基于OIF-VSR5-01.0规范,分析了12路并行40Gb/s甚短距离(VSR)光传输转换器模块的实现原理.采用top-down分析方法,使用硬件描述语言verilog,在可编程逻辑器件上完成了时钟数据恢复、基于字节对齐方案的帧同步、信道去斜移、比特间差奇偶校验(BIP)等功能模块的程序设计,实现了SFI-5与OIF-VSR5-01.0电信号格式的相互转换,并在Altera的Stratix II GX 系列的高速现场可编程门阵列(FPGA)上对功能模块进行了功能验证和联合仿真.结果表明所设计的各个功能模块满足系统应用要求,为下一步将系统设计转换为专用集成电路(ASIC)奠定了基础.

本学位论文报道了作为传统藏药材广泛使用的西藏产雪莲花化学成分的研究。论文由五章组成,第一章是三种西藏产雪莲花的化学成分的系统分离纯化和结构鉴定;第二章为西藏产雪莲花化学成分的液-质及串联质谱联用分析;第三章提出了以HPLC和TLC为检测方法的雪莲花药材质量标准草案;第四章给出了对西藏产雪莲花挥发油化学成分的气-质联用分析结果;第五章概述了雪莲花的化学成分及药理研究进展。 第一章包括三个部分。第一部分报道了绵头雪莲花(Saussurea laniceps Hand.-Mazz.)全草乙醇提取物化学成分的分离鉴定。采用正相硅胶柱层析及凝胶柱层析等分离方法,从西藏产绵头雪莲花的乙醇提取物中共分离鉴定出15个化合物。其中11个化合物为首次从该植物中分离得到,当中2个化合物系在凤毛菊属植物中首次发现。第二部分报道了水母雪莲花(Saussurea medusa Maxim.)全草乙醇提取物的化学成分。采用正、反相硅胶柱层析及凝胶柱层析等分离方法,共分离鉴定出15个化合物,其中1个为新化合物,另有4个化合物为首次从该植物中分离得到。新化合物结构通过质谱和一维及二维核磁共振等波谱解析方法及碱水解反应确定为巴豆酰基-高车前苷(M-7)。第三部分报道了三指雪莲花 (Saussurea tridactyla Sch.-Bip. ex Hook. f.)全草乙醇提取物的化学成分。采用正相硅胶柱层析及凝胶柱层析等分离方法,共分离鉴定出7个化合物,其中1个化合物为首次从该植物中分离得到。 第二章也包括三个部分。首先是采用液-质联用(HPLC-DAD-ESI-MSn)分析方法,对7个西藏不同产地的三指雪莲花化学成分进行了分析,通过与标准品的 UV和MS数据比较,共鉴定出14个峰,并对其中8个共有成分进行了定量测定。其次是关于八种西藏产雪莲花化学成分的液-质联用(HPLC-DAD-ESI-MSn)分析,通过与标准品的UV和MS数据比较,共鉴定出15个峰,并对其中8个共有成分进行了定量检测。最后通过对八种西藏产雪莲花主要化学成分的多级串联质谱(ESI-MSn)分析,快速、灵敏地鉴定出10个黄酮和3个香豆素化学成分。 第三章同样包括三个部分。首先是以绵头雪莲花中主要香豆素成分东莨菪素和伞形花内酯为对照品,通过TLC定性检测和HPLC含量测定,草拟出较严谨的药材质量标准。其次是将绵头雪莲花、三指雪莲花和雪兔子作为一个药材看待,草拟了以东莨菪素和伞形花内酯的TLC检测为指标的药材质量标准。最后是针对水母雪莲花,以主要黄酮成分芹菜素-7-O-b-D-葡萄糖苷为对照品作TLC检测,并草拟出该药材的质量标准草案。 第四章报道了西藏产雪莲花挥发油的化学成分分析。采用传统水蒸气蒸馏法分别从八种雪莲花全草中提取挥发油,利用气相色谱-质谱联用技术分别从水母雪莲花、绵头雪莲花、槲叶雪莲花、云状雪兔子、拉萨雪兔子、小果雪兔子、雪兔子和三指雪莲花中分别鉴定出83、83、56、34、21、20、24和20个化学成分,分别占其挥发油总量的70.7%、76.0%、82.2%、55.4%、49.7%、70.4 %、76.2%和 76.7%。 第五章为综述,总结和概括了雪莲花的化学和药理研究进展。 The dissertation reports the investigation of the chemical constituents of the genus Saussurea. Quite a lot of species in this genus are traditional Tibetan medicinal plants, and hence have been widely used in traditional Tibetan medicine. This dissertation consisted of five chapters. The first chapter is on the chemical constituents of three Saussurea plants. The second section is about the analysis of chemical constituents of Saussurea plants using HPLC-MS and ESI-MS/MS. In the third chapter, we proposed quality-control standards for the Genus Saussurea based on TLC (thin layer chromatography) and HPLC. The fourth chapter is about chemical compositions of the essential oil from the whole plant of Saussurea plants. The last chapter reviews the research progress of the Genus Saussurea. The first chapter consists of three parts. The first part is about chemical constituents of ethanol extracts from whole plant of Saussurea laniceps Hand.-Mazz. Fifteen compounds were isolated by column chromatography on normal phase silica gel and Sephadex LH-20. Among them, eleven compounds were isolated from this plant for the first time, and two compounds were isolated from Genus Saussurea for the first time. The second part is about chemical constituents of ethanol extracts from whole plant of Saussurea medusa Maxim. Fifteen compounds were isolated by column chromatography on normal phase, reversed phase silica gel and Sephadex LH-20. Five of them were isolated from this plant for the first time, and there is one new flavonoid glucoside which was identified as 6″-O-crotonoyl-homoplantaginin (M-7) based on the evidence of one- and two-dimensional nuclear magnetic resonance, mass spectrometry analysis, and alkaline hydrolysis reaction. The last part is about chemical constituents of ethanol extracts from whole plant of Saussurea tridactyla Sch.-Bip. ex Hook. f.. Seven compounds were isolated by column chromatography on normal phase silica gel and Sephadex LH-20. There is one compound which was isolated from this plant for the first time. The second chapter consists of three parts. In the first part, we analyzed the chemical constituents of S. tridactyla collected from seven different places in Tibet using HPLC-DAD-ESI-MSn. Fourteen peaks in the HPLC were identified by comparison of UV and MS spectra with those of authentic compounds, among which eight common peaks were quantified. In the second part, we analyzed the chemical constituents of eight Saussurea species using HPLC-DAD-ESI-MSn method. Fifteen peaks in the HPLC were identified by comparison of UV and MS spectra with those of authentic compounds and eight main peaks of them were quantified. In the last part, we analyzed the chemical compounds of the above eight Saussurea plants directly by ESI-MS/MS. Thirteen major compounds, including 10 flavonoids and 3 coumarins were easily rapidly identified. The third chapter consists of three parts. In the first part, we proposed a comparative high quality-control standard for S. laniceps, based on quality detection by TLC and quantity analysis by HPLC using two major compounds (umbelliferone and scopoletin) as standard compounds. In the second part, in viewing S. laniceps, S. tridactyla and S. gossypiphora as the members of one family of medicinal herbs, we suggested a quality-control standard based on the TLC detection of the two major compounds (umbelliferone and scopoletin). In the last part, we proposed a quality-control standard for S. medusa based on the TLC detection of its major component (apigenin 7-O-glucoside). The four chapter analyzed the chemical constituents of essential oil of eight Saussurea species. The essential oils were extracted from the whole plants of these samples with water stream distillation. By GC-MS analysis, we identified eighty-three compounds from S. medusa, eighty-three from S. laniceps, fifty-six from S. quercifolia, thirty-four from S. aster, twenty-one from S. kingii, twenty from S. simpsoniana, twenty-four from S. gossypiphora, and twenty from S. tridactyla respetively, which accounted for 70.7%, 76.0%, 82.2%, 55.4%, 49.7%, 70.4 %, 76.2% and 76.7% of the total essential oil, respectively. The last chapter reviews the research progress of the Genus Saussurea.

