The aim of our study was to provide an innovative HS-GC/MS method applicable to the routine determination of butane concentration in forensic toxicology laboratories. The main drawback of the GC/MS methods discussed in literature concerning butane measurement was the absence of a specific butane internal standard necessary to perform quantification. Because no stable isotope of butane is commercially available, it is essential to develop a new approach by an in situ generation of standards. To avoid the manipulation of a stable isotope-labelled gas, we have chosen to generate in situ an internal labelled standard gas (C(4)H(9)D) following the basis of the stoichiometric formation of butane by the reaction of deuterated water (D(2)O) with Grignard reagent butylmagnesium chloride (C(4)H(9)MgCl). This method allows a precise measurement of butane concentration and therefore, a full validation by accuracy profile was presented.

Obesity is heritable and predisposes to many diseases. To understand the genetic basis of obesity better, here we conduct a genome-wide association study and Metabochip meta-analysis of body mass index (BMI), a measure commonly used to define obesity and assess adiposity, in up to 339,224 individuals. This analysis identifies 97 BMI-associated loci (P < 5 × 10(-8)), 56 of which are novel. Five loci demonstrate clear evidence of several independent association signals, and many loci have significant effects on other metabolic phenotypes. The 97 loci account for ∼2.7% of BMI variation, and genome-wide estimates suggest that common variation accounts for >20% of BMI variation. Pathway analyses provide strong support for a role of the central nervous system in obesity susceptibility and implicate new genes and pathways, including those related to synaptic function, glutamate signalling, insulin secretion/action, energy metabolism, lipid biology and adipogenesis.

A new Early Triassic marine fauna is described from the Central Oman Mountains. The fauna is Griesbachian in age, on the basis of abundant conodonts and ammonoids, and was deposited in an oxygenated seamount setting off the Arabian platform margin. It is the first Griesbachian assemblage from a well-oxygenated marine setting and thus provides a test for the hypothesis that widespread anoxia prevented rapid recovery. The earliest Griesbachian (parvus zone) contains a low-diversity benthic fauna dominated by the bivalves Promyalina and Claraia. A similar level of recovery characterizes the immediate postextinction interval worldwide. However, the middle upper Griesbachian sedimentary rocks (isarcica and catinata zones) contain an incredibly diverse benthic fauna of bivalves, gastropods, articulate brachiopods, a new undescribed crinoid, echinoids, and ostracods. This fauna is more diverse and ecologically complex than the typical middle to late Griesbachian faunas described from oxygen-restricted settings worldwide. The level of postextinction recovery observed in the Oman fauna is not recorded elsewhere until the Spathian. These data support the hypothesis that the apparent delay in recovery after the end-Permian extinction event was due to widespread and prolonged benthic oxygen restriction: in the absence of anoxia, marine recovery is much faster.

The present review compiles positive MS fragmentation data of selected carotenoids obtained using various ionization techniques and matrices. In addition, new experimental data from the analysis of carotenoids in transgenic maize and rice callus are provided. Several carotenes and oxygen-functionalized carotenoids containing epoxy, hydroxyl, and ketone groups were ionized by atmospheric pressure chemical ionization (APCI)-tandem mass spectrometry (MS/MS) in positive ion mode. Thus, on the basis of the information obtained from the literature and our own experiments, we identified characteristic carotenoid ions that can be associated to functional groups in the structures of these compounds. In addition, pigments with a very similar structure were differentiated through comparison of the intensities of their fragments. The data provide a basis for the structural elucidation of carotenoids by mass spectrometry (MS).

