligées et ce sujet précis est peu abordé dans la littérature. Un lien direct peut être fait entre cette non-considération et des travaux de stabilisation réalisés sur des ouvrages hydrauliques. Cette étude a comme objectif la caractérisation de la cohésion réelle dans le but de déterminer s’il est sécuritaire d’incorporer sa contribution dans l’évaluation de stabilité des barrages-poids. Pour ce faire, il est nécessaire d’évaluer les comportements en traction et en cisaillement de l’interface et d’analyser comment ils sont affectés par des paramètres importants telle la rugosité de l’interface. Cette caractérisation est faite à l’aide d’un programme expérimental sur 66 répliques d’interfaces béton-rocher en mortier. La rugosité est évaluée à l’aide d’un profilomètre laser et du paramètre Z2. Les répliques ont fait l’objet d’essais de traction directe, de traction par pression de fluide et de cisaillement direct. L’influence de la rugosité d’interface et de la résistance à la compression uniaxiale (UCS) des matériaux sur les résistances à la traction et au cisaillement est évaluée grâce à l’analyse des variances (ANOVA). Des essais supplémentaires ont permis d’approfondir la compréhension du mécanisme de rupture en cisaillement. Les résultats indiquent une résistance à la traction moyenne de l’interface liée de 0,62 MPa et une cohésion (en cisaillement) moyenne de 3,1 MPa. L’ANOVA montre une augmentation significative de la résistance à la traction avec la rugosité et une augmentation significative de la résistance au cisaillement au pic avec la rugosité, l’UCS et la contrainte normale. Il a aussi été observé que le pas d’échantillonnage a un impact important sur la valeur de Z2. Les résultats suggèrent qu’une valeur minimale de cohésion de 100 à 200 kPa pourrait être utilisée dans la mesure où il peut être démontré que l’interface est liée. Cette condition pourrait d’ailleurs constituer un sujet de recherche s’inscrivant dans la continuité des travaux réalisés.

Résumé : La capacité de décider parmi plusieurs possibilités d'actions, grâce à l'information sensorielle disponible, est essentielle à un organisme interagissant avec un environnement complexe. Les modèles actuels de sélection d'une action soutiennent que le cerveau traite continuellement l'information sensorielle afin de planifier plusieurs possibilités d'action en parallèle. Dans cette perspective, ces représentations motrices, associées à chaque possibilité d'action, sont en constante compétition entre elles. Afin qu'une alternative puisse être sélectionnée pour le mouvement, une valeur de pondération, intégrant une multitude de facteurs, doit être associée à chacun des plans moteurs afin de venir moduler la compétition. Plusieurs études se sont intéressées aux différents facteurs modulant la sélection de l'action, tels que la disposition de l'environnement, le coût des actions, le niveau de récompense, etc. Par contre, il semble qu'aucune étude n'ait rapporté ce qu'il advient lorsque la valeur de pondération de chacune des actions possibles est identique. Dans ce contexte, quel est l'élément permettant de venir moduler la sélection de l'action? De ce fait, l'objectif principal de mon projet de maitrise est d'investiguer le facteur permettant au cerveau de sélectionner une action lorsque tous les facteurs rapportés dans la littérature sont contrôlés. De récentes données ont montré que les oscillations corticales lentes dans la bande delta peuvent servir d'instrument de sélection attentionnelle en modulant l'amplitude de la réponse neuronale. Ainsi, les stimuli arrivant dans le cortex pendant une phase en delta de forte excitabilité sont amplifiés, tandis que ceux arrivant lors d'une phase en delta de faible excitabilité sont atténués. Ceci dit, il est possible que la phase en delta dans laquelle se trouve le cerveau au moment d'effectuer la sélection d'une action puisse influencer la décision. Utilisant une tâche de sélection de main, cette étude teste l'hypothèse que la sélection de la main est associée à la phase en delta des ensembles neuronaux codant le mouvement de chacune des mains, lorsque tous les facteurs connus influençant la décision sont contrôlés. L'électroencéphalographie (EEG) fut utilisée afin d'enregistrer les signaux corticaux pendant que les participants effectuaient une tâche de sélection de main dans laquelle ils devaient, à chaque essai, atteindre une cible visuelle aussi rapidement que possible en utilisant la main de leur choix. La tâche fut conçue de façon à ce que les facteurs spatiaux et biomécaniques soient contrôlés. Ceci fut réalisé enidentifiant premièrement, sur une base individuelle, l'emplacement de la cible pour laquelle les mains droite et gauche avaient une probabilité équivalente d'être choisies (point d'égalité subjective, PSE). Ensuite, dans l'expérience principale, les participants effectuaient plusieurs mouvements d'atteinte vers des cibles positionnées près et loin du PSE, toujours avec la main de leur choix. L'utilisation de cinq cibles très près du PSE a permis de collecter de nombreux essais dans lesquels la main droite et la main gauche furent sélectionnées en réponse à un même stimulus visuel. Ceci a ainsi permis d'analyser les signaux des deux cortex dans des conditions d'utilisation de la main droite et gauche, tout en contrôlant pour les autres facteurs pouvant moduler la sélection de la main. Les résultats de cette recherche révèlent que l'hémisphère cortical se trouvant dans la phase la plus excitable en delta (près du pic négatif), lors de l'apparition du stimulus, est associé à la fois à la main qui sera sélectionnée ainsi qu'au temps de réaction. Ces résultats montrent que l'excitabilité corticale momentanée (phase du signal) pourrait agir comme un facteur modulant la sélection d'une action. Dans cette optique, ces données élargissent considérablement les modèles actuels en montrant que la sélection d'une action est en partie déterminée par l'état du cerveau au moment d'effectuer un choix, d'une manière qui est indépendante de toutes les variables de décision connues.
