The environmental factors that contribute to the development of autoimmune diseases are largely unknown. Endemic pemphigus foliaceus in humans, known as Fogo Selvagem (FS) in Brazil, is mediated by pathogenic IgG4 autoantibodies against desmoglein 1 (Dsg1). Clusters of FS overlap with those of leishmaniasis, a disease transmitted by sand fly (Lutzomyia longipalpis) bites. In this study, we show that salivary Ags from the sand fly, and specifically the LJM11 salivary protein, are recognized by FS Abs. Anti-Dsg1 monoclonal autoantibodies derived from FS patients also cross-react with LJM11. Mice immunized with LJM11 generate anti-Dsg1 Abs. Thus, insect bites may deliver salivary Ags that initiate a cross-reactive IgG4 Ab response in genetically susceptible individuals and lead to subsequent FS. Our findings establish a clear relationship between an environmental, noninfectious Ag and the development of potentially pathogenic autoantibodies in an autoimmune disease. The photophysics of the 1-nitronaphthalene molecular system, after the absorption transition to the first singlet excited state, is theoretically studied for investigating the ultrafast multiplicity change to the triplet manifold. The consecutive transient absorption spectra experimentally observed in this molecular system are also studied. To identify the electronic states involved in the nonradiative decay, the minimum energy path of the first singlet excited state is obtained using the complete active space self-consistent field//configurational second-order perturbation approach. A near degeneracy region was found between the first singlet and the second triplet excited states with large spin-orbit coupling between them. The intersystem crossing rate was also evaluated. To support the proposed deactivation model the transient absorption spectra observed in the experiments were also considered. For this, computer simulations using sequential quantum mechanic-molecular mechanic methodology was used to consider the solvent effect in the ground and excited states for proper comparison with the experimental results. The absorption transitions from the second triplet excited state in the relaxed geometry permit to describe the transient absorption band experimentally observed around 200 fs after the absorption transition. This indicates that the T-2 electronic state is populated through the intersystem crossing presented here. The two transient absorption bands experimentally observed between 2 and 45 ps after the absorption transition are described here as the T-1 -> T-3 and T-1 -> T-5 transitions, supporting that the intermediate triplet state (T-2) decays by internal conversion to T-1. Myofibril proteins have excellent filmogenic properties. The objective of this article was to study the effect of the thermal treatment, of the pH and of the plasticizer concentration (Cp) of the filmogenic solution (FS), using over some physical properties of edible films, using a surface and response methodology (SRM). Films were made of lyophilized myofibril proteins (LMP) extracted from bovine muscle, employing the technique of solubility obtained from diluted saline solutions. The films were elaborated from FS containing 1 g of LMP/100g of FS and from Cp of 50 g to 79 g of glycerin/100 g of LMP. The LMP was dispersed in water under moderate agitation, and the pH was kept at 2.5-3.5 with the use of acetic acid. The FS were submitted to thermal treatment at different temperatures for 45 minutes. Films were dried in ventilated oven at 37 degrees C/18hr, conditioned at 75% of relative humidity at 25 degrees C/48 hr before analysis of: mechanical properties by puncture test; apparent opacity by spectrophotometer; solubility by immersion in water; and water vapor permeability by the gravimetric method. In general, films showed good appearance, translucent, easily handled and touchable, except for the films formed with pH 2.5 and at a low temperature (35 degrees C), with a medium thickness of 0.400 +/- 0.005 mm. The pH of the FS significantly affected all the physical properties under study. The temperature of the thermal treatment of the FS greatly affected the force at the rupture, solubility and water vapor permeability. This treatment can promote intermolecular interactions through the formation of disulphide bonds; however a very intense treatment can reverse this effect by irreversible structural alterations in the proteins. The glycerol concentration affected considerably all the properties under study, with the exception of the apparent opacity. Plasticizer increases the mobility of macromolecules with consequences in all physical properties.

The impact of leucine supplementation and resistance exercise (RE) on plasma lipid profile was evaluated in adult rats treated with dexamethasone, an experimental model of dyslipidemia. Total cholesterol did not differ among groups. Furthermore, leucine supplementation did not promote improvement in the plasma total cholesterol and LDL-c of the animals. However, plasma TG and VLDL-c were significantly decreased and HDL-c increased after 7 days of leucine supplementation combined with RE. In conclusion, leucine supplementation combined with RE, but not isolated, improved the plasma lipid profile of dexamethasone-induced dyslipidemic rats.