GRP78 (78 kDa glucose-regulated protein), also known as BiP (immunoglobulin heavy-chain-binding protein), is an essential regulator of endoplasmic reticulum (ER) homeostasis because of its multiple functions in protein folding, ER calcium binding, and controlling of the activation of transmembrane ER stress sensors. In this report, we cloned the full length cDNA of GRP78 (FcGRP78) from Chinese shrimp Fenneropenaeus chinensis. This cDNA revealed a 2,325 bp with 1,968 bp open reading frame encoding 655 amino acids. This is the first reported GRP78 gene in Crustacea. The deduced amino acid sequence of FcGRP78 shared high identity with previously reported insect GRP78s: 86, 87 and 85% identity with GRP78s of Drosophila melanogaster, Aedes aegypti and Bombyx mori, respectively. Northern blot analysis shows that FcGRP78 is ubiquitously expressed in tissues of shrimp. Heat shock at 35A degrees C significantly enhanced the expression of FcGRP78 at the first hour, reached the maximum at 4 h post heat shock, dropped after that and resumed to the normal level until 48 h of post recovery at 25A degrees C. Additionally, differential expression of FcGRP78 was detected in haemocytes, hepatopancreas and lymphoid organ when shrimp were challenged by white spot syndrome virus (WSSV). We inferred that FcGRP78 may play important roles in chaperoning, protein folding and immune function of shrimp.

