Abstract: The Enterobacteriaceae family contains potentially zoonotic bacteria, and their presence in canaries is often reported, though the current status of these in bird flocks is unknown. Therefore, this study aimed to identify the most common genera of enterobacteria from canaries (Serinus canaria) and their antimicrobial resistance profiles. From February to June of 2013, a total of 387 cloacal swab samples from eight domiciliary breeding locations of Fortaleza city, Brazil, were collected and 58 necropsies were performed in canaries, which belonged to the Laboratory of Ornithological Studies. The samples were submitted to microbiological procedure using buffered peptone water and MacConkey agar. Colonies were selected according to their morphological characteristics on selective agar and submitted for biochemical identification and antimicrobial susceptibility. A total of 61 isolates were obtained, of which 42 were from cloacal swabs and 19 from necropsies. The most isolated bacteria was Escherichia coli with twenty five strains, followed by fourteen Klebsiellaspp., twelve Enterobacterspp., seven Pantoea agglomerans, two Serratiaspp. and one Proteus mirabilis. The antimicrobial to which the strains presented most resistance was sulfonamides with 55.7%, followed by ampicillin with 54.1% and tetracycline with 39.3%. The total of multidrug-resistant bacteria (MDR) was 34 (55.7%). In conclusion, canaries harbor members of the Enterobacteriaceae family and common strains present a high antimicrobial resistance rate, with a high frequency of MDR bacteria.

Tutkielman tavoitteena on ensinnäkin selvittää, miten New Public Management (NPM) -doktriini ja muutokset yksityisen ja julkisen sektorin välistä kilpailuneutraliteettiongelmaa korjaavassa lainsäädännössä ovat vaikuttaneet kuntien toimintojen yhtiöittämiseen. Toisena tutkimuksen kohteena on selvittää erilaiset tavat yhtiöittää liiketoimintoja sekä niiden valintaan vaikuttavat tekijät. Empiirinen osio on toteutettu kvalitatiivisella ja deskriptiivisellä tutkimusmetodologialla. Yhtiöittämisiä tarkastellaan tapaustutkimuksina, johon on valittu tarkemman tutkimuksen kohteeksi kaksi toimialaa: tilahallinto ja satamateollisuus. Kyseisten yhtiöittämistapausten avulla pyritään tuomaan monipuolisesti esille erilaisia yhtiöittämisen taustatekijöitä ja yhtiöittämistapoja. On varsin helppo havaita ne tapaukset, joissa yhtiöittämisen taustalla on ollut tulevaan yhtiöittämisvelvollisuuteen varautuminen. Tähän ryhmään kuuluvat muun muassa lähes kaikki kuntien liikelaitosmuodossa toimivat satamat. Puolestaan kaikki tehokkuuteen, päätöksenteon nopeutumiseen ja muihin liikemaailmasta omaksuttuihin elementteihin perustuvat tavoitteet yhtiöittämisen taustalla on pääsääntöisesti helppo luokitella NPM-doktriiniin liittyviksi. Kuntien tilahallinnot kuuluvat tähän ryhmään, koska niitä ei pääsääntöisesti koske yhtiöittämisvelvollisuus. Yhtiöittämisessä on otettava huomioon useita eri aspekteja, ja myös yhtiöittämistapojen välillä on huomattavia eroja. Tämän vuoksi on tarpeen kartoittaa kaikki kulloiseenkin tilanteeseen vaikuttavat tekijät, jotta voidaan löytää optimaalisin yhtiöittämistapa.