It is well established that autoantibodies against desmoglein 3 and desmoglein 1 (Dsg1) are relevant in the pathogenesis of pemphigus vulgaris and pemphigus foliaceus, including its endemic form fogo selvagem (FS). Isolated reports have shown that in certain patients with these diseases, autoantibodies against other desmosomal cadherins and E-cadherin may also be present. The goal of this investigation was to determine whether FS patients and normal individuals living in endemic areas possess autoantibodies against other desmosomal cadherins and E-cadherin. By testing a large number of FS and endemic control sera by ELISA, we found a consistent and specific autoantibody response against Dsg1 and other keratinocyte cadherins in these individuals, which is quite different from healthy individuals from the United States (US controls). Overall, the highest correlations among the autoantibody responses tested were in the endemic controls, followed by FS patients, and lowest in the US controls. These findings suggest that multiple, perhaps cross-reactive, keratinocyte cadherins are recognized by FS patients and endemic controls.

The aim of the present study was to estimate genetic parameters for flight speed and its association with growth traits in Nellore beef cattle. The flight speed (FS) of 7,402 yearling animals was measured, using a device composed of a pair of photoelectric cells. Time interval data (s) were converted to speed (m/s) and faster animals were regarded as more reactive. The growth traits analyzed were weaning weight (WW), ADG from weaning to yearling age, and yearling scrotal circumference (SC). The (co)variance components were estimated using REML in a multitrait analysis applying an animal model. The model included random direct additive genetic and residual effects, fixed effects of contemporary groups, age of dam (classes), and age of animal as covariable. For WW, the model also included maternal genetic and permanent environmental random effects. The direct heritability estimate for FS was 0.26 +/- 0.05 and direct heritability estimates for WW, SC, and ADG were 0.30 +/- 0.01, 0.48 +/- 0.02, and 0.19 +/- 0.01, respectively. Estimates of the genetic correlation between FS and the growth traits were -0.12 +/- 0.07 (WW), -0.13 +/- 0.08 (ADG), and -0.11 +/- 0.07 (SC). Although the values were low, these correlations showed that animals with better temperaments (slower FS) tended to present better performance. It is possible to infer that long-term selection for weight and scrotal circumference can promote a positive genetic response in the temperament of animals. Nevertheless, to obtain faster genetic progress in temperament, it would be necessary to perform direct selection for such trait. Flight speed is an easily measured indicator of temperament and can be included as a selection criterion in breeding programs for Nellore cattle.

Topical glucocorticoid (GC) therapy has been successfully used in the treatment of several common cutaneous diseases in clinical practice for a long time, and skin atrophy is one of the most typical cutaneous side effects of this therapy. The aim of this study was to evaluate the potential of noninvasive fluorescence spectroscopy (FS) technique in the detection and classification of GC-induced skin atrophy. A total of 20 male Wistar rats were used in the experimental protocol under controlled environmental conditions and with free access to food. One group received topical application of clobetasol propionate 0.05% for 14 days to induce cutaneous atrophy (atrophic group) and the other (control) group received only vehicle application following the same protocol and schedule. Histological analyses and FS measurements with laser excitation at both 532 nm and 408 nm were obtained on days 1 and 15. The FS results were classified as "normal" or "atrophic" according by histological analysis. Fluorescence spectra obtained with excitation at 408 nm allowed a clear distinction between the control and atrophic groups, and were more informative than the those obtained at 532 nm. Our results reveal that, if correctly applied, FS allows noninvasive evaluation of corticosteroid-induced skin atrophy, and thus represents an important step towards better monitoring of undesirable side effects of cutaneous therapy.