HSP70 chaperones mediate protein folding by ATP-dependent interaction with short linear peptide segments that are exposed on unfolded proteins. The mode of action of the Escherichia coli homolog DnaK is representative of all HSP70 chaperones, including the endoplasmic reticulum variant BiP/GRP78. DnaK has been shown to be effective in assisting refolding of a wide variety of prokaryotic and eukaryotic proteins, including the -helical homodimeric secretory cytokine interferon- (IFN-). We screened solid-phase peptide libraries from human and mouse IFN- to identify DnaK-binding sites. Conserved DnaK-binding sites were identified in the N-terminal half of helix B and in the C-terminal half of helix C, both of which are located at the IFN- dimer interface. Soluble peptides derived from helices B and C bound DnaK with high affinity in competition assays. No DnaK-binding sites were found in the loops connecting the -helices. The helix C DnaK-binding site appears to be conserved in most members of the superfamily of interleukin (IL)-10-related cytokines that comprises, apart from IL-10 and IFN-, a series of recently discovered small secretory proteins, including IL-19, IL-20, IL-22/IL-TIF, IL-24/MDA-7 (melanoma differentiation-associated gene), IL-26/AK155, and a number of viral IL-10 homologs. These cytokines belong to a relatively small group of homodimeric proteins with highly interdigitated interfaces that exhibit the strongly hydrophobic character of the interior core of a single-chain folded domain. We propose that binding of DnaK to helix C in the superfamily of IL-10-related cytokines may constitute the hallmark of a novel conserved regulatory mechanism in which HSP70-like chaperones assist in the formation of a hydrophobic dimeric "folding" interface.

A novel undecapeptide has been isolated and structurally characterized from the venoms of three species of New World pit vipers from the subfamily, Crotalinae. These include the Mexican moccasin (Agkistrodon bilineatus), the prairie rattlesnake (Crotalus viridis viridis), and the South American bushmaster (Lachesis muta). The peptide was purified from all three venoms using a combination of gel permeation chromatography and reverse-phase HPLC. Automated Edman degradation sequencing and MALDI-TOF mass spectrometry established its peptide primary structure as: Thr-Pro-Pro-Ala-Gly-Pro-Asp-Val-Gly-Pro-Arg-OH, with a non-protonated molecular mass of 1063.18 Da. A synthetic replicate of the peptide was found to be an antagonist of bradykinin action at the rat vascular B2 receptor. This is the first bradykinin inhibitory peptide isolated from snake venom. Database searching revealed the peptide to be highly structurally related (10/11 residues) with a domain residing between the bradykinin-potentiating peptide and C-type natriuretic peptide domains of a recently cloned precursor from tropical rattlesnake (Crotalus durissus terrificus) venom gland. BIP thus represents a novel biological entity from snake venom.