The objective of this work was to combine the advantages of the dried blood spot (DBS) sampling process with the highly sensitive and selective negative-ion chemical ionization tandem mass spectrometry (NICI-MS-MS) to analyze for recent antidepressants including fluoxetine, norfluoxetine, reboxetine, and paroxetine from micro whole blood samples (i.e., 10 microL). Before analysis, DBS samples were punched out, and antidepressants were simultaneously extracted and derivatized in a single step by use of pentafluoropropionic acid anhydride and 0.02% triethylamine in butyl chloride for 30 min at 60 degrees C under ultrasonication. Derivatives were then separated on a gas chromatograph coupled with a triple-quadrupole mass spectrometer operating in negative selected reaction monitoring mode for a total run time of 5 min. To establish the validity of the method, trueness, precision, and selectivity were determined on the basis of the guidelines of the "Société Française des Sciences et des Techniques Pharmaceutiques" (SFSTP). The assay was found to be linear in the concentration ranges 1 to 500 ng mL(-1) for fluoxetine and norfluoxetine and 20 to 500 ng mL(-1) for reboxetine and paroxetine. Despite the small sampling volume, the limit of detection was estimated at 20 pg mL(-1) for all the analytes. The stability of DBS was also evaluated at -20 degrees C, 4 degrees C, 25 degrees C, and 40 degrees C for up to 30 days. Furthermore, the method was successfully applied to a pharmacokinetic investigation performed on a healthy volunteer after oral administration of a single 40-mg dose of fluoxetine. Thus, this validated DBS method combines an extractive-derivative single step with a fast and sensitive GC-NICI-MS-MS technique. Using microliter blood samples, this procedure offers a patient-friendly tool in many biomedical fields such as checking treatment adherence, therapeutic drug monitoring, toxicological analyses, or pharmacokinetic studies.

Producing high quality products and services is one of the key concerns in order to keep up with the competition in the global markets. Companies are putting a great effort on preventing customers having faulty products and services by any means. However, the total elimination of mistakes in manufacturing processes has always been a great challenge for quality management. In this thesis the applicability of poka-yoke methodology in reducing the number of quality failures in the case company has been studied. Poka-yoke stands for the mistake-proofing and is mainly developed for the purpose of eliminating human errors in manufacturing processes. Inspection techniques; judgment, informative and source inspection are in the core of this methodology. Mass customization and large configurability of products leads to situation where the root causes of quality problems may vary a lot. To study the effects of these factors extensive analysis of quality failures was conducted. Recommendations and proposals for further actions regarding problem solving processes and utilization of mistake-proofing methods were provided on the basis of the analysis.

In this thesis the main objective is to examine and model configuration system and related processes. When and where configuration information is created in product development process and how it is utilized in order-delivery process? These two processes are the essential part of the whole configuration system from the information point of view. Empirical part of the work was done as a constructive research inside a company that follows a mass customization approach. Data models and documentation are created for different development stages of the configuration system. A base data model already existed for new structures and relations between these structures. This model was used as the basis for the later data modeling work. Data models include different data structures, their key objects and attributes, and relations between. Representation of configuration rules for the to-be configuration system was defined as one of the key focus point. Further, it is examined how the customer needs and requirements information can be integrated into the product development process. Requirements hierarchy and classification system is presented. It is shown how individual requirement specifications can be connected for physical design structure via features by developing the existing base data model further.

A low-protein diet leads to functional and structural pancreatic islet alterations, including islet hypotrophy. Insulin-signaling pathways are involved in several adaptive responses by pancreatic islets. We determined the levels of some insulin-signaling proteins related to pancreatic islet function and growth in malnourished rats. Adult male Wistar rats (N = 20 per group) were fed a 17% protein (normal-protein diet; NP) or 6% protein (low-protein diet; LP), for 8 weeks. At the end of this period, blood glucose and serum insulin and albumin levels were measured. The morphometric parameters of the endocrine pancreas and the content of some proteins in islet lysates were determined. The β-cell mass was significantly reduced (≅65%) in normoglycemic but hypoinsulinemic LP rats compared to NP rats. Associated with these alterations, a significant 30% reduction in insulin receptor substrate-1 and a 70% increase in insulin receptor substrate-2 protein content were observed in LP islets compared to NP islets. The phosphorylated serine-threonine protein kinase (pAkt)/Akt protein ratio was similar in LP and NP islets. The phosphorylated forkhead-O1 (pFoxO1)/FoxO1 protein ratio was decreased by 43% in LP islets compared to NP islets (P < 0.05). Finally, the ratio of phosphorylated-extracellular signal-related kinase 1/2 (pErk1/2) to total Erk1/2 protein levels was decreased by 71% in LP islets compared to NP islets (P < 0.05). Therefore, the reduced β-cell mass observed in LP rats is associated with the reduction of phosphorylation in mitogenic-related signals, FoxO1 and Erk proteins. The cause/effect basis of this association remains to be determined.