Tämän työn tarkoituksena on tutkia esimiestoiminnan haasteita, joita syntyy liikelaitoksen yhtiöittämisessä. Yhtiöittämisen perusteet kumpuavat juridisesta toimintakentästä, joka muodostaa pakon muutokselle. Työn teoriaosassa käydään läpi lyhyesti liikkeen luovutuksen merkitys työsuhteen sääntelyn kannalta ja perehdytään johtamiseen, esimiestyöhön sekä muutoksen aiheuttamien tunteiden huomioimiseen. Tätä teoriaviitekehystä tutkitaan konkreettisen case-yrityksen avulla. Case-yrityksenä on Helsingin Satama, jonka esimiehiä on haastateltu empiirisen aineiston saamiseksi. Tutkimuskysymyksenä tässä työssä on, miten suurta muutosta tulisi johtaa yksilön tunteet huomioiden. Työssä havaittiin, että vaikka viestintävälineet ovat kunnossa ja vaikka niiden käyttö on vähintäänkin kohtalaisella tasolla, ei viestin omaksuminen silti ole niin helppoa. Syitä tähän löydettiin mm. viestimiskulttuurin epämääräisyydestä ja työntekijätason haluttomuudesta käsitellä muutosta. Muutosvastarinta on yrityksessä erittäin vahvaa, mikä kielii epäonnistumisesta edellisen muutoksen johtamisessa. Onnistuneen muutoksen johtaminen tapahtuu vuorovaikutuksessa henkilöstön kanssa. Yhtiöittämismuutos on pyritty viestimään paremmin kuin edellinen suuri muutos. Yhtiöittämisen koettiin tuovan pääasiassa positiivisia asioita, kuten byrokratian vähenemistä. Positiivisiin asioihin keskittyminen parantaa muutoksen onnistumista.

Soil islands on rocky surfaces often harbor aggregated vegetation that consists of insular plant communities. These islands are typical of the rocky outcrops and in various parts of Brazil form the so-called "campos rupestres" vegetation. Four of such sites have been selected in the state of Bahia, Northeast Brazil, for this comparative study on floristics and vegetation structure: three areas situated inside the "Parque Nacional da Chapada Diamantina" (Guiné, Fumaça and "Gerais da Fumaça") and one is at the border of the Environmental Protection Area of "Marimbus-Iraquara" ("Mãe Inácia"). All occurring vegetation islands were studied in four random plots of 10 × 10 m per site. Soil was often shallow, sandy and acidic. Vascular plant species were determined, with respective life forms and canopy coverage areas. The total number of species when all four sites were added was 135, and the number of species per island varied from 2 to 32. The areas of the 214 soil islands varied from 0.015 to 91.9 m², totaling 568 m² in the four sites. Monocotyledon families were dominant, essentially Velloziaceae, as well as Orchidaceae, Bromeliaceae, Amaryllidaceae and Cyperaceae. Among the eudicotyledons, dominant families were mainly Clusiaceae, Asteraceae and Melastomataceae. The biological spectra revealed that phanerophytes and hemicryptophytes predominated among the life forms, while chamaephytes had the largest coverage area. Epilithic and desiccant chamaephytes composed the most conspicuous interspecific associations, and were probably related to early successional processes. Sites closest to one another were not the most similar in structure, indicating that other factors more relevant than distance might be involved in the abundance of species in space.

Acute promyelocytic leukemia (APL) is characterized by the expansion of blasts that resemble morphologically promyelocytes and harbor a chromosomal translocation involving the retinoic acid receptor a (RARa) and the promyelocytic leukemia (PML) genes on chromosomes 17 and 15, respectively. The expression of the PML/RARa fusion gene is essential for APL genesis. In fact, transgenic mice (TM) expressing PML/RARa develop a form of leukemia that mimics the hematological findings of human APL. Leukemia is diagnosed after a long latency (approximately 12 months) during which no hematological abnormality is detected in peripheral blood (pre-leukemic phase). In humans, immunophenotypic analysis of APL blasts revealed distinct features; however, the precise immunophenotype of leukemic cells in the TM model has not been established. Our aim was to characterize the expression of myeloid antigens by leukemic cells from hCG-PML/RARa TM. In this study, TM (N = 12) developed leukemia at the mean age of 13.1 months. Morphological analysis of bone marrow revealed an increase of the percentage of immature myeloid cells in leukemic TM compared to pre-leukemic TM and wild-type controls (48.63 ± 16.68, 10.83 ± 8.11, 7.4 ± 5.46%, respectively; P < 0.05). Flow cytometry analysis of bone marrow and spleen from leukemic TM identified the asynchronous co-expression of CD34, CD117, and CD11b. This abnormal phenotype was rarely detected prior to the diagnosis of leukemia and was present at similar frequencies in hematologically normal TM and wild-type controls of different ages. The present results demonstrate that, similarly to human APL, leukemic cells from hCG-PML/RARa TM present a specific immunophenotype.