Im Rahmen der Untersuchung wurde aufgezeigt, welchen Stellenwert die sozial-kommunikative Kompetenz neben den übrigen Basiskompetenzen der personalen Kompetenz, Methoden- und Fachkompetenz und der Aktivitäts- und umsetzungsbezogenen Kompetenz für den "Erfolg" von Versicherungsberatern hat. Das Forschungsfeld wurde auf den Arbeitsbereich des Versicherungsberaters eingegrenzt, wobei besonderes Augenmerk auf seine diversen Fähigkeiten und Fertigkeiten im Kundenkontakt gelegt wurde. Als erstes Ziel der Untersuchung galt es soziale Kompetenz theoretisch zu erheben und durch die Erkenntnisse zu einer für die Arbeit passenden Arbeitsdefinition zu gelangen. In welchem Zusammenhang diese Größe mit dem „Erfolg“ von Versicherungsberatern steht, wurde als zweites Hauptziel der Arbeit empirisch geprüft. Dazu wurden in einer Vorstudie die für die Beratertätigkeit relevanten Aspekte anhand der qualitativen Forschungsmethode des problemzentrierten Interviews mit 11 Kunden und 11 Beratern unterschiedlichen Alters, Bildung und Organisationszugehörigkeit erhoben, die Aussagen dem Kompetenzatlas von Heyse&Erpenbeck (2004) zugeordnet und die Daten mit dem Datenverarbeitungsprogramm MAXQDA reduktiv frequenzanalytisch ausgewertet. Ein Fragebogen, der alle Aspekte der Befragung beinhaltet, wurde erstellt, erprobt und durch den Adjektiv-Ratingbogen von SYMLOG und dem Bochumer Inventar zur berufsbezogenen Persönlichkeitsbeschreibung (BIP) erweitert. 45 Versicherungsberater von drei unterschiedlichen Versicherungsunternehmen in Österreich wurden mittels der Forschungsmethode des 360° Feedbacks untersucht, wobei neben der Selbsteinschätzung die Fremdeinschätzung vom unmittelbaren Vorgesetzten, zwei Arbeitskollegen und drei Kunden eingeholt wurde. Die Beurteilung des „Erfolges“ durch den Vorgesetzten schloss die Erhebungsphase ab. Bei den Auswertungen der Fragebögen konnten unterschiedliche Einschätzungstendenzen bei den sehr erfolgreichen Versicherungsberatern und den wenig erfolgreichen bei der Beurteilung ihres Arbeitsverhaltens, ihres sozialen Interaktionsverhaltens und ihrer berufsbezogenen Persönlichkeitsbeschreibung aufgezeigt werden. Weiters wurden die Bedeutung der Schulungsmaßnahmen, die Versicherungsprodukte und das Image der Unternehmen für einen Verkaufsabschluss untersucht. Die Bedeutung der Arbeit für die Praxis erschließt sich vor allem in der Hilfestellung für die wenig erfolgreichen Versicherungsberater.

En Colombia el sector pecuario, es uno de los más representativos para el crecimiento y sostenibilidad de la economía del país. El sector aporta el 9% del PIB, sus ventas al exterior representan el 21% del valor de las exportaciones totales y genera el 19% del empleo a nivel nacional y el 66% en las zonas rurales. En el contexto del municipio de Sesquilé, la producción en el sector pecuario ha generado la inconformidad por parte de los habitantes y productores del municipio. Este trabajo de grado se desarrolla bajo la Metodología de Matriz de Marco Lógico, la cual presenta una dinámica de identificación del problema central que afecta al sector pecuario y permite plantear diferentes ejes para encontrar las alternativas más óptimas de mejoramiento al mismo, con el apoyo de la Alcaldía Municipal, los Presidentes de la Juntas de Acción Comunal y los habitantes. Para cumplir con el objetivo de caracterizar el sector pecuario del Municipio de Sesquilé, se tiene en cuenta variables productivas y logísticas, para la identificación de alternativas que permitan la generación de empleo y de ingresos en la población. Se identificaron problemas de carácter social, político, económico, tecnológico y logístico, a los cuales se establecen una serie de recomendaciones con pautas de cómo mejorar y aprovechar las oportunidades potenciales que brinda el sector.

En este artículo se habla de la experiencia llevada a cabo en la Escola Bressol Municipal de Bip, en la que a lo largo de los últimos años, ha demostrado que los niños y niñas de la etapa infantil, y específicamente los que acuden a la escola Bressol, tienen curiosidades en relación con el mundo que les rodea, las cuales son tanto o más complejas que las de las personas adultas. A lo largo del artículo tratan de describir cómo han convertido en acción didáctica las evidencias de los niños y niñas de primera infancia.

La presente comunicación quiere dar a conocer una importante experiencia de innovación docente desarrollada en el curso 2009/2010 en la que participan 53 profesores y 785 estudiantes. A partir del uso de una herramienta de autoevaluación del aprendizaje del alumnado universitario, a la que se denominó Aprender Pensando nivel Universitario (HAPu) adaptada a formato electrónico por parte de BIP Asesoría Tecnológica y Logiciel Software Factory en colaboración con la Universidad de Valladolid. El objetivo perseguido es promover la mejora del proceso de aprendizaje a partir del entrenamiento en autorregulación

Recently, various approaches have been suggested for dose escalation studies based on observations of both undesirable events and evidence of therapeutic benefit. This article concerns a Bayesian approach to dose escalation that requires the user to make numerous design decisions relating to the number of doses to make available, the choice of the prior distribution, the imposition of safety constraints and stopping rules, and the criteria by which the design is to be optimized. Results are presented of a substantial simulation study conducted to investigate the influence of some of these factors on the safety and the accuracy of the procedure with a view toward providing general guidance for investigators conducting such studies. The Bayesian procedures evaluated use logistic regression to model the two responses, which are both assumed to be binary. The simulation study is based on features of a recently completed study of a compound with potential benefit to patients suffering from inflammatory diseases of the lung.