The fragmentation processes in the mass spectra of a series of organophosphorus, organochlorine, thio and dithiocarbamate as well as a number of miscellaneous pesticides have been studied i n detail by using the Bendix timeof- flight, MS-12 single-focussing and MS-30 double-focussing mass spectrometers. Interpretation of all the spectra have been presented; their mode s of dissociation elucidated, aided by metastable transitions wherever possible and the structures of the various f ragmentation species postulated wherever f easible. The fragmentation mechanisms are based on the concepts of inductive, resonance and steric ef~ects. Multiple bond cleavages accompanied by simultaneous bond formation and rearrangement reactions involving cycli c t r ansition states have clarified t he formation of various ions . Due emphasis has been placed on the effect of the functional groups or substituents in altering the mass spectral behaviour of the pesticides as they form the basis for the identifi cation of the otherwise identical pesticides. The organophosphorus pesticides which have been studied include i) the phosphates (eg: DDVP and Phosdrin ); ii) phosphorothionates (eg: Parathion, 0-2, 4 dichloro phenyl 0, O-diethyl thionophosphate); iii) phosphorothioites (eg: Tributyl phosphorotrithioite); i V) phosphorothioates (eg: Ethion) and v) phosphorodithioates (eg: Carbophenolthion). Cleavages and rearrangements of the ester moiety dominate the spectrum of phosdrin while that of DDVP is + dominated by t he fragmentation modes of the (OH30)2P=0 + moiety. Fragmentation §f the (CH30)2P=S characterises the spectrum of (OH30)2"P -Cl while cleavages of the + (C2H50 )2P=S species mark the spectra of parathion and 0-2, 4- di chlorophenyl O, O-diethyl thiophosphate. The 0(, cl eavages of the thioether f unction rather than + cleavages of the (C2H50)2P=S signify the spectrum of carbophenolthion. Tributyl phosphorotrithioite behaves more like an aliphatic hydrocarbon than like the corresponding phosphites. The isopropyl and butyl esters of 2, 4 dichlorophenoxy acetic acid show cleavage and rearrangement ions typical of an ester. In spite of its structural similari ty to pp' - DDT and pp' - DDD, Kalthane has a completely different mass spectral behaviour due to the influence of its hydroxyl function. The thiocarbamate pesticides studied include Eptam and Perbulate. Both are structurally similar but having different alkyl substituents on nitrogen and sulphur. This structurQlsimilarity leads to similar types of (N-C), (O-S) and (S-alkyl cleavages). However, perbulate differs from Eptam in showing a rearrangement ion at mle 161 and in forming an isocyanate ion as the base peak. In Eptam the base peak i s the alkyl ion. The dithiocarbamate, Vegadex, resembles the thiocarbamates in undergoing simple cleavages but it differs from them in having a weak parent ion; in the formation of its base peak and in undergoing a series of rearrangement reactions. The miscellaneous pesticides studied include 1-Naphthalene acetic aCid- methyl ester, Fiperonyl butoxide and Allethrin. The ester i s stable to electron impact and shows only fewer ions. Piper onyl butoxide, a polyether, shows characteristics of an et her, alcohol and aldehyde . Allethrin is regarded as an ester of the type R-C-O-R1 with n R being a substituted cyclopr opane moiety and o Rt, a substituted cyclopentenone mOiety. Accordingly it shows cleavage ions typical of an aliphatic ester and undergoes bond ruptures of the cyclic moieties to give unusual ions. Its base peak is an odd electron ion, quite contrary to expectations.