This study evaluated the effect of the systemic use of sodium alendronate in rats in vivo. Forty-five Wistar rats aged 36 to 42 days and weighing 200 to 230 g were randomly assigned to a control group (n = 20), which received distilled water, and an experimental group (n = 25), which received 2 weekly doses of 1 mg/kg of chemically pure sodium alendronate. The animals were killed after 60 days of treatment. The tibias were removed for analysis of bone mineral density by dual-energy X-ray absorptiometry (DXA). Then, the maxillary incisors were extracted for analysis of the mineralized dental tissues using fluorescence spectroscopy (FS), scanning electron microscopy (SEM), bright field microscopy (BFM), and cross-sectional microhardness (CSMH) testing. DXA and CSMH data were subjected to statistical analysis by Kruskal-Wallis test (5% significance level). The experimental group presented higher bone mineral density than the control group by DXA. FS analysis revealed presence of alendronate in the mineralized dental tissues of the specimens of the experimental group. Significant morphological differences were not found by SEM and BFM. Enamel and dentin (100 and 300 mu m from the dentinoenamel junction) CSMH data did not show significant difference between the control and experimental groups. Based on the obtained results, we conclude that while alendronate increased the bone mineral density and was incorporated into the mineralized dental tissues it did not cause significant alterations in the morphology and microhardness of rat incisor enamel and dentin. A variety of seemingly unrelated processes, such as core-mantle interaction, desulfurization, and direct precipitation from a silicate melt have been proposed to explain the formation of Ru-Os-Ir alloys (here referred to as osmiridiums) found in terrestrial mantle rocks. However, no consensus has yet been reached on how these important micrometer-sized phases form. In this paper we report the results of an experimental study on the solubilities of Ru, Os and Ir in sulfide melts (or mattes) as a function of alloy composition at 1300 degrees C. Considering the low solubilities of Ru, Os, and Ir in silicate melts, coupled with their high matte/silicate-melt partition coefficients, our results indicate that these elements concentrate initially at the ppm level in a matte phase in the mantle source region. During partial melting, the extraction of sulfur into silicate melt leads to a decrease in fS(2) that triggers the exsolution of osmiridiums from the refractory matte in the residue. The newly formed osmiridiums may persist in the terrestrial mantle for periods exceeding billions of years. (C) 2012 Elsevier Ltd. All rights reserved.

Abstract Background In the tephritids Ceratitis, Bactrocera and Anastrepha, the gene transformer provides the memory device for sex determination via its auto-regulation; only in females is functional Tra protein produced. To date, the isolation and characterisation of the gene transformer-2 in the tephritids has only been undertaken in Ceratitis, and it has been shown that its function is required for the female-specific splicing of doublesex and transformer pre-mRNA. It therefore participates in transformer auto-regulatory function. In this work, the characterisation of this gene in eleven tephritid species belonging to the less extensively analysed genus Anastrepha was undertaken in order to throw light on the evolution of transformer-2. Results The gene transformer-2 produces a protein of 249 amino acids in both sexes, which shows the features of the SR protein family. No significant partially spliced mRNA isoform specific to the male germ line was detected, unlike in Drosophila. It is transcribed in both sexes during development and in adult life, in both the soma and germ line. The injection of Anastrepha transformer-2 dsRNA into Anastrepha embryos caused a change in the splicing pattern of the endogenous transformer and doublesex pre-mRNA of XX females from the female to the male mode. Consequently, these XX females were transformed into pseudomales. The comparison of the eleven Anastrepha Transformer-2 proteins among themselves, and with the Transformer-2 proteins of other insects, suggests the existence of negative selection acting at the protein level to maintain Transformer-2 structural features. Conclusions These results indicate that transformer-2 is required for sex determination in Anastrepha through its participation in the female-specific splicing of transformer and doublesex pre-mRNAs. It is therefore needed for the auto-regulation of the gene transformer. Thus, the transformer/transfomer-2 > doublesex elements at the bottom of the cascade, and their relationships, probably represent the ancestral state (which still exists in the Tephritidae, Calliphoridae and Muscidae lineages) of the extant cascade found in the Drosophilidae lineage (in which tra is just another component of the sex determination gene cascade regulated by Sex-lethal). In the phylogenetic lineage that gave rise to the drosophilids, evolution co-opted for Sex-lethal, modified it, and converted it into the key gene controlling sex determination.

O objetivo deste estudo foi analisar a reprodutibilidade dos parâmetros biomecânicos da curva força-tempo do estilo "Crawl" em um protocolo de 10 s no nado atado. Dezesseis nadadores do sexo masculino (idade: 20,4 ± 4,0 anos; tempo na prova de 100 m livre: 53,68 ± 0,99 s) realizaram dois esforços máximos de 10 s no nado atado. Os parâmetros força pico, força média, taxa de desenvolvimento de força, impulso, duração da braçada, tempo para atingir a força pico e força mínima foram representados pela média de oito braçadas consecutivas obtidas em cada tentativa. Utilizou-se o teste t para observar as diferenças entre os esforços para cada parâmetro. O nível de significância estabelecido foi de 5%. A reprodutibilidade relativa foi medida pelo coeficiente de correlação de Pearson e a consistência entre as duas tentativas pelo coeficiente de correlação intraclasse (CCI). A reprodutibilidade absoluta foi verificada pelo coeficiente de variação (CV). Não foi demonstrada diferença estatisticamente significante para nenhum parâmetro biomecânico quando comparados os dois esforços. Os elevados CCI e baixos CV indicaram alta consistência interna dos parâmetros analisados. Conclui-se que os parâmetros biomecânicos analisados a partir do nado atado são reprodutíveis quando empregado protocolo de curta duração o que demonstra a possibilidade de utilização do protocolo com alto grau de confiabilidade, por parte de treinadores e atletas.