Helicobacter pylori adhesion to gastric epithelial cells constitutes a key step in the establishment of a successful infection of the gastric mucosa. The high representation of outer membrane proteins in the bacterial genome suggests the relevance of those proteins in the establishment of profitable interactions with the host gastric cells. Gastric epithelial cells are protected by a mucous layer gel, mainly consisting of the MUC5AC and MUC6 mucins. In addition to this protective role, mucins harbor glycan-rich domains that constitute preferential binding sites of many pathogens. In this article we review the main players in the process of H. pylori adhesion to gastric epithelial cells, which contribute decisively to the high prevalence and chronicity of H. pylori infection. The BabA adhesin recognizes both H-type 1 and Lewis b blood-group antigens expressed on normal gastric mucosa of secretor individuals, contributing to the initial steps of infection. Upon colonization, persistent infection induces an inflammatory response with concomitant expression of sialylated antigens. The SabA adhesin mediates H. pylori binding to inflamed gastric mucosa by recognizing sialyl-Lewis a and sialyl-Lewis x antigens. The expression of the BabA and SabA adhesins is tightly regulated, permitting the bacteria to rapidly adapt to the changes of glycosylation of the host gastric mucosa that occur during infection, as well as to escape from the inflammatory response. The growing knowledge of the interactions between the bacterial adhesins and the host receptors will contribute to the design of alternative strategies for eradication of the infection.

This study aimed to evaluate the presence of enterotoxigenic S. aureus in the endogenous starter and in Artisan Minas cheeses from the Serra da Canastra. Sixteen samples of endogenous starters and cheese were collected during the rainy and dry seasons. The isolation and enumeration of S. aureus were performed using the PetrifilmTM-Rapid S. aureus Plate Count method. The presence of enterotoxin in the cheese samples was analyzed by the Optimal Sensitivity Plate (OSP) method and the ELFA-VIDAS®-Staph enterotoxin-II assay. S. aureus strains were tested for their ability to produce enterotoxins using the Optimal Sensitivity Plate (OSP) method and the polymerase chain reaction (PCR) assay for the classical enterotoxin genes. The Optimal Sensitivity Plate (OSP) method data showed that staphylococcal enterotoxin A (SEA) was detected in 75% of the cheese samples, but no toxin was detected with the ELFA-VIDAS method. It was found that 12.5% of the isolated strains produced staphylococcal enterotoxin A (SEA) and staphylococcal enterotoxin C (SEC). When using the the polymerase chain reaction (PCR) assay, only one isolate was found to harbor an enterotoxin gene, contrary our expectations. However, discrepancies between the immunological and molecular assays are not uncommon. Despite the fact that most isolates did not produce classical enterotoxins, high S. aureus counts in the cheese samples causes concern since there is a risk of the presence of non-classical enterotoxins.

The microenvironment within the tumor plays a central role in cellular signaling. Rapidly proliferating cancer cells need building blocks for structures as well as nutrients and oxygen for energy production. In normal tissue, the vasculature effectively transports oxygen, nutrient and waste products, and maintains physiological pH. Within a tumor however, the vasculature is rarely sufficient for the needs of tumor cells. This causes the tumor to suffer from lack of oxygen (hypoxia) and nutrients as well as acidification, as the glycolytic end product lactate is accumulated. Cancer cells harbor mutations enabling survival in the rough microenvironment. One of the best characterized mutations is the inactivation of the von Hippel-Lindau protein (pVHL) in clear cell renal cell carcinoma (ccRCC). Inactivation causes constitutive activation of hypoxia-inducible factor HIF which is an important survival factor regulating glycolysis, neovascularization and apoptosis. HIFs are normally regulated by HIF prolyl hydroxylases (PHDs), which in the presence of oxygen target HIF α-subunit to ubiquitination by pVHL and degradation by proteasomes. In my thesis work, I studied the role of PHDs in the survival of carcinoma cells in hypoxia. My work revealed an essential role of PHD1 and PHD3 in cell cycle regulation through two cyclin-dependent kinase inhibitors (CKIs) p21 and p27. Depletion of PHD1 or PHD3 caused a cell cycle arrest and subjected the carcinoma cells to stress and impaired the survival.