Mit dieser Arbeit wurde die Selbstassemblierung von dia- und paramagnetischen Molekülen sowie Einzelmolekülmagneten auf Goldsubstraten und magnetisch strukturierten Substraten untersucht. Dazu wurden drei verschiedene Klassen an Phthalocyaninderivaten verwendet: Diamagnetische Subphthalocyanine, paramagnetische Phthalocyaninatometalle und Diphthalocyaninatolanthanidkomplexe. Alle synthetisierten Verbindungen sind peripher thioethersubstituiert. Die Alkylketten (a: n-C8H17, b: n-C12H25) vermitteln die Löslichkeit in vielen organischen Solventien und sorgen für eine geordnete Assemblierung auf einer Oberfläche, wobei die Bindung auf Gold hauptsächlich über die Schwefelatome stattfindet. Die aus Lösung abgeschiedenen selbstassemblierten Monolagen wurden mit XPS, NEXAFS-Spektroskopie und ToF-SIMS untersucht. Bei der Selbstassemblierung auf magnetisch strukturierten Substraten stehen die Moleküle unter dem Einfluss magnetischer Streufelder und binden bevorzugt nur in bestimmten Bereichen. Die gebildeten Submonolagen wurden zusätzlich mit X-PEEM untersucht. Die erstmals dargestellten Manganphthalocyanine [MnClPc(SR)8] 1 wurden ausgehend von MnCl2 erhalten. Hier fand bei der Aufarbeitung an Luft eine Oxidation zu Mangan(III) statt; +III ist die stabilste Oxidationsstufe von Mangan in Phthalocyaninen. Der Nachweis des axialen Chloridoliganden erfolgte mit Massenspektrometrie und FIR- sowie Raman-Spektroskopie. SQUID-Messungen haben gezeigt, dass die Komplexe 1 vier ungepaarte Elektronen haben. Bei den Subphthalocyaninen [BClSubpc(SR)6] 2 wurde der axiale Chloridoligand mit dem stäbchenförmigen Phenolderivat 29-H substituiert und die erfolgreiche Ligandensubstitution durch NMR- und IR-Spektroskopie sowie Massenspektrometrie an den Produkten [BSubpc(SR)6(29)] 30 belegt. Der Radikalcharakter der synthetisierten Terbiumkomplexe [Tb{Pc(SR)8}2] 3 wurde spektroskopisch nachgewiesen; SQUID-Messungen ergaben, dass es sich um Einzelmolekülmagnete mit einer Energiebarriere U des Doppelpotentialtopfs von 880 K oder 610 cm-1 bei 3a handelt. Zunächst wurden die SAMs der Komplexverbindungen 1, 2, 30 und 3 auf nicht magnetisch strukturierten Goldsubstraten untersucht. Die Manganphthalocyanine 1 bilden geordnete SAMs mit größtenteils flach liegenden Molekülen, wie die XPS-, NEXAFS- und ToF-SIMS-Analyse zeigte. Die Mehrzahl der Thioether-Einheiten ist auf Gold koordiniert und die Alkylketten zeigen ungeordnet von der Oberfläche weg. Bei der Adsorption findet eine Reduktion zu Mangan(II) statt und der axiale Chloridoligand wird abgespalten. Das beruht auf dem sog. Oberflächen-trans-Effekt. Im vorliegenden Fall übt die Metalloberfläche einen stärkeren trans-Effekt als der axiale Ligand aus, was bisher experimentell noch nicht beobachtet wurde. Die thioethersubstituierten Subphthalocyanine 2 und 30 sowie die Diphthalocyaninatoterbium-Komplexe 3 sind ebenfalls für SAMs geeignet. Ihre Monolagen wurden mit XPS und NEXAFS-Spektroskopie untersucht, und trotz einer gewissen Unordnung in den Filmen liegen die Moleküle jeweils im Wesentlichen flach auf der Goldoberfläche. Vermutlich sind bei diesen Systemen auch die Alkylketten größtenteils parallel zur Oberfläche orientiert. Im Gegensatz zu den Manganphthalocyaninen 1 tritt bei 2b, 30a, 30b und 3b neben der koordinativen Bindung der Schwefelatome auf Gold auch eine für Thioether nicht erwartete kovalente Au–S-Bindung auf, die durch C–S-Bindungsbruch unter Abspaltung der Alkylketten ermöglicht wird. Der Anteil, zu dem dieser Prozess stattfindet, scheint nicht mit der Molekülstruktur zu korrelieren. Selbstassemblierte Submonolagen auf magnetisch strukturierten Substraten wurden mit dem diamagnetischen Subphthalocyanin 2b hergestellt. Der Nachweis der Submonolagen war schwierig und gelang schließlich durch eine Kombination von ToF-SIMS, NEXAFS Imaging und X-PEEM. Die Analyse der ToF-SIMS-Daten zeigte, dass tatsächlich eine Modulation der Verteilung der Moleküle auf einem unterwärts magnetisch strukturierten Substrat eintritt. Mit X-PEEM konnte die magnetische Struktur der ferromagnetischen Schicht des Substrats direkt der Verteilung der adsorbierten Moleküle zugeordnet werden. Die Subphthalocyanine 2b adsorbieren nicht an den Domänengrenzen, sondern vermehrt dazwischen. Auf Substraten mit abwechselnd 6.5 und 3.5 µm breiten magnetischen Domänen binden die Moleküle bevorzugt in den Bereichen geringster magnetischer Streufeldgradienten, also den größeren Domänen. Solche Substrate wurden für die ToF-SIMS- und X-PEEM-Messungen verwendet. Bei größeren magnetischen Strukturen mit ca. 400 µm breiten Domänen, wie sie aufgrund der geringeren Ortsauflösung dieser Methode für NEXAFS Imaging eingesetzt wurden, binden die Moleküle dann in allen Domänen. Die diamagnetischen Moleküle werden nach dieser Interpretation aus dem inhomogenen Magnetfeld über der Probenoberfläche heraus gedrängt und verhalten sich analog makroskopischer Diamagnete. Die eindeutige Detektion der Moleküle auf den magnetisch strukturierten Substraten konnte bisher nur für die diamagnetischen Subphthalocyanine 2b erfolgen. Um die Interpretation ihres Verhaltens bei der Selbstassemblierung in einem inhomogenen Magnetfeld weiter voranzutreiben, wurde das Subphthalocyanin 37b dargestellt, welches ein stabiles organisches TEMPO-Radikal in seinem axialen Liganden enthält. Das paramagnetische Subphthalocyanin 37b sollte auf den magnetisch strukturierten Substraten in Regionen starker magnetischer Streufelder binden und damit das entgegengesetzte Verhalten zu den diamagnetischen Subphthalocyaninen 2b zeigen. Aus Zeitgründen konnte dieser Nachweis im Rahmen dieser Arbeit noch nicht erbracht werden.