Este estudo investigou a aprendizagem de uma tarefa motora seriada em diferentes estágios de desenvolvimento. Quinze crianças, 14 adultos e 13 idosos praticaram a tarefa de rastrear uma sequência de seis estímulos luminosos durante 10 blocos de tentativas ou até descobrir a sequência, constituindo a fase de estabilização e mais dois blocos de tentativas, referentes as fases de adaptação I e II. O desempenho foi mensurado por meio das respostas funcionais e não-funcionais e das sequências funcionais. Os resultados indicaram que os adultos foram superiores aos demais participantes, e idosos apresentaram melhor desempenho que crianças apenas no início da prática, sugerindo que o estágio de desenvolvimento interage com o processo de aprendizagem motora.

O objetivo deste estudo foi investigar os efeitos de diferentes padrões respiratórios no desempenho e na organização temporal das braçadas do nado "Crawl" de atletas de elite. Vinte e um jovens atletas do gênero masculino participaram de forma voluntária do estudo. Eles foram filmados nadando 25 m do nado "Crawl", em velocidade máxima, em quatro condições de respiração: inspiração para o lado preferido (LP); para o lado não preferido (LNP); bilateral (B); e, sem respiração (S). Duas filmadoras digitais (60 fps) captaram as imagens que foram analisadas através do "software" Kinovea 0.8.7 nos 10 metros centrais do percurso. Para análise foram consideradas medidas de desempenho e de organização temporal das braçadas. Os resultados permitiram concluir que o padrão respiratório afeta o desempenho de atletas jovens de elite. Mostraram que, em termos de velocidade, tempo, frequência e comprimento de braçada, é mais vantajosa a condição de nadar "Crawl" em apneia (S) que em qualquer das condições com inspiração (B, LP e LNP). Portanto, o padrão preferido de respiração, não necessariamente, corresponde ao mais eficiente em termos do desempenho. Assim, professores e técnicos de Natação deveriam incentivar a prática e o emprego do padrão em apneia como estratégia tática para nadadores velocistas. Em relação à braçada, os resultados mostraram que diante da modificação do padrão respiratório, nadadores peritos jovens mantêm a estrutura temporal das braçadas direita e esquerda (aspectos invariantes da braçada) bem como a coordenação entre os braços inalterada enquanto efetuam ajustes na fase aérea da braçada (aspectos variantes). Assim sendo, atletas jovens de elite apresentam braçada com relativa autonomia frente ao componente respiração e nadam efetuando ajustes considerando a economia de recursos.

O presente ensaio tem por objetivo analisar os conceitos de abordagem de ensino e tendências pedagógicas no campo da Educação Física Escolar. A partir da exploração da literatura especializada, foi possível indagar sobre o grau de consistência interna das teorias pedagógicas produzidas nos anos 80 e 90 do século passado, como propostas sistematizadas para o ensino da Educação Física e suas implicações para o processo de formação docente. Desta forma, é proposto que a partir de uma aproximação acadêmica e profissional, seja possível promover um avanço na direção da sistematização do conhecimento.

O objetivo do presente estudo foi investigar a contribuição dos parâmetros biomecânicos para o desempenho do salto vertical com contramovimento (SV) e SV precedido de corrida (SVcorrida) em 19 jogadoras da seleção brasileira adulta de basquetebol feminino (26,2 ± 4,7 anos; 1,81 ± 0,07 m; 75,6 ± 12,6 kg; 20,4 ± 6,0% de gordura). Foram considerados os picos de força passiva (PFPa) e propulsão (PFP), tempo para alcance dos picos de força passiva (TPFPa) e propulsão (TPFP), "load rate" (LR), taxa de desenvolvimento de força (TDF), tempo de fase excêntrica (Texc) e concêntrica (Tcon). A análise de componentes principais revelou que 50,86% da altura de SV foi explicada por PFPa, TPFPa, LR, Texc e TPFP, e que 43,28% de SVcorrida foi explicada por PFPa, TPFPa, LR, PFP. Esses resultados sugerem que parâmetros temporais parecem contribuir de maneira mais significativa para o desempenho de salto, porém diferentes tipos de salto podem demandar comportamentos distintos de parâmetros biomecânicos.