Perchlorate-reducing bacteria fractionate chlorine stable isotopes giving a powerful approach to monitor the extent of microbial consumption of perchlorate in contaminated sites undergoing remediation or natural perchlorate containing sites. This study reports the full experimental data and methodology used to re-evaluate the chlorine isotope fractionation of perchlorate reduction in duplicate culture experiments of Azospira suillum strain PS at 37 degrees C (Delta Cl-37(Cr)--ClO4-) previously reported, without a supporting data set by Coleman et al. [Coleman, M.L., Ader, M., Chaudhuri, S., Coates,J.D., 2003. Microbial Isotopic Fractionation of Perchlorate Chlorine. Appl. Environ. Microbiol. 69, 4997-5000] in a reconnaissance study, with the goal of increasing the accuracy and precision of the isotopic fractionation determination. The method fully described here for the first time, allows the determination of a higher precision Delta Cl-37(Cl)--ClO4- value, either from accumulated chloride content and isotopic composition or from the residual perchlorate content and isotopic composition. The result sets agree perfectly, within error, giving average Delta Cl-37(Cl)--ClO4- = -14.94 +/- 0.15%omicron. Complementary use of chloride and perchlorate data allowed the identification and rejection of poor quality data by applying mass and isotopic balance checks. This precise Delta Cl-37(Cl)--ClO4-, value can serve as a reference point for comparison with future in situ or microcosm studies but we also note its similarity to the theoretical equilibrium isotopic fractionation between a hypothetical chlorine species of redox state +6 and perchlorate at 37 degrees C and suggest that the first electron transfer during perchlorate reduction may occur at isotopic equilibrium between art enzyme-bound chlorine and perchlorate. Prediction of the solar wind conditions in near-Earth space, arising from both quasi-steady and transient structures, is essential for space weather forecasting. To achieve forecast lead times of a day or more, such predictions must be made on the basis of remote solar observations. A number of empirical prediction schemes have been proposed to forecast the transit time and speed of coronal mass ejections (CMEs) at 1 AU. However, the current lack of magnetic field measurements in the corona severely limits our ability to forecast the 1 AU magnetic field strengths resulting from interplanetary CMEs (ICMEs). In this study we investigate the relation between the characteristic magnetic field strengths and speeds of both magnetic cloud and noncloud ICMEs at 1 AU. Correlation between field and speed is found to be significant only in the sheath region ahead of magnetic clouds, not within the clouds themselves. The lack of such a relation in the sheaths ahead of noncloud ICMEs is consistent with such ICMEs being skimming encounters of magnetic clouds, though other explanations are also put forward. Linear fits to the radial speed profiles of ejecta reveal that faster-traveling ICMEs are also expanding more at 1 AU. We combine these empirical relations to form a prediction scheme for the magnetic field strength in the sheaths ahead of magnetic clouds and also suggest a method for predicting the radial speed profile through an ICME on the basis of upstream measurements.

Objectives: Our objective was to test the performance of CA125 in classifying serum samples from a cohort of malignant and benign ovarian cancers and age-matched healthy controls and to assess whether combining information from matrix-assisted laser desorption/ionization (MALDI) time-of-flight profiling could improve diagnostic performance. Materials and Methods: Serum samples from women with ovarian neoplasms and healthy volunteers were subjected to CA125 assay and MALDI time-of-flight mass spectrometry (MS) profiling. Models were built from training data sets using discriminatory MALDI MS peaks in combination with CA125 values and tested their ability to classify blinded test samples. These were compared with models using CA125 threshold levels from 193 patients with ovarian cancer, 290 with benign neoplasm, and 2236 postmenopausal healthy controls. Results: Using a CA125 cutoff of 30 U/mL, an overall sensitivity of 94.8% (96.6% specificity) was obtained when comparing malignancies versus healthy postmenopausal controls, whereas a cutoff of 65 U/mL provided a sensitivity of 83.9% (99.6% specificity). High classification accuracies were obtained for early-stage cancers (93.5% sensitivity). Reasons for high accuracies include recruitment bias, restriction to postmenopausal women, and inclusion of only primary invasive epithelial ovarian cancer cases. The combination of MS profiling information with CA125 did not significantly improve the specificity/accuracy compared with classifications on the basis of CA125 alone. Conclusions: We report unexpectedly good performance of serum CA125 using threshold classification in discriminating healthy controls and women with benign masses from those with invasive ovarian cancer. This highlights the dependence of diagnostic tests on the characteristics of the study population and the crucial need for authors to provide sufficient relevant details to allow comparison. Our study also shows that MS profiling information adds little to diagnostic accuracy. This finding is in contrast with other reports and shows the limitations of serum MS profiling for biomarker discovery and as a diagnostic tool

It has been known for decades that the metabolic rate of animals scales with body mass with an exponent that is almost always <1, >2/3, and often very close to 3/4. The 3/4 exponent emerges naturally from two models of resource distribution networks, radial explosion and hierarchically branched, which incorporate a minimum of specific details. Both models show that the exponent is 2/3 if velocity of flow remains constant, but can attain a maximum value of 3/4 if velocity scales with its maximum exponent, 1/12. Quarterpower scaling can arise even when there is no underlying fractality. The canonical “fourth dimension” in biological scaling relations can result from matching the velocity of flow through the network to the linear dimension of the terminal “service volume” where resources are consumed. These models have broad applicability for the optimal design of biological and engineered systems where energy, materials, or information are distributed from a single source.
